A summary of the resource.
A modified and miniaturized SweEt/QuEChERS method for pesticide residue analysis applied to vine leaves is presented. The deep-frozen plant material was cryogenically processed. A 2 g analytical portion was hydrated for 30 min and extracted with acidified ethyl acetate after buffering with NaHCO3 and adding Na2SO4. A dispersive solid-phase (d-SPE) cleanup step with primary–secondary amine (PSA) was performed. The pesticide residues were determined using GC-MS/MS. The whole procedure was validated for 54–59 pesticides at 0.01, 0.02, 0.1, and 0.2 mg/kg in fresh vine leaves (Vitis vinifera). The key method performance parameters investigated were specificity, linearity, trueness, within laboratory repeatability and reproducibility, limit of detection, limit of quantitation, and matrix effects. Recoveries for the 59 pesticides tested ranged from 60 to 110%, and the RSDs were lower than 20% for the majority of the pesticides studied.

Rapanea ferruginea antioxidant and antitumoral properties were not explored before in literature. This study aimed to investigate these biological activities for the R. ferruginea leaf extract and correlate them with its phenolic content and influence in biological membrane dynamics. Thus, in this study, anti/pro-oxidative properties of R. ferruginea leaf extract by in vitro DPPH and TBARS assays, with respect to the free radical reducing potential and to its activity regarding membrane free radical-induced peroxidation, respectively. Furthermore, preliminary tests related to the extract effect on in vitro glioma cell viability were also performed. In parallel, the phenolic content was detected by HPLC-DAD and included syringic and trans-cinnamic acids, quercetrin, catechin, quercetin, and gallic acid. In an attempt to correlate the biological activity of R. ferruginea extract and its effect on membrane dynamics, the molecular interaction between the extract and a liposomal model with natural-sourced phospholipids was investigated. Location and changes in vibrational, rotational, and translational lipid motions, as well as in the phase state of liposomes, induced by R. ferruginea extract, were monitored by Fourier-transform infrared spectroscopy, nuclear magnetic resonance, differential scanning calorimetry, and UV-visible spectroscopy. In its free form, the extract showed promising in vitro antioxidant properties. Free-form extract (at 1000µ g/mL) exposure reduced glioma cell in vitro viability in 40%, as evidenced by MTT tests. Pro-oxidant behavior was observed when the extract was loaded into liposomes. A 70.8% cell viability reduction was achieved with 500 µg/mL of liposome-loaded extract. The compounds of R. ferruginea extract ordered liposome interface and disorder edits a polar region. Phenolic content, as well as membrane interaction and modulation may have an important role in the oxidative and antitumoral activities of the R. ferruginea leaf extract.

In this work, highly reactive carbon–iron composites were prepared using a waste, i.e. tar, as carbon precursor and a simple iron oxide, i.e. hematite. Tar was impregnated on Fe2O3 with different tar/hematite weight ratios of 1:1; 2:1 and 4:1, and thermally treated under N2 atmosphere (400◦C, 600◦C and 800◦C). Mössbauer, XRD andmagnetizationmeasurements suggested that treatment at 400◦C and 600◦C produces Fe3O4 but treatment at 800◦C produced mainly Fe◦. Raman and TG analyses of the different composites suggested the formation of carbon contents of 18, 24 and 32 wt.% as amorphous and graphitic highly dispersed on the Fe surface. The composites obtained at 800◦C showed high efficiency to reduce Cr(VI) as CrO2− 4 in aqueous medium with much better results compared to finely ground commercial Fe◦.

A summary of the resource.
A simulated therapeutic equivalence scenario was created between two multisource drug products (tablets marketed in Uruguay) containing a poorly water-soluble compound, flunitrazepam, and the reference product (Rohypnol®). The simulated study was built from an in vitro - in vivo dissolution-pharmacokinetic C-level (IVIV) correlation and from an in vivo pharmacokinetic-pharmacodynamic (PKPD) correlation, using experimental and bibliographic data. In vitro parameters were experimentally obtained from the dissolution profile for each commercial brand at the three biorelevant dissolution media (pH 1.2, 4.5 and 6.8) whereas in vivo parameters were acquired from the bibliography. From IVIV correlation plasma drug concentration-time profiles were simulated. These concentrations were used to simulate the intensity of drug effect throughout time in order to predict relative clinical responses of each brand. It could be concluded that from the three brands of flunitrazepam marketed in Uruguay, one was revealed as having a very poor sedative effect based on its simulated low pharmacodynamic-time profile, reinforcing hence the ineffective clinical experience claimed by physicians.

