The aim of the present work was to explore children’s reaction to sugar reduction in the context of the imple mentation of nutritional warnings in Uruguay. The study was composed of two sessions involving three evalu ation conditions: tasting without package information, package evaluation without tasting, and tasting with package information. A total of 122 children, ages ranging between 6 and 13 years old (47% girls) were involved in the study. In the first session, children’s hedonic and emotional response to a regular and a sugar-reduced chocolate dairy dessert (without other sweeteners) was evaluated. In the second session, children first evalu ated their expected liking, emotional associations and choice of packages differing in the presence of warning labels for excessive sugar content and cartoon character (2x2 design). Finally, they tasted the selected sample in the presence of the package and evaluated their liking, emotional associations, and intention to re-taste it. Although sugar reduction led to a significant reduction in overall liking, the dessert with 40% sugar reduction showed a mean overall score of 6.5 in a 9-point hedonic scale and was described using positive emoji ( , , ). When the desserts were tasted with package information, no significant differences in the expected overall liking of the regular and sugar-reduced dessert were found. Regarding the effect of packaging elements, the presence of a warning label highlighting excessive content of sugar did not have a significant effect on children’s choice. Instead, children’s choices were defined by the presence of a cartoon character. Results from the present work provide additional evidence about the feasibility of reducing the sugar content and sweetness of dairy products targeted at children and stress the need to regulate the use of cartoon characters on products with an unfavorable nutritional profile.Methodological recommendations for sensory and consumer research with children are also discussed. The serious widespread development of nematode resistance has motivated the use of combined anthelmintic formulations. However, the advantages/disadvantages of the combined use of anthelmintics require further scientific characterization. The goals of the current trial were a) to characterize the pharmacokinetics of closantel (CLO) and moxidectin (MXD) administered both subcutaneously (sc) and orally either separately or co administered (CLO + MXD) to lambs; b) to compare the nematodicidal activity of both molecules given indi vidually or co-administered to lambs infected with resistant nematodes. Seventy (70) Corriedale lambs naturally infected with multiple resistant gastrointestinal nematodes were involved in the pharmacokinetic and efficacy trials. The animals were allocated into six groups (n = 10) and treated with either CLO, MXD, or with the CLO + MXD combined formulation by both the oral and sc routes. Additionally, an untreated control group (n = 10) was included for the efficacy trial. The efficacy was estimated by the faecal egg count reduction test (FECRT). Higher systemic exposure of both CLO and MXD was observed after the sc compared to the oral administration in lambs. The combined administration of CLO + MXD did not markedly alter their disposition kinetics. At 13 days post treatment, the administration of both molecules as a single active principle reached efficacy levels ranging be tween 80% (MXDoral), 84% (CLOoral), 85% (CLOsc), and 92% (MXDsc). The combined oral and sc treatments reached 99% efficacy. No adverse effects were observed after the combined treatment of CLO + MXD, and their co-administration did not show any adverse pharmacokinetic interaction. The combined effect of CLO + MXD successfully restored the maximum efficacy levels, which were not reached by the individual active ingredients. Los vegetales son una fuente rica en macro y micronutrientes. En particular, la papa y el boniato se consumen amplia mente, siendo de los cultivos más importantes a nivel mundial. Por tanto, la contaminación de estos productos debido a la presencia de contaminantes inorgánicos es de gran preocupación. Considerando la creciente tendencia de consumo saludable de estos tubérculos y raíces con su cáscara, rica en fibra y otros nutrientes, el análisis del alimento entero podría brindar información valiosa con relación a su seguridad. Debido a esto, se estudió la presencia de As, Cd y Pb en papas y boniatos pelados y enteros. Para ello, se optimizaron y validaron métodos analíticos según los requerimientos de la Association of Official Analytical Collaboration International (AOAC) para análisis de alimentos. A pesar de que el contenido de arsénico (As), cadmio (Cd) y plomo (Pb) fue en la mayoría de los casos menor a un tercio de los valores máximos permitidos para estos contaminantes, la presencia de Cd se detectó en todos los boniatos enteros y no así en los pelados. El mismo comportamiento se observó para la presencia de Pb en la mayoría de las muestras de boniato analizadas. Esto señala la