Abstract
A summary of the resource.
Sensory characterization provides a complete description of the sensory characteristics of food products. Descriptive Analysis is the most common methodology for this purpose. However, due to the cost and time associated with its application, several alternative methods have been recently developed. These methods do not require training, can be performed by trained assessors or consumers, and have been reported to be a good option when quick information about the sensory characteristics of a set of products is needed. There are basically four types of methodologies: methodologies based on the evaluation of specifi c attributes, on global differences among products, on the comparison with references, and methodologies that provide a verbal description of the products. In the present chapter these novel methodologies for sensory characterization of food products are described. Advantages and disadvantages of each methodology are discussed and recommendations for their application are provided.

Abstract
A summary of the resource.
Hybrid compounds containing hydrazones and benzofuroxan pharmacophores were designed as potential Trypanosoma cruzi-enzyme inhibitors. The majority of the designed compounds was successfully synthesized and biologically evaluated displaying remarkable in vitro activity against different strains of T. cruzi. Unspecific cytotoxicity was evaluated using mouse macrophages, displaying isothiosemicarbazone 10 and thiosemicarbazone 12 selectivity indexes (macrophage/parasite) of 21 and 27, respectively. In addition, the mode of anti-trypanosomal action of the derivatives was investigated. Some of these derivatives were moderate inhibitors of cysteinyl active site enzymes of T. cruzi, cruzipain and trypanothione reductase. ESR experiments using T. cruzi microsomal fraction suggest that the main mechanism of action of the trypanocidal effects is the production of oxidative stress into the parasite.

This study provides insights into the pathogenesis of Ralstonia solanacearum, in particular with regards to strains belonging to phylotype IIB, sequevar 1 (IIB-1) and their interaction with potato, its natural host. We performed a comparative genomic analysis among IIB-1 R. solanacearum strains with different levels of virulence in order to identify candidate virulence genes. With this approach, we identified a 33.7-kb deletion in a strain showing reduced virulence on potato. This region contains a cluster of six genes putatively involved in type IV pili (Tfp) biogenesis. Functional analysis suggests that these proteins contribute to several Tfp-related functions such as twitching motility and biofilm formation. In addition, this genetic cluster was found to contribute to early bacterial wilt pathogenesis and colonization fitness of potato roots.

The new dioxo(semicarbazone)vanadium(V) complexes cis-VO2L, where L = salicylaldehyde semicarbazone (L1), salicylaldehyde 4-n-butylsemicarbazone (L2), or salicylaldehyde 4-(2-naphthyl)semicarbazone (L3), have been synthesized, characterized by 1H and 13C NMR and FTIR spectroscopy and tested for bioactivity as potential insulin-mimetic agents. All dioxovanadium(V) complexes exhibited essentially no in vitro insulin-mimetic activity, but the VO2L2 complex developed activity in the presence of ascorbic acid, similar to that of vanadyl sulfate. The molecular structure of the novel complex VO2L1 has been solved by X-ray diffraction methods. It crystallizes in the tetragonal space group P42/n with a = 12.7674(7), c = 11.5308(5) Å, and Z = 8. The vanadium atom is in a distorted square-pyramidal coordination, with L1 acting as a tridentate ligand through its azomethyne nitrogen atom, carbonyl oxygen atom and deprotonated phenol oxygen atom. The coordination sphere is completed by two oxo ligands at cis positions.

The three novel mononuclear copper(II) complexes with dipicolinate and pyridyl-based ligands [Cu(dipic)(L)(OH2)] (L=4-picoline, vinylpyridine, 4-styrylpyridine; dipic2-=dipicolinate) were afforded and structurally characterized. X-ray diffraction studies accounted for slight distorted square-pyramidal structures in which the dianion dipic2- acts as a tridentate ligand in a mer-fashion, the N-donor species occupy an in-plane position, and a water molecule was detected apically coordinated. To assess the effect of the nature of the pyridyl-substituent (para position) on electronic properties, other complexes were also synthesized: [Cu(dipic)(py)(OH2)], [{Cu(dipic)(OH2)}2(μ-pyz)] and [{Cu(dipic)(OH2)}(μ-pypy){Cu(dipic)}] (py=pyridine, pyz=pyrazine, pypy=(E)-1,2-bis(pyridine-4-yl)ethane). Absorptive behavior in the UV-VIS region was studied in solution and in the solid state (reflectance measurements). Additionally, geometry and population analyses were conducted by means of DFT calculations. Electronic UV-VIS spectra were simulated for both dinuclear complexes in the framework of the TD-DFT methodology to assign the origin of the absorption bands.

