The heterocyclic N atom nearest to the Cl atom and the Nsulfonamide atom could be coordination sites for the silver ion in the Ag–SCP–SCN complex. The Ag–SCP complex is a polymeric compound with metal–metal bonds, and the heterocyclic and sulfonamide N atoms are points of coordination for AgI. Both complexes showed activity against all the tested bacteria, and in the cases of Escherichia coli and Pseudomonas aeruginosa, the action was better than that of SCP. In all cases, both silver–SCP complexes showed better antifungal activity than SCP, which was inactive against the tested fungi. Notably, the activity against P. aeruginosa, a nosocomial multidrug-resistant pathogen, was better than that of the reference antibiotic cefotaxim. Both silver–sulfa complexes displayed moderate activity against the tested yeast, especially for C. neoformans, which is an important fact considering the incidence of cryptococcosis, mainly in immune-deficient patients. No chromosomal aberrations were observed with the Allium cepa test, which is auspicious for further study of these complexes as potential drugs. Chagas disease is an endemic illness in Latin America caused by the parasite Trypanosoma cruzi. Current chemotherapies are old and inadequate, and the emergence of drug-resistant strains underscores the need of new drugs. Platinum-based complexes have been shown to be a promising approach against parasitic diseases. In this work, the effect of 1,1'-bis(diphenylphosphino)ferrocene pyridine-2-thiolate-1-oxide Pt(II) hexafluorophosphate, Pt-dppf-mpo, was studied on T. cruzi. A promising antitrypanosomal activity was determined for the CL Brener strain with a low cytotoxicity determined using in vitro-cultured mammal cells. The compound uptake in parasites treated with concentrations of 1× and 10× the IC50 value reached ~75% and 19%, respectively. Pt-dppf-mpo induced necrosis after 24 hr of parasite incubation. This event was preceded by depolarization of mitochondrial membrane potential. Cell vitality assays showed high esterase activity in treated parasites. However, despite this increase in metabolic activity, treated epimastigotes showed rounded morphology and loss of flagellum with a reduction in mobility as compound concentration and/or time of incubation was increased. At last, we demonstrate that Pt-dppf-mpo incubation also affects the trypomastigote infection process as well as the infection persistence evaluated as the number of amastigotes per cell in a dose-dependent manner. There is an urgent need to develop new drugs against Chagas’ disease. In addition, the mechanisms of action of existing drugs have not been completely worked out at the molecular level. High throughput approaches have been demonstrated to be powerful tools not only for understanding the basic biology of Trypanosoma cruzi, but also for the identification of drug targets such as proteins or pathways that are essential for parasite infection and survival within the mammalian host. Here, we have applied these tools towards the discovery of the effects of two organometallic compounds with trypanocidal activity, Pd–dppf–mpo and Pt–dppf–mpo, on the transcriptome and proteome of T. cruzi epimastigotes. These approaches have not yet been reported for any other prospective metal-based anti T. cruzi drug. We found differentially expressed transcripts and proteins in treated parasites. Pd–dppf–mpo treatment resulted in more modulated transcripts (2327 of 10 785 identified transcripts) than Pt–dppf–mpo treatment (201 of 10 773 identified transcripts) suggesting a mechanism of action for Pd–dppf–mpo at the transcriptome level. Similar numbers of differentially expressed proteins (342 and 411 for Pd–dppf–mpo and Pt–dppf–mpo respectively) were also observed. We further functionally categorized differentially expressed transcripts and identified cellular processes and pathways significantly impacted by treatment with the compounds. Transcripts involved in DNA binding, protein metabolism, transmembrane transport, oxidative defense, and the ergosterol pathways were found to be modulated by the presence of the compounds. Our transcriptomic dataset also contained previously validated essential genes. Los helmintos parásitos son un grupo de metazoarios muy diverso y complejo, cuya principal característica en común es su habilidad de generar infecciones crónicas en sus