We studied the accumulation and depuration of microcystin-LR (MCLR) in the hepatopancreas of the crab Neohelice granulata fed twice weekly with either non toxic or MCLR-producing Microcystis aeruginosa (strain NPDC1 or NPJB, respectively) during seven weeks. We also analyzed MCLR effects on the oxidative stress- and detoxification-related variables, superoxide dismutase and glutathione-S-transferase activities, and the levels of reduced glutathione and lipid peroxidation (as thiobarbituric acid reactive substances, TBARS). Hepatopancreas MCLR content slightly increased during the first three weeks, up to 8.81±1.84 ng g−1 wet tissue mass (WTM) and then started to decrease to a minimum of 1.57±0.74 ng g−1 WTM at the seventh week (p<0.05 with respect to that in the first week). TBARS levels were about 55% higher in treated than in control N. granulata (p<0.001 and p<0.05) during the first three weeks of the experimental period. GSH content became 50% lower than in control individuals (p<0.01) during weeks 6 and 7. SOD activity was increased by about 2-fold (p<0.05 or p<0.001) from week 3 to 7 in treated crabs with respect to control ones, while GST activity was about 70% higher in treated than in control crabs from week 4 to week 7 (p<0.05). Our data suggest that in the hepatopancreas of N. granulata MCLR accumulation and oxidative damage are limited and reversed by detoxification-excretion and antioxidant mechanisms. Antioxidant systems, Fe, Cu, Zn, Mn and Ni levels and proteomics in soybean seeds of different performance in field were assessed. These results were correlated with the in vitro tests required by regulatory organisms, and the actual behavior in field. Basal superoxide content, superoxide dismutase (SOD) activity (20-80 U mg-1 protein) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activity (4.6-7.0 mg antioxidant per g dry seed) permitted to study the oxidative stress of seeds. Essential metal levels were in accordance to previous reports, with the exception of Zn that was higher in some batches of good in vitro quality, but poor performance in field. Proteomic profile showed no differences between batches. Two of the studied parameters (basal superoxide content and Zn level) contribute to a better assessment of the health state of the seeds and predict a possible poor performance in field. Their activity is evaluated on Trypanosoma cruzi. The lipophilicity, as structural descriptor related to bioactivity, of the whole [VIVO(L-2H)(NN)] series is determined. Furthermore, the antiproliferative effect of those new compounds showing activity against T. cruzi is evaluated on the genetically related parasite T. bruceiwith the aimto develop broad spectrumagents. The new[VIVO(L-2H)(dppz)] complexes are about ten to fifteen times more toxic to T. cruzi than the bipy analogues and show quite good in vitro activity on T. brucei brucei. They are shown to interact with DNA, suggesting that this biomolecule may be the parasite target. The stability of the VIVO-complexes in solution is accessed by several techniques. Globally the data suggest that the relevant species for biological activity are the [VIVO(L-2H)(NN)] compounds, their order of activity being dependent on the NN nature, but not much on the substitution on the salicylaldehyde semicarbazone moiety. A parabolic relationship between biological response and lipophilicity (determined as RM = log [(1 / Rf) − 1] by a TLC method) is obtained. From this correlation an optimum RM value, close to 1.44, was found, which may be used as design guide for future development of antitrypanosomal compounds. A novel SNS tridentate ligand, the N,N-bis(2-mercaptoethyl)-N',N'-diisopropylethylenediamine was synthesized and the corresponding oxorhenium and oxotechnetium mixed ligand complexes using 3 monodentate aromatic thiols as coligands were prepared and characterized. Rhenium complexes were obtained as crystals with high yield by substitution of ligand and coligand on the trichlorobis(triphenylphosphine)- rhenium(V) oxide, using a molar ratio ligand:coligand:precursor of 1. Characterization was performed by HPLC analysis, UV-vis and IR spectra, elemental analysis and X-ray diffraction. Results were consistent with the ReOLC structure. The complexes adopted a distorted trigonal bipiramidal geometry and were neutral due to ionization of all sulphur atoms upon complexation. Consequently 99mTc complexes were also prepared and evaluated in mice as potential brain imaging agents. Labelling was performed by substitution using 99mTc-glucohepionate as precursor. The labelling yield was over 85% and the radiochemical purity of the complexes over 90%. Biodistribution in mice demonstrated high uptake and retention in brain. However comparison with previously reported mixed ligand complexes showed that the incorporation of the diisopropylaminoethyl group in the SNS/S backbone is not enhancing the biological characteristics of this type of complexes. Marketing of unhealthy products has been identified as one of the main characteristics of the food environment that negatively affects children's eating patterns. Restrictions