Se estudia una técnica de destrucción que asegura la mineralización completa de la materia orgánica, pudiendo servir en forma general para la investigación de los tóxicos denominados minerales o metálicos en química toxicológica. La técnica se desarrolla en 3 etapas y coordina: I) La acción solubilizante por degradación ácida y oxidación del ácido nítrico diluido. II) La concentración de la solución, que completa a la acción de dicho ácido y facilita luego el ataque sulfonítrico. III) Ataque sulfonítrico sobre el residuo anterior. Se usa  en un pequeño y simple aparato de destilación que permita separar tres facciones: a) el mercurio por volatilización durante el ataque sulfonítrico; b) el arsénico por destilación con HCI; c) en el residuo del ácido sulfúrico quedan los metales tóxicos de importancia secundaria: plomo, bario, cobre, zinc, etc. La técnica se conduce asegurando el máximo de rendimiento de reactivos lo que reduce el costo y evita posibilidades de contaminación. Como las pérdidas son mínimas, operando sólo con 100 gramos de masa visceral, es posible asegurar la identificación de los tóxicos metálicos dentro de límites aceptables, en los casos frecuentes que la falta de antecedentes no permitan orientar la investigación y exige el empleo de métedos generales. Trabajando con 100 gramos de mezcla orgánica (carne. hígado y riñón) es posible identificar por reacciones corrientes hasta: mg: 0,1 de arsénico (mineral y orgánico); mg:01 de mercurio (mineral y orgánico); 2mg. de plomo, bario y cobre; 4mg. de zinc, cadmio y bismuto. La Enfermedad de Chagas es una enfermedad no resuelta, causada por el parasite Trypanosoma cruzi. Existe desde hace miles de anos y afecta a millones de personas, mayoritariamente en America Latina. Con el objetivo de buscar terapias apropiadas para est a problematica, en este trabajo se disenaron, sintetizaron y caracterizaron ochenta y cinco nuevos compuestos. Se determinaron sus capacidades tripanosomicidas in vitro obteniendose mas de veinte derivados con destacada actividad anti-Trypanosoma cruzi. Se estudi6 la toxicidad de los mismos frente a celulas de mamffero determinandose la selectividad de los derivados biol6gicamente activos. Se realizaron estudios de mutagenicidad in vitro. Se realizaron estudios in vivo con tres de los derivados mas relevantes. Se identific6 un derivado con potencialidad a farmaco que posee varias ventajas. Desde el punta de vista qufmico: sencillez sintetica, producto escalable y de producci6n econ6mica. Desde el punta de vista biol6gico: activo en diferentes estadfos y cepas del parasito, baja toxicidad inespedfica, ausencia de mutagenicidad, actividad in vivo similar a la del farmaco de referencia y ausencia de genotoxicidad in vivo. Los nuevos compuestos han sido desarrollados empleando procedimientos sinteticos sencillos y eficientes y la caracterizaci6n estructural de los mismos se ha realizado mediante tecnicas espectrosc6picas habituales. Estos compuestos hibridos se han evaluado en diferentes modelos bio16gicos y a traves de ellos ha sido posible identificar nuevos lfderes con elevados indices de selectividad frente a Mycobacterium tuberculosis, Trypanosoma cruzi y Leishmania amazonensis. Los estudios fannaco16gicos han permitido identificar nuevos compuestos con actividades analgesicas y anti-inflamatorias que actuan principalmente a traves de su capacidad de inhibir la via NF-KB y la liberaci6n de Interleuquina-8. Los compuestos que en este trabajo se identificaron con mejor perfil bio16gico, presentan en general, baja citotoxicidad y no son mutagenicos. Esto los ubica como nuevos prototipos para la realizaci6n de futuras optimizaciones moleculares. En el presente trabajo de tesis doctoral se realizó el diseño, síntesis, evaluación biológica frente al parásito, estudios de inhibición enzimática y estudios de relación estructura-actividad inhibitoria de nuevos compuestos híbridos derivados del sistema benzofuroxano y 1,3-dióxido de benzimidazol conteniendo agrupamientos semicarbazona, tiosemicarbazona y amidinohidrazona (Serie I) o agrupamientos arilguanidina (Serie II) como potenciales inhibidores de cruzipaína. Los nuevos compuestos desarrollados se prepararon empleando procedimientos sintéticos eficientes. La caracterización estructural se realizó por técnicas espectroscópicas habituales. Los nuevos compuestos se evaluaron sobre la forma epimastigota de T. cruzi y se estudió su toxicidad frente a glóbulos rojos humanos y macrófagos murinos. Los derivados de la Serie II mostraron