Research on behavioral and social science has demonstrated that transitions throughout a person’s life course, as the life event of becoming a parent, represent an opportunity for dietary changes. However, research in this area has been mostly restricted to developed European and North American countries and has shown ambiguous results. The present work aimed to gain an in-depth understanding on the changes in eating habits caused by the transition to parenthood and to explore factors influencing those changes in Uruguay, a Latin American country. Forty-two in-depth interviews with Uruguayan parents from diverse social-economic backgrounds were conducted. The narrations by the interviewees evidenced mostly positive changes during three stages in the transition: pregnancy, the first months with the baby, and the beginning of complementary feeding. Most informants perceived changes mainly in terms of increased consumption of fruits, vegetables, and pulses; increased consumption of homemade meals; and decreased consumption of ultra-processed foods, fast foods, and fried foods. Reasons for these changes included adaptation of meals to the child’s needs and schedule, feeling of responsibility for the child’s health, and willingness to be a good role model. Factors influencing changes in eating habits were identified in some levels of the socioecological model and included socioeconomic status; price of fruits, vegetables, and fish; low accessibility of good quality fish; perceived time pressure; father’s low interest on healthy eating; social support; and access to nutrition information. Findings suggest that parenthood represents a window of opportunity for favorable changes in eating habits. However, barriers constraining those changes should be addressed by stakeholders. In this context, the aims of the present work were: i) to explore the automatic mental associations raised by packages of ultra-processed products among parents of children under 12 months of age, ii) to estimate the valence of the affective reactions elicited by ultra-processed products, and iii) to explore the influence of socio-demographic variables and feeding styles on the valence of parents’ associations with ultra-processed products. A total of 419 parents of children under 12 months of age were recruited at 20 health facilities that offer well-child services, selected using probability proportional to size sampling based on the number of births at each institution. Participants were presented with the labels of six ultra-processed products (corn snack, frankfurters, milk dessert, orange juice, wafer cookies, yogurt) and were asked to indicate the first words that came to their mind. The products raised mixed mental associations, which were mainly related to pleasure and health. Responses from the majority of participants (61–82%) evidenced negative associations with corn snacks, frankfurters and wafer cookies, whereas responses from 35 to 45% of the participants suggested a positive affective reaction towards orange juice, milk dessert and yogurt. Likelihood of having a positive affective reaction towards the evaluated ultra-processed products was higher for parents of 1 to 4 months infants, as well as those from low educational level and low socio-economic status. In addition, beliefs about three feeding practices (restriction on diet quality, indulgence in screen use and pressure for finishing) significantly influenced the valence of the associations. Results from the present work suggest the need to implement regulatory measures to reduce positive associations with ultra-processed products among parents and contribute to reduce their consumption among infants. Sodium reduction strategies that fully or partially compensate for the effect of reducing sodium on the sensoy profile of foods have been proposed as a cost‐effective alternative to decrease dietary sodium intake at the population level. The present work aimed at exploring the effect of partially replacing NaCl with KCl on the sensory characteristics and hedonic perception of toast bread. Equivalent salty concentration of NaCl : KCl blends were estimated relative to different NaCl concentrations, using paired comparisons with a panel of trained assessors. The influence of partially replacing NaCl by KCl on the dynamic sensory profile of toast breads was assessed using the Temporal Check‐all‐that‐apply approach with a trained panel. Finally, a consumer study was carried out to evaluate consumers' sensory and hedonic perception of toast breads produced by partially replacing 30% NaCl with KCl. Results