El objetivo general de este trabajo fue la identificacion de algunas especies de rnicroalgas dulceacuicolas 0 subaereas existentes en nuestro pais, el estudio de su composicion lipidica y su dependencia cuali-cuantitativa de las condiciones de cultivo experimentales Su cumplimiento implico desarrollar metodos, procesos y tecnicas que permitieron determinar las condiciones optimas de cultivos, segitn los fines perseguidos, para algunas de las eventuales aplicaciones de las microalgas encontradas en nuestro pais, asi como poner a punta las tecnicas bioquimicas de analisis requeridas. Los objetivos especijicos fueron: 1) Colectar, purificar, aislar e identificar especies de microalgas de agua dulce del Uruguay. 2) Realizar cultivos de mantenimiento unialgal de las especies identificadas. 3) Poner a punta de ensayos analfticos para la determinacion cuali-cuantitativa de los lipid os algales 4) Determinar la combinacion optima de panlmetros fisicos y quimicos que permitieron la obtencion de una mayor biomasa en el menor tiempo posible, con una riqueza relativamente mayor en acid os grasos poliinsaturados (PUFAs). Se intento poner en marcha un area y una metodologia de trabajo nuevos para el pais, con amplias posibilidades de desarrollo y aplicacion y con un costa relativamente bajo en infraestructura. En este trabajo se cultivaron microalgas de diferentes Familias, colectadas en el pais, bajo distintas condiciones de temperatura, luz y nutrientes Para esto se dedico una primera etapa a optimizar los muestreos, aislamiento de las cepas colectadas y su mantenimiento en el laboratorio. Se estudiaron tres Chlorophyceae (Chlorella vulgaris, Oocystis sp. y Scenedesmus quadricauda), y dos Cyanophyceae (Chroococcus minutus y Chroocopcidiopsis sp.). En algunos casos se logro identificar la especie y en otros se llego solamente al Genero. Para ilustrar mejor los resultados obtenidos, las microalgas se agrupan por Clase. La Chlorella vulgaris se obtuvo de una pileta de oxidacion de aguas industriales; se purifico y cultivo en medio ISO 8692, luz continua, 28°C y aireacion. Se construyo la curva de crecimiento y se determino la composicion en acid os grasos de las muestras colectadas en las J jases de latencia, exponencial y de muerte. A partir de ello se determino que la colecta mas • conveniente resulta a los 15 dias de inoculado el cultivo, cuando aun este se encuentra en lase exponencial. _ Se realizaron modificaciones en la condiciones de cultivo: se trabajo con jotoperiodo de 16: 8 y se cambio la relacion C!!I y la fuente de nitrogeno. Se obtuvo un mayor porcentaje de PUFAs parajotoperiodos 16: 8 y 17 mg/l de urea como fuente de nitrogeno en el medio ISO 8692. Tambien se trabajo a 200C y luz natural, pero no se obtuvo resultados positivos. La Oocystis sp y la Scenedesmus quadricauda se obtuvieron de una fuente ornamental de jardin, mezcladas con otras microalgas. La Oocystis sp. se aislo y cultivo en jorma similar a la Chlorella vulgaris. Con la Scenedesmus quadricauda no se obtuvieron buenos resultados en el aislamiento, por 10 que no pudo completar su estudio. En los cultivos de Oocystis sp. se obtuvo el mayor porcentaje en PUFAs, en el medio ISO 8692 con aporte de nitrogeno de 17 mg/l de urea, tanto a 280C y jotoperiodos 16: 8 como a 20°C y luz natural. De las dos Cyanophyceae estudiadas, el Chroococcus minutus se obtuvo del raspado de la corleza de un arbol y la Chroocopcidiosis sp. de una pared Ambas se cultivaron en medios CgIO y SOT diluido al cuarto, tanto a 280C y jotoperiodos 16: 8 como a 20°C y luz natural. En ambas el mayor porcentaje de PUFAs se logr6 con medio SOT/4, a 20°c y luz natural. Para poder evaluar mejor los resultados obtenidos en todos los cultivos, se defini6 un parinnetro que relaciona La cantidad de biomasa en el momenta de fa colecta (15 dins de cultivo), Sit porcentaje de lipidos y el porcentaje en PUFAs en ello. A este panimetro se Ie llam6 Productividad en PUFAs. Se observo que la mayor Productividad es para el Chroococcus minutus cultivado con medio SOT/4 a 20°C y luz natural. Como resumen general de este trabajo se puede indicar que se logr6 adaptar al cultivo in vitro a cuatro especies de microaLgas de agua dulce colectadas en el Uruguay. Su estudio estuvo orientado al contenido en lipidos y su composici6n en acid os grasos,. en vista de su posible uso como fuente natural de los mismos. La aplicaci6n de variaciones en las condiciones de cultivo permitieron modificar La composici6n en acid os grasos de las cepas estudiadas. Como corolario