Single-phase oxygen stoichiometric LaMnO3 and doped La0.8A0.2MnO3 (A = Ca, Sr, Ba) perovskites have been prepared by a simple one-step auto-combustion method. Cation-deficient LaMnO3+ and La0.8A0.2MnO3+ were obtained by calcination of the former samples in air at 750 C. The samples were characterized by X-ray powder diffraction, X-ray photoelectron spectroscopy, temperature-programmed reduction, temperature-programmed oxygen desorption, and N2 physisorption in order to apply them as catalysts in the complete catalytic oxidation of acetone as a model volatile organic compound. The studied phases show the expected orthorhombic and rhombohedral perovskite crystal structures. Catalytic experiments performed with all the samples show measurable activity already at 100 C. At 200 C, doped La0.8A0.2MnO3 samples show higher activity than undoped LaMnO3, with increasing conversion with larger A-cation size. Calcined samples also show higher activity than as-prepared ones making La0.8Ba0.2MnO3+ the best catalyst at this temperature. All doped samples show >95% acetone conversion at T 250 C with a weak dependence on the sample processing or A cation doping. The collected evidence confirms that the most important factors for the catalytic activity of these oxides are the Mn4+/Mn3+ molar ratio on the surface of the samples and the cation-deficiency of the bulk perovskite structure. In addition, increasing the symmetry of the bulk crystal structure appears to have an additional favourable effect. Despite the observation of the presence of surface carbonates, we show that it is possible to use the as-prepared samples without further thermal treatment with good results in the oxidation of acetone. To better understanding the consumer choice of functional foods, a choice-based conjoint study was carried out to investigate the influence of three non-sensory factors on consumer choice of functional yogurts over regular ones. Four attributes were considered in the conjoint study: type of yogurt (regular, enriched with antioxidants and enriched with fibre), brand (national familiar, national unfamiliar, and foreign familiar), price (low, regular and high) and claim (with and without ‘reduced disease risk’ health claim). Eighteen choice sets, each composed of three stimuli (regular yogurt, enriched with antioxidants and enriched with fibre and varying the brand, price and claim levels) were evaluated by 103 consumers. For each choice set, consumers had to indicate which yogurt would normally buy. A questionnaire on consumer attitude towards health and nutrition was answered by participants after the choice task. Brand, price and health claims highly significantly affected consumer choice of functional yogurts. The relative importance of brand was similar to that of type of yogurt. Therefore, results suggested that non-sensory factors such as brand and price could have an important impact on consumer perception of functional foods. The impact of brand, price and health claims was affected by consumer interest in health issues. Time orientation can influence health-related behaviors, including food consumption. The aim of the present work was to study the influence of time orientation on food choice, using cookie labels as case study. A choice-conjoint task was designed using labels differing in type of cookie (chocolate chips vs. granola), front-of-pack nutrition information (nutritional warnings vs. Facts Up Front system) and nutritional claim (no claim vs. “0% cholesterol. 0% trans fat”). An online study was conducted, in which 155 participants evaluated 8 pairs of cookie labels and selected the one they would buy if they were in the supermarket. Then, they were asked to complete a consideration of future consequences scale (CFC) adapted to eating habits, as well as a questionnaire about socio-demographic characteristics. Time orientation influenced participants' choices of cookies labels; particularly the importance attached to type of cookie. Participants with greater consideration of future consequences preferred the granola cookies, associated with health, while those who prioritized immediate consequences preferred chocolate chip cookies. In addition, nutritional warnings discouraged choice regardless of participants' time orientation. Results from the present work provide additional evidence of the influence of time preferences on food choices and suggest that strategies to stimulate and generate a more future-oriented perspective on eating habits could contribute to more healthful food choices. Leaf removal is beginning to receive consideration for many growers as an important