A combination of three-dimensional quantitative structure-activity relationship (3D-QSAR), and molecular modelling methods were used to understand the potent inhibitory NAD(P)H:quinone oxidoreductase 1 (NQO1) activity of a set of 52 heterocyclic quinones. Molecular docking results indicated that some favourable interactions of key amino acid residues at the binding site of NQO1 with these quinones would be responsible for an improvement of the NQO1 activity of these compounds. The main interactions involved are hydrogen bond of the amino group of residue Tyr128, π-stacking interactions with Phe106 and Phe178, and electrostatic interactions with flavin adenine dinucleotide (FADH) cofactor. Three models were prepared by 3D-QSAR analysis. The models derived from Model I and Model III, shown leave-one-out cross-validation correlation coefficients (q2LOO ) of .75 and .73 as well as conventional correlation coefficients (R2 ) of .93 and .95, respectively. In addition, the external predictive abilities of these models were evaluated using a test set, producing the predicted correlation coefficients (r2pred ) of .76 and .74, respectively. The good concordance between the docking results and 3D-QSAR contour maps provides helpful information about a rational modification of new molecules based in quinone scaffold, in order to design more potent NQO1 inhibitors, which would exhibit highly potent antitumor activity.

En el presente trabajo se describe el desarrollo, en nuestro laboratorio, de una herramienta didáctic para la enseñanza de la Química Orgánica 3D y su implementación en el aula de las primeras asignaturas del curso de grado de Química Orgánica. El mismo se basó en la creación, utilizando una serie de herramientas informáticas, de simulaciones en formato electrónico de ciertas temáticas del contenido curricular de dichas asignaturas. Los tópicos desarrollados incluyeron análisis conformacional, mecanismos de reacción, estereoquímica y grupos funcionales. Estas simulaciones se aplicaron de forma exitosa y con una valoración positiva a clases presenciales y se implementaron en un soporte electrónico para clases no-presenciales (red y disco compacto)

Whey proteins are known for their high nutritional value and bioactivity, notably great potential is found on α-lactalbumin. The objective of this study was to evaluate antioxidant, antiglycant and ACE (angiotensin converting enzyme)-inhibitory activity of α-lactalbumin peptides obtained from enzymatic hydrolysis with Alcalase through different hydrolysis conditions. The optimization of enzymatic hydrolysis was studied by response surface methodology variating enzyme:substrate ratio (0.0050%, 0.0525% and 0.1000% w/w) and time (0, 30 and 60 minutes) measuring antioxidant activity by ABTS and ORAC-FL (response variables), founding an augment of this activity with time. Characterization of seven samples showed increasing hydrolysis with time. Hydrolysate obtained with 0.1000% w/w and 60 minutes demonstrated higher antioxidant activity related to greater hydrolysis. This hydrolysate did not show antiglycant activity but promoted advanced glycation end products formation with methylglyoxal and glucose. In contrast, this sample showed 30% of ACE inhibition when compared to Captopril, having great potential by enhancing hydrolysis.

Twelve yeast Debaryomyces hansenii strains were characterized for their ability to inhibit in vitro growth of Penicillium italicum, production of lytic enzymes (b-1.3-glucanase, chitinase and protease) and postharvest biological control of blue mold decay (caused by P. italicum) on Mexican lemon fruits. The D. hansenii strains inhibited in vitro the growth of P. italicum in different pHs (4.2, 4.6 y 5.0), effect related to killer toxins. The enzyme activity (of b-1.3- glucanase, chitinase and protease) was detected on Yeast Nitrogen Base (YNB) medium inoculated with cell wall fragments of P. italicum, the range enzymatic activity of all strains was between 9 and 13 days after inoculated culture medium. All D. hansenii strains significantly reduced blue mold on Mexican lemon during fifteen days of storage at 13 8C, but the LL1 strain was distinctly more effective under these conditions. These results showed that D. hansenii is an alternative for biological control of blue mold decay of Mexican lemon fruits.

