Aspergillus spp. and Penicillium spp. are the main producers of ochratoxin A (OTA), a mycotoxin responsible for fatal human diseases. Some authorities have established a maximum of 2 μg/L of OTA in wine. Although the incidence and occurrence of OTA in grapes and wine is highly related to climate conditions, as has been extensively documented, there is no conclusive information on the effects of cultivation systems on the presence of OTA. This study focuses on determining the effect of the trellis system, planting density and cordon height on plant microclimate and thus on Aspergillus spp. contamination and OTA production in Tannat wines in Southern Uruguay. Two experiments were conducted during the 2010–2011 growing season: (1) a strip split plot design with five replicates and two cordon heights (CH) (0.5 m and 1.0 m above the soil) were compared in two planting densities (PD) (0.8 and 1.5 m between plants); (2) a randomised complete block design, vertical shoot positioning (VSP) versus Lyra trellis systems were evaluated. The results suggest that, even the macro- and micro-climate growing conditions play an important part in Aspergillus developing on grapes. Agronomical practices also have an undoubted impact on the risk and control of OTA accumulation in wine. Un capítulo completo se dedica a su calidad sensorial, al entrenamiento de jueces y características de su cata. The larvae of the cestodes belonging to the genus Echinococcus dwell primarily in mammalian liver. They are protected by the laminated layer (LL), an acellular mucin-based structure. The glycans decorating these mucins constitute the overwhelming majority of molecules exposed by these larvae to their hosts. However, their decoding by host innate immunity has not been studied. The effect of pure Zn2+ and combined Zn2+ and Mn2+ supplements on the growth and ligninolytic activity of wood-rotting basidiomycetes was studied. Based on measurements of minimum inhibitory concentration and minimum decolourisation-inhibiting concentration available from cultures of Inocutis sp., Trametes versicolor and Gloeophyllum trabeum, and a comparison of such results with those obtained in previous work for P. chrysosporium and P. atropurpurascens, metal supplements were shown to inhibit fungal physiology at concentrations within the study range. Average minimum inhibitory concentration values were consistently lower for ZnSO4 than for ZnSO4/MnSO4 (2 mM Zn2+), demonstrating the antagonistic effect of the tested cations. Furthermore, because the ligninolytic system of P. atropurpurascens had shown a high tolerance to metals, its ligninolytic and germinative behaviour was compared with that of P. chrysosporium. Conidial germination was inhibited at 1 mM of Zn2+ in the two strains, whereas extracellular activity was inhibited at concentrations above 2 mM. Sensory and consumer scientists disagree on the practice of concurrently obtaining sensory information in hedonic tests. This is in part due to different mindsets about what consumers are able to do and evidence that such co-elicitation may bias hedonic scores. Check-all-that-apply (CATA) questions have been claimed to have a smaller effect on hedonic scores than other attribute such as just-about-right or intensity scales. In this research, nine studies using consumers as participants examined effects on hedonic product scores when sensory attribute information was co-elicited using CATA questions. The use of CATA concurrently with hedonic was benchmarked against concurrent attribute liking scores, attribute intensity scores and just-about-right scaling. Across a range of product categories (beer, fresh fruit, tea, flavoured water, crackers, savoury dips), only weak and transient evidence of bias of hedonic scores when concurrently using CATA questions was established. This effect was independent on whether samples, on average were moderately liked or moderately disliked, and replicated when samples were assessed partially by the sense of smell only or via full product assessment (appearance, aroma, flavour, taste, aftertaste, mouthfeel). The present research suggests that co-elicitation of hedonic scores and product attribute information using CATA questions may bias the hedonic scores, but not that it certainly will do so. This needs to be recognised, leading to more widespread acceptance that co-elicitation has merit. Investigators should decide on whether or not to co-elicit product attribute information using CATA questions on a case-by-case basis, acknowledging that bias may occur. A long-standing pathomechanistic model proposes that the polyglutamine (polyQ)-length-dependent toxicity threshold observed in all polyQ diseases is triggered by a conformational change within the monomer that occurs only above a certain polyQ length. If true, this yet undefined and elusive mutant-specific toxic conformation would constitute a direct therapeutic target. Three anti-polyQ antibodies—MW1, 1C2 and 3B5H10—have been extensively used to probe the conformation of polyQ. The crystal structure of the MW1 epitope reveals a linear, non-pathogenic polyQ. In contrast, although the detailed structure of its epitope is unknown, the 3B5H10 antibody is widely advertised and used as a conformational antibody that recognizes the toxic conformation of expanded polyQ. We solved the crystal structure of the 1C2 antigen-binding domain (1C2-Fab) and performed a direct comparison between the 1C2, MW1 and 3B5H10 structures. The MW1 and 1C2 antibodies have similar sequences and structures, consistent with their binding to short polyQ and