As a contribution to the development of novel vanadium complexes with pharmacologically interesting moieties, new dioxovanadium( V) semicarbazone complexes with the formula cis-VO2L, where L = 5-bromosalicylaldehyde semicarbazone and 2-hydroxynaphtalen- 1-carboxaldehyde semicarbazone have been synthesized. and characterized by 1H and 13C NMR, Raman and FTIR spectroscopies. Results were compared with those previously reported for other three analogous complexes of this series. The five complexes were tested in three different human tumor cell lines for bioactivity as potential anti-tumor agents, showing selective cytotoxicity on TK-10 cell line. Results showed that structural modifications on the semicarbazone moiety could have a significant effect on the anti-tumor activity of the vanadium complexes. In addition, the electrochemical behavior of all the complexes was studied. No apparent correlation could be demonstrated between reduction potentials of the complexes and their anti-tumor activities. The molecular structure of the novel [VVO2(5-bromosalicylaldehyde semicarbazone)] complex was solved by X-ray diffraction methods. The vanadium atom shows a distorted square pyramidal coordination sphere. The (VO2)+ cation is coordinated to a nearly planar (L)� anion acting as a tridentate ligand through both oxygen and one nitrogen atoms. The new dioxo(semicarbazone)vanadium(V) complexes cis-VO2L, where L = salicylaldehyde semicarbazone (L1), salicylaldehyde 4-n-butylsemicarbazone (L2), or salicylaldehyde 4-(2-naphthyl)semicarbazone (L3), have been synthesized, characterized by 1H and 13C NMR and FTIR spectroscopy and tested for bioactivity as potential insulin-mimetic agents. All dioxovanadium(V) complexes exhibited essentially no in vitro insulin-mimetic activity, but the VO2L2 complex developed activity in the presence of ascorbic acid, similar to that of vanadyl sulfate. The molecular structure of the novel complex VO2L1 has been solved by X-ray diffraction methods. It crystallizes in the tetragonal space group P42/n with a = 12.7674(7), c = 11.5308(5) Å, and Z = 8. The vanadium atom is in a distorted square-pyramidal coordination, with L1 acting as a tridentate ligand through its azomethyne nitrogen atom, carbonyl oxygen atom and deprotonated phenol oxygen atom. Background: Brazil is currently debating the implementation of front-of-package labels. This study tested if Warning labels (WLs) improved consumer understanding, perceptions, and purchase intentions compared to Traffic-Light labels (TLLs) in 1607 Brazilian adults. Methods: In this online, randomized controlled experiment participants saw images of 10 products and answered questions twice—once in a no-label, control condition and then again in a randomly assigned label condition. The relative differences in responses between WLs and TLLs between control and label conditions were estimated using one-way ANOVAs or Chi-square tests. Results: Presenting WLs on products compared to TLLs helped participants: (i) improve their understanding of excess nutrient content (27.0% versus 8.2%, p < 0.001); (ii) improve their ability to identify the healthier product (24.6% versus 3.3%, p < 0.001); (iii) decrease perceptions of product healthfulness; and (iv) correctly identify healthier products (14.0% versus 6.9%, p < 0.001), relative to the control condition. With WLs, there was also an increase in the percentage of people: (v) expressing an intention to purchase the relatively healthier option (16.1% versus 9.8%, p < 0.001); and (vi) choosing not to buy either product (13.0% versus 2.9%, p < 0.001), relative to the control condition. The participants in the WL condition had significantly more favorable opinions of the labels compared to those in the TLL group. Conclusions: WLs would be more effective, compared to the TLL, at improving consumer food choices. To select the best biomarkers of tacrolimus pharmacodynamics, we explored the strength and variability of signal transduction and the influence of polymorphisms along the calcineurin pathway. </div> <div id="sec-2" class="subsection"> METHODS: Peripheral blood mononuclear cells from 35 healthy volunteers were incubated with tacrolimus (0.1–50 ng/mL) and stimulated ex vivo. Inhibition of NFAT1 (nuclear factor of activated T cells 1) translocation to the nucleus and intracellular expression of interleukin-2 in CD4+ and CD8+ T cells and the surface activation marker CD25 in CD3+cells were measured by flow cytometry. We sequenced the promoter regions of immunophilins and calcineurin subunits and characterized selected single nucleotide polymorphisms in the genes of the calcineurin pathway with allelic discrimination assays. </div> <div