A summary of the resource.
Sixteen healthy volunteers (8 women and 8 men) participated in a 2-period, 2-treatment crossover study. A delayed-release gastroresistant formulation of ketoprofen was administered under fasting and fed conditions. Cmax, AUC, Cmax/AUC, and kel obtained after food coadministration did not differ from those calculated under fasting administration. Ninety-five percent confidence intervals for fed/fasting geometric mean ratio of Cmax/AUC and AUC were 0.80–1.14 and 0.80–1.23, respectively. A significant difference (P < .01) was found between lag-time medians (T0), with a longer T0 after food intake (5.5 vs 2.5 hours). Also, a significant difference between the medians of Tmax was found (P < .01), being 7.0 hours after food coadministration and 4.0 hours under fasting administration, but this difference disappeared once T0 was subtracted from Tmax. Cmax/AUC, which is related to drug absorption rate, showed significant differences between sexes. Men showed higher (P =.006) Cmax/AUC means (0.468 ± 0.094 vs 0.361 ± 0.087 h−1. Tmax was also significantly different (P < .05), being 4.0 (3.0–5.0) hours for men and 8.0 (5.0–10.0) hours for women. In conclusion, men showed a faster intestinal absorption rate with earlier time-to-peak plasma concentration of ketoprofen. Food coadministration extended the gastric residence time of formulation but exerted no effect on its intestinal absorption pattern.

Los medicamentos que contienen el mismo principio activo se ofrecen en el mercado farmacéutico como “similares” o como “alternativas farmacéuticas”. Dentro de los primeros se hallan los que originalmente se comercializaron como innovaciones y los que luego de un tiempo aparecieron como copias. Entre los segundos encontramos aquellos que contienen diferentes ingredientes que rinden el mismo principio activo, y aquellos que conteniendo el mismo ingrediente lo vehiculizan en diferentes formas farmacéuticas. Tanto pacientes como prescriptores se encuentran frente ofertas de similares cuyas biodisponibilidades relativas son desconocidas en nuestro país, en virtud de que la evaluación de las mismas quedó relegada por años, y aunque actualmente existe una base regulatoria [1] que intenta subsanar este rezago, el proceso de su elucidación sigue un curso demasiado lento

New dynamic combinatorial libraries (DCLs) were generated using the reversible aminothiol exchange reaction of thiazolidines and aromatic aldehydes. The reaction proceeded in aqueous buffered media at pH 4 and room temperature to generate thermodynamically controlled mixtures of heterocycles. The synthesis of an enantiomerically pure thiazolidinyloxazolidine is also reported. The oxazolidine moiety could be exchanged in CH2Cl2 in the presence of catalytic p-TsOH.

The reversibility of the thiol-thioester linkage has been broadly employed in many fields of biochemistry (lipid synthesis) and chemistry (dynamic combinatorial chemistry and material science). When the transthioesterification is followed by a S-to-N acyl transfer to give an amide bond, it is called Native Chemical Ligation (NCL), a high-yield chemoselective process used for peptide synthesis. Recently, we described thioglycolic acid (TGA) as a useful reagent for thioester deprotection both in solution and anchored to a solid-support under mild conditions. Inspired by NCL, in this work, we extended this approach and explored the use of 2-aminothiols for the deprotection of thiols bearing an acyl group. The best results were obtained using cysteamine or L-cysteine in an aqueous buffer pH 8 at room temperature for 30 min. The described approach was useful for S-acetyl, S-butyryl, and S-benzoyl heterocycles deprotection with yields up to 84%. Employing this methodology, we prepared six new analogs 2 of mercaptomethyl bisthiazolidine 1, a useful inhibitor of a wide-range of Metallo-β-Lactamases (MBLs). Compared with the previous methodologies (TGA polymer supported and TGA in solution), the biomimetic deprotection herein described presents better performance with higher yields, shorter reaction times, less timeconsuming operations, easier setup, and lower costs.

