En este trabajo, a partir de cultivos puros de Hanseniaspora vineae, Hanseniaspora opuntiae y Hanseniaspora guillermondi, se realizaron microvinificaciones en mosto de uva tinta. En los vinos elaborados se midieron índices espectrofotométricos de color y polifenoles. Los resultados obtenidos mostraron diferencias significativas entre las cepas para antocianos totales (p < 0,001), intensidad colorante (p< 0,01), matiz (p < 0,001), y polifenoles totales (p < 0,05). Los valores mayores de antocianos totales fueron obtenidos para la cepa H. opuntiae (T06/19G), mientras que los valores mayores de intensidad colorante se obtuvieron para la cepa H. vineae (T02/19F), H. guillermondi (T06/09G), y H. opuntiae (T08/01G). En cuanto a poder fermentativo, dos de las cepas de H. vineae (02/5 F y 02/25F), fueron las que presentaron los valores mayores. Estos resultados demuestran la capacidad que poseen algunas cepas no-Saccharomyces, del género Hanseniaspora, de influir sobre la composición polifenólica de los vinos tintos. El uso desmedido de fuentes de energía fósiles ha provocado desbalances en la composición atmosférica lo que ha causado un efecto invernadero alterando la temperatura del planeta. Esta problemática ambiental, sumada al esperado incremento del precio de los combustibles fósiles en el futuro próximo, plantea la necesidad de explorar nuevas fuentes de energía y por consiguiente desarrollar las tecnologías adecuadas para su producción. Una alternativa para la producción de energía y combustibles es la biomasa. Sin embargo, suele tener un alto contenido de humedad y de grupos oxigenados en su estructura química, lo que hace que su poder calorífico sea inferior al de los combustibles fósiles y su estabilidad temporal escasa. El uso de tratamientos termoquímicos o biológicos permite transformar la biomasa en biocombustibles con mayor poder calorífico inferior (PCI), mayor estabilidad temporaria y en algunos casos adecuarlos a los sistemas existentes utilizados para combustibles fósiles. Uno de los tratamientos termoquímicos utilizados es el procesamiento hidrotérmico. En especial, la licuefacción hidrotérmica (LHT) es un proceso termoquímico que consiste en someter la biomasa a temperaturas entre 250-400 oC y alta presión (10-25 MPa) durante tiempos de residencia cortos (0-60 min) en medio acuoso [1] produciendo biocombustibles sólidos, líquidos y gaseosos, conocidos como hydrochar, biocrudo y gas, respectivamente y una solución acuosa rica en compuestos orgánicos disueltos. Al ser un proceso en medio acuoso permite el uso de biomasa húmeda sin necesidad de una operación previa de secado evitando con esto pérdidas energéticas. Por otro lado, la biomasa a utilizar para fabricación de biocombustibles debe ser abundante y cercana al lugar de transformación para evitar altos costes de transporte. En el caso de Uruguay se estima una producción de residuos lignocelulósicos de 2.2 a 2.8 millones de m3 por año ( 1.5 a 2 millones de toneladas por año) [2] siendo el eucalipto y el pino las maderas más producidas. Es por esta disponibilidad de biomasa que se decidió estudiar la producción de biocombustibles mediante LHT utilizando estas dos maderas. 3 4 En este trabajo se estudió el efecto de las condiciones de reacción en la distribución de los productos de la LHT de madera de pino y eucalipto, así como la influencia de dichas condiciones sobre el PCI de los productos. Para ello se estudió el efecto del tiempo de residencia, la temperatura, la relación solución acuosa:biomasa y el uso de carbonato de potasio y bicarbonato de potasio como catalizadores, así como un análisis de la factibilidad energética del proceso. Se determinó que los parámetros de reacción influyen en la distribución de productos, siendo los de mayor influencia la relación solución:biomasa a la cual se trabaja y el uso de los catalizadores. Bacterial adenosine 5’-diphosphate-ribosylating toxins are encoded by several human pathogens, such as Pseudomonas aeruginosa (exotoxin A (ETA)), Corynebacterium diphtheriae (diphtheria toxin (DT)), and Vibrio cholerae (cholix toxin (CT)). The toxins modify eukaryotic elongation factor 2, an essential human enzyme in protein synthesis, thereby causing cell death. Targeting external virulence factors, such as the above toxins, is a promising alternative for developing new antibiotics, while at the same time avoiding drug resistance. This study aims to establish a reliable computational methodology to find a “silver bullet” able to target all three toxins. Herein, we have undertaken a detailed analysis of the active sites of ETA, DT, and CT, followed by the determination of the most appropriate selection of the size of the docking sphere. Thereafter, we tested two different approaches for normalizing the docking scores and used these to verify the best target (toxin) for each ligand. The results indicate that the methodology is suitable for identifying selective as well as multitoxin inhibitors, further validating the robustness of inverse docking for target-fishing experiments. Cardiomyopathy due to monoclonal light chain deposits is a complication of plasma cell disorders. The deposits may be either fibrillar as in light chain amyloid or nonfibrillar as in light chain deposition disease. The reasons for these structural differences are still unknown. We characterized the myocardial