A summary of the resource.
Lignocellulosic materials represent a sustainable source of chemicals and fuels because of their lowcost and ample supply. The current limitations on bioconversion of lignocellulosic biomass include poorenzyme stability and inhibition by secondary or final products. Here, we report the biochemical char-acterization of a novel, rumen metagenome-derived glucanase and xylanase, named EndoG. From thededuced amino acid sequence it was assigned to glycoside hydrolase family 5. EndoG showed similarityto non-characterized proteins, and its parental organism is likely related to the genus Prevotella. The1146pb ORF encoding EndoG was over-expressed in Escherichia coli and the protein purified. The recom-binant EndoG displayed a wide range of pH activity with a maximum at pH 5.0 and at least 65% activityat pH between 4.5 and 7.5. The enzyme was highly stable at 55◦C for 1 h, and retained 81% activity at 4 MNaCl. EndoG was also active in the presence of diverse divalent cations, detergents, EDTA, acetate, fur-fural, imidazolium ionic liquids, and ethanol. Glucose or cellobiose had no effect in EndoG performance.EndoG behaved as a multifunctional endo- and exo-glucanase, as well as xylanase, displaying activity on4-methylumbelliferyl-�-d-cellobioside, p-nitrophenyl-�-d-cellobioside, carboxymethylcellulose (CMC),phosphoric acid swollen cellulose, avicel, xylan, lichenan and sugar cane bagasse. To evaluate its biotech-nological potential, ethanologenic E. coli MS04 cells expressing either EndoG or a Thermobifida fusca�-glucosidase were co-cultured in minimal media with avicel or CMC as the sole carbon source attaining2.0 g/L and 3.3 g/L of ethanol, respectively.

Abstract
A summary of the resource.
The essential oil of aerial parts of Porophyllum ruderale (Jacq.) Cass. was analysed by HRGC, HRGC–MS (quadrupole) and 26 components (93% of the total composition) were identified. The enantiomeric distribution of β-pinene, sabinene, 4-terpineol and α-terpineol was studied by multidimensional HRGC–HRGC. The major component was sabinene (64%) with an enantiomeric purity of 97% for (+)-sabinene.

La industria del cuero se encuentra entre las seis industrias mas importantes del pais por su nivel ocupacional, incidencia en el PBI nacional y su capacidad exportadora. El volumen de efluentes producidos por el sector se estima en 2:960.000 m3 por afio y se caracteriza por su elevada carga organica e importante nivel de toxicidad debido a su contenido en sulfuro y cromo. En el Departamento de Bioingenieria se ha desarrollado un proceso de depilado enziIruitico de pieles ovinas (con extensi6n futura a pieles bovinas) que permite disminuir la carga organica y t6xica de estos efluentes al recuperar el pelo y eliminar el uso de sulfuros en los bafios de depilado. El objetivo de esta tesis es profundizar en el conocirniento de tres aspectos de este proceso: I) el rnicroorganismo, II) las enzimas presentes en el extracto depilante y III) el proceso de su producci6n. La cepa seleccionada como buena productora de enzimas depilantes se identificó como perteneciente a la especie B. subtilis segUn estudios morfo16gicos, culturales y fisio16gicos realizados. Se estudió la incidencia en la producción de proteasas de los componentes del medio de cultivo: caldo nutritivo, peptona, triptona, extracto de carne, extracto de levadura, glucosa, ~Cl, Mg2+, Ca2+, Fe2+, Mn2+ y Zn2+. Los resultados obtenidos mostraron que el caldo nutritivo es un medio apropiado para sustentar el crecirniento y la producción de proteasas de este Bacillus, que la actividad proteolitica producida aument6 con la adici6n al medio de cultivo de extracto de levadura (3 g/l), glucosa (5 g/l), Ca2+(3 mM), Mn2+ (0.8 mM) y disminuy6 con ~Cl (4 g/l) y Fe2+ (0.1 mM), que la actividad del extracto crudo producido se mantiene al suplantar el caldo nutritivo por dos de sus componentes la triptona y el extracto de carne En la busqueda de un medio de cultivo econórnico que permita su utilización a nivel industrial, se 10gr6 una producci6n de proteasas del orden de la obtenida con caldo nutritivo en un medio que contiene peptona y glucosa en cultivo por lote alimentado donde la concentraci6n de glucosa en el medio de cultivo varió entre - 0-2.5g/l. Del estudio de los mecanismos regulatorios que intervienen en la producción de proteasas se vió que: I) la sintesis de parte 0 el total de las proteasas es inducible y esta sujeta a represi6n catab6lica por glucosa; II) la producci6n de proteasas sigue una cinetica de formaci6n de producto mixta; III) las enzimas contenidas en el extracto crudo estan invoiucradas en por 10 menos dos procesos, proteasas ligadas a la hidr6lisis de fuentes de carbono y nitr6geno complejas y proteasas vinculadas a la esporulaci6n. Mediante SDS-PAGE y electroforesis de actividad se caracterizaron las proteasas presentes en el extracto crudo, 1 serino proteasa y 3 proteasas cuyas caracteristicas se asirnilan al de las metalo proteasas. El PM relativo de las metalo proteasas es de 151, 106 Y 39 kDa, su rango de actividad se extiende entre pH 5 Y 10 y su actividad se vio inhibida parcialmente con PMSF y totalmente con EDT A. La serino proteasa tiene un PM relativo de 80 kDa, su rango de actividad se limita a pH 7-8, se inhibe parcialmente con pCMB y totalmente con PMSF y EDTA y su actividad depende del ion Ca 2+.