hospederos naturales. Dentro de las helmintiasis, la equinococosis quística (EQ) es una parasitosis causada por el estadío larvario de Echinococcus granulosus, y es considerada una zoonosis de gran relevancia por afectar tanto al ser humano como a una amplia variedad de especies ganaderas productivas. Interesantemente, la infección por E. granulosus es capaz de durar varios años de manera asintomática en sus hospederos intermediarios, por lo que se ha postulado que el parásito tiene la habilidad de modular la respuesta del hospedero en favor de su sobrevida. En este sentido, los fenómenos inmunológicos responsables de la resistencia o susceptibilidad del hospedero a la infección han sido escasamente estudiados. Por lo tanto, el presente trabajo se realizó a efectos de profundizar en el conocimiento de las respuestas inmunes potencialmente protectoras desencadenadas por hospederos intermediarios. Para ello, se realizaron estudios en humanos y modelos murinos focalizados en demostrar que las respuestas inmunes montadas por ciertos hospederos tendrían un papel crucial en limitar el establecimiento y/o desarrollo de la EQ. En primer lugar, se caracterizó el perfil serológico de una cohorte de individuos residentes en una zona endémica para EQ en Uruguay; que si bien fueron serológicamente positivos, no fueron diagnosticados imagenológicamente como pacientes con EQ durante un período de seguimiento de 2 años. En forma paralela, se caracterizaron también sendas cohortes de pacientes con EQ confirmada por cirugía y de donantes sanos. Los resultados mostraron que los individuos seropositivos pero normales por ultrasonido para EQ, presentaron un perfil serológico particularmente interesante, capaz de diferenciarlos de los dos grupos control. Asimismo, las características de dicho perfil sugieren que son capaces de mediar potenciales funciones efectoras antiparasitarias. Humoral immunity wanes during healthy ageing, increasing susceptibility to infections in the elderly. In this sense, information about parasite infections and human immunosenescence is scarce. Cystic echinococcosis (CE) is an infectious disease caused by the larval stage of the cestode parasite Echinococcus granulosus, whose prevalence in humans shows an increase with host age. Susceptibility to human CE has been associated with humoral immunity to some extent; and therefore, we have here analyzed the influence of host age on the serological profile of young, middle-aged, and aged patients. Our results highlighted the detrimental influence of ageing on the intensity and quality of the anti-parasite antibody response. Remarkable differences in serological profiles between young and aged individuals were observed. In this sense, through Principal Components Analysis we identified aged patients as those exhibiting overall less intense antibody responses, mainly in isotypes/subclasses supposed to exert efficient anti-parasite activities (e.g. IgE and IgG1). Thus, these humoral defects could at least partially explain the reported increase in CE prevalence among older individuals, since a weaker immune response in the elderly might facilitate the establishment and maintenance of the parasite infection. Finally, a possible association between age-dependent susceptibility to CE and host immunosenescence is discussed. Binding studies show that both soluble and membrane-bound forms of CD5 and CD6 bind to intact viable protoscoleces from E. granulosus s.l. through recognition of metaperiodate-resistant tegumental components. Proteomic analyses allowed identification of thioredoxin peroxidase for CD5, and peptidyl-prolyl cis-trans isomerase (cyclophilin) and endophilin B1 (antigen P-29) for CD6, as their potential interactors. Further in vitro assays demonstrate that membrane-bound or soluble CD5 and CD6 forms differentially modulate the pro- and anti-inflammatory cytokine release induced following peritoneal cells exposure to E. granulosus s.l. tegumental components. Importantly, prophylactic infusion of soluble CD5 or CD6 significantly ameliorated the infection outcome in the mouse model of secondary cystic echinococcosis. Recombinant ectodomains of human scavenger receptors CD5 (rshCD5) and CD6 (rshCD6) were previously reported to bind its tegumental antigens and to exert prophylactic effects in a murine model of infection. Although the properties of mammalian