A summary of the resource.
In this work we report a conformational study of [ReVO2(pn)]2+ (pn = 1,2-propylenediamine) in aqueous solution using experimental and theoretical approaches. The studies in aqueous solution were carried out by 1H and 13C NMR spectroscopy and applying the Altona’s equation. Theoretically, the studies were performed by means of the Density Functional Theory (DFT) and the Gauge-Including Atomic Orbitals (GIAO) method. All 12 spatial dispositions arising from the relative orientation of the CH3 substituents were studied. The results are consistent with the predominance of complexes with λλ ligand conformation. The computation analysis shows that only the λλ conformers exhibit a 1H-signal pattern in agreement with the experimental evidence. We proposed that only two main species can predominate in an aqueous solution of {ReVO2[(R)-pn]2}+. They should have a λλ conformation of the (R)-pn ligands and the energy difference (PCM (Polarizable Continuum Model) methodology) between them is only 1.5 kJ mol−1.

Chiral cis-1,2-dihydrocatechols have been extensively used as starting materials for the synthesis of complex organic molecules. The preparation of these high added value compounds is carried out mainly by enzymatic dihydroxylation of arenes since no other efficient chemical method is known. In this paper we describe a detailed study of the biotechnological production at a preparative scale of cis-3-bromo- 1,2-dihydrocatechol (BDC) from bromobenzene using Escherichia coli JM109 (pDTG601), a recombinant strain that harbors the toluene dioxygenase genes from Pseudomonas putida F1. High cell-density cultures of the microorganism (65 g/L cdw) were achieved in a 5 L bioreactor using fed-batch cultures in aerobic conditions. The influence of the biomass concentration in the volumetric and specific productivity of the system, as well as the kinetics of the biotransformation, was thoroughly studied. The use of liquid paraffin as a second phase to relieve bromobenzene’s toxicity resulted in an increased specific productivity for the bi-phasic protocol, which resulted in higher final BDC concentrations. An improvement of 3.2-fold was obtained for BDC production compared to previous reports using this organism. Furthermore, storage strategies of the biocatalytic system as a reagent for organic synthesis are presented.

New 1,8-dioxooctahydroxanthenes with substituents in the 2-, 3-, and 9-positions were obtained by cyclization with iodine from tandem Michael/Michael adducts. Abstract—A series of synthetic chalcones, flavanones, and flavones has been synthesized and evaluated for antitumor activity against the human kidney carcinoma cells TK-10, human mammary adenocarcinoma cells MCF-7 (estrogen receptor-positive), and human colon adenocarcinoma cells HT-29. The most active series is the chalcone ones with the best results against TK-10 and HT-29 cells. Fourteen out of 53 analyzed compounds resulted very active against at least two of the studied tumoral cells. Alkaline single cell gel electrophoresis, comet assay, was performed as a study of the chromosomal aberrations promoted by the compounds on normal cells. Four active and two inactive chalcones were studied in the comet assay against normal human kidney cells (HK-2). A structure–activity relationship analysis of these compounds was performed and for 4- and 3,4-disubstituted derivatives a quantitative correlation was obtained in the case of anti-HT-29 activity

A summary of the resource.
Myrcianthes cisplatensis (Cambess.) O. Berg (Myrtaceae) grows freely in Uruguay especially in the banks of rivers and streams. It is locally known as guayabo colorado and it's fruits are edible and used for the preparation of marmalades. In this work we present the results of the bioguided isolation and structural elucidation of the main active compound as well as it's antibacterial activity. Through repeated chromatography a pure compound could be obtained. The compound was studied by different spectroscopic techniques and could be unambiguously identified as α-methyl-1-(2′, 4′, 6,-trimethoxyphenyl)-1-propanone. When assaying for antistaphylococcal activity, it showed MICs of 62.5 μg/mL for the sensible strain (ATC 6538p) and 250 μg/mL for the multirresistant ones (ATCC 43300 and ATC 700699). This shows that the bioguided fractionation is appropriate even when not very active compounds are isolated

