The objective of the current study was to examine the safe use of a daily standard dose of omeprazole (20 mg) regarding magnesium levels in blood in hospitalized patients. A total of 51 patients (15 women, 36 men) with different characteristics (pathologies, comedications, age, habits, etc) and taking a standard dose of omeprazole for more than three months were included. 17.6 % of the patients showed mild hypomagnesemia but we concluded that the observed low levels of this electrolyte could be attributed to comedications, age and different pathologies rather than the 20-mg dose of omeprazole. So, hypomagnesemia does not eliminate proton-pump inhibitors as a reasonable option; it just requires clinicians to be aware of this problem and use them safely at conventional doses. We studied the filling state and reloading rate of the Brindley's glands in the haematophagous Chagas disease vector Triatoma infestans (Hemiptera: Reduviidae). Quantitative analyses of isobutyric acid, the main secretion component, were carried out with glands dissected from adults under different scenarios of development, number of discharging events and feeding conditions. The alarm-pheromone function of the gland secretion was also assessed in bioassays with conspecific nymphs. Although pharate adults have their glands completely developed, these were not full until imaginal ecdysis. If kept undisturbed, the adults maintained a constant gland load, and discharged about 75% of the gland contents upon one disturbance event. While the glands can be discharged several times, full replenishing was not complete after one week, unless the insect had access to food. The escape behavior of nymphs in bioassays correlated with the chemical analyses, with nymphs showing significant avoidance only toward gland discharges from undisturbed or disturbed/fed adults. Ins(1,2,3)P3 is probably an important safe chelator of iron cations not strongly bound to proteins. In order to clarify its biological functions, Ins(1,2,3)P3 chemistry under physiological conditions must be completely elucidated. The protonation and complexation behaviour of Ins(1,2,3)P3 has been recently studied under these conditions by potentiometry. Under simulated physiological conditions it forms the protonated species H2L4� and H3L3�. The presence of high concentrations of potassium in intracellular compartments causes the formation of two predominant Ins(1,2,3)P3 complexes: [K(HL)]4� and [K(H2L)]3�, in the absence of iron. In this work we expand part of this macroscopic knowledge to the inframolecular level, by 31P NMR measurements and focusing on the protonation and complexation of this biologically relevant molecule to potassium. We complete this study with theoretical calculations which lead us to predict the geometries of every form of the ligand and their relative stabilities. The influence of the ring conformation in protonated and complexed forms is also discussed. Significance: Major pathogenic enterobacteria and protozoan parasites from the phylum Euglenozoa, such as trypanosomatids, are endowed with glutathione (GSH)-spermidine (Sp) derivatives that play important roles in signaling and metal and thiol-redox homeostasis. For some Euglenozoa lineages, the GSH-Sp conjugates represent the main redox cosubstrates around which entire new redox systems have evolved. Several proteins underwent molecular adaptations to synthesize and utilize the new polyamine-based thiols. Recent Advances: The genomes of closely related organisms have recently been sequenced, which allows mining and analysis of gene sequences that belong to these peculiar redox systems. Similarly, the three-dimensional structures of several of these proteins have been solved, which allows for comparison with their counterparts in classical redox systems that rely on GSH/glutaredoxin and thioredoxin. Critical Issues: The evolutionary and structural aspects related to the emergence and use of GSH-Sp conjugates in Euglenozoa are reviewed focusing on unique structural specializations that proteins developed to use N1,N8-bisglutathionylspermidine (trypanothione) as redox cosubstrate. An updated overview on the biochemical and biological significance of the major enzymatic activities is also provided. Future Directions: A thiol-redox system strictly dependent on trypanothione is a feature unique to trypanosomatids. The physicochemical properties of the polyamine-GSH conjugates were a major driving force for structural adaptation of proteins that use these thiols as ligand and redox cofactor. In fact, the structural differences of indispensable components of this system can be exploited toward selective drug development. With the aim to