necesidad de generar información sobre el contenido de estos contaminantes en el alimento entero y disponer de metodologías analíticas para ello, de manera de poder realizar una evaluación de riesgo de acuerdo con los hábitos de consumo. One hundred twenty-five yeast strains isolated from table grapes and apples were evaluated for the control Botrytis cinerea of in vitro and in vivo. Ten strains were selected for their ability to inhibit mycelial growth of B. cinerea in vitro. In the in vivo assays, these yeasts were tested at 20 ◦C on ‘Thompson Seedless’ berries for 7 days; only three were selected (m11, me99 and ca80) because they significantly reduced the incidence of gray mold. These three yeast strains were then evaluated at different concentrations (1 × 107 , 1 × 108 and 1 × 109 cells mL−1 ) on ‘Thompson Seedless’ grape berries at 20 ◦C. The strains m11, me99 and ca80 reduced the incidence of B. cinerea to 11.9, 26.1 and 32.1%, respectively, when the berries were submerged in a yeast suspension at a concentration of 1 × 109 cells mL−1 24 h before inoculation with B. cinerea. The most favorable pH for antifungal activity was 4.6 in the three isolates. The three yeast strains secreted the hydrolytic enzymes chitinase and β-1-glucanase, and two strains (me99 and ca80) produced siderophores. The three yeast strains exhibited low oxidative stress tolerance and only strain m11 had the ability to produce biofilms. The strains were identified using 5.8S-ITS rDNA PCR-RFLP and correspond to the Meyerozyma guilliermondii (m11) and Aureobasidium pullulans (me99 and ca80) species. Objective: To study the quality of potato chips for sale in the Uruguayan market. Theoretical framework: For potato chips, the regulation controls moisture, peroxide value (VP) and acidity (%A). It was also considered important to evaluate the fatty material contained in the potatoes as frying oil, where polar compounds (PC) are controlled. The composition of CPs and total polymers (TP) was also evaluated. This set of measures took into account all the deterioration processes to which these lipids are subjected, so as to have a complete idea of their quality. Method: Six different brands of potato chips for sale in the Uruguayan market and the oil absorbed during their elaboration were evaluated. The lipid content and moisture content of the potatoes were measured. In the oil, VP, %A, CP and its composition, PT, the composition in fatty acids and the content of cabbage were measured. Results and conclusion: All the products analyzed comply with the current regulations, except for sample E, which presented high %A. Nevertheless, it was observed that in some cases the oils have an advanced deterioration. Likewise, the sample obtained through vacuum frying with a significantly lower lipid content than the others was highlighted. Implications of the research: This paper seeks to inform the quality of these foods that are for sale in the market. Originality/Value: The quality of food has a direct influence on the health of the population. Antigen tests have been crucial for managing the COVID-19 pandemic by identifying individuals infected with SARS-CoV-2. This remains true even after immunity has been widely attained through natural infection and vaccination, since it only provides moderate protection against transmission and is highly permeable to the emergence of new virus variants. For this reason, the widespread availability of diagnostic methods is essential for health systems to manage outbreaks effectively. In this work, we generated nanobodies to the virus nucleocapsid protein (NP) and after an affinity-guided selection identified a nanobody pair that allowed the detection of NP at sub-ng/mL levels in a colorimetric two-site ELISA, demonstrating high diagnostic value with clinical samples. We further modified the assay by using a nanobody-NanoLuc luciferase chimeric tracer, resulting in increased sensitivity (detection limit = 61 pg/mL) and remarkable improvement in diagnostic performance. The luminescent assay was finally evaluated using 115 nasopharyngeal swab samples. Receiver Operating Characteristic (ROC) curve analysis revealed a sensitivity of 78.7% (95% confidence interval: 64.3%-89.3%) and specificity of 100.0% (95% confidence interval: 94.7%-100.0%). The test allows the parallel analysis of a large number of untreated samples, and fulfills our goal of producing a recombinant reagent-based test that can be reproduced at low cost by other laboratories with recombinant expression capabilities, aiding to build diagnostic capacity. Introduction: The identification of chemical compounds that interfere with SARS CoV-2 replication continues to be a priority in several academic and pharmaceutical laboratories. Computational tools and approaches have the power to integrate, process and analyze multiple data in a short time. However, these initiatives may yield unrealistic results if the applied models are not inferred from reliable data and the resulting predictions are not confirmed by experimental