El estudio de los contaminantes en los alimentos y el medioambiente es uno de los campos que mayor desarrollo ha experimentado en los últimos tiempos tanto en aspectos regulatorios como analíticos. La diversificación de las actividades económicas y humanas tendientes hacia una sociedad de consumo, ha generado una variación en las características y clases de contaminantes orgánicos que atentan contra la calidad y sustentabilidad de los recursos. El presente proyecto de tesis tuvo por objetivo el desarrollo y evaluación de métodos y estrategias analíticas cualitativas y cuantitativas para la determinación de compuestos orgánicos de relevancia sanitaria y/o ambiental presentes a bajas concentraciones en diferentes matrices como materias primas farmacéuticas, alimentos y muestras ambientales. Se han realizado aportes al análisis de compuestos orgánicos trazas, en matrices de diverso origen y complejidad. La presente tesis ha sido dividida a efectos prácticos en tres capítulos dependiendo el tipo de matriz de trabajo: materias primas farmacéuticas, alimentos y medio ambiente. En el primer capítulo, se destacan procedimientos analíticos originales para la determinación de residuos de pesticidas en materias primas farmacéuticas de origen natural, para los cuales los métodos oficiales o son obsoletos conceptualmente o simplemente no existen. Se desarrollaron dos metodologías basadas en la técnica MSPD para el análisis multirresiduo de pesticidas en lanolina y propóleos, matrices de compleja composición química donde la etapa de purificación es crítica para una buena determinación. El segundo capítulo aborda el análisis de agroquímicos en alimentos. En primera instancia, se comprobó la posibilidad de determinación de reguladores de crecimiento en vegetales empleando tratamientos de muestra multirresiduo habituales (métodos Luke, QuEChERS y acetato de etilo) para el análisis de con determinación por sistemas LC-MS/MS. Por otro lado se realizó una evaluación preliminar a la contribución del consumo de yerba mate a la ingesta diaria de pesticidas por la población uruguaya, a partir de un dispositivo específico para evaluar la extracción de pesticidas en condiciones de ingesta típica del mate. RESUMEN 13 Se comprobó que la extractabilidad de residuos y por lo tanto la ingesta de de pesticidas se puede relacionar con las propiedades fisicoquímicas de los pesticidas en estudio. En el tercer capítulo, se describe la principal vía de transformación ambiental del antibiótico amoxicilina monitoreada en aguas residuales y aguas de río. Posteriormente se expone la posibilidad de evaluación de contaminantes emergente en aguas empleando diversos arreglos instrumentales. Son expuestas las fortalezas y debilidades de diversas estrategias analíticas especialmente enfocadas hacia instrumentos QTOF en experimentos dedicados a confirmación de identidad y estrategias de screening automatizado combinado con motores de búsqueda basados en bases de datos de masas exactas. Se ha observado que las estrategias basadas en detección asistida por software generalmente necesitan una etapa adicional de confirmación. Un instrumento QTOF en modo MS/MS permitió confirmar o rechazar la identidad de la mayoría de los hallazgos de screening de contaminantes emergentes en muestras reales a pesar de que existen limitaciones de sensibilidad analítica y efecto matriz. Se compararon además estrategias cuantitativas y cualitativas basada en el análisis por inyección directa de muestras acuosas por LC-MS/MS empleando instrumentos QLIT para un grupo representativo de contaminantes emergentes operando en modo SRM y en modo IDA. La mayor sensibilidad de los instrumentos QLIT-MS/MS ofrece la posibilidad de realizar análisis cuantitativo de variedad de contaminantes por inyección directa de la muestra, reduciendo el efecto de la matriz y aumentando de la confiabilidad de los resultados. Durante esta tesis, se ha logrado cubrir los distintos aspectos del análisis de trazas contemplando desde estrategias de preparación de muestra, expansión del alcance de métodos preexistentes y caracterización de contaminantes antropogénicos, empleando distintas técnicas instrumentales y de tratamientos de datos que permiten la realización de experimentos útiles tanto en cuantificación, identificación preliminar, confirmación y elucidación estructural de contaminantes orgánicos. Free-range livestock can be exposed to toxic cyanobacteria by drinking water from eutrophic natural sources. This study reports on the analysis of livestock drinking water sources from ranches in an eutrophic subtropical basin of South America (33ºS, 57°W), where frequent Microcystis spp. blooms and