In the search for acetylcholinesterase inhibitors as a potential target for the discovery of anthelmintic drugs, a series of 27 pyridinic and pyrazinic compounds have been designed on the basis of molecular hybridization of two known AChE inhibitors, namely, tacrine and (-)-3-O-acetylspectaline, and on the concept of isosterism. The synthesized compounds generally presented moderate anticholinesterasic activities when compared with the positive control physostigmine, but one compound (ethyl 2-[(6-chloropyrazin-2-yl)sulfanyl] acetate, 11) exhibited an in vitro ability to immobilize the root-knot nematode Meloidogyne incognita that was highly comparable to that of the positive control Temik. Moreover, in anthelmintic assays against the gastrointestinal parasitic nematode Nippostrongy-lus brasiliensis (L4), some of the compounds, such as (6-chloropyrazin-2-yl)sulfanyl ethanol (32, EC50 = 33 nM), presented activities that were considerably stronger than that of the positive control albendazole (EC50 = 340 nM). In the light of the positive results obtained in the anthelmintic evaluations, the acute oral toxicity of the representative compound diethyl 2,2'-[(3-nitropyridine-2,6-diyl) bissulfanediyl] diacetate (7) was determined in rats, and the drug was shown to be non-toxic at a dose of 2000 mg/kg. These results, allied with the relatively simple structures of the active compounds and their facile synthesis, highlight their potential use as anthelmintic or nematicidic agents. In several grape varieties, the dominating aryl alkyl alcohols found are the volatile group of phenylpropanoid-related compounds, such as glycosylated benzyl and 2-phenylethyl alcohol, which contribute to wine with floral and fruity aromas after being hydrolysed during fermentation. Saccharomyces cerevisiae is largely recognized as the main agent in grape must fermentation, but yeast strains belonging to other genera, including Hanseniaspora, are known to predominate during the first stages of alcoholic fermentation. Although non-Saccharomyces yeast strains have a well-recognized genetic diversity, understanding of their impact on wine flavour richness is still emerging. In this study, 11 Hansenisapora vineae strains were used to ferment a chemically defined simil-grape fermentation medium, resembling the nutrient composition of grape juice but devoid of grape-derived secondary metabolites. GC–MS analysis was performed to determine volatile compounds in the produced wines. Our results showed that benzyl alcohol, benzyl acetate and 2-phenylethyl acetate are significantly synthesized by H. vineae strains. Levels of these compounds found in fermentations with 11 H. vineae different strains were one or two orders of magnitude higher than those measured in fermentations with a known S. cerevisiae wine strain. The implications for winemaking in response to the negative correlation of benzyl alcohol, benzyl acetate and 2-phenylethyl acetate production with yeast assimilable nitrogen concentrations are discussed. Apiculate yeasts of the genus Hanseniaspora/Kloeckera are the main species present on mature grapes and play a significant role at the beginning of fermentation, producing enzymes and aroma compounds that expand the diversity of wine color and flavor. Ten species of the genus Hanseniaspora have been recovered from grapes and are associated in two groups: H. valbyensis, H. guilliermondii, H. uvarum, H. opuntiae, H. thailandica, H. meyeri, and H. clermontiae; and H. vineae, H. osmophila, and H. occidentalis. This review focuses on the application of some strains belonging to this genus in co-fermentation with Saccharomyces cerevisiae that demonstrates their positive contribution to winemaking. Some consistent results have shown more intense flavors and complex, full-bodied wines, compared with wines produced by the use of S. cerevisiae alone. Additionally, HHP reduced native aerobic microbiota to <1 log CFU/g and slightly affected a* and b* color values. Montecarlo simulation was used to estimate potential initial counts of STEC that allow compliance with existing regulatory limits after applying HHP, showing that absence of STEC studied can be achieved in 65 g of patties provided initial loads are ~2 log CFU/g. Finally, HHP treatments at 600 MPa and mild temperatures can be considered a valid non-thermal processing technology to achieve 5 log CFU/g reductions. The harmful bloom-forming cyanobacterium Cylindrospermopsis raciborskii grows in freshwaters over a wide range of light conditions. This species has increased its global distribution recently. The influence of ultraviolet radiation (UVR) on the fitness and toxin production of C. raciborskii has not previously been explored. We performed short-term experiments with three C. raciborskii strains (MVCC19, LB2897, and CYP011 K), and we compared their responses with other bloom-forming species (Microcystis sp.1 and Plankthotrix agardhii) to determine the impact of UV-B radiation on pigments, biomass, and morphological traits. In addition, we analyzed the effect of UV-B on the saxitoxin content and sxtU gene expression in the strain