Tapeworms (Cestoda) cause neglected diseases that can be fatal and are difficult to treat, owing to inefficient drugs. Here we present an analysis of tapeworm genome sequences using the human-infective species Echinococcus multilocularis, E. granulosus, Taenia solium and the laboratory model Hymenolepis microstoma as examples. The 115- to 141-megabase genomes offer insights into the evolution of parasitism. Synteny is maintained with distantly related blood flukes but we find extreme losses of genes and pathways that are ubiquitous in other animals, including 34 homeobox families and several determinants of stem cell fate. Tapeworms have specialized detoxification pathways, metabolism that is finely tuned to rely on nutrients scavenged from their hosts, and species-specific expansions of non-canonical heat shock proteins and families of known antigens. We identify new potential drug targets, including some on which existing pharmaceuticals may act. SUMMARY According to the literature, cyclosporine in healthy volunteers yielded significant lower clear- ances than in patients with renal failure, uremic, or with kidney transplant. This could be due to a de- creased renal blood flow these patients have and the negligible renal metabolism cyclosporine exhibits. However, in the case of tacrolimus, literature shows almost the same clearance in healthy volunteers and patients with renal impairment. This fact might be related with its dual ability of being eliminated, splanchnic and renal metabolism, and the relevant impact of CYP3A5 polimorfism. Eighty eight kidney transplant recipients under tacrolimus treatment were enrolled. Whole blood tacrolimus levels were determined and evolution of apparent clearance with serum creatinine was assessed. In some patients an increase in serum creatinine produced an increase in tacrolimus clearance, whereas others displayed an inverse effect with increasing serum creatinine. This fact gives new evidence about the impact renal metabolism would have on tacrolimus pharmacokinetics. Temporal Check-All-That-Apply (TCATA) is a multi-attribute temporal approach that extends CATA questions. It is based on continuous selection of the sensory attributes that are perceived as applicable for describing a focal sample during consumption. Compared to CATA, TCATA is a relatively intense and demanding task for consumers who have to focus their attention on the evolution of the sensory characteristics of samples during consumption. The present work evaluated a short familiarization step (7–10 min) where participants became acquainted with the list of sensory terms and applied these to a practice sample using a CATA task and also completed a TCATA task using a warm-up sample. All participants, including those who did not take part in the familiarization step, watched a demonstration video that explained how to use the TCATA software and to complete the task. Three consumer studies, each involving &sim;100 participants, were conducted using dried apricots, peanuts, and milk chocolate. The TCATA curves when data were collected with and without the familiarization step were not systematically different. However, a small increase in sample discrimination was established and on this basis familiarization should be considered when using TCATA with consumers. A need to explore other familiarization protocols than the one used here exist in order to develop guidelines for implementation. To perform a properly approach on this issue the determination of total concentration of minerals is not sufficient, it is necessary to know about the bioaccessibility of each element. Due to the widespread pollution problems, food is not only a source of essential minerals but also contributes to the intake of toxic elements by human beings. To asses both, nutritional properties and food safety of raw edible globe artichoke heads, the concentrations of total arsenic (As), cadmium (Cd), copper (Cu), iron (Fe), nickel (Ni), lead (Pb) and zinc (Zn) were determined in 10 samples from Uruguayan markets. Analytical determinations were performed using flame and electrothermal atomic absorption spectrometry. In all samples, the decreasing sequence of the mean total concentrations was: Fe > Zn > Cu > Ni with values ranges between 35.8 and 57.8 mg kg− 1 (Fe), 27.7 to 42.2 mg kg− 1 (Zn), 5.4 to 7.5 mg kg− 1 (Cu) and 1.9 to 3.4 mg kg− 1 (Ni). In vitro bioaccessibility assays showed that the decreasing sequence of bioaccessibility for these elements was Cu > Zn > Ni > Fe with bioaccessibility factors percentages ranged between 71.3 and 82.3% (Cu), 51.2–64.2% (Zn), 41.3–54.7% (Ni) and 39.5–49.7% (Fe). Total concentrations of As, Cd and Pb were far below the maximum limits established by regional regulations in all samples, presenting negligible bioaccessibility. Results demonstrate that raw edible artichoke heads are a good source mainly of Cu, Zn with an interesting contribution to the daily intake of these essential elements. This novel information provides a starting point for more complex and in-depth evaluations from the nutritional point of view, including the establishment of a