A summary of the resource.
Lapachol acetate [systematic name: 3-(3-methyl­but-2-en­yl)-1,4-dioxonaph­thalen-2-yl acetate], C17H16O4, was prepared using a modified high-yield procedure and its crystal structure is reported for the first time 80 years after its first synthesis. The full spectroscopic characterization of the mol­ecule is reported. The mol­ecular conformation shows little difference with other lapachol derivatives and lapachol itself. The packing is directed by inter­molecular π–π and C—H...O inter­actions, as described by Hirshfeld surface analysis. The former inter­actions make the largest contributions to the total packing energy in a ratio of 2:1 with respect to the latter.

<span class="hit">A</span> double <span class="hit">mutant</span> strain (UR3) of R. <span class="hit">meliloti</span> L5-30 was isolated from <span class="hit">a</span> phosphoglucose isomerase <span class="hit">mutant</span> (UR1) on the basis of its resistance to fructose inhibition when grown on fructose-rich medium. UR3 lacked both phosphoglucose isomerase and <span class="hit">fructokinase</span> activity. <span class="hit">A</span> <span class="hit">mutant</span> strain (UR4) lacking only the <span class="hit">fructokinase</span> activity was derived from UR3; it grew on the same C sources as the parent strain, but not on fructose, mannitol, or sorbitol. <span class="hit">A</span> spontaneous revertant (UR5) of normal growth phenotype contained <span class="hit">fructokinase</span> activity. <span class="hit">A</span> fructose transport system was found in L5-30, UR4, and UR5 grown in arabinose-fructose minimal medium. No fructose uptake activity was detected when L5-30 and UR5 were grown on arabinose minimal medium, but this activity was present in strain UR4. Free fructose was concd. intracellularly by UR4 >200-fold above the external level. <span class="hit">A</span> partial transformation of fructose into mannitol and sorbitol was detected by enzymic anal. of the uptake products. Polyol dehydrogenase activity was detected in UR4 grown in arabinose-fructose minimal medium. The induction pattern of polyol dehydrogenase activities in this strain might be due to slight intracellular fructose accumulation.

Abstract

Background: Invasive fungal diseases represent an unmet clinical need that could 29 benefit from novel immunotherapeutic approaches. Host pattern-recognition receptors 30 (e.g., Toll-like receptors, C-type lectins or Scavenger receptors) that sense conserved 31 fungal cell wall constituents may provide suitable immunotherapeutic antifungal agents. 32 Thus, we explored the therapeutic potential of the lymphocyte class I scavenger receptor 33 CD5, a non-redundant component of the antifungal host immune response that binds to 34 fungal β-glucans. 35 Methods: antifungal properties of the soluble ectodomain of human CD5 (shCD5) were 36 assessed in vivo in experimental models of systemic fungal infection induced by 37 pathogenic species (Candida albicans and Cryptococcus neoformans). In vitro 38 mechanistic studies were performed by means of fungal-spleen cell co-cultures. 39 Results: shCD5-induced survival of lethally infected mice was dose and time-dependent 40 and concomitant with reduced fungal load and increased leukocyte infiltration in 41 primary target organ. Additive effects were observed in vivo after shCD5 was combined 42 with sub-optimal doses of fluconazole. Ex vivo addition of shCD5 to fungal-spleen cell 43 co-cultures increased release of pro-inflammatory cytokines involved in antifungal 44 defence (TNF-α and IFN-γ) and reduced the number of viable C. albicans. 45 Conclusions: The results prompt further exploration of the adjunctive therapeutic 46 potential of shCD5 in severe invasive fungal diseases.

A summary of the resource.
The use of biodiesel has generated a diversification of the energy matrix, which has led to a lower dependence on fossil fuels and a concomitant reduction of the adverse effects on the environment generated by their use. Currently, biodiesel is mainly produced by transesterification with short-chain alcohols of oils from different sources, in particular from oil crops, such as soybean and canola. Since the use of crops for the generation of fuels competes with their use as food, it seems appropriate to explore new oil sources for the production of biodiesel, such as the intracellular triglycerides accumulated by oleaginous yeasts. In this case, to achieve a competitive process, yeast production should be carried out using low-cost fermentable substrates such as industrial wastes or by-products. In this work we optimized the production of intracellular lipids by a previously selected oleaginous yeast strain in a fed-batch culture in presence of crude glycerol as a source of carbon and energy. In the optimized conditions an important improvement of lipid concentration was achieved, obtaining 12.2 g L−1 of fatty acid methyl esters (FAMEs) and 26.6 g L−1 of dry biomass in 360 h of culture.

