Human Mn-containing superoxide dismutase (hMnSOD) is a mitochondrial enzyme that metabolizes superoxide radical (O2(•-)). O2(•-) reacts at diffusional rates with nitric oxide to yield a potent nitrating species, peroxynitrite anion (ONOO(-)). MnSOD is nitrated and inactivated in vivo, with active site Tyr34 as the key oxidatively modified residue. We previously reported a k of &sim;1.0 × 10(5) M(-1) s(-1) for the reaction of hMnSOD with ONOO(-) by direct stopped-flow spectroscopy and the critical role of Mn in the nitration process. In this study, we further established the mechanism of the reaction of hMnSOD with ONOO(-), including the necessary re-examination of the second-order rate constant by an independent method and the delineation of the microscopic steps that lead to the regio-specific nitration of Tyr34. The redetermination of k was performed by competition kinetics utilizing coumarin boronic acid, which reacts with ONOO(-) at a rate of &sim;1 × 10(6) M(-1) s(-1) to yield the fluorescence product, 7-hydroxycoumarin. Time-resolved fluorescence studies in the presence of increasing concentrations of hMnSOD provided a k of &sim;1.0 × 10(5) M(-1) s(-1), fully consistent with the direct method. Proteomic analysis indicated that ONOO(-), but not other nitrating agents, mediates the selective modification of active site Tyr34. Hybrid quantum-classical (quantum mechanics/molecular mechanics) simulations supported a series of steps that involve the initial reaction of ONOO(-) with Mn(III) to yield Mn(IV) and intermediates that ultimately culminate in 3-nitroTyr34. The data reported herein provide a kinetic and mechanistic basis for rationalizing how MnSOD constitutes an intramitochondrial target for ONOO(-) and the microscopic events, with atomic level resolution, that lead to selective and efficient nitration of critical Tyr34. BACKGROUND: The search for novel chemical entities targeting essential and parasite-specific pathways is considered a priority for neglected diseases such as trypanosomiasis and leishmaniasis. The thiol-dependent redox metabolism of trypanosomatids relies on bis-glutathionylspermidine [trypanothione, T(SH)2], a low molecular mass cosubstrate absent in the host. In pathogenic trypanosomatids, a single enzyme, trypanothione synthetase (TryS), catalyzes trypanothione biosynthesis, which is indispensable for parasite survival. Thus, TryS qualifies as an attractive drug target candidate. METHODOLOGY/PRINCIPAL FINDING: A library composed of 144 compounds from 7 different families and several singletons was screened against TryS from three major pathogen species (Trypanosoma brucei, Trypanosoma cruzi and Leishmania infantum). The screening conditions were adjusted to the TryS´ kinetic parameters and intracellular concentration of substrates corresponding to each trypanosomatid species, and/or to avoid assay interference. The screening assay yielded suitable Z' and signal to noise values (≥0.85 and ~3.5, respectively), and high intra-assay reproducibility. Several novel chemical scaffolds were identified as low μM and selective tri-tryp TryS inhibitors. Compounds displaying multi-TryS inhibition (N,N'-bis(3,4-substituted-benzyl) diamine derivatives) and an N5-substituted paullone (MOL2008) halted the proliferation of infective Trypanosoma brucei (EC50 in the nM range) and Leishmania infantum promastigotes (EC50 = 12 μM), respectively. A bis-benzyl diamine derivative and MOL2008 depleted intracellular trypanothione in treated parasites, which confirmed the on-target activity of these compounds. CONCLUSIONS/SIGNIFICANCE: Novel molecular scaffolds with on-target mode of action were identified as hit candidates for TryS inhibition. Due to the remarkable species-specificity exhibited by tri-tryp TryS towards the compounds, future optimization and screening campaigns should aim at designing and detecting, respectively, more potent and broad-range TryS inhibitors. Gastric cancer (GC) is the third most common cause of cancer death worldwide. Natural killer cells play an important role in the immune defense against transformed cells. They express the activating receptor NKG2D, whose ligands belong to the MIC and ULBP/RAET family. Although it is well established that these ligands are generally expressed in tumors, the association between their expression in the tumor and gastric mucosa and clinical parameters and prognosis of GC remains to be addressed. In the present study, MICA and MICB expression was analyzed, by flow cytometry, in 23 and 20 pairs of gastric tumor and adjacent non-neoplasic gastric mucosa, respectively. Additionally, ligands expression in 13 tumors and 7 gastric mucosa samples from GC patients were evaluated by immunohistochemistry. The mRNA levels