Due to their size, small molecules cannot be simultaneously bound by two antibodies, precluding their detection by noncompetitive two-site immunoassays, which are superior to competitive ones in terms of sensitivity, kinetics, and working range. This has prompted the development of anti-immune complex antibodies, but these are difficult to produce, and often exhibit high crossreactivity with the unliganded primary antibody. This work demonstrates that anti-immune complex antibodies can be substituted by phage particles isolated from phage display peptide libraries. Phages bearing specific small peptide loops allowed to focus the recognition to changes in the binding area of the immune complex. The concept was tested using environmental and drug analytes; with improved sensitivity and ready adaptation into on-site formats. Peptides specific for different immune complexes can be isolated from different peptide libraries in a simple and systematic fashion allowing the rapid development of noncompetitive assays for small molecules

The impact of the use of herbicides in agriculture can be minimized by compliance with good management practices that reduce the amount used and their release into the environment. Simple tests that provide real time on-site information about these chemicals are a major aid for these programs. In this work, we show that phage anti-immunocomplex assay (PHAIA), a method that uses phage-borne peptides to detect the formation of antibody-analyte immunocomplexes, is an advantageous technology to produce such field tests. A monoclonal antibody to the herbicide clomazone was raised and used in the development of conventional competitive and noncompetitive PHAIA immunoassays. The sensitivity attained with the PHAIA format was over 10 times higher than that of the competitive format. The cross-reactivity of the two methods was also compared using structurally related compounds, and we observed that the two-site binding of PHAIA "double-checks" the recognition of the analyte, thereby increasing the assay specificity. The positive readout of the noncompetitive PHAIA method allowed adaptation of the assay into a rapid and simple format where as little as 0.4 ng/mL clomazone (more than 10-fold lower than the proposed standard) in water samples from a rice field could be easily detected by simple visual inspection.

Competitive immunoassays for the detection of small analytes, such as pesticides and their metabolites, use haptens that compete with the target compounds for binding to the antibody. This competing hapten can be either the same as the immunizing hapten (homologous assay) or structurally modified mimics of the immunizing hapten (heterologous assay). Polyclonal antibody-based heterologous immunoassays have shown superior sensitivities to homologous ones, but the synthesis of heterologous haptens may be time-consuming, requiring expertise in synthetic chemistry. In this work we demonstrate that phage display peptide libraries can be used as a source of phage-borne peptidomimetics to facilitate the development of sensitive heterologous assays. Different strategies for the isolation of these peptides were explored using two metabolites of pyrethroid insecticides. The sensitivities of the best competitive phage heterologous enzyme-linked immunosorbent assays were 13 fold and 100 fold better than the homologous assay, for the glycine conjugate of trans-3-(2,2-dichlorovinyl)-2,2-dimethylcyclopropane-1-carboxylic acid and 3-phenoxy-benzoic acid, respectively. The phage particles were highly versatile as tracer reagents, allowing the use of enzymatic, chemiluminescent, or immuno-polymerase chain reaction detection. The data presented here shows a new systematic procedure that enables the fast generation of several competing haptens for the rapid development of sensitive heterologous immunoassays.

Three cos-type virulent Streptococcus thermophilus phages were isolated from failed mozzarella production in Uruguay. Genome analyses showed that these phages are similar to those isolated elsewhere around the world. The CRISPR1 and CRISPR3 arrays of the three S. thermophilus host strains from Uruguay were also characterized and similarities were noted with previously described model strains SMQ-301, LMD-9 and DGCC7710. Spontaneous bacteriophage-insensitive S. thermophilus mutants (BIMs) were obtained after challenging the phage-sensitive wild-type strain Uy02 with the phage 128 and their CRISPR content was analyzed. Analysis of 23 BIMs indicated that all of them had acquired at least one new spacer in their CRISPR1 array. While 14 BIMs had acquired spacer at the 5'-end of the array, 9 other BIMs acquired a spacer within the array. Comparison of the leader sequence in strains Uy02 and DGCC7710 showed a nucleotide deletion at position -1 in Uy02, which may be responsible for the observed ectopic spacer acquisition. Analysis of the spacer sequences upstream the newly acquired ectopic spacer indicated presence of a conserved adenine residue at position -2. This study indicates that natural strains of S. thermophilus can also acquire spacers within a CRISPR array.

