<p>Biocatalysts have emerged as promising tools, with aldolases being among the most employed. Unfortunately, the features of their active sites that modulate the mechanistic details have been poorly assessed. Here, the electronic and structural factors associated with the substrates binding and catalytic mechanism of rhamnulose-<br />1-phosphate aldolase (RhuA) from E. coli are computationally analysed. The results allowed us to furnish a holistic view of the RhuA’s entire turnover cycle. The active site bears three pockets that anchor different moieties of the substrates: L-rhamnulose-1-phosphate (R1P), dihydroxyacetone phosphate (DHAP) and (S)-lactaldehyde (LLA). The results predict a 5-step catalytic mechanism, being the CeC bond breaking (Ea =24.2 kcal mol−1) and the DHAP deprotonation (Ea =22.0 kcal mol−1) the rate-determining steps for the retro- and aldolic<br />reactions, respectively. The residues E117, E171′, G31 and N29, and the Zn2+ cofactor, play a major role in stabilizing the correct substrates’ orientation, securing the stereocontrol at C4. Our findings suggest the presence of two acid/base catalytic residues, E117 and E171′, the latter of which being also involved in assisting the CeC<br />bond formation through a concerted protonation of the transition state.</p>

Abstract
A summary of the resource.
Current knowledge on the role of oxygen radicals in the development of chemotherapy for Chagas' disease is reviewed. This includes a) a brief description of the disease and its prevalence in the endemic area; b) applicability and toxicity of nifurtimox and benznidazole; c) the production of oxygen radicals by nitrocompounds, specially by nifurtimox; d) the action of nifurtimox and benzidazole on DNA, RNA and protein synthesis and degradation in Trypanosoma cruzi, including nuclear DNA and kinetoplast DNA cleavage and repair; e) nifurtimox reduction by lipoamide dehydrogenase; J) the role of T cruzi trypanothione system; g) the chemical and biological consequences of replacing the tetrahydrothiazine group of nifurtimox by unsaturated nitrogen heterocycles; h) the electronic properties of the new compounds, in relation to their capability for nitroanion radical production and trypanocidal action in vitro (QSAR) and in vivo; i) evidence for the possible role of oxygen radicals in the action of nifurtimox on T. cruzi and the human host. It is concluded that the chemotherapy of Chagas'disease remains an unsolved problem and, therefore, the search for new drugs must continue.

Reaction fl ow (RF) chromatography with fl uorescamine reagent and UV – vis detection was used for the analysis of amino acids. The performance advantage of RF chromatography was tested against the conventional post- column derivatisation (PCD) methods. The results of the study veri fi ed that greater sensitivity could be obtained using RF columns compared to conventional PCD methods. The reaction loops employed in the conventional PCD methods did not need to be used, which resulted in more ef fi cient separations using RF columns. The selective detection of amino acids in this RF study reduced limits of detection (LOD) and limits of quanti fi cation (LOQ) by about 50% compared to conventional methods of PCD. The calibration data of the RF-PCD technique also yielded R 2 ≥ 0.99 and %RSD in peak areas ranging from 3.0 – 8.5%. The RF method also enabled multiplexed detec- tion since both the derivatised and non-derivatised fl ow streams could be analysed simultaneously.

Ready-to-eat fruit and vegetables comprise a group of increasingly demanded value-added products. Broccoli is a highly perishable vegetable with unique nutritional characteristics. Development of minimally processed broccoli products demands varietal-specific knowledge as to which are the packaging conditions that preserve quality throughout shelf life. ‘Legacy’ cultivar broccoli florets were washed, disinfected, packaged in polypropylene and stored at 4, 8 and 15 °C for 21 d. Weight loss, internal atmosphere composition, respiration rate, color, texture, glucosinolate content, antioxidant capacity (AOC) and sensory attributes were evaluated throughout storage time. Results showed that 4 °C helped preserve sensory quality, texture, total glucosinolate content and AOC for 21 d. Temperature fluctuations reaching 15 °C resulted in loss of total glucosinolate content and unacceptable sensory quality. MAP helped mitigate temperature effects, especially at 8 °C. MAP is therefore an appropriate technology which can be applied to extend the shelf-life of ready-to-eat broccoli florets.