The title complex, {[Ni(hab)]K[(hab)Ni]}·SCN (H 2 hab is bis(2-hydroxy-3-methoxybenzylidene)-1,2-diaminobenzene) crystallizes in the Monoclinic space group P2 1 /c with unit cell dimensions a = 11.5891 (13) Å, b = 17.307 (2) Å, c = 19.781 (2) Å,β = 92.368 (10)°, V = 3964.2 (8) Å 3 , Z = 4, R 1 = 0.106 and wR 2 = 0.305. The Ni II cation is in a N 2 O 2 inner and has a distorted square-planar coordination environment. The Ni II cation is linked by two imino nitrogen atoms and two bridged phenolate oxygen atoms. The potassium cation is octacoordinated in O 8 donor site and linked by four bridged phenolate oxygen atoms and four methoxy oxygen atoms. In the crystal, the {[Ni(hab)]K[(hab)Ni]} + cations form layers parallel to the ab plane. The SCN-anions are located in the interlayer space and connect the layers via C-H…N and C-H…S hydrogen bonds, forming three dimensional network.

The synthesis and structural characterization of the three novel nickel(II) complexes [Ni(OOCPh)2 (mpkoH)2] (1), [Ni(NO3)2(mpkoH)2] (2) and [Ni(mpkoH)3](NO3)2�½H2O (3�½H2O), with mpkoH = methyl(2-pyridyl)ketone oxime is reported. Geometry optimization and population analyses were performed by means of DFT calculations for the previously mentioned compounds as well as for [NiCl2 (mpkoH)2] (4). Electronic UV–vis spectra were also simulated in the TD-DFT framework to assign the origin of the absorption bands and in doing so, to have a clear picture of the absorptive features of the coordination compounds under investigation.

Abstract
A summary of the resource.
<p>NiLaM (M = Ce and/or Zr) catalysts were prepared by the polymerized complex method and evaluated in the dry, oxidative dry, and tri-reforming of a synthetic biogas (CH4/CO2 = 1.5) at 800° C and atmospheric pressure. The catalysts that possessed the highest number of total basic sites and stronger Ni-support<br />interaction showed better activity and coke resistance. A lower level of carbon deposits, with higher reactivity, was found for these catalysts in tri-reforming experiments. However, catalyst deactivation was observed for NiLaCe, attributed to nickel re-oxidation.</p>

La mitocondria es un el sitio principal de generacion de superoxido y otras especies reactivas y es uno de los principales sitios de formacion de 3-nitrotirosina proteica tanto en condiciones basales como patologicas. En este trabajo se estudiaron dos proteinas mitocondriales, el citocromo c y la superoxido dismutasa de manganese (MnSOD) y de cobre y cinc (CuZnSOD), estudiando tanto la nitracion de estas proteinas como cambios estructurales en condiciones similares alas encontradas in vivo. En el caso del citocromo c se estudio la nitracion de tirosinas proteicas evaluando la presencia de un lipide mitocondrial, la cardiolipina. En los experimentos realizados se observo por diversas tecnicas que en presencia de este Iipido la nitracion de citocromo c expuesto a peroxinitrito se encuentra aumentada, siendo este aumento mediado por un cambio conformacional mediado por la cardiolipina. A nivel estructural, se observaron los cambios sufrido por citocromo 0 en presencia de cardiolipina por tecnicas de NMR para y diamagnetico. Estos experimentos, si bien no fueron concluyentes mostraron que la proteina sufre un cambio conformacional global. Estudios de la reactividad de cada tirosina de citocromo c utilizando mutantes mostro que, en presencia de cardiolipina, tirosinas que no son preferencialmente nitradas 0 modificadas en ausencia de este Iipido estan involucradas en procesos oxidativos tales como la actividad peroxidasa de citocromo c mediada por cardiolipina. Esta actividad peroxidasa se estudio tam bien utilizando un derivado carboximetilado en me\ionina de ci\ocmmo c que, a di~efencia de\ ci\ocmmo c sin modi~icaf pefmi\e \a deteccion de un Compuesto I de peroxidasa. EI agregado de cardiolipina a citocromo c carboximetilado expuesto a peroxido de hidrogeno aumenta la formacion de compuesto I de citocromo c, y por 10 tanto su actividad peroxidasa. Para profundizar el estudio de los cambios estructurales sufridos por citocromo c frente a diferentes oxidantes se sintetizo y purifico citocromo c mononitrado a nivel de Tyr 97 y 74, para generar anticuerpos monoclonales contra formas conformacionalmente modificadas de esta proteina que permitan la deteccion de las mismas tanto en estudios in vitro como in vivo En el caso de las superoxido dismutasas (SOD) se estudiaron las modificaciones sufridas en presencia de flujos de oxido nitrico y superoxido evaluando la capacidad de esta enzima de competir con el oxido nitrico por su sustrato, observandose que en presencia de estos flujos se forma peroxinitrito en concentraciones capaces de inactivar a la enzima y en el caso de MnSOD capaz de nitrarla. Por otra parte se reeestudio la reaccion de MnSOD con peroxinitrito determinando un nuevo valor para la constante de reaccion de 1,9 x 104 M·1s·1 por tetramero. A nivel estructural se estudio el efecto de la nitracion en la accesibilidad del sustrato al sitio activo utilizando metodos in silico donde se determino que la nitracion de Tyr 34, responsable de la inactivacion de MnSOD por peroxinitrito genera un aumento en la barrera de energia libre para el acceso del superoxido al sitio activo, 10 cual impide el acceso al sustrato, tanto por repulsion esterica como electrostatica. Tambien a nivel estructural, se determino el efecto de mutaciones puntuales de MnSOD en la estructura cuaternaria de la enzima, evaluando, a su vez, el efecto de estas mutaciones en la sensibilidad de sus tioles. ----- No se encontraron virus en este mensaje. Activated carbon monoliths and oxidized carbon monoliths are prepared from Eucalyptus grandis wood by pyrolysis and subsequent activation under CO2 (aCMs) and further functionalization using nitric acid (ox-aCMs). Their textural and chemical properties are compared and the electrochemical characteristics as supercapacitor electrodes are tested. The nitric acid treatment does not produce substantial changes in the textural properties of the monolith obtained. Elemental analysis and X-ray photoelectron spectroscopy (XPS) reveal that the oxidized monolith presents a high content of oxygenated functional groups that can participate in pseudocapacitive reactions. This determines that the oxidized monoliths have higher electrical capacitance (80 F cm−3) at low current densities and higher energy density (up to Wv = 2.7 W h L−1) than the non-oxidized ones. The supercapacitor cell containing the oxidized monoliths presents higher total resistance values and response time compared to the non-oxidized monolith. The non-oxidized monolith showed the highest power density (up to PV = 94.5 W L−1).