MVCC19. C. raciborskii strains were stressed differentially by UV-B exposure as evidenced by changes in growth, morphology, and heterocytes number. A significant increase in saxitoxin concentration and sxtU gene expression under UV-B suggests that toxin production in C. raciborskii can be a response to UV-B stress. In comparison, Microcystis sp.1 was more tolerant, while P. agardhii was severely impacted by UV-B, indicating also different sensitivities among cyanobacteria to UVR. Our results underscore the influence of UVR on C. raciborskii and the differences between strains which showed phenotypic plasticity, which potentially could affect its distribution in freshwaters. Fungal lectins constitute excellent ligands for development of affinity adsorbents useful in affinity chromatography. In this work, a lectin was purified from Pycnoporus sanguineus (PSL) mycelium using 3 procedures: by affinity chromatography, using magnetic galactosyl-nanoparticles or galactose coupled to Sepharose, and by ionic exchange chromatography (IEC). The highest lectin yield was achieved by IEC (55%); SDS-PAGE of PSL showed 2 bands with molecular mass of 68.7 and 55.2 kDa and IEC displayed 2 bands at pi 5.5 and 5.2. The lectin agglutinates rat erythrocytes, exhibiting broad specificity toward several monosaccharides, including galactose. The agglutination was also inhibited by the glycoproteins fetal calf fetuin, bovine lactoferrin, bovine transferrin, and horseradish peroxidase. The lectin was then used to synthesize an affinity adsorbent (PSL-Sepharose) and the interaction with glycoproteins was evaluated by analyzing their chromatographic behaviors. The strongest interaction with the PSL-derivative was observed with transferrin, although lower interactions were also displayed toward fetuin and lactoferrin. These results indicate that the purified PSL constitutes an interesting ligand for the design of affinity adsorbents to be used (i.e., in glycoprotein purification). El microbioma asociado a las plantas de interés agrícola, especialmente a nivel de las raíces, juega un papel fundamental en el reciclado de nutrientes y contribuye mediante distintos mecanismos al crecimiento y salud de la planta. En el cultivo de arroz irrigado, la inundación ocasiona una disminución en el potencial redox, generándose rápidamente condiciones anaerobias en el sistema. Mientras el suelo se mantiene en condiciones anóxicas, la entrada de oxígeno a través del aerénquima a nivel de la rizósfera de la planta de arroz crea un ambiente parcialmente óxico en donde conviven procesos anaerobios y aerobios. En este sistema, las bacterias desnitrificantes y las bacterias reductoras de sulfato (BRS) presentan gran relevancia ambiental, ya que participan en el reciclado de nutrientes como el carbono y el nitrógeno e intervienen en los procesos microbianos de producción y consumo de gases de efecto invernadero y de intermediarios metabólicos implicados en la emisión de estos gases. En Uruguay, el arroz es uno de los principales rubros de exportación. El sistema de producción utilizado tradicionalmente en nuestro pa´ıs que se basa en la rotación del cultivo de arroz con varios años de pasturas es considerado de baja intensidad y reducido impacto ambiental, y se cree que éste ha sido la base para la obtención de los excelentes rendimientos alcanzados. La reciente intensificaci´on en los sistemas de producción arrocera puede modificar la disponibilidad de nutrientes y, por lo tanto, la composición de las poblaciones microbianas presentes tanto en suelo como asociadas a la planta. En esta tesis se llevó a cabo el estudio de las comunidades microbianas de suelo y asociadas a plantas de arroz provenientes de un ensayo de campo, utilizando distintas herramientas moleculares. Se seleccionaron tres sistemas de rotación arrocera con diferente grado de intensificación y se tomaron muestras en dos etapas del desarrollo de la planta y manejo de agua. Mediante secuenciación masiva de genes 16S rRNA, se estudió la dinámica del microbioma bacteriano de suelo, rizósfera y raíces de arroz en los distintos sistemas de rotaci´on a lo largo del ciclo de cultivo, y se evaluó el efecto de la intensificación sobre estas comunidades. Los resultados obtenidos en este trabajo mostraron que el microbioma asociado al cultivo de arroz se vio determinado principalmente por el nivel de influencia de la planta sobre los distintos nichos asociados a las raíces (suelo, rizósfera y endósfera) y en segundo lugar por las condiciones redox del suelo establecidas luego de la inundación. Los distintos compartimientos presentaron comunidades claramente diferentes, presentando diferentes phyla dominantes. Mientras las comunidades del suelo se vieron dominadas por bacterias de los phyla Acidobacteria (23 %), Proteobacteria (23 %) y Actinobacteria (14 %), el microbioma rizosf´erico se vio marcadamente enriquecido en proteobacterias (37 %), principalmente Beta y Deltaproteobacteria, y Bacteroidetes (9 %). La comunidad rizosférica resultó significativamente más diversa que la de suelo y endósfera de raíces, presentando la mayor abundancia relativa