A summary of the resource.
Serum copper levels in dairy cattle have been studied in eight locations throughout Department of Salto, in Uruguay. The overall incidence of hypocupraemia lies in 30% which is not homogeneously distributed since some farms are almost exempt from hypocupraemical individuals. Copper complexes with Cu(Phe)2, Cu(Gly)2.H2O and Cu(Val)2 were evaluated as injectable sources of supplemental Cu in the most hypocupraemic herd and Cu(Phe)2 proved to be the best at improving Cu status. The efficacy of Cu(Phe)2 was tested in a further hypocupraemic herd, showing that the treated cattle maintained the serum copper level at an adequate level for at least 100 days. At the same time, analyses of copper, molybdenum, sulphur and phosphorous levels in grass were done along the year. Our results show that the copper content in grass was under adequate levels most part of the year, and the deficiency was more critical in autumn. Although the molybdenum and sulphur levels were low enough not to expect them to interfere in copper absorption, the Mo content may be enhancing the copper deficiency.

Abstract
A summary of the resource.
The aim of this study is to evaluate the possible incidence in the dosage adjustment of Theophylline. An in vivo study was carried out with 6 healthy volunteers after the administration of an infusion of "mate herb" for a period of 7 days. Plasma Theophilline and Caffeine levels were analysed by high performance liquid chromatography (HLPC) |b revealing the formation of Theophylline as a metabolism product of Caffeine. The analysis of the infusion showed presence of Caffeine and Theobromine |b but not Theophylline. After 7 days of administration the Theophylline concentration oscillated between 0.62 and 1.59 ug/ml for the different individuals; these values can be increased with concomitant intake of other beverages containing xanthines.

El objetivo general es poder comprender las relaciones entre los procesos de configuración de la epistemología ambiental y la educación ambiental, los enfoques metodológicos y los diseños curriculares, en ambas instituciones. En el desarrollo de este escrito se exponen referencias al proceso de institucionalización de la educación ambiental en la Educación Superior de la República Argentina y en la República del Uruguay, enmarcado en el contexto de reflexión de la educación universitaria en el MERCOSUR. Luego se exponen reflexiones sobre la conformación de la red y los diversos intercambios y movilidades efectuadas. Finalmente se hace hincapié en la experiencia colaborativa del trabajo en red que apunta a la visión de interdisciplina puesta en juego. Estamos interesados en contextualizar la reflexión sobre nuestras prácticas en un proceso de integración creciente a nivel de la Educación Superior en el MERCOSUR, en este caso específico, referido a la educación ambiental.

In spite of high plant growth regulator application rates, little has been reported in the literature on determination of their residues in fruits and vegetables. This would be useful in monitoring good manufacturing practices and overall safety through the enforcement of maximum residue levels (MRLs). The present work describes method validation for the determination of 1-naphthylacetic acid (NAA) and 2(1-naphthyl)acetamide (NAAm) in tomato and zucchini using the mini-Luke, ethyl acetate (EtOAc) and acetate-buffered quick, easy, cheap, effective, rugged, and safe (QuEChERS) methods. Samples were spiked at two different levels: 50 and 100 μg/kg for NAA and 20 and 100 μg/kg for NAAm. These compounds were analyzed within the same chromatographic run with LC coupled to triple quadrupole MS (LC/(QqQ)MS/MS) in positive and negative electrospray ionization [ESI(+) and ESI(–)] modes for NAAm and NAA, respectively. For analyte confirmation, LC/ESI(-)QTOF-MS was also investigated given that NAA has only one multiple reaction monitoring transition (185.1→140.9 m/z). These three common methods were used to determine linearity, recoveries, precision (RSD), matrix effects, repeatability, and reproducibility (n = 5) for the selected matrixes. In terms of the Directorate-General for Health and Consumers (DG-SANCO) guidelines, only insignificant differences were found for the multiresidue methods tested, regardless of the commodity. Matrix-matched calibration was used, and LODs were below 10.1 μg/kg for NAA and 6.0 μg/kg for NAAm, which were lower than the MRLs established in current European Union legislation for these compounds. Obtained recoveries for NAA ranged from 87 to 107% with RSD values below 10% for mini-Luke, 83 to 107% with RSD 0.996) for all methods used. Good repeatability and reproducibility in terms of RSD were obtained (11.3–17.8%) for these analytical methods. The results were acceptable within the studied ranges for both analytes in all methods. As these multiresidue methods are used at the present time in many laboratories, this survey indicates that these analytes should be included in their monitoring scope.