The reaction of reactive oxygen and nitrogen species with the [4Fe–4S]2+cluster of mitochondrial (m-) and cytosolic (c-) aconitases leads to loss of catalytic activity and, in the case of the c-aconitase, triggers total cluster disruption to yield the iron-regulatory protein-1 (IRP-1). Herein we have studied the relative contribution and interplay of reactive oxygen species (O [equation] and H2O2), nitric oxide (•NO), and peroxynitrite in the modulation of m- and c-aconitase and IRP-1 activities in V79-M8 mammalian fibroblasts, identifying key variables that control the various reactivities at the cellular level. Extracellular production of H2O2led to inactivation of both m- and c-aconitase and IRP-1 activation, while extracellular [equation] had no effect. However, increased intracellular production of [equation] caused a loss in m- and c-aconitase activity and IRP-1 activation. Nitric oxide released from NOC-12 had a more complex effect on aconitase and IRP-1 activities. Mitochondrial aconitase was more sensitive than c-aconitase to•NO-mediated inactivation and minimal activation of IRP-1 was observed during a 30-min exposure to the•NO donor. The action of•NO was down- or upregulated by the presence of extra- or intracelular [equation], respectively. Extracellular [equation] decreased the•NO-mediated inactivation of aconitases, due to the preferential extracellular decomposition and the lower diffusivity of peroxynitrite compared to•NO. On the other hand,•NO exposure concomitant with enhanced intracellular [equation] fluxes lead to intracellular peroxynitrite formation as evidenced by Western blot analysis of nitrated proteins, which increased the effects observed with•NO alone. Peroxynitrite-mediated aconitase inactivation, IRP-1 activation, and cellular protein nitration were more pronounced in cells with low GSH content such as V79-M8 glutathione-depleted cells as well as in pGSOD4 cells which contain 32% of the GSH of the parental strain. Mechanistically, our results imply that the differential actions of the studied reactive species toward cellular aconitases depend on at least three critical factors: (i) their reaction rates with aconitases, (ii) the cellular compartment where they are formed, and (iii) the intracellular status of glutathione.

Lipid nitration has been observed during oxidative/nitrative stress conditions generating a variety of biomolecules capable of modulating cellular responses. This concept has grown as a result of studies with nitro-derivatives of long-chain unsaturated fatty acids containing a nitroalkene group (nitro-fatty acids). This review focuses on the interactions of nitro-fatty acids with members of the peroxisome proliferator-activated receptors (PPARs) family. These nuclear receptors belong to a superfamily of ligand-activated transcription factors, which serve as sensors of lipophilic molecules and regulate the expression of a set of genes involved in glucose and lipid metabolism. Here we discuss how nitro-fatty acids bind and activate PPARs, including the current knowledge of the molecular interactions and cell signaling events involved as well as their therapeutic potential associated with chronic inflammation and metabolic disorders.<br /><br />

A summary of the resource.
Nitroalkenes derivatives of free as well as esterified unsaturated fatty acids are present in human plasma and tissue, representing novel pluripotent cell signaling mediators. Lipid nitration occurs in response to pro-inflammatory stimuli as an adaptative mechanism to downregulate inflammatory responses. This chapter first discusses the generation of nitroalkenes during macrophage activation following chemical and biological characterization. In particular, it describes procedures for (a) synthesizing and characterizing esterified (cholesteryl-nitrolinoleate, CLNO2) as well as free (nitroarachidonate, AANO2) nitroalkenes, (b) determining nitration of cholesteryl linoleic acid during macrophage activation by inflammatory stimuli, (c) examining the modulatory effects of nitroalkenes on the expression of inducible enzymes by activated macrophages, and (d) discussing the signaling pathways involved in nitroalkene-mediated anti-inflammatory actions.