cultural practice for bunch rots control. It is normally applied after veraison and almost only on red varieties because of the high sunburn susceptibility of white grapes varieties under Uruguayan climatic conditions. The aim of this study was to determine the time and intensity of leaves removal impact on bunch rots and grapes quality on Sauvignon blanc. In parallel, the effects of kaolin (Surround® WP) applications in reducing the negative impact of excessive exposure to high temperatures were evaluated. Different intensities and times of defoliation, in combination with kaolin applications, were evaluated over 2006/07, 2007/08 and 2008/09 seasons. All defoliation treatments and mainly those made after fruit set, significantly reduced the occurrence of bunch rots, having a critical impact on those metabolites responsible for the final taste of wines. Early defoliation was significantly more effective controlling Botrytis incidence and improving berry quality than later in the season. Leaf removal had a determining impact over fruit quality and primary and secondary metabolites contents. An increase in soluble solids and glycosides concentrations was observed, while IBMP grape content was reduced, thus affecting the flavor of Sauvignon blanc wines. Kaolin applications significantly reduced berries temperature, sunburn and other berry damages associated with clusters solar exposure. In sum, leaves removal and exogenous kaolin application in Sauvignon blanc showed great potential as Botrytis mitigation strategy because it positively impacts berry quality as a result of many molecular and biochemical changes in key secondary metabolic pathways. The harmful bloom-forming cyanobacterium Cylindrospermopsis raciborskii grows in freshwaters over a wide range of light conditions. This species has increased its global distribution recently. The influence of ultraviolet radiation (UVR) on the fitness and toxin production of C. raciborskii has not previously been explored. We performed short-term experiments with three C. raciborskii strains (MVCC19, LB2897, and CYP011 K), and we compared their responses with other bloom-forming species (Microcystis sp.1 and Plankthotrix agardhii) to determine the impact of UV-B radiation on pigments, biomass, and morphological traits. In addition, we analyzed the effect of UV-B on the saxitoxin content and sxtU gene expression in the strain MVCC19. C. raciborskii strains were stressed differentially by UV-B exposure as evidenced by changes in growth, morphology, and heterocytes number. A significant increase in saxitoxin concentration and sxtU gene expression under UV-B suggests that toxin production in C. raciborskii can be a response to UV-B stress. In comparison, Microcystis sp.1 was more tolerant, while P. agardhii was severely impacted by UV-B, indicating also different sensitivities among cyanobacteria to UVR. Our results underscore the influence of UVR on C. raciborskii and the differences between strains which showed phenotypic plasticity, which potentially could affect its distribution in freshwaters. La produccion biologica de metano a partir de materia organica es un procedimiento eficaz para reducir la contaminacion de efluentes domesticos e industriales. Las bacterias que intervienen en el proceso componen diferentes grupos troficos que actuan en forma secuencial, la ultima etapa de las cuales es la metanogenesis. En esta tesis se investigo el efecto del Cr+3, un toxico potencial presente en las aguas residuales y en los lodos de las curtiembres, sobre las bacterias de estos diferentes grupos troficost Se establecio un orden de sensibilidad y se determino que las mas afectadas fueron las bacterias metanogenicas degradadoras de acetato. Se encontro correlacion entre la actividad metabolica de estos microorganismos en cultivo puro y formando parte de un ecosistema bacteriano complejo. Ademas de la ,concentracion de cromo, totros parametros como la concentracion de biomasa 0 la presencia de ion ferrico modificaron el comportamiento del sistema y podrfan manejarse para mejorar la eficiencia del proceso Se establecio que las bacterias metanogenicas acetotrofas sobreviven a concentraciones de cromo muy superiores alas inhibitorias de la metanogenesis y que tienen cierta capacidad de adaptacion. Estos resultados tambien pueden contribufr a minimizar la inhibicion de la metanogenesis en un digestor anaerobio. Los ensayos con cultivos puros de bacterias metanogenicas acetotrofas permitieron inferir algunos aspectos del mecanisme de acci6n del cromo. La inhibici6n de la metanogenesis depende de la concentración de ión libre 0 quelado, no