The aim of this work was to evaluate consumer visual processing of food labels when evaluating the salt content of pan bread labels and to study the influence of label design and nutritional labelling format on consumer attention. A total of 16 pan bread labels, designed according to a full factorial design, were presented to 52 participants, who were asked to decide whether the sodium content of each label was medium or low, while their eye movements were recorded using an eye tracker. Results showed that most participants looked at nutrition labels and the traffic light system to conclude on the salt content of the labels. However, the average percentage of participants who looked at the actual sodium content was much lower. Nutrition information format affected participants’ processing of nutrition information. Among other effects, the inclusion of the traffic light system increased participants’ attention towards some kind of nutrition information and facilitated its processing, but not its understanding.

Results from conjoint analysis do not allow to conclude if a certain variable is not relevant for consumers or if it did not catch their attention. The incorporation of eye-tracking to conjoint analysis seems to be a proper way to overcome this limitation. The aim of the present work was to evaluate the contribution of eye-tracking techniques to the interpretation of results from conjoint analysis using a case study on consumers’ healthfulness perception of yogurt labels. Sixteen yogurt labels (designed according to a full factorial design with four 2-level factors – label background, fat content, sugar content and traffic light system –) were presented to 51 consumers, who were asked to rate their healthfulness. Participants’ eye movements when evaluating the labels were recorded using an eye tracker. Fat content was the only variable that significantly affected perceived healthfulness. Eye tracking provided an insight on how consumers visually processed the labels and allowed to conclude that consumers visually processed all the information responsible for differences among labels.

The cestode Echinococcus granulosus, the agent of hydatidosis/echinococcosis, is remarkably well adapted to its definitive host. However, the molecular mechanisms underlying the successful establishment of larval worms (protoscoleces) in the dog duodenum are unknown. With the aim of identifying molecules participating in the E. granulosus-dog cross-talk, we surveyed the transcriptomes of protoscoleces and protoscoleces treated with pepsin at pH 2. This analysis identified a multigene family of secreted monodomain Kunitz proteins associated mostly with pepsin/H+-treated worms, suggesting that they play a role at the onset of infection. We present the relevant molecular features of eight members of the E. granulosus Kunitz family (EgKU-1 – EgKU-8). Although diverse, the family includes three pairs of close paralogs (EgKU-1/ EgKU-4; EgKU-3/EgKU-8; EgKU-6/EgKU-7), which would be the products of recent gene duplications. In addition, we describe the purification of EgKU-1 and EgKU-8 from larval worms, and provide data indicating that some members of the family (notably, EgKU-3 and EgKU-8) are secreted by protoscoleces. Detailed kinetic studies with native EgKU-1 and EgKU-8 highlighted their functional diversity. Like most monodomain Kunitz proteins, EgKU-8 behaved as a slow, tight-binding inhibitor of serine proteases, with global inhibition constants (KI *) versus trypsins in the picomolar range. In sharp contrast, EgKU-1 did not inhibit any of the assayed peptidases. Interestingly, molecular modeling revealed structural elements associated with activity in Kunitz cation-channel blockers. We propose that this family of inhibitors has the potential to act at the E. granulosus-dog interface and interfere with host physiological processes at the initial stages of infection

The effectiveness was evaluated for an established wood preservative, zinc borate, prepared in an agitating bead mill to extensively reduce its particle size. The generated micro- or nano-particles of zinc borate were characterized by X-ray powder diffraction, and the particle size distribution was determined to evaluate the effect of milling. Then the fungicidal effectiveness of the zinc borate of both milled and unmilled samples were assayed against brown- and white-rot fungi, on culture medium and on conifer and hardwood as substrates. Treated wood samples were subjected to leaching tests. Scanning electron microscopic images of wood samples were examined to analyze the distribution of zinc borate within the wood. The micronized zinc product kept its crystal structure intact, and it increased the proportion of particles with diameters below 100 nm by 25% when compared to the unmilled product. Malt extract-agar medium supplemented with 2.5% of w/w milled and unmilled zinc borate inhibited fungal growth tested. Both milled and unmilled zinc borate protected the wood when not subjected to leaching. The milled sample of zinc borate improved resistance to leaching, which would allow its application in environments of high moisture content; however, it did not improve the fungicidal action against decay fungi.

