A summary of the resource.
Chitooligosaccharides possessing remarkable biological properties can be obtained by enzymatic hydrolysis of chitin. In this work, the chitosanase activity of soluble and immobilized glycosyltransferase (Branchzyme) toward chitosan and biochemical characterization are described for the first time. This enzyme was found to be homotetrameric with a molecular weight of 256 kDa, an isoelectric point of 5.3, and an optimal temperature range of between 50 and 60 °C. It was covalently immobilized to glutaraldehyde–agarose with protein and activity immobilization yields of 67% and 17%, respectively. Immobilization improved enzyme stability, increasing its half-life 5-fold, and allowed enzyme reuse for at least 25 consecutive cycles. The chitosanase activity of Branchzyme on chitosan was similar for the soluble and immobilized forms. The reaction mixture was constituted by chitooligosaccharides with degrees of polymerization of between 2 and 20, with a higher concentration having degrees of polymerization of 3–8.

A telephone survey (n = 601) was conducted in Uruguay to explore participants’ mental associations with meat and the underlying reasons changes in the quantity and type of meat consumed. Participants mentioned mostly positive associations with meat’s culinary and cultural aspects and its nutritional value as source of high-quality protein. Other associations were related to meat production, specific types of meat and cuts, meat consumption and pleasure, while price was perceived negatively. Animal welfare concerns emerged as a relevant association, but minor references were made to the effects of meat production on the environment. Changes in the amount and type of meat consumed (higher pork, chicken and cheaper cuts and beef) in the last year were mainly due to price followed by health reasons. Results emphasize the strong meat culture among Uruguayans and reveal that changes in the amount and type of meat consumed last year relate to external (price) or egoistic (health) rather than altruistic motives (ethical or environmental concerns).

Abstract
A summary of the resource.
The production manufacture of juices generates a large amount of waste consisting mainly of fruit peels. The peels are source of molecules such as coumarins and flavonoids. In this study, the peels of Citrus limonia, Citrus deliciosa, Citrus latifolia and Citrus sinensis were the raw material for supercritical fluid extraction. The Box- Behnken design was used to optimize the global yield in function of process variables. The chemical analysis of the extract was carried out by LC-MS and GC-MS. CFD was applied to the mass and momentum conservation equations in the study of supercritical extraction using citrus peels, which allowed the determination of unknown parameters of the bidimensional mathematical model. The flavonoids, nobiletin, sinensetin and tangeretin, as well as the coumarin, citropten, were identified in all species investigated. The maximum global yield obtained were 3.93 % for Citrus limonia, 4.18 % for Citrus deliciosa, 3.48 % for Citrus latifolia and 5.32 % for Citrus sinensis.

Azospirillum sp promotes the growth of many important crop plants. We demonstrated lectin binding activity in outer-membrane protein extracts of A. brasilense Sp7 by hemagglutination assays. The lectin specifically recognised the exopolysaccharide (EPS) produced by aggregated cells. Affinity chromatography using EPS-Sepharose was used to identify a 67 kDa outer-membrane lectin (OML) that recognised a binding region in the extracellular polysaccharide. Results show the specific recognition and binding between EPS and OML. The potential relationship between cell-to-cell aggregation and the OML–EPS interaction is discussed.

A summary of the resource.
nfancia 2mil20 se constituyó como un grupo de investigación en el ámbito del Espacio Interdisciplinario de la Universidad de la República de Uruguay. El tema de investigación definido por el grupo es el consumo de sustancias de uso recreativo y sustancias de abuso durante la gestación y sus efectos sobre la mujer, el feto y el desarrollo posterior del niño. La profundización del conocimientosobre la temática en las diferentes investigacionesrealizadas en 10 años de trabajopermitió el crecimiento del grupo de investigación desde una etapa de integración interdisciplinaria entre los clínicos (médicos) y el conocimiento de las materias básicas (químicos). Sobre estos logros, en principio desde un objetivo meramente teórico, se incorpora el diálogo con otros saberes, en particularcon las ciencias antropológicas, la sociología, el servicio social y el derecho con perspectiva de género. De este modo se ha transitado desde una formulación de la problemática y, por tanto, de conocimiento de la realidad, focalizada en los aspectos básico-clínicos, a una perspectiva más amplia, en donde se consideran las circunstancias de vida de estas mujeres y cómo en relación a estas se vive la maternidad, sus vínculos afectivos, familiares y con el sistema sanitario. Este libro es el resultado de las investigaciones desarrolladas por el grupo y de los esfuerzos de sus integrantes por lograr una visión más comprensiva de la problemática de estudio. Con respecto al consumo de pasta base de cocaína existe al momento actual una obra que integre los conocimientos en salud con otros saberes. La presente obra está centrada en comunicar el conocimiento actual sobre el consumo de pasta base de cocaína en embarazadas y en las vivencias posteriores al parto, con foco en la población de mayor vulnerabilidad socioeconómica que se atiende en el Hospital de la Mujer.