improve the performance of enzyme bound to hydrophilic solid phases, their immobilization with polyethylene glycol (PEG) tether have been studied. Sweet potato �-amylase, which hydrolyses the high molecular weight substrate starch and �-galactosidase, which acts on low molecular weight substrates, were used as model enzymes and beaded thiol&ndash;agarose as solid phase. Several two step methods for the introduction of the tether using a bis-oxirane homobifunctional PEG as well as a heterobifunctional derivative with a hydroxysuccinimide ester and a maleimide group have been evaluated. Amino groups, native and de novo thiol groups in the enzymes were utilized for immobilization. The best approach was found to be to first introduce the PEG derivative via one of its reactive groups to the enzyme. Subsequently the formed conjugate was bound to the solid phase by the remaining reactive group. Attempts to first introduce the PEG tether into the solid phase were not successful. Ahigh degree of substitution with PEG chains on the enzyme leads to high immobilization yields for both �-amylase and �-galactosidase, but relatively lower gel-bound activity for the former enzyme which is acting on a high molecular weight substrate and thus more sensitive for steric shielding effects. With optimal degree of PEG substitution (which occurred at five times molar excess of the heterobifunctional reagent) the gel-bound activity of �-amylase was increased from 12% (for the derivative without tether) to 31%. Objective: Systemic inflammatory response syndrome is characterized by increased urinary excretion of selenium and low serum concentration. Repletion by parenteral selenite is the most efficacious form of supplementation. However, the optimum safe dose and mode of administration remain controversial. We aimed to determine pharmacokinetic and pharmacodynamic profiles of selenite and estimate a safe dose to optimize selenium status. Methods: A prospective, randomized, pilot study in 20 patients with systemic inflammatory response syndrome compared a high-dose (HD) group that received a loading dose of selenium as selenite 15.18 mmol over 2 h and thereafter 10.12 mmol/d as a continuous intravenous infusion (CIV) for 10 d with a very&ndash;high-dose (VHD) group that received a loading dose of 25.30 mmol over 2 h and thereafter 20.24 mmol as a CIV for 10 d. Clinical outcome was evaluated by length of stay in the intensive care unit, incidence of ventilator-associated pneumonia, and Sequential Organ Failure Assessment score. Results: Patients in group HD (n &frac14; 10, age 54 � 23 y) had an Acute Physiology and Chronic Health Evaluation II score of 23 � 5 and a Sequential Organ Function Assessment score of 10 � 2. Those in group VHD (n &frac14; 10, age 41 � 19 y) had scores of 21 � 7 and 8 � 3, respectively. Pharmacokinetic concentration/time curves for serum selenium overlapped but were independent of dose, whereas the pharmacodynamics were different, showing maximum glutathione peroxidase activity only with VHD. Glutathione peroxidase decreased after day 7 independently of the selenium dose. Clinical outcomes were similar in both groups. In the present work the performance of global projective mapping and partial projective mapping based on texture and flavor for sensory characterization of a novel food category (satiating food product)was compared. Eight different fresh-cheese pie formulationswere designed to obtain different sensory (principally texture) characteristics which could affect expected satiating perception. Three groups of consumers evaluated the samples using one of the following methodologies: global projective mapping (G-PM) (n = 47), partial projective mapping based on flavor (F-PM) (n=53), and partial projective mapping based on texture (T-PM) (n=61). In addition, the expected satiating capacity of each cheese pie sample was scored on a nine-point scale. Results showed that the vocabulary used by consumers for describing the sensory characteristics of samples did not largely differ between global and partial projective mapping tasks. However, T-PM and F-PM tasks provided more detailed information than G-PM in each specific modality. Results suggested that when consumers performed the global projective mapping task they mainly took into account flavor characteristics for evaluating global similarities and differences among samples. In addition, hedonic attributes were more frequently mentioned in G-PM. Fresh cheese, sugar, corn starch and egg were the basic ingredients that conferred the fresh-cheese pies their typical, characteristic flavor and texture. The addition ofwhey or soy proteins,wheat bran or glucomannan increased expected