Solid solutions could represent a viable alternative to better understand and control structure–property relationships of drugs, in order to optimize their properties for practical applications. Lamivudine (3TC) and emtricitabine (FTC) are nucleoside analogue reverse transcriptase inhibitor antiretroviral drugs, which have similar molecular structures, differing by a single fluorine atom, which is only present in the FTC molecule. Due to these similarities in structure and molecular resemblances, lamivudine and emtricitabine are good candidates for producing a solid solution with physicochemical properties controlled by the stoichiometry. Following this hypothesis, the formation of a nonconventional solid solution was verified, whose crystalline structure is not defined by any of the individual constituents but by the one of the lamivudine hydrate with emtricitabine as a solute. The crystalline structures of two members of the solid solution were determined showing a nonuniform distribution of the solute among the independent molecules of the asymmetric unit of the lamivudine hydrate structure. Thermal analysis investigations confirmed that physicochemical properties could be controlled through the variation of the emtricitabine content.

Here, we report a synergistic in situ synthesis of lithium-ion poly (ethylene citrate) with embedded titania nanoparticles in which the esterification process itself triggered the hydrolysis of the titanium alkoxide precursor to form a promising fluorine-free lithium-ion solid polymer electrolyte. Structural characterization by means of X-ray diffraction, small angle X-ray scattering and also confocal Raman spectroscopy revealed the formation of the polyester with embedded titania nanoparticles in agreement with our Raman spectra simulation by means of DFT methodologies. Our theoretical modeling of the polyester also envisaged the interaction of lithium-ion via a tridentate coordination with oxygen atoms of carboxylate (–COO-) and ester (–COO–) groups. There is a slight increase in the degree of polymerization and thermal stability for our lithium-ion polyester with increasing titania nanoparticles concentration. In consequence, a two-order magnitude increase in the ionic conductivity was observed, yielding 1.74× 10−4 S cm−1 for a 20% weight fraction of titania nanoparticles. Thus, involving a novel approach based in the synergistic in situ preparation of lithium-ion poly (ethylene citrate) with titania nanoparticles, this procedure contributes to solve the many safety and environmental issues of the constitutive materials of lithium-ion solid polymer electrolytes.

A summary of the resource.
A series of new hybrid compounds was prepared combining tetrahydropyran rings with different aromatic systems by means of a 1,2,3-triazole, using a building block strategy. The design of these structures was guided by Lead-Likeness and Molecular Analysis (LLAMA) software, adding modifications to our most potent scaffold (the tetrahydropyran ring) to generate promising “lead-like” candidates, which were subsequently compared against reported anticancer compounds. Our synthesized compounds demonstrated significant antiproliferative activity when compared with the standards cisplatin and 5-fluorouracil, across a panel of six different tumor cell lines. Moreover, compared with our group's previous hybrid compounds, these new structures exhibit similar activity while offering simpler synthesis and greater potential for structural diversification, a fact that was previously an issue. Further investigations on the most active compounds included assessments of reproductive cell survival, inhibition of cell migration, and effects on nuclear morphology, indicating potential diverse mechanisms of action for these compounds. Pharmacokinetic properties were also calculated for the whole series of compounds using the pkCSM online software.

Neurodegenerative diseases affect millions worldwide, posing significant challenges to public health. Oxidative stress and increased caspase-3 activity are key features in the development and progression of neurological disorders leading to neuronal damage and cell loss. As part of our ongoing efforts to identify new molecules with enhanced neuroprotective potential, we have synthesized six novel thiazole-nitrone hybrid compounds, designed as dual-action antiapoptotic and antioxidant agents. The nitrone moiety is known for its spin-trapping ability, which confers antioxidant and neuroprotective properties, while thiazole-containing compounds have been described as antioxidants and potent caspase-3 inhibitors. Our findings demonstrate that these hybrid compounds effectively reduce intracellular reactive oxygen species (ROS) levels and mitigate apoptosis rates in a hippocampal cell line by decreasing active caspase-3 activity, confirming their antioxidant and antiapoptotic properties. Notably, two of the six evaluated compounds exhibited the most promising neuroprotective profiles, including favorable blood-brain barrier permeability. Altogether, these results position the identified molecules as excellent candidates for further structure-based optimization, contributing to the development of more efficient therapeutic strategies against neurodegenerative disorders.

