Tannat is a red variety of Vitis vinifera that has become the emblematic red wine variety of Uruguay. Tannat wine is characterized by its high total phenolic content, which provides a good potential for aging and differential sensory characteristics. The aim of the present thesis was to characterize the astringency of Uruguayan Tannat wines from both a physicochemical and sensory perspective. Astringency is a complex and timedependent attribute, which exhibits buildup and carry over effects upon repeated ingestions, and is related to several subtle sensations that can be perceived simultaneously. This poses several challenges for the accurate sensory characterization of Tannat wine astringency, which have been addressed during the development of the thesis. First, qualitative studies with consumers were carried out to understand their conceptualization of astringency and the vocabulary they use for describing this complex attribute. Basic research about the applicability of different palate cleansers for wine astringency evaluation was also conducted. The astringency of 40 commercial wines was characterized using static and dynamic sensory methods: Time-Intensity (TI), checkall- that-apply (CATA) questions and Temporal Dominance of Sensations (TDS). The TI evaluations performed by a panel of trained assessors showed that commercial Tannat wines mainly differed in intensity-related parameters rather than in astringency development over time, although the variability was moderate. CATA questions involving 16 astringency sub-qualities proved to be a valuable tool to describe and differentiate a large sample set of Tannat wine samples based on their astringency characteristics. Different styles of Tannat wine were identified, which were described using a wide range of sub-qualities, from silky and velvety to harsh and aggressive. The phenolic profile of the 40 commercial Tannat wines was also characterized using reversed phase high performance liquid chromatography coupled with a mass spectrometer (HPLC-MS) and its relationship with sensory astringency was explored using boosted regression trees. The total content of condensed tannins as well and some specific phenolic compounds were identified as the main predictors of sensory astringency. Finally, the comparison of astringency evaluation of trained assessors and experts showed that their intensity assessments were partly related, but their description of astringency sub-qualities differed. Insights on the astringency sub-qualities related to expert’s perception of astringency quality were also revealed. Los inhibidores peptídicos de la enzima convertidora de angiotensina-I (ECA-I) son muy interesantes por ser compuestos naturales con efecto antihipertensivo. Muchos péptidos bioactivos han sido obtenidos por hidrólisis enzimática de proteínas alimentarias y las proteínas lácteas han tenido un papel destacado. Sin embargo, las peptidasas empleadas han sido fundamentalmente enzimas comerciales de origen animal o microbiano. El uso de peptidasas de plantas, obtenidas de fuentes menos convencionales, podría liberar nuevos péptidos inhibidores de ECA-I o aquellos que se sabe forman parte de la secuencia de las proteínas del LS pero que aún no han sido liberados con las enzimas clásicas. Las peptidasas de una especie de Bromeliácea autóctona, recientemente caracterizadas por nuestro grupo de investigación, presentan características interesantes para la hidrólisis del LS y aún no han sido utilizadas para la preparación in vitro de hidrolizados de proteínas. En este sentido, se estudió el potencial de las peptidasas de Bromelia antiacantha Bertol., para hidrolizar el LS y evaluar la actividad inhibidora de la ECA-I. Demostramos que las peptidasas de B. antiacantha fueron capaces de hidrolizar a las proteínas del LS con un importante efecto del pH de reacción para modificar la susceptibilidad de hidrólisis de α-lactalbumina (ALA) y β-lactoglobulina (BLG), las proteínas mayoritarias del LS. Se obtuvieron hidrolizados de LS, ALA y BLG con potente actividad inhibitoria de ECA-I. En todos los casos, la fracción peptídica de tamaño menor a 2 kDa presentó una actividad inhibitoria de ECA-I potente y esta fue resistente a un proceso de digestión gastrointestinal simulado in vitro. A partir del hidrolizado de ALA se capturaron por cromatografía de afinidad y posteriormente se identificaron por espectrofotometría de masas dos nuevos péptidos derivados de ALA