El arroz (Oryza sativa) es el cereal cultivado más importante a nivel mundial (672×106 toneladas /año). Su destino prioritario es la alimentación humana como arroz blanco. El Uruguay es un exportador neto de arroz blanco, generando anualmente 130 mil toneladas de salvado de arroz como residuo. Aunque el 10% de este salvado tiene como destino la producción de aceite y una parte se utiliza para la alimentación animal, es de gran interés estudiar su uso potencial en la alimentación humana. Las enfermedades no transmisibles son responsables de la muerte de 40 millones de personas en el mundo cada año. Uno de los factores de riesgo más relevante son el sobrepeso y la obesidad. Estos están directamente relacionados un mayor riesgo de padecer diabetes tipo 2, enfermedades cardiovasculares, entre otras. Dados los actuales hábitos alimenticios, de alto consumo de azúcares libres se prevé un incremento de la incidencia global de estas patologías en los próximos años. La presente tesis doctoral se enfoca en la posibilidad de aprovechar una materia prima sub-valorada como el salvado de arroz, con el objetivo de desarrollar nuevos ingredientes alimentarios para el consumo humano que contribuyan a la reducción del riesgo de padecer estas patologías crónicas no trasmisibles. Para ello, se desarrolló un extracto rico en antioxidantes polifenólicos cuyas condiciones de producción fueron optimizadas a través de un diseño experimental central compuesto (CCD) 2k = 23 = 16 tratamientos, con 3 variables independientes (% de etanol, tiempo y temperatura) a 3 niveles (-1, 0 y +1) y utilizando tecnologías simples y fácilmente escalables. El residuo insoluble generado tras la obtención de dicho extracto fue caracterizado por análisis de su composición nutricional y capacidad antioxidante demostrando ser una fracción rica en fibra dietética antioxidante. x Las condiciones de extracción optimizadas (maceración simple con 35% de etanol en la solución extractora, a 20°C por 105 minutos) permitió obtener un extracto de salvado de arroz principalmente rico en proteínas (12,74 g/100g de MS) y en carbohidratos (77,0 g/100g MS). Bajo dichas condiciones, se recuperó el total de fibra dietética soluble (2,23 ±0,10 g/100g MS) que compone el salvado de arroz; así como, un contenido de lípidos totales de 3,38 ±0,07 g/100g MS y de cenizas de 4,82±0,06 g/100g MS. El extracto antioxidante resultó ser rico en polifenoles (71,42±1,60 mg de GAE/ g de extracto) y presentó elevada capacidad antioxidante (451±14 μmol de TE/g muestra frente a radical ABTS.+ y 912±31 μmol de TE/g muestra frente a radicales peroxilos). El análisis cromatográfico por UHPLC indicó que los componentes fenólicos mayoritarios presentes en extracto son ácido clorogénico (44, 84 ± 2, 25 mg / 100 g de materia seca) y ácido ferúlico (12,44 ± 0,60 mg / 100 g de materia seca). El extracto presentó valores IC50 de 0,14 ± 0,01 y 4,50 ± 0,08 mg/mL contra radicales peroxilos y ABTS.+, respectivamente. Para su mejor caracterización el extracto seleccionado se fraccionó mediante ultrafiltración empleando una membrana con tamaño de poro uniforme de 3 kDa. Aplicando este procedimiento se encontró que el extracto está constituido por un 68% de compuestos con tamaño molecular mayor a 3 kDa (FAPM) y 32% de compuestos con tamaño menor a este valor (FBPM). El análisis cromatográfico de ambas fracciones indica que la FAPM contiene principalmente ácido ferúlico mientras que el compuesto fenólico mayoritario que compone a la FBPM es ácido clorogénico. El estudio de bioaccesibilidad de los componentes bioactivos que forman el extracto antioxidante de salvado de arroz llevado a cabo por simulación gastrointestinal in vitro indicó una pérdida significativa (p6g/100g de galleta). Se utilizó stevia como edulcorante no nutritivo resultando ser un alimento que no contiene azúcar añadido y; por lo tanto, presenta potencial para reducir el riesgo de diabetes. El alimento presentó una capacidad antioxidante de 3653 ± 150 μmol TE/ porción considerando los compuestos xii extraíbles e hidrolizables presentes y de 1125 ± 73 μmol de TE/porción de galleta (30g) por el método de ABTS-QUENCHER. El % de bioaccesibilidad de los polifenoles presentes en las galletas fue de 17% respecto a los polifenoles totales presentes en una porción de 30g de alimento (33,07±1,59 mgGAE/porción). El promedio de aceptabilidad general de las nuevas galletas libres de gluten, fuente de fibra dietética y antioxidantes fue de 51% (88 evaluaciones). La formulación debe considerarse un prototipo inicial sobre el cual debe trabajarse para obtener un alimento de óptima calidad sensorial. En conclusión, se puede afirmar con un 95% de confianza que es posible obtener 2 nuevos ingredientes alimentarios de calidad nutricional y funcional a partir de salvado de arroz: extracto antioxidante (fracción soluble) y la fracción insoluble rico en fibra dietética antioxidante. El proceso de extracción propuesto es de gran simplicidad y bajo costo de inversión por lo que se alinean al concepto de “extracción verde” destacándose el hecho de no generar nuevos residuos de producción siendo innovadora, eco-sostenible y de promisoria implementación en la industria uruguaya. Las galletas elaboradas permiten afirmar que se ha logrado un alimento tradicional de gran consumo regional que reúne varias de las condiciones recomendadas sobre una dieta saludable por la OMS (2014 y 2015) para la reducción del riesgo de padecer enfermedades crónicas no transmisibles en un solo alimento. Las galletas no contienen gluten, ni azúcar añadido (endulzado con stevia como edulcorante no nutritivo), son fuente de fibra alimentaria antioxidante (3 g /porción) y presentan capacidad antioxidante siendo un primer prototipo de alimento funcional que valida la recuperación del salvado de arroz.