La investigación científica en la Antártida se ha incrementado en los últimos años, fundamentalmente en áreas como microbiología, biología y oceanografía. Existen pocos reportes sobre los niveles de contaminantes inorgánicos y el impacto antrópico relacionado a ellos en el ecosistema. Al ser un territorio prácticamente virgen, es de particular interés realizar esta evaluación. En este trabajo se analizaron por primera vez y como primera aproximación al abordaje del tema seis muestras de aguas y seis muestras de suelos obtenidas en seis puntos de muestreo próximos a la base uruguaya en Antártida. Los elementos determinados para ambas matrices fueron: cromo, níquel, cobalto, arsénico, estroncio, cadmio, mercurio, plomo, uranio. En agua también se determinaron torio, estaño y antimonio. La preparación de las muestras de suelos se realizó mediante un tratamiento asistido con radiación de microondas; las muestras de agua fueron analizadas directamente. Las determinaciones analíticas se realizaron mediante espectrometría de emisión atómica de plasma de acoplamiento

The serodiagnosis of hydatid disease is a valuable instrument for clinical diagnosis and epidemiological surveillance of high-risk populations. In the past decade a wealth of reports on the diagnostic performance of numerous antigens have been produced. However, their diagnostic value has been estimated under different conditions, using different serum collection, therefore precluding their direct comparison. Here we report an unbiased comparison of the same batch of six major E. granulosus antigens, namely, hydatid cyst fluid (HCF), native antigen B (AgB), two recombinant AgB subunits, an AgB-derived synthetic peptide, and recombinant cytosolic malate dehydrogenase from E. granulosus (EgMDH), against the same serum collection. The doubleblind analysis was performed using a standardized protocol and receiver operating characteristic (ROC) data analysis by a network of six South American laboratories. High intercenter reproducibility was attained, and the intralaboratory analysis allowed the comparative ranking of the antigen panel. HCF, AgB, and its AgB8/1 subunit exhibited equivalent diagnostic efficiencies, 81.4% � 0.5%, 81.3% � 0.6%, and 81.9% � 2.0%, respectively; with a more favorable balance toward specificity in the case of the last antigen. The diagnostic efficiencies for the other three antigens were 76.8% � 6.8%, 69.1% � 2.7%, and 66.8% � 2.1%, for the peptide, the AgB8/2 subunit, and the EgMDH, respectively. The study also included an analysis of batch-to-batch variation in the diagnostic performance of different HCF regional preparations. Based on these results, a suggested recommendation on the use of these antigens was drawn.

