A summary of the resource.
The negative effect of lead exposure on children’s intelligence is well-documented. Less is known about the impact of lead on the use of executive functions to self-regulate behavior. We measured blood lead level (BLL) in a sample of first grade children from Montevideo, Uruguay (n = 206, age 6.7 ± 0.5 years, 59.7% boys). Behavior was assessed with teacher versions of the Conners Rating Scale (CRS) and the Behavior Rating Inventory of Executive Functions (BRIEF). Mean BLL was 4.2 ± 2.1 μg/dL; 10% had mild-to-severe ratings of Attentional Deficit with Hyperactivity Disorder (ADHD) (T score > 65). In negative binomial regression, BLL was not associated with CRS sub-scales, but was associated with a poorer ability to inhibit inappropriate behaviors, prevalence ratio (PR) [95% CI]: 1.01 [1.00, 1.03] as measured by the BRIEF. In covariate-adjusted models, the association with BLL was attenuated. When stratified by sex, the covariate-adjusted association between BLL, hyperactivity, poorer inhitibion, emotional control, and behavioral regulation was marginally significant for girls but not boys. In summary, among children with low lead-exposure, we found some, but nonetheless modest, evidence of a relationship between higher BLL and child behavior. If confirmed by larger studies and other objective measures of behavior, such links could have implications for learning and social interaction, particularly among girls.

La inocuidad de los alimentos engloba diferentes acciones con el objetivo de garantizar la máxima seguridad posible de los mismos. Casi la totalidad de las frutas y algunas hortalizas pueden ser consumidas sin necesidad de cocción, pero pueden contener contaminantes que llegan al producto en cualquier punto, desde su cultivo, cosecha, y hasta en la cadena de procesamiento que la lleva al consumidor final. Con el objetivo de monitorear los contaminantes inorgánicos, arsénico (As), cadmio (Cd) y plomo (Pb) de forma rápida, confiable y con métodos analíticos en acuerdo con los principios de la Química Analítica Verde, se desarrollaron y validaron dos metodologías para la extracción simultánea de estos elementos. Un método asistido con microondas, en un solo paso y utilizando ácido diluido. Y un método asistido por ondas de ultrasonido y ácido diluido. Se aplicaron los métodos validados para la determinación de estos elementos a cuatro productos: zanahoria, lechuga, manzana y tomate. Los parámetros de desempeño de las metodologías validadas indicaron que ambas resultaron adecuadas para monitorear estos contaminantes en los referidos alimentos tomando como referencia la reglamentación vigente. Es importante destacar que los límites de detección obtenidos con el método asistido con ondas de ultrasonido fueron uno o dos órdenes de magnitud más bajos que los alcanzados con el método asistido con microondas, lo que la convierte a la extracción asistida por ondas de ultrasonido en una técnica simple y muy eficiente para los fines propuestos. Todas las muestras analizadas arrojaron resultados comparables por ambos métodos, y los niveles de Cd, Pb y As en todas ellas estuvieron por debajo de la concentración máxima permitida (LMR), establecida por el Reglamento Técnico MERCOSUR y el Codex Alimentarius. Ambas metodologías pueden ser aplicadas a estudios de inocuidad, esto es de relevancia y deja disponible una plataforma analítica al servicio del sector productivo y organismos de contralor nacional.

