A Gram-stain-positive bacterium, strain LZ-22T, was isolated from a rhizosphere of moss Leptobryum sp. collected at the shore of Lake Zub in Antarctica. Cells were motile, straight or pleomorphic rods with sizes of 0.6-1.0×3.5-10 µm. The novel isolate was a facultatively anaerobic, catalase-positive, psychrotolerant mesophile. Growth was observed at 3-41 °C (optimum 24-28 °C), with 0-7 % (w/v) NaCl (optimum 0.25 %) and at pH 4.0-9.0 (optimum pH 7.8). The quinone system of strain LZ-22T possessed predominately menaquinone MK-9(H4). The genomic G+C content was 70.2 mol%. Strain 10J was isolated from a biofilm of sediment microbial fuel cell, in Uruguay and had 99 % 16S rRNA gene sequence similarity to strain LZ-22T. DNA-DNA-hybridization values of 84 % confirmed that both strains belonged to the same species. Both strains grew on sugars, proteinaceous compounds, and some amino- and organic acids. Strain LZ-22T uniquely grew on D-enantiomers of histidine and valine while neglecting growth on L-enantiomers. Both strains were sensitive to most of the tested antibiotics but resistant to tested nitrofurans and sulfanilamides. Phylogenetic analyses of the 16S rRNA gene sequences indicated that the strains were related to members of the family Propionibacteriaceae (~93-94 % 16S rRNA gene sequence similarity) with formation of a separate branch within the radiation of the genera Granulicoccus and Luteococcus. Based on phenotypic and genotypic characteristics, we propose the affiliation of both strains into a novel species of a new genus. The name Raineyella antarctica gen. nov., sp. nov. is proposed for the novel taxon with the type strain LZ-22T (=ATCC TSD-18T=DSM 100494T=JCM 30886T). Algunos clústeres de espín muestran una relajación de la magnetización muy lenta a bajas temperaturas, que origina una histéresis magnética puramente molecular. Estos imanes moleculares (en inglés, single-molecule magnets, SMM) se comportan como nanopartículas magnéticas de forma y tamaño perfectamente definidos, que teóricamente podrían cumplir funciones de almacenamiento de información; presentan propiedades magnéticas únicas y son sistemas ideales para la observación de fenómenos cuánticos como el efecto túnel en la magnetización. Para que un clúster de espín se comporte como un SMM debe presentar un estado electrónico fundamental con un valor de espín (S) elevado, resultante de las interacciones magnéticas de canje entre los iones que constituyen la molécula y una anisotropía axial tipo Ising caracterizada por un valor alto y negativo del parámetro de desdoblamiento a campo cero (D).1 Dado el estado actual del conocimiento, es imposible a priori controlar tanto la nuclearidad del clúster como la naturaleza del estado fundamental o el valor resultante de anisotropía magnética. We report herein the antitumor actions of three copper(II) complexes on MG-63 human osteosarcoma cells. The three complexes: Cu-sac, Cu-gln and Cu-sac-gln (sac= saccharinate, gln= glutamine) caused a decline in cell viability. The half-maximal inhibitory concentration in MG-63 cells for Cu-sac-gln is 170 µM, showing the strongest antiproliferative effect. Moreover, only Cu-sac-gln caused a decrease of the mitochondrial activity from 100 μM. Our results indicate that the copper(II) complexes studied here produce DNA damage and suggest that the rise of reactive oxygen species (ROS) is the central mechanism action. Genotoxicity studied by the Cytokinesis-block micronucleus (MN) assay and the Single cell gel electrophoresis (comet assay) could be observed in MG-63 cells treated with Cu-sac-gln from 100 and 50 μM, respectively. Cu-sac and Cu-gln also induced DNA damage; however their effect was definitively weaker. The generation of reactive oxygen species increased from 50 μM of Cu-sac-gln and Cu-sac and only from 250 μM of Cu-gln, as well as a reduction of the GSH/GSSG ratio from 50 μM. When cells were treated with several concentrations of the complexes in addition to a combination of 50 μM of vitamin C plus 50 μM of vitamin E, a total recovery in cell survival was obtained for Cu-gln in the whole range of tested concentrations while only a partial viability recovery was obtained from 250 μM of Cu-sac and Cu-sac-gln. Overall, our results point to a differential cyto- and genotoxicity of the three copper(II) complexes and demonstrate that the complexation with both ligands confers the most potent antitumor action in human osteosarcoma cells. Although non-nutritive sweeteners have been extensively used to reduce the added sugar content of these products, increasing evidence about the existence of compensatory energy intake mechanisms makes it necessary to develop alternative strategies to achieve rapid sugar reductions. In this