A summary of the resource.
The opilionid, Parampheres ronae, has a pair of orange markings on its carapace at a location where some other opilionids discharge yellowish, quinone-containing defensive fluid. P. ronae itself produces a defensive secretion, but the fluid is quinone-free and nearly translucent. We suggest that the orange markings in P. ronae are aposematic in the sense that they are imitative of the glandular emissions of quinone-producing opilionids.

Abstract
A summary of the resource.
In this mini-review, a comprehensive discussion on the state of the art of hybrid organic–inorganic mixed ionic–electronic conductors (hOI-MIECs) is given, focusing on conducting polymer nanocomposites comprising inorganic nanoparticles ranging from ceramic-in-polymer to polymer-in-ceramic concentration regimes. First, a brief<br />discussion on fundamental aspects of mixed ionic–electronic transport phenomena considering the charge carrier transport at bulk regions together with the effect of the organic–inorganic interphase of hybrid nanocomposites is presented. We also make a recount of updated instrumentation techniques to characterize structure, microstructure,<br />chemical composition, and mixed ionic–electronic transport with special focus on those relevant for hOI-MIECs. Raman imaging and impedance spectroscopy instrumentation techniques are particularly discussed as relatively simple and versatile tools to study the charge carrier localization and transport at different regions of hOI-MIECs including both bulk and interphase regions to shed some light on the mixed ionic–electronic transport mechanism. In addition, we will also refer to different device assembly configurations and in situ/operando measurements experiments to analyze mixed ionic–electronic<br />conduction phenomena for different specific applications. Finally, we will also review the<br />broad range of promising applications of hOI-MIECs, mainly in the field of energy storage<br />and conversion, but also in the emerging field of electronics and bioelectronics.

Abstract
A summary of the resource.
The impacts of the modern, agrochemicals based agriculture that threatens the overall systems sustainability, need to be monitored and evaluated. Seeking for agroecosystems monitors, the present article focus in the occurrence and abundance of aquatic macroinvertebrates, that have been frequently used as bioindicators of water quality due to their relationship with land use. Some of these organisms are on the top of the food chain, where bioaccumulation and biomagnification processes can be observed, and they can turn into secondary pollution sources of systems and terrestrial organisms as well. Odonate nymphs, which belong to the functional group of predators, were selected for this study. A methodology to determine 73 pesticide residues in odonate nymphs by LC-MS/MS and GC-MS/MS was developed. A QuEChERS sample preparation strategy was adapted. As it is complex to obtain samples especially in disturbed ecosystems, the method was minimized to a sample size of 200 mg of fresh nymphs. The method was validated and good recoveries (71–120%) with RSDs below 20% for the majority of the studied pesticides at least at two of the assayed levels 1, 10 and 50 µg kg−1 were obtained. For 32 analytes the limit of quantitation was 1 µg kg−1 and 10 µg kg−1 for the others. The lineal range was observed between 1–100 µg kg−1 in matrix-matched and solvent calibration curves for most of the assessed pesticides. LC-MS/MS matrix effects were evaluated, 40% of the analytes presented low or no signal suppression. Only flufenoxuron presented high matrix effects. The obtained methodology is adequate for pesticide multiresidue analysis in aquatic macroinvertebrates (odonates) aiming to contribute to the ecological state evaluation of freshwater ecosystems.