recommended daily dietary intake for a non-traditional food. All pesticides were analyzed at or below their Maximum Residue Levels fixed for globe artichoke by the European Union. The method was successfully applied for the analysis of commercial samples. Maintaining redox homeostasis is a major adaptive problem faced by parasites and its disruption can shift the biochemical balance toward the host. Platyhelminth parasites possess a streamlined thiol-based redox system in which a single enzyme, thioredoxin glutathione reductase (TGR), a fusion of a glutaredoxin (Grx) domain to canonical thioredoxin reductase (TR) domains, supplies electrons to oxidized glutathione (GSSG) and thioredoxin (Trx). TGR has been validated as a drug target for schistosomiasis. Recent Advances: In addition to glutathione (GSH) and Trx reduction, TGR supports GSH-independent deglutathionylation conferring an additional advantage to the TGR redox array. Biochemical and structural studies have shown that the TR activity does not require the Grx domain, while the glutathione reductase and deglutathionylase activities depend on the Grx domain, which receives electrons from the TR domains. The search for TGR inhibitors has identified promising drug leads, notably oxadiazole N-oxides. Critical Issues: A conspicuous feature of platyhelminth TGRs is that their Grx-dependent activities are temporarily inhibited at high GSSG concentrations. Six from twelve volunteers, to whom 600 mg every 72 h or 100 mg every 12 h of phenytoin were administered during 10 days, completed a two-way, two-period crossover study. Two fractions of stimulated saliva were collected simultaneously with plasma samples throughout a scheduled 24-hour period on day 10, and during the subsequent 4-day washout period. Average plasma (P) concentrations of both the parent drug (p<0.01) and the main metabolite (p-hydroxyphenylphenytoin, p<0.05) showed higher values when 600 mg each 72 h were given. Also, the saliva first-fraction (S1) average concentration was higher (p<0.05) after 600 mg every 72 h. Conversely, S1/P average concentration ratio was higher (p<0.01) after 100 mg every 12 h. Half-lives, metabolic ratios, saliva second-fraction (S2) average concentrations, and S1/S2 ratios of average levels were not significantly different between treatments. Results reveal that, even though the input rate was kept constant, higher doses given at long dosing intervals displayed higher systemic exposures and lower saliva/plasma exposure ratio than lower doses given at short intervals . Our hypothesis, supported by studies carried out with rats and theoretical approaches, is that efflux transporter overexpression at hepatocytes and at salivary cells would be induced by the higher local phenytoin exposures provoked by 600 mg every 72 h and by the persistent systemic exposures provoked by 100 mg every 12 h, respectively. The maintenance of systemic phenytoin concentration would be foreseen as one of the causes leading to antiepileptic treatment refractoriness, since a higher brain-to-plasma transportation of drug could be induced. Maybe once-daily dosing of phenytoin would be preferable to twice or three times a day administration regimens. Despite the fact that avoiding peak-trough oscillation was always the aim of pharmacotherapy, it might be more appropriate for patients under phenytoin treatments to increase both dose and dosing interval in order not to contribute to the installation of pharmacoresistance. A large body of data exists about the interaction between curved π systems and C60 or C70. However, little is known about the interaction with fullerenes smaller than C60 or larger than C70. To fill that gap, we studied, by means of density functional theory (M06-2X), the interaction between corannulene, pentaindenocorannulene, C60H28 buckycatcher and the following fullerenes: C44, C50, C80, C90, C100, C180 and C240. For fullerenes smaller than C60, their high reactivity facilitated the covalent addition to the hosts assayed. Yet, the reaction energies determined for the covalent addition were comparable to those calculated for the formation of supramolecular complexes. Thus, the receptor may host a fullerene and at least have another one attached. As expected, for fullerenes larger than C70, supramolecular complexes were preferred over covalent assemblies. The binding energies with bowls increased with the size of the fullerenes in a non-monotonic fashion since they depended on the shape of the fullerene. Indeed, for one C80 isomer, it is possible to find a region which forms a complex with corannulene that is stronger than C60@corannulene, while another region exists whose interaction with corannulene is weaker. As the size of the fullerene becomes larger, ball–socket interactions are weakened, and CH–π interactions become important, accounting for the large interaction determined for corannulene and graphene. Finally, for the buckycatcher, the maximum encapsulation energy among the fullerenes assayed was displayed by C90. Efficiency of in-situ alkaline transesterification of dry biomass from a crop of Spirulina sp., using