occasionally dense scums were found. To minimise monitoring costs, microcystins (MCs) were determined in serum and water with an in-house developed enzyme linked immunosorbent assay (ELISA). The water results were compared with those of liquid chromatography – tandem mass spectrometry (LC-MS/MS) for the most common congeners (MC-LR, MC-RR; MC-YR, MC-LA). Both methods were strongly correlated along a broad range of concentrations. The concentrations of MCs in the livestock drinking water were up to 3,700 μg/L. This is about 1,000 times higher than the Australian and New Zealand livestock MCs guidance values (cattle = 4.2; sheep = 3.9 μg/L) as well as the California-EPA sub-chronic action level (3 μg/L) and implies a high risk of poisoning. The animals exposed to MCs concentrations 70–100 times higher than these guidelines showed no significant alterations in liver enzymes. Nevertheless, serum concentrations of up to 0.11 μg/L MCs were recorded by ELISA. These results reveal the potential of MCs-ELISA in serum as a sensitive, minimally invasive method for risk assessment in integrated environmental monitoring strategies. Treatment for Chagas disease, a parasitosis caused by Trypanosoma cruzi, has always been based on two drugs, nifurtimox and benznidazole, despite the toxic side effects described after prolonged prescription. In this work, we study a new prospective antitrypanosomal drug based on vanadium, here named VIVO(5Brsal)(aminophen). We found a good IC50 value, (3.76 ± 0.08) μM, on CL Brener epimastigotes. The analysis of cell death mechanism allowed us to rule out the implication of a mechanism based on early apoptosis or necrosis. Recovery assays revealed a trypanostatic effect, accompanied by cell shape and motility alterations. An uptake mostly associated with the insoluble fraction of the parasites was deduced through vanadium determinations. Concordantly, no drastic changes of the parasite transcriptome were detected after 6 h of treatment. Instead, proteomic analysis uncovered the modulation of proteins involved in different processes such as energy and redox metabolism, transport systems, detoxifying pathways, ribosomal protein synthesis, and proteasome protein degradation. Overall, the results here presented lead us to propose that VIVO(5Brsal)(aminophen) exerts a trypanostatic effect on T. cruzi affecting parasite insoluble proteins. Five heteroleptic compounds, [VVO(IN-2H)(L-H)], where L are 8-hydroxyquinoline derivatives and IN is a Schiff base ligand, were synthesized and characterized in both the solid and solution state. The compounds were evaluated on epimastigotes and trypomastigotes of Trypanosoma cruzi as well as on VERO cells, as a mammalian cell model. Compounds showed activity against trypomastigotes with IC50 values of 0.29–3.02 �M. IN ligand and the new [VVO2(IN-H)] complex showed negligible activity. The most active compound [VVO(IN-2H)(L2-H)], with L2 = 5-chloro-7-iodo- 8-hydroxyquinoline, showed good selectivity towards the parasite and was selected to carry out further biological studies. Stability studies suggested a partial decomposition in solution. [VVO(IN-2H)(L2-H)] affects the infection potential of cell-derived trypomastigotes. Low total vanadium uptake by parasites and preferential accumulation in the soluble proteins fraction were determined. A trypanocide effect was observed when incubating epimastigotes with 10 � IC50 values of [VVO(IN-2H)(L2-H)] and the generation of ROS after treatments was suggested. Fluorescence competition measurements with DNA:ethidium bromide adduct showed a moderate DNA interaction of the complexes. In vivo toxicity study on C. elegans model showed no toxicity up to a 100 �M concentration of [VVO(IN-2H)(L2-H)]. This compound could be considered a prospective anti-T. cruzi agent that deserves further research. The infrared and electronic spectra as well as the thermal analysis of all the compounds were recorded and discussed. The thiosaccharinate anion acts in three different coordination forms, one of then reported for the first time. The crystal structures of complexes 2 and 3 have been determined by single crystal X-ray diffractometry. In complex 2, two monomeric moieties are joined together forming a symmetric bis-μ-sulphur bridged dimer by interaction of two lead(II) atoms through the exocyclic sulphur atoms of two thiosaccharinate ligands. The seven-fold coordination sphere of each lead atom is completed by two pyridine nitrogen atoms and by another sulfur and two nitrogen atoms of the thiosaccharinate anions. In complex 3, the lead(II) atom is coordinated by four nitrogen atoms of two 1,10-phenantroline molecules and by the sulfur