An analytical methodology was adjusted and validated for the analysis of 15 selected pesticides currently employed in extensive agriculture. The main application of this methodology is studying the pesticides degradation behavior using biobeds as a friendly environmental tool for the treatment of wastewaters generated in fields. The scope of the method was selected based on the most used pesticides in soybean crops in Uruguay. The novelty of this work is the inclusion of neutral and acidic herbicides such as 2,4-D, clethodim, dicamba, together with fungicides and insecticides which are usually included in Multi Residue Methods. An acetonitrile extraction methodology without a clean-up step yielded acceptable results for all the analytes. The instrumental analysis was performed using HPLC-MS/MS. The selected methodology was validated according to the SANTE guidelines. The recoveries were between 65 and 130% with RSD < 20%. The instrumental LOQs were fixed at 1 µg/L for all the compounds except for clethodim, and the method LOQs were 1 mg/kg in biomixture dry basis. These LOQs values are acceptable for biodegradation studies in biobeds. A multiresidue methodology was: • Validated for 15 pesticides in biomixture. • Acidic herbicides were included in the scope. • The method was employed for the environmental monitoring of pesticide degradation in biobeds. Two digestive proteases from Parona signata (Palometa) were isolated from gastric and pyloric caeca tissues and characterized. The stomach enzyme was inhibited by pepstatin and was classified as pepsin. Palometa pepsin purified from a Sephadex G-100 column appeared as two bands on an SDS-PAGE gel and exhibited optimum activity at pH 3.5, and 37C. Palometa pepsin was inhibited by pepstatin to a similar exient as porcine pepsin. An enzyme isolated from the pyloric caeca was shown to be trypsin based on its molecular weight, its ability to hydrolyze the synthetic substrates, N-α-benzoyl-arginine-p-nitroanilide (BAPA), and tosyl-arginine methyl ester (TAME) and inhibition by the known trypsin inhibitors, SBTI, TLCK, PMSF and benzamidine. The purified trypsin from palometa pyloric caeca yielded a single band with a molecular mass of 24 kDa. Optima trypsin activity was obtained at pH 8.5 and the enzyme was stable over a pH range of 3 to 11. Palometa trypsin activity was stable for 30 min at 50C, but lost 50% of its activity after 30 min at 60C. The optimum temperature for activity was 65C. The biochemical properties of Palometa trypsin and pepsin were discussed in relation to the varying environment of Palometa in the Rio de la Plata estuary. In the search for new compounds with antitumor activity, coordination complexes with different metals are being studied by our group. This work presents the synthesis and characterization of six copper complexes with general stoichiometry [Cu(L-dipeptide)(phen)]·nH2O (were phen= 1,10-phenanthroline) and their cytotoxic activities against tumor cell lines. To characterize these systems, analytical and spectroscopic studies were performed in solid state (by UV–visible, IR, X-ray diffraction) including the crystal structure of four new complexes (of the six complexes studied): [Cu(Ala-Phe)(phen)]·4H2O, [Cu(Phe-Ala)(phen)]·4H2O, [Cu(Phe-Val)(phen)]·4.5H2O and [Cu(Phe-Phe)(phen)]·3H2O. In all of them, the copper ion is situated in a distorted squared pyramidal environment. The phen ligand is perpendicular to the dipeptide, therefore exposed and potentially available for interaction with biological molecules. In addition, for all the studied complexes, structural information in solution using EPR andUV– visible spectroscopieswere obtained, showing that the coordination observed in solid state ismaintained. The lipophilicity, DNA binding and albumin interaction were also studied. Biological experiments showed that all the complexes induce cell death in the cell lines: HeLa (human cervical adenocarcinoma),MCF-7 (human metastatic breast adenocarcinoma) and A549 (human lung epithelial carcinoma). Among the six complexes, [Cu(Ala-Phe)(phen)] presents the lowest IC50 values. Taken together all these data we hypothesize that [Cu(Ala-Phe)(phen)] may be a good candidate for further studies in vivo. ETHNOPHARMACOLOGICAL RELEVANCE: Ophidian accidents are a serious public health problem in Argentina; the Bothrops species is responsible for 97% of these accidents, and in particular, B. diporus is responsible for 80% of them. In the northeast of the country (Corrientes Provinces), Cissampelos pareira L. (Menispermaceae) is commonly used against the venom of B. diporus; its use is described in almost all ethnobotanical literature from countries where the plant grows. AIM OF THE STUDY: In this study, the in vitro and