An increasing global concern regarding energy saving and a growing pharmaceutical interest towards the adoption of industrial continuous processing generate an appropriate scenario for the development of solid dosage forms by moisture activated dry granulation (MADG) processes, which eliminate granulate drying and sizing, and minimize granulation end point control by making it less sensitive to shear and processing time. The aim of this study was to contribute to further enlighten formulation aspects of an attractive granulation process such as MADG, using statistical techniques. A 2^(4-1) fractional factorial design was introduced to study the effect of formulation variables on granulates obtained and on their tablets. Multivariate techniques were applied in the evaluation of the products. Lactose monohydrate and mannitol, povidone K15 and copovidone, and microcrystalline cellulose, partially pregelatinized corn starch and amorphous silica were selected, as diluents, binders and moisture absorbents, respectively. Contact angle and drop penetration time of diluents and moisture absorbents were used to study their wettability. All of the granulates yielded tablets with appropriate aspect and tensile strength, low friability, short disintegration time and in most of the cases, good weight uniformity. The statistical techniques showed the influence of excipient selection in the granulation process. Lactose produced granulates which presented lower median size and more rounded granules, leading to tablets which showed greater weight uniformity than the other diluent mannitol. The binder PVP K15 increased granule mechanical strength, producing finished tablets with higher tensile strength and longer disintegration time than copovidone. The use of a second moisture absorbent improved granulate flowability and contributed to prevent granulate caking. Wettability studies provided useful information for guidance on preliminary excipient selection. In summary, MADG is an innovative and economical process which facilitates quality by design approach.

En esta tesis se estudi6 la aplicaci6n de la granulaci6n por fusi6n como alternativa frente a los procesos por via humeda y via seca comunmente utilizados en la industria farmaceutica. Se investigaron excipientes de uso frecuente en formas farmaceuticas s61idas de administraci6n oral y equipos adecuados entre los de uso habitual en la industria farmaceutica local. Se disenaron dos procesos de granulaci6n por fusi6n que se diferencian en la fuerza de corte impartida allecho s6lido a granular durante la etapa de fusi6n: 1. proceso en lecho m6vil, que utiliza la tecnologia mas clasicamente empleada para granulaci6n por via humeda en la industria farmaceutica local: mezcladores de velocidad media 0 fuerza de corte media, en este caso con camisa, para formar masas aglomeradas, seguido de equipos formadores de granulos, como los granuladores oscilantes. Impone movimiento de fuerza de corte media al s6lido. 2. proceso en lecho estatico, que emplea un secador de cabina para la primera operaci6n tecnol6gica, seguido de una regularizaci6n por tamizado manual para terminar la granulaci6n. No imparte movimiento al s6lido. Los procesos fueron aplicados al s6lido inactivo almid6n de maiz, asi como a los principios activos carbamacepina y ketoprofeno. Los granulados fueron caracterizados en relaci6n a sus propiedades fisicomecanicas, incluyendo la aptitud para la compresi6n. Los comprimidos asi obtenidos tambien fueron caracterizados en relaci6n a sus propiedades fisicomecanicas. En el caso de los comprimidos conteniendo principios activos, la caracterizaci6n incluy6 ademas, las propiedades fisicoquimicas, en particular la liberaci6n de los activos. Entre los estudios de compresi6n se evalu6 tambien, la capacidad de estos procesos de granulaci6n por fusi6n para generar granulados inactivos aptos para ser empleados como excipientes de compresi6n directa. Para ello se fabricaron comprimidos de liberaci6n inmediata con la droga modele hidroclorotiazida. Los agentes granulantes seleccionados para lIevar a cabo las granulaciones por fusi6n mediante los procesos disenados comprenden excipientes tanto hidr6filos como hidr6fobos, usados solos 0 en combinaci6n: polietilenglicol 6000, povidona K30, palmitoestearato de glicerilo, acido estearico y monoestearato de glicerilo. A 10 largo del trabajo se emplearon herramientas estadisticas multivariadas, en concreto el analisis de componentes principales (peA) y el analisis de cluster, de forma de evaluar su uso para el estudio de los resultados de las propiedades farmacotecnicas ensayadas. Mediante el empleo de estas herramientas se busc6 contar con tratamientos de datos que facilitaran el analisis del importante numero de variables y la gran cantidad de datos generados normalmente por la cantidad de muestras que se ensayan en la evaluaci6n de granulados y comprimidos. Los resultados del presente trabajo permiten concluir que los dos procesos de granulaci6n por fusi6n (proceso en lecho m6vil y proceso en lecho estatico): J.- son capaces de dar productos de calidad farmacotecnica aceptable para varias materias primas de uso farmaceutico, mejorando el flujo y la compresibilidad de las mismas. J.- son sencillos, emplean equipos simples y con uso bien establecido en la industria farmaceutica, requieren control de pocas variables durante la producci6n y anticipan un escalado y una validaci6n de proceso simples. J.- pudieron utilizarse para producir comprimidos de Iiberaci6n inmediata con carbamacepina y con ketoprofeno, asi como de liberaci6n prolongada con ketoprofeno. J.- son capaces de dar granulados para ser usados como diluyentes de compresi6n directa. Por otra parte, las herramientas multivariadas seleccionadas para la evaluaci6n de los productos obtenidos probaron ser de utilidad para la interpretaci6n de los datos recogidos, facilitando el estudio de las propiedades de los granulados y la selecci6n de procesos de producci6n mas eficientes.

