The larval stage of the cestode Echinococcus granulosus causes cystic echinococcosis in humans and livestock. This larva is protected by the millimeter-thick, mucin-based laminated layer (LL), from which materials have to be shed to allow parasite growth. We previously reported that dendritic cells (DCs) respond to microscopic pieces of the mucin gel of the LL (pLL) with unconventional maturation phenotypes, in the absence or presence of Toll-like receptor (TLR) agonists, including lipopolysaccharide (LPS). We also reported that the presence of pLL inhibited the activating phosphorylation of the phosphatidylinositol 3-kinase (PI3K) effector Akt induced by granulocyte-macrophage colony-stimulating factor or interleukin-4. We now show that the inhibitory effect of pLL extends to LPS as a PI3K activator, and results in diminished phosphorylation of GSK3 downstream from Akt. Functionally, the inhibition of Akt and GSK3 phosphorylation are linked to the blunted upregulation of CD40, a major feature of the unconventional maturation phenotype. Paradoxically, all aspects of unconventional maturation induced by pLL depend on PI3K class I. Additional components of the phagocytic machinery are needed, but phagocytosis of pLL particles is not required. These observations hint at a DC response mechanism related to receptor-independent mechanisms proposed for certain crystalline and synthetic polymer-based particles; this would fit the previously reported lack of detection of molecular-level motifs necessary of the effects of pLL on DCs. Finally, we report that DCs exposed to pLL are able to condition DCs not exposed to the material so that these cannot upregulate CD40 in full in response to LPS.

Hemp (Cannabis sativa L.) seeds are composed of a high oil content rich in polyunsaturated fatty acids and antioxidants such as tocopherols and carotenes, resulting in an interesting nutritional aspect. Oil from hemp seeds extracted using supercritical carbon dioxide (scCO2) as the solvent preserves the qualities described above. However, the high operating pressures and the low solubility of the oil in scCO2 significantly increase operating costs. Generally, the use of pressurized n-propane as a solvent for extraction of vegetable oils is an alternative to overcome these challenges without changing their nutritional qualities. The main objective of this work was to study the extraction of the oil from dehulled hemp seeds using scCO2, pressurized n-propane and organic solvents at atmosphere conditions, to compare some physicochemical properties of the extracts. The extraction conditions using scCO2 as the solvent were 40 and 60 °C and 30 and 40 MPa. These operating conditions of extraction are similar to those available in the literature. For pressurized n-propane used as a solvent, temperatures of 40, 50 and 60 °C and pressures of 6, 8 and 10 MPa were used. The values of the extraction pressures with n-propane were defined from the knowledge of the phase behavior measured for the system n-propane (1) / hemp seed oil (2). The values of the mass percentage yields were similar for both pressurized solvents. However, the solubility of the oil in n-propane was higher than in scCO2. The kinetics of the extractions were satisfactorily using the Sovová modeling.

Doping leading to low electrical resistivity in electrodeposited thin films of Cu2O is a straightforward requirement for the construction of efficient electronic and energy devices. Here, Bi (7 at. %) doped Cu2O layers were deposited electrochemically onto Si(100) single-crystal substrates from aqueous solutions containing bismuth nitrate and cupric sulfate. X-ray photoelectron spectroscopy shows that Bi ions in a Cu2O lattice have an oxidation valence of 3+ and glancing angle X-ray diffraction measurements indicated no presence of secondary phases. The reduction in the electrical resistivity from undoped to Bi-doped Cu2O is of 4 and 2 orders of magnitude for electrical measurements at 230 and 300 K, respectively. From variations in the lattice parameter and the refractive index, the electrical resistivity decrease is addressed to an increase in the density of Cu vacancies. Density functional theory (DFT) calculations supported the experimental findings. The DFT results showed that in a 6% Bi doped Cu2O cell, the formation of Cu vacancies is more favorable than in an undoped Cu2O one. Moreover, from DFT data was observed that there is an increase (decrease) of the Cu2O band gap (activation energy) for 6% Bi doping, which is consistent with the experimental results.