A summary of the resource.
Development of new antimicrobial compounds against different microorganisms is becoming critically important, as infectious diseases are still one of the leading causes of death in the world. The pharmaceutical industry is searching for new lead compounds with novel chemical structures to overcome the increasing resistance to known antibiotics. Plants can be a useful source of these lead compounds. Xanthium cavanillesii Schouw (Asteraceae) grows wild in Uruguay, and its infusion is used in popular medicine as skin antiseptic. In this work, we present the study of the antimicrobial activity of several extracts of X. cavanillesii against different microorganisms. In the agar diffusion assays, the plant extracts showed an interesting antimicrobial activity, including activity against Mycobacterium smegmatis and Candida albicans. The extracts showed low toxicity in the acute oral assay performed with no deaths at a 200 mg=kg dose.

A summary of the resource.
A recycling process which recovers the main components of rejected zinccarbon and alkaline domestic batteries has been developed. The product of recycling is a solution of zinc (32 g.l-1) and manganese (15 g.l-1) salts, free of heavy metals, the use of this solution as wood preservative is being studied. The fungicide potential of the solution is known from previous screening plate tests and tests using wood blocks, in the presence of white and brown rot fungi. Changes were introduced in the recycling process and their effects on fungicide and insecticide properties of the solution are studied in the laboratory, leaching of wood preservative is also analyzed. The evaluation of the solution at four different retention levels was carried out on Pinus taeda and Eucalyptus grandis (sapwood) specimens. Fungicide effect was tested with Fomitopsis palustris and Trametes versicolor. The insecticide potential was tested with Coptotermes formosanus. It is concluded that the changes introduced in the recycling process are not adequate, the solution kept its insecticide properties, but mass losses in fungal tests were higher than expected. In addition, some kind of fixation of the preservative is needed, because leaching leaves wood with low protection.

A summary of the resource.
(2S,3R)-Sitophilate, the male-produced aggregation pheromone of the granary weevil Sitophilus granarius (L.) was prepared stereoselectively using a novel chemoenzymatic approach in 50% overall yield. The synthetic design was based on an enantioselective fungal reduction of ethyl 2-methyl-3-oxopentanoate with a strain of Aureobasidium pullulans (CCM H1), followed by a Mitsunobu inversion at C3. The last step in the synthetic sequence was a lipase-mediated transesterification using the commercially available Candida antarctica B lipase (CaL B, Novozym 435) using microwave irradiation under solvent-free conditions.

A summary of the resource.
The use of oxidoreductases as biocatalysts for the production of a wide number of chiral building blocks is presently a mature (bio-)technology. In this context some industrial applications are currently performed by means of those enzymatic approaches, and new examples are expected to be realized. Moreover, oxidoreductases provide an interesting academic platform to undertake fundamental research in enzymology, to acquire a better understanding on catalytic mechanisms, and to facilitate the development of new biocatalytic applications. Within this area, a wide number of oxidoreductases from genus Candida spp. have been characterized and used as biocatalysts. These enzymes are rather diverse, and are able to carry out many useful reactions, like highly (enantio)selective keto-reductions, (de)racemizations and stereoinversions, and promiscuous catalytic imine reductions. In addition, some Candida spp. dehydrogenases are very useful for regenerating the cofactors, with the aid of sacrificial substrates. Addressing those features, the present paper aims to give an overview of these enzymes, by focusing on practical applications that these biocatalysts can provide. Furthermore, when possible, academic insights on the enzymatic performances will be discussed as well.

The 3-oxygenated-cis-dialkyl-2,5-substituted tetrahydrofuran system, present in several natural products, was prepared with good selectivity by acidic cyclization of 5-alkene-1,2,4-triol derivatives. The starting alkenol was obtained in few steps from a chiral cis-diol resulting from microbial oxidation of bromobenzene. The study of the cyclization allowed the rationalization of all experimental results by assuming a complete ionization at the allylic position and a model close to the one proposed by Labelle for homoallylic induction in five-membered ring closures.