context, the aim of the present work was to evaluate aroma-related cross modal interactions for sugar reduction in vanilla milk desserts. In particular, the influence of increasing vanilla concentration and the joint increase of vanilla and starch concentration on consumer sensory and hedonic perception was assessed. Two studies with 100 consumers each were conducted, in which a total of 15 samples were evaluated. For each sample, consumers rated their overall liking and answered a check-all-that-apply (CATA) question comprising 12 flavour and texture terms. Sugar reduction caused significant changes in the flavour and texture characteristics of the desserts. An increase in vanilla concentration had a minor effect on their sensory characteristics. However, increasing both vanilla and starch concentration led to an increase in vanilla flavour and sweetness perception and reduced changes in consumer hedonic perception. These results showed the potential of aroma-related cross modal interactions for minimizing the sensory changes caused by sugar reduction. The effect of the addition of gelatin and starch on the rheological and sensory texture properties of sweetened plain stirred yogurt was studied. Six samples were prepared: two with gelatin (3 and 6 mg/g), three with starch (1, 5 and 10 mg/g) and a control sample without thickener. Rheological measurements were performed and flow curves were modelled according to the Herschel–Bulkley rheological model. Also, sensory evaluation was carried out using a trained panel. The addition of gelatin and starch significantly affected the instrumental texture parameters (Herschel–Bulkley's model parameters and syneresis) and the perceived sensory texture of yogurts. All sensory descriptors were well predicted by the partial least squares regression of the instrumental parameters. The addition of thickeners significantly reduced syneresis. Samples produced with 6 mg/g of gelatin did not show syneresis and showed the highest sensory viscosity, creaminess and mouth-feel. Therefore, the use of 6 mg/g of gelatin could be a reasonable concentration to use in the production of low-fat yogurts. Interest in consumer-based methodologies for sensory characterization has largely increased in the last decade. However, research on their applicability and reliability is still necessary. In this context, the aim of the present work was to compare three consumer-based methodologies and descriptive analysis for sensory characterization, considering as case study four sets of orange-flavoured powdered drinks, differing in the number of samples and the extent to which they represented the whole product category. A trained assessor panel evaluated sample sets using descriptive analysis, while groups of 100 consumers evaluated samples using one of three methodologies: CATA questions, projective mapping (PM) and polarized sensory positioning (PSP). Across the four sample sets, the three-consumer based methodologies provided similar information regarding the main similarities and differences among samples, which did not largely differ from that obtained using descriptive analysis. The main difference among the methodologies was related to the relative importance given to the sensory characteristics when evaluating differences among samples, which led to differences in the dimensionality of the sensory spaces. Sample configurations from PM and PSP required more dimensions than those from descriptive analysis and CATA questions to fully explain differences among samples. None of the consumer-based methodologies outperformed descriptive analysis in terms of ability to discriminate samples, being CATA questions the methodology that showed the lowest discrimination. Los colorantes sintéticos son ampliamente utilizados en la industria textil. La presencia de estos compuestos recalcitrantes en los efluentes industriales representa un problema ambiental si no se realiza un tratamiento previo para su eliminación antes de su vertido en los cursos de agua. Estos xenobióticos presentan una elevada resistencia a la degradación química y/o biológica, causan la disminución de entrada de luz en ecosistemas acuáticos retardando la fotosíntesis y modifican la solubilidad de gases en el agua afectando el desarrollo de organismos acuáticos. Adicionalmente, muchos son tóxicos, carcinógenos o mutagénicos. La aplicación de lacasas en el tratamiento enzimático de colorantes utilizados en la industria textil surge como alternativa a los tratamientos con métodos tradicionales, los cuales no siempre son ambientalmente amigables, accesibles desde el punto de vista económico, ni completamente efectivos. Estas oxidorreductasas son muy versátiles debido a su baja especificidad, pueden oxidar diversos