En la actualidad, el consumo de vegetales en diversas formas y con mínimo procesamiento, es una práctica cada vez más extendida por la población, pues son reconocidos como fuente de principios activos benéficos para la salud. En general, son cultivados empleando prácticas agrícolas diversas, convencionales, integradas u orgánicas, con un claro predominio de la forma de cultivo convencional, donde el uso de agroquímicos es común. Considerando además la capacidad de estos vegetales de absorber metales y otros elementos del suelo donde se cultivan, algunos de ellos esenciales para la vida y otros potencialmente tóxicos, es que el interés en conocer los niveles de estos elementos se vuelve cada vez mayor. El desafío de este trabajo de posgrado fue obtener una visión global de los aportes tanto benéficos como negativos para la salud que el consumo de estos vegetales podía tener, mediante un estudio de la composición química de los nutracéuticos, así como de los contaminantes orgánicos e inorgánicos presentes en una planta con reconocidas propiedades benéficas como lo es la alcachofa. El fruto de la misma es un alimento muy valorado debido a sus propiedades nutricionales. Su elevada humedad, bajo contenido calórico, la práctica ausencia de grasas y su naturaleza fibrosa la han convertido en uno de los alimentos dietéticos por excelencia. A esto se suma los efectos antioxidantes, coleréticos y adelgazantes reportados en la literatura. Se estudiaron las propiedades de los frutos y de las hojas de alcachofa, así como las propiedades de las correspondientes infusiones de las hojas, debido a su amplio consumo bajo forma de té y a su empleo en productos fitoterápicos. Para lograr esta evaluación integrada, se estudió el contenido de elementos con valor nutricional como ser algunos metales esenciales (Cu, Fe, Ni y Zn) y algunos metabolitos secundarios como los polifenoles de reconocida capacidad antioxidante. También se estudió el contenido de aquellos elementos que podían representar un riesgo para la salud, como ser As, Cd y Pb, y los residuos de algunos agroquímicos comúnmente utilizados para proteger los cultivos y asegurar rendimiento y calidad en la producción. Glycerol is a side product of biodiesel production which is available in the world market at relatively low cost and could be used to obtain other valuable products. H2 and H2-CO mixtures can be obtained from glycerol by steam reforming using Ni based catalysts. NiLaZr catalysts prepared by a coprecipitation technique were tested in the steam reforming of pure glycerol. The tests were performed at 500 and 650 °C and at glycerol:water molar ratios 1:6 and 1:12. Glycerol conversion to gas phase products and H2 yields were greatly improved with the increase of Ni loading from 5 to 15 %. The addition of 0.5 % of Rh is also very effective at low temperature to attain almost total glycerol conversions and high H2 yields. The catalyst containing 15 % of Ni and the NiO and La2Zr2O7 phases showed the best catalytic performance with glycerol conversion above 99 % and a H2 yield close to the thermodynamic value at 650 °C. This catalyst also showed the best results in term of stability and low carbon formation. Fast deactivation of the Rh impregnated catalyst is ascribed to its ability to yield carbon by dissociation of C–C and C–H bonds in glycerol and other C1–C3 intermediates. Carbon morphology is greatly influenced by Ni interactions with the rest of the catalyst structure. A common approach to deal with this phenomenon during astringency evaluation is the use of palate cleansers. The aim of this work was to evaluate palate cleansers for red wine astringency evaluation, considering both reduction of astringency buildup and sample discrimination. Fourteen trained panelists evaluated two sets of four Tannat wine samples using time-intensity methodology. In the first sample set the same wine was presented four times, while the second one comprised four samples with different added concentrations of added grape seed tannins. Each assessor evaluated both sample sets with five different palate cleansers (still mineral water, plain unsalted crackers, skimmed milk, drinkable plain sweetened yogurt and 2 g/L pectin solution) in triplicate. Astringency is one of the most important sensory characteristics of red wine. It involves several mouth-feel sensations, which have been commonly used to describe red wines. However, the dynamics of astringency sensations have not been previously studied. In this context, the aim of the present work was to obtain a dynamic description of the astringency of red wines. Seven commercial Uruguayan Tannat wines were evaluated in triplicate by a panel of 9 trained assessors. They were asked to describe the astringency of the wines during 40 s in a Temporal Dominance of Sensations (TDS) task comprising a list of 8 terms: ‘dry’, ‘fine emery’, ‘harsh’, ‘mouthcoating’, ‘puckery’, ‘rough’, ‘silky’, and ‘velvety’. After completing the TDS task they were asked to rate global astringency intensity using an unstructured scale. The wines significantly differed in their average global astringency intensity. Between two and three terms were significantly dominant to describe the astringency of each of the seven wines and enabled to discriminate samples with different astringency characteristics. Samples differed in the dominance of the terms and the time elapsed until they became dominant. Wines which did not significantly differ in their average astringency rating showed different dynamic astringency profiles, which