El arroz (Oryza sativa) es el cereal cultivado más importante a nivel mundial (672×106 toneladas /año). Su destino prioritario es la alimentación humana como arroz blanco. El Uruguay es un exportador neto de arroz blanco, generando anualmente 130 mil toneladas de salvado de arroz como residuo. Aunque el 10% de este salvado tiene como destino la producción de aceite y una parte se utiliza para la alimentación animal, es de gran interés estudiar su uso potencial en la alimentación humana. Las enfermedades no transmisibles son responsables de la muerte de 40 millones de personas en el mundo cada año. Uno de los factores de riesgo más relevante son el sobrepeso y la obesidad. Estos están directamente relacionados un mayor riesgo de padecer diabetes tipo 2, enfermedades cardiovasculares, entre otras. Dados los actuales hábitos alimenticios, de alto consumo de azúcares libres se prevé un incremento de la incidencia global de estas patologías en los próximos años. La presente tesis doctoral se enfoca en la posibilidad de aprovechar una materia prima sub-valorada como el salvado de arroz, con el objetivo de desarrollar nuevos ingredientes alimentarios para el consumo humano que contribuyan a la reducción del riesgo de padecer estas patologías crónicas no trasmisibles. Para ello, se desarrolló un extracto rico en antioxidantes polifenólicos cuyas condiciones de producción fueron optimizadas a través de un diseño experimental central compuesto (CCD) 2k = 23 = 16 tratamientos, con 3 variables independientes (% de etanol, tiempo y temperatura) a 3 niveles (-1, 0 y +1) y utilizando tecnologías simples y fácilmente escalables. El residuo insoluble generado tras la obtención de dicho extracto fue caracterizado por análisis de su composición nutricional y capacidad antioxidante demostrando ser una fracción rica en fibra dietética antioxidante. x Las condiciones de extracción optimizadas (maceración simple con 35% de etanol en la solución extractora, a 20°C por 105 minutos) permitió obtener un extracto de salvado de arroz principalmente rico en proteínas (12,74 g/100g de MS) y en carbohidratos (77,0 g/100g MS). Bajo dichas condiciones, se recuperó el total de fibra dietética soluble (2,23 ±0,10 g/100g MS) que compone el salvado de arroz; así como, un contenido de lípidos totales de 3,38 ±0,07 g/100g MS y de cenizas de 4,82±0,06 g/100g MS. El extracto antioxidante resultó ser rico en polifenoles (71,42±1,60 mg de GAE/ g de extracto) y presentó elevada capacidad antioxidante (451±14 μmol de TE/g muestra frente a radical ABTS.+ y 912±31 μmol de TE/g muestra frente a radicales peroxilos). El análisis cromatográfico por UHPLC indicó que los componentes fenólicos mayoritarios presentes en extracto son ácido clorogénico (44, 84 ± 2, 25 mg / 100 g de materia seca) y ácido ferúlico (12,44 ± 0,60 mg / 100 g de materia seca). El extracto presentó valores IC50 de 0,14 ± 0,01 y 4,50 ± 0,08 mg/mL contra radicales peroxilos y ABTS.+, respectivamente. Para su mejor caracterización el extracto seleccionado se fraccionó mediante ultrafiltración empleando una membrana con tamaño de poro uniforme de 3 kDa. Aplicando este procedimiento se encontró que el extracto está constituido por un 68% de compuestos con tamaño molecular mayor a 3 kDa (FAPM) y 32% de compuestos con tamaño menor a este valor (FBPM). El análisis cromatográfico de ambas fracciones indica que la FAPM contiene principalmente ácido ferúlico mientras que el compuesto fenólico mayoritario que compone a la FBPM es ácido clorogénico. El estudio de bioaccesibilidad de los componentes bioactivos que forman el extracto antioxidante de salvado de arroz llevado a cabo por simulación gastrointestinal in vitro indicó una pérdida significativa (p6g/100g de galleta). Se utilizó stevia como edulcorante no nutritivo resultando ser un alimento que no contiene azúcar añadido y; por lo tanto, presenta potencial para reducir el riesgo de diabetes. El alimento presentó una capacidad antioxidante de 3653 ± 150 μmol TE/ porción considerando los compuestos xii extraíbles e hidrolizables presentes y de 1125 ± 73 μmol de TE/porción de galleta (30g) por el método de ABTS-QUENCHER. El % de bioaccesibilidad de los polifenoles presentes en las galletas fue de 17% respecto a los polifenoles totales presentes en una porción de 30g de alimento (33,07±1,59 mgGAE/porción). El promedio de aceptabilidad general de las nuevas galletas libres de gluten, fuente de fibra dietética y antioxidantes fue de 51% (88 evaluaciones). La formulación debe considerarse un prototipo inicial sobre el cual debe trabajarse para obtener un alimento de óptima calidad sensorial. En conclusión, se puede afirmar con un 95% de confianza que es posible obtener 2 nuevos ingredientes alimentarios de calidad nutricional y funcional a partir de salvado de arroz: extracto antioxidante (fracción soluble) y la fracción insoluble rico en fibra dietética antioxidante. El proceso de extracción propuesto es de gran simplicidad y bajo costo de inversión por lo que se alinean al concepto de “extracción verde” destacándose el hecho de no generar nuevos residuos de producción siendo innovadora, eco-sostenible y de promisoria implementación en la industria uruguaya. Las galletas elaboradas permiten afirmar que se ha logrado un alimento tradicional de gran consumo regional que reúne varias de las condiciones recomendadas sobre una dieta