Since its early introduction as a marker for the immunodiagnosis of hydatid disease, antigen 5 (Ag5) has been regarded as one of the more relevant antigens of Echinococcus granulosus, and it is still widely used in different confirmation techniques. In this work we prepared 2 recombinant forms of the antigen, namely, rAg5 (corresponding to the unprocessed polypeptide chain of the antigen) and rAg5-38s (corresponding to its 38 kDa subunit). Their antigenicities were compared to that of the native antigen using a human serum collection. There was a major drop in the reactivity of the sera, particularly against rAg5-38s, which was confirmed by analysis of the cross-reactivity of 2 panels of monoclonal antibodies specific for rAg5-38s and the native antigen. Using the chemically deglycosylated native antigen, we demonstrated that the reduced antigenicity of the recombinants is due to the loss of the sugar determinants, and not to their misfolding. Inhibition experiments using phosphorylcholine confirmed that this moiety also contributes to the reactivity of the antigen, but to a much lesser extent. The presence of immunodominant highly cross-reactive glycan moieties in the Ag5 molecule may involve a parasite evasion mechanism.

A summary of the resource.
Antigen 5 (Ag5) is a dominant secreted component of the larval stage of Echinococcus granulosus, and is highly immunogenic in human infections. Although the diagnostic value of Ag5 has been thoroughly evaluated, there has been little progress in its molecular characterization and the understanding of its biological role. In the present study, the Ag5 gene was cloned by reverse transcription-PCR on the basis of the amino acid sequences of tryptic fragments. The nucleotide sequence indicates that Ag5 is synthesized as a single polypeptide chain that is afterwards processed into single disulphide-bridged 22 and 38 kDa subunits. Whereas the 22 kDa component contains a highly conserved glycosaminoglycan-binding motif that may help to con®ne Ag5 in the host tissue surrounding the parasite, the 38 kDa subunit is closely related to serine proteases of the trypsin family.

Essential oils obtained by hydrodistillation from aerial parts of Hedyosmum angustifolium (Ruiz & Pav´on) Solms-Laubach and H. scabrum (Ruiz & Pav´on) Solms-Laubach from Bolivia were analysed by GC–FID and GC–MS. Monoterpene hydrocarbons, especially ˛- and ˇ-pinene, and oxygen containing monoterpenes, mainly 1,8-cineole and linalool, were the major constituents found in the oils of H. angustifolium. The oil of H. scabrum was mainly made up of monoterpenes, in particular υ-3-carene, the sesquiterpene hydrocarbons ˛- gurjunene and germacrene D, and 30, 40-dimethoxypropiophenone. For both oils, the enantiomeric distribution of ˛-pinene, sabinene, ˇ-pinene, limonene, linalool, terpinen-4-ol and ˛-terpineol was studied by multidimensional HRGC–HRGC. The main difference between the oils corresponded to the reverse enantiomeric distribution found for linalool, indicating a clear index of different metabolic response in each species.