Grape pomaces have a wide and diverse antioxidant phenolics composition. Six Uruguayan red grape pomaces were evaluated in their phenolics composition, antioxidant capacity, and anti inflammatory properties. Not only radical scavenging methods as DPPH· and ABTS·+ were employed but also ORAC and FRAP analyses were applied to assess the antioxidant potency of the extracts. The antioxidant reactivity of all extracts against hydroxyl radicals was assessed with ESR. The phenol profile of the most bioactive extract was analyzed by HPLC-MS, and a set of 57 structures were determined. To investigate the potential anti-inflammatory activity of the extracts, Nuclear Factor kappa-B (NF-κB) modulation was evaluated in the human colon cancer reporter cell line (HT-29-NF-κB-hrGFP). Our results suggest that Tannat grapes pomaces have higher phenolic content and antioxidant capacity compared to Cabernet Franc. These extracts inhibited TNF-alpha mediated NF-κB activation and IL-8 production when added to reporter cells. A molecular docking study was carried out to rationalize the experimental results allowing us to propose the proactive interaction between the NF-κB, the grape extracts phenols, and their putative anti-inflammatory bioactivity. The present findings show that red grape pomace constitutes a sustainable source of phenolic compounds, which may be valuable for pharmaceutical, cosmetic, and food industry applications.

We combine molecular modeling and NMR methods to better understand intramolecular hydrogen bonding (H-bonding) in a frequently used arylamide foldamer building block, ortho-methoxy-N-methylbenzamide. Our results show that solvents have a profound influence on the cumulative number and stabilizing effects of intramolecular H-bonds, and thus conformational preferences, of foldamers based on this compound. While intramolecular H-bonds are conserved in aprotic environments, they are significantly disrupted in protic solvents. Furthermore, these solvent effects can be accurately quantified using the computational approach presented here. The results could have significant implications in foldamer design, particularly for applications in aqueous environments

A summary of the resource.
The electron spin resonance (ESR) spectra of free radicals obtained by electrolytic or microsomal reduction of several potential antiprotozoal 1,2,5-oxadiazoles were characterized and analyzed. Ab initio molecular orbital calculations were performed to obtain the optimized geometries and the theoretical hyperfine constant was carried out using ZINDO semiempirical methodology. Density functional theory was used to rationalize the reduction potentials of these compounds.

Los glicósidos son moléculas ampliamente distribuidas en la naturaleza, que han demostrado poseer variadas actividades biológicas. En algunos casos esa actividad viene dada por la estructura global de la molécula; en otros, el residuo glicosídico puede ser importante para la actividad biológica, ya sea para aumentar la potencia de la misma o para mejorar los parámetros farmacocinéticos de la molécula cuando es utilizada como fármaco. La glicosilación enzimática es una potente herramienta para la generación de glicósidos. Las glicosidasas, usadas en las condiciones adecuadas, son capaces de transferir una unidad de grupo glicosilo de una molécula dadora a otra que lo acepta. En esta tesis, con el fin de sintetizar glicósidos con potencial actividad biológica, se estudiaron sistemas de transglicosilación utilizando como catalizadores la -galactosidasa de Aspergillus oryzae y la -glucosidasa de almendras. Como moléculas aceptoras de grupo glicosilo se seleccionaron dos alcoholaminas (3-amino-1-propanol y 1-amino-2-propanol) e hidroxiurea. Se logró sintetizar, purificar e identificar el compuesto 3-aminopropil-β-Dgalactopiranósido utilizando lactosa como dador de grupo galactosilo, 3-amino-1-propanol como aceptor y -galactosidasa (6 UE/mL), a pH 5.5, 50°C y 24 hs de reacción. Las concentraciones óptimas seleccionadas fueron 100 mM de lactosa y 500 mM de 3- aminopropanol. La lactosa también demostró ser aceptor del sistema de transglicosilación dando lugar a la formación de un trisacárido que fue identificado como o-β-Dgalactopiranosil-( 1,4)-β-D-galactopiranosil-(1,4)-α-D-glucopiranósido. Si bien los dos productos formados no mostraron ser mejores inhibidores de galectina 1 que la lactosa, lo que los descarta como posibles agentes antitumorales, el 3-amino-propil--Dgalactopiranósido es una molécula muy interesante desde el punto de vista químico, dado que constituye una galactosa funcionalizada con un grupo amino de alta reactividad. Tanto el 1-amino-2-propanol como la hidroxiurea no pudieron ser glicosilados utilizando los sistemas de transglicosilación propuestos, lo que hace pensar que no son buenos aceptores para el sistema. El 1-amino-2-propanol es un alcohol secundario lo que estaría confirmando el hecho de que esta -galactosidasa tiene alta selectividad para alcoholes primarios que son más reactivos. . El hecho de que la hidroxiurea no sea buen aceptor en ambos sistemas de transglicosilación podría estar relacionado con su estructura, ya que es probable que la cercanía del grupo amino, afecte la nucleofilia del hidroxilo. A su vez, es posible que el grupo carbonilo fuertemente electrófilo limite la electronegatividad del grupo hidroxilo disminuyendo así su nucleofilia. En estos casos, ambos aceptores, el 2- amino-1-propanol y la hidroxiurea demostraron ser inhibidores de la -galactosidasa de Aspergillus oryzae . Sin embargo ésta no es la causa de que no sea posible la glicosilación de los mismos, ya que los valores de las constantes de inhibición son muy elevados y en ambos casos se observó la síntesis del trisacárido antes mencionado. El desarrollo de esta tesis generó un mayor conocimiento de los sistemas de transglicosilación estudiados, lo que constituye un insumo para el diseño de próximas síntesis de glicósidos catalizadas por estas enzimas.