Various microbial groups are well known to produce a range of extracellular enzymes and other secondary metabolites. However, the occurrence and importance of investment in such activities have received relatively limited attention in studies of Antarctic soil microbiota. Sixty-one yeasts strains were isolated from King George Island, Antarctica which were characterized physiologically and identified at the molecular level using the D1/D2 region of rDNA. Fifty-eight yeasts (belonging to the genera Cryptococcus, Leucosporidiella, Rhodotorula, Guehomyces, Candida, Metschnikowia and Debaryomyces) were screened for extracellular amylolytic, proteolytic, esterasic, pectinolytic, inulolytic xylanolytic and cellulolytic activities at low and moderate temperatures. Esterase activity was the most common enzymatic activity expressed by the yeast isolates regardless the assay temperature and inulinase was the second most common enzymatic activity. No cellulolytic activity was detected. One yeast identified as Guehomyces pullulans (8E) showed significant activity across six of seven enzymes types tested. Twenty-eight yeast isolates were classified as oleaginous, being the isolate 8E the strain that accumulated the highest levels of saponifiable lipids (42 %).

Forest plantations inUruguay have doubled in the past decade, with Eucalyptus spp. leading this growth. The bronze bug, Thaumastocoris peregrinus (Heteroptera: Thaumastocoridae), originally restricted to Australia, is an important emerging pest of Eucalyptus plantations in the Southern hemisphere. T. peregrinus feeds onmature Eucalyptus leaves, causing them to turn brown and often fall from the tree. Although population dynamics and behavioural patterns are not clearly understood, circumstantial observations suggest that males and nymphs aggregate. We used gas chromatography coupled to mass spectrometry to analyze volatile organic compounds emitted by virgin males and females, and characterized a male-specific compound, 3-methylbut-2-enyl butanoate, based on mass spectral data and chromatographic comparison with a synthetic standard. We also performed Y-olfactometer bioassays to test the attraction of virgin males and females toward live virgin males, male volatile extracts, and synthetic 3-methylbut- 2-enyl butanoate. Males were attracted toward conspecific males, while virgin females showed no preference, suggesting that male volatiles are not involved in sexual communication. Further olfactometer tests showed that males were attracted to male volatile extracts and to synthetic 3-methylbut-2-enyl butanoate. The ecological significance of this compound and its potential use for the management of T. peregrinus in Eucalyptus forests will be further investigated.

Two new copper(II) complexes containing the methyl(2-pyridyl)ketone oxime ligand (mpkoH) [Cu3(OH)(ClO4)2(mpko)3]·CH3OH (1) and [Cu(ClO4)(mpko)(mpkoH)]n (2) have been prepared from Cu(ClO4)2 and mpkoH in different metal-to-ligand molar ratios. In addition, the compound [Cu{(mpko)2BF2}(H2O)](BF4) (3) [(mpko)2BF2 is the fluoroboration product of the oxime] has been obtained when replacing Cu(ClO4)2 by Cu(BF4)2. Compound 1 is an isolated triangle with a {Cu3(µ3-OH)}5+ core, whereas 2 is a chain of CuII ions linked by anionic mpko− bridges. 1 exhibits strong antiferromagnetic competing interactions, as well as antisymmetric exchange. On the other hand, very weak ferromagnetic interactions are found in 2. The magnetic properties of these compounds have been analyzed by magnetic measurements and EPR spectra.

wo new mixed-valence iron complexes with 2-pyridyl oximes, [Fe(mpko)3Fe(H2O)2(NO3)](NO3)·2H2O (1) (mpko− = methyl(2-pyridyl)ketone oximate) and [{Fe(dpko)3}2Fe](ClO4)·4H2O (2) (dpko− = bis(2-pyridyl)ketone oximate), have been prepared by reaction of FeIII with mpkoH in methanol (1) and FeII with dpkoH in methanol/water (2). Dinuclear FeII(low-spin)FeIII(high-spin) and trinuclear FeII(low-spin)FeIII(high-spin)FeII(low-spin) cations are present in the crystal structure of 1 and 2, respectively. Intermolecular hydrogen bonds in 1 lead to weak antiferromagnetic interactions between pairs of neighboring FeIII centers, which allows observation of single-ion zero-field splitting effects. Dinuclear class II CoIII/FeII mixed-valence complexes of type [LnCoIII(m-NC)FeII(CN)3L2]m� (where Ln represents a pentadentate macrocycle and L2 a bpy ligand or two cyanides) have electronic properties that make them possible sensitizers in DSSC (dye sensitized solar cells). For this purpose the new complex Na2[{trans-L14COOCoIII(m-NC)}FeII(CN)5] has been prepared and characterized by the usual methods and its sensitizer properties compared with those of the already known [{trans-L15CoIII(m-NC)}FeII(CN)3(bipy)eq,ax](ClO4). The complex [{trans- L14COOCoIII(m-NC)}FeII(CN)5]2� has been designed for the actuation of an electron injection from the cobalt centre on MMCT, while the [{trans-LnCoIII(m-NC)}FeII(CN)3(bipy)]+ structure can produce a tuned injection from the iron centre via an MLCT electronic state, as described for similar systems. The characterization on solid oxide semiconductor supports has been carried out for these two complexes and the results have been compared with the behaviour observed in aqueous solution and in solvents of varying polarities. Their use in DSSC has been checked and, while a sensitizer response is observed for [{trans-L14COOCoIII(m-NC)}FeII(CN)5]2�, complex [{trans-L15CoIII(m-NC)}FeII(CN)3(bipy)eq,ax]+ does not produce any electrical current on illumination. The low efficiency of the built DSSC can be easily related both with the very low value of the extinction coefficient of the MMCT band responsible for the electron injection, and with the small driving force for the reduction of the complex with the standard I2/I3� system used after electron injection.