saludable por la OMS (2014 y 2015) para la reducción del riesgo de padecer enfermedades crónicas no transmisibles en un solo alimento. Las galletas no contienen gluten, ni azúcar añadido (endulzado con stevia como edulcorante no nutritivo), son fuente de fibra alimentaria antioxidante (3 g /porción) y presentan capacidad antioxidante siendo un primer prototipo de alimento funcional que valida la recuperación del salvado de arroz. This study characterised the chemical and sensory properties of 15 non-spreadable goat cheeses (eight fresh and seven ripened). The majority of the cheeses had a short ripening period. Hence, low free fatty acid (FFA) contents were detected. The main difference among the cheeses was the short-chain FFA content, which was higher in the ripened cheeses, and responsible for their distinctive odours and flavours. In both types of cheeses (fresh and ripened), lactic acid was the most abundant organic acid (OA), followed by propionic acid. Hierarchical cluster analysis identified three distinct groups of samples with marked differences in their texture and flavour attributes. The percentage of moisture was positively correlated to sensory attributes such as surface film, solubility, moistness and sourness, and negatively correlated to the firmness and saltiness. No significant correlation was found among sensory attributes and total fat percentage or any of the OAs. Pesticides can be found in bee products as they are usually employed to protect the beehive, but they can also reach the hive due to environmental contamination. These contaminants could be present in processed foods. One of the most common and consumed comfitures based in bee products are honey and propolis candies, for which no analytical method has yet been developed. This work presents the development of an ethyl acetate based extraction method followed by dispersive clean up using Primary and Secondary Amine (PSA) plus Graphitized Carbon Black (GCB) with Gas Chromatography coupled to Mass Spectrometry (GC-MS) determination for pesticide residue monitoring candies containing honey and propolis from the Mercosur region. Sixteen pesticides found in bee products as well as three pesticide metabolites of toxicological significance were evaluated including acaricides and insecticides (pyrethroids, organophosphates and some organochlorines). The method was validated, with a LOQ of 0.01 mg/kg for most analyzed pesticides, showing a recovery rate of 70-120% with <20% RSD. Real samples from the Mercosur countries were analyzed. This enzyme immobilization approach involves the formation of disulfide (−S–S–) bonds with the support. Thus, enzymes bearing exposed nonessential thiol (SH) groups can be immobilized onto thiol-reactive supports provided with reactive disulfides or disulfide oxides under mild conditions. The great potential advantage of this approach is the reversibility of the bonds formed between the activated solid phase and the thiol-enzyme, because the bound protein can be released with an excess of a low-molecular-weight thiol (e.g., dithiothreitol [DTT]). This is of particular interest when the enzyme degrades much faster than the adsorbent, which can be reloaded afterwards. The possibility of reusing the polymeric support after inactivation of the enzyme may be of interest for the practical use of immobilized enzymes in large-scale processes in industry, where their use has often been hampered by the high cost of the support material. Disulfide oxides (thiolsulfinate or thiolsulfonate groups) can be introduced onto a wide variety of support materials with different degrees of porosity and with different mechanical resistances. Procedures are given for the preparation of thiol-activated solid phases and the covalent attachment of thiol-enzymes to the support material via disulfide bonds. The possibility of reusing the polymeric support is also shown. Marketing of unhealthy products has been identified as one of the main characteristics of the food environment that negatively affects children's eating patterns. Restrictions on advertising of unhealthy foods to children have already been imposed in different countries. However, marketing strategies are not limited to broadcast and digital advertising, but also include package design. In this context, the current study aimed to describe the food products targeted at children and sold in supermarkets in Montevideo, Uruguay, in terms of package design and nutrient profile. Two supermarkets in Montevideo were selected for data collection. In each supermarket, all products targeted at children were identified. Products were analyzed in terms of package design and nutritional profile, considering the Pan American Health Organization Nutrient Profile Model. A total of 180 unique products were identified, which included a wide range of product categories. The great majority of the products corresponded to ultra-processed products with excessive amounts of sodium, free sugars, total fat, saturated fat, and/or trans fat, which are not recommended for frequent consumption. Several marketing strategies were identified in the design of packages to attract children's attention and drive their preferences. The most common strategies were the inclusion of cartoon characters, bright colors, childish lettering, and a wide range of claims related to health and