satiating capacity which could be related to changes in texture (harder, more compact) leading to longer orosensory exposure. The addition of glucomannan caused the largest changes in the sensory characteristics of the cheese pies which in turn would be related to a decrease in perceived flavor intensity. Studying the interplay between formulations, sensory characteristics, expected satiating capacity and consumer liking could largely contribute to the development of this novel food category. DDRs are uniquely positioned to function as sensors for extracellular matrix (ECM) and to regulate a wide-range of cell functions from migration and proliferation to cytokine secretion and ECM homeostasis/remodeling. While activation of DDRs by ECM collagens is required for normal development and tissue homeostasis, aberrant activation and function of these receptors following injury or in disease is detrimental. Both DDRs are indicated to play key roles in development and metastasis of various types of cancer. In spite of this, the mechanisms whereby DDRs contribute to tumor formation and cancer progression are poorly understood. Among reproductive system cancers, epithelial ovarian cancer (EOC) and prostate cancer (PCa) result in high rates of morbidity and mortality. In EOC and PCa, atypical expression of DDRs are indicated to function in malignancy and metastasis. The molecular mechanisms underlying how DDRs contribute to these and other pathologies need to be understood to find new markers and for development of therapeutic agents for treatment of disease. This is particularly the case for EOC in which mechanisms explaining the atypically high levels of DDR1 at initial and late stages of disease have not been described. We first outline studies showing an essential role for DDR2 in steroidogenesis and gamete development in ovary and testes. We then focus on what is known on the role of DDR1 and DDR2 in PCa and DDR1 in EOC. Finally, we speculate on possible functions DDR1 could be playing in different stages of EOC disease. Interactions between ECM proteins and cell surface receptors are well-known to play key roles in regulation of cell behavior and determining physiological function. The switch from DDR2 expression in healthy ovaries to that of DDR1 in initial and late stages of EOC disease provides an experimental system to investigate to what extent ECM and DDR signaling enables malignant transformation. Supercapacitors have been studied due to their promising as powerful storage devices. The development of highly efficient electrodes is one way to manufacture supercapacitors with high energy and power efficiencies. Binary composites based on activated carbon fiber felt (ACFF) and polypyrrole (PPy) have been investigated to increase capacitance performance. ACF, when applied as a bulk electrode has limited electric characteristics while the PPy suffers from limited stability during cycling that reduces the initial performance. Based on the synergism effect, this work proposes the production of a binary composite based on ACFF produced from a low-cost precursor textile polyacrylonitrile (textile PAN) and the facile synthesis of PPy from the chemical route of polymerization. The binary composite formed by a 3D network with the interconnection of a more conductive form of PPy coated on ACFF was obtained. The resulting architecture was determinant to significantly increase the energy storage capacities when compared to the bulk ACFF electrode. Supercapacitors have received great attention in science and technology, due to its important qualities as the high cycle life, high power density, and variety of applications. The main challenge for the development of better devices is the electrode material. Developing electrode materials with high specific electrical capacitance and low electrical resistance enables an increase of energy accumulated in the supercapacitor. In addition, it is expected that the electrode material presents a simple procedure for preparation with low production cost and be friendly to the environment. In this work, a cheap and simple electroless process was involved to deposit chemical species of iron on a no expensive textile activated carbon fiber felt with interesting properties for application as supercapacitor electrode. The resulting binary composite (Fe@ACF) supercapacitor electrode possesses a high specific capacitance of 237 F g&minus;1 at a current density of 0.2 A g&minus;1 and only 3% of loss of its initial specific capacitance after 3000 charge-discharge cycles. These results reveal that the activated carbon fiber felt/iron composites are promising materials for supercapacitor electrode applications.
