Single domain heavychain binders (nanobodies) obtained from camelid antibody libraries hold a great promise for immunoassay development. However, there is no simple method to select the most valuable nanobodies from the crowd of positive clones obtained after the initial screening. In this paper, we describe a novel nanobody-based platform that allows comparison of the reactivity of hundreds of clones with the labeled antigen, and identifies the best nanobody pairs for two-site immunoassay development. The output clones are biotinylated in vivo in 96-well culture blocks and then used to saturate the biotin binding capacity of avidin coated wells. This standardizes the amount of captured antibody allowing their sorting by ranking their reactivity with the labeled antigen. Using human soluble epoxide hydrolase (sEH) as a model antigen, we were able to classify 96 clones in four families and confirm this classification by sequencing. This provided a criterion to select a restricted panel of five capturing antibodies and to test each of them against the rest of the 96 clones. The method constitutes a powerful tool for epitope binning, and in our case allowed development of a sandwich ELISA for sEH with a detection limit of 63 pg/mL and four log dynamic range, which performed with excellent recovery in different tissue extracts. This strategy provides a systematic way to test nanobody pairwise combinations and would have a broad utility for the development of highly sensitive sandwich immunoassays.

<p>Turmeric (Curcuma longa L.) is an economically important food and medicinal plant that grows in tropical and subtropical regions consumed worldwide. Turmeric is a difficult matrix for residue/contaminant analysis due to the high content of potentially interfering polyphenols (curcuminoids) and essential oils with physicochemical properties similar to target analytes. To solve this problem, a simple and straightforward sample preparation protocol based on ethyl acetate extraction and dispersive solid phase clean-up, followed by gas and liquid<br />chromatography coupled to tandem mass spectrometry analysis, was optimized and validated, after selecting a<br />representative blank material using gas chromatography coupled to ion mobility spectrometry and multivariate<br />data analysis. Method validation was performed according to Codex guidelines for 67 pesticide residues, including 12 persistent organic pollutants, 4 aflatoxins and 3 dyes at 0.01, 0.02 and 0.05 mg kg</p>

<p>Affective studies are one of the most important research fields within the Sensory Evaluation area and their main objective is to determine the consumers’ acceptance of certain food products.<br />Acceptance and preference tests are widely used at the development and launching stages of new products and during market studies.<br />Usually, studies involving consumers have used 9-point hedonic scales in order to determine how much consumers like certain food products.<br />However, this methodology has some drawbacks, which have led some researchers to question its validity. The same happens with traditional<br />paired comparison tests. In this Chapter, we will gather the questions raised by various authors regarding the validity of both tests and the<br />suggestions made to improve them.</p>

A summary of the resource.
Interest in new methodologies for sensory characterization, which can be performed with panels with different degrees of training or with consumers, has largely increased in the last decade. Compared to classic Descriptive Analysis these methodologies have been applied for a limited period of time and in a small number of applications. Therefore, before new methodologies get established as standard alternatives for sensory characterization, it is necessary to develop guidelines for best practice. The aim of the present work is to provide an overview of the remaining challenges related to the implementation of new methodologies for sensory characterization and to discuss recent developments. In particular, three issues will be given special attention: understanding the cognitive processes involved in sample evaluation, development of tools for evaluating reliability, and the identification of the limitations of new methodologies. Hopefully, this work will encourage research on methodological issues of new tools for sensory characterization that contributes to the development of best practice.

A summary of the resource.
Cross-cultural research is becoming increasingly relevant in sensory and consumer science. The design of cross-cultural studies involves several methodological challenges that are not commonly faced in studies involving a single culture. However, several of these challenges have not yet received enough attention in the field, which poses several limitations to the validity and generalizability of the findings of cross-cultural studies. In this context, the aim of the present work is to review some of the most relevant methodological issues that should be considered when designing cross-cultural studies. In particular, five topics are addressed: sampling procedures, conceptual equivalence, linguistic equivalence, data collection procedures and cultural differences in response style. Suggestions and recommendations are discussed for each of the topics, which are expected to encourage greater methodological rigor and contribute to more theoretical robust cross-cultural studies in the field.

