The 5-(n)-alkylresorcinol fraction of the epicuticular waxes of Hordeum vulgare seeds appeared to be responsible for their in-born resistance to pathogenic fungi such as Aspergillus niger and Penicillium crysogenum. The antifungal properties of this fraction were evaluated qualitatively and quantitatively with a novel bioassay where the extreme lipophilicity of these compounds was taken into account. The minimum inhibitory concentration in the fungi tested ranged from 5.6 to 10 micrograms cm-2 for the alkyresorcinols. The behaviour of the different cultivars against these fungi could be predicted by measuring the natural amount of resorcinols of each variety by TLC-scanning densitometry. The ranking of cultivars thus established correlated well with the field behaviour of each cultivar, providing a useful and rapid method for predicting the behaviour against fungi of new varieties being developed. Sulfonamides, derivatives of sulfanilamide, are classified as antimicrobialdrugs. Several authors have reported modified pharmacological and toxicological properties when some of these drugs are administered in the form of copper complexes. Taking into account the importance of these compounds, we have prepared and characterized the copper (II) complexes with sulfameter, sulfamethazine, sulfamerazine, succinylsulfathiazole, sulfadiazine. sulfamethoxypyridazine and sulfachlopyridazine and investigated their solution behavior. Characterization by means of elemental analysis, TGA, IR and UV-Vis spectroscopy showed that all complexes coordinate through the oxygen of the sulfonic acid and the heterocyclic nitrogen. We are also reporting the superoxide dismutase mimetic activity. The average values of IC50 is 250 11M . The complexes were screened for their activity against E.coli and S. aureus, showing a significant antimicrobial activity. The levels of superoxide dismutase (SOD) were determined in detergent-soluble, somatic and excretion–secretion (E–S) preparations from adult Fasciola hepatica using the xanthine oxidase system and visualized in substrate gels. Compared to detergent-soluble and somatic extracts, E–S products showed the highest SOD activity (88 ·5 U/mg), indicating active release to the medium in which parasites were maintained. SOD specific activity was also detected at high levels in E–S products from 3-week-old and 5-week-old immature migrating flukes (25 and 143 U/mg, respectively). In all preparations except for the somatic extract, the activity was characterized as cyanide-sensitive CuZn SOD. Differences in SOD isoenzyme profiles between the extracts were observed in native polyacrylamide gel electrophoresis: the somatic and detergent-soluble extracts exhibited 1 band of activity while the E–S products from immature and adults flukes contained 2 and 3 migrating bands, respectively. SOD was purified from the detergent-soluble extract and E–S products of adult worms by a combination of ultrafiltration, gel filtration on Sephacryl S-200 HR and ion-exchange chromatography on QAE Sephadex A-50. The SOD from detergent-soluble extract showed, by SDS–PAGE analysis, 1 band of 16 kDa apparent molecular weight. The SOD from E–S products showed 2 bands of 16 and 60 kDa apparent molecular weight. N-terminal sequence analysis of the 16 kDa band from the detergent-soluble preparation showed some similarity with Schistosoma mansoni cytoplasmic SOD. These enzymes may have a potential role in the evasion of the oxidative burst killing mechanism by immune cells. During a screen of Neisseria gonorrhoeae clinical isolates obtained in Uruguay for susceptibility to azithromycin, we noticed that approximately 10% of the strains examined displayed decreased susceptibility to azithromycin and erythromycin due to the mtr(CDE)-encoded efflux pump system, but remained susceptible to Triton X-100. Molecular dynamics simulations of the zinc finger domain of protein Zif268, in a complex with a high affinity DNA sequence, yields a globally stable system with small yet significant readjustments with persistence time of the order of 1.1ns. The results confirm the quality of the standard GROMOS87 force field with a corrected solvent-to-solute interaction that does not affect the water-water SPC interactions nor the intra-molecular cohesive forces. The factors contributing to the molecular stability found in GROMOS' potential energy function appear to be sufficient to yield stable fluctuation patterns when surrounding medium effects are properly included. Complement depletion by treatment with cobra venom factor caused a 79% reduction in cyst numbers (P < 0.05). Possible explanations of this unexpected result are discussed. The results presented suggest that lysis or opsonization by host complement are not effective against the establishing parasite in this model. They also indicate the significance of complement activation by the parasite needs to be studied at a local level. Among them, only α-amiryn is incorporated at as high levels as cholesterol into DPPC bilayers. At 50 mol%, it results in the phase transition vanishing. Pentacyclic monoalcohols are more potent than cholesterol to decrease the DPPC gel phase order, but less potent than it to increase the liquid–crystalline state order. They also produce more packing defects than cholesterol in DPPC bilayers. These effects correlate with the absence of planarity in the pentacyclic skeleton, which would obstruct the `all trans' packing of acyl chains in the gel state and Van der Waals interactions in the liquid–crystalline state. Taraxerone has little effect on DPPC bilayers, probably due to its difficulty to form hydrogen bonds. In this work we analysed the modulation of the antibody response to Echinococcus granulosus antigens via anti-idiotype (Ab2) administration. In a first set of experiments, we determined the antibody response to hydatid cyst fluid antigen (HCFA) of mice immunized with Ab2. Results showed that Ab2 elicited anti-HCFA antibodies in mice that had never been exposed to HCFA before. Moreover, that response was characterized by