compuestos fenólicos y aminas aromáticas, así como compuestos más complejos utilizando mediadores redox como co-sustrato. En esta Tesis se trabajó con dos lacasas producidas por cepas nativas de los basidiomicetes Trametes villosa y Pycnoporus sanguineus aislados de zonas forestadas con Eucalyptus globulus en Uruguay. Se optimizó la producción de ambas enzimas en un medio de cultivo semi-sólido utilizando como sustrato un sub-producto de la industria forestal y las mismas fueron purificadas y caracterizadas. Estas lacasas presentaron una alta capacidad de degradación de distintos colorantes sintéticos, incluso en algunos casos sin necesidad de un mediador redox. Los estudios realizados representan una contribución importante al área ya que no se ha reportado previamente el tratamiento de efluentes coloreados con lacasas producidas por hongos de colecciones locales. Por otro lado, dadas las ventajas que ofrecen los procesos de inmovilización de proteínas, las lacasas fueron inmovilizadas para la obtención de nuevos biocatalizadores insolubles. La inmovilización hace posible la separación del derivado enzimático de los productos de reacción así como la disminución de los costos globales del proceso al permitir el re-uso del biocatalizador. Como método de inmovilización se seleccionó el covalente reversible. El mismo combina una unión fuerte que evita la Resumen XX pérdida de enzima durante la aplicación del biocatalizador insoluble y posibilita regenerar el soporte por reversibilidad de la unión, lo cual contribuye a la reducción del costo global del proceso. Es importante remarcar que el desarrollo de un biocatalizador insoluble en base a lacasa inmovilizada en el soporte tiolsulfinato-agarosa es totalmente novedoso y de gran potencial para futuras aplicaciones biotecnológicas. En particular, con el derivado obtenido por inmovilización de lacasa de T. villosa se logró alcanzar altos rendimientos de degradación de colorantes tipo azoico y antraquinónico, los cuales representan las principales clases de colorantes empleados a nivel industrial. Por último, se debe tener en cuenta que si bien la disminución del color del efluente puede ser el principal objetivo de los tratamientos realizados en la industria no debe ser el único, ya que se debe considerar asimismo la toxicidad de los productos obtenidos. En este sentido, los estudios realizados aportan información significativa acerca de la ecotoxicidad de dos colorantes azoicos (Acid Red 88 y Acid Black 172) tratados con el sistema lacasa-mediador. De esta forma, el trabajo realizado contribuye al desarrollo de técnicas para el tratamiento de efluentes industriales coloreados que contemplen diferentes aspectos relacionados a la preservación del medio ambiente. La digestión anaerobia (DA) es una biotecnología que tiene por objetivo la degradación de materia orgánica y la producción de metano. El proceso es llevado a cabo por un consorcio de microorganismos anaerobios. La DA ha tenido un gran impacto en el tratamiento de aguas residuales con alto contenido en materia orgánica y residuos urbanos, ya que además de tratar el efluente se obtiene energía. En nuestro país se está aplicando esta tecnología en algunas industrias dentro del sector lácteo y de bebidas, entre otros. Si bien la DA es una tecnología consolidada y con gran aplicación, aún se conoce poco la microbiología de estos sistemas en escala real y en particular hay muy pocos estudios de la microbiología de reactores durante su período de arranque. Aprovechando la instalación y puesta en marcha de dos reactores anaerobios en industrias de nuestro país surge esta tesis, en la cual se planteó entre otros objetivos, conocer la dinámica de las comunidades microbianas presentes en dos reactores UASB a escala real durante el período de “arranque”. Ambos reactores fueron inoculados con lodos provenientes de una laguna anaerobia, uno de ellos trató vinaza, generada en la producción de bioetanol a partir de caña de azúcar y el otro efluente lácteo, rico en grasas. Se estudió la comunidad microbiana en muestras de biomasa colectadas durante aproximadamente un año. Se realizaron análisis fisicoquímicos y se analizó la comunidad microbiana por secuenciación masiva el gen del rARN de 16S procariota. Para estudiar la evolución de la población metanogénica se realizaron actividades metanogénicas y se cuantificó por PCR cuantitativo el gen del rARN de 16S de arqueas y el gen mcrA, un gen funcional marcador de la metanogénesis. Los resultados muestran que en términos generales el efecto del tipo de efluente sobre la selección de los grupos de microorganismos -que luego se volvieron dominantes- fue el fenómeno más evidente. En lo que respecta a los consorcios bacterianos, en ambos reactores