Infection by larval Echinococcus granulosus is usually characterized by tight inflammatory control. However, various degrees of chronic granulomatous inflammation are also observed, reaching a high point in infection of cattle by the most prevalent parasite strain worldwide, which is not well adapted to this host species. In this context, epithelioid and multinucleated giant macrophages surround the parasite, and the secreted products of these cells often associate with the larval wall. The phagocyte-specific S100 proteins, S100A8, S100A9 and S100A12, are important non-conventionally secreted amplifiers of inflammatory responses. We have analysed by proteomics and immunohistochemistry the presence of these proteins at the E. granulosus larva-host interface. We found that, in the context of inflammatory control as observed in human infections, the S100 proteins are not abundant, but S100A9 and S100A8 can be expressed by eosinophils distal to the parasite. In the granulomatous inflammation context as observed in cattle infections, we found that S100A12 is one of the most abundant host-derived, parasite-associated proteins, while S100A9 and S100A8 are not present at similarly high levels. As expected, S100A12 derives mostly from the epithelioid and multinucleated giant cells. S100A12, as well as cathepsin K and matrix metalloproteinase-9, also expressed by E. granulosus-elicited epithelioid cells, are connected to the Th17 arm of immunity, which may therefore be involved in this granulomatous response

Abstract
A summary of the resource.
Environmental exposure to active pharmaceutical ingredients (APIs) can have negative effects on the health of ecosystems and humans. While numerous studies have monitored APIs in rivers, these employ different analytical methods, measure different APIs, and have ignored many of the countries of the world. This makes it difficult to quantify the scale of the problem from a global perspective. Furthermore, comparison of the existing data, generated for different studies/regions/continents, is challenging due to the vast differences between the analytical methodologies employed. Here, we present a global-scale study of API pollution in 258 of the world’s rivers, representing the environmental influence of 471.4 million people across 137 geographic regions. Samples were obtained from 1,052 locations in 104 countries (representing all continents and 36 countries not previously studied for API contamination) and analyzed for 61 APIs. Highest cumulative API concentrations were observed in sub-Saharan Africa, south Asia, and South America. The most contaminated sites were in low- to middle-income countries and were associated with areas with poor wastewater and waste management infrastructure and pharmaceutical manufacturing. The most frequently detected APIs were carbamazepine, metformin, and caffeine (a compound also arising from lifestyle use), which were detected at over half of the sites monitored. Concentrations of at least one API at 25.7%of the sampling sites were greater than concentrations considered safe for aquatic organisms, or which are of concern in terms of selection for antimicrobial resistance. Therefore, pharmaceutical pollution poses a global threat to environmental and human health, as well as to delivery of the United Nations Sustainable Development Goals.

The phosphatidylinositol 3-kinase (PI3K) pathway is known to down-regulate inflammatory cytokine responses in dendritic cells and macrophages stimulated with TLR agonists. This is due to class I PI3Ks causing the activation of Akt, which in turn inactivates GSK3, a kinase that promotes the transcription of IL-12 and represses that of anti-inflammatory IL-10. Using bone marrow-derived dendritic cells we find that whereas pharmacological inhibition of Akt or GSK3 has the expected effects on these cytokines, the widely used PI3K inhibitor wortmannin causes a paradoxical increase in the production of IL-10. Wortmannin inhibits all PI3K classes, including PI3K class III, involved in endosomal function and autophagy, for which specific inhibitors were until recently not available. Using inhibitors specific for PI3K class III vs class I, we show that whereas inhibition of class I PI3K has the expected opposing effects on IL-10 and IL-12 production, inhibition of class III PI3K enhances the production of both of these, plus further cytokines. This explains the paradoxical inhibition of IL-10 production by wortmannin.

A summary of the resource.
The chemistry of furoxans (1, 2, 5-oxadiazole-2-oxides) and benzofuroxans (benzo[1, 2-c]1, 2, 5- oxadiazole-1-oxides) is very well known. These systems are widely used in organic chemistry as intermediate compounds for the synthesis of numerous heterocycles. In the other hand, furoxan and benzofuroxan derivatives were extensively studied as bioactive compounds. They possess remarkable biological activities, such as anti-microbial and anti-parasitic properties, mutagenic, immunosuppressive and anticancer effects, anti-aggregating and vasorelaxant activity, among others. In some cases, molecular mode of action was proposed. Recently, the research and development in the medicinal chemistry of these systems have produced hybrid compounds in which furoxan or benzofuroxan moieties together with a classical drug moieties are present in a single molecule. So, new anti-ulcer drugs, calcium channel modulators and vasodilator derivatives were described and they are currently in study. In this presentation recent developments in the medicinal chemistry of furoxans and benzofuroxans will be reviewed.