Background and Objectives Ketoprofen, a potent nonsteroidal anti-inflammatory drug, is clinically administered as a racemic mixture. One of the possible metabolism routes of ketoprofen is the inversion of the R- to S-enantiomer in the gastrointestinal tract. Ketoprofen, as a weak acid drug, might undergo recirculation through pancreatic/intestinal juices. The aim of the work was to investigate if a plasma-gastrointestinal tract recirculation of ketoprofen could explain its R-to-S chiral inversion after the oral administration of two modified-release formulations: a gastro-resistant delayed-release tablet (Reference) and an extended-release-plus-immediate-release bilayer tablet (Test). Methods Sixteen healthy Caucasian volunteers (eight women and eight men) participated in a ketoprofen bioequivalence study. Both formulations were administered with and without food. In both cases, standard meals were given throughout the experiment. R- and S-enantiomers were measured separately using a validated HPLC–UV chiral method. Mean concentration–time profiles of ketoprofen enantiomers in plasma were obtained for men and women. Area under the plasma concentration–time curve, maximum ketoprofen plasma concentration, and time-to-peak were also computed for both isomers, both modes of administration, and both sexes. S/R concentration ratio was assessed as an indicator of enantiomer chiral inversion rate. Results Differences in the pharmacokinetics of S- and R-ketoprofen enantiomers were found after the Test administration. S-Ketoprofen presented a lower plasma exposure compared to R-enantiomer. However, the S/R concentration ratio increased 1 h (in men) and 2 h (in women) after meal intakes. This was related to pancreatic and/or intestinal and/or biliary secretions of the drug, followed by reabsorption and conversion of the R- to the S-isomer. The lower intestinal pH reported for men would lead to a higher oral bioavailability of the Test formulation and a higher reabsorption of both ketoprofen isomers in this sex. Hence, a higher rise of the S/R concentration ratio could be observed in men. No significant differences between isomers exposure were detected in both sexes after the Reference administration. Different lag times were observed after fed and fasting administration of this formulation; however, drug absorption coincided with food ingestion. Then, drug recirculation affected the S/R ratio from the beginning of drug exposure, minimizing the difference between isomers disposition. Conclusions R-to-S conversion rate could be mainly associated with several passages of the drug through the intestinal mucosa. The concentration–time profiles of ketoprofen in plasma after the administration of both formulations evidenced R-to-S conversion of recirculating drug following meal intakes.