id="sec-3" class="subsection"> RESULTS: All responses closely fitted an I/Imax sigmoid model. Large interindividual variability (n = 30) in I0 and IC50 was found for all biomarkers. Les inhibiteurs de la calcineurine (ICN) sont les immunosuppresseurs les plus employés en transplantation d’organe et, malgré leur toxicité et leur efficacité imparfaite, ils resteront les premières options thérapeutiques dans un avenir proche. Leurs effets présentent une large variabilité intra- et inter-individuelle, qui n’est pas expliquée par les différences de doses, de concentrations ou d’aires sous la courbe des concentrations en fonction du temps, ce qui limite les bénéfices du suivi thérapeutique pharmacologique et suggère que d’autres facteurs contribuent à la variabilité de la réponse. Comme les ICN ciblent la fonction des lymphocytes T (LT), la concentration des ICN dans les LT a été étudiée comme marqueur intermédiaire de la fonction immunitaire. Effectivement, les concentrations intra-lymphocytaires de TAC chez des transplantés hépatiques montraient une plus forte association avec le rejet aigu que les concentrations dans le sang total, probablement parce qu’elles intègrent la variabilité inter-individuelle d’entrée du médicament dans les LT. De la même manière, des études antérieures ont montré que les concentrations intra-hépatiques de TAC étaient très bien corrélées avec les scores de rejet de Banff, alors que les concentrations résiduelles dans le sang ne l’étaient pas. Les deux pourraient représenter des biomarqueurs pharmacocinétiques intéressants, quoique difficiles à mesurer. Pour se qualifier comme index d’activité immunosuppressive des ICN, un biomarqueur pharmacodynamique (PD) devrait être directement affecté par l’activité du médicament, c'est-à-dire ne pas être trop éloigné de la cible du médicament et en même temps proche de ses effets cliniques. Comme de nombreuses preuves accréditent le concept que la suppression des LT est le mécanisme clé par lequel les ICN induisent une immunosuppression permettant de prévenir le rejet cellulaire, la fonction et l’activation des LT sont des candidats séduisants pour les stratégies de suivi PD. Toutefois, à ce jour aucun biomarqueur PD ne présente tous les prérequis idéaux, c'est-à-dire est à la fois non-invasif, fiable, sensible, spécifique, reproductible et disponible rapidement. Afin d’identifier des biomarqueurs PD très spécifiques de l’inhibition de la calcineurine et reflétant une part importante de la variabilité inter-individuelle, nos travaux avaient pour objectifs d’explorer la pharmacodynamie des ICN, la force et la variabilité de la Ofelia Noceti Penza | Thèse de doctorat | Université de Limoges | 2015 9 of 298 transmission du signal le long de l’axe calcineurine, ainsi que les étapes où les sources de variabilité PD internes (génétiques) ou externes sont les plus influentes. Dans ce but, nous avons mesuré simultanément la translocation de NFAT1 dans le noyaux des cellules mononuclées du sang périphérique (PBMC) (NFTA1 étant l’isoforme prédominante de NFAT dans les LT quiescents ou activés), l’expression intracellulaire d’IL-2 dans les sous-populations lymphocytaires CD3+, CD4+ et CD8+, et l’expression membranaire de CD25 (IL-2Rα), un marqueur de surface de l’activation des LT. Un essai clinique non-interventionnel a été mis en place chez des volontaires sains (VS), des patients inscrits en liste d’attente de transplantation hépatique (PLA) et des patients transplantés hépatiques (PTH). Une question différente a été étudiée dans chaque groupe : HS et PLA ont été inclus pour étudier la réponse des LT au tacrolimus (TAC) exvivo dans des conditions de stimulation, ainsi que les sources génétiques potentielles de variabilité PD, respectivement dans des conditions physiologiques et pathologiques. Les PTH ont été recrutés pour étudier les relations pharmacodynamiques, pharmacogénétiques et pharmacocinétiques dans des situations d’administration de tacrolimus ou de ciclosporine, c'est-à-dire d’activité PD résiduelle mesurée ex-vivo en conditions de stimulation ou non. Le petit groupe de PTH (n=6) inclus juste avant la transplantation et suivi de façon répétitive pendant la première année post-greffe était prévu pour explorer les relations entre PD des ICN et réponses cliniques. L’étude chez les VS (n=35) : a exploré la PD du TAC au long de la voie calcineurine en exposant des PBMC ex-vivo au médicament ; a modélisé la transmission du signal au long de cette voie ; a examiné la variabilité inter-individuelle des paramètres pharmacodynamiques du TAC ; et a étudié les sources de cette variabilité et leur contribution à chaque étape de la voie calcineurine. De plus, elle nous a permis d’évaluer la variabilité analytique