Research background. New sources of docosahexaenoic acid have recently been investigated aiming at infant formula fortification and dietary supplementation, among which the single cell oil with 40–50 % of this acid. Experimental approach. For this purpose, such an oil was blended with caprylic acid in amount substance ratio ranging from 1:1 to 5:1 and the blends were interesterified using either Novozym 435 or Lipozyme TL IM as the catalyst. The influence of the amount of excess free caprylic acid in the substrate, as well as the type of enzyme on the triacylglycerol rearrangement resulting from the synthesis of the structured lipids were evaluated. Results and conclusions. The regiospecific lipase Lipozyme TL IM seemed to induce transesterification among single cell oil triacylglycerols preferably by acidolysis with caprylic acid, which was directly proportional to the ratio of this acid in the substrate. In reactions catalyzed by the non-regiospecific lipase Novozym 435, a higher incorporation of caprylic acid into single cell oil triacylglycerols was observed than when using Lipozyme TL IM, independently of the oil/caprylic acid molar ratio. Novelty and scientific contribution. These results revealed the importance of combining the choice of the type of lipase, either regiospecific or not, with the amount ratios of free fatty acids and the substrate in acidolysis when aiming to produce structured lipids as a source of docosahexaenoic acid.

Discovery, characterisation and use of novel yeast strains for winemaking is increasingly regarded as a way for improving quality and to provide variation, including subtle characteristic differences in fine wines. The objective of this work was to evaluate the use of a native apiculate strain, selected from grapes, Hanseniaspora vineae (H. vineae) 02/5A. Fermentations were done in triplicate, working with 225 L oak barrels, using a Chardonnay grape must. Three yeast fermentation strategies were compared: conventional inoculation with a commercial Saccharomyces cerevisiae strain, ALG 804, sequential inoculation with H. vineae and then strain ALG 804 and spontaneous fermentation. Yeast strain identification was performed during fermentation, in which the apiculate strain was found to be active, until 9% of alcohol in volume, for the co-fermentation and the spontaneous fermentation was completed by three native S. cerevisiae strains. Basic winemaking parameters and some key chemical analysis, such as concentration of glycerol, biogenic amines, organic acids, and aroma compounds were analysed. Sensory analysis was done using a trained panel and further evaluated with professional winemakers. Sequential inoculation with H. vineae followed by S. cerevisiae resulted in relatively dry wines, with increased aroma and flavour diversity compared with wines resulting from inoculation with S. cerevisiae alone. Wines produced from sequential inoculations were considered, by a winemaker’s panel, to have an increased palate length and body. Characteristics of wines derived from sequential inoculation could be explained due to significant increases in glycerol and acetyl and ethyl ester flavour compounds and relative decreases in alcohols and fatty acids. Aroma sensory analysis of wine character and flavour, attributed to winemaking using H. vineae, indicated a significant increase in fruit intensity described as banana, pear, apple, citric fruits and guava. GC analysis of the relative accumulation of 23 compounds to significantly different concentrations for the three fermentation strategies is discussed in relation to aroma compound composition

In this study, several approaches were tested to optimise the production and recovery of the widely used anticancer drug Taxol® (paclitaxel) from culturable vascular stem cells (VSCs) of Taxus baccata, which is currently used as a suc‑ cessful cell line for paclitaxel production. An in situ product recovery (ISPR) technique was employed, which involved combining three commercial macro-porous resin beads (HP-20, XAD7HP and HP-2MG) with batch and semi-continu‑ ous cultivations of the T. baccata VSCs after adding methyl jasmonate (Me-JA) as an elicitor. The optimal resin combi‑ nation resulted in 234±23 mg of paclitaxel per kg of fresh-weight cells, indicating a 13-fold improved yield compared to the control (with no resins) in batch cultivation. This resin treatment was further studied to evaluate the resins’ removal capacity of reactive oxygen species (ROS), which can cause poor cell growth or reduce product synthesis. It was observed that the ISPR cultivations had fourfold less intracellular ROS concentration than that of the control; thus, a reduced ROS concentration established by the resin contributed to increased paclitaxel yield, contrary to previous studies. These paclitaxel yields are the highest reported to date using VSCs, and this scalable production method could be applied for a diverse range of similar compounds utilising plant cell culture.