in vivo antivenom activities of C. pareira extracts were evaluated against B. diporus venom, with a particular focus on the local effects associated with envenoming. The seasonal influence on the chemical composition of the active extracts was also studied, in order determine the associated range of variability and its influence on the antivenom activity. MATERIALS AND METHODS: This research was conducted using aerial parts (leaves, flowers, tender stems) and roots of Cissampelos pareira collected from two different phytogeographic regions of Corrientes (Argentina); Paso de la Patria and Lomas de Vallejos. In addition, to perform a seasonal analysis and to evaluate the metabolic stability, material was collected at three different growth stages. In vivo and in vitro anti-snake venom activities were tested, and a bio-guided chromatographic separation was performed in order to determine the active chemicals involved. The fractions obtained were analyzed by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and the chemical profile of the most active constituent was analyzed by ultra high-performance liquid chromatography coupled to quadrupole/high-resolution mass spectrometry (Q-Orbitrap). RESULTS: The alcoholic extract was found to be the most active The bio-guided fractionation allowed selection one fraction to be analyzed by UHPLC-MS in order to identify the components responsible for the activities found; this identified five possible flavonoids. CONCLUSIONS: Our studies of the activity of C. pareira against the venom of B. diporus have confirmed that this species possesses inhibitory effects in both in vitro and in vivo models. Moreover, the present data demonstrate that certain flavonoids may mitigate some of the venom-induced local tissue damage. The qualitative and quantitative composition of the essential oil from aerial parts of Aloysia virgata var. plathyphylla (Verbenaceae) has been investigated for the first time. This species grows naturally in northeast Argentina, where is employed in folk medicine. The essential oils were obtained by steam distillation of leaves and young stems at different stages and analyzed by GC-MS. The Pechini sol-gel method has been employed for the synthesis of pure and (181Hf→)181Ta-doped Y2O3 nanopowders. We performed a structural characterization from the micro to the subnanoscopic scale by means of scanning electron microscopy, energy dispersive x-ray spectroscopy, x-ray diffraction, and time-differential perturbed γ–γ angular correlation (PAC) spectroscopy. The results show the formation of the cubic bixbyite structure after a thermal treatment at 1473 K. For the synthesized 181Ta-doped Y2O3, the PAC experiments demonstrate that the impurities are mainly located at both substitutional cationic sites of the bixbyite structure. The experimental investigation was complemented by performing first-principles electronic structure calculations for Ta atoms localized at the two cationic sites of the Y2O3 semiconductor structure, which allow the study of the structural and electronic modifications induced in the host system when the impurities are introduced. These calculations confirm that the measured electric-field gradients for the synthesized 181Ta-doped Y2O3 correspond to double-ionized impurities located at substitutional defect-free cationic sites of the bixbyite host structure and indicate the site occupancy preference for 181Hf(→181Ta) doping. The behaviour of the site preference of 181Ta impurities with temperature is also discussed. In addition to an extensive structural and electronic characterization of pure and Ta-doped Y2O3 semiconductor, our results demonstrate that the Pechini sol-gel process is an affordable and effective way to successfully synthesize these PAC substitutional doped samples. Pure and Ta-doped ZnO were prepared by the sol-gel method in acetic medium using different annealing temperature treatments. The effects of low Ta doping on the crystallinity and electrical properties of ZnO nanoparticles were analyzed by means of X-ray diffraction and micro-Raman, UV–visible, and impedance spectroscopies. We also performed first principles calculations in order to study the predicted changes in the structural, vibrational and electronic properties induced by the inclusion of the impurities, and to complement the experimental measurements. We showed that above thermal treatments at 600 °C the precursor samples take the hexagonal wurtzite ZnO phase with high crystallinity. For the doped samples, we found that the synthesizing method has good Ta doping efficiency of the ZnO host structure. Also, Ta doping substantially decreases the resistivity compared to pure ZnO. These results confirm that Ta impurities can substitute