There is scarce information on the vibrational and thermal properties of small Ni clusters. For theses clusters a comparison is presented between the closed shell symmetric octahedral (Oh) and the icosahedral (Ih) geometries. The results indicate that the Ih isomers are lower in energy. Besides, ab initio molecular dynamics runs at T = 300K show that Ni13 and Ni55 clusters transform from their initial Oh geometries towards the corresponding Ih ones. For Ni13, we also consider the lowest energy less symmetric layered 1-3-6-3 structure, and the cuboid, recently observed experimentally for Pt13, which is competitive in energy but is unstable, as phonon analysis reveals. We calculate their vibrational density of states (νDOS) and heat capacity, and compare with the Ni FCC bulk counterpart. The characteristic features of the νDOS curves of these clusters are interpreted in terms of the clusters’ sizes, the interatomic distance contractions, the bond order values as well as the internal pressure and strains of the clusters. We find that the softest possible frequency of the clusters is size and structure-dependent, being the smallest for the Oh ones. We identify mostly shear, tangential type displacements involving mainly surface atoms for the lowest frequency of the spectra of both Ih and Oh isomers. For the maximum frequencies of these clusters the central atom shows anti-phase movements against groups of nearest neighbor atoms. An excess of heat capacity at low temperatures with respect to the bulk is found, while at high temperatures a constant limiting value, close but lower to the Dulong and Petit value, is determined.

Background: The sudden changes (increase in capillar permeability, edema formation, vasodilation and hypotension) observed in septic patients and the measurements taken in order to revert this situation make vancomycin concentrations difficult to interpret. Therapeutic drug monitoring (TDM) of vancomycin is routinely performed at steady state, target concentrations are peaks between 20 and 40 mg/l and troughs of 5 – 10 mg/l. Lately, continuous infusion of vancomycin (CIV) has been used as an alternative mode of administration mainly in critically ill patients with sepsis or septic shock. Despite this novel mode of administration, the need of drug monitoring in this population is under discussion. Objective: The aim of our study was to test the usefulness of a multi-compartment model in order to understand the rapid changes that occur in critically ill patients. Materials and methods: A prospective, cohort study was carried out in the intensive care unit of the University Hospital. 25 intensive care unit adults patients with severe sepsis or septic shock receiving vancomycin in CIV modality for documented gram-positive infections were included in the study. Once the infusion was started, blood samples were drawn periodically and analized by fluorescence polarisation immunoassay (FPIA, TDx, Abbott Laboratories, Chicago, IL, USA). A multi-compartment model was used to predict vancomycin level evolution throughout the treatment of patients with sepsis. Results: High doses of vancomycin were administered in order to rescue patients from septic shock. Plasma drug concentration dropped while clinical condition of patients worsened. Conversely, drug levels increased spontaneously once the infection was reverted. The theoretical model provided greater insight into pharmacokinetic features related with the use of vancomycin in septic patients. Conclusions: There was consistency between the model based prediction and the experimental data so dose adjustment was performed in order to reach target concentrations above 20 mg/l and an initial dose of 3 grams of vancomycin per day was recommended to reach these levels.

<p><strong class="sub-title"> Background: </strong> Current therapy of neurological disorders has several limitations. Although a high number of drugs are clinically available, several subjects do not achieve full symptomatic remission. In recent years, there has been an increasing interest in the therapeutic potential of L-carnitine (LCAR) and acetyl-L-carnitine (ALCAR) because of the multiplicity of actions they exert in energy metabolism, as antioxidants, neuromodulators and neuroprotectors. They also show excellent safety and tolerability profile.</p>
<p><strong class="sub-title"> Objective: </strong> To assess the role of LCAR and ALCAR in neurological disorders.</p>
<p><strong class="sub-title"> Methods: </strong> A meticulous review of the literature was conducted in order to establish the linkage between LCAR and ALCAR and neurological diseases.</p>
<p><strong class="sub-title"> Results: </strong> LCAR and ALCAR mechanisms and effects were studied for Alzheimer's disease, depression, neuropathic pain, bipolar disorder, Parkinson's disease and epilepsy in the elderly. Both substances exert their actions mainly on primary metabolism, enhancing energy production, through β-oxidation, and the ammonia elimination via urea cycle promotion. These systemic actions impact positively on the Central Nervous System state, as Ammonia and energy depletion seem to underlie most of the neurotoxic events, such as inflammation, oxidative stress, membrane degeneration, and neurotransmitters disbalances, present in neurological disorders, mainly in the elderly. The impact on bipolar disorder is controversial. LCAR absorption seems to be impaired in the elderly due to the decrease of active transportation; therefore, ALCAR seems to be the more effective option to administer.</p>
<p><strong class="sub-title"> Conclusion: </strong> ALCAR emerges as a simple, economical and safe adjuvant option in order to impair the progression of most neurological disorders.</p>