The chemistry of indazole and its N-oxide derivatives is very well-known. Indazole derivatives were<br />extensively studied as bioactive compounds, such as anti-aggregatory and vasorelaxant activity by NO release<br />and increase of cGMP levels and anticancer effects, antimicrobial and antiparasitic properties, among others.<br />Recently, the research and development in the medicinal chemistry of these systems have produced<br />compounds with contraceptive activities for men, for the treatment of osteoporosis, inflammatory disorders<br />and neurodegenerative diseases. On the other hand, indazole N-oxide derivatives were poorly studied as<br />bioactive compounds, but recently compounds with antiparasitic properties were produced. In this<br />presentation, recent developments in the chemistry and medicinal chemistry of indazole and its N-oxide<br />derivatives will be reviewed.

Heterogeneous nucleation and further growth of HgI2 onto glass were studied by the physical vapor deposition (PVD) method, with and without an argon atmosphere. Glass substrates 2″×2″ in area were used for nucleation and growth. Supersaturation strongly determined not only the nuclei population and size, but also the HgI2 nucleation phase. A range of nucleation–growth temperatures between 303 and 333 K for the substrate, and a range of initial argon pressure between 1.3 and 2.6×104 Pa were found appropriate for nucleation and growth in the α (red) phase of HgI2. Other conditions, such as an argon pressure higher than 4×104 Pa, determined the nucleation and growth in the β (yellow) metastable phase. After a nucleation growth time of 30 min, clusters of about 0.5 μm for β-HgI2 and 1 μm for α-HgI2 and a minimum distance between the clusters of about 1 and 10 μm were, respectively, obtained. Coalescence was performed by annealing the clusters at a temperature between 303 and 333 K and from 5 min to 24 h, at an initial argon pressure of 104 Pa. They coalesced to form larger clusters, which, in the case of the β-phase also transformed to the α-stable phase. From the experimental nucleation–growth and further coalescence conditions, correlations between supersaturation conditions and phase were established. Also, considerations about interface free energies and nucleation phase were made. Future work will be conducted to diminish nuclei size and enlarge nuclei population, seeking for early stages of nucleation.

The synthesis and evaluation as hypoxic selective cytotoxins of 2-amino- or 2-hydroxyphenazine 5,10-dioxide derivatives and reduced analogues are reported. In vitro cytotoxicities on V79 cells under hypoxic and aerobic conditions were determined. Some derivatives, such as 7(8)-bromo-2-hydroxyphenazine 5,10-dioxide, showed selective toxicity toward hypoxic cells and along with derivatives 7(8)-bromo-2-aminophenazine 5,10-dioxide and 7(8)-chloro-2-aminophenazine 5,10-dioxide behave as hypoxic trigger cytotoxins. These compounds represent interesting leads for further chemical modifications and biological studies.

A summary of the resource.
Objective: To determine the pharmacokinetic parameters of phenobarbital in infants, using saliva as a biologic fluid, and to correlate the parameters obtained from saliva data with those obtained from plasma data. Design: A prospective, randomized study. Setting: Hospitalized patients at the Medical Center Pereira Rossell, a pediatric hospital in Montevideo, Uruguay. Patients: Sixteen infants with seizure disorders were included in the study. None of them was treated with other medications. Interventions: A direct intravenous loading dose of phenobarbital 10 mg/kg was administered, followed by a maintenance dosage of 5 mg/kg/d (once- or twice-daily dosing) given 12 hours after the loading dose. Main Outcome Measures: Saliva and plasma samples were obtained 6 and 12 hours after the loading dose and 3 days after the initiation of the maintenance dose (trough sample). The samples were analyzed by HPLC and the elimination half-life (t1/2), the volume of distribution (Vd), and the percentage of unbound drug in plasma (UDP) were calculated. Results: The t1/2 obtained from plasma and saliva data was 30 hours; the Vd was 0.73 L/kg from plasma data and 2.4 L/kg from saliva data; and the UDP was 75 percent. Trough concentrations showed no significant difference between treatments. Conclusions: Saliva is a useful biologic fluid to determine phenobarbital pharmacokinetic parameters, mainly in pediatric patients. Moreover, a single daily dose of phenobarbital is sufficient to obtain therapeutic concentrations.