Las galectinas, son proteínas que poseen alta afinidad por β-galactósidos. Dichas proteínas poseen un dominio conservado denominado Dominio de Reconocimiento a Carbohidrato (DRC), donde se da la interacción con sus ligandos. En particular las galectinas 1 y 3 han sido ampliamente estudiadas, ya que se encuentran vinculadas a distintos procesos biológicos, entre otros adhesión celular, extravasación celular y metástasis, favoreciendo el desarrollo tumoral. Por esta razón se han convertido en un blanco prometedor para la terapia contra el cáncer ya que se podría inhibir su capacidad pro-tumorogénica bloqueando el sitio de unión a carbohidratos con glicósidos que compitan con sus ligandos naturales. En este marco el objetivo general de esta tesis fue establecer una estrategia que permitiera el diseño racional de inhibidores de galectinas con potencial actividad antitumoral. Para ello se propuso el desarrollo in silico de modelos computacionales, capaces de predecir la naturaleza de la interacción entre los ligandos seleccionados y las galectinas. Por otro lado se realizó la síntesis enzimática de dichos compuestos y la evaluación de su actividad biológica mediante estudios in vitro. Se sintetizaron enzimáticamente cinco galactósidos: β-D-Galp-(16)-β-D-Galp-(14)-D-Glcp (2), la mezcla β-D-Galp-(16)-β-D-Glcp-(14)-D-Glcp (3) y β-D-Galp-(13)-β-D-Glcp-(14)-D-Glcp (4), β-D-Galp-(16)-β-D-GlcNAcp-(14)-D-GlcNAcp (6) y el galactósido de carácter aromático bencil-β-D-galactopiranósido (5), aplicando el sistema de transglicosilación catalizado por la enzima β-galactosidasa de Aspergillus oryzae. Posteriormente se estudió la afinidad y especificidad de unión in silico de las galectinas 1 y 3 y los β-galacto-compuestos sintetizados junto con otros disacáridos de origen comercial. Para ello se aplicaron distintas simulaciones de Dinámica Molecular, en combinación con experimentos de Docking Molecular, en la búsqueda de generar modelos computacionales capaces de reproducir y predecir las interacciones entre las galectinas y sus ligandos. A su vez, se estimaron las energías libres de unión (ΔG) de cada complejo galectina-ligando obtenido. Con el objetivo de validar los modelos teóricos obtenidos, se estudió la interacción galectina-ligando mediante dos ensayos biológicos in vitro: ensayo en fase sólida e ITC. Los resultados obtenidos, mostraron una óptima correlación entre los resultados experimentales y teóricos, validando los modelos generados previamente. A pesar de la complejidad intrínseca del modelado de carbohidratos y lo expuesto al disolvente del DRC, en esta tesis se obtuvieron modelos teóricos preliminares, capaces de sortear dichas dificultades. Si bien los ligandos sintetizados no mostraron afinidades superiores a las de los ligandos de referencia (lactosa y N-acetil-lactosamina), los resultados obtenidos, constituyen un estudio detallado de la naturaleza de dichas interacciones. De hecho se pudieron establecer diversos aspectos de la naturaleza química del ligando que influyen en la interacción con la galectina. Por un lado la naturaleza del enlace entre la galactosa y el segundo azúcar es fundamental, siendo los enlaces β-(14) y β-(13) los que presentan mayor afinidad. Otro aspecto a tener en cuenta es la identidad de dicho azúcar, determinándose que la presencia del grupo N-acetilo en el carbono 2 del residuo glucopiranósico, mejora la interacción con el subsitio D de las galectinas, tanto 1 como 3. Finalmente, la adición de un tercer azúcar no resulta beneficioso respecto a la energía de unión. Esto no quiere decir que ligandos más grandes no puedan ser más afines por la galectina, sino que en principio estructuras de tipo trisacáridos no sería suficiente y no evidencian diferencias respecto a los disacáridos. Finalmente los modelos teóricos generados en esta tesis representan un importante insumo para el diseño y síntesis de inhibidores de galectinas más potentes.