In this work, principal component analysis and cluster analysis were applied as helping tools to extract useful information in the development of formulations and manufacturing processes of melt granulates. Two melt granulation processes that differ in the shear stress applied to the solid bed during melting step were designed. These processes employ equipment frequently used in the local pharmaceutical industry. The selected binders include both hydrophilic and hydrophobic excipients, which were used alone or in binary mixtures. Granulates were characterized regarding their physicomechanical properties, including their compaction behavior. The resulting tablets were also evaluated. The selected multivariate statistical methods proved to be useful in facilitating the interpretation of the collected data and the study of the properties of granulates and tablets, as well as the selection of more efficient production processes.

The aim of the present work was to study the main formulation variables that influence attributes of bioadhesive emulgels based on a combination of polymers, using response surface methodology (RSM). Bioadhesive products continue to gain attention in topical cutaneous administration as they allow long residence times on the application site, which is important when a long dermal action and a reduced product administration frequency are desired. A Box-Behnken design of experiments (DoE) was introduced to study the effect of formulation variables on quality attributes of the emulgels. The effects of concentration of carbomer interpolymer type A (Polym1), xanthan gum (Polym2) and mineral oil (Oil) on detachment force (Fdetch), spreadability (Spread), and phase separation by mechanical stress (PhSep) were investigated. RSM and desirability functions were applied for data analysis. Emulgels were further characterized by viscosity and extrudability measurements. Polym1 showed a positive effect on Fdetch, while the increase in concentrations of Polym2 and Oil decreased this property. Polym1 and Polym2 favored emulgel PhSep. However, their interaction effect decreased it. The combination of 0.4-0.6% of carbomer and 0.2-0.3% of gum was able to produce easy-to-spread bioadhesive emulgels with mineral oil as discontinuous phase in the presence of a low surfactant concentration. Based on the DoE results, value ranges for the variables, which could achieve for the experimental domain to get the critical quality attributes of emulgels jointly within the specification limits, were able to be identified using RSM supported by desirability functions.

The chemistry of indazole and its N-oxide derivatives is very well-known. Indazole derivatives were<br />extensively studied as bioactive compounds, such as anti-aggregatory and vasorelaxant activity by NO release<br />and increase of cGMP levels and anticancer effects, antimicrobial and antiparasitic properties, among others.<br />Recently, the research and development in the medicinal chemistry of these systems have produced<br />compounds with contraceptive activities for men, for the treatment of osteoporosis, inflammatory disorders<br />and neurodegenerative diseases. On the other hand, indazole N-oxide derivatives were poorly studied as<br />bioactive compounds, but recently compounds with antiparasitic properties were produced. In this<br />presentation, recent developments in the chemistry and medicinal chemistry of indazole and its N-oxide<br />derivatives will be reviewed.

Quantitative structure-activity relationship (QSAR) studies, using the Comparative Molecular Field Analysis (CoMFA), on a series of 6,7-diarylpteridine derivatives showing nematocide properties have been carried out. The CoMFA model generated in the study has been used to estimate the namatocide activity (MIC50) of seven 6,7-diarylpteridines related to those previously studied. The model is highly predictive for all pteridine derivatives in the test set. Moreover, this model also predicts satisfactorily the nematocide activity of other [6] + [6] fused pyrazines (quinoxalines and pyridopyrazines) which were not represented in the training set. The analysis revealed the importance of steric factors (64.8%) and then the electrostatic ones (35.8%). Most pteridines under study were previously synthesized and tested as nematocide agents. Now, the synthesis and biological evaluation of 6,7-di-(2′-thienyl)-4(3H)-thioxo-pteridine 36 are reported.