Microencapsulation of the Schinus molle Rev L. (Anacardiaceae) leaves essential oil (EO) has been employed to control the release of active ingredients, protecting them from the external environment, with the concurrent improvement of its insecticidal potential on Haematobia irritans. Four microcapsule formulations (EEO1 to EEO4) of S. molle EO were prepared by spray-drying, using a mini spray dryer and gum Arabic/maltodextrin (AG/MDX) as the carrier in different proportions, at a ratio of 4:1 (MDX/AG:EO). Encapsulation efficiency (EE: 96–100%), powder morphology and particle size distribution were analyzed as responses. An interesting correlation was found between EO free and microcapsules (EEO) in the preliminary and comparative studies of stability (at 45 °C) and in the insecticide activity on H. irritans. In fact, a very slow liberation profile of the microencapsulated EO (EEO4) was observed over a period of 366 h (71% of EO retention), as well as a slower time-dependent insecticide effect (32 and 73% of dead flies at 2 and 4 h of exposure time) compared to the free EO (96% of dead flies at 2 h).

In the present study, we explored the reactivity of 5- hydroxymethylfurfural (5-HMF), a renewable platform chemical, as the aldehyde component in the Groebke-Blackburn-Bienaymé (GBB) multicomponent reaction. Given the limited stability of 5-HMF under conventional conditions used for this reaction, acidic milieu in combination with high temperature or long reaction times, we explored factors which influence the course of the reaction (solvent, catalyst, time, and temperature) within the guidelines of previously reported green reaction conditions. Mild conditions and short reaction times were achieved to obtain good conversion and selectivity values. The reaction scope was exemplified by preparing a small compound library based on the optimized conditions varying the amidine and the isocyanide component. The hydroxymethylfuran moiety present in the GBB products, imidazo[1,2-a]heterocycles, provides useful functionalization for further transformations, which could expand the diversity of the chemical space.

<p><em>Lithraea molleoides</em> (Anacardiaceae) is a tree with a broad area of distribution in the southern region of South America, including Southern Brazil, Southern, and Eastern Bolivia, Southern Paraguay, Northern, and Central Argentina, and<br />reaches its southernmost distribution limit in Northern and Northeastern Uruguay. It presents considerable morphological variability. It is used as medicinal plant and as a sweetener in traditional infusions. There have been reports on its antibacterial, antiviral, antioxidant, anti-inflammatory and antinociceptive activities. It is traditionally known that it may induce allergic contact dermatitis in sensitive persons by<br />direct and/or airborne contact, i.e. when used as firewood. Many of its biological activities have been associated with the reported presence of 5-alkyl/alkenyl resorcinols. Its allergenic activity has been ascribed to 3-alkyl/alkenyl catechols, as it<br />is well known for other species of the Anacardiaceae family. The wide distribution of the species, its morphological plasticity, the traditional medicinal uses, and toxic properties indicate the necessity of a more extensive and systematic study of the<br />species to assess its medicinal and biological potential.</p>

El objetivo de este estudio fue desarrollar, caracterizar y comparar nano y microemulsiones estables incorporando quercetina
como compuesto bioactivo. Se estudiaron emulsiones aceite:agua con beta-lactoglobulina, utilizando dos procesos
de elaboración: homogeneizador de alta velocidad (Ultra-turrax) y homogeneizador de alta presión (HPH, Emulsiflex-C5,
500-1200bar). Se evaluó la variación en la concentración de emulsionante, fracción volumétrica de aceite e incorporación
de hidrocoloides. La formación y la estabilidad de las emulsiones se analizaron por su perfil de tamaño de gota (Malvern-
Zetasizer Nano ZS) y constantes cinéticas de desestabilización obtenidas a partir del estudio de la retro dispersión de la
luz en el tiempo (Turbiscan-Classic). Se determinó que utilizando el Ultra-turrax se logra formar nanoemulsiones con
mayor estabilidad a una concentración de 2.2 % (m/v) de proteína y 0.2 % (m/v) de goma guar, mientras que utilizando
HPH con una concentración de 1.5 % (m/v) de beta-lactoglobulina y sin necesidad de hidrocoloides se logra una emulsión
de adecuado tamaño de gota (d<500nm) y mayor estabilidad (α<0.05). Las nanoemulsiones elaboradas con HPH presentaron
mayor porcentaje de encapsulación de quercetina