and nitrogen atoms of one thiosaccharinate ion. The second anion has an electrostatic interaction with the nucleus. Apiculate yeasts belonging to the genus Hanseniaspora are predominant on grapes and other fruits. While some species, such as Hanseniaspora uvarum, are well known for their abundant presence in fruits, they are generally characterized by their detrimental effect on fermentation quality because the excessive production of acetic acid. However, the species Hanseniaspora vineae is adapted to fermentation and currently is considered as an enhancer of positive flavour and sensory complexity in foods. Since 2002, we have been isolating strains from this species and conducting winemaking processes with them. In parallel, we also characterized this species from genes to metabolites. In 2013, we sequenced the genomes of two H. vineae strains, being these the first apiculate yeast genomes determined. In the last 10 years, it has become possible to understand its biology, discovering very peculiar features compared to the conventional Saccharomyces yeasts, such as a natural and unique G2 cell cycle arrest or the elucidation of the mandelate pathway for benzenoids synthesis. All these characteristics contribute to phenotypes with proved interest from the biotechnological point of view for winemaking and the production of other foods. This paper reports the growth of bismuth tri-iodide thick films intended for direct and digital X-ray imaging. Films were grown by the vertical physical vapor deposition method, onto glass substrates 2″x 2″ in size, with gold previously deposited as rear electrode. The film thickness was up to 33 μm (±5 %). Optical microscopy and SEM were performed on the films and grain size resulted to be up to 40 μm. A strong correlation was found between the microcrystals growth orientation and the growth temperature. At low temperatures, microcrystals grow with their c axis parallel to the substrate, whereas at higher temperatures, they grow with their c axis perpendicular to the substrate. The higher the growth temperature, the lower the dark current of the film, and the higher the resistivity, which was from 1013 to 1015 Ωcm. A sensitivity to X-rays of 6.9 nC/R.cm2 was measured irradiating the films with X-rays from a mamographer. Film properties were correlated with the growth temperature, with previous results for bismuth tri-iodide films and monocrystals and with data for films of alternative materials such as lead and mercuric iodide. Owing to their reproducibility, stability, and cost-effective production, the recombinant variable domains of heavy-chain-only antibodies (VHHs) are becoming a salient option as immunoassay reagents. Recently, there have been several reports describing their application to the detection of small molecules (haptens). However, lacking the heavy-light chain interface of conventional antibodies, VHHs are not particularly apt to bind small analytes and failures are not uncommon. Here we describe the construction of a VHH phage display library against the cyanobacterial hepatotoxin microcystin LR and its selection using competitive panning and two novel panning strategies. The outcome of each strategy was evaluated by a large-scale screening using in vivo biotinylated nanobodies. The three methods selected for different nonoverlapping subsets of VHHs, allowing one to optimize the immunodetection of the toxin. The best results were obtained by promoting the isolation of VHHs with the slowest koff (off-rate selection). Among these, the biotinylated nanobody A2.3 performed in ELISA with excellent recovery and high sensitivity, IC50 = 0.28 μg/L, with a limit of detection that is well below the most rigorous guidelines for the toxin. While it may be case-specific, these results highlight the importance of exploring different panning strategies to optimize the selection of antihapten nanobodies. La eutrofización de los ambientes acuáticos promueve la proliferación de cianobacterias, que producen una amplia variedad de compuestos altamente tóxicos para animales, las cianotoxinas. En Uruguay, los principales sistemas acuáticos (Río Negro, Río Uruguay y Río de la Plata) están seriamente afectados por floraciones de cianobacterias de géneros productores de microcistinas. Estas cianotoxinas son heptapéptidos cíclicos de elevada toxicidad, que interfieren en los principales usos del agua: producción de agua potable, recreación, producción agropecuaria, pesca, entre otros. Para evaluar y gestionar los riesgos asociados, se requieren métodos sencillos, rápidos y validados, que viabilicen su determinación a bajo costo. En esta tesis se desarrolló y validó un kit de