Background Breast cancer is the second leading cause of cancer death worldwide. Nanotechnology approaches can overcome the side effects of chemotherapy as well as improve the efficacy of drugs. Dendrimers are nanometric size polymers which are suitable as drug delivery systems. To the best of our knowledge, studies on the application of PAMAM G4.5 (polyamidoamine half generation 4) dendrimers as potential drug delivery systems in breast cancer have not been reported. In this work we developed a PAMAM G4.5 dendrimer containing FITC (fluorescein isothiocyanate) dye to study their uptake by murine breast cancer cells and BALB/c mice breast tumors. Results We performed a reaction between FITC and PAMAM G4.5 dendrimers which were previously derivatized with piperazine (linker molecule), characterized them by 1H NMR (proton nuclear magnetic resonance) spectroscopy and MALDI-TOF (matrix-assisted laser desorption/ionization- time-of-flight) mass spectrometry. The experimental data indicated that 2 FITC molecules could be bound covalently at the PAMAM G4.5 dendrimer surface, with 17 FITC molecules probably occluded in PAMAM dendrimers cavity. PAMAM-FITC dendrimer (PAMAM G4.5-piperazinyl-FITC dendrimer) size distribution was evaluated by DLS (dynamic light scattering) and TEM (transmission electron microscopy). The nanoparticle hydrodynamic size was 96.3 ± 1.4 nm with a PdI (polydispersion index) of 0.0296 ± 0.0171, and the size distribution measured by TEM was 44.2 ± 9.2 nm. PAMAM-FITC dendrimers were neither cytotoxic in 4T1 cells nor hemolytic up to 24 h of incubation. In addition, they were uptaken in vitro by 4T1 cells and in vivo by BALB/c mice breast tumors. PAMAM G4.5-piperazinyl-FITC dendrimer intracellular distribution was observed through histologic analysis of the tumor by laser confocal microscopy.

The kinetics of reversible 3D aggregation processes was studied for different interparticle potentials by means of simulations. In previous work (Phys. ReV. E 2002, 65, 031405),1 freely diffusing particles were considered that aggregate whenever a collision occurs but disintegrate only with a single given breakup probability. The DLVO theory, however, predicts also interparticle potentials showing two minima of different depths separated by an energetic barrier. Hence, two different kind of bonds, primary and secondary ones, can be formed and should be treated separately. In the present work, this behavior was implemented by considering bonds with different breakup probabilities. The data obtained from simulations were compared with the stochastic solutions of the corresponding master equation. For this purpose, the Brownian kernel was employed together with novel fragmentation kernels. The agreement between the simulations and the kinetic description was found to be quite satisfactory. Moreover, studying the time evolution of the bond population showed that cluster aging appears as a natural consequence of the employed model.

Two limiting regimes for colloidal particle aggregation are well described in the literature: diffusion-limited cluster aggregation and reaction-limited cluster aggregation. Between these two limiting regimes, a vast transition region is expected. In this paper, the transition region is studied by means of static and dynamic light scattering. Therefore, a system of latex particles is aggregated at different electrolyte concentrations. The time dependence of the average diffusion coefficient is fitted considering the Brownian kernel and the kernel proposed by Schmitt et al. [Phys. Rev. E62, 8335 (2000)]. The first fits the experimental data only at high electrolyte concentrations while the latter, which considers multiple cluster–cluster contacts, is found to fit the complete set of experimental data.

Colloidalaggregation processes arising at different electrolyte concentrations were studied by means of experiments and confronted with theoretical predictions of different kinetic aggregationmodels. For this purpose, aqueous dispersions of relatively large polystyrene microspheres were chosen as experimental systems.Aggregation was induced by adding KBr electrolyte to the initially stable particle dispersions. During the aggregation processes, the cluster-size distribution was monitored by means of single cluster light scattering. Analyzing the time evolution of the monomer concentration, we found that the processes arising even at moderate electrolyte concentrations cannot be described by pure time-independent irreversible aggregationmodels. Hence, alternative models such as time-dependent irreversible aggregation and several reversible aggregationmodels were also tested. The model that considers a time-dependent sticking probability was found to fit the data quite satisfactorily. Nevertheless, the fitted was so slow that it seems not very likely to find such a behavior in real systems. The aggregation-fragmentation models reported in the literature were unable to reproduce the experimental observations. Hence, a more realistic reversible aggregationmodel was developed. This model accounts also for reenforced or double bonds between the constituent particles. The corresponding fit improved significantly and reached the same quality as the time-dependent model. Moreover, the obtained fitting parameters were in qualitative agreement with the DLVO predictions and so, reversible aggregation seems to be a more reasonable explanation for the experimental data than time-dependent irreversible aggregation. However, no definite statement on the possible secondary bond fragmentation mechanism may be made since both the applied shear stress in the measuring cell and thermal fluctuations can cause weaker bonds to break.

