Polychlorinated dibenzo-p-dioxins (PCDD) and dibenzofurans (PCDF) are persistent, toxic, and bioaccumulate in the environment. Due to their high analytical costs, these compounds are hardly regulated and mostly not monitored in the Third World. To overcome this, bioassays have been proposed as low-cost alternative methods. Two of the most established bioanalytical tools, the dioxin antibody-based enzyme-linked immunosorbent assay ELISA and the micro-EROD bioassay are evaluated and compared to high resolution gas chromatography and high resolution mass spectrometry (HRGC/HRMS) analytical methodology. The methods were tested using thirteen soils and sediment samples selected from diverse sites in Montevideo, Uruguay. The WHO₂₀₀₅ total toxic equivalent (WHO₂₀₀₅-TEQ) of soils ranged from 2.4 to 2212 (ng WHO₂₀₀₅-TEQ/kg dry sample) and from 0.14 to 9.4 (ng WHO₂₀₀₅-TEQ/kg dry sample) in sediments. This study shows significant contamination related to dioxin-like compounds, particularly in sites where uncontrolled burnings were carried out. ELISA and micro-EROD bioassay correlated well with HRGC/HRMS, R Spearman 0.773 and 0.913, respectively and were highly correlated to each other, R Spearman 0.879. Preliminary threshold values of bioassay toxic equivalents of 330 (ng/kg dry sample) for the micro-EROD bioassay and 220 (ng/kg dry sample) for ELISA are proposed. Las feromonas son compuestos qUlmlcos emitidos por un organismo y que ejercen una respuesta de comportamiento 0 fisiol6gica sobre otro organismo de la misma especie. Dentro de este grupo, las feromonas sexuales son aquellas que permiten el encuentro de los sexos para favorecer la c6pula. Los insectos, especialmente los del orden Lepidoptera (mariposas y polillas), son el grupo de seres vivos en los cuales estos mediadores qUlmicos han side mas estudiados. Los lepid6pteros son un orden muy numeroso y presentan gran cantidad de especies consideradas plagas agricolas. EI usa de feromonas sexuales tiene un rol muy importante dentro del manejo integrado de plagas y representa una alternativa de control amigable con el ser humane y el medio ambiente. Se utilizan tanto en metodologias de monitoreo como de control. En esta tesis se presenta la ruta de sintesis de cuatro componentes mayoritarios de las feromonas sexuales de Bonagota salubricola, Cryptoblabes gnidiella y Crocidosema aporema, plagas regionales de manzanos, vid y soja respectivamente, con la finalidad de utilizarlos en el monitoreo de las mismas en cultivos de nuestro pais. Los compuestos objetivo de este trabajo presentan una funci6n oxigenada terminal (alcohol primario, acetato 0 aldehido), largo de cadena entre 12 y 16 carbon os y una 0 dos insaturaciones. Los pasos c1aves de las rutas son la introducci6n de los grupos funcionales term in ales y la obtenci6n de los dobles enlaces con la estereoquimlca definida, ya que la relaci6n ZIE es muy importante en la actividad biol6gica de estos compuestos. Food products targeted at children are usually marketed using persuasive elements aimed at creating positive hedonic and emotional associations. For this reason, changes in children's emotional associations with unhealthy food products can discourage their consumption. In this context, the aim of the present study was to compare children's emotional associations with food products featuring different front-of-pack (FOP) nutrition labelling schemes. A total of 492 children (6&ndash;12 years old) were randomly divided into three groups, each of which evaluated a series of packages featuring different FOP nutrition labelling schemes: guidelines daily amount (GDA), traffic light system (TLS) and nutritional warnings. For each of the six packages and three unpackaged products, children were asked to select all the emoji from a list that described how they would feel eating the product. Data were analyzed using generalized linear models. FOP nutrition labelling significantly influenced the frequency of use five of the 16 emoji. In general, children who evaluated packages with directive and semi-directive schemes used emoji associated with positive emotions less frequently than those who evaluated packages with the GDA system. The effect of FOP nutrition labelling scheme on emotional associations was moderated by age and type of school. The effect of directive and semi-directive FOP nutritional schemes, in special nutritional warnings, tended to be higher for younger children and children from public schools. These results suggest that FOP nutrition labelling may contribute to discouraging consumption