Abstract
A summary of the resource.
This chapter presents a review of the scientific literature concerning the determination of the mineral contents of edible fats and oils. The search was carried out by means of the Scopus and Google Scholar internet search engines without publishing date restrictions, and produced 84 references comprising book chapters and journal articles. Detection techniques and sample preparation are discussed in detail. As expected, the majority of the articles refer to the use of atomic spectrometric techniques such as atomic absorption and emission and plasma source mass spectrometry. Sample preparation techniques range from simple dilution in organic solvent to various wet-ashing approaches including microwave-assisted digestion and formation of emulsions and microemulsions, either batch-wise or by means of flow injection analysis. Dry ashing is also reported. Based on this review, it can be concluded that the determination of the mineral contents of edible oils and fats may be relevant from the technological and toxicological points of view but not from the nutritional one. It should also be noted that almost all the references report the determination of total element regardless of its speciation.

<p>Hanseniaspora vineae is an apiculate yeast that can be used for the production of interesting commercial wines, due to its contribution of fermentative volatiles. This paper presents a detailed comparative study of the use of H. vineae, compared to pure fermentations of S. cerevisiae in Tempranillo and Albillo rosé wines. Fermentations were carried out in oak barrels and stainless steel barrels. The results indicated that fermentation with H. vineae resulted in wines with residual sugars below 3.4 g/L and similar general characteristics, compared to S. cerevisiae. However, H. vineae wines contain up to 44% more total anthocyanins, resulting in an appreciable improvement in colour. In addition, H. vineae produced up to 65% more 2-phenylethyl acetate in stainless steel barrels and 2.5 times more terpene alcohols in oak barrels. Therefore, the use of H. vineae results</p>

La cisteína endopeptidasa de T. cruzi llamada cruzipaína, también conocida como cruzaína, es la enzima responsable de la mayor actividad proteolítica en todos los estadios del ciclo de T. cruzi. En las últimas décadas, estudios independientes la validan como un blanco prometedor contra dicho parásito. Numerosas son las publicaciones en las que se describen varias clases de nuevos inhibidores irreversibles o reversibles de esta enzima. A summary of the resource.
In the search for new therapeutic tools against Chagas’ disease (American Trypanosomiasis) two series of new platinum(II) complexes with bioactive 5-nitrofuryl containing thiosemicarbazones as ligands were synthesized, characterized and in vitro evaluated. Most of the complexes showed IC50 values in the μM range against two different strains of Trypanosoma cruzi, causative agent of the disease, being as active as the anti-trypanosomal drug Nifurtimox. In particular, the coordination of L3 (4-ethyl-1-(5-nitrofurfurylidene)thiosemicarbazide) to Pt(II) forming [Pt(L3)2] lead to almost a five-fold activity increase in respect to the free ligand. Trying to get an insight into the trypanocidal mechanism of action of these compounds, DNA and redox metabolism (intra-parasite free radical production) were evaluated as potential parasite targets. Results suggest that the complexes could inhibit parasite growth through a dual mechanism of action involving production of toxic free radicals by bioreduction and DNA interaction.

A summary of the resource.
The synthesis of the title Tröger's base analogue, C23H22N6, was undertaken in order to study the influence of a methyl substituent on the structure. Minor differences were found in the bond lengths of the title structure in comparison with the values for free pyrazole and for the first pyrazolic Tröger's base reported in the literature. There are two mol­ecules of opposite chirality in the asymmetric unit and the packing in the lattice is characterized by a non-crystallographic n-glide plane relating these mol­ecules.

Abstract
A summary of the resource.
In this work we present the study of the elastic properties of polycrystalline samples of superconducting YBa2Cu3O7�d prepared by the sol–gel method. The quality of all samples was checked by X-ray diffraction and scanning electronic microscopy while their physical properties were verified by transport and magnetic measurements. The elastic study was performed using the standard pulse–echo technique through measuring the phase velocity and the attenuation of ultrasonic waves (in the range of a few MHz) as a function of temperature. We have focused this study on the low temperatures interval (T < 200 K). The obtained results show a strong hysteretic behavior in the ultrasonic attenuation (in addition to usually observed hysteretic behavior for the velocity) which strongly supports the existence of a martensitic-like phase above the superconducting critical temperature Tc. We argue that this peculiar behavior can be attributed to the granularity present in the samples.