Background Individual variability is among the causes of toxicity and interruption of treatment in acute lymphoblastic leukemia (ALL) and severe non-Hodgkin lymphoma (NHL) patients under protocols including Methotrexate (MTX): 2,4-diamino-N10-methyl propyl-glutamic acid. Methods 41 Uruguayan patients were recruited. Gene polymorphisms involved in MTX pathway were analyzed and their association with treatment toxicities and outcome was evaluated. Results Genotype distribution and allele frequency were determined for SLC19A1 G80A, MTHFR C677T and A1298C, TYMS 28 bp copy number variation, SLCO1B1 T521C, DHFR C− 1610G/T, DHFR C-680A, DHFR A-317G and DHFR 19 bp indel. Multivariate analysis showed that DHFR-1610G/T (OR = 0.107, p = 0.018) and MTHFR677T alleles (OR = 0.12, p = 0.026) had a strong protective effect against hematologic toxicity, while DHFR-1610CC genotype increased this toxicity (OR = 9, p = 0.045). No more associations were found. Conclusions The associations found between gene polymorphisms and toxicities in this small cohort are encouraging for a more extensive research to gain a better dose individualization in adult ALL and NHL patients. Besides, genotype distribution showed to be different from other populations, reinforcing the idea that genotype data from other populations should not be extrapolated to ours.

A novel methanotroph, strain E10T, was isolated from a rice paddy field in Uruguay. Strain E10T grows on methane or methanol as its sole carbon and energy source. It is a Gram-negative, non-motile, non-pigmented, slightly curved rod showing type I intracytoplasmic membranes arranged in stacks. It is neutrophilic and mesophilic (optimum growth at 30-35 °C and with no growth above 37°C). The strain only possesses a particulate methane monooxygenase (pmoA). Phylogenetic analysis based on the 16S rRNA gene sequence indicated that it is most closely related to the moderately thermophilic strains Methylocaldum szegediense OR2T (91.6 % similarity) and Methylococcus capsulatus Bath (91.5 % similarity). Comparative sequence analysis of the pmoA genes also confirmed that strain E10T forms a new lineage among the genera Methylocaldum and Methylococcus (89 and 84% derived amino acid sequence identity to Methylococcus capsulatus Bath and Methylocaldum gracile VKM-14LT, respectively). The DNA G+C content is 63.1 mol% and the major cellular fatty acid is 16:0 (62.05%). Thus, it is proposed that strain E10T (= JCM 16910T=DSM 23452T) represents the type strain of a novel species within a new genus, Methylogaea oryzae gen. nov., sp. nov.

El presente trabajo propone una metodología para el aprendizaje de física moderna, principalmente para estudiantes que se encuentran cursando los dos últimos años de bachillerato de enseñanza secundaria. Enfoque constructivista del aprendizaje, en donde los estudiantes tienen un rol activo, son responsables de su proceso de construcción de conocimientos y, en donde los docentes actúan como generadores de situaciones de enseñanza para el aprendizaje. La metodología propuesta se fundamenta en el estudio del estado del arte del aprendizaje de física moderna en la enseñanza secundaria de Uruguay así como del análisis de diferentes fuentes de información: programas de cursos de física moderna de nivel terciario universitario y de formación docente y sus libros de texto más comunes, la opinión de informantes claves, programas de física y química de bachillerato de secundaria y metodologías de enseñanza con un marco conceptual coherente con el constructivismo. La propuesta metodológica incorpora experimentos que requieren responder preguntas desafiantes para estudiantes secundarios. Varios de los experimentos aquí desarrollados fueron implementados en dos cursos de tercero de bachillerato de secundaria, uno de opción científico-matemático y el otro de opción ciencias-biológicas. Los ejecutores de las propuestas fueron los docentes de estos cursos, en el científico-matemático un profesor de Química y en la opción ciencias-biológicas un profesor de Física. El análisis de los resultados de su implementación en el aula muestra que los estudiantes se ven atraídos y motivados por el desafío de responder preguntas a través de la experimentación y aprender con y del otro. Los estudiantes fueron capaces de medir cuidadosamente y analizar los datos obtenidos utilizando planillas de cálculo. Es importante destacar que, la introducción de Tecnologías de la Información y Comunicación (PC y teléfonos inteligentes, entre otros dispositivos) para realizar los experimentos propuestos, permite disponer de un laboratorio simple, económico y al alcance de todos. Permitiendo realizar experimentos con distintos grados de Metodología para el aprendizaje de Física Moderna para Educación Secundaria con Química 8 José Di Laccio (2018) sofisticación, que los estudiantes lo pueden realizar en el aula, un parque, en sus propias casas o en el lugar que lo deseen y en el momento que lo prefieran. Varios de los proyectos desarrollados en el contexto de la presente tesis fueron publicados en diversas revistas. Las propuestas educativas desarrolladas aquí, han tenido buena acogida en la comunidad educativa regional, en su presentación en diferentes congresos de enseñanza de las ciencias. Algunas de las actividades experimentales innovadoras de esta tesis, fueron premiadas en la convocatoria para presentar experiencias didácticas de Física organizada por la Universidad Nacional de Tucumán y la Asociación Física Argentina.