de nos techniques ainsi que la variabilité intra-individuelle des paramètres PD du TAC. Les PLA (n = 19) nous ont permis de confirmer les résultats obtenus chez les VS, ainsi que de tester l’influence potentielle de leur maladie initiale sur la pharmacodynamie ex-vivo du TAC. Les buts de l’étude transversale chez des PTH (n=80) étaient d’explorer la variabilité inter-individuelle de la PD des ICN dans des situations cliniques réalistes et les sources pharmacogénétiques potentielles de cette variabilité. Ofelia Noceti Penza | Thèse de doctorat | Université de Limoges | 2015 10 of 298 Nous avons mesuré par cytométrie en flux la présence de NFTA1 transloqué dans le noyau des PBMC (qui est très proche de l’activité phosphatase de la calcineurine et probablement plus spécifique que la mesure « directe » de son activité, qui nécessite d’inhiber d’autres activités phosphatases), ainsi que la fonction (IL-2) et l’activation (CD25) des LT comme marqueurs du statut de l’immunité cellulaire. Les PBMC ont été séparés par gradient de centrifugation à partir d’échantillons de sang total recueillis avant l’administration du médicament et à jeun. Des aliquots frais de 106 PBMC dans 100 μl de RPMI supplémenté ont été employés pour toutes les analyses. Les PBMC des VS et des PLA ont été exposés ex-vivo pendant 30 min à des concentrations de TAC entre 0 et 50 ng/mL avant chaque mesure. Les mesures de NFAT1 étaient précédées d’une stimulation polyclonale par du phorbol 12-myristate 13-acétate (PMA) à 50 ng/mL et du calcium ionophore (I) à 2.5 μg/ml, à 37°C dans une atmosphère humidifiée avec 5% CO2 pendant 30 min, pour garantir la translocation de NFAT1 vers le noyau. Après deux lavages avec du PBS 1X froid, les cellules étaient incubées dans la glace pendant 30 min avec du tampon de lyse PIPE (pH 7,4 dans le PBS 1X) pour perméabiliser les noyaux cellulaires. Après deux nouveaux lavages avec de la BSA à 1% dans le PBS 1X, 50 μL d’un mélange d’anticorps anti-NFAT1 à 5μg/ml dans le PBS 1X étaient ajoutés au milieu pour 30 min d’incubation dans la glace. Puis des Ig polyclonales à 0,33% marquées au PE (fragment F(ab’)2 de chèvres anti- IgG(H+L) de souris) étaient ajoutées. Après 15 min d’incubation dans la glace à l’abri de la lumière, le signal fluorescent résultant des noyaux de PBMC marqués a été mesuré par cytométrie de flux et l’intensité de fluorescence moyenne de NFTA1 dans les noyaux isolés utilisée pour les analyses statistiques. La mesure d’IL-2 intra-cellulaire a nécessité 5h d’activation polyclonale avec du PMA/I et de la bréfeldine à 1 μg/ml (pour arrêter la sécrétion de protéines) à 37% en atmosphère humidifiée avec 5% de CO2. Les cellules étaient alors : i) marquées avec des marqueurs de surface anti-LT humain (CD3 PerCP-Cy5.5, CD4 FITC, CD8 PE-Cy7); ii) fixées : iii) perméabilisées (IntraPrep) ; iv) marquées avec des anticorps PE de rat anti-IL-2 humains et avec des IgG2a κ PE de rat comme contrôle isotypique. Pour la mesure d’expression de CD25, des aliquots de PBMC ont été incubés pendant 72h à 37°C et 5% de CO2 en présence de 7,5 μg/ml de Concanavaline A dans du RPMI 1640 1X. L’analyse par cytométrie de flux était pratiquée après 30 min d’incubation dans le noir et deux lavages avec de la BSA dans du PBS 1X. BACKGROUND: We sought to evaluate, in patients on a liver transplantation waiting list, potential biomarkers of the base calcineurin pathway activity with use of a new model of nonstimulated peripheral blood mononuclear cells (PBMC) and ex vivo response to tacrolimus (TAC). METHODS: The calcineurin pathway activity was explored ex vivo in stimulated and nonstimulated PBMC from 19 patients. The inhibition of NFAT1 translocation to PBMC nuclei, expression of intracellular IL-2, and membrane CD25 in different T-cell subsets were measured by multiparametric flow cytometry before and after exposure to TAC. We also studied the influence on the individual response of polymorphisms in 3 key genes of the calcineurin pathway: PPIA, PPP3CA, and IL2RA. RESULTS: All pharmacodynamics profiles closely fitted an I/Imax sigmoid model. Interindividual variability was higher in nonstimulated than in stimulated conditions, as well as in the presence of TAC. IL-2+CD8+ cells at TAC Imax showed the highest interindividual variability, suggesting its usefulness as a biomarker of individual TAC effects integrating many different sources of regulation and variability. Moreover, in the absence of TAC, patients with end-stage liver disease exhibited lower NFAT1 translocation and T-cell activation than healthy volunteers from a previous study under similar conditions. Multivariate statistical analysis showed strong and significant associations between TAC pharmacodynamic