of MICA in 9 pairs of tumor and mucosa were determined by quantitative PCR. Data were associated with the clinicopathological characteristics and the patient outcome. MICA expression was observed in 57% of tumors (13/23) and 44% of mucosal samples (10/23), while MICB was detected in 50% of tumors (10/20) and 45% of mucosal tissues (9/20). At the protein level, ligand expression was significantly higher in the tumor than in the gastric mucosa. MICA mRNA levels were also increased in the tumor as compared to the mucosa. However, clinicopathological analysis indicated that, in patients with tumors >5 cm, the expression of MICA and MICB in the tumor did not differ from that of the mucosa, and tumors >5 cm showed significantly higher MICA and MICB expression than tumors ≤5 cm. Patients presenting tumors >5 cm that expressed MICA and MICB had substantially shorter survival than those with large tumors that did not express these ligands. Our results suggest that locally sustained expression of MICA and MICB in the tumor may contribute to the malignant progression of GC and that expression of these ligands predicts an unfavorable prognosis in GC patients presenting large tumors. La Química de Coordinación permite el desarrollo de compuestos variados desde el punto de vista estructural y de la reactividad. En este trabajo se estudió el efecto de diferencias estructurales en complejos ternarios de cobre(I) y cobre(II) en el comportamiento de los mismos en solución acuosa, así como la modulación de sus propiedades fisicoquímicas, de los modos de interacción con el ADN (como molécula blanco para la acción terapéutica) y la actividad citotóxica in vitro. Tomando como base el núcleo cobre‐fenantrolina, con alta actividad antitumoral in vitro, se eligieron ligandos diimínicos aromáticos relacionados a la fenantrolina y coligandos adecuados para la obtención de compuestos heterolépticos de cobre(I) y cobre(II), ligandos aniónicos derivados del iminodiacetato para el primero y trifenilfosfina para el segundo. De forma de determinar el efecto del núcleo cobre‐diimina sobre las propiedades y la actividad biológica de los complejos se sintetizó compuestos homolépticos cobre(II)‐diimina. Los compuestos fueron caracterizados en estado sólido mediante análisis elemental, espectroscopía infrarroja, difracción de rayos X, espectroscopía de resonancia paramagnética electrónica, termogravimetría y calorimetría diferencial de barrido. Los compuestos de cobre(I) obtenidos presentan geometrías tetraédricas cuyo grado de distorsión depende del ligando diimínico. Los compuestos de cobre(II) presentan geometrías que varían desde octaédrica y pirámide de base cuadrada con diferentes grados de distorsión. El comportamiento de los compuestos en solución acuosa se estudió mediante conductimetría, espectroscopía electrónica y de resonancia paramagnética electrónica. Todos los compuestos sufren disociación parcial, observándose que para la mayoría la especie predominante en solución es la ternaria neutra observada en estado sólido. La lipofilia es una magnitud de interés en compuestos para aplicación biológica. Los compuestos obtenidos abarcan un rango amplio de lipofilia. Ésta se encuentra modulada tanto por el ligando diimínico y el coligando, observándose una mayor dependencia respecto al segundo para los complejos de cobre(II). Todos los compuestos obtenidos se unen al ADN. Los compuestos de cobre(I) causan hidrólisis de la biomolécula mientras que los compuestos de cobre(II) producen cambios conformacionales relacionados con procesos de intercalación parcial o unión a los surcos. Los cambios conformacionales más observados para estos compuestos son de la forma B del ADN a la C, Z y A dependiendo del ligando diimínico. Los estudios de actividad citotóxica in vitro de los compuestos de cobre(I) obtenidos, cobre(II)‐diimina y una de las familias de compuestos ternarios de cobre(II) muestran que se trata de potentes agentes citotóxicos. Sensory evaluation has traditionally been divided into two clearly defined areas: analytical tests, aimed at objectively evaluating the sensory characteristics of products, and hedonic tests, in which consumers evaluate their acceptance/preference. One of the central dogmas of the field has been matching these two types of tests to different types of assessors respectively: selected and trained assessors and regular consumers of the target products. For years, consumers have been regarded as not capable of performing analytical tasks. However, the development of various alternative methods for sensory characterization in the last couple of decades has agitated the debate about the use of untrained assessors for analytical tasks in sensory science. Lately, the line between