con actividad inhibitoria de ECA-I. A partir de estudios de cinética de inhibición de ECA-I fue posible establecer que el péptido Thr-Thr-Phe-His-Thr-Ser-Gly-Tyr (TTFHTSGY) se comportó como un inhibidor total, mientras que el péptido Gly-Tyr-Asp-Thr-Gln-Ala-Ile-Val-Gln (GYDTQAIVQ) fue capaz de inhibir parcialmente a ECA-I y a concentraciones elevadas de inhibidor por lo que no se continuó su caracterización. Sin embargo, hay dos secuencias con actividad inhibitoria de ECA-I reportada encriptadas en el péptido GYDTQAIVQ (GY e IVQ) que podrían ser liberadas durante su digestión gastrointestinal, convirtiéndolo en productos potencialmente más activos in vivo. TTFHTSGY mostró una cinética de inhibición compatible con un inhibidor competitivo. The aim of the study was to evaluate the influence of intrinsic and extrinsic characteristics of the regular and probiotic version of two milk products on consumer overall liking and perceived wellbeing. In Study 1, consumers evaluated milk samples with different label design. In Study 2, consumers evaluated samples of chocolate flavored milk differing in sugar content and nutrition information. In both studies, consumers rated their overall liking and completed a check-all-thatapply question composed of expressions related to different dimensions of wellbeing. Overall liking scores were significantly affected by the sensory characteristics of samples but not by the characteristics of the labels. In Study 2, the highest average overall liking scores were found for samples with high sugar content (6.2 vs. 6.0). The frequency with which consumers used some of the wellbeing terms was influenced by both intrinsic and extrinsic variables. In Study 1, the term “It is nutritious” was significantly more used to described probiotic milk compared to regular milk. In Study 2, sugar content and the information included on the labels influenced the frequency of use of terms related to physical and psychological wellbeing, such as “It is good for wellbeing” and “It makes me feel guilty.” Practical applications Although hedonic aspects of food consumption are major determinants of consumer food choices, results from the present work showed that consumer perceived wellbeing enabled the discrimination of samples that did not significantly differ in their overall liking scores. This suggests that wellbeing measurements can provide complementary information about consumer perception of food products to overall liking scores. Projective mapping (PM), one of the most holistic product profiling methods in approach, is increasingly being used to uncover consumers' perception of products and packages. Assessors rely on a process of synthesis for evaluating product information, which would determine the relative importance of the perceived characteristics they use for mapping them. Individual differences are expected, as participants are not instructed on the characteristics to consider for evaluating the degree of difference among samples, generating different perceptual spaces. Individual differences in cognitive style can affect synthesis processes and thus their perception of similarities and differences among samples. In this study, the influence of the cognitive style in the results of PM was explored. Two consumer studies were performed, one aimed at describing intrinsic sensory characteristics of chocolate flavoured milk and the other one looking into extrinsic (package only) of blueberry yogurts. Consumers completed the wholistic-analytic module of the extended Verbal Imagery Cognitive Styles Test & Extended Cognitive Style Analysis-Wholistic Analytic Test, to characterize their cognitive style. Differences between wholistic and analytic consumers in how they evaluated samples using projective mapping were found in both studies. Analytics separated the samples more in the PM perceptual space than wholistic consumers, showing more discriminating abilities. This may come from a deeper analysis of the samples, both from intrinsic and extrinsic point of views. From a sensory perspective (intrinsic), analytic consumers relied on more sensory characteristics, while wholistic mainly discriminated samples according to sweetness and bitterness/chocolate flavour. In the extrinsic study however, even if analytic consumers discriminated more between packs, they described the products using similar words in the descriptive step. One important recommendation coming from this study is