evidence. Methods: We undertook a drug discovery campaign against the essential major protease (MPro) from SARS-CoV-2, which relied on an in silico search strategy –performed in a large and diverse chemolibrary– complemented by experimental validation. The computational method comprises a recently reported ligand-based approach developed upon refinement/learning cycles, and structure-based approximations. Search models were applied to both retrospective (in silico) and prospective (experimentally confirmed) screening. Results: The first generation of ligand-based models were fed by data, which to a great extent, had not been published in peer-reviewed articles. The first screening campaign performed with 188 compounds (46 in silico hits and 100 analogues, and 40 unrelated compounds: flavonols and pyrazoles) yielded three hits against MPro (IC50 ≤ 25 μM): two analogues of in silico hits (one glycoside and one benzo-thiazol) and one flavonol. A second generation of ligand-based models was developed based on this negative information and newly published peer-reviewed data for MPro inhibitors. This led to 43 new hit candidates belonging to different chemical families. From 45 compounds (28 in silico hits and 17 related analogues) tested in the second screening campaign, eight inhibited MPro with IC50 = 0.12–20 μM and five of them also impaired the proliferation of SARS-CoV-2 in Vero cells (EC50 7–45 μM). Discussion: Our study provides an example of a virtuous loop between computational and experimental approaches applied to target-focused drug discovery against a major and global pathogen, reaffirming the well-known “garbage in, garbage out” machine learning principle. Global warming has a strong impact on the polar regions, in particular, the Antarctic Peninsula and nearby islands. Methane (CH4) is a major factor in climate change and mitigation of CH4 emissions can be accomplished through microbial oxidation by methanotrophic bacteria. Understanding this biological process is crucial given the shortage of research carried out in this geographical area. The aim of this study was to characterise psychrophilic enrichment cultures of aerobic methanotrophs obtained from lake sediments of the Fildes Peninsula (King George Island, South Shetland Islands) and revealing the distribution of the genus Methylobacter in different lake sediments of the peninsula. Four stable methanotrophic enrichment cultures were obtained and analysed by metagenome-assembled genomes (MAGs). The phylogeny of methanotroph MAGs recovered from these enrichment cultures based on the 16S rRNA gene showed that K-2018 MAG008 and D1-2020 MAG004Ts clustered within the Methylobacter clade 2, with high similarity to Methylobacter tundripaludum SV96T (97.88 and 98.56% respectively). However, the average nucleotide identity (ANI) and digital DNA-DNA hybridization (dDDH) values with M. tundripaludum were < 95% (84.8 and 85.0%, respectively) and < 70% (30.2 and 30.3%, respectively), suggesting that they represent a putative novel species for which the name ‘Ca. Methylobacter titanis’ is proposed. This is the first species of clade 2 of the genus Methylobacter obtained from Antarctica. The bacterial diversity assessed by 16S rRNA gene sequencing of 21 samples of different lakes (water column and sediments) revealed 54 ASVs associated with methanotrophs and the genus Methylobacter as the most abundant. These results suggest that aerobic methanotrophs belonging to the Methylobacter clade 2 would be the main responsible for CH4 oxidation in these sediments. En este sentido, es necesario realizar un control sobre los niveles de Cd en estos alimentos, contando con métodos analíticos confiables y amigables con el medio ambiente. Con ese objetivo se desarrolló y validó un método analítico para la determinación de Cd en cocoas puras y en su preparación con leche, la forma de consumo más popular en nuestro país. El método resultó adecuado para el fin definido y presentó como ventaja un menor consumo de ácidos concentrados que el método de referencia de la AOAC International (2016b). Se aplicó para un sondeo del contenido de Cd en muestras disponibles en el mercado local de Montevideo, Uruguay. The β-lactoglobulin protein from cow’s milk is a common allergen present in human milk that leads to serious allergic reactions in susceptible infants. This creates a serious safety issue in Human Milk Banks, and the best practice to assure the safety of susceptible newborns would be to classify the donated human milk according to its content of β-lactoglobulin. To provide a simple and fast assay to detect this molecule in human milk we developed a nanobody-based sandwich ELISA. The capture and detection nanobodies were isolated by phage display from a llama immunized with the antigen using a sequential selection strategy for antibody pair iden tification. This allowed to attain a highly sensitive test with a limit of quantitation of 40 pg/mL and extended lineal range of up to 3000 pg/mL, showing an excellent performance with human milk. For these