In search of prospective agents against infectious diseases, 1,1'-bis(diphenylphosphino)ferrocene pyridine-2-thiolato-1-oxide M(ii) hexafluorophosphate compounds [M(mpo)(dppf)](PF6), where M = palladium or platinum, were synthesized and fully characterized in the solid state and in solution using experimental and DFT computational techniques. The compounds are isomorphous and the M(ii) transition metal ions are in a nearly planar trapezoidal cis-coordination bound to the pyridine-2-thiolato-1-oxide (mpo) and to the 1,1'-bis(diphenylphosphino)ferrocene molecules, both acting as bidentate ligands. Both compounds showed high cytotoxic activity on Trypanosoma cruzi and Mycobacterium tuberculosis (MTB) and acceptable selectivities towards MTB, but good to excellent selectivity index values as anti-T. cruzi compounds. The inclusion of the ferrocene moiety (dppf ligand) improved the selectivity towards the parasite when compared to the previously reported [M(mpo)2] complexes. Related to the probable mechanism of action of the complexes, molecular docking studies on modelled T. cruzi NADH-fumarate reductase (TcFR) predicted that both be very good inhibitors of the enzyme. The effect of the compounds on the enzyme activity was experimentally confirmed using T. cruzi protein extracts. According to all obtained results, both [M(mpo)(dppf)](PF6) compounds could be considered prospective anti-trypanosomal agents that deserve further research.

A protease classified as trypsin was isolated and purified from the pyloric caeca of two species of fishes: Brevoortia spp (menhaden) and Mugil spp. (mullet). The characterization of both enzymes as trypsin was based on their molecular weight (24 kDa) determined by SDS-PAGE, their inhibition by some known trypsin inhibitors (PMSF, SBTI, TLCK and benzamidine), and their ability to hydrolyze the synthetic trypsin substrate N-α-Benzoyl-DL-arginine-p-nitroanilide (BAPA). Menhaden trypsin had maximum activity at pH 9.5 and was stable for 30 min between pH 6.0 and 10.0 at 0, 10 and 25C. On the other hand, mullet trypsin showed maximum activity at pH's from 7.8 to 9.0, and was stable over a wider pH range (7.0–10.0). The optimum temperature for menhaden and mullet trypsin was 63C and 60C respectively. Thermostability for menhaden trypsin was up to 50C, whereas mullet trypsin was stable up to 60C. The Km(BAPA) values for menhaden trypsin were conserved in the temperature range from 10 to 30C, while for mullet trypsin the Km(BAPA) was conserved between 10 and 25C.

Solubilization of muscle from South American menhaden, Brevoortia spp., was done with and without added proteolytic enzymes. Pepsin and trypsin were isolated from the stomach and the pyloric caeca of croaker (Micropogonias furnieri) respectively, and used for enzymatic solubilization. Digestion of muscle proteins was carried out at 8C for 20 h with the pH maintained at either 3, 4, 5, 6, 7, 8, 9 or 10. The hydrolytic action was evaluated by measuring the concentration of peptides and proteins solubilized from previously frozen muscle or from lyophilized muscle. The protein solubility of menhaden muscle was optimum at basic pH; approximately 50–60% of the previously frozen or lyophilized muscle was soluble after incubation without added enzyme at pH 9.0. All of the protein was solubilized from lyophilized menhaden muscle with purified trypsin at pH 10.0. Addition of croaker pepsin to menhaden muscle increased protein solubility by only 10–15%. Digestion of menhaden muscle with both pepsin and trypsin from croaker resulted in a similar increase in protein solubility as obtained by addition of only trypsin. Similar results were obtained with crude croaker stomach extracts and with purified pepsin, and also with crude pyloric caeca extract and with purified trypsin. Complete solubilization of menhaden muscle with croaker pyloric caeca extract may be an inexpensive way to separate the protein from lipids and prepare a protein isolate from this underutilized specie. Additional work is needed to characterize the functional properties of the menhaden protein extract obtained by digestion of the flesh at alkaline pH with croaker trypsin.

Trypsins from the pyloric caeca of menhaden (Brevoortia spp.) and croaker (Micropogonias furnieri) were used for hydrolysis of both myofibrillar and sarcoplasmic fractions from menhaden muscle. Digestion of muscle proteins was carried out at 37C for 20 min with the pH maintained at either 3, 4, 5, 6, 7, 8 or 9; or for 60 min at the pH's of 5, 7, 8 or 9 at 30C and 37C. The hydrolytic action was evaluated based on the concentration of peptides solubilized. Solubility after digestion of myofibrillar, sarcoplasmic fraction and hemoglobin was optimum at basic pH. The sarcoplasmic fraction was solubilized more than the myofibrillar fraction by both fish trypsins. Similar results were obtained with both crude pyloric extracts and purified trypsins from menhaden and white croaker. Thus, the preparation of the crude pyloric caeca may be preferentially used, because of its low cost as well as the simplicity of the preparative procedure.