El objetivo de esta tesis fue el estuóio y la caracterización de lipasas de semillas. Se seleccionó un grupo de especies de la familia de las Cucurbitaceas: zapallo (CuGurbita pepo), zapallito (CuGurbita maxima), pepino (Cucumis'satlvus) melon (Cucumis melo) y sandia (Citrolus lanatus). Esta familia se caracteriza por utilizar triacilgliceroles como fuente de carbono y energia, por 10 tanto es de esperar que durante la germinación de sus semillas aparezcan lipasas en sus tejidos de reserva. Para caracterizar las lipasas en su comportamiento en la hidrólisis endógena del aceite de reserva se estudió la variación de su composición y de su contenido en distintos estadios de la germinación. El estudio se realizó a varias temperaturas de germinación para determinar el efecto de esta variable. El efecto de la temperatura sobre las velocidades de germinación de estas especies, medidas en función del tiempo que demora una plantula en alcanzar un cierto estadio en su desarrollo, es muy notorio. Se establecieron correlaciones entre la velocidad de germinaci6n y la velocidad de consumo del aceite alas distintas temperaturas de trabajo. La composición del aceite varió muy poco durante la gerrninaci6n, aunque se observó un aumento en los <icidos grasos que 10 integran en forma minoritaria. Para poder utilizar estas lipasas, se las extrajo de los tejidos y se estudió su comportamiento en la hidr6lisis de un aceite comercial. Como los extractos obtenidos por 10s metodos utilizados comUnmente en la extracci6n de lipasas de semillas (crudos 0 polvos de acetona) presentaron actividad muy baja para todas las especies estudiadas, se optimizó un metodo diferente para lograr su extracción, y a los preparados de lipasas, obtenidos por ese metoda, se les llamó concentrados en polvo (C. P.) .. La actividad lipolitica de los C. P. se estudió en función del estadio y de la temperatura de germinación de las semillas. Las mayores actividades lipoliticas se obtuvieron en estadios de germinación altos y cuando la temperatura es la óptima para la germinación de las semillas.

<p>Se estudiaron las distintas condiciones de extracción de beta-anilasa de Ipomea batata.</p>
<p>El extracto fue purificado en dos etapas sucesivas por intercambio iónico en DEAE Sephadex A-50 y por el gel filtración en Sephadex G-200.</p>
<p>En la etapa de intercambio iónico se determinaron las condiciones óptimas de pH, fuerza iónica y cantidad de intercambiador para retener el máximo de impurezas.</p>
<p>Previo a la gel filtración la enzima, parcialmente purificada fue concentrada por ultrafiltración con membrana Amicon 10.</p>
<p>La purificación obtenida en las distintas etapas fue controlada por electroforesis en gel de poliacrilamida y por determinación del aumento de actividad específica.</p>
<p>Por este método se obtuvo una preparación de la beta-amilasa altamente purificada como lo demuestra la homogeneidad en la electroforesis en gel de poliacrilamida y la alta actividad específica alcanzada.</p>