La respuesta farmacocinética de un fármaco es variable y depende de múltiples procesos. Esta respuesta puede complejizarse, por ejemplo, debido a procesos de recirculación plasma – tracto gastrointestinal – plasma del fármaco. Las moléculas secretadas pueden volverse a absorber, y en dicho caso no se hablaría de excreción sino de circulación entre la sangre y el fluido hacia el cual se pretendió eliminar. El proceso más ampliamente reconocido en farmacología es la recirculación entero-hepática. Sin embargo, en la presente tesis se profundiza en otros posibles circuitos de recirculación. Un trayecto del fármaco podría ser a través del jugo gástrico, en donde partiendo desde la sangre, alcanzaría dicho fluido para luego ingresar al medio interno desde la luz intestinal cuando el mismo se vacíe desde el estómago tras la apertura del píloro. Otro podría ser mediante el jugo pancreático, el cual es secretado a través del ducto pancreático que se conecta con el hepático inmediatamente antes de vaciar su contenido hacia el duodeno. Varios mililitros de ambos jugos son vertidos en pocos minutos hacia el tracto gastrointestinal, activados esencialmente por la llegada de alimentos al estómago y al intestino, respectivamente. Estos trayectos adquieren relevancia en moléculas ionizables dado el pH que posee cada uno de estos fluidos. Existe un gradiente importante de pH entre ambos y el plasma (pH 7,4). De este modo, las drogas básicas preferirán la transferencia desde el plasma hacia el jugo gástrico de pH entre 1,2 y 2,3, para luego verse impedido su retorno a plasma por ionización a este pH ácido. En tanto las drogas ácidas, desde el plasma hacia el jugo pancreático de pH 8 aproximadamente. El objetivo de la tesis fue estudiar la circulación plasma-tracto gastrointestinal-plasma de moléculas ácidas y básicas, que a su vez presenten estereoisomería. Los fármacos estereoisómeros, si bien poseen las mismas propiedades físico- químicas, pueden diferir en su farmacocinética y farmacodinamia. Los procesos de recirculación pueden afectar directamente la exposición a los diferentes isómeros ya que una posible ruta de metabolización para este grupo de fármacos es justamente la inversión quiral y al atravesar el enterocito puede ocurrir la conversión de un isómero en el otro. La investigación realizada demostró la recirculación a la que son sometidos algunos fármacos. Además, repetidamente se observó que luego de la ingesta de comidas, el cociente de concentraciones de isómeros modificaba la tendencia que seguía. Muchas veces los procesos de recirculación aumentan el tiempo de residencia de las moléculas en el organismo, prolongado su acción, sin embargo, en algunos fármacos, como para Metadona, el isómero S podría no favorecerse de este proceso ya que cada vez que atraviesa el enterocito aumenta su eliminación. Para los AINES, la ingesta de comidas podría en algunos casos promover la relación entre isómeros que resulta más beneficiosa para la terapia, al favorecer la conversión R-en-S, pero en otros casos, como para Ibuprofeno, podría no impactar sobre la exposición al isómero activo ya que la mayor tasa de conversión no logra compensar el gran aumento de clearance intestinal que se ve para ambos isómeros cuando se administran comidas frecuentes. Debe considerarse cada caso en particular para poder hacer una recomendación respecto, por ejemplo, al modo de administración. Por esto es interesante destacar, que este tipo de 5 investigaciones, si bien aportan valiosa información respecto a la farmacocinética de determinados fármacos, principalmente constituyen herramientas poderosas para el entendimiento de la fisiología, lo que permite la aplicación del conocimiento a diferentes casos. Otro factor que demostró condicionar de manera importante la farmacocinética de los fármacos estudiados, fueron las diferencias fisiológicas entre hombre y mujeres. Se observó el más lento vaciado gástrico de las mujeres respecto los hombres, su mayor pH intestinal y el impacto del tamaño corporal sobre procesos de metabolización saturables. Resumiendo, se demostró la disímil exposición a los isómeros que puede obtenerse tras la administración de una mezcla racémica y cómo diferentes variables impactan sobre la relación entre los isómeros. Por más que la terapia estereoselectiva resulte beneficiosa, por ejemplo, porque solo uno de los isómeros sea farmacológicamente activo, la mayoría de los fármacos estereoisómeros que se encuentran en el mercado, se administran como racematos debido al coste económico y a la complejidad de la separación de los isómeros. Esto introduce la necesidad de considerar el seguimiento de la terapia con estos medicamentos a través de la determinación de los isómeros por separado, ya que, como se mencionó, el perfil farmacocinético y farmacodinámico de los isómeros puede ser diferente y, por lo tanto, carece de lógica el análisis de la mezcla racémica. En el presente trabajo, se pone de manifiesto la ventaja de la determinación quiral. De otra manera, no hubiese sido posible la detección de los procesos estudiados y el correcto análisis de los resultados. Por otro lado, la información recabada podría ser de utilidad para el desarrollo de formas farmacéuticas innovadoras que permitan un más eficiente uso de la dosis racémica contenida. Es de destacar la importancia de la investigación, en este caso en farmacocinética y en los factores que impactan sobre la misma, además del entendimiento de diferentes aspectos de la fisiología humana, para lograr un mejor aprovechamiento de los fármacos disponibles en el mercado, promoviendo el uso racional de los mismos, y para guiar e incentivar el desarrollo de nuevos fármacos a nivel nacional, siempre con el fin último de mejorar los perfiles de seguridad y eficacia de los tratamientos farmacológicos en beneficio de los usuarios

<p>Invasive fungal diseases represent an unmet clinical need that could benefit from novel immunotherapeutic approaches. Host pattern recognition receptors (e.g., Toll-like receptors, C-type lectins, or scavenger receptors) that sense conserved fungal cell wall constituents may provide suitable immunotherapeutic antifungal agents. Thus, we explored the therapeutic potential of the lymphocyte class I scavenger receptor CD5, a nonredundant component of the antifungal host immune<br />response that binds to fungal �-glucans. Antifungal properties of the soluble ectodomain of human CD5 (shCD5) were assessed in vivo in experimental models of systemic fungal infection induced by pathogenic species (Candida albicans and Cryptococcus neoformans). In vitro mechanistic studies were performed by means of fungus-spleen cell cocultures. shCD5-induced survival of lethally infected mice was dose and time dependent and concomitant with reduced fungal load and increased leukocyte infiltration in the primary target organ. Additive effects were observed in vivo after shCD5 was combined with suboptimal doses of fluconazole. Ex vivo addition of shCD5 to fungus-spleen cell cocultures increased the release of proinflammatory cytokines involved in antifungal defense (tumor necrosis factor alpha and gamma interferon) and reduced the number of viable C. albicans organisms. The results prompt further exploration of the adjunctive therapeutic potential of shCD5 in severe invasive fungal diseases.</p>