Eucalyptus grandis sawdust, a major waste from the growing Uruguayan wood industry, was used in previous work to prepare powdered activated carbon (PAC). In the present work, granular activated carbon (GAC) was prepared by mixing PAC, carboxymethyl cellulose as a binder, and kaolin as reinforcer. Ultimate analysis and surface characterization of GAC and PAC were performed. Phenol adsorption was used as a way to compare the characteristics of different PAC and GAC preparations. Kinetics and isotherms of the different GAC and PAC were performed in a shaking bath at 100 rpm and 298 K. Phenol concentrations were determined by UV spectroscopy. Some kinetics parameters were calculated; from kinetics results, external resistance to mass transfer from the bulk liquid can be neglected as the controlling step. Isotherms were fitted to Langmuir and Freundlich models, and corresponding parameters were determined. Maximum phenol uptakes for all carbons were determined and correlated with carbon characteristics. Thermogravimertic analysis (TGA) determinations were performed in order to study adsorption characteristics and conditions for GAC regeneration after its use. The results showed that phenol is preferentially physisorbed on the carbon of the granules, though some chemisorption was detected. No adsorption was detected in the kaolin–carboxymethyl cellulose mixture.

A summary of the resource.
Detailed composition of phenolic compounds in the stems, seeds and flesh (including peel and core) recovered from crab apple pomace (seven single-varieties) was studied. The flesh constituted of 95.8–98.6%, whereas, the seeds and stems accounted for the remaining 1.0–3.7% and 0.4–0.7%, respectively, of the total apple pomace expressed in dry weight basis (dwb). The highest content of polyphenols was detected in seeds and stems 3592.6–23606.8 and 6747.2–59335.4 mg/kg dwb, respectively. In the flesh, the concentration of polyphenols was between 1654.8–5314.1 mg/kg dwb. Chlorogenic acid (405.3–2298.8 mg/kg dwb) was the main polyphenol detected in the flesh. For the seeds and stems – phloridzin was the main identyfied phenolic compound (2899.0–19600.3 and 3017.7–40488.3 mg/kg dwb, respectively). A dihydrochalcone – phloretin dihexoside was detected only in the seeds, and 3-hydroxyphloridzin was mainly identified in the stems, hence, these two dihydrochalcones may play an important role as quality markers of the apple pomace.

A summary of the resource.
A study on the phenolic content and antioxidant properties of Uruguayan propolis is presented. Identification and quantification was carried out by Reverse Phase High Performance Liquid Chromatography (RP-HPLC). Antioxidant activity was determined by Antiradical Activity Determination (AAD) measured with DPPH and Microsomal Lipid Peroxidation (MLP) assays. Two phenolic acids (gentistic and p-coumaric) and eight flavonoids (fisetin, myricetin, luteolin, quercetin, kaempferol, pinocembrin, chrysin and tectochrysin) were identified. The existence of five other phenols is postulated. The large difference between range of values of scavenger activity AAD (between 60% and 100%) as compared with lipoperoxidation inhibition (between 15% and 92%), suggests that the complexity of the biologic matrix could modulate the antioxidant effect. From a commercial standpoint, research such as the one presented here endows the product with added value, making it more appealing to the interest of industry and entrepreneurs.

Persicaria maculosa (Polygonaceae) (known as lady’s thumb) is an annual morphologically variable weed that is widely distributed in Chile. The purpose of this study was to investigate the antifeedant potential of methanolic (MeOH), ethanolic (EtOH), and dichloromethane (DCM) extracts from the aerial parts of this plant collected in the Valparaíso and Curicó provinces (Chile) and relate this activity to the antioxidant capacity and the presence of phenolic compounds in the extracts. A novel approach for the analysis of phenolics in tea was developed using multiplexed simultaneous detection. This is the first study employing a short reaction flow (RF) chromatography column (length 50 mm) for the analysis of phenolics in complex samples (not a standard mixture of compounds). RF separation efficiency was high as the detectors are multiplexed simultaneously, rather than sequentially. The delay time between RF detection responses is minor (1.2 s) providing simple peak assignment between the phenolic detection and conventional detectors, such as the UV and fluorescence detectors (FLD). Thirty-nine phenolics were detected amongst the five different tea samples (three from Camellia sinensis and two herbal teas). The highest number of phenols detected was 24 for English Breakfast tea (four highly abundant), 19 for Earl Grey (six highly abundant), 13 in Pure Green (two highly abundant) and 9 in both Fruit of the Forest (two highly abundant) and Camomile (none highly abundant) herbal teas. Numerous low concentration phenolic compounds in the Fruit of the Forest tea were not included due to their abundance being only slightly above the noise level of the detection process. The relative response factor (RRF) analysed at 520 and 500 nm aided to identify unique phenolics to each sample with similar retention times; three phenolics were unique to the Pure Green tea (2.30, 4.11 and 5.80 min) and two to the Earl Grey Tea (4.11 and 5.80 min). The Fruit of the Forest tea and the Camomile tea were both distinctly different from all other teas. This approach may be used for phenolic profiling of food, beverages or natural products.