se puede decir que, a partir de esta tesis, se cuenta con la capacitaci6n y el dominio de los metados de cultivo y de anillisis de microalgas que permitiran proseguir estudios futuros, no s610 continuando el relevamiento propuesto sino utilizando cepas de colecci6n y realizando trabajos con otros grupos latinoamericanos, tanto con algas de agua dulce como marinas. The color of 30 honey samples, both from unifloral and multifloral origin, was characterized using Pfund scale and CIE L*a*b* tristimulus method. A panel of 140 consumers evaluated the color acceptability of the samples using a 9-point structured scale. The color of the samples was differentiated (P < 0.001) by both instrumental methods, which confirms the validity of using them to characterize the color of honey. Consumers showed highly significant different degrees of liking for the color of the evaluated honeys. Two clusters (cluster 2 and cluster 3) with different preference patterns were identified; one liked intermediate-colored honeys, and the other preferred dark reddish honeys. External preference mapping was used to model color acceptability for consumers in each cluster using the first two principal components of the principal component analysis of the instrumental color data. Conformational diseases are a group of diverse disorders that have been associated with misfolding of specific proteins, leading to their aggregation in particular cell tissues. Despite their relevance, the mechanisms involved in neurodegenerative processes remains poorly understood. Mutations in Cu,Zn superoxide dismutase (SOD1) are implicated in death of motor neurons in amyotrophic lateral sclerosis. Among others, the SOD1G93A mutation is known to weaken the structure and this could lead to conformational variations of the protein. As an approach to understand the tissue-specific propensity of protein aggregation, we developed an experimental procedure allowing rapid extraction of variants of human SOD1 (hSOD1) produced in different tissues. Using an antibody-based affinity chromatography procedure enzymatically active hSOD was extracted, indicating preservation of its native conformation. Analysis of the eluted fractions of hSOD extracted from the brain and liver of transgenic hSODG93A rats provided evidence about heterodimers rSOD–hSODG93A formation in both extracts. Moreover, when characterized by 2-DE and MALDI-TOF/TOF MS, the extracted hSODG93A showed a complex profile suggesting the existence of various covalent modifications of the enzyme in both tissues. Thus, this method should allow following post-translational modifications of hSOD1 produced in various tissues. EI presente trabajo establece un metoda cuantitativo rapido de analisis de ceras de distintas v ariedades de semillas de trigo 10 que permitirfa realizar una identificaci6n y clasificaci6n de las variedades en base a Ios porcentajes de los compuestos qufmicos mayoritarios presentes en dichas ceras. El metoda de anaIisis por TLC-Scanner, establece ademas, una alternativa a los metodos clasicos de GC 0 HPLC, que presentan serias dificultades, debido a la diversidad de compuestos presentes en las ceras (Hidrocarburos, aldehfdos y cetonas, esteres, terpenos, alcoholes, acid os, etc.) para un analisis directo. El objetivo del presente trabajo consisti6 en la identificaci6n de reductasas con actiyidad anti Prelog y el desarrollo de cepas recombinantes que expresaran las mismas de modo de completar el espectro de biocatalizadores disponibles para la sintesis de a-alquil-~-hidroxiesteres quirales. Para lograr identificar nuevas enzimas que permitieran construir estos biocatalizadores, se desarro1l6 un metoda rflpido de screening para la identificaci6n de genes correspondientes a enzimas con actividad reductasa. Se utiliz6 como modelo la cepa Paucimonas lemoignei, la cual expresa una ~-cetoester reductasa con actividad anti Prelog, aunque no se conoce el gen que la codifica. Utilizando este metodo fueron identificados genes que codifican para posibles ~-cetoester reductasas, uno de los cuales fue clonado y expresado en diyersas cepas de E. coli. Las cepas recombinantes generadas fueron utilizadas en diversos ensayos de biotransformaci6n con el objetivo de caracterizar su especificidad de sustrato y su diastero y enantioselectividad. Uno de los biocatalizadores desarrollados present6 actividad ~-cetoester reductasa sobre todos los a-alquil-B-cetoesteres utilizados como sustratos, exhibiendo actiyidad anti Prelog en la reducci6n de a-alil y a-propargil