Local and systemic immune responses were studied in six dogs experimentally infected with the dog/sheep tapeworm Echinococcus granulosus. All dogs developed similar IgG antibody response to parasite antigens. In contrast, IgE and IgA responses differed widely. No relationship between IgA responses and parasite burden at the end of the infection were observed. Further, clear differences in the anti-parasite IgA response in serum as compared with specific IgA forming cells in mesenteric lymph nodes were observed within the same dog. An inverse association of anti-parasite IgE and parasite load seemed to be present, with the strongest IgE response in the one dog that had no worms in the intestine at the end of the experiment. No differences were observed in the numbers of intestinal mast cells and goblet cells among all infected dogs. However, the dog with no detectable parasite load had a marked reduction of detected mast cells in the submuscular and muscular layer of the mucosa. Our data give new insight into the immune response of dogs during E. granulosus infection and provide information that may be useful for the rational design of vaccines for the control of hydatid disease.

<p>The evaluation of oxygenation status of solid tumors is an important field of radiopharmaceutical research. With the aim to develop new potential 99mTc-radiopharmaceuticals for imaging hypoxia, we have synthesized two novel isocyanide derivatives of metronidazole, which has demonstrated high affinity for hypoxic tumors in vitro and in vivo. Methods: Metronidazole derivatives 4-isocyano-N-[2-(2-methyl-5-nitro-1H-imidazol-1-yl)ethyl]butanamide (M1) and 1-(4-isocyanobutanoyl)- 4-[2-(2-methyl-5-nitro-1H-imidazol-1-yl)ethyl]piperazine (M2) were synthesized, and labeling was performed through preparation of their corresponding 99mTc-(4+1) complexes, 99mTc-NS3M1 and 99mTc-NS3M2. The structure of the technetium complexes was corroborated by preparation and characterization of the corresponding rhenium complexes. We have studied the main physicochemical properties (stability, lipophilicity and plasma protein binding). Biological behavior in HCT-15 cells both in oxia and in hypoxia was assessed. Biodistribution in normal mice and in animals bearing induced 3LL Lewis murine lung carcinoma was also studied. Results: Metronidazole derivatives were successfully synthesized. Labeling with high radiochemical purity was achieved for both ligands. 99mTc complexes were stable in labeling milieu and human plasma. However, presence of the piperazine linker in M2 resulted in higher lipophilicity and protein binding. Although cell uptake in hypoxic conditions was observed for both radiotracers, 99mTc-NS3M2 biodistribution was considered unsuitable for a potential radiopharmaceutical due to high liver uptake and poor blood clearance. However, 99mTc-NS3M1 demonstrated a very favorable in vivo profile both in normal mice and in mice bearing induced tumors. Conclusion: Selective uptake and retention in tumor together with favorable tumor/muscle ratio make 99mTc-NS3M1 a promising candidate for further evaluation as potential hypoxia imaging agent in tumors.</p>

Glucose 9 and 2-deoxyglucose 10 were successfully synthesized and radiolabeled with [99mTc(CO)3(H20)3]+ intermediate in high yield. The complexes were characterized by HPLC and its stability with histidine over time was challenged. Cell uptake and biodistribution studies in melanoma-bearing C57BL/6 mice were performed. Both compounds showed accumulation in tumor tissue with high tumor-to-muscle ratios. Thus, D-glucose- and D-2-deoxyglucose-99mTc complex could be considered as agents for melanoma diagnosis.

As a contribution to the identification of the relevant species for biological activity and the understanding of structure– activity relationships of [VIVO(L-2H)(NN)] antitrypanosomal complexes (NN is a bidentate polypyridyl DNA intercalator; L is a tridentate salicylaldehyde semicarbazone derivative), new [VVO2(L-2H)] complexes and [VIVO(L-2H)(NN)] complexes including bipy or dppz (dipyrido[3,2-a: 2′,3′-c]phenazine) co-ligands are prepared and characterized in the solid state and in solution. Their activity is evaluated on Trypanosoma cruzi. The lipophilicity, as structural descriptor related to bioactivity, of the whole [VIVO(L-2H)(NN)] series is determined. Furthermore, the antiproliferative effect of those new compounds showing activity against T. cruzi is evaluated on the genetically related parasite T. bruceiwith the aimto develop broad spectrumagents. The new[VIVO(L-2H)(dppz)] complexes are about ten to fifteen times more toxic to T. cruzi than the bipy analogues and show quite good in vitro activity on T. brucei brucei. They are shown to interact with DNA, suggesting that this biomolecule may be the parasite target. The stability of the VIVO-complexes in solution is accessed by several techniques. Globally the data suggest that the relevant species for biological activity are the [VIVO(L-2H)(NN)] compounds, their order of activity being dependent on the NN nature, but not much on the substitution on the salicylaldehyde semicarbazone moiety. A parabolic relationship between biological response and lipophilicity (determined as RM = log [(1 / Rf) − 1] by a TLC method) is obtained. From this correlation an optimum RM value, close to 1.44, was found, which may be used as design guide for future development of antitrypanosomal compounds.