characteristics, the ELISA developed might be useful for the classification of donated human milk according to their β-lacto globulin content, improving the quality of milk destined to susceptible newborns. A pilot annual monitoring survey (April 2018–March 2019) was conducted to investigate the presence of pesticides in superficial water and fish in Laguna del Cisne, one of the most critical drinking water sources in Uruguay. A total of 25 pesticide residues were detected in superficial water (89.3 % of the samples). Pesticide's temporal distribution was as sociated with crops and livestock practices, with higher occurrences in spring and summer than in autumn and winter. The most frequent compounds in superficial water were the insecticide chlorantraniliprole, and the herbicides glyph osate (including its metabolite AMPA) and metolachlor. The levels of Organochlorine pesticide, p,p′-DDT, was in some cases two order of magnitude above the international water quality guidelines for Ambient Water Criteria. In fishes, eight different pesticides were detected, at concentrations from 1000 to 453,000 ng·kg−1 . The most frequent pesticides found were propiconazole, chlorpyrifos, and p,p′-DDE. The widespread occurrence of pesticides in fish suggests poten tial exposure effects on fish populations and the aquatic ecosystem. The sampling approach of this work allowed mon itoring the continuous concentrations of several pesticides in surface waters and fishes to establish the influence from past and current agriculture practices in Laguna del Cisne basin. For safety measures, continuous monitoring programs must be performed in this system to prevent toxicity impacts on aquatic organisms and human health. Penicillium digitatum is the main species responsible for postharvest losses in citrus fruit and, therefore, rapid and accurate methods are needed for its early detection. For this purpose, a new procedure based on real-time quantitative PCR (qPCR) was developed to detect and quantify P. digitatum in lemons. The procedure included a rapid extraction of fungal DNA using the commercial Quick-DNA Fungal/Bacterial Miniprep Kit; the design of a specific primers pair by selecting the conserved region of the calmodulin A (cmdA) gene; and the design and evaluation of a qPCR system based on SYBR-Green dye detection chemistry. The functionality of the developed method was demonstrated by the high linear correlation of the standard curve constructed over the entire range of fungal DNA concentrations used (R2 > 0.99), indicating precision and accuracy of the qPCR. The protocol was developed aiming to evaluate the biotechnological potential of the biocontrol yeast Clavispora lusitaniae AgL21, by quantifying the fungus P. digitatum in lemon wounds. The yeast was shown to restrict the pathogen growth, reaching fungal DNA concentrations of up to 0.038 ng/wound after 4 days of incubation, compared to 173.06 ng/wound when the fungus was alone. No inhibition of qPCR due to food matrices was observed. The developed qPCR protocol could be considered as a suitable tool to detect P. digitatum in food products and can be used to specifically test the ability of new biological control microorganisms against green mold in lemons. Hv exhibited fermentation kinetics comparable to controls when its nutritional requirements were satisfied. Volatile acidity in Hv from one batch was significantly lower (–0.3 g/L) compared to the controls, while in the other two batches the levels were equal. Interestingly, Hv trials showed a significant degradation of malic acid (–0.9 g/L on average) without variations of pH. Even though the matrix influenced the volatile composition, the characteristic features of H. vineae remained intact, with 24-fold higher levels of 2-phenylethyl acetate. There was no higher production of 2-aminoacetophenone, albeit tryptophan (known as a 2-aminoacetophenone precursor) is a preferred amino acid for H. vineae. Results suggest the potential of H. vineae to produce base wine with a broad spectrum of aroma compounds that increase flavor complexity, if obtained from nutrient-balanced grape musts. Kefir is a fermented probiotic drink obtained by placing kefir granules in a suitable substrate. The kefir granules are a consortium of bacteria and yeasts embedded in a exopolysaccharide matrix. The aim of this research was the isolation and identification of yeasts from kefir of different origin, the evaluation of their antifungal capacity against Aspergillus spp. and the characterization of virulence related traits. Using RFLP of ITS1/ITS4 region, D1/D2 region sequencing and RAPD techniques, 20 kefir isolates were identified as Geotrichum candidum, Pichia kudriavzevii, P. membranifaciens, Saccharomyces cerevisiae and Candida ethanolica. Their antifungal capacity was evaluated by their conidia germination reduction, which allowed the selection of eight isolates with high to moderate conidia germination reduction against A. flavus and A. parasiticus. Furthermore, these selected isolates showed growth inhibition