Zn atoms and act as donor impurities in the host semiconductor. The Pechini-type sol-gel (PSG) process has been used for the preparation of doped oxides due to its capability to overcome most of the difficulties that frequently occur by using other producing methods. In this work we analyze the case of samples of pure and In-doped yttria (Y2O3) prepared by the PSG process. We experimentally characterize the synthesized samples by x-ray diffraction, micro-Raman spectroscopy, electrochemical impedance spectroscopy (EIS), and time-differential perturbed γ-γ angular correlation (PAC) spectroscopy, and we compare these results with those obtained starting from commercial oxide powders. We found that the PSG process can be used to successfully produce doped yttria in the cubic phase, with the impurities substitutionally located at the cationic sites of the structure. By the proposed PSG route, the inclusion of impurities does not affect the particle size nor the resistivity. However, when we compare the PSG samples with other samples produced from commercial powder, we found that the first have lower resistivities at grain interiors. On the other hand, PAC spectroscopy in 111In(→111Cd)-doped yttria allows the study of the dynamic hyperfine interactions observed by the radioactive 111Cd impurity-probe, which can be used to “sense” the host electron availability near the impurities after the electron-capture decay of 111In. Differences between PAC spectra for PSG samples and the commercial powder suggest that the PSG process introduces additional donor defects into the yttria electronic structure, which is consistent with the lower resistivity observed in the PSG samples by EIS spectroscopy. Pseudomonas fluorescens CFBP2392 has been recognized as a potential biocontrol agent due to its ability to suppress damping-off and root rot disease. This isolate has antibacterial activity in vitro as many other strains from the Pseudomonas fluorescens complex. In this work, the antibacterial and antifungal activity of the strain were explored. Dual culture assays evidenced the antifungal activity of the strain against different phytopathogens: Alternaria sp., Pythium ultimun, Fusarium oxysporum, and Rhizoctonia solani. Purification of an antifungal fraction was performed by preparative HPLC from the chemical extraction of growth media. The fraction showed altered R. solani growth and ultrastructure. Transmission electron microscopy revealed the purified compound induced hypertrophied mitochondria, membranous vesicles, and a higher number of vacuoles in R. salani cytoplasm. In addition, co-cultivation of P. fluorescens CFBP2392 with R. solani resulted in an enlarged and deformed cell wall. To gain genomic insights on this inhibition, the complete genome of P. fluorescens CFBP2392 was obtained with Oxford Nanopore technology. Different biosynthetic gene clusters (BGCs) involved in specialized metabolites production including a lokisin-like and a koreenceine-like cluster were identified. In accordance with the putative BGCs identified, sequence phylogeny analysis of the MacB transporter in the lokisin-like cluster further supports the similarity with other transporters from the amphisin family. Our results give insights into the cellular effects of the purified microbial metabolite in R. solani ultrastructure and provide a genomic background to further explore the specialized metabolite potential. Subject The topic of the resource ANTITRIPANOSOMAS ANTIPARASITARIOS NIFURTIMOX BIBLIOGRAFIA NACIONAL QUIMICA 2004 Abstract A summary of the resource. The in vitro growth inhibition activity of new thiosemicarbazone derivatives against the protozoan parasite Trypanosoma cruzi, the causative agent of American trypanosomiasis, are described. The designed compounds combine in the same molecule the thiosemicarbazone function, recently described as a potent cruzain-inhibitor moiety, and the recognised 5-nitrofuryl group, an oxidative stress promoter. Some of the derivatives were found to be very active against the cultured (epimastigote) form of the parasite, being 1.5–1.7-fold more active than the reference compound, Nifurtimox. Free radicals production was detected when the compounds were incubated in presence of mammalian-liver microsomes. The thiosemicarbazones� capacity to act as pharmacophore in the cruzain inhibition process was theoretically analysed. Frontier molecular orbital HOMO was found as an adequate descriptor in this process. Acute in vivo toxicity of two of the more active derivatives was evaluated. The results showed that these compounds are among the most potent 5-nitrofuryl derivatives tested against this parasite thus support further in vivo studies of some of these thiosemicarbazones.