acetoacetato de etilo. Esta cepa constituye el primer biocatalizador de celula entera para la sintesis de ami (2R, 3R) a-alquil-~-hidroxiesteres, ampliando el repertorio de biocatalizadores disponibles para la sintesis de ahidroxiesteres quirales. Aims: Identification, cloning, expression and characterization of a novel lipase – Lip I.3 – from strain Pseudomonas CR-611. Methods and Results: The corresponding gene was identified and isolated by PCR-amplification, cloned and expressed in Escherichia coli, and purified by refolding from inclusion bodies. Analysis of the deduced amino acid sequence revealed high homology with members of the bacterial lipase family I.3, showing 97% identity to a putative lipase from Pseudomonas fluorescens Pf0-1, and 93% identity to a crystallized extracellular lipase from Pseudomonas sp. MIS38. A typical C-terminal type I secretion signal and several putative Ca2+ binding sites were also identified. Experimental data confirmed that Lip I.3 requires Ca2+ ions for correct folding and activity. The enzyme differs from the previously reported family I.3 lipases in optimal pH, being the first acidophilic lipase reported in this family. Furthermore, Lip I.3 shows a strong preference for medium chain fatty acid esters and does not display interfacial activation. When tested for activity on secondary alcohol hydrolysis, Lip I.3 displayed higher efficiency on aromatic alcohols rather than on alkyl alcohols. Conclusions: A new family I.3 lipase with unusual properties has been isolated, cloned and described. This will contribute to a better knowledge of family I.3 lipases, a family that has been scarcely explored, and that might provide a novel source of biocatalysts. Significance and Impact of the Study: The unusual properties shown by Lip I.3 and the finding of activity and enantioselectivity on secondary alcohol esters may contribute to the development of new enzymatic tools for applied biocatalysis. A comparative study on the foaming properties and behavior at the air-water interface of soy and whey protein isolates were made. Foams were obtained by the method of gas bubbling. The initial rate of passage of liquid to the foam (vi) and the maximum volume of liquid incorporated to the foam (VLEmax) were determined. The destabilization process of the formed foams was analyzed by a biphasic second order equation. Measurements of equilibrium surface tension (water/air) and surface rheological properties were carried out in a dynamic drop tensiometer. The foaming capacity (vi and VLEmax) and the stability of foams prepared with the whey protein isolates (WPI) were better than those formulated with the soy protein isolates (SPI). WPI foams were more stable showing the lower values of rate constants of gravity drainage and disproportion. There were significant differences (P ≤ 0.05) in the dilatational modulus in the surface rheology measurements, which were higher at the interface with WPI, implying greater resistance of the film formed to collapse and disproportion. In conclusion, WPI formed better and more stable foams than the SPI. We have developed new surfactant agents based on hydrolyzed soybean proteins using papain, and we have studied their ability to form and stabilize emulsions. The interfacial behavior and the emulsifying properties were correlated to the structural changes that the proteins underwent. The hydrolysis reaction was stopped by dropping to pH 2 in one case, or rapidly dropping the temperature to -18ºC in the other. The structural and functional properties of the obtained products depended on the way the papain hydrolysis of the soy proteins was stopped. Hydrolysis did not have a beneficial effect on the emulsifying properties of those hydrolysates that were stopped by freezing. In the present work, new kinetics to describe the creaming stability of oil-in-water emulsions determined by backscattering measurements (BS) is proposed. The emulsions assayed exhibited a different backscattering profiles regarding creaming destabilization, hyperbolic and sigmoid one. Hyperbolic behavior can be described by a second order kinetics, where kh could be equaled to a rate constant that describes the creaming process and its values would indicate the stability of emulsions. While for the sigmoid BS pattern, kinetics with two terms, is adequate to describe the creaming process in contrast to kinetics previously reported in the literature. The kh value has the same meaning as before, and ks indicates the delaying effect on the creaming rate.