trained and consumer panels for analytical tests has blurred and is expected to continue to do so. The present opinion paper discusses some of the most relevant issues around the debate of whether consumer or trained assessor panels are appropriate for analytical testing in specific application and provides recommendations for practitioners on this respect. A pineapple vinification process was conducted through inoculated and spontaneous fermentation to develop a process suitable for a quality beverage during two successive vintages in Huambo, Angola. Wines obtained with the conventional Saccharomyces cerevisiae strain, were analysed by gas chromatography, and a total of 61 volatile constituents were detected in the volatile fraction and 18 as glycosidically bound aroma compounds. Concentration levels of carbonyl and sulphur compounds were in agreement with the limited information reported about pineapple fruits of other regions. We report, for the first time in pineapple wines, the presence of significant concentrations of lactones, ketones, terpenes, norisoprenoids and a variety of volatile phenols. Eight native yeast strains were isolated from spontaneous batches. Further single-strain fermentations allowed us to characterise their suitability for commercial fermentation. Three native strains (Hanseniaspora opuntiae, H. uvarum and Meyerozyma guilliermondii) were selected with sensory potential to ferment pineapple fruits with increased flavour diversity. Results obtained here contribute to a better understanding of quality fermentation alternatives of this tropical fruit in subtropical regions. A novel automated method for the determination of boron based on the use of pulsed flows was developed and applied to the determination of this element in samples of tap water, ocean water and eye drops. The method was implemented by means of a multi-pumping system consisting of three solenoid micropumps and a photometric detector and exploits the reaction of azomethine-H in the presence of boron. The system runs under control of an open-source microcontroller. The main operational parameters were optimized. Given the particular kinetics of the reaction, a stopped-flow period (1 or 5 min) was included to allow for color development. The method presents linearity in the range 0.35–3.0 mg L−1, good precision (sr<3%), and detection and quantification limits of 0.10 and 0.35 mg L−1 respectively. Samples of tap water or eye drops could be successfully analyzed employing a 1-minute stop time, providing a maximum sampling frequency of 32 samples h-1. In order to overcome matrix effect caused by the high saline concentration, ocean water samples required stop times of 5 min, providing a sampling frequency of 10 samples h-1. Recoveries of 102% (eye drops), 94% (drinking water) and 93% (ocean water) were obtained. The method was considered accurate and fit for the purpose. Molybdenum is an essential trace element of biological and environmental relevance. Monitoring its content in water sources is very important from the standpoint of health and environment. Given the very low concentrations of Mo in water, it is necessary to use highly sensitive techniques such as ICP-MS or elaborate methods including preconcentration prior to detection. In this work, a new method for the determination of molybdenum in groundwater was developed, incorporating a preconcentration step based on dispersive liquid–liquid microextraction (DLLME) prior to detection by electrothermal atomic absorption spectrometry (ETAAS). After the injection of a ternary mixture formed by an aqueous solution of the complexing agent (potassium ethyl xanthate) and extraction and dispersion solvents (carbon tetrachloride, acetonitrile), the formation of the Mo–xanthate complex occurs followed by its extraction in fine droplets of carbon tetrachloride. After separating the phases the molybdenum content is measured by ETAAS. Several experimental parameters were optimised and the method was validated, presenting a detection limit (3s/b) of 0.017 μg L−1, a quantification limit (10s/b) of 0.062 μg L−1, and an enrichment factor of 165 for a sample volume of 6.0 mL. The calibration curve was linear in the range of 0.06–1.1 μg L−1, the precision (repeatability, sr (%), n = 10) was 2.98% at 0.1 μg L−1 and the recovery of spiked samples was 98.5–103%. This method was successfully applied to the determination of molybdenum in groundwater and the results were found to be equivalent to those obtained by ICP-MS. Temporal Dominance of Sensations (TDS) and Temporal Check-all-that-apply (TCATA) are two multi-attribute methods for dynamic sensory characterization. Previous research has shown that both methodologies provide complementary information. However, it