deposits by immunohistochemical examination of sections and extraction and biochemical analysis of the tissue deposits in a patient (MCM) who died of myeloma and systemic light chain deposition disease. Amino acid sequence analysis of the extracted nonfibrillar MCM kappa-light chain reveals that it belongs to the L12a germline subset of the kappa(I) protein and contains five distinctive amino acid substitutions (three in the framework region III and two in the complementarity-determining region III) that have not been reported previously in the same positions in other kappa(I) light chains. The theoretically determined isoelectric point (pI 8.21) of the MCM light chain is high compared with the low isoelectric point of other Bence Jones proteins from subjects without light chain deposition disease. The diffuse binding to basement membranes and the high isoelectric point of the MCM kappa-light chain suggest electrostatic interaction as a possible mechanism of tissue deposition. Ralstonia pseudosolanacearum GMI1000 (Rpso GMI1000) is a soil-borne vascular phytopathogen that infects host plants through the root system causing wilting disease in a wide range of agroeconomic interest crops, producing economical losses. Several features contribute to the full bacterial virulence. In this work we study the participation of light, an important environmental factor, in the regulation of the physiological attributes and infectivity of Rpso GMI1000. In silico analysis of the Rpso genome revealed the presence of a Rsp0254 gene, which encodes a putative blue light LOV-type photoreceptor. We constructed a mutant strain of Rpso lacking the LOV protein and found that the loss of this protein and light, influenced characteristics involved in the pathogenicity process such as motility, adhesion and the biofilms development, which allows the successful host plant colonization, rendering bacterial wilt. This protein could be involved in the adaptive responses to environmental changes. We demonstrated that light sensing and the LOV protein, would be used as a location signal in the host plant, to regulate the expression of several virulence factors, in a time and tissue dependent way. Consequently, bacteria could use an external signal and Rpsolov gene to know their location within plant tissue during the colonization process. The full utilization of the main components in the lignocellulosic biomass is the major goal from a biorefinery point of view, giving not only environmental benefits but also making the process economically viable. In this sense the solid residue obtained in bioethanol production after steam explosion pretreatment, enzymatic hydrolysis, and fermentation of the lignocellulosic biomass, was studied for further valorization. Two different residues were analyzed, one generated by the production of cellulosic ethanol from an energy crop such as switchgrass (Panicum virgatum) and the other, from wood (Eucalyptus globulus). The chemical composition of these by-products showed that they were mainly composed of lignin with a total content range from 70 to 83% (w/w) and small amounts of cellulose and hemicellulose. The present work was focused on devising a new alternative for processing these materials, based on the ability of the ionic liquids (IL) to dissolve lignocellulosic biomass. The resulting mixture of biopolymers and IL constituted the raw material for developing new insoluble biocatalysts. Active hydrogels based on fungal laccase from Dichostereum sordulentum 1488 were attained. A multifactorial analysis of the main variables involved in the immobilization process enabled a more direct approach to improving hydrogel-bound activity. These hydrogels achieved a 97% reduction in the concentration of the estrogen ethinylestradiol, an emerging contaminant of particular concern due to its endocrine activity. The novel biocatalysts based on fungal laccase entrapped on a matrix made from a by-product of second-generation bioethanol production presents great potential for performing heterogeneous catalysis offering extra value to the ethanol biorefinery. The mortality of C. brevis was significantly higher than in untreated controls at concentrations above 50 mg/cm3 and similar to controls with starved termites. The value of LC50 was calculated as 32.76 mg/cm3. Also a residual action was evaluated six months after the blocks were treated. A deterrent but sub-lethal effect on termites was observed. The number of protozoa in termite intestines decreased significantly compared to control. The lime wax was effective against C. brevis, reducing the feeding activity and the survival of termites, and appears to have the potential to be used in the treatment of wood for its protection against powderpost termites. In recent years, the international demand for commodities has prompted enormous growth in agriculture in most South American countries. Due to intensive use of fertilizers, cyanobacterial blooms have become a recurrent phenomenon throughout the continent, but their potential health risk remains largely unknown due to the lack of analytical capacity. In this paper we report the main results and conclusions of more than five years of systematic monitoring of cyanobacterial blooms in 20 beaches of Montevideo, Uruguay, on the Rio de la Plata, the fifth largest basin in the world. A