Mercuric iodide platelets were grown in the HgI 2 }HI}H 2 O system by solvent evaporation at constant temperature. Di!erent solution temperatures, growing times and stirring conditions were studied. In order to determine the best growing parameters the chemistry of the HgI 2 }HI}H 2 O system was studied. Complexes [HgI 3 ]~ and [HgI 4 ]~2 were identi"ed and quanti"ed in the solution by visible and UV spectrometry, according to the R ratio ([HgI 2 ]/[HI]). Solution oxidation and I~3 interference were eliminated performing growth in high purity Argon atmosphere. Good mercuric iodide platelets, up to 6.0]7.0]1.5mm3 in size, were obtained with appropriate argon #ow, temperature and growing time, for several independent experiences. Platelets were observed by optical microscopy and the absence of possible dislocations was con"rmed by studying etch pits density after etching with ethanol}trichloroethylene. Nonexistence of occluded water and hydrocarbons was con"rmed by Fourier transform infrared spectroscopy and was related to results obtained for mercuric iodide crystals grown by the physical vapor transport method. Detectors were made with the platelets and their apparent resistivity, current density and X-ray performance were measured. Discussions about future improvements of growing mercuric iodide platelets in the HgI 2 }HI}H 2 O system and comparison of this method with others are included.

Soybean beta-amylase was immobilized on phenylboronate-agarose by strong interactive binding. The insoluble derivative was active and more stable to temperature changes than the free enzyme. The absence of enzyme leakage even in the presence of substrate was demostrated. Changes in pH over a wide range(4.0-8.0) did not affect the stability of the complex. The support could be recovered by sorbitol elution |b which demonstrated the reversibility of the binding. Since the enzyme was not reatained on phenylagarose under similar conditions |b we rejected hydrophobic interactions as a cause of the strong binding of the enzyme to phenylboronate-agarose. We suggest that the bonding of the enzyme to the phenylboronate ligand occurs by a charge transfer mechanism between the trigonal boronate and the side chain nitrogenated groups. It was concluded that phenylboronate-agarose has good properties as a support |b which recommends its use for the preparation of immobilized enzymes

The development of sensitive and easy-to-use biosensors that allow an adequate characterization of specific weak biological interactions like carbohydrate–lectin interactions still remains challenging today. Nanoparticles functionalized with carbohydrates are one of the most powerful systems for studying carbohydrate–lectin interactions, because they mimic the multivalent presentation of carbohydrates encountered in nature, for example when viruses and bacteria bind to cells. On the basis of the model system glucose–Concanavalin A (ConA), we explore the application of Transient Magnetic Birefringence (TMB) to study these weak interactions, using glucose-functionalized colloidal magnetite nanoparticles (NPs) as probes. We demonstrate that the binding dynamics can be monitored and derive a model to obtain the apparent cooperativity. For our studies, we use nanoparticles of 6 and 8 nm in diameter. The ConA-generated response shows apparent cooperativity, due to the cross-linking of nanoparticles by the ConA tetramer which has four binding sites. Cooperativity is higher for 6 nm NPs, possibly due to a better accessibility of all four ConA binding sites on smaller NPs, enhancing cross-linking. For this system, we find a detection limit of 3–23 nM.