ELISA sencillo y robusto, basado en anticuerpos policlonales, que ha permitido monitorear microcistinas en aguas de embalses y playas. Los resultados evidenciaron en espumas cianobacterianas, concentraciones extremadamente elevadas de microcistinas (>1000 veces el límite para agua recreacional), que impulsaron el uso de la bandera sanitaria y sustentaron la detección visual de colonias cianobacterianas dispersas y espumas, como un criterio simple de alerta para la toma de decisiones rápidas en la gestión de playas. En una segunda etapa se avanzó en la estandarización de la inmunodetección, generando anticuerpos monoclonales de llama (Nanobodies), implementando estrategias novedosas de selección para aislar los clones de mayor desempeño analítico. Esto posibilitó el desarrollo de un ELISA de segunda generación sensible y con elevada selectividad. Ambos ensayos se han validado rigurosamente y comparado con métodos analíticos de referencia como LC-MS/MS. El nuevo anticuerpo viabilizó el desarrollo de un ensayo inmunocromatográfico en tiras, que se realiza en minutos en el sitio de interés y es adecuado para analizar aguas ambientales y potables. Para generar mayor información sobre la presencia de congéneres individuales de microcistinas, se desarrollaron métodos de MALDI-TOF cualitativos, y posteriormente cuantitativos incorporando estándares internos de síntesis sencilla. In recent years, the international demand for commodities has prompted enormous growth in agriculture in most South American countries. Due to intensive use of fertilizers, cyanobacterial blooms have become a recurrent phenomenon throughout the continent, but their potential health risk remains largely unknown due to the lack of analytical capacity. In this paper we report the main results and conclusions of more than five years of systematic monitoring of cyanobacterial blooms in 20 beaches of Montevideo, Uruguay, on the Rio de la Plata, the fifth largest basin in the world. A locally developed microcystin ELISA was used to establish a sustainable monitoring program that revealed seasonal peaks of extremely high toxicity, more than one-thousand-fold greater than the WHO limit for recreational water. Comparison with cyanobacterial cell counts and chlorophyll-a determination, two commonly used parameters for indirect estimation of toxicity, showed that such indicators can be highly misleading. On the other hand, the accumulated experience led to the definition of a simple criterion for visual classification of blooms, that can be used by trained lifeguards and technicians to take rapid on-site decisions on beach management. La investigación de la correlación entre la estructura y las propiedades magnéticas de compuestos polinucleares de coordinación se ha incrementado, persiguiendo dos objetivos principales: a) sintetizar y caracterizar nuevos materiales con interesantes propiedades magnéticas al estado sólido y b) diseñar y estudiar nuevos compuestos de coordinación que puedan ser útiles como modelos para el estudio teórico de diferentes tipos de interacciones magnéticas. Esta tesis propone la síntesis y caracterización de nuevos compuestos de coordinación polinucleares de Re (II) con iones paramagnéticos 3d que puedan presentar propiedades magnéticas interesantes. Para llevar a cabo estos estudios, se prepararon nitrosil complejos mononucleares de renio(II) que pudieran potencialmente actuar como “ligandos complejo”. Catorce complejos mononucleares Re(II) de fórmula (NBu4)[Re(NO)Br4(L)] [NBu4+ = catión n-tetrabutilamonio y L = piridina (1), 4-piridincarbonitrilo (2), ácido nicotínico (3), ácido 3,5-piridindicarboxílico (4)], pirazina (5), pirazincarbonitrilo (6), 2-metilpirazina (7), ácido pirazincarboxílico (8), ácido 2,6 pirazinodicarboxílico (9), pirimidina (10), piridazina (11), imidazol (12), pirazol (13) y triazol (14)] se han preparado mediante una reacción de sustitución directa de la molécula de etanol en el complejo (NBu4)[Re(NO)Br4(EtOH)] por los ligandos orgánicos L. Las estructuras cristalinas determinadas por difracción de rayos X muestran que L siempre está unido al renio (II) por un átomo de nitrógeno heterocíclico. Posteriormente, los compuestos 3, 4 y 5 se ensayaron como "ligandos complejos" para obtener nuevos complejos heteropolincleares de renio(II) con iones metálicos de la primera fila de transición [Cu(II), Ni(II), Mn(II), Fe(II) o Co(II)]. Las estructuras cristalinas estudiadas por difracción de rayos X de monocristal muestran que los compuestos 3a-3e y 4a-4c están formados por unidades heterodinucleares discretas de Re(II)M(II) en donde las entidades [Re(NO)Br4(nic)]2- o [Re(NO)Br4(Hpydc)]2- actúan como ligandos