parameters and 2 polymorphisms in the gene-coding cyclophilin A (rs8177826 and rs6850). CONCLUSIONS: We show the feasibility of using nonstimulated PBMCs to explore the calcineurin pathway under more physiologic conditions and point toward potential biomarkers for TAC pharmacodynamic monitoring. Films of heavy metal halides (mercuric iodide, lead iodide, bismuth tri-iodide, lead bromide, mercuric bromide and mercuric bromide-iodide), 1” x 1” and 2” x 2” in area, have been grown by physical vapor deposition onto alumina and glass substrates with conductive coatings. From the point of view of film growth the materials was found to have similar behavior, which was evaluated by studying grain size and texture of the films as a function of growth temperature. Films grow oriented with the (0 0 l) crystalline planes parallel or perpendicular to the substrate, according to X-ray diffraction. The influence of film orientation on electrical properties and on response to radiation was evaluated by measuring resistivity and response to X-rays. All the layers give good linearity of response to an X-ray beam. The sensitivity of the layers (signal to dark relation / exposure rate) is maximum (1380) for mercuric iodide. For all the materials, the more oriented the films, the lowest the dark current, and the higher the sensitivity and the signal/dark relation. A superior correlation between electrical and response properties and the layer orientation can be deduced for films of the family of heavy metal halides, observed as a whole. El término “arenas negras” hace referencia a los minerales pesados que pueden ser encontrados en depósitos denominados Placer; estas arenas pueden ser generadas y concentradas por la acción de agentes naturales como corrientes de agua, y acción de las olas y el viento a lo largo de la playa, así como también por procesos extractivos. Sin importar si son de origen natural o de minería las arenas negras son producto de descomposición y ruptura de rocas, seguido por concentración por gravedad. En estos procesos en la mayoría de los casos la composición de la arena hace que tome color negro o marrón oscuro, de ahí la denominación. Debido a que se forman por desgaste de rocas los minerales componentes de las arenas negras son en general resistentes química y mecánicamente. Es entonces un yacimiento formado por la concentración de minerales en la arena de ríos, arroyos o playas como consecuencia de su elevada densidad (mayor que el cuarzo), minerales que deben ser al mismo tiempo resistentes a la abrasión. Los placeres se clasifican en depósitos residuales y depósitos transportados. En los primeros los minerales han sido más o menos liberados por desgaste y el depósito posee más o menos la misma composición que la roca original, de esta manera los minerales no han sido concentrados, mientras que en los segundos los minerales generados por desgaste se han desplazado de la roca original y se han re-depositado (generalmente por acción del agua corriente). En estos proceso de trasporte y re-deposición los minerales inestables se solubilizan, los minerales más livianos viajan más que los más pesados generándose depósitos concentrados. Estos últimos son los más estudiados, debido a su importancia comercial. Los minerales que se pueden encontrar en un depósito de arena negra dependen del tipo de roca de la cual el depósito proviene; las arenas negras por lo tanto en general son una mezcla de varios minerales pero pueden consistir enteramente de uno o dos minerales. Los minerales cuya densidad específica es menor a 6 g.cm-3 no pueden ser concentrados por procesos de transporte fluvial debido a que la diferencia de densidad con el sedimento base no es lo suficientemente grande para poseer modos de transporte diferencial en la mayoría de los regímenes hidráulicos turbulentos (de composición base cuarzo o feldespato cuya densidad específica es 2.7 g.cm-3). Este es el caso de los minerales detallados en la Tabla 1, pero que sí pueden ser concentrados por los sistemas de arenas costeras (Spencer Cronan 1999). Los depósitos de costa se acumulan en las playas, las estructuras son laminares o tubulares sub-horizontales, la acumulación de volúmenes significativos requiere de múltiples transgresiones o acción simultánea del viento. Los depósitos de mar adentro se producen por acciones de las olas durante las tormentas, removiendo materiales de 80 a 120 m de profundidad (Ferrando et al. 2003). A mixed Mg2+/Al3+ oxide featuring an incipient spinel phase was employed in the catalytic decomposition of a range of phosphate triesters, including the neurotoxic biocide chlorpyrifos methyl O-analog, with estimated catalytic effects up to 104-fold. The target reaction, namely, the