According to the hydrolysis performance "in vitro" and lipophylicity |b two Furosemide (F) prodrugs were chosen from a series of acyloxymethylesters of F synthesized previously: PI (acetyloxymethyl-4-chloro-N-furfuryl-5-sulfamoylanthranilate) and P4 (pivaloyloxymethyl-4-chloro-N-furfuryl-5-sulfamoylanthranilate). The bioavailability studies were assayed over two groups of eight male Wistar rats as a randomized two-way crossover and balanced design: group 1) a solution of PI in propylenglycol/ethyl acetate vs an aqueous solution of F |b and group 2) PI vs.P4 solutions in oleic acid (PI,P4).These assays showed a better absorption performance of PI and P4 than F |b while the two prodrugs showed a similar bioavailability. The oleic acid seems to be responsible for the delay in the recovery of 50 of the total amount of F excreted in urine (T50). When the monitoring is done in plasma after the administration of PI |b PI was not detected as circulating prodrug. The analytical determinations of F in urine and plasma were done by high performance liquid chromatography (HPLC). From the urinary excretion data |b a slope that indicates a slow elimination was found with a half-life of 12 hours approximately. A bioavailability study of one of these W/O emulsions was carried out choosing a conventional tablet commercialized in Uruguay as reference. A single-dose study was carried out in eight healthy volunteers in a crossover randomized test. Saliva and plasma samples were taken and analyzed by HPLC. Saliva/plasma ratios enable the observation of absorption inputs sometimes up to 48 hours after administraion. Plasma concentrations show sustained drug levels for the emulsion during the period studied. No significant differences were found for C max,T max and AUC 0-48 h. Similar bioavailability is concluded for both dosage forms. An AM1 and MNDO–PM3 semiempirical study of the reaction mechanism of glyoxalase I is presented. According to some authors, this enzyme serves as a detoxifying agent, transforming ketoaldehydes to hydroxiacids. According to others, however, the enzyme serves as a growth-control agent, destroying 2-oxopropanal (MG), which might be involved in some, mostly unknown, biological pathways. Following a previous study on the conformation of the hemimercaptal of MG and glutathione (HSG) done using molecular mechanics, we study here the possible reaction path of this substrate at a model reaction site in the enzyme. First, the active center of the enzyme was modeled using a Zn2+ cation with several ligands. Second, its complex with the hemimercaptal of MG and HSG (SAG) was calculated using semiempirical methods. The reaction proceeds from the substrate to the enediol intermediate in two steps: In the first one, a proton from the activated carbon in SAG is captured by a base–which we model in this work by HO−, CH3COO−, and imidazole anion. Our calculations show that the likeliness of the reaction is enhanced if imidazole is used as the base instead of an acid conjugate base or a free hydroxide anion. Moreover, we found a stable intermediate carbanion–described for the first time as an intermediate in this mechanism–whose existence is supported by some of the available experimental evidence. In the second step of the mechanism, a proton is transferred from a Zn-bound water to the intermediate carbanion, giving the enediol species, originally postulated as the intermediate in this mechanism, and leaving a hydroxide anion bound to the Zn2+ complex. Comparing this reaction with the same process when no metal complex is present–as if the reaction occurred in aqueous solution–it is shown that the metal ion activates water, facilitating the proton transfer. Our calculations point out the existence of two potential energy barriers to be overcome in the process: the need of the metal center to be coordinated to water as a prerequisite for the second proton transfer to take place, and the fine tuning caused in the first proton transfer by the very nature of the base that initially captures the proton. No significant differences in the qualitative characteristics of the reaction were found in using AM1 or PM3 as the method of calculation. Recently proposed modified MNDO methods (MNDO/M and MNDO-PM3) for the study of hydrogen-bonded complexes are tested in four simple systems: (H2O)2, NH4+ · H2O, formamide water and formamide-methanol dimers. A comparison is made with AM1 since this method, intended to be a general improvement of MNDO, was already shown to fail in the former two dimers. Bifurcated hydrogen-bonded structures were always preferred over linear ones. AM1 also shows this artifact in the formamide-water dimer but not in the formamide-methanol one. This is due to the inability of -CH3 to bind to the carbonyl oxygen. In this case the AM1 optimized global minimum is in excellent agreement with the Hartree-Fock ab initio DZP optimized structure. Moreover, when it is physically impossible to form bifurcated hydrogen bonds, AM1 is in good agreement with ab initio results. MNDO/M and MNDO-PM3 (an improvement on AM1 ) do not give any bifurcated structure either in the former two dimers or in the formamide complexes. MNDO/M and MNDO-PM3 structures are in