Recombinant single domain antibodies (nanobodies) constitute an attractive alternative for the production of neutralizing therapeutic agents. Their small size warrants rapid bioavailability and fast penetration to sites of toxin uptake, but also rapid renal clearance, which negatively affects their performance. In this work, we present a new strategy to drastically improve the neutralizing potency of single domain antibodies based on their fusion to a second nanobody specific for the complement receptor CD11b/CD18 (Mac-1). These bispecific antibodies retain a small size (~30 kDa), but acquire effector functions that promote the elimination of the toxin-immunocomplexes. The principle was demonstrated in a mouse model of lethal toxicity with tetanus toxin. Three anti-tetanus toxin nanobodies were selected and characterized in terms of overlapping epitopes and inhibition of toxin binding to neuron gangliosides. Bispecific constructs of the most promising monodomain antibodies were built using anti Mac-1, CD45 and MHC II nanobodies. When co-administered with the toxin, all bispecific antibodies showed higher toxin-neutralizing capacity than the monomeric ones, but only their fusion to the anti-endocytic receptor Mac-1 nanobody allowed the mice to survive a 10-fold lethal dose. In a model of delayed neutralization of the toxin, the anti- Mac-1 bispecific antibodies outperformed a sheep anti-toxin polyclonal IgG that had shown similar neutralization potency in the co-administration experiments. This strategy should have widespread application in the development of nanobody-based neutralizing therapeutics, which can be produced economically and more safely than conventional antisera.

Con el propósito de analizar la flexibilidad en dos diferentes planes de estudio de una misma carrera de grado a nivel universitario, se realizó una búsqueda bibliográfica de indicadores que dieran cuenta de la misma. Como resultado de esa búsqueda, en el presente trabajo se definen una serie de indicadores de flexibilidad curricular y se analizan los resultados de su aplicación. Se trabaja con cinco indicadores que dan cuenta de la comunalidad, de la autonomía y especificidad así como de las posibilidades de movilidad intra e intercarreras. Los resultados se presentan en forma de tablas comparativas para ambos planes y se comentan los aspectos más relevantes en relación a los valores obtenidos.

El objetivo de este trabajo es estudiar el comportamiento diferencial de dos indicadores de rendimiento académico mediante modelos de regresión lineal multivariante: el indicador tradicional –promedio de calificaciones– y un indicador novedoso –el avance en la carrera– definido como la razón entre los créditos acumulados por el alumno durante cierto período de tiempo y los créditos que teóricamente debió acumular en dicho período de acuerdo con el plan de estudios programado. Se trata de una investigación correlacional, de corte transversal, de variables no manipuladas experimentalmente. Participaron 587 estudiantes matriculados en carreras de Química de la Universidad de la República (UdelaR) durante 2000-2003, con al menos una asignatura aprobada durante el período comprendido entre marzo de 2003 y marzo de 2005. El grupo de participantes fue dividido aleatoriamente en dos muestras: estimación –se construyeron los modelos de regresión lineal incluyendo análisis de interacciones y diagnóstico de anomalías– y validación –los modelos fueron validados mediante contrastes de hipótesis de igualdad de parámetros. Los factores explicativos considerados fueron el rendimiento previo, las metas académicas de los estudiantes, la percepción de su propia capacidad y variables sociodemográficas. Los dos indicadores están notablemente correlacionados entre sí (r=0,615, p<0,001) y ninguno correlaciona significativamente con las metas de resultado (promedio, p=0,612; avance, p=0,358). Los modelos multivariantes fueron satisfactoriamente validados y muestran que los indicadores se ven igualmente afectados por el rendimiento previo, que resultó la variable dominante (promedio, p=0,493, p<0,001; avance, p=0,484, p<0,001). Se diferencian en que el promedio además resulta afectado por la capacidad percibida (p=0,175, p<0,001) y, de manera indirecta, por las metas de aprendizaje (p=0,16, p<0,05, controlando por capacidad percibida). El comportamiento de ambos indicadores es similar, siendo los estudiantes que obtienen las peores calificaciones los más susceptibles de presentar retraso curricular. Hay una dominancia del rendimiento previo como predictor de ambas formas de rendimiento futuro.