Consumer studies conducted under central location test (CLT) conditions continue to be dominant in product research and context evocation have been suggested as an avenue to partly mitigate the lack of real consumption settings. In this research the influence of evoked context on product acceptability was investigated in eight diverse consumer studies (138–268 participants per study) through the use of between-subjects designs that allowed the comparison of hedonic scores obtained with and without evoked context. In a departure from previous research, consumers mentally evoked their typical consumption contexts for the focal product categories and content analysis of descriptions of these situations showed them to often be idiosyncratic. Results were partly product- and situation-specific, and in this regard replicated past research. The evoked context only significantly modified hedonic scores in two of the eight studies, whereas it increased sample discrimination in three studies. Thus, accumulating evidence now supports the conclusion that evoked context is less rather than more likely to impact hedonic responses. Nonetheless, a benefit of context evocation is to give products a more complete meaning, and this may motivate their continued use in CLT settings. For researchers who wish to continue their use, key considerations in implementation are discussed, including relevance in home-use testing (HUT). By asking consumers to describe a typical eating occasion for the tested products, useful understanding of product use and pairings is gained. Such data can be easily elicited from consumers and obtained independently of context evocation.

Minthostachys verticillata essential oil (EO) is a natural product that reports immunomodulatory effects on human T cells as well as anti-inflammatory activity. Bovine mastitis is a worldwide disease, mainly caused by bacteria, affecting milk quality and yield, leading to high economic losses. Environmental pathogens, as Enterococcus faecium, are implicated in the disease. Antibiotic therapy is adequate, although it can leave residues in milk, causing problems in human health. The search of immunomodulatory substances for bovine mastitis treatment is a promising alternative strategy. The aim of this study was to characterize the effect of M. verticillata EO on macrophage phagocytosis and evaluate its immunomodulatory and protective effects in mice challenged with E. faecium. The results showed that EO activated macrophage phagocytosis mechanisms inducing reactive oxygen species production. Moreover, EO modulated the innate immune response in mammary glands of female Balb/c mice challenged with E. faecium decreasing the infiltration of polymorphonuclear neutrophils and IL-1β and TNF-α mRNA expression. In addition, EO increased the expression of IL-10 in the last hours of infection. Treatment with EO did not increase the number of activated CD4+ or CD8+ T cells or the production of specific antibodies. These results suggest that EO play an important role in helping to resolve the infection in the first hours without activating adaptive immunity. In addition, a marked decrease of the bacterial count in the glands of mice treated with EO was observed. A natural product such as M. verticillata EO could have a potential use to control bovine mastitis.

Heterometallic compounds containing La(III), bivalent cationsM(M = Co, Ni, Ca), and 2,20-oxydiacetate (oda) as connecting ligand have been prepared and characterized. The complexes can be formulated as [La2M3(oda)6(H2O)6] Æ 12H2O. The structure of [La2Co3(oda) 6(H2O)6] shows the presence of the La(III) coordinated by six carboxy and three ether oxygen atoms, and the Co(II) cation bonded to four carboxy oxygens and two molecules of water. An open 3D framework is observed, containing large hexagonal channels. The chemical systems were also investigated in solution (25.0 �C, I = 0.5 M Me4NCl) by potentiometry. The same kind of polynuclear species have been found in aqueous solution.

<p>Recent studies are being focused on establishing a link between structural and mixed ionic-electronic transport properties based on the characterization of organic mixed ionic-electronic conductors (OMIECs) under operation conditions in organic electrochemical transistor (OECT) devices. Here, we report the simultaneous access to microstructural, chemical, and physical information using impedance spectroscopy and micro-Raman imaging techniques to address the mixed ionic-electronic transport mechanisms in PEDOT:PSS-based OMIECs working as OECT channel materials. We focused on the out-of-plane and inplane mixed ionic-electronic transport mechanisms and complement our experimental studies with computational simulations using molecular dynamics (MD) and density functional theory (DFT) methodologies. Our studies revealed that the out-of-plane transport is based on ionic injection and diffusion mainly associated with the formation of nanopore channels in PEDOT:PSS exhibiting sulfonic groups toward the aqueous media. Thus, we also evidenced that the dedoping is not only due to the direct interaction of positive ions with PEDOT:PSS bulk region but also indirectly by the rearrangements of sulfonic groups from PSS toward the aqueous media. The in-plane transport in the OECT channel was properly described as a mixed ionic-electronic transport, with not only ionic transport of sodium (Na+) but also hydronium (H3O+) species mainly through nanopores and PEDOT:PSS bulk regions and with electronic transport mainly through the PEDOT:PSS bulk region. Although it is shown that the electronic transport monotonically declines with increasing ionic concentration, the Na+ diluted regime (1 mM) exhibits weak selectivity compared to H3O+ species, and the Na+ concentrated regime (100 mM) induces extreme porosity in PEDOT:PSS yielding to the destruction of the device. These results introduce some particular features for PEDOT:PSS-based OECT channel materials that need to be considered not only for ionic concentration and selectivity but also other related stability issues.</p>