ultrasound-assisted and/or the addition of different co-solvents to the reaction medium was studied. The starting biomass had a total lipid content of 7.0% (Folch method), comprised mainly by the following fatty acids: 16:0 (44.2%), 18:3 (n–6) (23.3%), and 18: 2 (n–6) (11.1%). When the transesterification reaction was carried out without ultrasound, it only extracted 12.8% of lipids (0.9% of the biomass) after 2 h of reaction, achieving only a 63.6% ester content. When the process was carried out under the same conditions except for the ultrasound-assisted (80 W), the weight yield remained mostly unchanged, but the ester content increased to 97.6%. Additionally, when chloroform was added to methanol as a co-solvent in a 2:1 ratio it was possible to extract 43% of lipids. However, the ester content was reduced to 69.7%. Moreover, with maximum ultrasound power (180 W) a high ester content (96.9%) and a relatively high extraction yield (26%) were obtained, even without the co-solvent. These results show that the ultrasound significant increased the yield of the in-situ alkaline transesterification of Spirulina sp. biomass, promoting a higher percent recovery of lipids as well as ester content from the starting material. A novel method was developed for the determination of zinc in wine by direct-injection electrothermal atomic absorption spectrometry (ETAAS). Unlike the OIV (Organisation Internationale de la Vigne et du Vin) reference method, which is based on flame atomic absorption spectrometry (FAAS), the proposed method does not require any sample preparation. Previous to the development of this method a reappraisal of the possible interferences found during the determination of zinc in wine by the reference OIV method was undertaken. Aiming at minimising sample preparation during FAAS method, alternatives such as the use of matched-matrix standards and the use of the Standard Additions Method (SAM) were studied, however different samples of Tannat wine produced in Uruguay exhibited widely different interference patterns, precluding the possible development of an alternate sample preparation-free FAAS method. Thus a new method based on direct-injection ETAAS was developed. The method is based on the atomic absorption of zinc at the alternate resonance wavelength of 307.6 nm. Samples are injected in the graphite furnace without preparation and the determination carried out by SAM. Quantification limit was 0.10 mg L− 1 and linearity was confirmed in the range 0.10–10 mg L− 1. Precision (repeatability, sr (%), n = 5) was calculated at three concentration levels and found to be better than 5%. When compared with the OIV reference method, results agreed within 10%. The method is fit for the purpose of routine analytical quality control, requiring no sample preparation and thus less labour and producing less chemical waste. The presence of the polymer host and interparticle Coulomb repulsive interactions was evaluated as an influence for the formation of ~3–8 nm equally-sized quantum dots independently of the concentration. The polyaniline polaron population showed an increase for the quantum dots diluted regime and the suppression at the concentrated regime, ascribed to the formation of chemical bonds at the interface, which was confirmed by theoretical simulations. In agreement with the previous observation, the in situ growth of ceramic quantum dots in polyaniline host via water vapor flow diffusion could be very useful as a novel approach to prepare electrode materials for energy conversion and storage applications. This study aimed to evaluate the waste cooking oil (WCO) hydrolysis in ultrasonic system using lipase as catalyst. Lipase was produced by the fungus Aspergillus niger via solid state fermentation (SSF) using canola meal as substrate. Prior to the hydrolysis reaction, the lipase behavior when subjected to ultrasound was evaluated by varying the temperature of the ultrasonic bath, the exposure time and the equipment power. Having optimized the treatment on ultrasound, the WCO hydrolysis reaction was carried out by evaluating the oil:water ratio and the lipase concentration. For a greater homogenization of the reaction medium, a mechanical stirrer at 170 rpm was used. All steps were analyzed by experimental design technique. The lipase treatment in ultrasound generated an increase of about 320% in its hydrolytic activity using 50% of ultrasonic power for 25 min. at 45 °C. The results of the experimental design conducted for ultrasound-assisted hydrolysis showed that the best condition was using an oil:water ratio of 1:3 (v:v) and enzyme concentration of 15% (v/v), generating 62.67 μmol/mL of free fatty acids (FFA) in 12 h of reaction. Thus, the use of Aspergillus niger lipase as a catalyst for hydrolysis reaction of WCO can be considered as a possible pretreatment technique of the oil in order to accelerate its degradation. The inhibitory capacity of trypsin and elastase of extracts prepared from Achyrocline atureioidehojas flowers, Baccharis trimera leaves, Eugenia uniflora fruits and Schinus molle fruits was studied. The extracts showed differences for the