<p>El cáncer de mama es la principal neoplasia en mujeres y la segunda causa de muerte por cáncer a nivel mundial (sólo el cáncer de pulmón mata a más mujeres).<br />Aproximadamente dos de cada tres cánceres de mama muestran una sobreexpresión de receptores de hormonas, en particular del receptor de estrógeno (RE) y receptor<br />de progesterona (RP).<br />El presente trabajo plantea contribuir al diagnóstico y caracterización del cáncer de mama receptor de estrógeno positivo (RE+), a través del desarrollo de potenciales radiofármacos para Imagenología molecular dirigidos al RE.<br />La primer etapa del trabajo consistió en la síntesis, purificación y formulación del 16α-[18F]fluoroestradiol ([18F]FES), radiofármaco considerado como de referencia en Imagenología de RE en un módulo automatizado de síntesis Synthra RNplus Research.<br />Una vez puesta a punto la misma y realizados tres lotes pilotos se procedió a efectuar estudios in vitro, en donde se empleó la línea celular MCF-7 portadora de RE, derivadas de un adenocarcinoma mamario humano. Los estudios demostraron una elevada captación celular, siendo la misma de 6.3 ± 0.4 %.<br />Al mismo tiempo, se trabajó en la síntesis de ligandos derivados de estradiol para la formación de compuestos de coordinación con 99mTc. Para la síntesis de los mismos, se tomó como reactivo de partida al etinilestradiol, fármaco agonista de los receptores de estrógenos. Se probaron distintas estrategias sintéticas para la obtención del ligando Cyclam-2-(2-(1H-1,2,3-triazol-1-il)acetamida) estradiol (L1) sin éxito, lo que llevó a proponer un nuevo ligando: Cyclam-3-(2-(2-(3-(1H-1,2,3 triazol)propoxi)etoxi)etoxi) propanamina estradiol (L2) que permite la formación de un complejo de 99mTc. Debido a que no fue posible su adecuada marcación, se diseñó un tercer ligando: 17[4-((2-azido-1-carboxietil)amino)-N-metilidino-4-oxobutano-1-1aminio)-1H-1.2.3-triazol-1-il] estradiol (L3), que permite la formación de un complejo de 99mTc(III) “4+1”, utilizando como coligando a el ácido 2,2',2''nitrilotriethanethiol-2-((2-carboxietoxi)metil)propano-1,3-di(oxo)dipropanoico (NS3(COOH)3). Dicho complejo fue obtenido con una adecuada pureza radioquímica. Los estudios fisicoquímicos demostraron una alta hidrofilia ya que presentó un log P de -2.05 ± 0.20. El complejo resultó ser estable en plasma a 37°C y en el medio de reacción durante al menos 4 horas. La unión a proteínas plasmáticas fue moderada siendo la misma de 32 ± 7 %. Los ensayos in vitro de unión a células MCF-7 indicaron ausencia de captación celular 0.10 ± 0.02 %, por lo cual no se realizaron estudios in vivo. Se planteó como posible hipótesis que la razón de ausencia de unión pudiera ser el bajo valor de lipofilicidad fuera del rango adecuado para atravesar barreras biológicas.<br />Los principales logros en el marco de esta tesis fueron: la obtención del [18F]FES, para su uso en clínica y como estándar de comparación de potenciales radiofármacos desarrollados y a desarrollar por nuestro grupo de investigación; por otro lado, se realizaron casi todas las etapas involucradas en el desarrollo de un potencial radiofármaco, desde la selección del farmacóforo y radionucleido, síntesis orgánica, marcación, estudios fisicoquímicos y estudios in vitro.</p>

Abstract

Las frutas y hortalizas pueden contener contaminantes que llegan al producto en cualquier punto de la cadena productiva. En este sentido, se debe controlar la acumulación de elementos potencialmente tóxicos como arsénico (As), cadmio (Cd) y plomo (Pb) para asegurar y controlar la inocuidad de estos alimentos. Con el objetivo de monitorear los contaminantes inorgánicos, de forma rápida, confiable y con métodos analíticos en acuerdo con los principios de la química verde, se desarrolló y validó una metodología para la extracción simultánea de estos elementos asistida con microondas, en un solo paso y utilizando ácido diluido.

<p>Complexes [Sb(2Ac4oClPh)Cl2] (1), [Sb(2Ac4oFPh)Cl2] (2), [Sb(2Ac4oNO2Ph)Cl2] (3), [Sb(2Bz4oClPh)Cl2] (4), [Sb(2Bz4oFPh)Cl2] (5) and [Sb(2Bz4oNO2Ph)Cl2] (6) were obtained with 2-acetylpyridine-N(4)-orthochlorophenyl thiosemicarbazone (H2Ac4oClPh) and its N(4)-ortho-fluor (H2Ac4oFPh) and N(4)-ortho-nitro (H2Ac4oNO2Ph) analogues, and with the corresponding 2-benzoylpyridine-derived thiosemicarbazones (H2Bz4oClPh, H2Bz4oFPh, H2Bz4oNO2Ph). The studied compounds are excellent inhibitors of Trypanosoma cruzi growth. H2Bz4oClPh and complexes (4) and (1) were the most trypanosomicidal. Upon coordination of H2Ac4oClPh to antimony(III) in 1, the therapeutic index (TI) goes from 10.58 to 14.35. However, the best values of TI were found for H2Bz4oClPh (TI = 1240) and H2Ac4oNO2Ph (TI = 773). Structure–activity relationship (SAR) studies did not allow the establishment of correlations between the anti-trypanosomal activity and physico-chemical parameters, but correlations were found between the cytotoxicities and physico-chemical properties.</p>