Diverse parasites and pathogens affect productivity and survival of honey bees. Plant secondary metabolites are potential alternative treatments, however, their effect has been little studied on microsporidian diseases. Furthermore, there is poor information about the toxicity resulting from prolonged oral administration of these substances to bees. In this research, we evaluated in vivo effects of different essential oils and main components (MCs) on bee survival and Nosema ceranae development under an ad libitum non-choice regimen. Substances administered on sucrose syrup to newly emerged bees at different concentrations were avidly consumed and caused different survival performances. Nevertheless, sublethal doses of substances did not control the parasite. Diversos parásitos y patógenos afectan la productividad y la supervivencia de las abejas melíferas. Los metabolitos secundarios de las plantas son tratamientos alternativos potenciales, sin embargo, su efecto ha sido poco estudiado sobre las enfermedades microsporidianas. Además, hay poca información sobre la toxicidad resultante de la administración oral prolongada de estas sustancias a las abejas. En esta investigación, evaluamos los efectos in vivo de diferentes aceites esenciales y componentes principales (CPs) sobre la supervivencia de abejas y el desarrollo de Nosema ceranae bajo un régimen de ad libitum sin elección. Las sustancias administradas en el jarabe de sacarosa a las abejas recién emergidas a diferentes concentraciones se consumieron con avidez y causaron diferentes resultados de supervivencia. No obstante, las dosis subletales de sustancias no controlaron el parásito.

A summary of the resource.
This work presents kinetic and thermal data of three different biomass wastes with high availability in Uruguay. Samples were analysed by proximate, ultimate and thermogravimetric analyses, transient plane source, and a cone calorimeter. DTG results at different heating rates allowed us to identify different stages during the thermogravimetric analysis. Kinetic modelling per stages was realized for each stage observed during the heating of the biomass samples. The kinetic was modelled minimizing the square difference between the experimental data and theoretical values obtained with the model. Activation energy values were obtained for each stage, with values in the range 164-217 kJ mol-1 for the decomposition of cellulose and hemicellulose, and between 190-507 kJ mol-1 for the decomposition of lignin

Abstract
A summary of the resource.
The first total synthesis of cyclotetrapeptide versicotide D has been achieved in 21% overall yield using solid phase peptide synthesis and solution cyclization. In addition, in the search for candidates of antimalarial new drugs, one cyclic tetrapeptide analog which differs in the sequence, and four cyclic pentapeptide containing N-methyl amino acids, were prepared. The obtained compounds were evaluated against P. falciparum 3D7. Versicotide D showed low micromolar antiplasmodial activity.

Abstract
A summary of the resource.
The syntheses of versicotides A–C, natural products containing anthranilic acid and NMe-Ala, were achieved by solid phase peptide synthesis on 2-chlorotrityl resin followed by solution phase macrocyclization. Using an oxyma additive, the difficult coupling reactions to the deactivated aromatic amine of o-aminobenzoic acid, were performed in high yield, avoiding anthranilic rearrangements or side reactions.

The aim of the present work was to evaluate the possibility of a fast, simple, and greener sample preparation method using electrospun fibers as SPE sorbents and its application for doping control analysis. Electrospun polymer fibers of polystyrene with smooth morphology (1.3 μm average diameter) were used as sorbent materials for the fabrication of miniaturized solid phase extraction devices. A microcolumn SPE device (10 mg of electrospun fibers packed in a 200 μL pipette tip) and a SPE disk device (10 mg of electrospun fibers packed in a 500 μL polypropylene barrel) were used. Their performance was evaluated using dexamethasone and betamethasone as model analytes for the analysis of drug residues in biological samples. A qualitative confirmatory method for the detection of these glucocorticosteroids by HPLC-MS/MS in human urine was developed using electrospun microfibers for sample clean-up and pre-concentration. The limit of detection for both analytes was 10 ng mL−1 in urine matrix, and the resulting analytical method is at the same time fit for purpose (complying with strict requirements of doping control international organizations), simple (using tip-based microcolumns) and has a greener footprint (using very small amounts of organic solvents).