remains an open question which of the two approaches better explains consumers' hedonic perception of products. In this context, the aim of the present work was to compare TDS and TCATA in terms of their ability to identify the influence of the dynamic sensory profile of food products on consumer overall liking scores. Two consumer studies were conducted using two different product categories (French bread and vanilla milk desserts). In each study, a between-subjects design was used to obtain dynamic sensory profiles using TDS and TCATA. After the dynamic sensory characterization tasks consumers rated their liking using a 9-point hedonic scale. Across the two studies, both methodologies provided similar information on the main drivers of liking and disliking, particularly when samples showed clear differences in liking. However, in one of the studies attribute applicability from TCATA provided additional insights on the influence of the dynamics of the sensory characteristics of products on consumers' liking. Results of the present work stress the complementarity between TCATA and TDS and highlight the potentiality of TCATA to provide a more detailed description of the dynamics of sensory perception during consumption. Methodologies that identify ways products differ from consumers’ ideal are commonly used to guide innovation. In this research the use of CATA questions for this purpose was compared to JAR scales, which are well established for use in product optimization efforts. Two CATA variants were considered: (i) CATA questions including terms with hedonic-intensity connotations (e.g., not enough sweet, much too sweet), and (ii) CATA questions pertaining to both the tasted and the ideal products. In six consumer studies (n = 939), spanning multiple product categories and consumer populations (Uruguay, New Zealand and USA), it was found that CATA questions and JAR scales provided similar insights regarding the most relevant deviations from ideal. However, several differences were also identified. In particular, CATA questions tended to identify fewer deviations than did JAR scales, especially when terms with hedonic-intensity connotations were used and when differences between samples were small. This difference is likely linked to facets of the two methodologies: only applicable terms are selected when using CATA questions, whereas responses must be provided for each JAR scale included in a study. Besides, the fact that only the extreme anchors of the JAR scale were included in the CATA question could have encouraged consumers not to indicate deviations from the ideal. Penalty analysis, performed using Partial-Least Squares (PLS) regression identified several significant deviations from the ideal. While the two methodologies established the same main differences, JAR scales identified more significant deviations from the ideal than CATA questions. Although results confirmed the potential for the use of CATA questions in product optimization research, careful consideration of purpose of the research and attention to terms included in the CATA questions is recommended. Purine transporters as substrate entry points in organisms, are involved in a number of cellular processes such as nitrogen source uptake, energy metabolism and synthesis of nucleic acids. In this study, two nucleobase transporter genes (phZ, phU) from Phanerochaete chrysosporium were cloned, identified, and functionally characterized. Our results show that PhZ is a transporter of adenine and hypoxanthine, and a protein belonging to the AzgA-like family, whilst PhU belongs to the NAT/NCS2 family, transporting xanthine and uric acid. No other sequences belonging to these families were detected in P. chrysosporium's genome. Phylogenetic analyses show that AzgA-like sequences form monophyletic groups for each major lineage (Ascomycota, Basidiomycota and Zygomycota). In contrast, Ascomycota and Basidiomycota NAT/NCS2 sequences do not form monophyletic groups and several copies of this protein are distributed across the tree. Expression of phU was significantly downregulated in the presence of a primary source like ammonium, and enhanced if purines were present or if the mycelium was nitrogen starved. phZ was clearly induced by its substrates (hypoxanthine, adenine), very lightly induced by xanthine, suppressed by urea and amino acids and expressed at a basal level when uric acid or ammonium was the nitrogen source or when the mycelium was starved for nitrogen. In order to perform substrate analyses, both P. chrysosporium proteins (PhZ, PhU) were expressed in Aspergillus nidulans. Epifluorescent microscopy showed that under inducing conditions, PhZ-GFP and PhU-GFP were present at the plasma membrane of