nutrition, as well as the products' sensory and hedonic characteristics. The study's findings provide additional evidence on the need to regulate packaging of products targeted at children. Each Re(II) ion is six-coordinate with four bromide ligands, a linear nitrosyl group and one monodentate nitrogen donor L building a tetragonally distorted octahedral surrounding. The magnetic properties of 1–4 have been investigated in the temperature range 1.9–295 K. They behave like quasi magnetically isolated spin doublets with very weak antiferromagnetic interactions through intermolecular Br⋯Br contacts. For most food products, shelf life is determined by changes in their sensory characteristics. A predetermined increase or decrease in the intensity of a sensory characteristic has frequently been used to signal that a product has reached the end of its shelf life. Considering all attributes change simultaneously, the concept of multivariate shelf life allows a single measurement of deterioration that takes into account all these sensory changes at a certain storage time. The aim of the present work was to apply survival analysis to estimate failure criteria in multivariate sensory shelf life testing using two case studies, hamburger buns and orange juice, by modelling the relationship between consumers' rejection of the product and the deterioration index estimated using PCA. In both studies, a panel of 13 trained assessors evaluated the samples using descriptive analysis whereas a panel of 100 consumers answered a “yes” or “no” question regarding intention to buy or consume the product. PC1 explained the great majority of the variance, indicating all sensory characteristics evolved similarly with storage time. Thus, PC1 could be regarded as index of sensory deterioration and a single failure criterion could be estimated through survival analysis for 25 and 50% consumers' rejection. The proposed approach based on multivariate shelf life testing may increase the accuracy of shelf life estimations. La depresión postparto (DPP) es el trastorno psiquiátrico materno más frecuente que puede determinar efectos negativos en el vínculo madre-hijo y sobre el desarrollo infantil. Nuestro objetivo fue analizar la asociación entre la DPP a los 6 meses post parto y alteraciones del neurodesarrollo a los 18 meses, en una población homogénea caracterizada por bajo nivel socioeconómico y cultural. Pacientes y Método: Estudio de cohorte. Se incluyeron 127 puérperas y recién nacidos sanos. Se realizó el seguimiento del binomio a los 6 meses del parto, donde se le aplicó a la madre el test de Beck para depresión y ansiedad. Se evaluó el neurodesarrollo de los niños a los 18 meses mediante el test de Brunet-Lezine Revisado. Resultados: La muestra quedo compuesta por 125 mujeres y sus hijos. La media de edad fue de 24,5 años (DE: 6,02), el 30,6% tenía menos de 6 años de educación. La incidencia de DPP moderada a severa a los 6 meses del nacimiento fue de 20%. La media de puntaje de desarrollo global fue de 73,5 (DE: 8,1) en la población con depresión y 76,97 (DE: 8,07) en la población sin depresión (p = 0,04). El coeficiente de desarrollo de lenguaje fue de 69,08 (DE: 10,35) en la población con depresión y 74,11 (DE: 0,67) en la población sin depresión (p = 0,01). Conclusiones: La incidencia de DPP moderada a severa fue 20%. La DPP persistente en un contexto de vulnerabilidad socioeconómica impacta sobre el desarrollo infantil. In this work, an upflow anaerobic packed bed reactor configuration to produce hydrogen using cheese whey as the substrate was tested. The microbiological composition was linked to the reactor operation. Three different organic loading rates of 22, 33, and 37 g COD/L-d were used at a fixed hydraulic retention time of 24 h. The increase of the organic loading rate from the initial value of 22 g COD/L-d to a value of 37 g COD/L-d and the adjustment of the pH to values greater than 5 had a positive effect on the hydrogen production and values of up to 1 L H2/L-d were achieved. The production of hydrogen was stable under all of the investigated conditions, and the problems frequently reported in this kind of reactors (bed clogging, methanogenesis and solvent production) were not observed during operation. The highest hydrogen yield was 1.1 mol H2/mol lactose far from the maximum theoretical value (8 mol H2/mol lactose), but similar to previous works using cheese whey as substrate. The microbiological analysis showed a mixed population with a low proportion of hydrogen-producing fermenters (Clostridium and Klebsiella) and other non-hydrogen-producing organisms. Debido a esta crítica situación y a la aparición de cepas resistentes a los fármacos usuales, la investigación de nuevas moléculas para el tratamiento de la TB se ha vuelto una prioridad. En este trabajo se reporta la síntesis y caracterización por análisis elemental, termogravimetría, medidas conductimétricas y espectroscopías UV-Vis, IR y EPR de los complejos [Cu(INH)(H2O)]SO4 ⋅2H2O (Cu-INH) y [CoCl(INH)2(H2O)]Cl⋅2.5H2O (Co-INH) con isoniacida (INH). Por otra parte, se determinó la actividad anti-Mycobacterium tuberculosis (CIMCu-INH = 0.78 µg/mL y CIMCo-INH = 0.19 µg/mL) y la citotoxicidad de los complejos (IC50 = 48.8 and 625 µg/mL respectivamente para los complejos de cobre y cobalto) y se calculó el índice de selectividad (62.5 para Cu-INH y 