from the present work suggest that KCl can be deemed effective at compensating for saltiness intensity in sodium reduced toast breads. Partial replacement of 30% NaCl with KCl did not significantly affect the sensory characteristics of toast breads. However, NaCl replacement equal or higher than 40% caused an increase in bitterness, off‐flavor, and metallic flavor. Heterogeneity in consumer hedonic reaction toward partial replacement of NaCl with KCl was observed, but mainly associated to differences in texture perception. Risk situations related to brownfield sites, contaminated lands and groups exposed to toxic substances are common in South America. In some of these situations, there is a lack of institutional response to the risks and consequences coupled with a failure to communicate the risks to those affected. Drawing on empirical research in 4 South American communities exposed to lead contamination, we investigated which strategies of risk communication (RC) have been used by researchers to communicate the risks of contamination, and the possible impacts of these strategies. Considering that risk situations of contaminated lands are characterized by uncertainty/strain/controversy, and that their residents are in a collective stress situation because of the physical conditions in their neighborhood and possible impacts on their health, in our study we argue for the need of participative RC in medical geology studies. The participative RC is based on cultural/social elements, trust, open dialogue and collective learning process to implement policies for mitigating risks and for a sustainable future. However, the analysis of these cases suggests that the efforts of RC have focused only on information transmission and public persuasion. We noticed that RC did not promote an open dialogue, or any discussion about the life styles of locals, which gave the impression that research results have not been properly released, and therefore, the general understanding (by assorted actors) is that environmental/health research is not being undertaken for the public good. The larval stage of the cestode Echinococcus granulosus causes cystic echinococcosis in humans and livestock. This larva is protected by the millimeter-thick, mucin-based laminated layer (LL), from which materials have to be shed to allow parasite growth. We previously reported that dendritic cells (DCs) respond to microscopic pieces of the mucin gel of the LL (pLL) with unconventional maturation phenotypes, in the absence or presence of Toll-like receptor (TLR) agonists, including lipopolysaccharide (LPS). We also reported that the presence of pLL inhibited the activating phosphorylation of the phosphatidylinositol 3-kinase (PI3K) effector Akt induced by granulocyte-macrophage colony-stimulating factor or interleukin-4. We now show that the inhibitory effect of pLL extends to LPS as a PI3K activator, and results in diminished phosphorylation of GSK3 downstream from Akt. Functionally, the inhibition of Akt and GSK3 phosphorylation are linked to the blunted upregulation of CD40, a major feature of the unconventional maturation phenotype. Paradoxically, all aspects of unconventional maturation induced by pLL depend on PI3K class I. Additional components of the phagocytic machinery are needed, but phagocytosis of pLL particles is not required. These observations hint at a DC response mechanism related to receptor-independent mechanisms proposed for certain crystalline and synthetic polymer-based particles; this would fit the previously reported lack of detection of molecular-level motifs necessary of the effects of pLL on DCs. Proliferation of macrophages is a hallmark of inflammation in many type 2 settings including helminth infections. The cellular expansion is driven by the type 2 cytokine interleukin-4 (IL-4), as well as by M-CSF, which also controls homeostatic levels of tissue resident macrophages. Cystic echinococcosis, caused by the tissue-dwelling larval stage of the cestode Echinococcus granulosus, is characterised by normally subdued local inflammation. Infiltrating host cells make contact only with the acellular protective coat of the parasite, called laminated layer, particles of which can be ingested by phagocytic cells. Here we report that a particulate preparation from this layer (pLL) strongly inhibits the proliferation of macrophages in response to IL-4 or M-CSF. In addition, pLL also inhibits IL-4-driven up-regulation of Relm-α, without similarly affecting Chitinase-like 3 (Chil3/Ym1). IL-4-driven cell proliferation and up-regulation of Relm-α are both