We studied Abcc mediated-transport in middle and posterior intestine of the rainbow trout, Oncorhynchus mykiss. Luminal and serosal transport were evaluated in everted and non-everted intestinal sacs, respectively, incubated with 1-chloro-2,4-dinitrobenzene (CDNB; 200 μM). CDNB enters the cells and is conjugated with glutathione via glutathione S-transferase (GST) to form 2,4-dinitrophenyl-S-glutathione (DNP-SG), a known Abcc substrate. DNP-SG concentration in the bath was recorded every 10 min, in order to calculate the mass-specific transport rate. For evaluating the possible involvement of Abcc proteins in microcystin-LR (MCLR) transport, 1.135 μM MCLR was added to the bath or inside the sacs, in everted or non-everted preparations, respectively. Both luminal and serosal DNP-SG efflux were significantly inhibited by MCLR. A concentration-response curve obtained using strips from middle intestine yielded an IC50 value of 1.33 μM MCLR. The Abcc inhibitor, MK571 produced concentration-dependent inhibition of DNP-SG similar to that produced by MCLR. Since competition of MCLR and CDNB as GST substrates could bias the DNP-SG transport results, we evaluated the effects of MCLR on calcein efflux, which does not depend on GST activity. We applied the non-fluorescent, cell-permeant compound calcein-AM (0.25 μM) to middle intestinal strips and recorded the efflux of its hydrolysis product, the fluorescent Abcc substrate calcein. 2.27 μM MCLR and 3 μM MK571 inhibited calcein efflux (17.39 and 20.2%, respectively). Finally, MCLR interaction with Abcc transporters was evaluated by measuring its toxic intracellular effects. Middle intestinal segments were incubated in saline solution with 1.135 μM MCLR (MC1), 2.27 μM MCLR (MC2), 3 μM MK571 (MK) or 1.135 μM MCLR+3 μM MK571 (MC1/MK). After 1h, GSH concentration, protein phosphatase 1 and 2A (PP1, PP2A) and GST activities were measured in each segment. MC1did not produce significant effect while MC1/MK and MC2 significantly inhibited PP1and PP2A in similar proportions (34-49%). MK alone significantly increased PP2A activity (40%) with no effect in any other variable. GST activity and GSH concentration were not affected by any treatment. Concentration-response curves for MCLR (1.135 to 13.62 μM) alone or plus 3 or 6 μM MK571 were obtained using PP1 activity as response variable. The IC50 values were 1.0, 0.52, and 0.37 μM, respectively. Our results suggest that O. mykiss enterocytes are capable of eliminating MCLR by GST-mediated conjugation and luminal excretion through an Abcc-like apical transporter. This mechanism would prevent toxic effects and reduce the toxin uptake into the blood, which is likely mediated by basolateral Abccs.

Abstract
A summary of the resource.
We studied the accumulation and depuration of microcystin-LR (MCLR) in the hepatopancreas of the crab Neohelice granulata fed twice weekly with either non toxic or MCLR-producing Microcystis aeruginosa (strain NPDC1 or NPJB, respectively) during seven weeks. We also analyzed MCLR effects on the oxidative stress- and detoxification-related variables, superoxide dismutase and glutathione-S-transferase activities, and the levels of reduced glutathione and lipid peroxidation (as thiobarbituric acid reactive substances, TBARS).<br /><br />Hepatopancreas MCLR content slightly increased during the first three weeks, up to 8.81±1.84 ng g−1 wet tissue mass (WTM) and then started to decrease to a minimum of 1.57±0.74 ng g−1 WTM at the seventh week (p<0.05 with respect to that in the first week). TBARS levels were about 55% higher in treated than in control N. granulata (p<0.001 and p<0.05) during the first three weeks of the experimental period. GSH content became 50% lower than in control individuals (p<0.01) during weeks 6 and 7. SOD activity was increased by about 2-fold (p<0.05 or p<0.001) from week 3 to 7 in treated crabs with respect to control ones, while GST activity was about 70% higher in treated than in control crabs from week 4 to week 7 (p<0.05).<br /><br />Our data suggest that in the hepatopancreas of N. granulata MCLR accumulation and oxidative damage are limited and reversed by detoxification-excretion and antioxidant mechanisms. The activation of these defensive mechanisms becomes evident at 3-4 weeks after the start of the intoxication.<br /><br />