locally developed microcystin ELISA was used to establish a sustainable monitoring program that revealed seasonal peaks of extremely high toxicity, more than one-thousand-fold greater than the WHO limit for recreational water. Comparison with cyanobacterial cell counts and chlorophyll-a determination, two commonly used parameters for indirect estimation of toxicity, showed that such indicators can be highly misleading. On the other hand, the accumulated experience led to the definition of a simple criterion for visual classification of blooms, that can be used by trained lifeguards and technicians to take rapid on-site decisions on beach management. Fungal decay, predominantly caused by Penicillium expansum, is the main cause of apple postharvest losses. To overcome this problem, synthetic fungicides are applied to fruit before storage. Public concern about this practice has raised interest in developing alternative biocontrol strategies for postharvest diseases. Debaryomyces hansenii F9D, a psychrotrophic Antarctic yeast, has been previously selected as a promising biocontrol agent due to its ability to reduce blue mold incidence in apples and pears. Previous in vitro studies indicated that the biocontrol mechanism of this yeast does not involve the production of antifungal metabolites or other inhibitory or competitive interactions with the pathogen. In this work, we conducted further studies in order to understand the interaction between Penicillium expansum and D. hansenii F9D within apple wounds. The growth of both mi croorganisms in apple wounds during cold storage was followed over time by q-PCR and viable cell counts. Scanning electron microscopy observations of co-inoculated and control wounds (inoculated only with the yeast or with the pathogen) were conducted to evaluate if the pathogen conidia could germinate in presence of the yeast and to determine the colonization patterns of P. expansum and the biocontrol yeast. Our results indicate that Debaryomyces hansenii F9D is able to prevent cold stored apples’ wounds rot by inhibiting the virulence factors produced by the pathogen without impeding its growth. These findings provide circumstantial evidence for a new biocontrol mechanism associated with the prevention of fungal decay in apples by biocontrol yeasts during cold storage of fruit. Liquid-phase oxidation of limonene was performed with molecular oxygen as the sole oxidizing agent under non-solvent conditions. The oxidation took place under atmospheric pressure and temperatures between 70 and 90 �C in the presence of three different nickel–aluminium hydrotalcites. The final limonene conversion at 80 �C and 6 h of reaction time was comprised between 40 and 50 %. Several oxygenated limonene derivatives such as endo- and exo-epoxides, carveol and carvone represent 45–60 % of the whole amount of products. In the absence of a catalyst, a higher initial content of peroxides in limonene lead to a higher reaction rate with no significant changes in product selectivities. Thermal decomposition of limonene peroxides took place as the initiation path of an autooxidation mechanism leading to the different products. The catalyst played an important role in the initial paths involving activation of both reactants as well as in the decomposition of limonene peroxide to form radicals. A long-standing pathomechanistic model proposes that the polyglutamine (polyQ)-length-dependent toxicity threshold observed in all polyQ diseases is triggered by a conformational change within the monomer that occurs only above a certain polyQ length. If true, this yet undefined and elusive mutant-specific toxic conformation would constitute a direct therapeutic target. Three anti-polyQ antibodies—MW1, 1C2 and 3B5H10—have been extensively used to probe the conformation of polyQ. The crystal structure of the MW1 epitope reveals a linear, non-pathogenic polyQ. In contrast, although the detailed structure of its epitope is unknown, the 3B5H10 antibody is widely advertised and used as a conformational antibody that recognizes the toxic conformation of expanded polyQ. We solved the crystal structure of the 1C2 antigen-binding domain (1C2-Fab) and performed a direct comparison between the 1C2, MW1 and 3B5H10 structures. The MW1 and 1C2 antibodies have similar sequences and structures, consistent with their binding to short polyQ and their polyQ length-discrimination properties. Unexpectedly, the 3B5H10 antibody also shares striking features with MW1 and 1C2, which prompted us to revisit its binding properties. We show that the 3B5H10 epitope is actually a short, non-pathogenic polyQ. All three antibodies MW1, 1C2 and 3B5H10 interact similarly with polyQ of various lengths, and bind small polyQ epitopes in similar linear and extended conformations. Together with studies published during the recent years, our work argues against the hypothesis that a mutant-specific conformation in monomeric polyQ molecules is the toxic entity responsible for polyQ diseases. In most organisms, thioredoxin (Trx) and/or glutathione (GSH) systems are essential for redox homeostasis and deoxyribonucleotide synthesis. Platyhelminth parasites have a unique and simplified thiol-based redox system, in which the selenoprotein thioredoxin-glutathione reductase (TGR), a fusion of a glutaredoxin (Grx) domain to canonical