excelentes actividades frente al parásito, identificándose derivados, pertenecientes a ambas series, con índices de selectividad mayores que 50, siendo este valor para las referencias, anfotericina B y nifurtimox, de 10 y mayor que 13, respectivamente. Los nuevos compuestos presentaron una actividad variable frente a cruzipaína, no encontrándose relación entre ésta y la actividad anti-T. cruzi. En general, los derivados de la Serie I fueron mejores inhibidores de cruzipaina que los de la Serie II. A fin de racionalizar la actividad frente a cruzipaína se realizaron estudios de interacción inhibidorenzima empleando técnicas de docking y de resonancia magnética nuclear, experimentos de variación del tiempo de relajación longitudinal, experimentos DOSY y experimentos de diferencia en la transferencia de saturación, que permitieron analizar en forma detallada las interacciones de los distintos ligandos con residuos clave localizados en el sitio activo de la enzima. Geometry optimization of all charge-transfer complexes and transition states has been done. Standard analysis of activation energies of addition reactions, vibrational and thermodynamical analysis, as well as Morokuma energy decomposition, BSSE correction, PMO analysis, and Pauling bond orders were used to explain the results. A subset of the reactions, including that of C2H4 as reference one and the two most favorable cases, was also studied at the MP2/6–31G(d,p)//HF/6–31G(d,p) level. The barriers so obtained are in agreement with the indirectly found from experimental data. It was found that the effect of the substituent is not monotonic for the additions. Decomposition of the interaction energy is shown to be adequate to explain this nonmonotonic behavior. The implications for laser chemistry of the addition of hydrogen halides to fluorosubstituted olefins is briefly discussed. Sensory and consumer scientists disagree on the practice of concurrently obtaining sensory information in hedonic tests. This is in part due to different mindsets about what consumers are able to do and evidence that such co-elicitation may bias hedonic scores. Check-all-that-apply (CATA) questions have been claimed to have a smaller effect on hedonic scores than other attribute such as just-about-right or intensity scales. In this research, nine studies using consumers as participants examined effects on hedonic product scores when sensory attribute information was co-elicited using CATA questions. The use of CATA concurrently with hedonic was benchmarked against concurrent attribute liking scores, attribute intensity scores and just-about-right scaling. Across a range of product categories (beer, fresh fruit, tea, flavoured water, crackers, savoury dips), only weak and transient evidence of bias of hedonic scores when concurrently using CATA questions was established. This effect was independent on whether samples, on average were moderately liked or moderately disliked, and replicated when samples were assessed partially by the sense of smell only or via full product assessment (appearance, aroma, flavour, taste, aftertaste, mouthfeel). The present research suggests that co-elicitation of hedonic scores and product attribute information using CATA questions may bias the hedonic scores, but not that it certainly will do so. This needs to be recognised, leading to more widespread acceptance that co-elicitation has merit. Investigators should decide on whether or not to co-elicit product attribute information using CATA questions on a case-by-case basis, acknowledging that bias may occur. The hydrogen storage capacities of the recently synthesized tetraindenocoranulene C44H18 and pentaindenocorannulene C50H20 have been investigated and compared with those of corannulene and C60. The results for the physisorption of a single H2 on the concave side indicate that the adsorption energies (Eads) for tetraindenocorannulene and pentaindenocorannulene are very similar, 2.9 and 3.0 kcal/mol, respectively. The later values are slightly larger than that estimated for corannulene, 2.4 kcal/mol, and about half of the encapsulation energy of H2 in C60, 5.4 kcal/mol. The differences in adsorption energies can be explained considering the dipole moments of the molecules. For C50H20 and C44H18 they are very similar, 4.0 and 3.7 Debye. However, it is much smaller in corannulene, 2.0 Debye, which explains the lower Eads observed for the later polyarene. The Eads presents a different behavior on the electron-poor convex side; they are