of products with high content of nutrients associated with non-communicable diseases among children. Excessive sugar intake is a global public health concern, and sugar-sweetened beverages (SSB) represent one of the main sources of added sugar in the diet. In this sense, the inclusion of front of pack (FOP) nutritional schemes and food reformulation are strategies that could contribute to reduce sugar consumption and improve population health status. The aims of this work were to evaluate adults and children&rsquo;s choice of sugar-reduced drinks in the context of the implementation of FOP nutrition labelling under different evaluation conditions, and to compare the influence of two FOP nutrition labelling schemes: the traffic-light system (TLS) and nutritional warnings. For that, 400 adults and 400 children (6&ndash;12 years old) divided in 2 groups (n = 200) each chose samples of grape nectar or chocolate flavoured milk (control and two sugar reduction levels) featuring different FOP nutritional schemes, under three experimental conditions: (a) blind, tasting the samples without any information, (b) expected, looking at the packages, and (c) informed, looking at the packages and tasting the samples. In each experimental condition, participants were asked to choose one of the three products. Both FOP schemes encouraged adults and children&rsquo;s to choose for healthier products under the expected condition. In the case of adults, nutritional warnings outperformed the traffic light system in the task involving grape nectars. However, when participants tasted the products (blind and informed conditions), choices were defined by their sensory characteristics and the control samples without sugar reduction were the most frequently selected. One hundred and seventeen 6&ndash;12 years old children participated in a two-part study: a 9-week study and a final liking test at the end of 11 weeks. In the 9-week study, children were divided in two sugar reduction strategies: stepwise and gradual. Children&rsquo;s sensory and hedonic perception was evaluated using 9-point hedonic scales and check-all-that-apply questions. There were no major differences between the gradual and stepwise reduction groups in children&rsquo;s sensory and hedonic perception. However, results showed that the gradual reduction strategy led to smaller changes in children&rsquo;s sensory and hedonic perception than the stepwise strategy. In the final test, children in the stepwise group gave significantly higher liking scores to the samples than those in the gradual sugar reduction strategy. Comparing the overall liking between the first time the children tasted the grape nectars and the final test, a significant difference was observed for the sample with lower sugar content only for the stepwise group, which was higher in the 11th week. Although differences between the sugar reduction strategies were small, the gradual reduction strategy seems to be recommended over the step-wise strategy. The larval stages of the cestodes Echinococcus multilocularis and Echinococcus granulosus cause the important zoonoses known as larval echinococcoses. These larvae are protected by a unique, massive, mucin-based structure known as the laminated layer. The mucin glycans of the E. granulosus laminated layer are core 1- or core 2-based O-glycans in which the core Galp&beta;1-3 residue can initiate a chain comprising one to three additional Galp&beta;1-3 residues, a motif not known in mammalian carbohydrates. This chain can be capped with a Galp&alpha;1-4 residue, and can be ramified with GlcNAcp&beta;1-6 residues. These, as well as the GlcNAcp&beta;1-6 residue in core 2, can be decorated with the Galp&alpha;1-4Galp&beta;1-4 disaccharide. Here we extend our analysis to the laminated layer of E. multilocularis, showing that the non-decorated cores, together with Galp&beta;1-3(Galp&alpha;1-4Galp&beta;1-4GlcNAcp&beta;1-6)GalNAc, comprise over 96% of the glycans in molar terms. This simple laminated layer glycome is exhibited by E. multilocularis grown either in vitro or in vivo. Interestingly, all the differences with the complex laminated layer glycome found in E. granulosus may be explained in terms of strongly reduced activity in E. multilocularis of a putative glycosyltransferase catalysing the elongation with Galp&beta;1-3. Comparative inter-species analysis of available genomic and transcriptomic data suggested a candidate for this enzyme, amongst more than 20 putative (non-core 1) Gal/GlcNAc &beta;1-3 transferases present in each