Chagas disease is caused by the trypanosomatid parasite Trypanosoma cruzi and threatens millions of lives in South America. As other neglected diseases there is almost no research and development effort by the pharmaceutical industry and the treatment relies on two drugs, Nifurtimox and Benznidazole, discovered empirically more than three decades ago. Nifurtimox, a nitrofurane derivative, is believed to exert its biological activity through the bioreduction of the nitro-group to a nitro-anion radical which undergoes redox-cycling with molecular oxygen. This hypothesis is generally accepted, although arguments against it have been presented. In the present work we studied the ability of Nifurtimox and five N-oxide-containing heterocycles to induce oxidative stress in T. cruzi. N-Oxide-containing heterocycles represent a promising group of new trypanosomicidal agents and their mode of action is not completely elucidated. The results here obtained argue against the oxidative stress hypothesis almost for all the studied compounds, including Nifurtimox. A significant reduction in the level of parasitic low-molecular-weight thiols was observed after Nifurtimox treatment; however, it was not linked to the production of reactive oxidant species. Besides, redox-cycling is only observed at high Nifurtimox concentrations (>400 mM), two orders ofmagnitude higher than the concentration required for anti-proliferative activity (5 mM). Our results indicate that an increase in oxidative stress is not the main mechanism of action of Nifurtimox. Among the studied N-oxide-containing heterocycles, benzofuroxan derivatives strongly inhibited parasite dehydrogenase activity and affected mitochondrial membrane potential. The indazole derivative raised intracellular oxidants production, but it was the least effective as anti-T. cruzi.