We investigated the accumulation and toxicity of microcystin-LR (MCLR) in the digestive gland of the freshwater clam Diplodon chilensis patagonicus. Treated clams were fed with a toxic strain of Microcystis aeruginosa (NPJB1) during 6 weeks and control clams received the non-toxic strain NPDC1. Filtration rate was estimated for both groups. Toxic effects were evaluated through the hepatosomatic index (HSI) and different oxidative stress biomarkers, lipid peroxidation (content of thiobarbituric reactive substances–TBARS), protein oxidation (carbonyl groups) and reduced glutathione (GSH) levels, and enzymatic activities of superoxide dismutase (SOD), catalase (CAT) and glutathione-S-transferase (GST). The extractable MCLR measured by ELISA in digestive gland extracts showed little or no change during the first 3 weeks and increased significantly at weeks 5 and 6. HSI was reduced by 30% in treated clams at weeks 5 and 6. No significant oxidative damage to lipids or proteins was. All the antioxidant defense parameters analyzed were significantly increased at week 5 or 6. GSH increased in treated clams at week 5, reaching 62% increase at week 6. SOD, CAT and GST activities were significantly increased in treated clams by 50%, 66% and 60%, respectively, at the end of the experiment. D. chilensis patagonicus can be exposed to prolonged cyanobacterial blooms accumulating significant quantities of MCLR, which could be a risk for mammals and birds, which feed on this species and, in a lesser extent, to humans

Uncontrolled combustion due to garbage recycling is a widespread activity among slum dwellers in distressed economy countries and has been indicated as a major source of dioxin contamination. However, because of the high cost and complexity of gas chromatography/high-resolution mass spectrometry (GC-HRMS) analysis, the magnitude of the problem remains largely unknown. The present study describes a first approach toward the use of a dioxin antibody-based enzyme-linked immunosorbent assay (ELISA) as the basis for a sustainable, simple, and low-cost monitoring program to assess the toxicological impact of uncontrolled combustion in slums. A panel of 16 samples was analyzed by GC-HRMS and ELISA on split extracts. Close to 20% of the analyzed samples showed dioxin concentrations up to almost twice the guidance level for residential soil in several countries, pointing out the need for performing a large-scale monitoring program. Despite the potential for variations in dioxin congener distribution due to the mixed nature of the incinerated material, there was a good correlation between the toxic equivalents as determined by GC-HRMS and ELISA. Furthermore, an interlaboratory ELISA validation showed that the capacity to perform the dioxin ELISA was successfully transferred between laboratories. It was concluded that the ELISA method performed very well as a screening tool to prioritize samples for instrumental analysis, which allows cutting down costs significantly.

A collection of 60 non-Saccharomyces yeasts isolated from grape musts in Uruguayan vineyards was screened for β-glucosidase activity and Metschnikowia pulcherrima was the best source of this enzyme activity. Its major β-glucosidase was successfully purified to homogeneity by ion-exchange chromatography on amino-agarose gel. The enzyme exhibited an optimum catalytic activity at 50°C and pH 4.5 and was active against (1 → 4)-β and (1 → 2)-β glycosidic linkages. In spite of preserving 100% of its activity and stability in the presence of 12% (v/v) ethanol and 5 g glucose/l, the enzyme was unstable below pH 4. We characterized the β-glucosidase from M. pulcherrima with a view to its potential applications in wine-making.

Entre las opciones más utilizadas para el diagnóstico de infecciones mediante imágenes encontramos radiografías, ultrasonido, CT, MRI complementados con leucocitos marcados u otros radiofármacos que proporcionan información anatómica y funcional para localizar los focos. Sin embargo no siempre conducen a una interpretación clínica sencilla y fiable debido a una baja especificidad y sensibilidad. En este trabajo se presentan los resultados del estudio comparativo del marcado de dos antifúngicos: Fluconazol y Voriconazol como posibles agentes de diagnóstico. La metodología utilizada es la sustitución a partir del precursor triaquotricarboniltecnecio(I). Se trabajó con modelos animales de ratones CD1 con lesiones causadas por inflamación estéril, infección por Candida albicans y por Aspergillus niger. Los resultados biológicos muestran que ambos complejos presentan buena especificidad en la diferenciación de inflamación y de infección. Las imágenes centellográficas, sin embargo, no logran alcanzar una sensibilidad adecuada para la detección de estos dos procesos.