A sequence for the fractionation of the amylasic components from a malted barley extract is proposed using two salt-promoted, adsorption processes: thiophilic interaction chromatography (TIC) and hydrophobic interaction chromatography (HIC). Two fractions containing α-amylase activity were recovered during the thiophilic chromatography; the first was resolved in to α-amylase I and β-amylase I by HIC on a phenyl-sepharose column; an enrichment factor of 32 was achieved for α-amylase I. The other amylasic component eluted from the thiophilic gel was characterized as α-amylase II. Although the adsorption of malt amylases on phenylsepharose and the thiophilic adsorbent is salt promoted, the interactions involved in each case are clearly distinguished by the different behaviour and disparate salt effects.

A summary of the resource.
The enantioselective preparation of three protected β-amino- γ -hydroxyesters from benzoic acid is described. The employed synthetic methodology involves the ipso, ortho cis-dihydroxylation of benzoic acid by the mutant strain Ralstonia eutropha B9, followed by a selective halonium induced beta lactamization. Modification of this novel β-lactam structure by the appropriate sequence of reactions allows for the selective preparation of the aforementioned β -amino- γ -hydroxyesters in a diastereodivergent manner. The overall transformation results in a selective formal aminohydroxylation of the diene moiety of the initial cis-cyclohexadienediol. The synthesized products are important building blocks and will allow for the selective preparation of aminoacids, inosamines and alkaloids from benzoic acid.

La adicción es un desorden neurobiológico en el que el uso repetido de sustancias de abuso corrompe los circuitos neuronales normales de recompensa y de conductas adaptativas, provocando cambios neuroplásticos en dicho circuito. Esta enfermedad crónica tiene graves y duraderas consecuencias para la salud del individuo y la sociedad.[1] El tratamiento farmacológico actual para la adicción a drogas de abuso está enfocado principalmente a disminuir los severos síntomas desarrollados durante la abstinencia o incluso a evitar la recaída. Sin embargo, hasta el momento no existen estrategias exitosas que logren revertir los cambios neurobiológicos implicados en el consumo crónico de una sustancia. A summary of the resource.
In the search for candidates of antiparasitic new drugs, macrocycle analogs and key fragments of aerucyclamides were obtained. The activities against Trypanosoma brucei brucei and cytotoxicities on murine macrophages (cell line J774) were evaluated. Two of the prepared compounds displayed similar anti-trypanosomal activities as aerucyclamides B and C. In addition, the compound with the highest activity presents a 150-fold selectivity (SI ¼ 150) for the parasite than against the murine macrophage

Abstract

This paper describes a new, improved systematic qualitative analysis of yucca saponins in commercial products by combined use of high-pressure liquid chromatography (HPLC) coupled to an evaporative light scattering detector (ELSD) and matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) mass spectrometry (MS). Three groups with four saponins in each were completely resolved by HPLC-ELSD under C18 reversed-phase conditions using a linear gradient composed of methanol and water. The selectivity of the described HPLC-ELSD method is given by the sapogenin differences at C12 (carbonyl group) and at C2 (hydroxyl group), followed by the presence or absence of an exomethylene group at C25, and the composition and length of the oligosaccharidic chain at C3 in the sapogenins. The saponins were identified by chemical and spectroscopic methods including MALDI-TOF MS1 and MS2, high resolution MS, and one and bi-dimensional nuclear magnetic resonance (NMR) experiments. Among the twelve saponins identified, eleven were previously reported, and one is reported for the first time in Yucca schidigera. An identification flowchart procedure based on the analysis of the sodium adducts of intact saponins [M+Na]+ and the oligosaccharidic chain ions obtained in the MALDI-TOF MS1 and the MS2 spectrums, was developed for the analysis of Y. schidigera saponins. Two intact isomeric saponins were differentiated by this method, which could also be applied to the structural assignment of other steroid and triterpenic saponins. Using the four-major saponins as reference standards, a C18 reversed-phase HPLC-ELSD method was validated for their specific analysis in commercial extracts of Y. schidigera. Finally, the applicability of the HPLC-ELSD method for the relative quantification of yucca saponins is discussed.

Yucca schidigera Roezl. (yucca) is one of the major industrial sources of steroid saponins approved by the Food and Drug Administration (FDA) as generally recognized as safe. Due to the increasing applications of yucca extracts, there is a need for a simple, low-cost method for the authentication of samples. This paper presents a method to obtain unique fingerprints of commercial products of yucca using a single-thin layer chromatography (TLC) system and two visualization reagents for the detection of phenolics and saponins, as a tool for the preliminary quality assessment of the products.