<p>En el marco de un intento activo de promover el desarrollo agrícola e industrial del Uruguay, durante la segunda presidencia de José Batlle y Ordoñez ¿1911=1015? se propicio la producciçon de carburantes que permitieran al país resolver su problema energético. A esos efectos el Instituto de Química Industrial =creado en 1912=, tuvo como uno de sus cometidos fundamentales la fabricación del "carburante nacionale" en base a alcohol. Como resultado de sus investigaciones, en 1923 se llevaron a cabo ensayos técnicamente exitosos, utilizando una mezcla de alcohol y nafta, sin embargo no tuvieron aplicación.</p>
<p>En los casi noventa años que han transcurrido desde entonces, el problema ha aparecido una y otra vez en la agenda política nacional, sin que se haya sin que se haya logrado su resolución hasta el presente. Buscando arrojar algo de luz sobre las posibles causas de este persistente estancamiento, consideramos importante realizar una aproximación a los veinte años posteriores a las investigaciones mencionadas, analizando la creación de la ANCAP (Administración Nacional de Combustibles, ALcohol y Pórtland), y el comportamiento que asumieron tanto ella como las compañías petrolíferas extranjeras establecidas en el país, respecto del tema en cuestión.</p>

Las enfermedades crónicas no trasmisibles (ECNT) son enfermedades no infecciosas ni contagiosas. Constituyen la principal causa de muerte tanto en Uruguay como en el mundo. Éstas incluyen: el cáncer, las enfermedades cardiovasculares (ECVs), la diabetes y las enfermedades respiratorias crónicas. Este trabajo se enfocó principalmente al estudio de la aterosclerosis, principal factor de riesgo para el desarrollo de ECVs. La aterosclerosis es una reacción inflamatoria crónica de la pared arterial, en cuyo origen y avance participan desde componentes del sistema inmune, hasta la dieta ingerida por el individuo. Los productos de la glicación avanzada (advanced glycation end products, AGEs), son un grupo de compuestos grande y heterogéneo, que se originan en la reacción de Maillard. Estos compuestos juegan un rol crucial en el desarrollo de ECNT mediante los siguientes mecanismos: a) el entrecruzamiento de proteínas alterando sus funciones y propiedades; b) activación de cascadas de señalización pro-inflamatorias mediante interacción con receptores específicos, o mediante mecanismos no dependientes de receptores; c) generación de compuestos oxidantes y pro-inflamatorios. Los compuestos fitoquímicos son compuestos bioactivos presentes en alimentos de origen vegetal. Entre éstos se encuentran los compuestos fenólicos, los cuales presentan una gran actividad antioxidante y antiglicante inhibiendo la formación de AGEs, y por ende disminuyendo el estrés oxidativo y las respuestas pro-inflamatorias. Incluso, se ha visto que algunos de estos compuestos presentan actividad antiaterómica, manteniendo la actividad antioxidante de partículas de lipoproteína de alta densidad (HDL), al evitar la oxidación de la enzima paraoxonasa 1 (PON-1) que contienen en su superficie. Estos compuestos con propiedades bioactivas, pueden ser utilizados en la formulación de alimentos funcionales, de forma de prevenir el desarrollo de ECNT. Por lo tanto, el principal objetivo de este trabajo fue el de obtener extractos inhibidores de la formación de los mismos a partir de hierbas medicinales de uso frecuente en Uruguay (Mentha x piperita L., Baccharis trimera Iless. y Peumus boldus Mol.), así como evaluar las diferentes vías de formación de compuestos AGEs provocados por la dieta. Conjuntamente, se estudió su comportamiento a lo largo del tracto gastrointestinal, de forma de determinar si la bioactividad se mantiene al alcanzar el intestino. Para efectuar este estudio, se realizaron extractos de las hierbas antes mencionadas utilizando dos solventes: agua destilada a 95°C (3 minutos) y etanol 95% a temperatura ambiente (24 horas). A su vez, se analizó la eficacia de un método de extracción alternativo (ultrasonido) a diferentes tiempos (1, 2 y 3 horas). Los resultados obtenidos demostraron en primer lugar, que la aplicación de ultrasonido no presenta diferencias significativas a la hora de extraer compuestos bioactivos, por lo que se seleccionaron los extractos en agua destilada a 95°C durante 3 minutos y en etanol 95% durante 24 horas. Todos los extractos seleccionados (figura 0), presentaron considerable XII contenido de compuestos fenólicos totales, así como una gran actividad antioxidante (frente a radicales ABTS•+ o peroxilo), antiglicante y anti-inflamatoria, incluso luego de haber sido sometidos al proceso de simulación gastrointestinal. Se sugiere que los extractos evitan la oxidación de la PON-1 en presencia de AAPH y SIN-1, manteniendo así su actividad arilesterasa. Asimismo, parecen afectar la distribución de subclases de partículas de HDL. En conclusión, surge de nuestros resultados que los extractos acuosos y etanólicos de las tres hierbas estudiadas, son muy buenos candidatos como fuente de compuestos fenólicos con capacidades bioactivas muy interesantes, pudiendo ser utilizados como ingredientes en alimentos funcionales.