interfiere con la captura de metales esenciales, y difiere cuantitativamente segun el sustrato utilizado. La diferencia de tolerancia que se observó entre las bacterias metanogenicas de los dos unicos Generos capaces de degradar acetato permite predecir el comportamiento de un digestor anaerobio alimentado con este tipo de efluente y configurar una hipótesis sobre el desarrollo de estos microorganismos en presencia de un grupo mas amplio de tóxicos. Los glicosidos han despertado un gran iDteres debido a su participacion en multiples procesos biologicos. EI uso de glicosidasas constituye tma alternativa atractiva a su compleja sintesis quimica, permitiendo la formaci6n de enlaces glicosidicos en un solo paso de reaccion y con un control completo de la configuracion del centro anomerico. Como la sintesis de giic6sidos frecuentemente requiere el uso de solventes organicos, las glicosidadas a see utilizadas deben ser estables en dichas condiciones. Una de las estrategias mas utilizadas para la estabilizaci6n de enzimas es su inmovilizacion. Esto a su vez facilita la separacion de la enzima del medio de reaccion, propiedad esencial en procesos de sintesis cineticarnente controlados. Se estudiaron dos estrategias de inmovilizacion de f3-galactosidasas. La primera a traves de la union de grupos amino de la enzima con grupos aldehido del soporte, y la segunda mediante la formaci6n de puentes disulfuro entre grupos tiol expuestos de la enzima y estructuras tiol-reactivas del soporte. La inmovilizacion sobre geles glutaraldehido genero derivados enzimciticos con rendimientos de inmovilizaci6n superiores al 60%, buenas actividades especificas y termicarnente mas estables que la enzima soluble. Se logro la estabilizacion de la f3-galactosidasa de K. lactis en presencia de acetona y DMF (18% v/v). La estabilidad del derivado de f3-galactosidasa de A. oryzae frente al efecto de los cosolventes, aunque no logro mejorar la correspondiente estabilidad de la enzima soluble, fue superior a la del derivado de K factis, Esto 10 biro potencialmente aplicable a la sintesis enzimatica en rnedio orgcinico. Las propiedades de los derivados obtenidos por inmovilización de la enzima sobre soportes tiolS\llfinato-agarosa fueeon difeeentes, dependiendo del origen de la enzima y de la modificación quimica realizada previa a la inmovilizacion. Se obtuvieron derivados con buenas actividades especificas y rendimientos de inmovilizacion superiores al 60 %, pero solo se logro una mayor estabilidad en presencia de cosolventes orgcinicos para el caso del derivado de la f3-galactosidasa de K. factio'S obtenido con la enzima reducida. Se estudio la sintesis de distintos galactósidos a partir de 0-nitrofellil-I3-D-galactopiran6sido (ONPG) y lactosa como dadores del grupo galactosilo y distintos aceptores (4-metilumbeliferona, xilosa, etilenglicol). Se obtuvo 4-metilumbiliferil-f3-D-galactopiranosido con un rendimiento muy bajo. Es destacable la sintesis de galactosil-xilosa con un rendirniento de un 16% para la enzirna soluble, el que aurnent6 a un 28% cuando se utilizó la enizrna inmovilizada, obteniendose una rnezcla de regiois6rneros (f3-D-galactopiranosil (1-1) D-xilopiranosa y f3-D-galactopiranosil (1-4) D-xilopiranosa 0 f3galactopiranosil (1-5) D-xilofuranosa). A su vez resulto interesante la produccion de galactosilletilenglicol con rendiIlljentos de hasta un 70% y 100% de transglicosilacion. En este trabajo se reporta la actividad catalítica de una arcilla pilareada con hierro (Fe-PILC) en la degradación de naranja de metilo mediante la técnica foto-Fenton heterogénea. Se prepararon Fe-PILCs a partir de una arcilla natural empleando diferentes temperaturas de calcinación: 350°C y 500°C. Los sólidos obtenidos se caracterizaron por análisis termogravimétrico y adsorción de nitrógeno, no observándose diferencias significativas de sus parámetros texturales (área específica y volumen de poros). Se estudió la influencia del pH del medio en el proceso catalítico y se verificó que la remoción de naranja de metilo fue efectiva a pH 3 pero no a pH 6. Este comportamiento puede explicarse por cambios en la polaridad de la molécula del colorante que afectan su adsorción sobre la arcilla. A pH 3 se estudió además la influencia de diferentes factores sobre el proceso catalítico. Al administrar un medicamento a un individuo se busca producir una respuesta clínica específica a través de un efecto farmacológico determinado en gran parte por la respuesta farmacocinética, es decir, por las cantidades de fármaco presentes a lo largo del tiempo en los diferentes tejidos y fluidos del cuerpo humano. Medicamento y organismo