Single-domain antibodies (sdAbs) found in camelids lack a light chain, and their antigen-binding site sits completely in the heavy-chain variable domain (VHH). Their simplicity, thermostability, and ease in expression have made VHHs highly attractive. Although this has been successfully exploited for macromolecular antigens, their application to the detection of small molecules is still limited to a very few reports, mostly describing low-affinity VHHs. Using triclocarban (TCC) as a model hapten, we found that conventional antibodies, IgG1 fraction, reacted with free TCC with a higher relative affinity (IC50 51.0 ng/mL) than did the sdAbs (IgG2 and IgG3, 497 and 370 ng/mL, respectively). A VHH library was prepared, and by elution of phage with limiting concentrations of TCC and competitive selection of binders, we were able to isolate high-affinity clones, KD 0.98�1.37 nM (SPR), which allowed development of a competitive assay for TCC with an IC50 = 3.5 ng/mL (11 nM). This represents a 100-fold improvement with regard to the performance of the sdAb serum fraction, and it is 100-fold better than the IC50 attained with other antihapten VHHs reported thus far. A cDNA clone from a Toxoplasma gondii tachyzoite cDNA library encoding a serine proteinase inhibitor (serpin) was isolated. The 1376 bp cDNA sequence encodes a 294 amino acid protein with a putative signal peptide of 23 amino acids resulting in a mature protein with a predicted mass of 30,190 Da and a pI of 4.86. This protein has internal sequence similarity of residues 30–66, 114–150, 181–217 and 247–283 indicating a four-domain structure. The four domains exhibit high identity to serine proteinase inhibitors belonging to the non-classical Kazal-type family. The gene is single copy in the tachyzoite haploid genome of RH strain and was amplified by polymerase chain reaction (PCR). Several introns were identified. The sequence encoding the mature protein was amplified by PCR, cloned into the pQE30 vector and expressed in Escherichia coli. Specific antiserum generated against the recombinant protein was used in immunoblot assay and two bands of 38 and 42 kDa were detected in a whole parasite homogenate. The recombinant protein showed trypsin-inhibitory activity, one of the two potential specificities. We discuss the possible roles that T. gondii serpin(s) may play in the survival of the tachyzoites in the host. El diseño y construcción de estructuras supramoleculares polinucleares de iones lantánidos es un campo de investigación de gran interés. Ellas presentan interesantes arquitecturas y propiedades, ya sea magnéticas, ópticas, eléctricas, etc. Los lantánidos (Ln) en su estado de oxidación +3, son iones ideales para el diseño de estas estructuras poliméricas, debido a sus altos índices de coordinación que permiten construir fascinantes estructuras térmicamente estables. El futuro de este tipo de complejos depende de continuar la búsqueda de nuevos compuestos y del cuidadoso diseño y control de la arquitectura final generada. La selección del ligando utilizado es crucial para el diseño de estos compuestos. Un tipo de ligandos de amplio estudio por formar complejos de gran estabilidad y ofrecer diversos modos de coordinación son los ligandos policarboxílicos. En particular el ácido iminodiacético (H2ida) ha mostrado una sorprendente versatilidad gracias a su gran flexibilidad y diversos modos de coordinación. Varias estructuras polinucleares se han reportado con este ligando, siendo relevante la serie [Ln(Hida)3]·xH2O. La particularidad de estos compuestos es la formación de estructuras tridimensionales que contienen canales de unos 11 Å de diámetro. Es de destacar que en estas estructuras se observa la capacidad de este ligando de pasar de ser un ligando tridentado ONO, a un ligando ditópico, que puede coordinar centros metálicos alejados. La protonación del átomo de N es la responsable de estos cambios. En esta misma línea, otras estructuras porosas han sido reportadas para complejos heteropolinucleares conteniendo iones lantánidos, metales de transición 3d y el ligando ida. Un ejemplo de estos son los complejos isoestructurales del tipo [Ln2Cu3(ida)6]·xH2O. Con estos antecedentes, el objetivo de este trabajo es sintetizar nuevas series de complejos polinucleares conteniendo iones lantánidos y ligandos aminodicarboxílicos N-sustituidos derivados del iminodiacetato y caracterizarlos del punto de vista estructural y de sus propiedades. Los ligandos aquí empleados son el ácido N-hidroxietiliminodiacético (H2hida), el ácido N-benciliminodiacético (H2bzlida), el ácido N-metiliminodiacético (H2mida) y el ácido Nmetiliminodipropiónico (H2midp). Con el fin de determinar las mejores condiciones de síntesis se realizaron estudios potenciométricos en solución acuosa. A partir de estos fue posible determinar las especies principales en solución y el rango de pH en el cual pueden ser aislados al estado sólido. Luego se pasó a la etapa de síntesis, donde se obtuvieron nuevos complejos polinucleares con todos los ligandos: [Ln(bzlida)(Hbzlida)]·H2O (Ln = La, Nd), [Ln2(bzlida)3]·3H2O (L n= Nd, Sm, Eu, Gd), [Ho(hida)(H2O)2](CF3SO3)·xH2O, [Sm(Hmida)2(H2O)2]·ClO4·H2O·1/6NaClO4 