Herein, we employed first-principle calculations to study the structure and reactivity of dual-doped graphene. The new materials were derived from graphene by replacing two carbon atoms with one 2p element (B, N or O), and one 3p element (Al, Si, P or S). A total of 12 dual-doped graphenes were used to perform a comparative study and identify the most promising materials for the development of new catalysts and anchoring nanoparticles. The structural analysis indicated that in all cases, except SiB, the dopants prefer to replace a CC bond. Yet, not always the dopants are bonded, presenting an edge-like bonding. As regards chemical reactivity, in general the introduction of two atoms remarkably increases reactivity as compared to graphene. The most prominent examples are Al–O, S–N, P–O and Si–B(para) codoped graphene which present reactivity higher than perfect and monodoped graphenes. This effect was attributed to the strong charge redistribution induced by the dopants. In most cases the heteroatoms are the most reactive sites, but in others the carbon atoms are more reactive, as in Al–O and Si–B codoped graphene. The combination of 2p and 3p dopants offers the possibility of adjusting over a wide range the reactivity of graphene.<br /><br />

Understanding the role of defects in the magnetic properties of the graphene buffer layer (BL) grown on substrates should be important to provide hints for manufacturing future graphene-based spintronic devices in a controlled fashion. Herein, density functional theory was applied to assess the structure and magnetic properties of defective BL on 6H-SiC(0001). Particularly, we conducted a thorough study of one and two vacancies and Stone–Wales defects in the BL. Our results reveal that the removal of a carbon atom in the BL framework that was originally bonded to a Si atom in the substrate is preferred over that of a sp2-bonded atom. As a result, a hexacoordinated silicon atom is formed with a slightly deviated octahedral geometry. A stable antiferromagnetic (AF) state was verified for the single vacancy system, with a quite different spin-density distribution to the one obtained for the perfect BL. Also, this AF state is nearly degenerate with the non-magnetic and low magnetic states. As for the Stone–Wales defect, the AF sate is almost degenerate with the most stable M = 2 mB magnetic configuration. However, the introduction of two vacancies in the carbon network of BL causes the loss of magnetism of the BL–SiC system. Our theoretical calculations support experimental predictions favoring the BL as the site for single vacancy formation rather than the epitaxial monolayer graphene, by 4.3 eV

The heteroatom doping of graphene is a useful method to amplify the outstanding properties of graphene. However, it is difficult to control the disposition of the dopants. By means of first-principles calculations, we show that the substrate-induced doping of epitaxial graphene leads to the formation of silicon doped graphene with a precise location of the dopants in the valley regions between crests. According to our results, in the valleys silicon dopants greatly prefer to replace the carbon atoms of the buffer layer which are bonded to the substrate by a large amount of energy. This selective doping suggests that a siligraphene layer in the g-SiC3 form would be an excellent model for a buffer layer grown on nanometer-sized areas of SiC, thus paving the way to the synthesis of a new two-dimensional material. We predict a stable antiferromagnetical (AF) ordering that behaves electronically as a semiconductor, degenerated with the highest ferromagnetic configuration of the system. A net magnetic moment of 0.65 μB per Si dangling bond was found for both the AF and the most stable ferromagnetic state of the SiC-siligraphene system. Finally, we demonstrate that quasi-free-standing epitaxial siligraphene layers on 6H-SiC(0001) could be obtained by the intercalation of argon atoms. We expect that this work can motivate new experimental studies pursuing the synthesis of SiC3 siligraphene.