scavenger receptors include the binding to diverse pathogen-derived structures, their interaction with helminth parasites has been scarcely explored. Therefore, we report here a search for CD5 and CD6 interactorswithin E. granulosus s.l. antigens.Mass spectrometry analysis of pull-downs fromsoluble tegumental componentswith biotinylated rshCD5 and rshCD6 resulted in 17 and 11 overrepresented interactors,<br />respectively, 8 of which were shared. The nteractors included previously reported protective molecules against E. granulosus s.l. and/or other helminths. Streptococcus pneumoniae (neumococo) continua siendo el principal agente etiológico de la neumonía adquirida en la comunidad en el mundo entero. Las defensas innatas son esenciales para la detección del neumococo y el control de la infección, en especial en etapas tempranas de la neumonía. Sin embargo, la bacteria ha desarrollado mecanismos de evasión que le permiten escapar de la inmunidad natural, resultando muchas veces en la progresión de la enfermedad. La creciente aparición de cepas resistentes a antibióticos, sumado al alto costo de los tratamientos y estrategias de profilaxis hace imperativo el desarrollo de nuevos tratamientos que puedan complementar los ya disponibles. Este trabajo se ha enfocado en el desarrollo de nuevas estrategias de tratamiento contra la neumonía neumocócica basadas en el uso de inmunoestimulantes del sistema innato, administrados por ruta mucosa. Partiendo de la hipótesis de que la activación de las defensas innatas es suficiente para impedir la progresión de la enfermedad, se evaluó la capacidad de diferentes agonistas de receptores innatos de conferir protección frente a la neumonía invasiva en ratones, empleando cepas clínicas de S. pneumoniae. Se demostró que la protección puede ser conferida tanto empleando agonistas de receptores activados normalmente por neumococo, como empleando agonistas de receptores que no son activados por neumococo durante una infección natural. Sin embargo, no todos los receptores activan respuestas protectoras y la estimulación por ciertas vías puede llegar incluso a tener consecuencias negativas. Flagelina de Salmonella enterica serovar Typhimurium, ligando de TLR5/NLRC4-NAIP5, fue uno de los agonistas estudiados. Flagelina confirió mayor protección que cualquiera de los otros agonistas probados, al ser administrado por ruta intranasal por lo que se realizó la caracterización de los mecanismos moleculares y celulares asociados a su capacidad de inducir protección contra la neumonía neumocócica. Los resultados sugieren que el efecto protector requiere del reclutamiento de neutrófilos hacia los espacios alveolares, lo que se logra a través de la secreción de citoquinas de la familia ELR-CXC por parte de las células epiteliales de la mucosa respiratoria y es independiente de células B, T y anticuerpos. La protección mostró ser dependiente de la presencia del motivo de unión a TLR5 en la molécula pero no de la región de interacción con NLRC4-NAIP5, sugiriendo que los efectos observados dependen del reconocimiento extracelular de flagelina a través de TLR5 pero no del inflamasoma. La terapia demostró ser eficaz contra dos serotipos diferentes de neumococo. En segunda instancia, se realizó la puesta a punto de un modelo de inmunoterapia sublingual y se evaluó la capacidad de flagelina de conferir protección frente al desafío intranasal con neumococo administrada por esta ruta, obteniéndose un buen grado de protección. Al igual que en el modelo de instilación intranasal, la protección inducida por administración sublingual de flagelina, requirió reclutamiento de neutrófilos y de la activación de la vía TLR5, pero no de la activación de inflamasoma vía NLRC4-NAIP5. 