y [Ln2(Hmidp)3(H2O)3](ClO4)3·3H2O (Ln = Nd, Sm, Eu, Gd, Tm, Dy, Ho). Los complejos fueron caracterizados completamente incluyendo su elucidación estructural mediante difracción de rayos X. Finalmente se evaluaron las propiedades ópticas y magnéticas de una selección de los compuestos obtenidos. Esto permitió relacionar las estructuras obtenidas con las propiedades al estado sólido. El estudio de sus propiedades ópticas permitió determinar que es posible una Resumen 2 transferencia de energía desde los ligandos a los centros metálicos, aunque la eficiencia de la transferencia es baja, siendo los compuestos de Eu los que arrojan los mejores resultados. Al evaluar las propiedades magnéticas se pudo racionalizar el origen del acoplamiento antiferromagnético en el compuesto [Gd2(bzlida)3]·3H2O. Addition of tags [such as His (histidine) tags] is extremely helpful for the affinity purification of recombinant proteins. In several cases, these tags must be removed before performing functional and structural studies. The enzyme most frequently used to cleave tags of recombinant proteins is the TEV-protease (tobacco-etch-virus NIa protease). The continuous production of this enzyme in soluble form is quite an expensive process and not easily accessible to many laboratories. Thus an interesting alternative is the use of TEV-protease in an immobilized form, which may be reutilized several times. The main objective of the present study was to obtain a TEV-protease in an immobilized form, by covalent immobilization on to solid supports through selective use of different amino acid residues, lysine or cysteine. High protein immobilization yields (75–97 %) were obtained with both strategies. The TEV-protease immobilized through its exposed cysteine thiol groups maintained its ability for cleaving a 20 kDa substrate. While the activity of the immobilized TEV-protease maintained only 30% of the activity of the enzyme in soluble form, its stability at 4 ◦C was improved three times. Moreover, this enzyme could be reutilized in at least five cycles of cleavage without loss of performance. Purpose: The current diagnostic methods for leptospirosis diagnosis are technically complex and expensive, with limited applicability to specialized laboratories. Furthermore, they lack diagnostic accuracy in the acute stage of the disease, which coincides with a period when antibiotics are highly effective. New simple and accurate tests are mandatory to decentralize and improve diagnosis. Here, we introduced a new lateral flow immunoassay (Lepto-LF) for human leptospirosis. Methods: We conducted a double-blinded assay using 104 serum samples from patients with confirmed or discarded diagnosis for leptospirosis. The diagnostic performance of Lepto-LF was estimated across different ranges of days from onset of symptoms (dpo), considering the diagnostic algorithm as reference standard. Additionally, it was compared with the screening methods enzyme-linked immunosorbent assay (IgM-ELISA) and the slide agglutination test using temperature-resistant antigen (SATR). Results: Lepto-LF exhibited perfect diagnostic performance with a Youden´s index J = 1 from 6 dpo in the acute phase. IgM-ELISA gave slightly lower accuracy with J = 0.91 and 95.5% of both sensitivity and specificity; while SATR showed a markedly inferior yield (J = 0.41, sensitivity = 95.5%, specificity = 45.5%). The performances remained consistent in the convalescence phase of the disease (> 10 dpo). Conclusion Lepto-LF was found to be a reliable test for simple, rapid and early diagnosis of leptospirosis, resulting a promising tool for decentralizing leptospirosis diagnosis and enabling timely treatment of patients. In addition, Lepto-LF may be employed as confirmatory test, especially in remote areas and vulnerable contexts where the standard MAT is not available. In freshwater, harmful cyanobacterial blooms threaten to increase with global climate change and eutrophication of surface waters. In addition to the burden and necessity of removal of algal material during water treatment processes, bloom-forming cyanobacteria can produce a class of remarkably stable toxins, microcystins, difficult to remove from drinking water sources. A number of animal intoxications over the past 20 years have served as sentinels for widespread risk presented by microcystins. Cyanobacterial blooms have the potential to threaten severely both public health and the regional economy of affected communities, particularly those with limited infrastructure or resources. Our main objectives were to assess whether existing water treatment infrastructure provides sufficient protection against microcystin exposure, identify available options feasible to implement in resource-limited communities in bloom scenarios and to identify strategies for improved solutions. Finally, interventions at the watershed level aimed at bloom prevention and risk reduction for entry into potable water sources were outlined. We evaluated primary studies, reviews and reports for treatment options for microcystins in surface waters, potable water sources and treatment