<p>En el marco de un intento activo de promover el desarrollo agrícola e industrial del Uruguay, durante la segunda presidencia de José Batlle y Ordoñez ¿1911=1015? se propicio la producciçon de carburantes que permitieran al país resolver su problema energético. A esos efectos el Instituto de Química Industrial =creado en 1912=, tuvo como uno de sus cometidos fundamentales la fabricación del "carburante nacionale" en base a alcohol. Como resultado de sus investigaciones, en 1923 se llevaron a cabo ensayos técnicamente exitosos, utilizando una mezcla de alcohol y nafta, sin embargo no tuvieron aplicación.</p>
<p>En los casi noventa años que han transcurrido desde entonces, el problema ha aparecido una y otra vez en la agenda política nacional, sin que se haya sin que se haya logrado su resolución hasta el presente. Buscando arrojar algo de luz sobre las posibles causas de este persistente estancamiento, consideramos importante realizar una aproximación a los veinte años posteriores a las investigaciones mencionadas, analizando la creación de la ANCAP (Administración Nacional de Combustibles, ALcohol y Pórtland), y el comportamiento que asumieron tanto ella como las compañías petrolíferas extranjeras establecidas en el país, respecto del tema en cuestión.</p>

Prospection for new sources of botanical pesticides has shown a revival in the last decades due not only to the fast development of resistance among different pests around the world but also by the need to use less eco-toxic products to control etiological agents of different pest-related problems found in agro-production. In this work, extracts from members of the families Simaroubaceae (Ailanthus altissima, Castela coccinea and Picrasma crenata) and Picramniaceae (Alvaradoa subovata and Picramnia sellowii) were evaluated for their toxicity against the cattle tick and for their antifeedant activity against insects. At the tested doses, none of the extracts exhibited a good toxicity against larvae of the common cattle tick. On the other hand, antifeedant activity was detected in various extracts. Foliage consumption was completely deterred in adults of the specialist Epilachna paenulata in the cases of the C. coccinea leaf and wood extracts and P. crenata wood extract. For larvae of the generalist Spodoptera frugiperda antifeedant effects were also detected, although in a lesser extent. Phytochemical analyses of the extracts showed the presence of alkaloids in P. crenata (including canthin-6-one) and in C. coccinea. Besides, GC/MS analyses of the wood extract from C. coccinea showed the presence of several steroids (ergot 5-en-3-ol-acetate, stigmastan-3,5 diene and stigmasta-3,5dien-7-one). Anthraquinones (emodin and chrysophanol) as well as chlorogenic acid were detected in the cases of A. subovata and P. sellowii extracts. The differences in deterrent activity could not be atributed to differences in the HPLC chemical profiles of the different extracts

An unsupplemented peach jam and four peach jam samples prepared by supplementing the former with increasing amounts of Aloysia citriodora aqueous extract were subjected to sensory testing. A trained panel of eight assessors initially identified (reaching consensus) the relevant sensory attributes and then rated the jam samples on an individual basis. The jam samples were later evaluated for overall liking and willingness to purchase by a group of 95 interested consumers, who also provided their individual appraisal of the sensory features of the various samples by responding to an open-ended question. Overall, consumers appeared not to accept the sensory modifications introduced in traditional peach jam by the addition of A. citriodora extract, even though they were expressly acquainted with its health benefits. The degree of acceptance of the extract supplement decreased significantly with increasing amounts of extract in the jam. The groups identified to have the least and the greatest tolerance to the sensory modifications introduced in the supplemented jams differed in gender distribution, with male consumers having a greater tolerance than females.