La hipoalfalipoproteinemia se define como un trastorno que afecta al metabolismo de las partículas de HDL, alterando su composición, concentración y el tamaño de las mismas. El término hipoalfalipoproteinemia primaria corresponde a concentraciones plasmáticas de HDL-Cpor debajo del percentil 10 con un componente genético. El bajo HDL es uno de los factores de riesgo aterogénico mejor conocidos y forma parte de las dislipemias más comunes generadoras de enfermedades cardiovasculares en la población occidental. En las últimas 6 décadas, estas enfermedades son la principal causa de muerte en nuestro país. Se sabe que existe una relación inversa entre los niveles plasmáticos de las lipoproteínas de alta densidad y el riesgo de cardiopatía isquémica. Se calcula que entre 40 y 60% de la variación en los niveles de las HDL está determinada genéticamente y existe evidencia de que son varios los genes que podrían participar en la regulación de este rasgo fenotípico. Se sabe cuáles son las causas genéticas de esta patología. La ApoAI es la apolipoproteina más importante del HDL constituyendo el 70 % de su masa proteica, siendo el gen apoAI uno de los candidatos más importantes para estudiar la variabilidad genética de los niveles de HDL. Una causa más rara de deficiencia de HDL se debe a mutaciones en el transportador de membrana ATP Binding Casette 1 (ABCA1). Esta proteína interactúa directamente con la ApoAI para promover el eflujo de colesterol celular. Otro gen candidato es el que codifica la lecitin colesterol acil transferasa (LCAT). Esta enzima es la encargada de esterificar el colesterol de las HDL para generar las partículas maduras. Estas tres moléculas, ApoAI, ABCA1 y LCAT, parecen tener un papel fundamental en el transporte reverso del colesterol desde las células periféricas hacia el hígado y otros tejidos. Nuestro objetivo ha sido elaborar una metodología que contribuya al conocimiento de los componentes genéticos de la hipoalfalipoproteinemia primaria. Esto representa un campo enorme de investigación de vital importancia para la prevención y el tratamiento de la aterosclerosis y la cardiopatía isquémica en nuestro país.