A summary of the resource.
In this paper, we present a stochastic model for obtaining the time evolution of the cluster size distribution for systems following coupled aggregation and sedimentation processes. Both, diffusion-limited cluster aggregation conditions and reaction limited cluster aggregation conditions are studied under the effect of sedimentation. For this purpose, the master equation is adapted for considering several slices where cluster aggregation and mass transport through the boundaries are taken into account. Furthermore, the kernel needed to solve the stochastic equation is adapted for considering the sticking probability, P, and the Peclet number, Pe, as parameters. The obtained solutions are then compared with the time evolution of the cluster size distribution obtained by means of computer simulations in Leone et al.

Smoluchowski’s equation is widely applied to describe the time evolution of the cluster-size distribution during aggregation processes. Analytical solutions for this equation, however, are known only for a very limited number of kernels. Therefore, numerical methods have to be used for describing the time evolution of the cluster-size distribution. In this work, we present a novel self-consistent method for solving Smoluchowski’s equation for any homogeneous kernel. The method considers dynamic scaling to be valid but does not need to assume a given form for the scaling distribution Φ(x). Moreover, the scaling distribution Φ(x) is obtained as a natural result of the algorithm. Due to the implementation of dynamic scaling concepts, the algorithm converges almost immediately with a minimal calculation effort. Comparing calculated size distributions with the corresponding analytical solutions shows the validity of the method. The method is then used to fit experimental data for diffusion limited aggregation. For this purpose, a fitting procedure is developed which allows us to fit the corresponding parameters for any given homogeneous kernel. As an application, a full comparison between the experimental data and the numerically obtained cluster-size distributions for the constant and the Brownian kernel was carried out.

In this work CD4-knockout mice were used as a model to analyse the role of CD4+ T cells in the antibody response against Echi­no­coccus granulo­sus immunization or experimental infection. Results obtained with mice immunized with protoscolex antigens indicated that these contain T-independent antigens. After infection, CD4-knockout mice and C57Bl/6 mice showed similar titres of specific antibodies indicating that T-independent antibody production was quantitatively important in early infection. We have also identified an antigenic fraction from protoscoleces (E4+) which induces CD4 T cell independent antibody response in early stages of infection. In conclusion, the results presented here directly support the existence of T-independent immunogens in E. granulo­sus protoscoleces and suggest that T-independent antibody response may be quantitatively important in early infection.

A summary of the resource.
Ibicella lutea is a ‘quasi-carnivorous’ plant that grows wild in Uruguay where it is used in popular medicine as an antiseptic for eye and skin infections. In an earlier screening, it showed a broad antibacterial spectrum. From the chloroform extract of the plant the main antibacterial compound has now been isolated and identified by several MS and NMR methods as a new compound, 11-O-(6%-O-acetyl-b-D-glucopyranosyl)-stearic acid.

A summary of the resource.
A diurnal variation study of carbamazepine (CBZ) and its metabolite carbamazepine-10,11-epoxide (EPOX) was carried out on six healthy volunteers, measuring drug levels in plasma and saliva throughout 34 hours. Four hundred milligrams of CBZ were given to subjects at 8:00 am or 8:00 pm, in a two-way crossover design. Drug and metabolite concentration in plasma and saliva stimulated with parafilm (after 24 hours post-dose) were higher during the night. The EPOX/CBZ metabolic ratio did not show any change between dark and light periods. The non-stimulated saliva/plasma concentration ratio (S/P) was higher during the day, but this quotient did not change when stimulated saliva was considered. Diurnal variation in the body blood flow seems to be a reasonable explanation for those differences. Factors such as protein binding or metabolic pathway are discarded as sources of carbamazepine chronopharmacokinetic variations.