Los biomateriales han evolucionado hacia la creación de dispositivos que se adapten al organismo biológico al cual se integran. Diferentes materiales han sido utilizados con el fin de mejorar su resistencia y biocompatibilidad. El presente trabajo de investigación explora una perspectiva diferente a la síntesis de materiales para sustitución ósea. En base a un modelo diseñado de interacción entre superficies (metal – proteína), se formuló como hipótesis de trabajo la idea que la dimensionalidad de la superficie y la dimensionalidad de la estructura debían ser diferentes para cumplir con una adecuada complementariedad de las superficies interaccionantes. Esta hipótesis se trasladó al desarrollo de biomateriales y al estudio de la interacción a nivel de la interface metal – cobertura de hidroxiapatita. Para ello se estudió la morfología y las propiedades fisicoquímicas de tres materiales con superficies modificadas a nivel macro y micro, con y sin hidroxiapatita por técnicas de SEM, FTIR, EDS, XPS, AFM, XRD y EIS. Los resultados mostraron la incidencia de la nanoestructuración del metal y la presencia de regiones no-apatíticas en los eventos de reconocimiento molecular a nivel de la interface. Con base en estos resultados, se discute la formulación de una nueva visión para el diseño de biomateriales implantables

Penicillium digitatum is the main species responsible for postharvest losses in citrus fruit and, therefore, rapid and accurate methods are needed for its early detection. For this purpose, a new procedure based on real-time quantitative PCR (qPCR) was developed to detect and quantify P. digitatum in lemons. The procedure included a rapid extraction of fungal DNA using the commercial Quick-DNA Fungal/Bacterial Miniprep Kit; the design of a specific primers pair by selecting the conserved region of the calmodulin A (cmdA) gene; and the design and evaluation of a qPCR system based on SYBR-Green dye detection chemistry. The functionality of the developed method was demonstrated by the high linear correlation of the standard curve constructed over the entire range of fungal DNA concentrations used (R2 > 0.99), indicating precision and accuracy of the qPCR. The protocol was developed aiming to evaluate the biotechnological potential of the biocontrol yeast Clavispora lusitaniae AgL21, by quantifying the fungus P. digitatum in lemon wounds. The yeast was shown to restrict the pathogen growth, reaching fungal DNA concentrations of up to 0.038 ng/wound after 4 days of incubation, compared to 173.06 ng/wound when the fungus was alone. No inhibition of qPCR due to food matrices was observed. The developed qPCR protocol could be considered as a suitable tool to detect P. digitatum in food products and can be used to specifically test the ability of new biological control microorganisms against green mold in lemons.

<span>This study compares the performance of Hanseniaspora vineae (Hv) under mixed culture fermentation with Saccharomyces cerevisiae, with a conventional control (Sc) for the production of base wine. Hv exhibited fermentation kinetics comparable to controls when its nutritional requirements were satisfied. Volatile acidity in Hv from one batch was significantly lower (–0.3 g/L) compared to the controls, while in the other two batches the levels were equal. Interestingly, Hv trials showed a significant degradation of malic acid (–0.9 g/L on average) without variations of pH. Even though the matrix influenced the volatile composition, the characteristic features of H. vineae remained intact, with 24-fold higher levels of 2-phenylethyl acetate. There was no higher production of 2-aminoacetophenone, albeit tryptophan (known as a 2-aminoacetophenone precursor) is a preferred amino acid for H. vineae. Results suggest the potential of H. vineae to produce base wine with a broad spectrum of aroma compounds that increase flavor complexity, if obtained from nutrient-balanced grape musts.</span>

A summary of the resource.
The plantation and natural forests of South America have been highly impacted by native and exotic pests in recent decades. The interaction of emerging invasive pests, climate change, and timber markets will define the region’s forests, with significant but uncertain ecological changes and economic losses expected. The Southern Cone Forest Health Group (SCFHG), a joint ad hoc initiative run by forest health professionals from Argentina, Brazil, Chile, and Uruguay, aims to strengthen relationships between the forestry industry, stakeholders, academia, and government agencies across the region. Here, we highlight regional strengths, weaknesses, threats, and opportunities to address forest health issues in the region. A regional approach with a strong communication network is relevant for future actions. In the current global scenario of invasive species and climate change, the implementation of practices that incorporate the resilience of forest ecosystems and sustainable management needs to be prioritized in forest policy across the region. Understanding that pests and pathogens do not recognize borders, we call on governments and organizations to support joint actions with agreements and adequate resources to enhance our regional capabilities.