thioredoxin reductase domains, is the sole enzyme supplying electrons to oxidized glutathione (GSSG) and Trx. This enzyme has recently been validated as a key drug target for flatworm infections. In this study, we show that TGR possesses GSH-independent deglutathionylase activity on a glutathionylated peptide. Furthermore, we demonstrate that deglutathionylation and GSSG reduction are mediated by the Grx domain by a monothiolic mechanism and that the glutathionylated TGR intermediate is resolved by selenocysteine. Deglutathionylation and GSSG reduction via Grx domain, but not Trx reduction, are inhibited at high [GSSG]/[GSH] ratios. We found that Trxs (cytosolic and mitochondrial) provide alternative pathways for deglutathionylation and GSSG reduction. These pathways are operative at high [GSSG]/[GSH] and function in a complementary manner to the Grx domain-dependent one. Despite the existence of alternative pathways, the thioredoxin reductase domains of TGR are an obligate electron route for both the Grx domain- and the Trx-dependent pathways. Overall, our results provide an explanation for the unique array of thiol-dependent redox pathways present in parasitic platyhelminths. Successful establishment of a parasite infection depends partially on the host intrinsic susceptibility to the pathogen. In cystic echinococcosis (CE), a zoonotic disease caused by the cestode parasite Echinococcus granulosus, the infection outcome in the murine model of secondary CE varies according to the mouse strain used. In this regard, intrinsic differences in susceptibility to the infection were previously reported for Balb/c and C57Bl/6 mice, being C57Bl/6 animals less permissive to secondary CE. Induction of parasite-specific antibodies has been suggested to play relevant roles in such susceptibility/resistance phenomena. Here, we report an in deep comparison of antibody responses induced in both mouse strains. Firstly, only C57Bl/6 mice were shown to induce specific-antibodies with efficient anti-parasite activities during early secondary CE. Then, through ImmunoTEM and Serological Proteome Analysis (SERPA), an evaluation of specific antibody responses targeting parasite tegumental antigens was performed. Both strategies showed that infected C57Bl/6 mice -unlike Balb/c animals- narrowed their IgG recognition repertoire against tegumental antigens, targeting fewer but potentially more relevant parasite components. In this sense, tegumental antigens recognition between Balb/c and C57Bl/6 mice, either by natural and/or induced antibodies, was analyzed through SERPA and MALDI-TOF/TOF studies. A total of 13 differentially recognized proteins (DRPs) uniquely targeted by antibodies from C57Bl/6 mice were successfully identified, wherein a subset of 7 DRPs were only recognized by infection-induced antibodies, suggesting their potential as natural protective antigens. In this regard, immunoinformatic analyses showed that such DRPs exhibited higher numbers of possible T cell epitopes towards the H-2-IAb haplotype, which is present in C57Bl/6 mice but absent in Balb/c animals. In summary, our results showed that the genetic predisposition to generate better T-dependent antibody responses against particular tegumental antigens might be a key factor influencing host susceptibility in the murine model of secondary CE. Sensory product characterisation by consumers is increasingly supplemented by measurement of emotional associations. However, studies that link products' sensory perception and emotional associations are still scarce. Five consumer studies were conducted using cashew nuts, peanuts, chocolate, fruit and processed tomatoes as the product categories. Consumers (n = 685) completed check-all-that-apply (CATA) questions to obtain sensory product perceptions and associations with emotion words. The latter were conceptualised and interpreted through a circumplex emotion model spanned by the dimensions of valence (pleasure to displeasure) and arousal (activation to deactivation). Through regression analysis, sensory terms were mapped to the circumplex model to represent statistical linkages with emotion words. Within a were interpretable. The most notable finding was the highly study-specific nature of the linkages, which was mainly attributed to the influence of product category. Methodological choices may also have been partly responsible for the differences. Three studies used a general emotion vocabulary (EsSense Profile®) and an identical number of sensory terms (n = 39). The less complete coverage of the emotional circumplex and the presence of synonymous sensory terms could have diminished the ability to interpret the results. Conversely, two studies used fewer emotion words and sensory terms and these, furthermore, were purposefully selected for the focal sets of samples. The linkages in these latter studies were more interpretable and this could suggest that customised vocabularies of modest length may be desirable when seeking to establish linkages between emotional associations and sensory characteristics of food/beverage stimuli. Purposeful inclusion of emotion words that fully span the circumplex emotion model may also be desirable. Overall, the research represents a new method for establishing linkages between the sensory properties and emotional association to food and beverage products.