Glutathione transferases (GSTs) perform several catalytic and non-catalytic roles in the defense against toxicities of electrophile compounds and oxidative stress, and therefore are involved in stress-response and cell detoxification. Previously, we have provided evidence indicating that EgGST2 and EgGST3, two phylogenetically distant Echinococcus granulosus GSTs, can naturally form a heterodimeric structure (EgGST2-3). In the present work, the recombinant heterodimer GST (rEgGST2-3) is characterized. Hence, rEgGST2-3 was able to conjugate GSH to three substrates: 1-chloro-2,4-dinitrobenzene (CDNB, general substrate for GSTs), 1,2-dichloro-4-nitrobenzene (specific substrate for mammalian Mu class) and trans,trans-deca-2,4-dienal (reactive carbonyl). The canonical activity was considerably reduced by all the conventional inhibitors (cybacron blue, triphenylthin chloride and bromosulfophthalein) and by other inhibitors (ellagic acid, alizarin and chenodeoxycholic acid). Besides this, rEgGST2-3 activity was inhibited by a number of anthelmintic drugs, where the halogenated phenolic drugs (mainly bithionol and hexachlorophene) acted as stronger inhibitors, suggesting they may bind to the EgGST2-3. Moreover, rEgGST2-3 exhibited glutathione-peroxidase activity, and its specific constant (kcat/KM) was calculated. Finally, rEgGST2-3 displayed the ability to bind non-substrate molecules, particularly anthelmintic drugs, suggesting that ligandin activity may have potential to act as a passive protection parasite mechanism. Overall, the rEgGST2-3 behavior was shown to be both complementary and redundant to that reported for rEgGST1, another characterized GST from E. granulosus. It would be appropriate that different enzymes in the same organism do not have exactly the same functional properties to develop a better adaptation to life in the host.

Echinococcus granulosus is a cestode parasite whose cytoskeleton plasticity allows it to enter and develop inside its hosts, completing thus its life cycle. We focused our attention on F-actin organization and distribution in E. granulosus protoscoleces (PSC) in order to contribute to the knowledge of the parasite cytoskeleton. In particular, we addressed some aspects of F-actin rearrangements in PSC at different stages of the evagination/invagination process. The use of light microscopy allowed us to identify different PSC structures and phalloidin staining displayed a parasite's highly organized F-actin cytoskeleton. Suckers exhibit an important musculature composed of a set of radial fibers. At the rostellum, the F-actin filaments are arranged in a bulbar shape with perforations that appear to be the attachment places for the hooks. Also, "circular" structures of F-actin were identified, which remind the flame cells. Furthermore, parasite F-actin filaments, unevenly distributed, seem to have remained substantially unchanged during the evagination/invagination process. Finally, we showed that the scolex of an evaginated E. granulosus PSC reinvaginates in vitro without any treatment.

The total synthesis of (+)-bromoxone and a formal synthesis of (�)-bromoxone were performed in a simple chemoenzymatic manner, starting from bromobenzene, and using an enantiodivergent strategy. The usefulness of chiral cyclohexadienediols derived from the biotransformations of monosubstituted-aromatic compounds as building blocks for the preparation of natural epoxyenones was confirmed.

Abstract
A summary of the resource.
La preparación de derivados de glucosa marcados con 99mTc reviste gran interés para la evaluación del consumo de glucosa in vivo en oncología y cardiología nuclear. Este trabajo presenta la marcación de un análogo de glucosa (GLU-DTC) mediante la formación de un complejo Tc(V)-nitruro simétrico. Para ello se incorporó a la biomolécula un grupo ditiocarbamato capaz de coordinar al metal. La marcación fue realizada mediante sustitución de ligandos, obteniéndose una única especie con pureza radioquímica superior al 90%, la que se mantuvo durante al menos 4 hs. La caracterización fisicoquímica y biológica muestra que el complejo 99mTc(V)-nitruro(GLU-DTC)2 es un compuesto estable y altamente hidrofílico, aunque su unión a proteínas plasmáticas es mayor a la esperada, hecho que justificaría la alta actividad retenida en sangre y en hígado durante la evaluación biológica en ratones CD1 normales. Estos resultados indican que la marcación con 99mTc de este derivado de glucosa produce una alteración significativa de su comportamiento biológico.