known to depend on the phosphatidylinositol (PI3K)/Akt pathway, which is dispensable for induction of Chil3/Ym1. Exposure to pLL in vitro inhibited Akt activation in response to proliferative stimuli, providing a potential mechanism for its activities. Our results suggest that the E. granulosus laminated layer exerts some of its anti-inflammatory properties through inhibition of PI3K/Akt activation and consequent limitation of macrophage proliferation. Application of passive modified atmosphere packaging (MAP) for shelf life extension of enzymatically and nonenzymatically peeled orange segments was evaluated. Oranges (Citrus sinensis cv. Valencia) were washed, sanitized and peeled by applying vacuum pulses only (vacuum peeling) and vacuum pulses followed by incubation in enzymatic solution (enzymatic peeling). Segments were packaged in polypropylene film and stored at 7C for 16 days. Package atmosphere composition, weight loss, respiration rate, color, instrumental texture, pH, soluble solids, sensory attributes, and bacterial and fungal growth were evaluated during storage. Microbiological limits (cfu/g > 106) were reached by enzymatically and vacuum-peeled segments on days 12 and 8, respectively. Integrity and characteristic odor had significantly changed by this day. Vacuum-peeled segments presented better texture attributes and integrity (50% higher scores in sensory evaluation). Both types of segments benefited from MAP and are an attractive alternative for commercialization of ready-to-eat oranges. Practical Applications The information presented in this paper can assist fresh-cut producers in developing ready-to-eat oranges. Quality of segments obtained with two peeling methods was evaluated, and potentials and limitations of the segments obtained by each process were pointed out. Passive MAP in polypropylene at a reasonable storage temperature (7C) resulted to be an inexpensive technology that can extend segment shelf life in eventual cold storage of orange segments. Streptococcus pneumoniae (neumococo) continua siendo el principal agente etiológico de la neumonía adquirida en la comunidad en el mundo entero. Las defensas innatas son esenciales para la detección del neumococo y el control de la infección, en especial en etapas tempranas de la neumonía. Sin embargo, la bacteria ha desarrollado mecanismos de evasión que le permiten escapar de la inmunidad natural, resultando muchas veces en la progresión de la enfermedad. La creciente aparición de cepas resistentes a antibióticos, sumado al alto costo de los tratamientos y estrategias de profilaxis hace imperativo el desarrollo de nuevos tratamientos que puedan complementar los ya disponibles. Este trabajo se ha enfocado en el desarrollo de nuevas estrategias de tratamiento contra la neumonía neumocócica basadas en el uso de inmunoestimulantes del sistema innato, administrados por ruta mucosa. Partiendo de la hipótesis de que la activación de las defensas innatas es suficiente para impedir la progresión de la enfermedad, se evaluó la capacidad de diferentes agonistas de receptores innatos de conferir protección frente a la neumonía invasiva en ratones, empleando cepas clínicas de S. pneumoniae. Se demostró que la protección puede ser conferida tanto empleando agonistas de receptores activados normalmente por neumococo, como empleando agonistas de receptores que no son activados por neumococo durante una infección natural. Sin embargo, no todos los receptores activan respuestas protectoras y la estimulación por ciertas vías puede llegar incluso a tener consecuencias negativas. Flagelina de Salmonella enterica serovar Typhimurium, ligando de TLR5/NLRC4-NAIP5, fue uno de los agonistas estudiados. Flagelina confirió mayor protección que cualquiera de los otros agonistas probados, al ser administrado por ruta intranasal por lo que se realizó la caracterización de los mecanismos moleculares y celulares asociados a su capacidad de inducir protección contra la neumonía neumocócica. Los resultados sugieren que el efecto protector requiere del reclutamiento de neutrófilos hacia los espacios alveolares, lo que se logra a través de la secreción de citoquinas de la familia ELR-CXC por parte de las células epiteliales de la mucosa respiratoria y es independiente de células B, T y anticuerpos. La protección mostró ser dependiente de la presencia del motivo de unión a TLR5 en la molécula pero no de la región de interacción con NLRC4-NAIP5, sugiriendo que los efectos observados dependen del reconocimiento extracelular de flagelina a través de TLR5 pero no del inflamasoma. La terapia demostró ser eficaz contra dos serotipos diferentes de neumococo. En segunda instancia, se realizó la puesta a punto de un modelo de inmunoterapia sublingual y se evaluó la capacidad de flagelina de conferir protección frente al desafío intranasal con neumococo administrada por esta ruta, obteniéndose un buen grado de protección. Al igual que en el modelo de instilación intranasal, la protección inducida por administración sublingual de flagelina, requirió reclutamiento de neutrófilos y de la activación de la vía TLR5, pero no de la activación de inflamasoma vía NLRC4-NAIP5. 