on contact in the dual culture assay for both Aspergillus species and 3 of them ‒belonging to S. cerevisiae and P. kudriavzevii species‒ generated volatile organic compounds which significantly affected the growth of both fungi. For the evaluation of virulence related traits, growth at high temperatures, enzymatic activities and the adhesion to Caco-2 cells were analyzed. The isolates did not present more than one positive virulence-related trait simultaneously. In particular, it is important to highlight that the adhesion capacity to the model of intestinal barrier was extremely low for all of them. According to the results obtained, further studies would be of interest for the possible use of these promising yeasts as biocontrol agents against fungi in food. Cold tolerance at germination and seedling stage is one of the most seeked traits in Southern Brazilian rice (Oryza sativa L.) cultivars, thus, the availability of improved methods for germplasm assessment is of high importance for breeding programs. The aim of this study was to validate an optimized Average Tolerance Index (ATI) for rice germplasm assessment under cold at germination and early seedling stage. A diverse panel composed by 124 rice accessions was assayed. Four genotypes with known performance under cold were used as controls (two tolerant and two sensitive). The genotypes were sown in roll papers, and evaluated at 13 °C for 28 days (cold stress) and 25 °C for seven days (control). Four traits were measured, germination percentage, coleoptile length, root length and shoot length, which were converted in terms of relative perfor‑ mance (cold/control). A PCA analysis was carried out. The ATI was calculated as a linear combination of all trait’s relative performance, weighted by the importance of each trait in explaining the genetic variability in the set, through including the PC1 vectors as trait coefcients. K means was applied for genotype classifcation. The panel showed expressive genetic variability for performance under low temperature. The ATI successfully distinguished the controls regarding their cold tolerance and allowed the formation of four classes of genotypes, sensitive, moderately sensitive, moderately tolerant and tolerant. Glutathione transferase enzymes (GSTs) are believed to be a major detoxification system in helminth parasites and have been associated with immunomodulation of the host response. Echinococcus granulosus sensu lato (s.l.) is a cestode parasite known to express at least five different GSTs, but no Omega-class enzymes have been reported in this parasite or in any other cestode. Herein we report the identification of a new member of the GST su perfamily in E. granulosus s.l., which is phylogenetically related to the Omega-class: EgrGSTO. Through mass spectrometry, we showed that the 237 amino acids protein EgrGSTO is expressed by the parasite. Moreover, we identified homologues of EgrGSTO in other eight members of the Taeniidae family, including E. canadensis, E. multilocularis, E. oligarthrus, Hydatigera taeniaeformis, Taenia asiatica, T. multiceps, T. saginata and T. solium. A manual sequence inspection and rational modification yielded eight Taeniidae’s GSTO sequences, each one encoding for a 237 aa polypeptide showing 80.2% overall identity. To the best of our knowledge, this is the first description of genes encoding for Omega-class GSTs in worms belonging to the Taeniidae family -that at least in E. granulosus s.l. is expressed as a protein- suggesting the gene encodes for a functional protein. Vaccines are among the most important advances in medicine throughout the human history. However, conventional vaccines exhibit several drawbacks in terms of design and production costs. Peptide-based vaccines are attractive alternatives, since they can be designed mainly in silico, can be produced cheaply and safely, and are able to induce immune responses exclusively towards protective epitopes. Yet, a proper peptide design is needed, not only to generate peptide-specific immune responses, but also for them to recognize the native protein in the occurrence of a natural infection. Herein, we propose a rational workflow for developing peptide-based vaccines including novel steps that assure the cross-recognition of native proteins. In this regard, we increased the probability of generating efficient antibodies through the selection of linear B-cell epitopes free of posttranslational modifications followed by analyzing the 3D-structure similarity between the peptide in-solution vs. within its parental native protein. As a proof of concept, this workflow was applied to a set of seven previously suggested potential protective antigens against the infection by Echinococcus granulosus sensu lato. Finally, two peptides were obtained showing the capacity to induce specific antibodies able to exert anti-parasite activities in different in vitro settings, as well as to provide significant protection in the murine model of secondary echinococcosis. Almond is a high-market value crop that