nearly identical for all of the molecules considered. The interaction energies on the convex side, determined for corannulene, tetraindenocorannulene, pentaindenocorannulene, and C60, are 1.2, 1.1, 1.1, and 1.2 kcal/mol, respectively. The hydrogen storage capabilities are discussed in terms of the results obtained for the sandwich complexes between pentaindenocorannulene with benzene or corannulene. As a by product, we estimated the complexation energies for the benzene-pentaindenocorannulene and corannulene-pentaindenocorannulene pairs as 6.0 and 8.3 kcal/mol, respectively. Finally, we replaced some carbon atoms in corannulene and pentaindenocorannulene with nitrogen atoms. The adsorption energies were increased; C16N4H10 has nearly the same adsorption energy as the much larger pentaindenocorannulene. In the case of pentaindenocorannulene, the introduction of six nitrogen atoms (12% of carbon atoms replaced) increases the adsorption energy, about 0.2 kcal/mol. The present results showed that the introduction of a small number of nitrogen atoms can increase the adsorption energy in carbon structures without increasing the weight of the material too much, but the increment seems to be strongly dependent on the position of the nitrogen atoms. As the use of check-all-that-apply (CATA) questions for consumers to aid sensory product characterization gains popularity the need for developing guidelines regarding best practice increases. An important issue in the design of studies including CATA questions is the number of consumers that are necessary to obtain stable results. In this context, the aim of the present work was to study the stability of sample and descriptor configurations obtained using CATA questions with consumers, by means of a bootstrapping resampling approach. Data from 13 consumer studies were analyzed, differing in number of consumers, product category, number of samples, degree of difference among samples and number and type of descriptors in the CATA question. For each study, 1000 random subsets of different number of consumers were generated from the original data set. For each consumer virtual panel, sample and descriptor configurations were obtained using Correspondence Analysis and the concordance between them and the reference configurations (obtained with all the consumers) were computed using the regression vector (RV) coefficient. The RV coefficients of sample and descriptor configurations increased with the number of consumers, suggesting an increase in the stability of the configurations. Results showed that the number of consumers necessary to yield fully stable configurations depended on the degree of difference among samples, the complexity of the sensory space and the type of descriptors included in the CATA question. Across the 13 data sets analysed, results suggested that when working with widely different samples, 60–80 consumers can be regarded as a reasonable compromise to get stable sample and descriptor configurations. However, it is recommended to check, a posteriori, the stability of sample and descriptor configurations using a stability index based on bootstrapping. Abstract Qualitative and quantitative determination of volatile markers in aqueous based fragrances assumes ever-increasing importance, because of both the need for quality control and the safety-regulatory limitations introduced for several compounds. This study reports and critically discusses the results of applying new water-compatible ionic-liquid (IL) GC stationary phases, based on phosphonium and imidazolium derivative cations combined with trifluoromethanesulphonate (Watercol™) to the direct quantitative analysis of aqueous samples in the perfume field with GC–MS. Narrow-bore columns of different lengths, especially prepared for this study, were adopted to minimize the amount of water reaching the MS detector after GC separation. All GC–MS analysis steps were investigated, to achieve results compatible with quality control requirements for the volatiles of interest in this field, in terms of LODs, LOQs, and repeatability. Reliability of the GC–MS results was demonstrated by determining volatile allergens in two commercial perfumes, as per EU regulations concerning no-declaration limits for leave-on (0.001%) and rinse-off (0.01%) cosmetic products. The catalytic activity of iron pillared montmorillonitic clays in the discoloration of methylene blue aqueous solutions by the heterogeneous photo- Fenton process was studied. Catalysts were prepared