One hundred twenty-five yeast strains isolated from table grapes and apples were evaluated for the control Botrytis cinerea of in vitro and in vivo. Ten strains were selected for their ability to inhibit mycelial growth of B. cinerea in vitro. In the in vivo assays, these yeasts were tested at 20 ◦C on ‘Thompson Seedless’ berries for 7 days; only three were selected (m11, me99 and ca80) because they significantly reduced the incidence of gray mold. These three yeast strains were then evaluated at different concentrations (1 × 107 , 1 × 108 and 1 × 109 cells mL−1 ) on ‘Thompson Seedless’ grape berries at 20 ◦C. The strains m11, me99 and ca80 reduced the incidence of B. cinerea to 11.9, 26.1 and 32.1%, respectively, when the berries were submerged in a yeast suspension at a concentration of 1 × 109 cells mL−1 24 h before inoculation with B. cinerea. The most favorable pH for antifungal activity was 4.6 in the three isolates. The three yeast strains secreted the hydrolytic enzymes chitinase and β-1-glucanase, and two strains (me99 and ca80) produced siderophores. The three yeast strains exhibited low oxidative stress tolerance and only strain m11 had the ability to produce biofilms. The strains were identified using 5.8S-ITS rDNA PCR-RFLP and correspond to the Meyerozyma guilliermondii (m11) and Aureobasidium pullulans (me99 and ca80) species.

<p>The association of autoimmune diseases with particular allellic products of the class-II major histocompatibility complex<br />(MHCII) region implicates the presentation of the offending self-antigens to T cells. Because antigen-presenting cells are<br />tolerogenic when they encounter an antigen under non-inflammatory conditions, the manipulation of antigen presentation may induce antigen-specific tolerance. Here, we show that, in mouse models of experimental autoimmune encephalomyelitis, type 1 diabetes and rheumatoid arthritis, the systemic administration of a single dose of nanobodies that recognize MHCII molecules and conjugated to the relevant self-antigen under non-inflammatory conditions confers long-lasting protection against these diseases. Moreover, co-administration of a nanobody–antigen adduct and the glucocorticoid dexamethasone, conjugated to the nanobody via a cleavable linker, halted the progression of established experimental autoimmune encephalomyelitis in symptomatic mice and alleviated their symptoms. This approach may represent a means of treating autoimmune conditions.</p>

Significant protective immunity against secondary hydatidosis in mice was achieved by immunization with a preparation of surface molecules of E. granulosus protoscoleces with Freund's incomplete adjuvant (PSEx-IFA). Study of PSEx-IFA immunogenicity demonstrates that glucidic epitopes evoke mainly IgM responses while peptidic epitopes evoke mainly IgG responses; however both types of epitopes elicit both types of responses. Analysis of the possible association between susceptibility or resistance to infection and antibody responses after challenge was also performed. Cyst fluid antigen (CFAg) specific antibody titres on month eight after challenge correlated with number and size of cysts. On the other hand no correlation was observed between protection and PSEx specific IgG titres either on the day of challenge or one month later. Nonetheless, immunoblot analysis revealed that some PSEx molecules were recognized on day 30 after challenge only by sera from immunized mice.

Electron spectroscopy for chemical analysis (ESCA) was performed for [Co(NH3)5Cl](ClO4)2, trans-[Co(NH3)4(NH2CH3)Cl]- (ClO4)2, [Co(NH2CH3)5Cl](ClO4)2 and trans-[Co(NH3)4(NH2CH3)(OSO3)](ClO4) complexes. Comparison of the results for the complexes [Co(NH3)5Cl](ClO4)2 and trans-[Co(NH3)4(NH2CH3)Cl](ClO4)2 shows clearly the electronic influence (qI effect) of the methylamine group on the cobalt and through this on the chlorine atom in trans position. Comparison of [Co(NH2CH3)5Cl](ClO4)2 with trans-[Co(NH3)4(NH2CH3)Cl](ClO4)2 shows that methylation of the four cis-NH3 ligands does not produce a proportional decrease in the binding energy of the cobalt atom, while the electron density of the chloro ligand is not affected. For the complex trans- [Co(NH3)4(NH2CH3)(OSO3)](ClO4) theqI effect is almost completely compensated by the presence of the sulfato group in trans position. Acid dissociation constants are also reported for [Co(NH3)5(OH2)]3q and trans-[Co(NH3)4(NH2CH3)(OH2)]3q ions. The implications of these results for the mechanism of the acid and base hydrolysis reactions of acidopentaaminecobalt(III) complexes are discussed. The preparation of the trans-[Co(NH3)4(NH2CH3)(OSO3)](ClO4) complex through the trans-[Co(NH3)4(NH2CH3)(SO3)]q precursor is also described. q2000 All rights reserved.