conforman un complejo sistema que debe ser correctamente interpretado para cumplir con los objetivos terapéuticos de eficacia y seguridad al diseñar un tratamiento farmacológico. La natural variabilidad fisiológica que presentan los individuos a lo largo de una población exige racionalizar la instauración de los mismos, ya que posee un gran impacto en el destino de un fármaco administrado. El sexo como característica fisiológica divide a la población en dos mitades, su relevancia en el campo de la medicina es altamente notoria, hombre y mujer al interaccionar con un medicamento componen sistemas diferentes. Es de esperar entonces que ciertas características específicas al sexo del paciente puedan condicionar el éxito de un tratamiento farmacológico. Durante esta tesis doctoral se analizó la influencia del sexo en la respuesta farmacocinética de principios activos correspondientes a diversos grupos farmacológicos dividiendo el análisis en dos bloques: su efecto sobre la evaluación de biodisponibilidad de medicamentos y su efecto sobre la disposición de fármacos. La diferente discriminación de marcas comerciales de medicamentos que pueden hacer hombres y mujeres, y sus consecuencias sobre las conclusiones de un ensayo de bioequivalencia promedio no habían sido analizadas hasta el momento en profundidad. El análisis realizado demuestra la importancia de tener en cuenta el sexo en la evaluación de bioequivalencia y de diseñar ensayos clínicos a modo de obtener conclusiones sobre ambos grupos. Los resultados hallados aumentan la evidencia respecto a diferentes respuestas farmacocinéticas en hombres y mujeres. Se incorpora la recirculación de fármacos como proceso de disposición relevante, y se introducen modelos no lineales de efectos mixtos para describir y cuantificar su contribución. Al indicar un medicamento, el sexo del paciente debe ser considerado ya que de acuerdo a las características del mismo podría tener un significativo efecto sobre el resultado clínico del tratamiento. The cupric reducing antioxidant capacity (CUPRAC) assay is an established procedure used to measure antioxidant content. Recently, the CUPRAC assay was adapted for use in post column derivatisation coupled with high performance liquid chromatography (HPLC-PCD). In prior works reported in the literature, CUPRAC HPLC-PCD assays used large post column mixing coils leading to a deterioration in the observed separation efficiency. In this work we used the CUPRAC PCD assay with reaction flow (RF) chromatography. No reaction loop was required, thus preserving the efficiency of the separation. We subsequently compared the performance of the CUPRAC assay to that of an antioxidant assay that we have previously converted to RF chromatography, namely the ferric reducing antioxidant potential (FRAP) assay, both in RF mode. The relative response factor with respect to trolox was measured for both RF assays via the separation of a standard mixture of 16 compounds. Sample analysis of coffee and green tea via both RF assays was used to compare their ability to screen for bioactive candidates in complex natural products. In general, the CUPRAC assay is recommended as the RF PCD methodology of choice as it provided a higher level of performance compared to the FRAP assay, demonstrating less baseline noise interference, greater sensitivity, wider linear dynamic range and better assay precision. The myo-inositol phosphates (InsPs) are specific signalling metabolites ubiquitous in eukaryotic cells. Although Ins(1,3,4,5,6)P5 is the second most abundant member of the InsPs family, its certain biological roles are far from being elucidated, in part due to the large number of species formed by Ins(1,3,4,5,6)P5 in the presence of metal ions. In light of this, we have strived in the past to make a complete and at the same time “biological-user-friendly” description of the Ins(1,3,4,5,6)P5 chemistry with mono and multivalent cations. In this work we expand these studies focusing on the inframolecular aspects of its protonation equilibria and the microscopic details of its coordination behaviour towards biologically relevant metal ions. We present here a systematic study of the Ins(1,3,4,5,6)P5 intrinsic acid–base processes, in a non-interacting medium, and over a wide pH range, analyzing the 31P NMR curves by means of a model based on the Cluster Expansion Method. In addition, we have used a computational approach to analyse the energetic and structural features of the protonation and conformational changes of Ins(1,3,4,5,6)P5, and how they are influenced by the presence of two physiologically relevant cations, Na+ and Mg2+.