Entire seeds of Chenopodium quinoa Willd are a rich protein source and are also well-known for their high saponin content. Due to their amphiphily quinoa saponins are able to form intricate micellar aggregates in aqueous media. In this paper we study the aggregates formed by self-association of these compounds from two quinoa saponin fractions (FQ70 and FQ90) as well as several distinctive nanostructures obtained after their complexation with different ratios of cholesterol (CHOL) and phosphatidylcholine (PC). The FQ70 and FQ90 fractions were obtained by reversed-phase preparative chromatography. The structural features of their resulting aggregates were determined by Dynamic Light Scattering (DLS) and Transmission Electron Microscopy (TEM). Novel nanosized spherical vesicles formed by self-association with mean diameter about 100-200 nm were observed in FQ70 aqueous solutions whereas worm-like micelles an approximate width of 20 nm were detected in FQ90 aqueous solutions. Under experimental conditions similar to those reported for the preparation of Quillaja saponaria ISCOM matrices, tubular and ring-like micelles arose from FQ70:CHOL:PC and FQ90:CHOL:PC formulations, respectively. However, under these conditions no cage-like ISCOM matrices were observed. The saponin composition of FQ70 and FQ90 seems to determine the nanosized structures viewed by TEM. Phytolaccagenic acid, predominant in FQ70 and FQ90 fractions, is accountable for the formation of the nanosized vesicles and tubular structures observed by TEM in the aqueous solutions of both samples. Conversely, ring-like micelles observed in FQ90:CHOL:PC complexes can be attributed to the presence of less polar saponins present in FQ90, in particular those derived from oleanolic acid.<br /><br />

Carotenoids are a large group of health-promoting compounds used in many industrial sectors, such as foods, feeds, pharmaceuticals, cosmetics, nutraceuticals, and colorants. Considering the global population growth and environmental challenges, it is essential to find new sustainable sources of carotenoids beyond those obtained from agriculture. This review focuses on the potential use of marine archaea, bacteria, algae, and yeast as biological factories of carotenoids. A wide variety of carotenoids, including novel ones, were identified in these organisms. The role of carotenoids in marine organisms and their potential health-promoting actions have also been discussed. Marine organisms have a great capacity to synthesize a wide variety of carotenoids, which can be obtained in a renewable manner without depleting natural resources. Thus, it is concluded that they represent a key sustainable source of carotenoids that could help Europe achieve its Green Deal and Recovery Plan. Additionally, the lack of standards, clinical studies, and toxicity analysis reduces the use of marine organisms as sources of traditional and novel carotenoids. Therefore, further research on the processing of marine organisms, the biosynthetic pathways, extraction procedures, and examination of their content is needed to increase carotenoid productivity, document their safety, and decrease costs for their industrial implementation.

Food losses, defined as a reduction in the quantity and quality of food during production and storage, impact food safety and security. Losses caused by plant pathogens are among the most significant. Chemical pesticides have been extensively used to prevent microbial diseases. Their toxicity and reduced efficacy, however, have encouraged investigators to develop alternatives. Alternatives based on microbial biopesticides tend to be safer and more environmentally benign than conventional pesticides. In recent years, formulations based on biopesticides have progressively increased in number and diversity and have attracted commercial interest. Understanding the mechanisms by which biopesticides control the disease is fundamental to achieving optimal disease control. Biocontrol mechanisms can be divided into two main categories: those related to the ability to inhibit pathogens or their virulence factors, and those that enhance host plant fitness and induce disease resistance. Here, the first type of strategy is reviewed, which is directly mediated by physical contact between biocontrol agents and pathogens or indirectly by exposure of a pathogen to antimicrobial or microbial-inhibiting compounds produced by the microbial antagonist. Mechanisms involving physical contact include mycophagy, destruction of pathogenic bacteria by bacteriophages or predation, and disease inhibition by topical applications of specific dsRNA. Indirect mechanisms that do not involve direct contact with a pathogen include the production of antimicrobial compounds, competition, and virulence factor suppression by quorum quenching. These topics are reviewed and discussed.

This study investigated the microbial community developed in a UASB reactor for hydrogen production and correlated it to reactor performance. The reactor was inoculated with kitchen waste compost and fed with raw cheese whey at two organic loading rates, 20 gCOD/Ld and 30 gCOD/Ld. Hydrogen production was very variable, using an OLR of 30 gCOD/Ld averaged 1.0 LH2/Ld with no methane produced under these conditions. The hydrogen yield was also very variable and far from the theoretical. This low yield could be explained by selection of a mixed fermentative population with presence of hydrogen producing organisms (Clostridium, Ruminococcus and Enterobacter) and other non-hydrogen producing fermenters (Lactobacillus, Dialister and Prevotella). The molecular analysis of the raw cheese whey used for feeding revealed the presence of three predominant organisms that are affiliated with the genera Buttiauxella (a low-yield hydrogen producer) and Streptococcus (a lactic acid-producing fermenter). Although these organisms did not persist in the reactor, the continuous addition of these fermenters could decrease the reactor’s hydrogen yield.