El Magnetismo Molecular ha sido un área de sostenido interés en los últimos 30 años. La búsqueda de la comprensión detallada de los fenómenos que influyen en el comportamiento magnético de especies de origen molecular ha sido su principal guía, con el objetivo de poder controlar factores como las interacciones y la anisotropía magnéticas, para lograr así especies que puedan presentar un mejor desempeño magnético con miras a potenciales aplicaciones tecnológicas. Este trabajo de tesis se enmarca en el Magnetismo Molecular y ha tenido como objetivo general la síntesis y caracterización estructural de nuevos compuestos de coordinación y el estudio de sus propiedades magnéticas. Para llevar adelante el trabajo se han adoptado dos diferentes estrategias de síntesis. La primera metodología comprende el uso de ligandos oxigenados con grupos alcoxo y carboxilato, los cuales presentan una gran flexibilidad con diferentes modos de coordinación en forma de puente. Cuando se mezclan directamente estos ligandos con iones metálicos 3d paramagnéticos, en reacciones que generalmente pueden proceder en un solo paso, es posible obtener diferentes especies de variadas topologías de espín de los centros metálicos. Si bien resulta difícil predecir el resultado final, esta metodología ha llevado a un avance importante en el magnetismo molecular ya que ha posibilitado el estudio de una gran diversidad de moléculas. Los resultados obtenidos siguiendo esta estrategia mediante el uso de iones metálicos 3d y ligandos tipo polipiridilcetona se muestran en la Sección II. En segundo lugar se ha llevado adelante una estrategia en la cual en una primera etapa se prepara un complejo mononuclear que contiene un ligando que puede funcionar como puente. Este precursor es usado luego, en una segunda etapa, como ligando frente a otro metal. Por otra parte, la mayoría de los estudios magnéticos se han realizado sobre compuestos Resumen conteniendo metales de la primera serie de transición, iones lantánidos o mezclas de ellos, mientras que relativamente muy poco trabajo se ha realizado sobre compuestos conteniendo iones 4d o 5d. Sin embargo, algunos iones de elementos de la segunda y tercera serie de transición presentan características químicas y magnéticas adecuadas para ser usados en compuestos precursores para coordinar a otros metales con miras a la síntesis de nuevos materiales magnéticos. En la Sección III se presentan resultados obtenidos a partir del uso de compuestos de Re(IV) como precursores de compuestos bimetálicos 5d/4f, 5d/3d y trimetálicos 5d/4f/3d. El contenido de esta tesis se organiza en tres secciones y cuatro apéndices. En la Sección I se encuentra la introducción y los objetivos, mientras que en Sección II y III ya mencionadas se presentan los resultados obtenidos. En la introducción realizada en la Sección I se presentan los aspectos generales de los materiales magnéticos moleculares que hacen de su estudio un área de interés, se detalla brevemente la evolución histórica del magnetismo molecular y se exponen algunos de los ejemplos más relevantes de estudios magnético-estructurales presentados en los últimos 30 años. Finalmente en esta sección se presentan los objetivos particulares de este trabajo. En la Sección II se presentan los resultados obtenidos trabajando con los iones metálicos Ni(II), Co(II) y Mn(II) con ligandos tipo polipiridilcetona, fundamentalmente con di-2-piridil cetona, (py)2CO, pero también con el ligando di-2,6(2-piridilcarbonil)piridina, pyCOpyCOpy. Los ligandos polipiridilcetona son atacados por el solvente en presencia del ion metálico generando nuevos ligandos tipo gem-diol o hemiacetal, ligandos que presentan una gran flexibilidad para la coordinación de hasta cinco centros metálicos por el mismo ligando. En la síntesis de los compuestos se ha usado además ligandos auxiliares como benzoato {O2CPh}- o picolinato (pic-). De esta forma se han obtenido los compuestos [M4{(py)2C(O)(OH)}4{O2CPh}4] ·2H2O (M = Ni(1) y