Infections by Candida spp and Aspergillus spp are the most common causes of invasive fungal infections. The main diagnostic methods are blood culture, and antigen-based techniques, but they are still suboptimal, leading to delays in the initiation of therapies and resulting in high mortality rates despite the availability of several new antifungal agents. The aim of this work was the development, synthesis and evaluation of a potential radiopharmaceutical enable the rapid and accurate diagnosis by scintigraphic images of fungal infection using caspofungin, a lipopeptide, radiolabelled with (99m)Tc. Caspofungin was radiolabeled with (99m)Tc-tricarbonyl precursor. The complex was assessed for in vitro stability, lipophilicity, plasma protein binding and plasma stability. Biological evaluation was conducted in four groups of CD1 female mice. G1 healthy animals, G2 was induced sterile inflammation with turpentine oil. G3 and G4 were infected with Candida albicans and Aspergillus Niger. Scintigraphicimages were acquired before sacrifice. The Caspofungin - tricarbonyl complex was obtained with RCP higher than 95%, it was stable in labeling milieu for at least 20 hours, and in plasma for 4 hours. Challenge with competitive agents showed no ligand exchange during 200 min. The product was well tolerated by mice and showed mainly hepatobiliar excretion. Lesion uptake was markedly higher in infected tissues than in sterile inflammation. Scintigraphic images clearly distinguished inflammation from infection. The high RCP yields and in vitro stability, the targeted biodistribution profile and good T/NT ratios, outlines this complex as a potential agent for rapid and specific diagnosis of infections caused by pathogenic yeasts.

In this work copper, molybdenum and sulfur determinations in grass, used as biomonitor of the soil, were performed during the period 2008-2009 and compared with the levels obtained in 2000-2001 in the same locations with the aim of observing environmental changes after the implementation of a program recommending oral and injected copper supplements in cows and after the use of fertilizers containing molybdenum and sulfur elements that can affect copper bioavailability. Several correlations with meteorological factors were also studied. The results showed that the copper level increased in grass but the concentrations remained below the recommended values. The mean copper concentrations in spring, summer, autumn and winter were 2.44, 3.12, 4.29 and 5.21 mg kg-1 DM, respectively, and for molybdenum were 0.20, 0.16, 0.22 and 0.17 mg kg-1 DM, respectively (DM = dry matter). In many samples, the molybdenum concentration was above the recommended range and consequently a secondary defi ciency can be observed. In addition, a seasonal variation in grass, an inverse correlation between copper and molybdenum concentrations and a direct correlation between copper levels and the relative humidity were observed permitting visualization of the most critical periods of hypocuprosis.

As a part of our studies of copper(II) complexes with possible antitumor activity, the synthesis and structural characterization of two new copper terpyridine (terpy) complexes: [Cu(terpy)(SO4)(H2O)]·2H2O and [Cu2(terpy)2(SO4)2]·4H2O are presented. The crystal structures show that the copper ion in both complexes is in a square pyramidal environment, coordinated equatorially to the terpy N atoms and completing the coordination with O atoms from the sulfate or water molecules. The second compound is a sulfate bridged dinuclear complex. In aqueous solution the coordination scheme is modified. Taking the crystal structure as a starting point the geometries of the complexes were optimized and vibrational spectroscopy calculations were performed allowing an assignation of the experimental IR spectra. Finally, the interaction of the complexes with DNA was studied, by UV titration and docking studies of the complexes. The results suggest that the interaction occur by minor groove interaction and partial insertion of the terpy into the DNA bases and that terpy interaction with DNA is strongly influenced by the Cu-coordination process.