EI gran interes que despiertan los compuestos basados en metales de transición como el platino en el tratamiento de una gran variedad de tumores sólidos y diserninados, debe su origen al descubrimiento de la acci6n antineophisica del Cisplatino en el ano 1970. A pesar de los excelentes resultados logrados con el Cisplatino, el mismo presenta lirnitaciones relacionadas con su toxicidad y con la aparici6n de resistencia intrinseca y/o adquirida en algunas lineas celulares. Esto ultimo sigue siendo cierto, con distintos perfiles farmacol6gicos, para varios compuestos de platino derivados. Todo ello ha fomentado la busqueda de anlilogos de eficacia similar 0 mayor en la quimioterapia del cancer, pero con perfiles farmacológicos que exhiban menor toxicidad y efectos laterales, y mas baja incidencia de resistencia. Entre cientos de compuestos emparentados sintetizados a la fecha s610 uno de ellos, el Carboplatino, ha sido aprobado mundialmente para su uso terapeutico. No obstante 10 anterior, la busqueda continua y algunos analogos ya lograron la aprobación local o regional. Los motivos estructurales que despiertan hoy en dia mayor interes en el area pueden dividirse en: i) compuestos de platino en conformaci6n trans; ii) grupos salientes en posici6n cis distintos de cloruro; iii) aminas secundarias, terciarias 0 heterociclicas, funcionando como ligandos mono y bidentados; iv) compuestos de Pt(IV); v) compuestos multi-nucleares de platino; y vi) compuestos con metales distintos al platina (Pd, Au, Ru, Re, etc.). Entre los compuestos de Pt(II) pueden destacarse dos grandes familias como muy prometedoras: 1) aquella donde los dos grupos amino (ligandos espectadores en el Cisplatino) se sustituyen por diaminociclohexano (dach) o un grupo aminociclohexil; y 2) aquella cuyos ligandos espectadores contienen piridina (Pi) 0 algun derivado. Por su lado, los complejos de Pd(II) iso-estructurales a los de Pt(1I) parecen ser una altemativa interesante ante la creciente evidencia experimental de varios compuestos de paladio menos tóxicos que los de platino y con buena actividad citotóxica. Con el fin de acercar guias para la busqueda de nuevos potenciales farmacos de Pt(1I) y Pd(lI), en este trabajo se utilizan los metodos y modelos de la quimica te6rica y computacional en sus versiones clasica, cuantica y mixta QMIMM (Quantum Mechanics/Molecular Mechanics) para desentrafiar intimidades de los aspectos estructurales y mecanismo de acci6n molecular de esta familia de compuestos. En el capitulo 3, un analisis estructural a nivel electr6nico de 27 compuestos de Pt(II)/ Pd(II), la identificaci6n de descriptores te6ricos de la estructura/reactiv~dad y la bUsqueda de indices experimentales permiten, con tecnicas de mineria de datos, ordenar/clasificar a los compuestos y finalmente establecer correlaciones estructura-propiedad. EI capitulo 4 enfoca las reacciones que pueden sufrir algunos compuestos, considerados representativos de los diferentes motivos estructurales presentes en el universo seleccionado, previa alcanzar el principal blanco molecular (el ADN nuclear). Se aborda el mecanismo de acción de las posibles transformaciones (isomerización cis-trans y acuación), analizando el efecto sobre la termodinamica y cinetica de la reacción que tienen: el cambio del metal y los sustituyentes espectadores. Los aspectos estructurales, energeticos y la reactividad de los aductos monofuncionales y bifuncionales formados entre los complejos metalicos y modelos quimicos del ADN de complejidad creciente, constituyen el centro del capitulo 5. Se aportan piezas claves de informaci6n para una mejor y mayor comprensi6n a nivel molecular de los aspectos estructurales y mecanismo de acci6n de un grupo de compuestos de Pt(II)/Pd(II). A su vez, el trabajo brinda evidencias que echan luz sobre varias contradicciones que existen de larga data a nivel experimental y que resultan importantes a la hora del disefio racional de nuevos farmacos. Una visi6n integradora, basada en el conocimiento de la quimica, farmacologia, bioquimica y biologia de los compuestos estudiados permite la predicci6n de propiedades y comportarnientos en un amplio rango de niveles de organización. Desde los pasos elementales de las reacciones mas sencillas hasta la predicción, para analogos del Cisplatino, de un post-procesamiento celular del ADN modificado diferente.