benefits from honeybee pollination services, even for self compatible varieties. Besides, it has been recently shown that the offering of food scented with floral mimic odors specific to different crops biased honeybee foraging preferences towards sunflowers, pear, or apple trees. Herein, we analyzed the floral volatiles of two almond self-compatible varieties to propose potential mimic odors. The mixture which bees discriminated the least from the natural floral scent in olfactory conditioning assays was chosen as almond mimic (AM). In the field, colonies fed AM-scented sucrose solution increased their foraging activity and amounts of stored pollen compared with colonies fed unscented food. Our results support the conditioning of honeybees to a floral mimic odor as a potential tool to bias their foraging prefer ences in almond, even applicable to self-compatible varieties. Quality control of essential oils is fundamental for verifying their authenticity and confor mity with quality standards, ensuring their safety and regulatory compliance, and monitoring their consistency. Companies that produce or market essential oils routinely evaluate the quality and au thenticity of their products. However, they also must deal with increasing attention to environmental sustainability as well as practical considerations such as productivity, cost, and simplicity of methods. In this study, enantioselective gas chromatography (GC) was adopted to evaluate the quality of sweet and bitter orange essential oils, used as a case study. The analytical conditions were optimized and translated to fast GC to evaluate the impact of this approach on the environmental footprint of the analyses. The greenness of fast GC, compared with conventional GC, was quantitatively evaluated using a dedicated metric tool (AGREE), and important improvements have been calculated. The developed methods were applied to a set of commercial essential oils, and the data about the enantiomeric composition and relative percentage abundance were elaborated through multivariate statistics (principal component analysis). The results showed that fast chiral gas chromatography enables the classification of citrus essential oil samples and can be considered an environmentally friendly and sustainable approach for evaluating their quality. Sodium titanates represent a promising alternative for the construction of anodes for sodium-ion batteries. To study the electrical response of nanostructured sodium titanate samples under plausible temperature conditions, we evaluate the electrical impedance in the range of temperatures from −30 ◦C to 50 ◦C, and the effect of consecutive temperature cycling. Strong changes in electrical properties are observed during the first temperature cycle, followed by a stabilization in the consecutive cycles. This behavior is reversible when the sample is exposed to external conditions and room temperature for less than an hour. Probably a structural and surface water removal and a surface water reabsorption occur, affecting the charge carriers and with it the impedance. Structurally, no differences are detected before and after the cycles, nor when evaluating only the vacuum process. However, when the optical properties are evaluated we find non-reversible changes after the temperature cycles. The optical behavior after the temperature cycling becomes uniform, the same type of electronic defects become predominant and similar direct and indirect band gap values are obtained, regardless of the starting synthesis condition. These results have significant implications for potential technological applications of these materials. Food losses and waste (FLW) is a global problem. Because household FLW is one of the biggest contributors to total FLW, strategies are being implemented to reduce per capita FLW in many countries. The present research contributed to this goal by investigating individual factors influencing self-reported household food waste in Australia. Using a web survey, data were obtained from 847 consumers. The average per capita food waste corresponded to 339 g/week and the median to 180 g/week. The most frequently wasted food categories were fresh vegetables and salads, fresh fruit, and bread. The percentage of participants reporting having disposed of these categories ranged between 45.9% and 65.9%. Although socio-demographic charac teristics have been identified as relevant influencers of household food waste, the evidence is still inconclusive about the strength of their effects. Results from the pre sent work identified significant effects of age and household size. For the former, the amount of per capita household food wasted tended to reduce with participants' age. Regarding the effect of household size, per capita food waste decreased with the number of people in the household. The present research also explored the effect of a series of psychological factors on the amount of food waste and five hypotheses derived based on the postulates of theories of consumer behavior were supported.