de la clase Deltaproteobacteria, conocida por comprender una gran variedad de bacterias anaerobias, muchas de ellas reductoras de sulfato. Por su parte, el microbioma endófito de raíces se vio ampliamente enriquecido en proteobacterias (66 %), principalmente por las clases Alpha, Beta y Gammaproteobacteria, y también Firmicutes, mayormente afiliados al orden Clostridiales. Diversas bacterias anaerobias fueron capaces de establecer interacciones fuertes y estables con la planta de arroz a lo largo del ciclo del cultivo (Bacteroidales, Clostridium, Anaeromyxobacter, Geobacter, Desulfovibrio, entre otras), algunos inclusive en etapas tempranas del cultivo previo a la inundación. Mediante un análisis de especies indicadoras (IndVal) se identificaron varias OTUs fuertemente asociadas a alguna de las rotaciones arroceras, tanto en rizósfera como en el interior de las ra´ıces. Estos resultados sugieren que el microbioma asociado a las raíces de arroz podría ser susceptible a la intensificación agrícola, particularmente en taxones que no se encuentran entre los dominantes de estas comunidades. Por otra parte, se optimizaron metodolog´ıas dirigidas a genes funcionales para estudiar la abundancia (qPCR) y diversidad (T-RFLP) de BRS y bacterias desnitrificantes asociados a la planta de arroz en el sistema estudiado. Tanto las BRS como las bacterias desnitrificantes fueron detectadas y cuantificadas en suelo y asociadas a las raíces de arroz como rizosféricos y endófitos, y su abundancia y distribución en los distintos nichos se vio afectadas por el manejo del cultivo y el sistema de rotaci´on empleado. Las comunidades de BRS y arqueas metanogénicas asociadas al cultivo se vieron fuertemente impactadas por la inundación, incrementando significativamente su abundancia (qPCR de genes dsrA y mcrA, respectivamente) en rizósfera y endósfera para las BRS, y en suelo y rizósfera para el caso de las metanogénicas. De acuerdo con el análisis de secuenciación masiva del gen 16S rRNA, varios grupos taxonómicos conocidos como BRS fueron encontrados formando parte del microbioma de rizósfera de la etapa inundada del cultivo, mientras que el género Desulfovibrio fue el taxón dominante en el microbioma end´ofito de raíces de arroz. Las comunidades desnitrificantes tipo nirK y nirS presentaron distinta dinámica a lo largo del ciclo y frente a las distintas rotaciones. De acuerdo a la cuantificaci´on de genes nirK y nirS realizada por qPCR, las poblaciones tipo nirK dominaron en todos los nichos estudiados frente a las tipo nirS, y su abundancia se vio afectada tanto por los cambios que se dan durante el ciclo del cultivo como por las distintas rotaciones evaluadas. Por el contrario, la abundancia de las poblaciones tipo nirS se mantuvo constante en cada uno de los nichos a lo largo del ciclo y en los diferentes sistemas de rotación. Aunque la comunidad tipo nirS endófita presentó  la menor abundancia, su diversidad (evaluada mediante T-RFLP de este gen) resultó significativamente mayor a la presente en rizósfera. Esta tesis aporta información relevante sobre la ecolog´ıa de las comunidades bacterianas asociadas al cultivo de arroz, y en particular, de grupos fisiológicos anaerobios fundamentales en las transformaciones biogeoquímicas de nutrientes esenciales en este ecosistema. Los resultados obtenidos sugieren que los microorganismos anaerobios rizosféricos y endóitos constituyen una parte importante del microbioma asociado a la planta de arroz y podrían desempeñar un rol relevante a lo largo del ciclo de cultivo. Helminths use an alternative mitochondrial electron transport chain (ETC) under hypoxic conditions, such as those found in the gastrointestinal tract. In this alternative ETC, fumarate is the final electron acceptor and rhodoquinone (RQ) serves as an electron carrier. RQ receives electrons from reduced nicotinamide adenine dinucleotide through complex I and donates electrons to fumarate through complex II. In this latter reaction, complex II functions in the opposite direction to the conventional ETC (i.e., as fumarate reductase instead of succinate dehydrogenase). Studies in Ascaris suum indicate that this is possible due to changes in complex II, involving alternative succinate dehydrogenase (SDH) subunits SDHA and SDHD, derived from duplicated genes. We analyzed helminth genomes and found that distinct lineages have different gene duplications of complex II subunits (SDHA, SDHB, SDHC, and SDHD). Similarly, we found lineage-specific duplications in genes encoding complex I subunits that interact with quinones (NDUF2 and NDUF7). The phylogenetic analysis of ETC subunits revealed a complex history with independent evolutionary events involving gene duplications and losses. Our results indicated that there is not a common evolutionary event related to ETC subunit genes linked to RQ. The free-living nematode Caenorhabditis elegans uses RQ and has two genes encoding SDHA (sdha-1 and sdha-2) and two genes encoding NDUF2 (nduf2-1 and nduf2-2). sdha-1 and nduf2-1 are essential genes and have a similar expression pattern during C. elegans lifecycle. Using knockout