El presente trabajo de tesis se enmarca en una línea de investigación vinculada a la enfermedad de Chagas y desarrollada por muchos años en el actual Centro de Bioinformática Estructural del DETEMA, de la Facultad de Química, Universidad de la República. Esta línea, iniciada con motivo de la tesis doctoral de la patrocinante, la Dra. Paulino, ha dado múltiples trabajos publicados, entre ellos “Modeling and study of cyclosporyn A and related compounds in complexes with Trypanosoma cruzi cyclophilin”, como producto de la primer parte del trabajo presentado en esta tesis. También la línea de investigación ha originado otras tesis de Maestría y Doctorado, entre ellas la de uno de los copatrocinantes, el Dr. Iribarne. Por otro lado no es ajena la mencionada línea al Laboratorio de Inmunotecnología dirigido por el otro copatrocinante, MsC. Battistoni. Mucho se ha investigado con el objetivo de obtener una cura para el Mal de Chagas. Esta Tesis es un aporte más, centrándose en el estudio de la molécula de ciclosporina (CsA), y compuestos estructuralmente relacionados. Estos compuestos han mostrado actividad inhibitoria sobre Trypanosoma cruzi. Si bien la CsA es una droga con actividad inmunosupresora, algunos de los compuestos estructuralmente relacionados activos contra T. cruzi no presentan efectos inmunosupresores. Se ha propuesto a CsA como compuesto líder para el desarrollo de nuevas drogas útiles en la farmacoterapias de la enfermedad de Chagas. Se conocen con cierto grado de detalle las bases bioquímicas de las actividades inmunosupresoras e inhibitorias sobre protozoarios de CsA. En ambas están involucradas una familia de proteínas, las ciclofilinas. Se han publicado datos de las actividades inhibitorias de CsA y compuestos relacionados sobre ciclofilinas presentes en T. cruzi, en particular sobre la ciclofilina mas expresada en T. cruzi, la TcCyP19. Las bases moleculares de la diferencia de actividad inmunosupresora y de inhibición sobre T. cruzi de CsA y de una serie análogos no han sido dilucidadas. Esta tesis pretende, mediante el uso de diversas herramientas de modelado molecular, docking y de mecánica molecular, estudiar el comportamiento de los complejos binarios de TcCyP19 con una serie de compuestos análogos a CsA para obtener información a nivel molecular que aporte a la comprensión de las diferencias encontradas. En forma complementaria, se estudian complejos de CsA y dos de sus análogos con una ciclofilina humana, así como el comportamiento de una proteína mutante de la ciclofilina humana con baja afinidad por CsA. Una vez elaborados los modelos de los sistemas a estudiar se analizaron los posibles sitios de unión de estos compuestos con las ciclofilina en estudio, así como los modos de unión a los mismos. Utilizando simulaciones de dinámica molecular, se obtuvo un muestreo del espacio de fases de los sistemas en estudio que permitió analizar estructuralmente y energéticamente dichos sistemas. Varios términos energéticos se calcularon, que incluyeron energías de interacción ligando-proteína y cálculos de energías libres de asociación por medio del método LIE (Linear Interacction Energy). Se discuten algunos aspectos metodológicos, como la necesidad de hacer varias simulaciones de dinámica molecular para obtener un muestreo mas apropiado del comportamiento del sistema y el cumplimiento de las premisas para usar el método LIE. Los resultados obtenidos muestran que se trata de sistemas con comportamiento muy similar, para los cuales los métodos de cálculo usados no son lo suficientemente sensibles para diferenciar energías libres de unión. Se demostró la posible existencia de lugares de unión de Modelado y estudio de complejos de Ciclosporina A y compuestos relacionados ….. 5 los compuestos estudiados diferentes al sitio catalítico, cuya relevancia deberá ser estudiada in vitro. Todo lo aprendido con estos sistemas permitirá abordar con mejores perspectivas ulteriores etapas del estudio de esta familia de compuestos que involucran complejos ternarios y sus potenciales actividades inmunosupresoras en el huésped o inhibitorias del crecimiento de T. cruzi.

In the present study a systematic approach was used to model the anti-T. cruzi activity of a series of N-oxide containing heterocycles belonging to four chemical families with a wide structural diversity. The proposed mode of action implies the reduction of the N-oxide moiety; however, the biochemical mechanism underlying the anti-T. cruzi activity is still unkown. For structural representation two types of descriptors were analyzed: quantum chemical (AM1) global descriptors and properties coded by radial distribution function (RDF). Both types of descriptors point to the relevance of electronic properties. The local-RDF (LRDF) identified an electrophilic center at 4.1-4.9 Å from the oxygen atom of the N-oxide moiety, although other properties are required to explain the biological activity. While the mode of action of N-oxide containing heterocycles is still unknown, the results obtained here strengthen the importance of the electrophilic character of the molecule and the possible participation of the heterocycle in a reduction process. The ability of these descriptors to distinguish among activity classes was assessed using Kohonen neural networks, and the best clustering descriptors were later used for model building. Different learning algorithms were used for model development, and stratified 10-fold cross-validation was used to evaluate the performance of each classifier. The best results were obtained using k-nearest neighbors (k-NN) and decision tree (J48) methods combined with global descriptors. Since tree-based methods are easily translated into classification rules, the J48 model is a useful tool in the de novo construction of new N-oxide containing heterocycle lead structures.