Ochratoxin A (OTA) is a nephrotoxic mycotoxinwhich has been foundworldwide as a contaminant inwines. It is produced on grapes mainly by molds fromAspergillus section Nigri. This study has demonstrated for the first time the occurrence of black aspergilli on Tannat grapes from Uruguay, in a two year survey. Aspergillus uvarum (uniseriate) and Aspergillus welwitschiae (from Aspergillusniger aggregate) were the prevalent species whereas Aspergillus carbonarius which is considered the main OTA producing species was not detected. OTA production in culture medium was evaluated for native isolates from A. niger aggregate and compared to levels produced by a type strain of A. carbonarius. Thiswork also includes the development of quick and easymolecular methods to identify black aspergilli to species level, avoiding sequencing.

<p>The results of an experiment to study the occurrence and distribution of pesticide residues during rice cropping and processing are reported. Four herbicides, nine fungicides, and two insecticides (azoxystrobin, byspiribac-sodium, carbendazim, clomazone, difenoconazole, epoxiconazole, isoprothiolane, kresoxim-methyl, propanil, quinclorac, tebuconazole, thiamethoxam, tricyclazole, trifloxystrobin, λ-cyhalotrin) were applied to an isolated rice-crop plot under controlled conditions, during the 2009−2010 cropping season in Uruguay. Paddy rice was harvested and industrially processed to brown rice, white rice, and rice bran, which were analyzed for pesticide residues using the original QuEChERS methodology and its citrate variation by LC-MS/MS and GC-MS. The distribution of pesticide residues was uneven among the different matrices. Ten different pesticide residues were found in paddy rice, seven in brown rice, and eight in rice bran. The highest concentrations were detected in paddy rice. These results provide information regarding the fate of pesticides in the rice food chain and its safety for consumers</p>

Pesticides can be found in bee products as they are usually employed to protect the beehive, but they can also reach the hive due to environmental contamination. These contaminants could be present in processed foods. One of the most common and consumed comfitures based in bee products are honey and propolis candies, for which no analytical method has yet been developed. This work presents the development of an ethyl acetate based extraction method followed by dispersive clean up using Primary and Secondary Amine (PSA) plus Graphitized Carbon Black (GCB) with Gas Chromatography coupled to Mass Spectrometry (GC-MS) determination for pesticide residue monitoring candies containing honey and propolis from the Mercosur region. Sixteen pesticides found in bee products as well as three pesticide metabolites of toxicological significance were evaluated including acaricides and insecticides (pyrethroids, organophosphates and some organochlorines). The method was validated, with a LOQ of 0.01 mg/kg for most analyzed pesticides, showing a recovery rate of 70-120% with <20% RSD. Real samples from the Mercosur countries were analyzed. Coumaphos and chlorpyrifos residues were detected in most of them. From these findings a preliminary toxicological evaluation of coumaphos via admissible daily intake (ADI) estimation was conducted.

Environmental sustainability of South American rainfed agroecosystems is of current concern. In this work, we evaluate the occurrence of multiple pesticide residues in muscle tissue of wild fish species from two large rivers in South America (Uruguay and Negro Rivers). Two sampling campaigns (representing summer and winter crops) were performed during 2015 targeting a wide biodiversity of fish species used for human consumption (ranging from migratory to non-migratory and from detritivorous to top-predators). Three different localities associated to rainfed agriculture were assessed, two of them enclosed to a RAMSAR site (National Park “Esteros de Farrapos e Islas del Rio Uruguay”). Pesticide residues occurred in muscle tissue of 143 from 149 sampled fishes (96%). Thirty different pesticides were detected at concentrations from <1 to 194 μg kg−1. Incidence of pesticides in fish were tightly related to: i) features of the contaminant: (Kow, environmental persistence and mobility) and ii) intensity of use of particular pesticides and land dedicated to rainfed agriculture. Trifloxystrobin, metolachlor and pyraclostrobin showed the highest rates of occurrence. Of great concern is that strobirulins have highest toxicity to fish from those detected compounds. From the pattern of pesticides occurring for non-migratory fish species it was possible to trend important spatial differences related to the intensity of rainfed agriculture. Results suggest a regular exposition of aquatic wild biota to sublethal concentrations of multiple semi-polar pesticides.