6 Como último tema se estudió la capacidad adyuvante de flagelina en vacunas de aplicación sublingual. Empleando ovoalbúmina como antígeno modelo se comprobó que la administración de flagelina en conjunto con el antígeno incrementa la respuesta de anticuerpos específicos en suero, observándose un perfil de subclases mixto con predominio del perfil Th2. Empleando un modelo de transferencia adoptiva de células T transgénicas para el receptor T y condicionada para reconocer el fragmento 323-339 de la ovoalbúmina en contexto MHC-II, se estudió la capacidad de flagelina de inducir su proliferación en respuesta a una única inmunización sublingual y se determinó la localización anatómica de la respuesta. Se estudió además la contribución de los receptores TLR5 y NLRC4-NAIP5 confirmándose que la señalización a través de uno solo es suficiente para el efecto adyuvante observado. Por último, se analizó la capacidad de flagelina de promover procesamiento antigénico y expresión de moléculas estimuladoras en células dendríticas derivadas de médula ósea. While it is known that toxic metals contribute individually to child cognitive and behavioral deficits, we still know little about the effects of exposure to multiple metals, particularly when exposures are low. We studied the association between children’s blood lead and hair arsenic, cadmium, and manganese and their performance on the Bayley Scales of Infant Development III. Ninety-two preschool children (age 13–42 months) from Montevideo, Uruguay, provided a hair sample and 78 had a blood lead level (BLL) measurement. Using latent class analysis (LCA), we identified four groups of exposure based on metal concentrations: (1) low metals, (2) low-to-moderate metals, (3) high lead and cadmium, and (4) high metals. Using the four-group exposure variable as the main predictor, and fitting raw scores on the cognitive, receptive vocabulary, and expressive vocabulary scales as dependent variables, both complete-case and multiple imputation (MI) analyses were conducted. We found no association between multiple-metal exposures and neurodevelopment in covariate-adjusted models. This study demonstrates the use of LCA together with MI to determine patterns of exposure to multiple toxic metals and relate these to child neurodevelopment. However, because the overall study population was small, other studies with larger sample sizes are needed to investigate these associations. One hundred and three yeasts isolated from soil samples from King George Island and Tierra del Fuego province were screened in relation with their capability to produce pectinolytic enzymes. Although all the yeasts showed well-developed colonies at 20 °C, only eight showed a clear halo around the colony, indicative of pectin degradation. A secondary screening demonstrated that only four yeasts were capable to produce pectinases at low temperatures (8 °C). It could be seen that the selected yeasts were able to grow and produce high levels of polygalacturonase activity when submerged fermentations were performed using pectin-containing fruit wastes as substrates. None of the strains produced neither lyase nor rhamnogalacturonan hydrolase activities. Regarding pectin esterase activity, it was only produced in lower amounts by G. pullulans 8E (0.022 U ml−1). A TLC analysis of the substrate cleavage pattern of the pectinolytic systems was consistent with an endo-type activity. The clarification of apple juice was only accomplished by G. pullulans pectinolytic system, with a clarification of 80% (%T650) using 4 U/ml of enzyme at 20 °C. As far as we concern this work describes for the first time the production of pectinases by the cold-adapted yeasts species Cystofilobasidium infirmominiatum, Cryptococcus adeliensis and G. pullulans. One of the most important steps of new product development process is product optimization, which aims at identifying consumers’ ideal products and directions for product reformulation. The present work proposes the application of a penalty analysis