A summary of the resource.
The origins of the biological complexity and the factors that regulate the development of community composition, diversity and richness in soil remain largely unknown. To gain a better understanding of how bacterial communities change during soil ecosystem development, their composition and diversity in soils that developed over c. 77 000 years of intermittent aeolian deposition were studied. 16S rRNA gene clone libraries and fatty acid methyl ester (FAME) analyses were used to assess the diversity and composition of the communities. The bacterial community composition changed with soil age, and the overall diversity, richness and evenness of the communities increased as the soil habitat matured. When analysed using a multivariate Bray–Curtis ordination technique, the distribution of ribotypes showed an orderly pattern of bacterial community development that was clearly associated with soil and ecosystem development. Similarly, changes in the composition of the FAMEs across the chronosequence were associated with biomarkers for fungi, actinomycetes and Gram-positive bacteria. The development of the soil ecosystem promoted the development of distinctive microbial communities that were reminiscent of successional processes often evoked to describe change during the development of plant communities in terrestrial ecosystems.

Cholesterol is a C27-sterol employed as starting material for the synthesis of valuable pharmaceutical steroids and precursors. The microbial transformations of cholesterol have been widely studied, since they are performed with high regio- and stereoselectivity and allow the production of steroidal compounds which are difficult to synthesize by classical chemical methods. In recent years, ongoing research is being conducted to discover novel biocatalysts and to develop biotechnological processes to improve existing biocatalysts and biotransformation reactions. The main objective of this review is to present the most remarkable advances in fungal and bacterial transformation of cholesterol, focusing on the different types of microbial reactions and biocatalysts, biotransformation products, and practical aspects related to sterol dispersion improvement, covering literature since 2000. It reviews the conversion of cholesterol by whole-cell biocatalysts and by purified enzymes that lead to various structural modifications, including side chain cleavage, hydroxylation, dehydrogenation/reduction, isomerization and esterification. Finally, approaches used to improve the poor solubility of cholesterol in aqueous media, such as the use of different sterol-solubilizing agents or two-phase conversion system, are also discussed.