Apiculate yeasts of the genus Hanseniaspora/Kloeckera are the main species present on mature grapes and play a significant role at the beginning of fermentation, producing enzymes and aroma compounds that expand the diversity of wine color and flavor. Ten species of the genus Hanseniaspora have been recovered from grapes and are associated in two groups: H. valbyensis, H. guilliermondii, H. uvarum, H. opuntiae, H. thailandica, H. meyeri, and H. clermontiae; and H. vineae, H. osmophila, and H. occidentalis. This review focuses on the application of some strains belonging to this genus in co-fermentation with Saccharomyces cerevisiae that demonstrates their positive contribution to winemaking. Some consistent results have shown more intense flavors and complex, full-bodied wines, compared with wines produced by the use of S. cerevisiae alone. Recent genetic and physiologic studies have improved the knowledge of the Hanseniaspora/Kloeckera species. Significant increases in acetyl esters, benzenoids, and sesquiterpene flavor compounds, and relative decreases in alcohols and acids have been reported, due to different fermentation pathways compared to conventional wine yeasts

A summary of the resource.
Currently, in addition to the use of olive oil in cosmetics, the use of olive-derived bioactives and their incorporation into cosmetics is a growing trend. The olive oil industry produces vast quantities of by-products, such as olive mill wastewater, olive pomace and leaves from which new ingredients may be obtained for cosmetic use. In this way, by-products are revalorized, which contributes to the implementation of a sustainable economy or upcycling. This review intends to provide a detailed overview of the different extraction techniques reported in order to obtain the bioactive compounds of cosmetic value that can be found in olive by-products: fatty acids, tocopherols, polyphenols, phytosterols and squalene. Different extraction techniques are presented, including some traditional techniques (solid–liquid extraction) and more novel or “greener” ones: ultrasound, microwave, supercritical extraction, pressurized fluids and deep eutectic solvents. Additionally, different applications of olive by-products in skin care products are explored: emollient, antioxidant, anti-age, anti-inflammatory, antiviral, antifungal and antibacterial, and the perspective of consumers is also considered since they increasingly demand products formulated with natural ingredients.

The species of genus Baccharis spp. (Compositae) are an important natural resource in the search for new bioactive compounds. This article presents an investigation of volatile compounds of two species of Baccharis that are native to southern Brazil (Baccharis dentata (Vell.) G.M. Barroso and Baccharis uncinella DC) collected in summer and autumn of 2011. The essential oils were extracted by steam distillation and analyzed by gas chromatography-mass spectrometry (GC-MS), olfactometry analysis (GC-O) and qualitative assessment of the antimicrobial potential using the indirect bioautography method against Staphylococcus aureus, Salmonella enterica subspecie enterica serovar Choleraesuis, Pseudomonas aeruginosa, Candida albicans, Aspergillus niger, Candida parapsilosis, Cryptococcus neoformans, Rhizopus stolonifer, Fusarium solani and Aspergillus oryzae. The results demonstrated that the harvest period has an influence on the composition of the essential oils, but little influence on extraction yield. It can also be observed that the joint analysis of GC-MS and GC-O resulted in complementary information about the essential oils studied because the intensities of volatile compounds detected by both methods are significantly different. Regarding antimicrobial assays, it was found that the essential oil of B. dentata was effective against S. aureus, A. niger, C. parapsilosis and R. stolonifer, whereas the essential oil of B. uncinella was effective against S. aureus, S. Choleraesuis, C. albicans, A. niger, C. parapsilosis, R. stolonifer and F. solani.

We have performed a comparative study on the reactivity of metallic and semiconducting nanotubes using infinite and finite models. Infinite models were created using periodic boundary conditions while finite ones were constructed by means of hydrogen terminated nanotubes sections. Cluster models systematically underestimate the reactivity of metallic single wall carbon nanotube (SWCNT)s. We have confirmed that metallic nanotubes are more reactive than semiconducting species, in disagreement with previous works. The differences can be attributed to the presence of an instability in the singlet ground state of the wavefunction corresponding to semiconducting nanotubes clusters. When lower electronic states of the pristine cluster are considered, semiconducting nanotubes become less reactive as compared with metallic SWCNTs. Particularly, if an antiferromagnetic solution is considered for the semiconducting (10,0) SWCNT cluster, it becomes less reactive than the (5,5) SWCNT, as observed for infinite models. Because semiconducting nanotubes are less reactive than metallic counterparts, their reaction energies converge faster to the values observed for graphene. For a 1.6-nm diameter semiconducting nanotube, the addition energy is comparable with graphene. Thus, semiconducting nanotubes with diameters larger than 1.6 nm are going to be as reactive as graphene and the effects of curvature will be unimportant.