plants. Because of the difficulty of removal of microcystins, prevention is ideal; once in the public water supply, the coarse removal of cyanobacterial cells combined with secondary carbon filtration of dissolved toxins currently provides the greatest potential for protection of public health. Options for point of use filtration must be optimized to provide affordable and adequate protection for affected communities. Copyright © 2013 John Wiley & Sons, Ltd. An increasing share of fine wines is marketed through retail stores. In this context, packaging plays a major role in attracting consumers’ attention and influencing their purchase decisions. The aim of the present work was to identify a specific bottle for Tannat red wine, the typical Vitis vinifera grape of Uruguay, by studying the effect of bottle shape and colour on consumers’ sensory expectations and willingness to purchase. A consumer study was carried out with 64 fine wine consumers, who were first introduced a list of bottle colours and shapes and were asked to answer a check-all-that-apply question. Then, participants were asked to complete a conjoint analysis task about Tannat wine bottles. Results from the check-all-that-apply question showed that consumers associated the shape and colour of bottles with specific types and styles of wine. Conjoint analysis suggested that Bordeaux dark green or dark gray bottles might be the most appropriate for Tannat wine. Interestingly, results shown here may indicate the existence of risks if deep innovation is made in bottle shape designs, since consumers clearly preferred Bordeaux bottles for red wines. BACKGROUND: When feeding preterm infants, donor milk is preferred if the mother's own milk is unavailable. Pasteurization may have detrimental effects on bioactivity, but more information is needed about its effects on the immunological compounds. Research aim: This work has two main aims: evaluate the antibody profile of colostrum and study the quantitative variations in the antibodies' level and specific reactivity after undergoing Holder pasteurization. The authors focused on immunoregulatory components of colostrum (antidietary antibodies and TGF-β2) in the neonatal gut. METHODS: This is a descriptive cross-sectional study of a convenience sample of 67 donated colostrum samples at different days after delivery, both raw and pasteurized. Antibody profiles were analyzed at different times during breastfeeding, and total and specific antibodies (IgM, IgA, and IgG subclasses) were compared with tetanus toxoid and ovalbumin using enzyme-linked immunosorbent assay. The processing effect on total and specific antibodies, as well as TGF-β2, was evaluated by paired analyses. RESULTS: No variations in immunological compounds were observed throughout the colostrum stage. The TGF-β2, antibodies' concentrations, and antibodies' specific reactivity after pasteurization did not vary significantly as days of lactation varied. Changes in antibody levels were dependent on isotype and IgG subclass, and IgG4 showed remarkable resistance to heating. Moreover, the effect of the pasteurization on specific reactivity was antigen dependent. CONCLUSION: The supply of relevant immunological components is stable throughout the colostrum stage. The effects of pasteurization on antibodies depend on isotype, subclass, and specificity. This information is relevant to improving the immunological quality of colostrum, especially for preterm newborns. Kefir is a fermented probiotic drink obtained by placing kefir granules in a suitable substrate. The kefir granules are a consortium of bacteria and yeasts embedded in a exopolysaccharide matrix. The aim of this research was the isolation and identification of yeasts from kefir of different origin, the evaluation of their antifungal capacity against Aspergillus spp. and the characterization of virulence related traits. Using RFLP of ITS1/ITS4 region, D1/D2 region sequencing and RAPD techniques, 20 kefir isolates were identified as Geotrichum candidum, Pichia kudriavzevii, P. membranifaciens, Saccharomyces cerevisiae and Candida ethanolica. Their antifungal capacity was evaluated by their conidia germination reduction, which allowed the selection of eight isolates with high to moderate conidia germination reduction against A. flavus and A. parasiticus. Furthermore, these selected isolates showed growth inhibition on contact in the dual culture assay for both Aspergillus species and 3 of them ‒belonging to S. cerevisiae and P. kudriavzevii species‒ generated volatile organic compounds which significantly affected the growth of both fungi. For the evaluation of virulence related traits, growth at high temperatures, enzymatic activities and the adhesion to Caco-2 cells were analyzed. The isolates did not present more than one positive virulence-related trait simultaneously. In particular, it is important to highlight that the adhesion capacity to the model of intestinal barrier was extremely low for all of them. According to the results obtained, further studies would be of interest for the possible use of these promising yeasts as biocontrol agents against fungi in food.