Abstract

Mixed strain dynamics are still not well or easily monitored although recently molecular identification methods have improved our knowledge. This study used a chromogenic differential plating medium that allows the discrimination of four of the main selected biofuel strains that are currently under development for ethanol production from cellulosic hydrolysates. Complete fermentation of hexoses and xylose was obtained with a yeast consortium composed of Spathaspora passalidarum, Scheffersomyces stipitis, Candida akabanensis and Saccharomyces cerevisiae. The results showed that C.akabanensis excessively dominated consortium balance. Reducing its inoculum from 33 to 4.8% improved population strain balance and fermentation efficiency. Comparison of the consortia with single strain fermentations showed that it optimize sugar consumption and ethanol yields. This simple and cheap method also has advantages compared with molecular methods, as the yeast strains do not need to be genetically marked and identified cell proportions are probably active in the fermentation system as compared to DNA determination methods.

This work reports a computational analysis of hydrogen‐bonded clusters of mono‐, di‐, tri‐, and tetrahydrates of the chlorohydrins CH3CHClOH (1ClEtOH) and CH2ClCH2OH (2ClEtOH). The goal of the study is to assess the role of the water solvent into the facilitation of the initial step for dehalogenation of these compounds, a process of interest in several contexts. Molecular orbital methods (MP2), density functional methods (B3LYP, M06, and ωB97X‐D), and composite model chemistries (CBS‐QB3, G4) were employed to investigate the structure, electronic distribution, and hydrogen‐bonded structure of seven monohydrates, six dihydrates, five trihydrates, and five tetrahydrates of both species. Standard reaction enthalpy and standard Gibbs free reaction energy (urn:x-wiley:00207608:media:qua25931:qua25931-math-0001) were computed for all aggregates with respect to n independent water molecules and with respect to the dimer, trimer, and tetramer of water, respectively, in order to evaluate stability and hydrogen bonding network. The influence of the water chains on the length and vibrational frequencies, especially of the CCl and OH bonds, was evaluated. Hydrogen bonding in the complexes is discussed at length.

Pathogenic Gram-negative bacteria resistant to almost all β-lactam antibiotics are a major public health threat. Zn(II)-dependent or metallo-β-lactamases (MBLs) produced by these bacteria inactivate most β-lactam antibiotics, including the carbapenems, which are “last line therapies” for life-threatening Gram-negative infections. NDM-1 is a carbapenemase belonging to the MBL family that is rapidly spreading worldwide. Regrettably, inhibitors of MBLs are not yet developed. Here we present the bisthiazolidine (BTZ) scaffold as a structure with some features of β-lactam substrates, which can be modified with metal-binding groups to target the MBL active site. Inspired by known interactions of MBLs with β-lactams, we designed four BTZs that behave as in vitro NDM-1 inhibitors with Ki values in the low micromolar range (from 7 ± 1 to 19 ± 3 μM). NMR spectroscopy demonstrated that they inhibit hydrolysis of imipenem in NDM-1-producing Escherichia coli. In vitro time kill cell-based assays against a variety of bacterial strains harboring blaNDM-1 including Acinetobacter baumannii show that the compounds restore the antibacterial activity of imipenem. A crystal structure of the most potent heterocycle (L-CS319) in complex with NDM-1 at 1.9 Å resolution identified both structural determinants for inhibitor binding and opportunities for further improvements in potency.

The grapevine (Vitis vinifera) cultivar Tannat is cultivated mainly in Uruguay for the production of high-quality red wines. Tannat berries have unusually high levels of polyphenolic compounds, producing wines with an intense purple color and remarkable antioxidant properties. We investigated the genetic basis of these important characteristics by sequencing the genome of the Uruguayan Tannat clone UY11 using Illumina technology, followed by a mixture of de novo assembly and iterative mapping onto the PN40024 reference genome. RNA sequencing data for genome reannotation were processed using a combination of reference-guided annotation and de novo transcript assembly, allowing 5901 previously unannotated or unassembled genes to be defined and resulting in the discovery of 1873 genes that were not shared with PN40024. Expression analysis showed that these cultivar-specific genes contributed substantially (up to 81.24%) to the overall expression of enzymes involved in the synthesis of phenolic and polyphenolic compounds that contribute to the unique characteristics of the Tannat berries. The characterization of the Tannat genome therefore indicated that the grapevine reference genome lacks many genes that appear to be relevant for the varietal phenotype.