using two different-sized aggregate fractions of the clay, ranging below 250 lm and within the range of 250–450 lm. Two calcination temperatures, 400 and 600 �C, were used. The solids were characterized by thermo-gravimetric analysis, nitrogen adsorption isotherms, elemental analysis, X-ray diffraction and iron content. A higher specific surface area and specific pore volume were found for the solid obtained from the finer aggregates of the clay and the lowest calcination temperature. Under photo-Fenton like conditions, this catalyst also displayed the highest catalytic activity which remained practically the same at pHs 3.0 and 5.6 and for H2O2 concentrations in the range from 5 to 20 mM. The observed differences in catalytic performance can be explained on the basis of the catalysts’ iron content and textural properties. Leaching of iron ions from the catalysts could not be detected. El término “arenas negras” hace referencia a los minerales pesados que pueden ser encontrados en depósitos denominados Placer; estas arenas pueden ser generadas y concentradas por la acción de agentes naturales como corrientes de agua, y acción de las olas y el viento a lo largo de la playa, así como también por procesos extractivos. Sin importar si son de origen natural o de minería las arenas negras son producto de descomposición y ruptura de rocas, seguido por concentración por gravedad. En estos procesos en la mayoría de los casos la composición de la arena hace que tome color negro o marrón oscuro, de ahí la denominación. Debido a que se forman por desgaste de rocas los minerales componentes de las arenas negras son en general resistentes química y mecánicamente. Es entonces un yacimiento formado por la concentración de minerales en la arena de ríos, arroyos o playas como consecuencia de su elevada densidad (mayor que el cuarzo), minerales que deben ser al mismo tiempo resistentes a la abrasión. Los placeres se clasifican en depósitos residuales y depósitos transportados. En los primeros los minerales han sido más o menos liberados por desgaste y el depósito posee más o menos la misma composición que la roca original, de esta manera los minerales no han sido concentrados, mientras que en los segundos los minerales generados por desgaste se han desplazado de la roca original y se han re-depositado (generalmente por acción del agua corriente). En estos proceso de trasporte y re-deposición los minerales inestables se solubilizan, los minerales más livianos viajan más que los más pesados generándose depósitos concentrados. Estos últimos son los más estudiados, debido a su importancia comercial. Los minerales que se pueden encontrar en un depósito de arena negra dependen del tipo de roca de la cual el depósito proviene; las arenas negras por lo tanto en general son una mezcla de varios minerales pero pueden consistir enteramente de uno o dos minerales. Monoterpenes are frequently the most common and distinctive components of essential oils. Most of them (the so-called regular monoterpenes) obey the Ruzicka’s Isoprene Biogenetic Rule, which means that the assembly of the bioactive isoprene building blocks (IPP and DMAPP) takes place in a head-to tail fashion. However, in some cases, the occurrence of completely different biogenetic assemblies gives rise to irregular monoterpenes that do not go from head-to-tail. The most common representatives of this type being artemisyl, chrysanthemyl, lavandulyl, and santolinyl skeletons. In addition, rearrangements from regular compounds are also frequently observed giving rise to a new category of irregular monoterpenes, the rearranged ones. Finally, other closely related group is made up by the degraded monoterpenes (or nor-monoterpenes or homoterpenes),compounds that lose carbon atoms during their biosynthesis and that can be considered also as “irregulars”. This chapter discusses the role of these three groups of compounds as all of them have eventually been identified in essential oils. Emphasis will be placed on rearranged monoterpenes, especially ortho-menthane derivatives, due to the paucity of literature dealing with this topic. Carquejol and piquerols, and some of their derivatives are reviewed according to their occurrence, possible biosynthesis, and biological properties. Semi-synthesis will also be discussed, as potentially bioactive compounds could be obtained. Finally, some practical aspects for the preparation of carquejol oxide (7,8-epoxy carquejol), a novel derivative of the ortho-menthane family, will be presented.