on advertising of unhealthy foods to children have already been imposed in different countries. However, marketing strategies are not limited to broadcast and digital advertising, but also include package design. In this context, the current study aimed to describe the food products targeted at children and sold in supermarkets in Montevideo, Uruguay, in terms of package design and nutrient profile. Two supermarkets in Montevideo were selected for data collection. In each supermarket, all products targeted at children were identified. Products were analyzed in terms of package design and nutritional profile, considering the Pan American Health Organization Nutrient Profile Model. A total of 180 unique products were identified, which included a wide range of product categories. The great majority of the products corresponded to ultra-processed products with excessive amounts of sodium, free sugars, total fat, saturated fat, and/or trans fat, which are not recommended for frequent consumption. Several marketing strategies were identified in the design of packages to attract children's attention and drive their preferences. The most common strategies were the inclusion of cartoon characters, bright colors, childish lettering, and a wide range of claims related to health and nutrition, as well as the products' sensory and hedonic characteristics. The study's findings provide additional evidence on the need to regulate packaging of products targeted at children. A negatively charged organic molecule acts as a tridentate ligand and binds to the metal through the pyridine nitrogen, the imine nitrogen and the sulfur atom. A chloride ion occupies the fourth coordination site. The planar complexes stack nearly parallel to one another in the lattice conforming a layered crystal structure. The cytotoxic activity of the thiosemicarbazones and their metal complexes was tested against the MCF-7, TK-10 and UACC-62 human tumor cell lines. The ligands exhibit lower values of GI50 and LC50 than the complexes, H2Bz4Ph being the most active with GI50 < 0.003 lM; LC50 = 13.4 lM; GI50 = 9.3 lM, LC50 = 12.9 lM; GI50 < 0.003, LC50 = 13.8 lM in the MCF-7, TK-10 and UACC-62 cell lines, respectively. Among the complexes, [Pd(2Bz4Ph)Cl] (3) exhibited the lowest values of GI50 in the three studied cell lines. Cystic echinococcosis is the infection by the larvae of cestode parasites belonging to the Echinococcus granulosus sensu lato species complex. Local host responses are strikingly subdued in relation to the size and persistence of these larvae, which develop within mammalian organs as ‘hydatid cysts’ measuring up to tens of cm in diameter. In a context in which helminth-derived immune-suppressive, as well as Th2-inducing, molecules garner much interest, knowledge on the interactions between E. granulosus molecules and the immune system lags behind. Here, we discuss what is known and what are the open questions on E. granulosus molecules and structures interacting with the innate and adaptive immune systems, potentially or in demonstrated form. We attempt a global biological approach on molecules that have been given consideration primarily as protective (Eg95) or diagnostic antigens (antigen B, antigen 5). We integrate glycobiological information, which traverses the discussions on antigen 5, the mucin-based protective laminated layer and immunologically active preparations from protoscoleces. We also highlight some less well-known molecules that appear as promising candidates to possess immune-regulatory activities. Finally, we point out gaps in the molecular-level knowledge of this infectious agent that hinder our understanding of its immunology. Objective: To explore the perception and experiences of parents of children and adolescents during the first year of the mandatory implementation of nutritional warning labels in Uruguay. Design: Qualitative research based on semistructured interviews. Setting: Montevideo, Uruguay. Participants: Twenty-eight parents of children and adolescents. Phenomenon of interest: Perception of and experiences with warning labels, a front-of-pack nutrition labeling scheme that has become increasingly popular in Latin America. Analysis: Content analysis of the transcripts based on a deductive-inductive approach. Results: Interview transcripts evidenced high awareness, acceptance, and understanding of warning labels among parents of children and adolescents. Most participants reported considering warnings for decision making and changing their purchase decisions because of their implementation, particularly when choosing foods for their children. They reported their children were aware of and understood warnings but did not tend to take them into account when choosing foods. Conclusions and Implications: Results from the present work contribute to the growing body of litera ture showing the effectiveness of warning labels. Lack of interest or perceived benefits and structural bar riers emerged as key motives for not using the warnings when making purchasing decisions, suggesting the need to develop additional strategies to increase policy effectiveness.