A. nidulans transformed strains, and were internalized in repressed conditions. Our results suggest that in the white-rot fungus P. chrysosporium, phU has a catabolic role and phZ, (less dependent of the nitrogen source), plays a key role in purine acquisition to provide biosynthetic components. The use of inorganic nanoparticles as electron acceptors in hybrid solar cells has been increased. Some advantages compared to electron acceptors based on organic materials, are better electron transfer and lower production costs. We report the synthesis of ββ-HgS nanoparticles by a combination of solution method using polyvinylpyrrolidone (PVP) as stabilizing agent, and hydrothermal treatment to diminish nanoparticles size and improve size distribution. The combination of a bottom-up and a top-down technique proved to be an appropriate method to obtain HgS nanoparticles with sizes in the order of the Bohr radius, essential for charge carrier generation in the solar cell. Additionally, PVP was replaced by hexadecanethiol (HDT). This exchange allowed the spin coating of a thin film of ββ-HgS dispersed in poly(3-hexylthiophene) (P3HT) onto a glass substrate. Characterizations showed that the HgS nanoparticles and the polymer were homogeneously mixed, but several aggregated HgS nanoparticles were still present. The lowering in the photoluminescence signal of the layer is an indicative of charge carrier transfer between the nanoparticles and P3HT. Our results are encouraging for the development of hybrid solar cells using ββ-HgS nanoparticles and P3HT. To our knowledge, these results are the first obtained for ββ-HgS nanoparticles, using a bottom-up/top-down approach. The present study examines the feasibility of producing hydrogen by dark fermentation using unsterilised cheese whey in a UASB reactor. A lab-scale UASB reactor was operated for more than 250 days and unsterilised whey was used as the feed. The evolution of the microbial community was studied during reactor operation using molecular biology tools (T-RFLP, 16S rRNA cloning library and FISH) and conventional microbiological techniques. The results showed that hydrogen can be produced but in low amounts. For the highest loading rate tested (20 gCOD/L.d), hydrogen production was 122 mL H2/L.d. Maintenance of low pH (mean = 5) was insufficient to control methanogenesis; methane was produced concomitantly with hydrogen, suggesting that the methanogenic biomass adapted to the low pH conditions. Increasing the loading rate to values of 2.5 gCOD/gVSS.d favoured hydrogen production in the reactor. Microbiological studies showed the prevalence of fermentative organisms from the genera Megasphaera, Anaerotruncus, Pectinatus and Lactobacillus, which may be responsible for hydrogen production. However, the persistence of methanogenesis and the presence of other fermenters, not clearly recognised as hydrogen producers indicates that competition for the substrate may explain the low hydrogen production. The harmonic and fundamental vibrational frequencies, rotational constants, vibration–rotation corrections and Zero point energies of HSO and SOH and their deuterated isomers (DSO and SOD) have been determined employing the CCSD(T) methodology in conjunction with the aug-cc-pV(X+d)Z and cc-pwCVQZ basis sets. The calculated fundamental frequencies of SOH are 830, 1150 and 3577 cm−1, for the υSO, υbend and υOH, respectively. In the case of HSO the computed fundamentals are 1002, 1077 and 2335 cm−1, for the υSO, υbend and υSH, respectively. The values are discussed in terms of the experimental determinations available. The rotational constants reported for HSO are in reasonable agreement with experiment; the computed values are 301,271, 20,557 and 19,192 MHz. In the case of SOH, for the first time, we report the rotational constants including vibration–rotation corrections, they are: 654,236, 16,621, 16,178 MHz. The force fields calculated allowed as to estimate accurate ZPEs, the suggested values are 6.48 and 8.19 kcal/mol for HSO and SOH, respectively. Finally, we recommend the following structural parameters for HSO rSO = 1.4924 Å, rSH = 1.3649 Å and &ang;HSO = 104.76°; whereas for SOH we recommend rSO = 1.6302 Å, rOH = 0.9629 Å and &ang;SOH = 107.97°. Herein, we report on a comparative study of the addition of hydrogen to graphene. The binding energies and structural parameters determined using Siesta are in excellent agreement with those calculated employing more sophisticated and computational expensive plane waves codes. The C–H binding energy between hydrogen and a graphene sheet containing Stone–Wales defects is 1.81 eV, twice as much the value computed for a perfect sheet. The single addition of hydrogen to this defect induces an