Inoculation of rabbits with a nonliving anti-Pseudomonas vaccine induced appreciable levels of agglutinating antibodies against strains of P. aeruginosa included or not included in the vaccine. Serum obtained from vaccinated rabbits was able to confer temporary protection to mice against challenge with homologous or heterologous strains of Pseudomonas. When two or three doses of serum were used, all mice survived the challenge dose for more than 48 hr, but some of the animals died 10 days after challenge. When five doses of serum were used, all mice survived this 10-day period, and even 4 months later they did not show any sign of infection. Serum treatment temporarily inhibited Pseudomonas activity and allowed for the activation of the immunogenic mechanisms of the animals. This was corroborated by the fact that mice treated with three doses of serum and surviving the challenge dose for more than 20 days were immune against a second challenge. Anti-Pseudomonas gamma globulin conferred a lower degree of protection against homologous or heterologous strains of Pseudomonas.

<p>El sistema de producción de frutilla en el litoral norte de Uruguay está basado en micro y macrotúneles, con el objetivo de producir fruta durante otoño, invierno y primavera. El objetivo del programa de mejoramiento de INIA es contar con variedades adaptadas a las condiciones ambientales, resistentes a enfermedades y plagas, de alta calidad, sabor y vida postcosecha. El objetivo del trabajo consistió en la evaluación de calidad de fruta de seis cultivares de frutilla (INIA Yvahé, INIA Guenoa, Yurí, Earlibrite, SGK50.4 y SGJ37.2) durante la estación productiva y la evaluación sensorial de los tres cultivares INIA disponibles en el mercado. Se trabajó con tres repeticiones por cultivar, evaluando calidad externa e interna en cosecha para cada fecha considerada (junio, setiembre y octubre). Se seleccionaron tres cultivares (INIA Yvahé, INIA Guenoa, Yurí) para su evaluación sensorial con consumidores en la cosecha de octubre. Yurí destacó por su calidad de fruta, siendo el cultivar más firme en todas las fechas de cosecha (20-55%). Respecto a la evaluación sensorial, los tres cultivares alcanzaron altos puntajes de aceptabilidad global. De acuerdo a la percepción de los consumidores, existieron claras diferencias en las características sensoriales de los cultivares, principalmente en su dulzor, acidez, tamaño, firmeza y forma. Yurí fue percibido por los consumidores como más dulce y firme que INIA Guenoa e INIA Yvahé. Es posible concluir que existen diferencias significativas en la calidad de fruta obtenida en función de la época de cosecha y que los cultivares evaluados tienen amplia aceptación por los consumidores uruguayos.</p>

The extensive use of imazalil (IMZ) in Uruguayan citrus packinghouses to control Penicillium spp. favored the selection and proliferation of resistant isolates. With the aim of detecting Penicillium digitatum biotypes that are not controlled by commercial doses of IMZ, the IMZ concentration within amended potato dextrose agar (PDA) plates was adjusted to 1.0mgL−1 IMZ. This concentration allowed the detection of resistant isolates that were not controlled by commercial applications of 3.0 g L−1 IMZ. These isolates were able to grow in fruit with IMZ residues of 0.92 and 3.08mgkg−1. Therefore, environmental monitoring of facilities where commercial dip applications containing 3.0 g L−1 IMZ are employed, should be done with 1.0mgL−1 of IMZ in Petri dishes. We can conclude that use of IMZ in postharvest applications of 3.0 g L−1 remains effective to control wild Uruguayan P. digitatum isolates but not for resistant ones. A survey of 26 P. digitatum isolates collected in Uruguay indicated IMZ resistant isolates occurred in packinghouses and not in citrus groves.