El proyecto de Maestría propone desarrollar y validar estrategias analíticas para el análisis confirmatorio de medicamentos veterinarios en muestras biológicas de origen equino para su uso en control de doping. Estas estrategias incluyen el desarrollo de nuevos métodos de extracción para el tratamiento de muestra en matrices biológicas de acuerdo a los principios de química verde y el estudio y optimización de patrones de fragmentación en experimentos de espectrometría de masa tándem para la detección de los analitos de interés. Los métodos desarrollados deberán cumplir con los requerimientos para la identificación de sustancias prohibidas en matrices biológicas de acuerdo a normativas aceptadas en control de doping. El trabajo fue desarrollado en el Instituto Polo Tecnológico de Pando de la Facultad de Química, cuya misión es la realización de proyectos I+D+i con distintos sectores de la industria. De manera de adaptarse a los requerimientos del usuario, los métodos desarrollados deben cumplir con el propósito de ser adecuados al uso (fit for purpose), incorporando adicionalmente los factores de la reducción del impacto ambiental sobre la salud humana y ser económicamente viables. En la preparación de muestra, se trabajó en la reducción en los tiempos de procesamiento, disminución de los volúmenes de solventes orgánicos y de muestra utilizados y la utilización de solventes ambientalmente amigables. En cuanto al análisis cromatográfico con detección por espectrometría de masa, se buscó la utilización de columnas más cortas, con menor tamaño de partícula o diámetro interno. Cada una de las moléculas seleccionadas para el trabajo representa una clase de compuestos buscados en control de doping. Los métodos desarrollados, pueden además tener aplicaciones en estudios farmacocinéticos para el monitoreo de drogas veterinarias y en el análisis de residuos para asegurar la calidad de productos alimenticios de origen animal.

This study reports a new octadentate tricarboxylate appended ligand (H4dphta) with enhanced relaxivity and high stability towards Gd(III) complex as an MRI probe. The designed ligand was synthesized in multiple steps. Both, the ligand and its complex were characterized by spectroscopic techniques. Structural analysis of Gd(III) complex by single crystal XRD confirms its mono-hydrated form adopting distorted capped square antiprism geometry. Protonation constants of the ligand and the thermodynamic stability constant of the [Gd(Hdphta)] and [Gd(H2dphta)]+ species have been found as log K110 = 18.6(1) and log K111 = 24.8(1) at 25.0 ◦C, and I = 0.15 mol⋅L

A preliminary bioavailability study of furosemide vs.a furosemide prodrug: acethyloxymethyl-4-chloro-N-furfuryl-5-sulfamoyl anthranilate was carried out using a randomized two-way crossover and balanced design. Furosemide levels in urine were analyzed by High Performance Liquid Chromatography (HPLC). The urine was collected up to 24 hours. A summary of the resource.
According to the hydrolysis performance "in vitro" and lipophylicity |b two Furosemide (F) prodrugs were chosen from a series of acyloxymethylesters of F synthesized previously: PI (acetyloxymethyl-4-chloro-N-furfuryl-5-sulfamoylanthranilate) and P4 (pivaloyloxymethyl-4-chloro-N-furfuryl-5-sulfamoylanthranilate). The bioavailability studies were assayed over two groups of eight male Wistar rats as a randomized two-way crossover and balanced design: group 1) a solution of PI in propylenglycol/ethyl acetate vs an aqueous solution of F |b and group 2) PI vs.P4 solutions in oleic acid (PI,P4).These assays showed a better absorption performance of PI and P4 than F |b while the two prodrugs showed a similar bioavailability. The oleic acid seems to be responsible for the delay in the recovery of 50 of the total amount of F excreted in urine (T50). When the monitoring is done in plasma after the administration of PI |b PI was not detected as circulating prodrug. The analytical determinations of F in urine and plasma were done by high performance liquid chromatography (HPLC). From the urinary excretion data |b a slope that indicates a slow elimination was found with a half-life of 12 hours approximately.