EgDf1 is a developmentally regulated protein from the parasite Echinococcus granulosus related to a family of hydrophobic ligand binding proteins. This protein could play a crucial role during the parasite life cycle development since this organism is unable to synthetize most of their own lipids de novo. Furthermore, it has been shown that two related protein from other parasitic platyhelminths (Fh15 from Fasciola hepatica and Sm14 from Schistosoma mansoni) are able to confer protective inmunity against experimental infection in animal models. A three-dimensional structure would help establishing structure/function relationships on a knowledge based manner. 3D structures for EgDf1 protein were modelled by using myelin P2 (mP2) and intestine fatty acid binding protein (I-FABP) as templates. Molecular dynamics techniques were used to validate the models. Template mP2 yielded the best 3D structure for EgDf1. Palmitic and oleic acids were docked inside EgDf1. The present theoretical results suggest definite location in the secondary structure of the epitopic regions, consensus phosphorylation motifs and oleic acid as a good ligand candidate to EgDf1. This protein might well be involved in the process of supplying hydrophobic metabolites for membrane biosynthesis and for signaling pathways.

Cyclophilins (CyPs) are enzymes involved in protein folding, catalyzing the isomerisation of peptidyl prolyl bonds in proteins and peptides between the cis- and trans-conformations. They are also the major cellular target for the immunosuppressive drug Cyclosporin A (CsA). In Trypanosoma cruzi, the most abundantly expressed CyP is an isoform of 19 kDa, TcCyP19, in which the enzymatic activity is inhibited by CsA. Among a reported set of CsA analogues, two non-immunosuppressive compounds, H-7-94 and F-7-62, proved to be the best inhibitors of TcCyP19 enzymatic activity as well as the most efficient trypanocidal drugs. With the objective of analysing, at the molecular level, how the structural differences between the three above-mentioned inhibitors justify their different inhibitory activity on TcCyP19, three-dimensional molecular modelling structures were generated to computationally simulate behaviours and interactions. An energy-minimized model of each binary complex in water with ions was obtained. These models were then used as starting point for molecular dynamic simulations, performed with GROMOS96 program. With the resulting set of co-ordinates and energies, a comparison of the interaction between CsA and both CsA analogues in T. cruzi and human cyclophilins were performed. Within the different magnitudes analysed, the total potential complex energy exhibited the best correlation with the experimental data.

Abstract
A summary of the resource.
The presence of defects can introduce important changes in the electronic structure of graphene, leading to phenomena such as C magnetism. In addition, vacancies are reactive and permit the incorporation of dopants. This paper discusses the electronic properties of defective graphene for O and B decoration. Phonon calculations allow us to address directly the stability of the systems under study. We show that it is possible to obtain magnetic solutions with and without dangling bonds, demonstrating that C magnetism can be achieved in the presence of B and O.

Abstract
A summary of the resource.
Se propone un modelo farmacocinético compartimental (SIMULFIS) en donde se consideran los flujos sanguíneos del organismo. Aplicado a la Digoxina, se estudiaron las consecuencias farinacocinéticas y los niveles plasmáticos y tisulares de la droga, al alterar el gasto cardíaco y los flujos sanguíneos en el cuerpo humano. Se analiza en base al modelo, la interacción Digoxina-Amiodarona. Los resultados de las simulaciones demuestran un aumento de la captación tisular y disminución de la concentración plasmática de Digoxina durante el ejercicio físico. El modelo asume un único mecanismo en la interacción Digoxina-Amiodarona, que explica el aumento de la biodisponibilidad oral, la disminución de la eliminación renal y hepática, y el inalterado volumen de distribución de la Digoxina.