La imagenología molecular es una disciplina que abarca innumerables herramientas. Entre sus aplicaciones se encuentra el diagnóstico in vivo del cáncer con agentes que reconocen marcadores tumorales y que a su vez poseen óptimas características para obtención de imágenes. Asimismo, proporciona información clínicamente fundamental, permitiendo una correcta selección del tratamiento a seguir y el monitoreo de sus efectos. Detalles como la localización, tamaño, morfología y cambios estructurales de un área afectada pueden ser detectados a través de los métodos convencionales de imagen. Sin embargo, técnicas de imagenología molecular como la SPECT, PET y FMT, entre otras, son cada vez más utilizadas. Debido a su característica no invasiva la imagen molecular permite evaluar la patología en su contexto, siendo clave para entender el proceso tumoral, sin perturbar el ambiente y proporcionando información adicional a los métodos convencionales. La estadificación tumoral, detección de metástasis, cirugía guiada, así como la cuantificación de una lesión son diferentes aplicaciones en este área. En este contexto, es muy importante el conocimiento de marcadores tumorales y el desarrollo de nuevos agentes de imagenología molecular específicos de estos. Los aptámeros son oligonucleótidos (DNA o RNA) que tienen la característica de reconocer su blanco con alta afinidad y especificidad. Similar a los anticuerpos en estas características, los aptámeros presentan ventajas que los hacen interesantes para su aplicación como agentes de imagen o terapia. El Sgc8-c es una secuencia truncada del aptámero sgc8, el cual muestra una unión específica y con alta afinidad al receptor PTK7. Sgc8-c tiene solamente 41 bases y una Kd de 0.78 nM para este receptor. Si bien este receptor está presente en células normales, su sobreexpresión ha sido observada en cáncer de colon, tumores gástricos, cáncer de pulmón, próstata, mama e incluso en metástasis. Si bien, su participación en la oncogénesis parece ser clara, aun no es un receptor extensamente caracterizado. El presente trabajo consistió en la investigación y desarrollo del aptámero Sgc8-c como agente de imagenología molecular en cáncer. Para ello Sgc8-c fue modificado en su extremo 5’ con HYNIC, DOTA y se marcó con Tecnecio-99m y Galio-67 respectivamente. Asimismo el aptámero Sgc8-c fue marcado con Alexa647, para su evaluación como agente de imagen óptica en el infrarrojo cercano. Todos los agentes desarrollados fueron evaluados fisicoquímicamente y frente a diferentes líneas celulares tumorales. Finalmente, las características de biodistribución y farmacocinética fueron estudiadas y tres modelos tumorales en ratón fueron seleccionados para realizar imágenes. Resumen 3 Molecular imaging is a discipline that involves many tools. One of its major applications is the in vivo diagnosis of cancer, using molecular probes that recognize tumor markers and have optimal characteristics for imaging. It also provides clinically important information, allowing the correct choice of treatment options and monitoring their effects. Details such as the location, size, morphology and structural changes of an affected area can be detected by conventional imaging methods. Moreover, molecular imaging techniques such as SPECT, PET and FMT, among others, are increasingly used. Due to its minimal invasive characteristics, molecular imaging allows the evaluation of the pathology in context, being key to understanding tumor process without disturbing the environment and providing additional information to conventional methods. Tumor staging, detection of metastasis, guided surgery, as well as quantification of disease are different applications in this area. In this context, knowledge of tumor markers and the development of new targeted molecular imaging agents is very important. Aptamers are oligonucleotides (DNA or RNA), which have the characteristics to recognize their target with high affinity and specificity. Similar to antibodies in their molecular recognition properties, aptamers offer advantages that make them interesting for use as imaging agents or therapy. Sgc8-c is a truncated aptamer sequence, which exhibits specific binding and high affinity for the receptor PTK7. The Sgc8-c aptamer has only 41 bases, but its Kd for its receptor is 0.78 nM. Although this receptor is present in normal cells, its overexpression has been observed in colon, gastric tumors, lung, prostate, breast cancer and even their metastases. While participation in oncogenesis seems clear, the receptor needs to be characterized more extensively in vivo as a valid biomarker. The work presented describes the research and development of aptamer Sgc8-c as a molecular imaging agent for cancer. Sgc8-c was modified at its 5 'end with radiometal chelators HYNIC and DOTA and labeled with technetium-99m and gallium-67 respectively. Also, the Sgc8-c aptamer was labeled with the near infrared dye Alexa647 for evaluation as an optical imaging agent. All agents were developed and evaluated physicochemically against different tumor cell lines. Finally, the pharmacokinetics and biodistribution characteristics of the labeled Sgc8-c aptamer were studied and three mouse tumor models were selected for imaging.