<p>El microbioma asociado a las plantas de interés agrícola, especialmente a nivel de las raíces, juega un papel fundamental en el reciclado de nutrientes y contribuye mediante distintos mecanismos al crecimiento y salud de la planta. En el cultivo de arroz irrigado, la inundación ocasiona una disminución en el potencial redox, generándose rápidamente condiciones anaerobias en el sistema. Mientras el suelo se mantiene en condiciones anóxicas, la entrada de oxígeno a través del aerénquima a nivel de la rizósfera de la planta de arroz crea un ambiente parcialmente óxico en donde conviven procesos anaerobios y aerobios.<br />En este sistema, las bacterias desnitrificantes y las bacterias reductoras de sulfato (BRS) presentan gran relevancia ambiental, ya que participan en el reciclado de nutrientes como el carbono y el nitrógeno e intervienen en los procesos microbianos de producción y consumo de gases de efecto invernadero y de intermediarios metabólicos implicados en la emisión de estos gases.<br />En Uruguay, el arroz es uno de los principales rubros de exportación. El sistema de producción utilizado tradicionalmente en nuestro pa´ıs que se basa en la rotación del cultivo de arroz con varios años de pasturas es considerado de baja intensidad y reducido impacto ambiental, y se cree que éste ha sido la base para la obtención de los excelentes rendimientos alcanzados. La reciente intensificaci´on en los sistemas de producción arrocera puede modificar la disponibilidad de nutrientes y, por lo tanto, la composición de las poblaciones microbianas presentes tanto en suelo como asociadas a la planta.<br />En esta tesis se llevó a cabo el estudio de las comunidades microbianas de suelo y asociadas a plantas de arroz provenientes de un ensayo de campo, utilizando distintas herramientas moleculares. Se seleccionaron tres sistemas de rotación arrocera con diferente grado de intensificación y se tomaron muestras en dos etapas del desarrollo de la planta y manejo de agua. Mediante secuenciación masiva de genes 16S rRNA, se estudió la dinámica del microbioma bacteriano de suelo, rizósfera y raíces de arroz en los distintos sistemas de rotaci´on a lo largo del ciclo de cultivo, y se evaluó el efecto de la intensificación sobre estas comunidades.<br />Los resultados obtenidos en este trabajo mostraron que el microbioma asociado al cultivo de arroz se vio determinado principalmente por el nivel de influencia de la planta sobre los distintos nichos asociados a las raíces (suelo, rizósfera y endósfera) y en segundo lugar por las condiciones redox del suelo establecidas luego de la inundación. Los distintos compartimientos presentaron comunidades claramente diferentes, presentando diferentes phyla dominantes. Mientras las comunidades del suelo se vieron dominadas por bacterias de los phyla Acidobacteria (23 %), Proteobacteria (23 %) y Actinobacteria (14 %), el microbioma rizosf´erico se vio marcadamente enriquecido en proteobacterias (37 %), principalmente Beta y Deltaproteobacteria, y Bacteroidetes (9 %). La comunidad rizosférica resultó significativamente más diversa que la de suelo y endósfera de raíces, presentando la mayor abundancia relativa de la clase Deltaproteobacteria, conocida por comprender una gran variedad de bacterias anaerobias, muchas de ellas reductoras de sulfato. Por su parte, el microbioma endófito de raíces se vio ampliamente enriquecido en proteobacterias (66 %), principalmente por las clases Alpha, Beta y Gammaproteobacteria, y también Firmicutes, mayormente afiliados al orden Clostridiales. Diversas bacterias anaerobias fueron capaces de establecer interacciones fuertes y estables con la planta de arroz a lo largo del ciclo del cultivo (Bacteroidales, Clostridium, Anaeromyxobacter, Geobacter, Desulfovibrio, entre otras), algunos inclusive en etapas tempranas del cultivo previo a la inundación.<br />Mediante un análisis de especies indicadoras (IndVal) se identificaron varias OTUs fuertemente asociadas a alguna de las rotaciones arroceras, tanto en rizósfera como en el interior de las ra´ıces. Estos resultados sugieren que el microbioma asociado a las raíces de arroz podría ser susceptible a la intensificación agrícola, particularmente en taxones que no se encuentran entre los dominantes de estas comunidades.<br />Por otra parte, se optimizaron metodolog´ıas dirigidas a genes funcionales para estudiar la abundancia (qPCR) y diversidad (T-RFLP) de BRS y bacterias desnitrificantes asociados a la planta de arroz en el sistema estudiado. Tanto las BRS como las bacterias desnitrificantes fueron detectadas y cuantificadas en suelo y asociadas a las raíces de arroz como rizosféricos y endófitos, y su abundancia y distribución en los distintos nichos se vio afectadas por el manejo del cultivo y el sistema de rotaci´on empleado. Las comunidades de BRS y arqueas metanogénicas asociadas al cultivo se vieron fuertemente impactadas por la inundación, incrementando significativamente su abundancia (qPCR de genes dsrA y mcrA, respectivamente) en rizósfera y endósfera para las BRS, y en suelo y rizósfera para el caso de las metanogénicas. De acuerdo con el análisis de secuenciación masiva del gen 16S rRNA, varios grupos taxonómicos conocidos como BRS fueron encontrados formando parte del microbioma de rizósfera de la etapa inundada del cultivo, mientras que el género Desulfovibrio fue el taxón dominante en el microbioma end´ofito de raíces de arroz. Las comunidades desnitrificantes tipo nirK y nirS presentaron distinta dinámica a lo largo del ciclo y frente a las distintas rotaciones. De acuerdo a la cuantificaci´on de genes nirK y nirS realizada por qPCR, las poblaciones tipo nirK dominaron en todos los nichos estudiados frente a las tipo nirS, y su abundancia se vio afectada tanto por los cambios que se dan durante el ciclo del cultivo como por las distintas rotaciones evaluadas. Por el contrario, la abundancia de las poblaciones tipo nirS se mantuvo constante en cada uno de los nichos a lo largo del ciclo y en los diferentes sistemas de rotación. Aunque la comunidad tipo nirS endófita presentó la menor abundancia, su diversidad (evaluada mediante T-RFLP de este gen) resultó significativamente mayor a la presente en rizósfera.<br />Esta tesis aporta información relevante sobre la ecolog´ıa de las comunidades bacterianas asociadas al cultivo de arroz, y en particular, de grupos fisiológicos anaerobios fundamentales en las transformaciones biogeoquímicas de nutrientes esenciales en este ecosistema.<br />Los resultados obtenidos sugieren que los microorganismos anaerobios rizosféricos y endóitos constituyen una parte importante del microbioma asociado a la planta de arroz y podrían desempeñar un rol relevante a lo largo del ciclo de cultivo.</p>