same plant material depending on using water or ethanol as solvents, and were obtained under different time and temperature conditions. The elastase inhibitory action was low (< 30%) or almost absent in the extracts tested. For trypsin, all extracts showed inhibitory action and significant differences depending on solvent used, regardless of the extraction temperature. Extracts of S. molle showed the most potent inhibitory activity for trypsin. The aqueous extract of this plant obtained at 100 °C was selected to determine IC 50 and inhibition type. We describe the first outbreak of Klebsiella pneumoniae carbapenemase-producing K. pneumoniae (KPC-KP), the infection control measures adopted and the shift in resistance patterns of isolates during antibiotic treatment. The ST258 KPC-KP strain exhibited a multiresistant antibiotic phenotype including co-resistance to gentamycin, colistin and tigecycline intermediate susceptibility. Isolates before and after treatment had different behaviour concerning their antibiotic susceptibility and the population analysis profile study. Introducción: el consumo de cafeína es un hábito extendido a nivel mundial, destacándose el mate en los países del cono sur como una fuente importante de ingesta. Objetivos: valorar la ingesta de cafeína en el embarazo, el aporte de las diferentes fuentes de ingesta y su relación con el peso al nacer. Metodología y población: se realizó un estudio observacional, analítico de corte transversal que incluyó 240 puérperas y sus recién nacidos sanos de 35 semanas de gestación o mayores. Resultados: la media de edad materna fue de 24,96 (DE=6,01), Estaban casadas o convivían con su pareja 179 mujeres (74,58%). De ellas 67 (28,03%) eran primigestas. La media de controles del embarazo fue 7,31 (DE=3,32) y nacieron por cesárea 70 (29,17%). Las características de los recién nacidos reflejan los criterios de inclusión referidos: 35 semanas de gestación o mayores, vigorosos, internados junto a su madre luego del nacimiento. Durante todo el embarazo la fuente principal de cafeína fue la ingesta de mate. La media de consumo de cafeína fue de 273,99 mg/día; el consumo de mate es la principal fuente de cafeína con una media de 240 mg/día, aportando un 85% de la ingesta diaria de cafeína. Se observó consumo de cafeína mayor de 300 mg/día en 77 embarazadas, 32% de la muestra. La ingesta de cafeína mayor a 300 mg/día se asocia al nacimiento de un niño pequeño para la edad gestacional, la ingesta de 100 mg de cafeína por día disminuye el peso al nacer en 38 g, independientemente del índice de masa corporal y el hábito de fumar. Conclusiones: el consumo de cafeína es elevado en las embarazadas en Uruguay, superando los 300 mg diarios en 32% de la muestra. La ingesta de mate es la fuente principal de cafeína en esta población. El consumo de cafeína mayor a 300 mg diarios se asocia a mayor riesgo de recién nacidos pequeños para la edad gestacional. En vistas a la evidencia científica actual debería implementarse una adecuada información para lograr un descenso del consumo de mate y cafeína durante la gestación. Biocatalytic approaches to the synthesis of optically pure chiral amines, starting from simple achiral building blocks, are highly desirable because such motifs are present in a wide variety of important natural products and pharmaceutical compounds. Herein, a novel one-pot ω-transaminase (TA)/monoamine oxidase (MAO-N) cascade process for the synthesis of chiral 2,5-disubstituted pyrrolidines is reported. The reactions proceeded with excellent enantio- and diastereoselectivity (>94 % ee; >98 % de) and can be performed on a preparative scale. This methodology exploits the complementary regio- and stereoselectivity displayed by both enzymes, which ensures that the stereogenic center established by the transaminase is not affected by the monoamine oxidase, and highlights the potential of this multienzyme cascade for the efficient synthesis of chiral building blocks. The main objective of this work was to develop a kinetic model to describe the variation of the surface tension in an air-water interface due to the adsorption of proteins from different origins and to identify quantitatively the relevant parameters. It was considered that the processes of adsorption, unfolding and reordering of the protein molecule in the interface occur simultaneously. The model used in the present work to calculate the surface tension postulates the existence of two simultaneous processes, adsorption and protein rearrangement represented with an equation of first order with two exponential components. The relevant parameter of the equation are ka and kr—the rate constants of the two first order kinetic phases that correspond to both conformational states of the protein, adsorption and rearrangement during the process of variation of the surface tension, and the amplitude parameters Aa and Ar. The results suggest that the kinetic model for the variation of the surface tension of protein solutions proposed in this work, with two simultaneous first order processes, is more appropriate than previous models to describe such variation.