their polyQ length-discrimination properties. Unexpectedly, the 3B5H10 antibody also shares striking features with MW1 and 1C2, which prompted us to revisit its binding properties. We show that the 3B5H10 epitope is actually a short, non-pathogenic polyQ. All three antibodies MW1, 1C2 and 3B5H10 interact similarly with polyQ of various lengths, and bind small polyQ epitopes in similar linear and extended conformations. Together with studies published during the recent years, our work argues against the hypothesis that a mutant-specific conformation in monomeric polyQ molecules is the toxic entity responsible for polyQ diseases. Distamycin, a natural polyamide containing three heterocycle rings with a polar end, has inspired several groups to prepare synthetic analogues, which proved to have anti-trypanosomal and anti-tumoral activity. We describe the synthesis of bi and tri thiazoles amides that harbor different substitutions at their ends and the evaluation of their anti-Trypanosoma brucei activity. The most active compound 10b showed better biological activity (EC50 310 nM and selectivity index 16) than the control drug nifurtimox (EC50 15 μM and selectivity index 10). Studies on the mode of action show that the parasiticidal activity of 10b originates from disruption of lysosomal homeostasis, which is followed by release of redox active iron, an increase in oxidizing species and collapse of cell membrane integrity. In this respect, our study suggests that non-charged lipophylic distamycins destabilize cell membranes. Projective techniques rely on a stimulus to elicit from respondents projections of their beliefs onto other people or objects, thus reflecting their own perceptions, feelings and motivations. In this work, the job-sorting technique, in combination with Aaker’s personality scale, was applied to the study of cream liqueur brand image in Uruguay and Costa Rica. Eight jobs with different levels of intellectual complexity, salary and physical demands were selected. In each of the two countries, three (two imported and one locally made) liqueurs were presented to 90 consumers with similar socio-demographic characteristics. Respondents were told to select the job, profession or occupation (JPO) that best fitted each brand. After JPO selection, respondents were asked to select from a list of 88 personality traits all the words that they considered applicable to each JPO selected. The JPOs associated with the different brands showed no similarity between Uruguayan and Costa Rican respondents, but the personality traits associated with the two imported brands were similar in both countries, supporting the use of JPOs as metaphors in the study of brand images. The brand image of locally made liqueurs was associated with traits that differed clearly from those of the imported brands. As a Medical Geology research issue, an environmental arsenic risk assessment study in the most important sedimentary aquifer in southern Uruguay is presented. The Raigón Aquifer System is the most exploited in Uruguay. It has a surface extent of about 1,800 square kilometres and 10,000 inhabitants in San Jose Department, where it was studied. Agriculture and cattle breeding are the main economic activities and this aquifer is the basic support. The groundwater sampling was done on 37 water samples of PRENADER (Natural Resources Management and Irrigation Development Program) wells. Outcropping sediments of Raigón Formation and the overlying Libertad Formation were also sampled in the Kiyú region. The analyses were performed by inductively coupled plasma-optical emission spectrometry (ICP-OES) and inductively coupled plasma-mass spectrometry (ICP-MS). The results showed 80% samples with arsenic levels exceeding the 10 μg/l of WHO as limit for waters, and 11% exceeds the 20 μg/l limit of uruguayan regulation. The median, maximum and minimum water arsenic concentrations determined have been 14.24, 24.19 and 1.44 μg/l, respectively. On the other hand, nine sediment samples of Raigón and Libertad Formations in Kiyú region were analysed and yielded median, maximum and minimum arsenic concentrations of 5.03, 9.82 and 1.18 ppm, respectively. This issue leads to the supposition that the population, as well as industrial and agricultural activities, are consuming water with arsenic concentrations over the national and international maximum recommended limit. Como una aproximación a la Geología Médica en Uruguay se presentan los resultados de un estudio sobre el estado del Sistema Acuífero Raigón en relación a la presencia de arsénico, en el entendido que es el acuífero más explotado en el país. En particular, el área cubierta por este trabajo es de unos 1800 km2 en un territorio con una población de unos 10,000 habitantes en el departamento de San José. La colecta de aguas del acuífero se realizó en perforaciones de la base de datos de PRENADER (Proyecto de Manejo de Recursos Naturales y Desarrollo del Riego) obteniéndose 37 muestras, y sobre 9 muestras de sedimentos tanto de la Formación Raigón como la suprayacente Libertad en afloramientos de la región de Kiyú. Los análisis fueron realizados mediante ICP-MS. Un 80% de las muestras de aguas analizadas mostraron contenidos mayores a los límites de la OMS (10 μg/l) y un 11% de las muestras superaron el límite de la OSE (20 μg/l). El valor medio, máximo y mínimo determinado en el caso del agua subterránea fue de 14.24, 24.19 and 1.44 μg/l, respectivamente. Por otro lado, las nueve muestras de sedimentos analizados mostraron concentraciones media, máxima y mínima de arsénico de 5.03, 9.82 