We investigated the accumulation and toxicity of microcystin-LR (MCLR) in the digestive gland of the freshwater clam Diplodon chilensis patagonicus. Treated clams were fed with a toxic strain of Microcystis aeruginosa (NPJB1) during 6 weeks and control clams received the non-toxic strain NPDC1. Filtration rate was estimated for both groups. Toxic effects were evaluated through the hepatosomatic index (HSI) and different oxidative stress biomarkers, lipid peroxidation (content of thiobarbituric reactive substances–TBARS), protein oxidation (carbonyl groups) and reduced glutathione (GSH) levels, and enzymatic activities of superoxide dismutase (SOD), catalase (CAT) and glutathione-S-transferase (GST). The extractable MCLR measured by ELISA in digestive gland extracts showed little or no change during the first 3 weeks and increased significantly at weeks 5 and 6. HSI was reduced by 30% in treated clams at weeks 5 and 6. No significant oxidative damage to lipids or proteins was. All the antioxidant defense parameters analyzed were significantly increased at week 5 or 6. GSH increased in treated clams at week 5, reaching 62% increase at week 6. SOD, CAT and GST activities were significantly increased in treated clams by 50%, 66% and 60%, respectively, at the end of the experiment. D. chilensis patagonicus can be exposed to prolonged cyanobacterial blooms accumulating significant quantities of MCLR, which could be a risk for mammals and birds, which feed on this species and, in a lesser extent, to humans

<p>Free-range livestock can be exposed to toxic cyanobacteria by drinking water from eutrophic natural sources. This study reports on the analysis of livestock drinking water sources from ranches in an eutrophic subtropical basin of South America (33ºS, 57°W), where frequent Microcystis spp. blooms and occasionally dense scums were found. To minimise monitoring costs, microcystins (MCs) were determined in serum and water with an in-house developed enzyme linked immunosorbent assay (ELISA). The water results were compared with those of liquid chromatography – tandem mass spectrometry (LC-MS/MS) for the most common congeners (MC-LR, MC-RR; MC-YR, MC-LA). Both methods were strongly correlated along a broad range of concentrations. The concentrations of MCs in the livestock drinking water were up to 3,700 μg/L. This is about 1,000 times higher than the Australian and New Zealand livestock MCs guidance values (cattle = 4.2; sheep = 3.9 μg/L) as well as the California-EPA sub-chronic action level (3 μg/L) and implies a high risk of poisoning. The animals exposed to MCs concentrations 70–100 times higher than these guidelines showed no significant alterations in liver enzymes. Nevertheless, serum concentrations of up to 0.11 μg/L MCs were recorded by ELISA. These results reveal the potential of MCs-ELISA in serum as a sensitive, minimally invasive method for risk assessment in integrated environmental monitoring strategies.</p>