BACKGROUND: Traditionally, breeding research has been focused on increasing yield and fruit size, improving resistance to diseases, behaviour during transport and increasing postharvest shelf-life. However, consumers’ liking is one of the biggest challenges for strawberry breeding programs. In this context, the aim of the present work was to evaluate the application of a check-all-that-apply (CATA) question to study consumers’ perception of new strawberry cultivars. RESULTS: Average liking scores were low for a highly appreciated product like strawberries, which could be explained considering unfavourable weather conditions prior to harvest. However, despite the small differences in overall liking scores, significant differences were found in the frequency in which 14 out of the 22 terms of the CATA question were used to describe samples. This suggests that the evaluated CATA questionwas able to detect differences in consumers’ perception of the sensory characteristics of the evaluated cultivars.Moreover, a significant correlation was found between laboratory analyses (firmness, colour, soluble solids and acidity) and consumers’ responses to the CATA question, which indicates their validity. CONCLUSIONS: Considering results from the present study, the use of CATA questions could be a simple and interesting methodology to gain insight into consumers’ perception of different fruit cultivars.

BACKGROUND: Increasing demand for sweet potato in regions with temperate climates has triggered interest in the development of new cultivars. Breeding of this crop should consider sensory characteristics in order to meet consumers’ expectations. This requires the application of simple and cost-effective methodologies that allow quality evaluation from a sensory perspective. RESULTS:With the objective of identifying the key sensory characteristics of different sweet potato genotypes, two commercial cultivars and seven clones were evaluated during three consecutive years using projectivemapping by an untrained consumer panel. This methodology allowed the discrimination of the genotypes, identifying similarities and differences among groups based on sensory terms selected by the assessors. Genotypes were differentiated in terms of texture and flavor characteristics (firmness, moisture, smoothness, creaminess, flavor intensity, sweetness and bitterness). Materials for future crossings were identified. CONCLUSIONS: The evaluation of the sensory characteristics of sweet potato clones and cultivars using projectivemapping is a quick, cost-effectiveandreliable tool for theselection ofnewadvancedsweet potato clones with superior sensory characteristics compared to the reference cultivars INIA Arapey and Cuarí.

<p>Earthworms have proved to contribute to plant health indirectly, by improvement of soil physical and chemical properties, as well as directly, through suppression of plant pathogens. Fusarium Head Blight, caused primarily by Fusarium graminearum, is one of the most important cereal diseases, with severe detrimental effects on yield and grain quality worldwide, and significant effect on grain safety due to the accumulation of mycotoxins produced by the fungus. Earthworms could reduce the residue-borne inoculum density of F. graminearum through different mechanisms, including direct competition by residue consumption, growth inhibition caused by the earthworm coelomic fluid, mechanical disruption of fungal hyphae, and burying crop residue which reduce the<br />chances for the fungus to release spores for head infection. Thus, the aim of this study was to assess the effects of anecic and epigeic earthworms on wheat straw and on F. graminearum inoculum. For this, PVC cylinders (microcosms 14.5 cm diameter, 30 cm height) were filled with moist soil mix, wheat straw was evenly distributed on the soil surface (inoculated with Fusarium, soil microorganisms or sterile) and three earthworm species (Aporrectodea longa, Lumbricus rubellus or Lumbricus terrestris) were tested. Since the response of earthworms may<br />change according to conditions of soil moisture and food availability, two different experiments were arranged. The first Experiment represented a sub-optimal situation for earthworms regarding soil moisture and feeding conditions, which was obtained by scarce watering of the microcosms with a consequent decrease in soil water content during incubation, and by not adding cow manure as supplementary food source. In the second experiment, soil was enriched with cow manure as feed for the earthworms and soil moisture was maintained<br />above 25%. Results from qPCR analysis revealed that F. graminearum inoculum on straw was reduced to undetectable<br />amounts by L. rubellus and A. longa when the feeding conditions were limited, while no significant differences compared with the control without earthworms were found when earthworms had high food availability (α = 0.05). Straw coverage on soil surface was reduced by L. rubellus (epigeic) in both experiments (p< 0.0001), while A. longa (anecic) just did so under optimal conditions. L. terrestris (anecic), only tested under optimal conditions, reduced soil cover significantly more than the other two species (p < 0.0001). Negative effect<br />of this fungus was not observed on the studied earthworms. Both anecic and epigeic earthworms tested showed potential to contribute to biological control of F. graminearum in wheat straw. This control may occur by reducing straw on surface, reducing the pathogen inoculum on straw, or both, depending on the environmental conditions and their response according to their ecological group and species. The possibilities to optimize earthworm capacity for biological control of fungal diseases in practical agriculture, under changing weather and organic matter supply conditions, are discussed.</p>