transesterification with 1-propanol, was conveniently chosen as a means to convert toxic organophosphorus substrates into trialkyl phosphates structurally related to a family of flame retardants. Catalytic efficiency depends upon the stereo-electronic properties of the substrate, with both leaving group ability and geometric factors displaying pivotal effects on the rate constants. To evaluate the adsorption of 1-propanol/propoxide and the phosphotriester methyl paraoxon over the MgAl2O4(100) surface, density functional theory calculations with periodic boundary conditions were performed, which showed that the most probable reactants’ conformation prior to the reaction is such that propoxide is bound to a Mg2+ center with a juxtapositioned methyl paraoxon molecule at a close, neighboring Al3+ site. Monitoring of veterinary drug residues in foods is often conducted using liquid chromatography/tandem mass spectrometry (LC/MS/MS). Results have high economic stakes for producers, but the ions monitored are usually selected due to signal intensities without structural interpretation. In this study, the ion transitions were characterized by high-resolution mass spectrometry. The 62 veterinary drugs from the LC/MS/MS method consisted of sulfonamides, β-lactams, phenicols, macrolides, tetracyclines, fluoroquinolones, non-steroidal anti-inflammatory drugs (NSAIDs), and corticosteroids. They were individually infused into a quadrupole time-of-flight (Q-TOF) mass spectrometer using electrospray ionization (ESI) operated in positive mode. The MS and collision-induced dissociation (CID) MS/MS spectra for each analyte were obtained for structural elucidation. The Q-TOF instrument was calibrated to obtain a mass accuracy error <5 ppm for the MS and MS/MS spectra. The use of high-resolution ESI-Q-TOF-MS for the generation of the MS/MS product ions allowed for the determination of chemical formulae for the analytes, some of which led to new findings. Assigned structures were based on rational interpretation of the most stable possible products with comparison with the scientific literature. In difficult cases, isotopically labeled drugs or hydrogen/deuterium (H/D) exchange experiments were used to help confirm the structures of the product ions. The use of ESI-Q-TOF-MS in this study has allowed structure elucidation of 186 MS/MS product ions previously selected for the LC/MS/MS analysis of 62 veterinary drugs. This serves to reduce the chances of false positives and negatives in the monitoring program, and provides justification and defense in regulatory enforcement actions. Nitrated fatty acids (nitroalkenes) have been recently detected and quantified in cell membranes and human plasma. However, nitration of arachidonate (AA), that could redirect AA-dependent cell signaling pathways, has not been studied in detail. Herein, we synthesized and determined for the first time the isomer distribution of nitroarachidonate (AANO2) and demonstrate its ability to modulate inflammation. Synthesis of AANO2 was achieved by AA treatment with sodium nitrite in acidic conditions following HPLC separation. Mass spectrometry (MS) analysis showed the characteristic MS/MS transition of AANO2 (m/z 348/301). Moreover, the IR signal at 1378.3 cm-1 and NMR studies confirmed the presence of mononitrated nitroalkenes. Positional isomer distribution was determined by NMR and MS fragmentation with lithium; four major isomers (9-, 12-, 14-, and 15-AANO2) were identified, which exhibited key anti-inflammatory properties. These include their ability to release biologically relevant amounts of nitric oxide, induce cGMP-dependent vasorelaxation, and down-regulate inducible nitric oxide synthase (NOS2) expression during macrophage activation, providing unique structural evidence and novel regulatory signaling properties of AANO2. Biocatalytic approaches to the synthesis of optically pure chiral amines, starting from simple achiral building blocks, are highly desirable because such motifs are present in a wide variety of important natural products and pharmaceutical compounds. Herein, a novel one-pot ω-transaminase (TA)/monoamine oxidase (MAO-N) cascade process for the synthesis of chiral 2,5-disubstituted pyrrolidines is reported. The reactions proceeded with excellent enantio- and diastereoselectivity (>94 % ee; >98 % de) and can be performed on a preparative scale. This methodology exploits the complementary regio- and stereoselectivity displayed by both enzymes, which ensures that the stereogenic center established by the transaminase is not affected by the monoamine oxidase, and highlights the potential of this multienzyme cascade for the efficient synthesis of chiral building blocks.