and 1.18 ppm, respectivamente, descartándose en principio un origen geogénico. In this work we present the synthesis and full structural characterization of hydrothermally synthesized titanate nanotubes (TNTs) for energy applications. It is of interest to characterize the building block of certain nanomaterials and their surface termination. This task is particularly difficult for nanotubes, where the exhibited surface curvature is a limitation for its study. We present the synthesis and thorough structural characterization through several experimental techniques for morphology and compositional characterization. Simulated X-rays powder diffraction patterns (XRPD) of real size nanotubes models are refined with experimental data. The building blocks for the TNT models were obtained from computational simulations by means of Density Functional Theory for H2Ti2O5·0.5H2O, H2Ti2O5·H2O, H2Ti3O7 and H2Ti3O7·0.67H2O. Our procedure confirms that open‐ended TNTs are obtained, with &sim;100 nm of length and 6(1) nm of inner diameter. By interpreting the XRPD data, the TNT building block was obtained and found to derive from H2Ti3O7, where the distance between [TiO6] octahedral layers is increased in accordance with H2Ti3O7·0.67H2O. This model fully agrees with all the structural characterizations, validating this methodology and suggesting its potential use to study other nanomaterials. A thiol-β-cyclodextrin was synthesized by a simple and environmentally friendly three-step method comprising epoxy activation of β-cyclodextrin, thiosulfate-mediated oxirane opening, and further reduction of the S-alkyl thiosulfate to a thiol group. The final step was optimized by using thiopropyl-agarose, a solid phase reducing agent with many advantages over soluble ones. β-Cyclodextrin thiolation was confirmed by titration with a thiol-reactive reagent, NMR studies, and MALDI-TOF/TOF. Thiolated cyclodextrin had an average value of one thiol group per molecule. Thiol-β-cyclodextrin proved to be an excellent agent for controlling polyphenol oxidase activity. This copper-containing enzyme is responsible for browning in fruits and vegetables. Under the same conditions, thiol-β-cyclodextrin generated a reductive microenvironment that increased the antibrowning effect on Red Delicious apples compared to unmodified β-cyclodextrin. A first principles simulation of the crystal structure of potassium peroxocarbonate is presented, using density functional methods (both local and semilocal) for the calculations. An experimental crystal structure with a seemingly inconsistent disposition of the peroxide bonds was used as initial input. Both geometry optimizations of the molecular structure and optimization of the cell size were performed. While cell parameters and heavier atom positions determined at the GGA level are very close to the experimental ones, there are important discrepancies in the positioning of the hydrogen atoms. As a result of these calculations, it was shown that the assignment of the peroxydic hydrogens and the peroxydic bond in the experimental structure was incorrect. A more accurate structure is presented and geometrical as well as cell parameters described. It is also shown that LDA is not accurate enough to describe this type of ionic crystals, because of overbinding of the ions, leading to incorrect cell parameters and volume. Our methodology was validated using DFT methods with several basis sets. Whey proteins are known for their high nutritional value and bioactivity, notably great potential is found on α-lactalbumin. The objective of this study was to evaluate antioxidant, antiglycant and ACE (angiotensin converting enzyme)-inhibitory activity of α-lactalbumin peptides obtained from enzymatic hydrolysis with Alcalase through different hydrolysis conditions. The optimization of enzymatic hydrolysis was studied by response surface methodology variating enzyme:substrate ratio (0.0050%, 0.0525% and 0.1000% w/w) and time (0, 30 and 60 minutes) measuring antioxidant activity by ABTS and ORAC-FL (response variables), founding an augment of this activity with time. Characterization of seven samples showed increasing hydrolysis with time. Hydrolysate obtained with 0.1000% w/w and 60 minutes demonstrated higher antioxidant activity related to greater hydrolysis. This hydrolysate did not show antiglycant activity but promoted advanced glycation end products formation with methylglyoxal and glucose. In contrast, this sample showed 30% of ACE inhibition when compared to Captopril, having great potential by enhancing hydrolysis. Chitooligosaccharides possessing remarkable biological properties can be obtained by enzymatic hydrolysis of chitin. In this work, the chitosanase activity of soluble and immobilized glycosyltransferase (Branchzyme) toward chitosan and biochemical characterization are described for the first time. This enzyme was found to be homotetrameric with a molecular weight of 256 kDa, an isoelectric point of 5.3, and an optimal temperature range of between 50 and 60 °C. It was covalently immobilized to glutaraldehyde–agarose with protein and activity immobilization yields of 67% and 17%, respectively. Immobilization improved enzyme stability, increasing its half-life 5-fold, and allowed enzyme reuse for at least 25 consecutive cycles. The chitosanase activity of Branchzyme on chitosan was similar for the soluble and immobilized forms. The reaction mixture was constituted by chitooligosaccharides with degrees of polymerization of between 2 and 20, with a higher concentration having degrees of polymerization of 3–8.