agreement with ab initio and experimental ones in almost all cases, except for the global minima in both the formamide-water and the formamide- methanol dimers. Singly hydrogen-bonded structures are predicted instead of the experimentally determined and ab initio calculated correct doubly hydrogen-bonded ones. MNDO-PM3, although not entirely correct, gives structures which are more correct than the MNDO/M ones. In conclusion, neither of these semiempirical methods is entirely correct. Overall, MNDO-PM3 seems to be more trustworthy than AM1 or MNDO/M, although these methods may be used when no bifurcated hydrogen bonds (AM1) or cycles (MNDO//M) exist. Failures of MNDO-PM3 indicate that a better semiempirical method is still needed for studies of solvation and hydrogen- bonded clusters. A recopilation of species of fishes used for the study of s-MDH isozymes was made. s-MDH of most fishes was shown to be codified by two gene loci. However, there are fishes with a more complex pattern, with s-MDH composed of subunits codified in three or four gene loci. s-MDH isozymes from ectothermic species proved useful in the study of biochemical adaptation to the environment. The central question addressed was whether adaptive difference in the effect of temperature on an important kinetic parameter, Km, reflect selection based on the range of temperatures experienced by the species, the highest temperature encountered by the species, or both of these factors. For the M4-LDHs of all 15 species, the Km of pyruvate was conserved between 0.1 and 0.4 mM pyruvate over the species' physiological temperature range. However, large increases in Km of pyruvate commonly were found at temperatures a few degrees C above the species' upper habitat temperature. Distinctions were found between the responses of the enzymes of eurythermal and stenothermal sciaenids. For the most eurythermal species, e.g., Sciaenops ocellatus, Micropogonias undulatus, and Roncador stearnsii, Km of pyruvate varied only about twofold between 10 and 30°C, the temperatures which encompass most of the habitat ranges of these species. For the most stenothermal species, e.g., Cynoscion striatus and Micropogonias furnieri, with habitat temperature ranges of 14–18°C, sharp increases in Km occurred at temperatures above 20°C. We hypothesize that thermal stability of Km values is more strongly selected by maximal habitat temperature than by the amount of variation in habitat temperature, and that adaptation to high temperatures may pre-adapt enzymes for eurythermal function. Se analizaron 362 sueros de novillos Hereford y sus cruces, procedentes de casi todos los departamentos del Uruguay, mediante las reacciones de Sabin y Feldman y de hemoaglutinación indirecta para toxoplasmosis. Inactivándolos a 60ºC. 30 minutos, los máximos títulos obtenidos fueron 1:16 y 1:64, respectivamente. Estos títulos no son considerados específicos. Se alimentaron dos gatos seronegativos a Toxoplasma con las muestras de 20 gr. de carne de cada uno de los 129 novillos analizados con la reacción S & F. Se incluyeron muestras de músculos de los cuatro novillos reaccionantes al S & F. No excretaron ooquistes toxoplasmáticos ni se seroconvirtieron después de transcurridos 16 días desde la ingesta supuestamente infectante. Discutiéndose estos resultados, junto con los obtenidos en otros países, se concluye que la carne de novillo no es fuente importante de infección toxoplásmica. Bismuth tri-iodide is a layered compound semiconductor which has suitable properties as material for ionizing radiation detection devices. Monocrystals and polycrystalline thin films have been studied for this application, but only recently, the development of nanostructures of this compound has emerged as an interesting alternative for using such nanostructures in new types of radiation detectors or for including them in other applications. Considering this, we present in this work BiI3 nanoparticles successfully synthesized by the hydrothermal method, using a Teflon-lined stainless steel autoclave, at a temperature of 180 °C during 8–20 h, with BiCl3 and NaI as source materials. We characterized the nanoparticles by X-ray diffraction (XRD), transmission electron microscopy (TEM) and electron dispersive spectroscopy (EDS). We obtained small rounded or hexagonal particles (10–20 nm in size) and larger structures. The maximum orientation of the nanostructures is along the (0 0 l) family planes and occurs after 16 h of synthesis, which arises as the best condition for obtaining BiI3 oriented nanostructures. When a 100 kV TEM electron beam was converged on the larger structures, we obtained highly oriented BiI3 hexagonal and rod shaped nanostructures. We found that particles’ shape does not depend on the synthesis time. In addition, results were compared with the ones obtained for nanoparticles synthesized from solution. The present work is an advance in the synthesis of BiI3 nanostructures by the hydrothermal method, and is also the first step on seeking the amenable control of morphology and size of such structures using electron beam irradiation. This last process may be particularly appropriate for producing nanostructures for future applications in new devices.