6 Como último tema se estudió la capacidad adyuvante de flagelina en vacunas de aplicación sublingual. Empleando ovoalbúmina como antígeno modelo se comprobó que la administración de flagelina en conjunto con el antígeno incrementa la respuesta de anticuerpos específicos en suero, observándose un perfil de subclases mixto con predominio del perfil Th2. Empleando un modelo de transferencia adoptiva de células T transgénicas para el receptor T y condicionada para reconocer el fragmento 323-339 de la ovoalbúmina en contexto MHC-II, se estudió la capacidad de flagelina de inducir su proliferación en respuesta a una única inmunización sublingual y se determinó la localización anatómica de la respuesta. Se estudió además la contribución de los receptores TLR5 y NLRC4-NAIP5 confirmándose que la señalización a través de uno solo es suficiente para el efecto adyuvante observado. Por último, se analizó la capacidad de flagelina de promover procesamiento antigénico y expresión de moléculas estimuladoras en células dendríticas derivadas de médula ósea. En suma, este trabajo destaca las ventajas de la potenciación de la inmunidad innata en mucosas mediante el empleo de Patrones Moleculares Asociados a Patógenos (PAMPs) para el desarrollo de nuevas estrategias de tratamiento y profilaxis de las infecciones respiratorias, y en particular las causadas por S. pneumoniae. We proposed to explore the pathogenesis of these oral defects, which are poorly studied. Sequence analyses of proteins from gluten (gliadins) and of prolinerich enamel proteins (amelogenin and ameloblastin) suggested the presence of common antigenic motifs. Therefore, we analyzed, by ELISA and western blotting, the reactivity of sera from patients with CD against gliadin and enamel-derived peptides. Correlation analyses between the levels of specific antibodies against gliadin and enamel derived peptides and inhibition experiments confirmed the presence of cross-reactive antibodies. Immunoblot analysis revealed that the most prominent component in enamel matrix derivative (of approximately 18.6 kDa), identified by an amelogenin-specific antibody, is recognized by sera from patients with CD; in addition, several fractions of pure gliadin were recognized by amelogenin-specific antibody. In agreement, sera from mice immunized with enamel matrix-derived proteins generated antibodies that recognized a peptide (of approximately 21.2 kDa) derived from gliadin. In conclusion, antibodies against gliadin generated in patients with CD can react in vitro with a major enamel protein. This study aimed to evaluate the waste cooking oil (WCO) hydrolysis in ultrasonic system using lipase as catalyst. Lipase was produced by the fungus Aspergillus niger via solid state fermentation (SSF) using canola meal as substrate. Prior to the hydrolysis reaction, the lipase behavior when subjected to ultrasound was evaluated by varying the temperature of the ultrasonic bath, the exposure time and the equipment power. Having optimized the treatment on ultrasound, the WCO hydrolysis reaction was carried out by evaluating the oil:water ratio and the lipase concentration. For a greater homogenization of the reaction medium, a mechanical stirrer at 170 rpm was used. All steps were analyzed by experimental design technique. The lipase treatment in ultrasound generated an increase of about 320% in its hydrolytic activity using 50% of ultrasonic power for 25 min. at 45 °C. The results of the experimental design conducted for ultrasound-assisted hydrolysis showed that the best condition was using an oil:water ratio of 1:3 (v:v) and enzyme concentration of 15% (v/v), generating 62.67 μmol/mL of free fatty acids (FFA) in 12 h of reaction. Thus, the use of Aspergillus niger lipase as a catalyst for hydrolysis reaction of WCO can be considered as a possible pretreatment technique of the oil in order to accelerate its degradation. Four novel alkali−manganese(II) trifluoroacetates were synthesized, and their potential as self-fluorinating precursors to layered perovskites A2MnF4 (A = K, Rb, and Cs) was demonstrated. Na2Mn(tfa)4, K4Mn2(tfa)8, Rb4Mn2(tfa)8·0.23H2O, and Cs3Mn2(tfa)7(tfaH) (tfa = CF3COO− and tfaH = CF3COOH) were grown as single crystals, and their crystal structures solved using X-ray diffraction. Chemically pure K4Mn2(tfa)8, Rb4 Mn2(tfa)8·0.23H2O, and Cs3Mn2(tfa)7(tfaH) were also prepared in polycrystalline form as confirmed by thermal analysis and powder X-ray diffraction. Thermolysis of these powders yielded the isostructural series K2MnF4, Rb2MnF4, and Cs2MnF4 at low temperatures (≈200−300 °C). Trifluoromethyl groups belonging to the trifluoroacetato ligands served as the fluorine source, thereby eliminating the need for external fluorinating agents. K2MnF4 and Rb2MnF4 were obtained as single-phase powders, whereas Cs2MnF4 crystallized along with CsMnF3. Access to polycrystalline Cs2MnF4 coupled to Rietveld analysis enabled elucidation of the crystal structure of this ternary fluoride, which had remained elusive. El trabajo