evidenced that the dynamics of astringency characteristics were not related to global astringency intensity. TDS seems to be an interesting methodological choice to characterize the dynamics of wine astringency and opens new possibilities to better understand this complex sensory characteristic. In projective mapping tasks assessors create an overall representation of the similarities and differences among samples by relying on a process of synthesis for analyzing and processing sensory information. Individual differences in consumers’ information processing and preference patterns could strongly affect which sensory characteristics they consider more relevant for estimating similarities and differences among samples. Therefore, low-dimensional consensus configurations (obtained via MFA or GPA) may not represent the perception of some consumer segments. This could lead to inaccurate conclusions about consumers’ sensory perception of the products or at least to the loss of valuable information about the perception of some consumer groups. In this context, the aims of the present work were to explore consumer segmentation in projective mapping. Datasets from nine studies with 81–102 consumers were analyzed to explore consumers’ segmentation. Through applying hierarchical cluster analysis on consumers’ coordinates in the first four dimensions of the MFA, between 2 and 4 groups of consumers were identified in each study. Sample configurations and consumers’ descriptions strongly differed among the groups, indicating heterogeneity in the relative relevance they gave to the sensory characteristics of the samples for estimating the similarities and differences among samples. In all cases it was observed that the consensus configuration was highly similar to the configuration of one of the groups, which was not necessarily the larger but the one with the highest explained variance by the first dimension of the MFA. These results suggest the need to explore segmentation when analyzing data from projective mapping tasks, and to further study the relationship between consumers’ holistic perception of products and preference patterns. Emotional responses to food and beverages has been established as a significant research topic within sensory and consumer science. The current research contributes to this activity by building new insights regarding consumers’ spontaneous expressions of food-related emotional experiences. This was done by analysing 12,260 tweets about breakfast, lunch, snack and dinner eating situations, previously retrieved by Vidal et al. (2015). A descriptive approach was adopted, wherein focus was direct to capturing frequency and diversity of emoticon and emoji use. It was found that consumers express a wide range of positive and negative emotions and that emoticon and emoji use is tailored to the content of the tweets. Emoji were used more frequently than emoticons to express emotions. While it was rare for tweets to include more than one emoticon or emoji, their use was almost exclusively in addition to other content of the tweet. Our results suggest that emoji and emoticon seem to be an easy and intuitive way to express emotions in a food context. This could represent an opportunity for development of non-verbal subjective methods to measure food-related emotions. The composition of fatty acids and triglycerides in the fat of Uruguayan goat cheeses was determined. Additionally, in the present study, triglyceride composition was determined in terms of partition number (PN) for fats from pure goat milk, pure cow milk and blends thereof (90:10, 80:20, and 50:50 blends of goat milk:cow milk). All Uruguayan goat cheeses showed high levels of conjugated linoleic acid and trans-vaccenic acid, well above those reported in cheeses from other countries. This is advantage in terms of nutritional quality. In fats from goat milk in Uruguayan cheeses, the highest average percentages of triglycerides correspond to PN = 36 (13.2%) and PN = 42 (13.6%). The results demonstrate that the triglyceride profile changes when cow milk is added to goat milk. When the amount of cow milk added to goat milk increases, triglycerides with PN values of 38, 40, and 42 tend to decrease, while triglycerides with PN values of 46, 48, and 50 tend to increase. These results suggest that the triglyceride profile could be used to determine whether a given goat cheese was adulterated with cow milk, which is cheaper. This was detected in the case of three Uruguayan goat cheeses that were analysed. Quillaja brasiliensis (Quillajaceae) is a saponin producing species native from southern Brazil and Uruguay. Its saponins are remarkably similar to those of Q. saponaria, which provides most of the saponins used as immunoadjuvants in vaccines. The immunostimulating capacities of aqueous extract (AE) and purified saponin fraction (QB-90) obtained from leaves of Q. brasiliensis were favorably comparable to those of a commercial saponin-based adjuvant preparation (Quil-A®) in experimental vaccines against bovine herpesvirus type 1 and 5, poliovirus and bovine viral diarrhea virus in mice model. Herein, the immunogenicity and protection efficacy of rabies vaccines adjuvanted with Q. brasiliensis AE and its saponin fractions were compared with vaccines adjuvanted with either commercial Quil-A or Alum. Mice were vaccinated with one or two doses (on days 0 and 14) of one of the different vaccines and serum levels of total IgG, IgG1 and IgG2a were quantified over time. A challenge experiment with a lethal