Five novel ruthenium(II)–arene complexes with polycyclic aromatic ligands were synthesized, comprising three compounds of the formula [RuCl(η6-p-cym)(L)][PF6], where p-cym = 1-isopropyl-4-methylbenzene and L are the bidentate aromatic ligands 1,10-phenanthroline-5,6-dione, 1, 5-amine-1,10-phenanthroline, 4, or 5,6- epoxy-5,6-dihydro-phenanthroline, 5. In the other two complexes [RuCl2(η6-p-cym)(L′)], the metal is coordinated to a monodentate ligand L′, where L′ is phenanthridine, 2, or 9-carbonylanthracene, 3. All compounds were fully characterized by mass spectrometry and elemental analysis, as well as NMR and IR spectroscopic techniques. Obtained ruthenium compounds as well as their respective ligands were tested for their antiparasitic and antitumoral activities. Even though all compounds showed lower Trypanosoma brucei activity than the free ligands, they also resulted less toxic on mammalian cells. Cytotoxicity assays on HL60 cells showed a moderate antitumoral activity for all ruthenium compounds. Compound 1 was the most potent antitumoral (IC50 = 1.26±0.78 μM) and antiparasitic (IC50=0.19±0.05 μM) agent, showing high selectivity towards the parasites (selectivity index N100). As complex 1was themost promising antitumoral compound, its interactionwith ubiquitin as potential target was also studied. In addition, obtained ruthenium compounds were found to bind DNA, and they are thought to interact with this macromolecule mainly through intercalation of the aromatic ligand.

Abstract
A summary of the resource.
In this article a potentiometric method for reducing sugar quantification is described. Copper(II) ion reacts with the reducing sugar (glucose, fructose, and others), and the excess is quantified using a copper wire indicator electrode. In order to accelerate the kinetics of the reaction, working conditions such as pH and temperature must be adjusted. However, a good relationship of mass of reducing sugar to potential was reached. Some of the advantages of the method are its simplicity, low cost, and that can be carried out even in colored and turbid samples. The analysis is safer than the standard Lane–Eynon method for determination of reducing sugars. To our knowledge, the use of a simple copper wire as electrode has not been reported previously for this type of determination, leading to a simplification of the experimental equipment.

<p>Objective: To assess the effects of three different front-of-package labelling (FOPL) schemes on objective understanding and intention to purchase of products, in Jamaica.</p>
<p>Setting: Supermarkets in Jamaica.</p>
<p>Participants: Adult supermarket shoppers in Jamaica (n=1206) aged 18 years old or older were included in the study, except for those visually impaired, or unable to give informed consent.</p>
<p>Design: Multiarm parallel-group randomised trial.</p>
<p>Interventions: Participants were randomly allocated to one of the three intervention groups or the control group. They were exposed to two-dimensional images of 12 mock-up products presented in random and balanced order. Participants assigned to the intervention groups were exposed to one FOPL scheme: black octagonal warning labels (OWL), magnifying glass high-in single icon (MGG) or traffic-light labelling (TFL). The control group was exposed to the nutrition facts up front.</p>
<p>Outcome measures: OR for correctly understanding nutritional information (correctly selecting the least harmful option, correctly identifying sugars, sodium and/or saturated fats found to be in excess) and choosing to purchase the least harmful option (purchase intention), more often.</p>
<p>Results: Compared with the control group, the odds for correctly selecting the least harmful option more often were 107% higher in the OWL group (OR 2.07, 95% CI 1.54 to 2.78; p<0.001), whereas the MGG (1.18, 95% CI 0.89 to 1.57; p=0.24) and the TFL (1.13, 95% CI 0.85 to 1.51; p=0.39) were inefficacious in improving such odds. OWL also resulted in the highest odds for correctly identifying a product with excessive amounts of sugars, sodium and/or saturated fats and for deciding to purchase the least harmful option or none of the options.</p>
<p>Conclusions: Octagonal warning labels performed best at improving the ability of adult shoppers in Jamaica to understand the nutrition information and at encouraging them to purchase the least harmful option more often.</p>