the need to consider higher dimensions in the interpretation of projective mapping tasks, as the first dimensions could underestimate the complexity of the perceptual space; currently, most applications of PM consider two dimensions only, which may not uncover the perception of specific groups of consumers. Introducción: el cáncer de mama representa la primera causa de muerte por cáncer en mujeres de Uruguay. Se estima que cerca de 7% son causados por mutaciones en el ácido desoxirribonucleico germinal. Los costos de la secuenciación genética han descendido dramáticamente gracias a la aparición de la secuenciación de nueva generación (NGS). El cambio tecnológico abrió una nueva etapa en el estudio del cáncer hereditario en nuestro país. Objetivo: comunicar los resultados de la utilización de tecnología NGS y paneles multigénicos en familias uruguayas con alto riesgo de cáncer de mama hereditario. Pacientes y método: se secuenciaron 135 familias de alto riesgo que provenían de la consulta de consejería genética que funciona en el Grupo Colaborativo Uruguayo: Investigación de afecciones oncológicas hereditarias. Cuando la historia familiar sugería claramente un síndrome de cáncer de mama y ovario hereditario se efectuó secuenciación NGS exclusiva de los genes BRCA1 y 2; cuando el patrón familiar no configuraba claramente se utilizó un panel multigénico. Resultados: se efectuó NGS exclusiva de genes BRCA1 y 2 en 62 familias y un panel multigénico en 73 familias. Se identificaron en total 29 mutaciones patógenas (21 en genes BRCA y 8 en otros genes). Dos de ellas fueron noveles y tres pueden considerarse recurrentes en la población uruguaya. Conclusiones: este trabajo es el primero en Uruguay en reportar el resultado de esta nueva tecnología en el cáncer de mama hereditario. El hallazgo de 29 mutaciones patógenas nos ayuda a delinear el perfil mutacional de nuestro país. BACKGROUND: We sought to evaluate, in patients on a liver transplantation waiting list, potential biomarkers of the base calcineurin pathway activity with use of a new model of nonstimulated peripheral blood mononuclear cells (PBMC) and ex vivo response to tacrolimus (TAC). METHODS: The calcineurin pathway activity was explored ex vivo in stimulated and nonstimulated PBMC from 19 patients. The inhibition of NFAT1 translocation to PBMC nuclei, expression of intracellular IL-2, and membrane CD25 in different T-cell subsets were measured by multiparametric flow cytometry before and after exposure to TAC. We also studied the influence on the individual response of polymorphisms in 3 key genes of the calcineurin pathway: PPIA, PPP3CA, and IL2RA. RESULTS: All pharmacodynamics profiles closely fitted an I/Imax sigmoid model. Interindividual variability was higher in nonstimulated than in stimulated conditions, as well as in the presence of TAC. IL-2+CD8+ cells at TAC Imax showed the highest interindividual variability, suggesting its usefulness as a biomarker of individual TAC effects integrating many different sources of regulation and variability. Moreover, in the absence of TAC, patients with end-stage liver disease exhibited lower NFAT1 translocation and T-cell activation than healthy volunteers from a previous study under similar conditions. Multivariate statistical analysis showed strong and significant associations between TAC pharmacodynamic parameters and 2 polymorphisms in the gene-coding cyclophilin A (rs8177826 and rs6850). CONCLUSIONS: We show the feasibility of using nonstimulated PBMCs to explore the calcineurin pathway under more physiologic conditions and point toward potential biomarkers for TAC pharmacodynamic monitoring. The enantioselective preparation of three protected β-amino- γ -hydroxyesters from benzoic acid is described. The employed synthetic methodology involves the ipso, ortho cis-dihydroxylation of benzoic acid by the mutant strain Ralstonia eutropha B9, followed by a selective halonium induced beta lactamization. Modification of this novel β-lactam structure by the appropriate sequence of reactions allows for the selective preparation of the aforementioned β -amino- γ -hydroxyesters in a diastereodivergent manner. The overall transformation results in a selective formal aminohydroxylation of the diene moiety of the initial cis-cyclohexadienediol. The synthesized products are important building blocks and will allow for the selective preparation of aminoacids, inosamines and alkaloids from benzoic acid.