dose of rabies virus was carried out with the formulations. Viral RNA detection in the brain of mice was performed by qPCR, and RNA copy-numbers were quantified using a standard curve of in vitro transcribed RNA. All Q. brasiliensis saponin-adjuvanted vaccines significantly enhanced levels of specific IgG isotypes when compared with the no adjuvant group (P ≤ 0.05). Overall, one or two doses of saponin-based vaccine were efficient to protect against the lethal rabies exposure. This stabilization of the Kubas-type complex could play a very important role for hydrogen storage in Pt-decorated carbon adsorbents with vacancies. The liver fluke Fasciola hepatica remains an important agent of food-borne trematode disease producing great economic losses due to its negative effect on productivity of livestock grazing in temperate areas. The prevailing control strategy based on anthelmintic drugs is unsustainable due to widespread resistance hence vaccination appears as an attractive option to pursue. In this study we evaluate the effect of vaccination in calves with a functional recombinant thioredoxin glutathione reductase (rFhTGR) from liver fluke, a critical antioxidant enzyme at the crossroads of the thioredoxin and glutathione metabolism in flatworms. The recombinant enzyme produced in Escherichia coli was tested in two vaccination experiments; in the first trial rFhTGR was administered in combination with Freund́s Incomplete Adjuvant (FIA) in a three-inoculation scheme on weeks 0, 4 and 8; in the second trial rFhTGR was given mixed with Adyuvac 50 or Alum as adjuvants on weeks 0 and 4. In both cases calves were challenged with metacercariae (400 in the first and 500 in the second trial) 2 weeks after the last inoculation. Our results demonstrate that two or three doses of the vaccine induced a non-significant reduction in worm counts of 8.2% (FIA), 3.8% (Adyuvac 50) and 23.0% (Alum) compared to adjuvant controls indicating that rFhTGR failed to induce a protective immunity in challenged calves. All vaccine formulations induced a mixed IgG1/IgG2 response but no booster was observed after challenge. Benzyl alcohol and other benzenoid-derived metabolites of particular importance in plants confer floral and fruity flavors to wines. Among the volatile aroma components in Vitis vinifera grape varieties, benzyl alcohol is present in its free and glycosylated forms. These compounds are considered to originate from grapes only and not from fermentative processes. We have found increased levels of benzyl alcohol in red Tannat wine compared to that in grape juice, suggesting de novo formation of this metabolite during vinification. In this work, we show that benzyl alcohol, benzaldehyde, p-hydroxybenzaldehyde, and p-hydroxybenzyl alcohol are synthesized de novo in the absence of grape-derived precursors by Hanseniaspora vineae. Levels of benzyl alcohol produced by 11 different H. vineae strains were 20–200 times higher than those measured in fermentations with Saccharomyces cerevisiae strains. These results show that H. vineae contributes to flavor diversity by increasing grape variety aroma concentration in a chemically defined medium. Feeding experiments with phenylalanine, tryptophan, tyrosine, p-aminobenzoic acid, and ammonium in an artificial medium were tested to evaluate the effect of these compounds either as precursors or as potential pathway regulators for the formation of benzenoid-derived aromas. Genomic analysis shows that the phenylalanine ammonia-lyase (PAL) and tyrosine ammonia lyase (TAL) pathways, used by plants to generate benzyl alcohols from aromatic amino acids, are absent in the H. vineae genome. Consequently, alternative pathways derived from chorismate with mandelate as an intermediate are discussed. The key transformations consist of an enzymatic dioxygenation using the toluene dioxygenase enzymatic complex, followed by an allylic azide double sigmatropic [3,3] shift to introduce the nitrogen functionality in the ring in a stereoselective manner. Azide reduction and further regioselective oxidation of the diene moiety afforded the desired inosamine in only eight steps from benzyl azide. Huntington's disease (HD) is a hereditary neurodegenerative disorder. The genetic cause is an expansion of CAG repeats located in the IT15 gene. Though the number of CAG repeats ((CAG)n) can largely explain the age at onset (AAO) of symptoms, a percentage of its variation could be attributed to modifier genes and to environmental factors. The study aimed to evaluate the influence of genetic modifiers of the AAO of HD including: (CAG)n and del2642 in the IT15 gene, ADORA2A rs5751876, HAP1 rs4523977, PGC1-α rs7665116 and UCH-L1 rs5030732. Eighteen patients with positive family history and HD-suggestive symptoms were recruited. The (CAG)n and gene polymorphisms were determined by different molecular biology techniques. We observed that the (CAG)n influenced in a 64.5% of the variability in the AAO. We also showed that the rs5751876 variant significantly affected this variability. However, the influence of UCH-L1, del2642, HAP1 and PGC1-α gene polymorphisms could not be replicated, perhaps due to small sample size. Genetic studies including the molecular determination of (CAG)n, in addition to other genetic modifiers involved in the variability of the AAO, were first performed in Uruguay. We could replicate in our cohort the anticipation effect on the AAO by the ADORA2A rs5751876. Our results confirm the usefulness of an expanded molecular characterization in HD patients.