3205 para Co-INH). Opal phytolith, geochemical and sedimentological data from two cores taken in Peña Lagoon, a small water body on the Atlantic shore of Uruguay, are presented in this paper. The choice of this specific sampling location aimed to obtain high resolution records to infer late Holocene environmental and climatic changes in southeastern Uruguay, in order to improve previous models for early and middle Holocene. The combined record of two cores extends to 2500 14C BP, with a sedimentation rate ranging between 0.6 and 2 mm y−1. A multiproxy approach allowed the identification of changes in dynamics, particularly organic matter and sediment input variability from the watershed. With this information, differences in the amount and extent of environmental signals were evaluated and a comprehensive climate change model was generated, especially for the last 1500 years. Three main climatic events were distinguished. The oldest, which lasted until 700 AD, was characterized by temperate and humid conditions. The second extended until 1200 AD and warmer and wetter conditions were inferred, but also a colder and drier pulse was observed in between. This second event was assigned to the Warm Medieval Period (WMP). During the third event, assigned to the Little Ice Age (LIA), which extended until the present, a high variability in climate was observed, but also three dry/cold phases reaching their maxima by 1300, 1600 and 1900 AD were inferred. Applying structure–activity relationships and principal component analysis strategies we were able to determine ring substituent effects and physicochemical properties that are important for the antichagasic activity of these novel derivatives, as well as get an insight into their possible mechanisms of action. Molecular docking studies revealed the binding orientation of the ligands in the active site of cruzipain providing new guidelines for the further design of better inhibitors. Conclusion: Compound 2a constitute a promising hit compound for novel anti-T. cruzi agents showing that the benzimidazole scaffold may represent an interesting therapeutic alternative for the treatment of Chagas disease. Measurement of consumers’ emotional associations to food/beverage stimuli is one way to obtain product insights that extend beyond hedonic responses. Survey methods are a popular way to obtain direct responses from consumers, but concerns over their ecological validity exist. In this research, a preliminary investigation and assessment of emoji-based questionnaires as a potential method for measuring food-related emotional associations was conducted. Six studies involving 1087 consumers in USA and China were conducted using names of foods and beverages as the stimuli. On average consumers selected 1–2 emoji per stimulus. The elicited data was able to discriminate between stimuli that span the hedonic continuum and generate detailed emotional product profiles. Less discrimination was obtained between hedonically similar stimuli, but meaningful emotional profiles were elicited nonetheless. Repeatability of emoji responses was high and data with good face validity was obtained from American and Chinese consumers. Emoji responses from groups of consumers who liked/disliked a focal stimulus were different (in the expected directions) and frequency of consumption also influenced emoji responses. The preliminary investigations reported here suggest that emoji may have potential as a method for direct measurement of emotional associations to foods and beverages. Additional research is required to further develop this emoji-based approach, including assessment of its pros and cons of and performance relative to existing tools. This research contributes new understanding of the role of consumption context. 12 studies involving 1727 consumers were conducted as central location tests (CLT). Using between-subjects experimental designs, the influence of evoked contexts relative to control condition for hedonic responses and sensory product characterization using check-all-that-apply (CATA) questions was evaluated. Contexts were evoked by asking consumers to think of the last time they ate/drank the focal product category or by imagining a specific consumption situation where product consumption would take place (e.g., breakfast on a weekend morning). In half of the studies, consumers were asked to provide a description of the consumption context they imagined. There was no consistent trend in the results regarding the influence of evoked context of hedonic responses. Effects were seen in a minority of studies, but not consistently showing differences in mean scores or changes in sample discrimination. The type of context that was evoked (last time ate/drank vs. specific situation) did not systematically influence the results, although the use of a description phase during context evocation was detrimental to hedonic sample discrimination. In all the studies where participants were asked to describe the context they imagined, hedonic discrimination was inferior to that achieved under no evoked context. Sensory responses to CATA questions were highly similar under evoked and no evoked context and suggested that product characterisations generated by consumers in a typical CLT provide a good proxy for sensory product experience in contextualised consumption situations.