species as a result of a taeniid-specific gene expansion. The candidate gene was experimentally verified to be transcribed at much higher levels in the larva of E. granulosus than that of E. multilocularis. There is increasing interest in the use of non-Saccharomyces yeasts in winemaking due to their positive attributes. The non-Saccharomyces yeast Hanseniaspora vineae is an apiculate yeast that has been associated with the production of wine with good fermentation capacity and an increase in aromatic properties. However, this yeast represents a concern in mixed culture fermentation because of its nutrient consumption, especially nitrogen, as its mechanisms of regulation and consumption are still unknown. In this study, we analyzed the nitrogen consumption, as well as the nitrogen catabolism repression (NCR) mechanism, in two genome-sequenced H. vineae strains, using synthetic must fermentations. The use of synthetic must with an established nitrogen content allowed us to study the NCR mechanism in H. vineae, following the amino acid and ammonia consumption, and the expression of genes known to be regulated by the NCR mechanism in S. cerevisiae, AGP1, GAP1, MEP2, and PUT2. H. vineae exhibited a similar amino acid consumption and gene expression profile to S. cerevisiae. However, the wine strain of S. cerevisiae QA23 consumed ammonia and valine more quickly and, in contrast, tyrosine and tryptophan more slowly, than the H. vineae strains. Our results showed a similar behavior of nitrogen regulation in H. vineae and S. cerevisiae, indicating the presence of the NCR mechanism in this Hanseniaspora yeast differentiated before the whole genome duplication event of the Saccharomyces complex. ABSTRACT: The quality of the grapes, and therefore of the wine, are the result of the interaction of different<br />factors involving biological, physical, climatic and cultural aspects; thus indicating the importance of the work<br />in the management of the vineyard. In this work, we demonstrate the application of chromatographic and<br />spectroscopic techniques as a tool for the enologic investigation able to predict, control and improve aspects<br />related with the quality of the wines and the management of the vineyard. Examples of extraction processes and<br />selective analysis are shown for volatile aglicones present in the grape as glycoconjugates (aromatic potential)<br />as well as anthocyanidins glycosides present in the wines obtained by microvinificaci&oacute;n of the same vineyards.<br />It is remarcable in the Vitis vinifera cv. Siderophore production confers to bacteria competitive advantages to colonize plant tissues and to exclude other microorganisms from the same ecological niche. This work shows that the community of endophytic siderophoreproducing bacteria (SPB) associated to Oryza sativa cultivated in Uruguayan soils is dynamic and diverse. These bacteria were present in grains, roots, and leaves, and their density fluctuated between log10 3.44 and log10 5.52 cfu g&minus;1 fresh weight (fw) during the plant growth. Less than 10% of the heterotrophic bacteria produced siderophores in roots and leaves of young plants, but most of the heterotrophic bacteria were siderophore-producers in mature plants. According to their amplified restriction DNA ribosomal analysis (ARDRA) pattern, 54 of the 109 endophytic SPB isolated from different plant tissues or growth stages from replicate plots, were unique. Bacteria belonging to the genera Sphingomonas, Pseudomonas, Burkholderia, and Enterobacter alternated during plant growth, but the genus Pantoea was predominant in roots at tillering and in leaves at subsequent stages. Pantoea ananatis was the SPB permanently associated to any of the plant tissues, but the genetic diversity within this species&mdash; revealed by BOX-PCR fingerprinting- showed that different strains were randomly distributed along time and plant tissue, suggesting that a common trait of the species P. ananatis determined the interaction with the rice plant. Several isolates were stronger IAA producers than Azospirillum brasilense or Herbaspirillum seropedicae. In vitro inhibition assays showed that SPB of the genus Burkholderia were good antagonists of pathogenic fungi and that only one SPB isolate of the genus Pseudomonas was able to inhibit A. brasilense and H. seropedicae. These results denoted that SPB were selected into the rice plant. P. ananatis was the permanent and dominant associated species which was unable to inhibit two of the relevant plant growth-promoting bacteria.