Abstract
A summary of the resource.
In a screening of plants used traditionally in Nepal to treat diseases that could be caused by viruses, twenty-one methanol extracts from twenty species were quantitatively assayed for activity against three mammalian viruses: herpes simplex virus, Sindbis virus and poliovirus. Assays were performed in ultraviolet (UV)-A or visible light, as well as dark, and cytotoxicity was also noted. Impressive antiviral activities were exhibited by species of Bauhinia (Fabaceae), Carissa (Apocynaceae), Milletia (Fabaceae), Mallotus (Fabaceae), Rumex (Polygonaceae), Streblus (Moraceae), Terminalia (Combretaceae) and Tridax (Asteraceae). The Carissa extract was the most active, showing activity against all three viruses at a concentration of 12 #g/ml. Many of the other extracts showed partial inactivation of one or more test viruses.

A summary of the resource.
New 5-(2-arylethenyl)-1, 2, 4-triazine N-oxide and N, N′-dioxide derivatives were synthesized in order to obtain compounds as selective hypoxic cell cytotoxins. The desired products were obtained when the 5-methyl heterocycle reacted with the corresponding iminium electrophiles. The new compounds were tested for their cytotoxicity in oxia and hypoxia. Some of them proved to be less active in hypoxic conditions than Tirapazamine, 3-aminobenzo[1, 2-e]1, 2, 4-triazine N1, N4-dioxide. Derivative 11, 6-methyl-5-[2-(5-nitrofuryl)ethenyl)-1, 2, 4-triazine N4-oxide, was the most cytotoxic compound, but it was non-selective. Some derivatives were studied as DNA-binding agents in oxic conditions showing poor affinity for this biomolecule. This result showed that the cytotoxic activity in oxia is DNA damage not dependent. Electrochemical and ESR spectroscopy studies were performed in order to determine the ability of compounds to produce radicals and the relation of these in the mechanism of cytotoxicity.

A summary of the resource.
Several novel semicarbazone derivatives were prepared from 5-nitro-2-furaldehyde or 5-nitrothiophene-2-carboxaldehyde and semicarbazides bearing a spermidine-mimetic moiety. All derivatives presented the E-configuration, as determined by NMR-NOE experiments. These compounds were tested in vitro as potential antitrypanosomal agents, and some of them, together with the parent compounds, 5-nitro-2-furaldehyde and 5-nitrothiophene-2-carboxaldehyde semicarbazone derivatives, were also evaluated in vivo using infected mice. Structure–activity relationship studies were carried out using voltammetric response and lipophilic–hydrophilic balance as parameters. Two of the compounds (1 and 3) displayed the highest in vivo activity. A correlation was found between lipophilic–hydrophilic properties and trypanocidal activity, high RM values being associated with low in vivo effects

A summary of the resource.
New 1,2,4-thiadiazolylnitrones and furoxanylnitrones were developed and evaluated as neuroprotective agents on a human neuroblastoma (SH-SY5Y) cells model. They inhibited at low micromolar concentrations the oxidative damage and the death induced by exposure to hydrogen peroxide. These heteroarylnitrones showed excellent peroxyl free radical absorbance capacities, analyzed by oxygen radical absorbance capacity (ORAC) assay with fluorescein as the fluorescent probe, ranging from 1.5- to 16.5-fold the value of the reference nitrone, α-phenyl-N-tert-butylnitrone (PBN). The electron spin resonance spectroscopy (ESR) demonstrated the ability of these derivatives to directly trap and stabilize oxygen, carbon, and sulfur-centered free radicals. These results demonstrated the potential use of these heteroarylnitrones as neuroprotective agents in preventing the death of cells exposed to enhanced oxidative stress and damage.<br /><br />

The relationship between the herbicidal activity of a number of novel 1,2,5- oxadiazole N-oxides and some physicochemical properties potentially related with this bioactivity, such as polarity, molecular volume, proton acceptor ability, lipophilicity, and reduction potential were studied. The semiempirical molecular orbital method AM1 was used to calculate theoretical descriptors such as dipolar moment, molecular volume, Mulliken´s charge and the octanol/water partition coefficients (log Po/w). The values of the reduction potentials (Er) were obtained by cyclic voltammetry. In addition, the retention factors (log k’w) on a reversed-phase high-performance liquid chromatography (RP-HPLC) column in pure aqueous mobile phases were measured for several N-oxide derivatives. The log k’w values show good correlation with the calculated values of log Po/w, showing that the chromatographic parameter can be used as lipophilicity descriptor for these compounds. The multiple regression analysis between the descriptors for the Noxide derivatives and the herbicide activity indicate that the variance in the biological activity can be explained by changes in the lipophilicity and in the reduction potential.