La química combinatoria dinámica ha surgido en las últimas dos décadas como una promisoria estrategia hacia la búsqueda de nuevos prototipos y la identificación de candidatos a fármacos. Ésta puede definirse como el conjunto de técnicas que permiten la síntesis simultánea de gran número de compuestos mediante la combinación sistemática de diversos precursores. La industria farmacéutica ha invertido considerables recursos en la búsqueda automatizada de compuestos que interaccionan con biomoléculas relevantes; dicha metodología se denomina ensayos de alto rendimiento (HTS por sus siglas en inglés). En los últimos años la academia ha comenzado a invertir en el desarrollo de dicha tecnología, hecho que se refleja en la aparición de los primeros prototipos activos en T. cruzi, T. brucei y Schistosoma mansonii, entre otros.

La academia ha reconocido la necesidad de tener en cuenta tanto las métricas ambientales como las económicas a la hora de desarrollar procesos que, en última instancia, encontrarán aplicación en la fabricación de productos químicos. Las aminas quirales son porciones de moléculas que constituyen un grupo de "estructuras preferidas" (“privileged scaffolds”) que son frecuentes en los productos naturales, fármacos, agroquímicos y otros productos químicos, y que se incluyen habitualmente en el diseño de bibliotecas para el descubrimiento de nuevas drogas. También se conoce que el 80 % de los 20 fármacos más recetados en medicinas son aminas quirales, lo que hace que estos compuestos sean valiosos componentes básicos en la industria farmacéutica. Esta tesis tiene como objetivo principal investigar nuevos biocatalizadores en síntesis para la producción de aminas quirales, utilizando tanto enzimas reportadas como nuevas enzimas aún no identificadas. Mediante análisis de motivos aminoacídicos conservados detectados en secuencias disponibles y análisis de motivos de iminoreductasas reportados en paralelo a nuestro trabajo, se realizó una búsqueda de nuevas IRED. Como resultado, se han encontrado varias secuencias que cumplen con las características para ser potenciales enzimas reductoras de iminas. Además, se aplicó un enfoque de “genome mining and motif análisis”, para la selección de potenciales secuencias R-ω-TA. A partir de 423 candidatos, la secuencia R-ω-TA de Capronia semiimmersa fue seleccionada, clonada y expresada en Escherichia coli . La enzima expresada demostró aceptar D-alanina así como también isopropilamina como grupo aminodonor y tener actividad sobre una variedad importante de cetonas. Ademas, ha sido demostrada la aplicación de una cascada quimioenzimática novedosa para la síntesis regio y estereoselectiva de un panel de pirrolidinas 2,5-disustituidas a partir de las correspondientes 1,4-dicetonas. Al mismo tiempo, se logró establecer un sistema de desracemización de aminas en una misma reacción, con cinco biocatalizadores actuando paralelamente de manera simple y práctica. De esta forma se logran obtener las aminas enantioméricamente puras a partir de una mezcla racémica o a partir de la amina con estereoquímica opuesta. Por otra parte, se logró ampliar los sistemas enzimáticos aplicados a la síntesis de aminas quirales, para ello se utilizó un nuevo tipo de transaminasa bifuncional denominada α,ω- DTAs, perteneciente a la metabolización de la putrescina. Este nuevo sistema, requiere solamente 3-5 equivalentes de aminodonor en el medio de reacción en contraste a los 50-500 equivalentes de grupo aminodonor utilizados normalmente.