Co(2)) de tipo cubano. Ambos compuestos neutros presentan interacciones ferromagnéticas. Por otra parte cuando se usa como ligando auxiliar picolinato se obtienen los compuestos [Ni4(pic)2{(py)2C(OH) O}2{(py)2C(OCH3)O}2](NO3)2 (3), [Ni4(pic)2{(py)2C(OH)O}4](NO3)2 (4) y Resumen [M4(pic)2{(py)2C(OH)O}2{(py)2C(OCH3)O}2](BPh4)2·2H2O (M = Ni(5), Co(6), y Mn(7)) los que presentan una estructura de tipo dicubano incompleto con un arreglo de los portadores de espín en forma de rombo. Los compuestos de Ni(II) presenta interacciones antiferromagnéticas dominantes mientras que el compuesto de Co(II) presenta interacciones ferromagnéticas y dependencia de la señal AC con la frecuencia. También ha sido posible para este sistema la preparación de soluciones solidas [MA xMB 4-x(pic)2{(py)2C(OH) O}2{(py)2C(OCH3)O}2](BPh4)2·2H2O (MA = Ni, MB = Co, Mn) (8-11). Estos compuestos bimetálicos son isoestructurales con los monometálicos anteriores y los valores de x dependen de la mezcla de partida de los iones Ni(II), Co(II) y Mn(II). Finalmente en esta sección también se informa la síntesis, resolución estructural y caracterización magnética del compuesto trinuclear angular [Ni3{pyC(OEt)(O)pyCOpy}2(O2CPh)2(EtOH)2](ClO4)2∙2EtOH (12) preparado a partir del ligando di-2,6(2-piridilcarbonil)piridina (pyCOpyCOpy), el cual presenta interacciones ferromagnéticas. En la Sección III se presentan los resultados obtenidos usando precursores de Re(IV). Este ion de la tercera serie de transición, que presenta generalmente entornos de coordinación octaédricos, con una relativa estabilidad frente a la sustitución de ligandos, así como inercia frente a procesos redox. Su estado fundamental presenta un estado con S = 3/2, y alta anisotropía magnética generada por efecto de la distorsión de la geometría octaédrica y del acoplamiento espín-órbita de segundo orden. El compuesto mononuclear NBu4[ReBr5(H2pydc)]∙2-PrOH (13) (H2pydc = ácido 3,5-pyridinedicarboxílico) ha sido obtenido por reacción de sustitución del ligando sobre el compuesto [ReBr6]2-. Su estructura y caracterización magnética son reportadas. El ligando H2pydc se encuentra coordinado al renio a través del nitrógeno piridínico, dejando disponibles los carboxiltaos para coordinar hacia otros metales. A partir de este precursor 13 se sintetizaron compuestos bimetálicos Re(IV)/Ln(III) siendo Ln = Dy, Pr y Sm. Los compuestos (NBu4)9{M3(H2O)2[ReBr5(pydc)]3[ReBr5(Hpydc)]4[ReBr5(H2pydc)]}∙7H2O, con M = Dy(14) y Sm(15) presentan estructuras en cadenas. El derivado de Dy(III) presenta dependencia de la frecuencia en la señal AC a baja temperatura, lo que sugiere un comportamiento de lenta relajación de la magnetización. El compuesto obtenido a partir del uso de praseodimio (NBu4)2{Pr(H2O)2(CH3OH)2[ReBr5(3,5pydc)][ReBr5(3,5pydcH)]} (16) también presenta una estructura monodimensional. El precursor 12 también se ha Resumen usado en la síntesis de compuestos trimetálicos 5d/4f/3d, llevando a la síntesis de las especies {ReBr5(H2pydc)}{NiGd(L)} (17) siendo L el ligando NiII(3-MeOsaltn)-(MeOH)(ac)DyIII(hfac)2], donde 3-MeOsaltn= N,N′-bis(3- metoxi-2-oxybenzilidene)-1,3-propanediaminato). En esta sección también se presenta la síntesis del compuesto {ReCl5(pyzc)}2{Ni([(py)2C(O)(OCH3)]2} (18) a partir del uso del precursor [ReCl5(pyzc)]2- (Hpyzc = ácido pirazincarboxílico), el ligando (py)2CO y Ni(II). En este compuesto tetranuclear ReNi2Re los dos átomos de níquel están unidos por dos ligandos [(py)2C(OCH3)O]- generando un módulo NiO2Ni con interacciones ferromagnéticas entre ambos iones 3d, y a su vez cada níquel está coordinado a un complejo de Re(IV) observándose interacciones ferromagnéticas entre renio y níquel. Al final de esta tesis, luego de las tres secciones, se presentan cuatro apéndices con datos cristalográficos (A1), detalles de las técnicas usadas (A2) y un breve repaso sobre la base teórica necesaria para comprender los resultados obtenidos (A3), así como los trabajos publicados en el marco de la Tesis (A4).