dominaron los mismos filos, aunque en distintas proporciones. Los filos Chloroflexi, Firmicutes, Proteobacteria, Synergistetes y Verrucomicrobia, junto con el cand. phyl, Hyd 24-12 fueron los grupos dominantes luego de la adaptación al efluente lácteo. Los Clostridiales fueron un grupo resistente y los Synergistetes fueron un grupo resiliente frente a una perturbación fuerte en el pH del reactor que trató efluente lácteo. En el reactor de vinaza los filos Firmicutes, cand. phyl. Hyd 24-12 y Verrucomicrobia demostraron ser resistentes a la vinaza y a las cargas orgánicas aplicadas. Por otra parte, la detención de la alimentación del reactor durante dos períodos de aproximadamente cinco meses no afectó a la comunidad microbiana que permaneció en un estado de latencia. Otro objetivo de esta tesis fue determinar cuáles son los grupos de metanogénicos predominantes en reactores a escala real. Este objetivo se fijó para revisar un postulado en la DA en el cual se estableció que el 70 % de la metanogénesis ocurre por la vía acetoclástica y el 30 % por el consumo de H2 y CO2. Aunque, datos preliminares de actividades viii metanogénicas realizadas en algunos reactores nacionales sugerían lo contrario. En esta parte, se analizaron dos reactores UASB a escala real de la industria láctea, uno de los cuales se encontraba durante su etapa de arranque y el otro durante su 8vo año de operación estable. Se colectaron muestras de biomasa durante aproximadamente un año y se cuantificaron los diferentes grupos de metanogénicos mediante PCR cuantitativo utilizando primers y sondas específicos para el gen del rARN de 16S de los diferentes grupos. Además se realizaron ensayos de actividad metanogénica a partir de acetato y a partir de H2 y CO2. Los resultados moleculares mostraron que las arqueas hidrogenotróficas fueron las dominantes. Estos resultados fueron concordantes con los resultados de las actividades metanogéncias que muestran una mayor actividad hidrogenotrófica. Los Methanobacteriales fueron el orden hidrogenotrófico dominante, seguido del orden Methanosarcinales. Si bien esto no es una demostración que pueda generalizarse a todos los reactores a escala real, es evidencia de que la dominancia de los acetoclásticos no sería un paradigma sino que se cumpliría en determinadas condiciones ambientales. Conocer las dinámicas de las comunidades microbianas durante períodos tan particulares como la etapa arranque brinda herramientas para poder mejorar dichos tiempos. Esto a su vez impacta en la reducción de costos y por lo tanto en la competitividad económica de la digestión anaerobia frente a otras tecnologías de tratamiento de residuos. The horn fly, Haematobia irritans (L.), is a blood-sucking ectoparasite that is responsible for sizeable economic losses in livestock. The salivary gland products facilitate blood intake. Taking advantage of the identification of novel H. irritans salivary antigens (Hematobin, HTB and Irritans 5, IT5), we investigated the parasite loads, H. irritans blood intake, and antibody response of naturally infected bovines during the fly season. Fly loads and fly hemoglobin content fluctuated during the trial. Each time horn fly loads exceeded 200 flies per cattle, a reduction in horn fly blood intake was observed three weeks later. All of the cattle elicited an antibody response against HTB and IT5 that declined once the fly season was over. Cattle anti-IT5 titers were positively correlated with parasite loads and negatively correlated with fly blood intake. These results suggest that the natural changes in the H. irritans blood intake observed in this study were associated with a natural host response against horn fly salivary antigens. The effect of lactose hydrolysis on the sensory profile of dulce de leche and consumer reaction to this process using overall acceptability data and survival analysis calculations was examined. Lactose hydrolysis caused marked changes in the sensory profile of dulce de leche, particularly for flavor and texture, leading to a decrease in consumer overall acceptability and intent to purchase the product. Survival analysis methodology was used to estimate the consumer rejection percentage to purchase dulce de leche as a function of lactose hydrolysis percentage. Using a Weibull distribution, lactose hydrolysis percentage corresponding to a 10% consumer rejection was calculated as 4.6 ± 0.4%, in agreement with calculations from least significant difference in overall acceptability, suggesting that both methodologies might provide similar results. Survival analysis could be used to estimate limits for ingredient replacement or changes in manufacturing processes that cause complex sensory changes in the product, leading to consumer rejection.