La alta incidencia de floraciones de cianobacterias productoras de microcistinas en el país y la región representa un riesgo muy elevado para humanos y animales. A fin de estudiar el impacto y la presencia de las microcistinas (MCs) en animales, es importante disponer de métodos simples de bajo costo. Como primera aproximación a estos objetivos en peces, se estudiaron Astraloheros facetus (Castañetas) expuestas a una floración de Microcystis spp (MCs 60 y 600 μgMCs/L) en un bioensayo subcrónico (18 días). Si bien no hubo mortalidad, la histopatología mostró infiltración grasa en el hígado, más relevante en los peces expuestos a la mayor concentración. Para analizar MCs en pescados se optimizaron dos métodos inmunoquímicos sensibles basados en un anticuerpo recombinante de llama (nanobody) de alta especificidad: ELISA y MALDI-TOF cuantitativo, utilizando partículas magnéticas funcionalizadas. Los métodos fueron recientemente desarrollados localmente. La excelente correlación ELISA/MALDI-TOF (rSpearman = 0,988, p< 10-7) resalta el potencial de este ELISA como herramienta simple y costo-efectiva para minimizar las muestras a analizar por métodos de referencia. Las concentraciones de MCs en las Castañetas fueron relevantes, acordes con bioensayos en otras especies y peces de la naturaleza. Esto destaca la importancia de analizar MCs en pescado para consumo

Abstract
A summary of the resource.
Con un extracto crudo (EC) de importante actividad proteolítica, obtenido a partir de frutos maduros de Bromelia antiacantha se trataron epiginos de Apopyllus iheringi a fin de remover, mediante hidrólisis, los tejidos blandos presentes en estas estructuras. La acción enzimática sobre las muestras fue evaluada en distintas condiciones de: pH, actividad y tiempo de biocatálisis. Luego de la acción biocatalítica, se observaron las muestras bajo aumento, tomándose registros fotográficos. De todas las condiciones evaluadas, los mejores resultados de biocatálisis fueron obtenidos a pH 6.0, condición óptima de actividad de estos biocatalizadores con sustratos solubles. La digestión tisular completa, observada a través del Microscopio Electrónico de Barrido, se obtuvo a pH 6.0, 42ºC, actividad específica de 308.3 UE/ mg y 8 h de incubación. Utilizando las mismas condiciones de catálisis fueron tratados epiginos de otras dos especies de arañas obteniendo muy buenos resultados.

Por ejemplo, el ion ferroso en la hemoglobina es fundamental para el transporte de oxígeno, los iones calcio son un componente vital de los huesos y otros elementos de transición son esenciales para el funcionamiento de enzimas [1].