The competition between selected or commercial killer strains of type K2 and sensitive commercial strains of Saccharomyces cerevisiae was studied under various conditions in sterile grape juice fermentations. The focus of this study was the effect of yeast inoculation levels and the role of assimilable nitrogen nutrition on killer activity. A study of the consumption of free amino nitrogen (FAN) by pure and mixed cultures of killer and sensitive cells showed no differences between the profiles of nitrogen assimilation in all cases, and FAN was practically depleted in the first 2 days of fermentation. The effect of the addition of assimilable nitrogen and the size of inoculum was examined in mixed killer and sensitive strain competitions. Stuck and sluggish wine fermentations were observed to depend on nitrogen availability when the ratio of killer to sensitive cells was low (1:10 to 1:100). A relationship between the initial assimilable nitrogen content of must and the proportion of killer cells during fermentation was shown. An indirect relationship was found between inoculum size and the percentage of killer cells: a smaller inoculum resulted in a higher proportion of killer cells in grape juice fermentations. In all cases, wines obtained with pure-culture fermentations were preferred to mixed-culture fermentations by sensory analysis. The reasons why killer cells do not finish fermentation under competitive conditions with sensitive cells are discussed.

<p>La enfermedad de Alzheimer (EA) es la forma más común de demencia en el mundo, con alrededor de 47.5 millones de personas afectadas. Actualmente no existe cura y los únicos fármacos aprobados para su tratamiento tienen como efecto una mejora transitoria de los síntomas de la enfermedad, pero no son capaces de revertir o detener el daño neuronal asociado. Esto se debe a que los procesos bioquímicos que desencadenan la enfermedad aún se desconocen. Un rasgo distintivo y común en los procesos neurodegenerativos es el aumento intracelular de especies reactivas del oxígeno y del nitrógeno, lo que desencadena una serie de eventos que conducen a la apoptosis.</p>
<p>La enzima caspasa-3 ha sido identificada como un blanco terapéutico interesante, ya que la misma no sólo actúa como un mediador clave en la apoptosis neuronal, sino que también participa en procesos no apoptóticos mediante la formación de fragmentos tóxicos de proteínas específicas que afectan la sinapsis y la plasticidad neuronal en etapas tempranas de la EA. Se ha observado que la caspasa-3 no sólo se activa selectivamente en células de hipocampo de ratones transgénicos modelo de EA alterando la función sináptica que da inicio a la pérdida de memoria, sino que también en cerebros de pacientes con EA existen niveles elevados de esta enzima.</p>
<p>En la búsqueda de nuevos agentes terapéuticos capaces de modular la sobreactivación de caspasa-3, nuestro grupo de investigación se ha enfocado en el desarrollo de inhibidores reversibles y selectivos de caspasa-3 evidenciando que aquellos compuestos derivados de tiazol y nitrona (imina N-óxido) son buenos inhibidores de caspasa-3 y presentan elevada capacidad antioxidante. En base a estos antecedentes, en el presente trabajo se realizó la síntesis de nuevas moléculas híbridas conteniendo agrupamientos nitrona y tiazol diseñadas para potenciar la selectividad y actividad inhibitoria frente a caspasa-3. Se determinó el efecto neuroprotector antioxidante, antiapoptótico, la capacidad inhibitoria frente a caspasa-3 y caspasa-7 in vitro y se realizaron estudios teóricos de docking y dinámica molecular para estudiar la interacción a nivel atómico de los compuestos en la caspasa-3 y caspasa-7. Los resultados obtenidos muestran que los compuestos evaluados son excelentes agentes antioxidantes y antiapoptóticos, y moderados inhibidores de caspasa-3. El análisis teórico indica que los derivados se unen en la zona de la interfase cercano al sitio alostérico interaccionando con residuos clave para mantener la conformación activa de la enzima caspasa-3 y con menor afinidad en caspasa-7. Las nitronas a4 y a6 presentan muy buena actividad neuroprotectora e inhiben al menos un 20% la actividad caspasa-3. Esto las posiciona como nuevos hits para el rediseño y desarrollo de futuros potenciales fármacos contra EA y otras enfermedades neurodegenerativas.</p>