based on consumer responses to CATA questions to identify drivers of liking and directions for product reformulation. Two studies were conducted in which 74 and 119 consumers evaluated a set of samples (5 apples and 8 yogurts) using a check-all-that-apply question related to sensory characteristics and were also asked to check all the terms they considered appropriate to describe their ideal product. Data were analyzed by counting the number of consumers who did not check an attribute as they did for their ideal product, and its associated mean drop. A dummy variable transformation approach was proposed to make linear regression models between CATA terms and overall liking scores using Partial Least Squares (PLS). Juiciness, sweetness, apple flavor, firmness and crispiness were the most relevant attributes for consumers in the apple study. Meanwhile, in the yogurt study smoothness, homogeneity and creaminess were the main drivers of liking and were responsible for the highest penalization on overall liking (more than 1 in the 9-point hedonic scale). PLS regression enabled the identification of the attributes which deviation from the ideal caused a significant decrease in overall liking. Penalty analysis on CATA questions proved to be a simple and useful approach to identify drivers of liking and directions for improving the products in both studies. The development of new radiopharmaceuticals for the detection of hidden infection foci has great relevance for early detection and the selection of the correct treatment, particularly in immunosuppressed patients. In that sense, the labelling of antimicrobial peptides (AMPs) that are capable of binding specifically to the pathogenic microorganism which causes the infection, should provide a sufficiently specific agent, able to distinguish an infection from a sterile inflammation. Defensins are particularly interesting molecules with antimicrobial activity, the EcgDf1 defensin was identified from the genome of a Uruguayan native plant, Erythrina crista-galli, the ‘Ceibo’ tree. Our group has previously reported a synthetic biologically active short analogue EcgDf21 (ERFTGGHCRGFRRRCFCTKHC) successfully labelled with 99mTc. Herein we present a shorter analogue which also preserves the γ-core domain, as a pharmacophore for a potential infection detection agent. This peptide was derivatized with the bifunctional chelating agent 1,4,7-triazacyclononane-1,4,7-triacetic acid (NOTA) through a lysine linker in the amino-terminal group (NOTA-KGHCRGFRRRC) and radiolabelled with 68Ga ([68Ga]Ga-NOTA-K-EcgDf1(10)). The [68Ga]Ga-NOTA-K-EcgDf1(10) labelling procedure rendered a product with high radiochemical purity and stability in the labelling milieu. The Log P value indicated that the complex has a hydrophilic behaviour, confirmed by the biodistribution profile. The [68Ga]Ga-NOTA-K-EcgDf1(10) complex demonstrated specific binding to cultures of Candida albicans and Aspergillus niger. Its biodistribution showed renal elimination and low accumulation in the rest of the body. It was possible to successfully differentiate sterile inflammation from infection by PET images in nude mice with a target/non-target ratio of 3.3 for C. albicans and 3.7 for A. niger, respectively. La técnica de asociación de palabras fue utilizada para realizar un estudio preliminar de la percepción sobre los tipos de carnes picadas comercializadas en Montevideo. 60 consumidores recibieron cinco tarjetas con las palabras: Carne Picada Común, Magra, Premium, de Ternera y Especial y se les solicitó que escribieran las cuatro primeras imágenes, asociaciones, pensamientos o sentimientos que vinieran a su mente al leer cada tarjeta. Posteriormente, evaluaron cuán saludable consideraban que era cada tipo de carne picada. Las respuestas fueron agrupadas en 16 categorías. La Carne Picada Común fue percibida como un alimento con alto contenido de grasa y evaluada como poco saludable. Las carnes picadas Magra y Premium fueron percibidas como productos con bajo contenido graso, nutritivas, saludables y de precio elevado. La tipo Premium fue considerada, además, como un producto de calidad. La carne picada de Ternera fue percibida principalmente como un producto con una textura tierna, suave y blanda, mientras que en la carne picada Especial se encontraron contradicciones en las palabras utilizadas por los consumidores para su descripción (cara-económica, con poca grasa- con mucha grasa). Por medio de esta técnica se identificaron los atributos más relevantes en el proceso de selección y toma de decisiones de compra de este tipo de producto.