didentados o monodentados a través de un grupo carboxilato hacia el metal catiónico 3d. Por otro lado, la pirazina en [Re(NO)Br4(pyz)]- no pudo actuar como un ligando puente hacia un segundo catión metálico, y por consiguiente se obtuvieron las sales 5a-5f. Las propiedades magnéticas de la mayoría de los complejos mononucleares y heterobinucleares se investigaron en el rango de temperatura de 1,9 a 300 K. El primero se comporta como dobletes de espín cuasi magnéticamente aislados con interacciones antiferromagnéticas muy débiles a través de contactos intermoleculares Br⸱⸱⸱Br o interacciones π-π. Sus propiedades magnéticas se discuten a través de un análisis profundo de la influencia del campo de ligando, acoplamiento espín-órbita, distorsión tetragonal y efectos de covalencia. Los valores del paramagnetismo independiente de la temperatura para estos complejos mononucleares también se corroboran y se comparan con los informados previamente en sistemas relacionados. Los complejos heterobinucleares también exhiben interacciones antiferromagnéticas débiles. La vía de intercambio intramolecular en esta familia se descarta debido a la simetría del orbital magnético del ión Re (II) (dxy) impide cualquier deslocalización de espín en los orbitales puentes nic o Hpydc. Por tanto, las interacciones magnéticas observadas probablemente estén mediadas por interacciones de tipo π-π entre los ligandos quelantes N bidentados periféricos o los contactos Br⸱⸱⸱Br que se producen en ellos. Phenazine 5,10-dioxides (PDOs) are a new class of bioreductive cytotoxins, which could act towards tumours containing hypoxic regions. The PDOs selective-hypoxic bioreduction was probed in vitro; however, the mechanism of action has not been completely explained. Besides, PDOs in vivo antitumour activities have not been demonstrated hitherto. We study the mechanism of hypoxic/normoxic cytotoxicity of PDO representative members. Electron spin resonance is used to confirm •OH production, alkaline comet assay to determine genotoxicity, and gel electrophoresis and flow cytometry to analyze DNA fragmentation and cell cycle distribution. Chemically induced rat breast tumours are employed to evaluate in vivo activities. For the most selective cytotoxin, 7(8)-bromo-2-hydroxyphenazine 5,10-dioxide (PDO1), exclusive hypoxic •OH production is evidenced, while for the unselective ones, •OH is produced in both conditions (normoxia and simulated hypoxia). In normoxia (Caco-2 cells), PDO1 induces cell-cycle arrest and DNA fragmentation but does not significantly induce apoptosis neither at IC50 nor IC80. No difference in the comet-assay scores are observed in normoxia and simulated hypoxia being the unselective 2-amino-7(8)-bromophenazine 5,10-dioxide (PDO2) the most genotoxic. The in vivo efficacy with the absence of systemic toxicity of PDO1 and PDO2 is checked out. Results from this study highlight the potential of PDOs as new therapeutics for cancer. The crystal structures of 1 and 2 are reported herein together with the cryomagnetic investigation of 1–4 in the temperature range of 2.0–300 K. 1 is a mononuclear compound whose structure is made of [Re(NO) Br4(H2pydc)]- complex anions, tetra-n-butylammonium cations and isopropanol molecules of crystallization. Each rhenium(II) ion in 1 is six-coordinate with four bromide ligands in the equatorial positions and a nitrosyl group and one pyridyl-nitrogen atom from an H2pydc ligand filling the axial sites. 2 is a neutral heterobimetallic compound where the [Re(NO)Br4(Hpydc)]2- complex anion acts as a monodentate ligand towards the [Cu(4,4′ -dmbipy)2]2+ complex cation through one carboxylate–oxygen atom. The rhenium(II) ion is six-coordinate in a somewhat distorted octahedral surrounding similar to that in 1. The copper(II) ion in 2 is five-coordinate with four nitrogen atoms from two bidentate 4,4′ -dmbipy ligands and one carboxylate–oxygen atom, describing a surrounding intermediate between square pyramidal and trigonal bipyramidal. A novel halogenation reaction from sulfonates catalyzed by ironACHTUNGTRENUNG(III) is described. The reaction can be performed as a stoichiometric or a catalytic version. This reaction provides a convenient strategy for the efficient access to structurally diverse secondary chlorides, bromides and iodides. The stereochemical course of the reaction is governed by the substrate and the experimental conditions. Secondary alcohols modified as quisylates or pysylates are substantially more reactive. Aliphatic quisylates proceed with overall inversion of configuration under catalytic conditions. Chemoselectivity in bismesylates was observed in favour of the secondary mesylate.