booster effect, avidity maturation and an inverse correlation between avidity and Ab2 dose. We infected these mice and measured the titre and avidity of anti-HCFA antibodies during 250 days of infection. Lack of avidity maturation was the most important feature observed, suggesting that the parasite (either protoscolex or cyst) could modulate the antibody response of its host. In a second set of experiments, we investigated the presence of regulatory anti-idiotypes in the Ab2 preparation. In this context we treated neonates with different doses of Ab2, immunized them with HCFA 12 weeks later and determined their anti-HCFA titres. A specific and Ab2 dose-dependent suppression of the response to HCFA was observed, suggesting the existence of regulatory anti-idiotypes in the Ab2 preparation. Although this Ab2 could be involved in the regulation of the anti-HCFA response, other anti-idiotypes could also be involved. Our results show that it may be possible to improve the avidity of the anti-HCFA response via the administration of the anti-idiotype Ab. However, the live parasite could successfully revert this effect by mechanisms not yet characterized. We describe the avidity maturation of IgGs in human toxoplasmosis using sequential serum samples from accidental and natural infections. In accidental cases, avidity increased continuously throughout infection while naturally infected patients showed a different profile. Twenty-five percent of sera from chronic patients having specific IgM positive results could be appropriately classified using exclusively the avidity test data. It has been reported that restricted and unrestricted MP2 ab initio calculations of the energy difference between the formaldehyde (H2CO+) and hydroxymethylene (HCOH+) radical cations are seriously in error because of poor convergence of the perturbation series. We had reported a study of the formaldehyde—water radical cation and found MP2 to be reliable. A study of the correlation energy contribution to the isomeric structures formaldehyde—water and hydroxymethylene—water radical cations is reported. Both perturbation theory (from MP2 to MP4SDTQ) and configuration interaction (CISD and CISD(Q)) calculations were performed with the 6-311 + G(d, p) basis set at the MP2/6-311 + G(d, p) optimum geometries. The MP2 flaw in formaldehyde/hydroxymethylene is not present in the radical cations derived from the complexes with water. Analysis of our results for formaldehyde/hydroxymethylene shows that the flow is mainly connected with poor convergence of the perturbation series for the formaldehyde radical cation (and not for the hydroxymethylene). Since proton transfer occurs in the complexes with water, this species is not present in our calculations, so the results are reliable. The hydroxymethylene—water radical cation is predicted to be more stable than the formaldehyde—water radical cation by 29 kJ/mol on the basis of CISD(Q)/6-311 + G(d, p)//MP2/6-311 + G(d, p) calculations. The kinetics of the oxygen electroreduction reaction (OERR) were investigated on (1 1 1)-type and (1 0 0)-type faceted, and polycrystalline platinum electrodes in aqueous (0.05–1.0)m trifluoromethanesulfonic acid (TFMSA) using the rotating disc and ring-disc electrode techniques at 25°C. Reaction orders with respect to oxygen close to either 1/2 or 1 were found, depending on the TFMSA concentration and platinum surface morphology. At all TFMSA concentrations the formation of H2O2 was enhanced at (1 0 0)-type platinum surfaces. The difference in the electrocatalytic activity of platinum surfaces can be explained through data derived from the OERR formalism proposed by Damjanovicet al. The rate of the direct O2 to H2O electroreduction reaction increased steadily with the cathodic overvoltage irrespective of the platinum surface morphology, whereas a maximum H2O2 formation rate was found at about 0.5 V, depending on the TFMSA concentration. The H2O2 decomposition rate on (1 0 0)-type platinum electrode yielding H2O approached zero within a certain potential range. The lipid content and the fatty acid composition of the fillets of the six fishes burriqueta (Menticirrhus americanus), white croaker (Micropogonias furnieri), black drum (Pogonias cromis), Brazilian menhaden (Brevoortuaspp.), pampano (Parona signata), and king weakfish (Microdon ancylodon) of the Rı́o de la Plata in Uruguay were analyzed and evaluated as sources of polyunsaturated fatty acids in comparison with fish oil capsules and hake (Merluccius hubbsi) liver oil. The lipid content of the fish fillets was found to vary over a wide range (0.56 to 13.6 g/100 g fillet), while the protein level varied very little (19.2 to 20.7 g/100g fillet). The percentage of docosahexaenoic acid (DHA) exceeded that of the eicosapentaenoic acid (EPA) in all the fish species analyzed. The actual content of these acids in the fillets indicated that Brazilian menhaden, burriqueta, and pampano provide the recommended daily intake of 1 g of EPA + DHA with reasonable amounts of fillet. The energy input for a daily gram intake of EPA + DHA provided by fish fillets ranged from 300 kJ for fatty fishes to more than 3000 kJ for lean fishes. For the same dose of EPA + DHA, the energy input provided by fish oil capsules is less than 250 kJ. Hake liver oil is a rich source of polyunsaturated fatty acids and a potential raw material for the production of fish oil capsules. Essential oils obtained from the dried plants harvested in full bloom of Origanum x applii (Domin) Boros from Uruguay, were analyzed by GC. Three different local populations (O.A. 1030, O.A. 1031 and O.A. 1032) were considered and statistically compared. Twenty-seven oil constituents were identified. The major oil components for O.A. 1030 and O.A. 1031 were respectively thymol (26.41%), linalyl acetate (16.64–20.04%) and γ-terpinene (9.04–9.19%), while O.A. 1032 had higher percentages of p-cymene (20.80%), terpinen-4-ol (17.00%) and sabinene (5.32%). A clear chemotypic partition was present between O.A. 1032 and the two other populations.