desarrollado en la presente tesis doctoral se enmarcó en una línea de investigación interdisciplinaria del grupo en la búsqueda y desarrollo de nuevos antihelmínticos. Dentro de los antecedentes del mismo, fueron sintetizados derivados de 2-amino-δ-valerolactamas, una nueva serie de compuestos que presentaron una importante actividad antihelmíntica in vitro, usando el nemátodo de rata Nippostrongylus brasiliensis, larva L4, pero con muy baja actividad in vivo, en ratas infectadas con dicho nemátodo. Mediante estudios de difusión intraparasitaria ex vivo en N. brasiliensis, se confirmó la muy baja o nula capacidad de estos derivados valerolactama de ingresar al medio intraparasitario. Estos estudios sentaron las bases para plantear la hipótesis de que la baja eficacia de los compuestos valerolactama se debe a su muy baja capacidad de difusión intraparasitaria. Considerando las evidencias experimentales mencionadas, en la presente tesis se diseñó la síntesis de nuevos derivados con el objetivo de modificar ciertas propiedades fisicoquímicas, como la lipofilia, y mejorar así los perfiles de difusión intraparasitaria. Para obtener estos nuevos compuestos se recurrió a la estrategia de hibridación molecular, sintetizando una serie de derivados híbridos valerolactama- benzimidazol. Complementariamente se evaluó la actividad antihelmíntica in vitro de los nuevos compuestos, en el modelo de N. brasiliensis L4, así como la actividad in vivo de uno de los derivados híbridos más promisorios, en ratas infectadas con el mismo nemátodo. Además, se estudiaron las propiedades fisicoquímicas y electrónicas, mediante cálculos computacionales ab initio. Se optimizó la geometría de los compuestos sintetizados y se obtuvieron descriptores fisicoquímicos y electrónicos. La lipofilia fue determinada de forma experimental, mediante método de HPLC de fase reversa. Finalmente, se desarrollaron y validaron dos modelos de difusión intraparasitaria ex vivo, uno de ellos sobre el céstodo Mesocestoides vogae (larva tetrathyridia) y otro con el nemátodo de interés en animales de producción, Haemonchus contortus (estadio adulto). A través de estos modelos fue posible evaluar la capacidad de difusión intraparasitaria de las moléculas híbridas, y corroborar el acierto de la estrategia de hibridación molecular para mejorar la difusión intraparasitaria. Estos modelos fueron de utilidad además, para estudiar de forma preliminar, la biotransformación que sufren los compuestos ensayados en ambos helmintos, ya que el metabolismo aumentado puede servir para la defensa de los parásitos contra tales fármacos, y por tanto resultar un mecanismo complementario de resistencia a antihelmínticos. Microtubules are non-covalent cylindrical polymers formed by alpha- and beta-tubulin heterodimer units, crucial for cell division, intracellular transport, motility and differentiation. This makes them very attractive pharmacological targets exploited to develop different drugs such as anthelmintics, antifungals, and antineoplastics. In this work, in order to establish an in vitro target-based screen to integrate to the search for new anthelmintics, we explored the extraction of native assembly-competent tubulin from two helminth parasites: Mesocestoides vogae tetrathyridia (syn. corti, Cestoda: Cyclophyllidea), a useful cestode biological model, and Haemonchus contortus, a sheep gastrointestinal nematode of interest in livestock production. For this purpose, a novel tubulin affinity chromatography procedure was employed, based on the binding capacity of TOG (Tumor Overexpressed Gene) domain from MAPs (microtubule-associated proteins). The TOG domain of the protein Stu2 from Saccharomyces cerevisiae fused to GST (glutathione S- transferase) were produced in E. coli, and the immobilized recombinant proteins allowed for native tubulin extraction from parasites. The binding capacity of TOG1 affinity column (3.6%) was estimated using commercial porcine brain tubulin. A total amount of up to 126 μg of M. vogae tubulin was purified, whereas H. contortus tubulin co-eluted with glutamate dehydrogenase enzyme. The identity of tubulins was confirmed by western blotting and mass spectrometry. The abundance of tubulin estimated in M. vogae was 10% soluble extract, which probably could explain differences observed between tubulin purification results of both helminth parasites.