strains, we found that sdha-2 and nduf2-2 are not essential, even in hypoxia. Yet, sdha-2 and nduf2-2 expression is increased in the early embryo and in dauer larvae, stages where there is low oxygen tension. Strikingly, sdha-1 and sdha-2 as well as nduf2-1 and nduf2-2 showed inverted expression profiles during the C. elegans life cycle. Finally, we found that sdha-2 and nduf2-2 knockout mutant strain progeny is affected. Our results indicate that different complex I and II subunit gene duplications provide increased fitness to worms. Two Pb(II)-resistant bacteria isolated from a soil containing 2,500 mg/kg of Pb were identified by 16S rRNA sequencing analysis as Delftia sp. and designated as 3C and 6C. Both isolates grew at a Pb(II) concentration of 62 mg/L and at the stationary phase showed a Pb(II)-sorption capability of 10 ± 1.5 (3C) and 5 ± 0.8 (6C) mg/g of biomass. Biochemical properties related to heavy metal resistance and plant growth promotion were analyzed and compared with the Cr(VI)-resistant plant growth-promoting Delftia sp. JD2, previously reported by our group. Both isolates and JD2 were resistant to Cr(VI), Pb(II) and many antibiotics, produced siderophores and the phytohormone indole-3-acetic, and showed clover growth-promoting activity in greenhouse conditions. Interestingly, the occurrence of integron class 1 was shown in all isolates. Our results add to previous reports and suggest that bacteria of the genus Delftia could be consider as good candidates for the design of technologies for cleaning up contaminated environments and/or the production of biofertilizers. Background Microbial oils produced by diverse microorganisms are being considered as alternative sources of triglycerides for biodiesel production. However, the standalone production of biodiesel from microorganisms is not currently economically feasible. In case of yeasts, the use of low-value nutrient sources in microbial production and the implementation of cost-efficient downstream processes could reduce costs and make microbial lipids competitive with other commodity-type oils in biodiesel production. Industrial biodiesel synthesis from oleaginous seeds is currently based on a multistep process. However, a simple process called in situ transesterification (ISTE), which takes place within the biomass without a previous lipid extraction step, is receiving increasing interest. In this work, the optimal conditions for an ISTE process to obtain biodiesel from previously selected oleaginous yeast (Rhodotorula graminis S1/S2) were defined using the response surface methodology (RSM). Results Using the RSM approach, the optimal conditions for the maximum yield with minimum reaction time included a methanol-to-biomass ratio of 60:1, 0.4 M H2SO4, and incubation at 70°C for 3 h. The optimized in situ process yield was significantly higher (123%) than that obtained with a two-step method in which fatty acids from saponifiable lipids were first extracted and then esterified with methanol. The composition of the fatty acid methyl ester mixture obtained from R. graminis S1/S2 by ISTE met Uruguayan standards for biodiesel. Conclusion The characteristics achieved by the optimized method make microbial oil a potential alternative for biodiesel production from yeast at an industrial scale. Achyrocline satureioides (Lam) D.C (Compositae) is a native medicinal plant of South America traditionally utilized for its anti-inflammatory, sedative and anti-atherosclerotic properties among others. Neuroprotective effects have been reported in vivo and could be associated to its elevated content of flavonoid aglycones. In the present study we performed the isolation and structure elucidation of the major individual flavonoids of A. satureioides along with the in vitro characterization of their individual antioxidant and neuroprotective properties in order to see their putative relevance for treating neurodegeneration. Exact mass, HPLC-MS/MS and 1H NMR identified dicaffeoyl quinic acid isomers, quercetin, luteolin, isoquercitrin, and 3-O-methylquercetin as the mayor polyphenols. Flavonoids intrinsic redox properties were evaluated in the presence of the endogenous antioxidants GSH and Ascorbate. Density Functional Theory (DFT) molecular modeling and electron density studies showed a theoretical basis for their different redox properties. Finally, in vitro neuroprotective effect of each isolated flavonoid was evaluated against hydrogen peroxide-induced toxicity in a primary neuronal culture paradigm. Our results showed that quercetin was more efficacious than luteolin and isoquercitrin, while 3-O-methylquercetin was unable to afford neuroprotection significantly. This was in accordance with the susceptibility of each flavonoid to be oxidized and to react with GSH. Overall our results shed light on chemical and molecular mechanisms underlying bioactive actions of A. satureioides main flavonoids that could contribute to its neuroprotective effects and support the positive association between the consumption of A. satureioides as a natural dietary source of polyphenols, and beneficial health effect.