important roughness on the graphene sheet. Thus, we suggest that the Stone–Wales defects are likely to be the sites where the hydrogenation of graphene originates. Finally, we have approximately estimated the free energy correction for the addition of hydrogen to graphene, with a value of 0.5 eV. The free energy change for the single H addition to perfect graphene at 298 K is spontaneous by 0.39 eV. Interventions aimed at changing dietary patterns should be designed based on the main motives underlying the food choices of specific target populations. The aim of the present study was to identify motives underlying food choice and barriers to healthy eating among consumers in two socioeconomic levels in Uruguay. Eleven focus groups were carried out with a total of 76 participants. Six of the groups involved low income participants and the others were conducted with middle income participants. Discussions were held around frequently consumed products, motives underlying food choices and barriers to healthy eating. Results confirmed the strong influence of income level on motives underlying food choice and barriers to the adoption of healthy eating. Low income participants described their choices as mainly driven by economic factors and satiety, whereas convenience was the main determinant of food selection for middle income participants. Implications for the design of public policies targeted at each group are discussed. The development of strategies to reduce sugar intake of children and adolescents has been deemed necessary to cope with the global obesity epidemic. In this context, the aims of the present work were to study attitudes towards sugar reduction in three dairy products of children and adolescents of two income levels and to assess if these attitudes were modulated by the inclusion of the traffic light system on labels. A total of 646 children and adolescents from three primary/secondary schools in Montevideo (Uruguay) participated in the study. A total of twelve labels were designed following a full factorial design with three variables: dairy product (3 levels: yogurt, chocolate-flavoured milk and vanilla milk dessert), sugar reduction claim (2 levels: absent and present "20% less sugar") and traffic light system (2 levels: absent and present). Children had to evaluate their expected liking of the labels and to answer a check-all-that-apply question composed of terms related to tastiness, healthfulness and fun. Sugar claim significantly increased children and adolescents' expected liking scores (p=0.01), suggesting they had a positive attitude towards sugar reduction in the evaluated dairy products. However, the influence of sugar reduction claim on hedonic expectations and healthfulness perception was modulated by the inclusion of the traffic-light system on labels (p=0.01). This front-of-pack nutrition labelling scheme increased their unhealthfulness perception of products with high sugar content and could potentially discourage their consumption. Age and income modulated the influence of these strategies on hedonic and healthfulness perception (p<0.05). El presente proyecto tiene como objetivo optimizar las metodologías analíticas para evaluar la presencia de metales y semimetales tóxicos en la matriz cabello a los efectos de ser utilizado como matriz biológica para la determinación de elementos tóxicos, evaluando su desempeño como posibles biomarcadores de exposición en población presuntamente expuesta. Para ello, en primera instancia, se puso a punto una metodología para el monitoreo y tratamiento de tóxicos metálicos y semimetálicos en una matriz compleja como lo es el cabello. Se optimizaron las metodologías para el tratamiento de la muestra y para las determinaciones de los elementos en estudio se utilizó la técnica de espectroscopía de absorción atómica. Se logró desarrollar un protocolo de digestión para determinación de cuatro elementos, plomo, mercurio, arsénico y manganeso, en forma simultánea y su posterior validación. Luego se realizó un estudio en población presuntamente expuesta a los tóxicos estudiados en el asentamiento Aquiles Lanza, con un enfoque basado en los pilares de investigación de la Ecosalud: transdisciplinaridad, participación social, pensamiento sistémico, equidad social y de género, sustentabilidad y acción. En el estudio poblacional, se encontraron niveles de plomo cuantificables, mientras que del resto de los analitos, si bien se detectaron, los niveles estuvieron por debajo de los límites de cuantificación. Del análisis estadístico, surge que podrían existir correlaciones con el sexo de los niños entre otras variables estudiadas. El estudio realizado, ha contemplado una integración global, utilizando los principios de la Ecosalud.