Polyphenols are a popular functional ingredient used in foods due to their antioxidant activity. The leaves of Acca sellowiana, a South American native plant, have a much higher polyphenol content than other popular native plants in the region. In this context, five strawberry jams were developed with increasing amounts of A. sellowiana extract. A panel of trained sensory assessors initially evaluated the sensory profiles of the obtained products. Subsequently, three jams were selected and were evaluated by 96 consumers in two instances. Coded samples were presented in the first session; in the second session, the consumers were presented with information on whether the jams contained antioxidants or not. In both sessions, the acceptability was determined and a description was obtained by means of a check-all-that-apply question. The original jam without A. sellowiana extract presented a polyphenol content of 310.0 mg gallic acid equivalents/100 g and an acceptability higher than 6 in both sessions. A jam was obtained in which the polyphenol content increased to 467.5 mg gallic acid equivalents/100 g, with a sensory profile and acceptability that were similar to those of the original product. Information on enrichment with antioxidants did not affect the acceptability, but it did modify the consumer’s perception, as the consumer showed a predisposition to perceive negative sensory attributes when the product was presented with such information but also perceived it as novel and a good complement to the diet.

A summary of the resource.
Efficiency of in-situ alkaline transesterification of dry biomass from a crop of Spirulina sp., using ultrasound-assisted and/or the addition of different co-solvents to the reaction medium was studied. The starting biomass had a total lipid content of 7.0% (Folch method), comprised mainly by the following fatty acids: 16:0 (44.2%), 18:3 (n–6) (23.3%), and 18: 2 (n–6) (11.1%). When the transesterification reaction was carried out without ultrasound, it only extracted 12.8% of lipids (0.9% of the biomass) after 2 h of reaction, achieving only a 63.6% ester content. When the process was carried out under the same conditions except for the ultrasound-assisted (80 W), the weight yield remained mostly unchanged, but the ester content increased to 97.6%. Additionally, when chloroform was added to methanol as a co-solvent in a 2:1 ratio it was possible to extract 43% of lipids. However, the ester content was reduced to 69.7%. Moreover, with maximum ultrasound power (180 W) a high ester content (96.9%) and a relatively high extraction yield (26%) were obtained, even without the co-solvent. These results show that the ultrasound significant increased the yield of the in-situ alkaline transesterification of Spirulina sp. biomass, promoting a higher percent recovery of lipids as well as ester content from the starting material.

The nitrilotripropionic acid (H3ntp) is a flexible polycarboxylate ligand that has been used in the preparation of polynuclear compounds with several metal cations. In this work, we present the in solution study of the interaction of ntp with a wide range of metal ions (M): Mn(II), Fe(II), Co(II), Ni(II), Cu(II), Zn(II), Cd(II), Pb(II) and Fe(III). The species [Mntp]−, [M(Hntp)], and [M3(Hntp)2]2+ were detected for most metal ions. In the case of Cu(II), the potentiometric model was also supported through additional thermodynamic characterization by spectroscopy. We also performed computational calculations at the UB3LYP/LANL2DZ SMD level of theory that allowed us to predict the electronic spectra of the proposed species. The computational model showed a satisfactory performance, as demonstrated by the calculated electronic spectra fit to the experimental data. A possible assembly mechanism for the reported capsule-shaped complex [Cu3(Hntp)2]2+ is proposed.

A new mononuclear complex of zinc(II), [Zn(HL)2]∙2DMF (H2L = (E)-N′-((E)-(hydroxyimino)butan-2-ylidene)salicyloylhydrazide, DMF = N,N-dimethylformamide), was prepared and characterized. Single-crystal X-ray crystallography revealed a six-coordinate zinc(II) surrounded by nitrogen of the oxime function and oxygen and distal nitrogens of the acylhydrazone group. This entity also exists in solution as demonstrated by 1H-NMR and potentiometric titrations. The computational analysis showed that the molecular orbitals involved in the main electronic transitions of the complex species in solution are centered on the ligand with negligible contribution of the metal ion. The photophysical properties of the complex were evaluated in solution and in the solid state. Luminescence studies showed that the solid has a strong emission at 550 nm with a large Stokes shift with respect to absorption. The solid state fluorescence emission is ascribed to ligand-centered and/or ligand-to-ligand charge transfer transitions, following the DFT results in solution. A comparison with a previously reported mononuclear [Zn(HL)2] allowed the investigation of the influence of DMF molecules in the structural packing and the luminescence properties.
