Actualmente la mayoría de las vacunas son diseñadas con subunidades las cuales poseen una inmunogenicidad limitada, por lo que es necesario el uso de adyuvantes en su formulación. A pesar de los avances en inmunología, el empleo de compuestos de aluminio como adyuvantes sigue siendo la opción más aceptada. Éstos presentan desventajas, lo que ha impulsado el desarrollo de nuevos adyuvantes, que es en la actualidad un campo de gran actividad científica. En esta área se enmarca el presente proyecto de tesis. Las suspensiones FCC-S06 y HAP-S08, sintetizadas a partir de calcita (carbonato de calcio), suministradas por una empresa extranjera fueron evaluadas como posible adyuvante. La FCCS06 es una suspensión de micropartículas funcionalizadas con grupos fosfato. Obteniéndose, según la empresa, micropartículas donde se incorpora hidroxiapatita en la superficie, resultando en un incremento considerable en la superficie específica. La HAP-SO8 fue sintetizada utilizando otro proceso de síntesis con el objetivo de obtener una suspensión de partículas con un diámetro promedio inferior al mínimo obtenido utilizando la síntesis estándar de FCCs. La empresa informó que la suspensión resultante teóricamente está compuesta por partículas que son 100% hidroxiapatita. Como está demostrada la eficacia de la hidroxiapatita en la adsorción de una gran variedad de proteínas, es biocompatible y se ha utilizado como adyuvante de inmunización se eligieron ambas suspensiones como candidatas para su evaluación como adyuvantes. En el presente trabajo se confirmó la hipótesis de que la suspensión FCCS06 es biocompatible ya que no se observó toxicidad en los estudios in vitro ni in vivo, siendo menos tóxica que el adyuvante de referencia. Está formada por un core de carbonato de calcio con una sal de la familia de los fosfatos de calcio precipitada en su superficie, cristalina, de morfología esferoide con estructuras en forma de placas laminares que sobresalen de la superficie formando poros intrapartículas, lo que aumenta considerablemente la superficie específica. Las formulaciones con FCC-S06 tienen la capacidad de activar al sistema inmune 9 innato, estimulando la activación y maduración de células presentadoras de antígenos, la presentación antigénica, el reclutamiento de componentes del sistema inmune al sitio de inoculación y la secreción de citoquinas proinflamatorias. Por lo anterior, se podría considerar como un posible candidato adyuvante. Lo antes mencionado modula la respuesta adaptativa hacia un perfil Th17 y Th1. En cuanto a la suspensión HAP-S08 no se pudo identificar la sal de la familia de los fosfatos de calcio que la compone. Se determinó que está formada por partículas amorfas sin poros intrapartículas. Los resultados permiten asegurar que no es tóxica ni in vitro ni in vivo, y sugieren que las formulaciones con HAPS08 inducen in vitro la activación y maduración de células presentadoras de antígenos, y en concordancia con el perfil de citoquinas secretado por estas células, estimulan la activación de una respuesta inmune adaptativa polarizada hacia un perfil Th17. Sin embargo, poseen una gran limitante, son muy inestables y sedimentan en periodos cortos de tiempo siendo difícil su re-dispersión. Lo anterior hace que su aplicación en el diseño de vacunas no sea recomendable. Por lo cual para seguir avanzando en el proceso de desarrollo de un nuevo adyuvante es necesario realizar un nuevo proceso de síntesis que no se base en la precipitación. The marine bacterium Stenotrophomonas rhizophila was assessed in vitro and in vivo as biocontrol agent against anthracnose disease of mango fruit caused by Colletotrichum gloeosporioides. The results showed that in vitro inhibition of the colony diameter and spore germination of the phytopathogen was due to the production of VOCs, competition for nutrients, and lytic enzymes. When a concentration of 1 × 108 cells ml−1 of the antagonist bacterium was applied to the fruit, disease incidence was reduced by 95%, and the lesion diameter of anthracnose decreased by 85%, which offered greater protection than the synthetic fungicide. This is the first report of antagonistic mechanisms of the marine bacterium S. rhizophila against anthracnose disease in mango, which in this study was found to be more effective than the synthetic fungicide. Most of the fourteen Raman lines expected for this compound according to group theory analysis were observed in the spectrum. The effect of storage time on quality attributes of refrigerated fresh-cut mints (Mentha � piperita and M. spicata) was studied. Atmosphere composition, respiratory activity, weight loss, surface colour, total chlorophyll, carotenoids, browning potential, total phenols, flavonoids, radical-scavenging activity, ascorbic acid and essential oil yield and composition were analysed. Respiratory activity of peppermint and spearmint samples diminished moderately (42% and 28%, respectively) after 21 days at 0 �C. A slight modification of the internal atmosphere was achieved. Surface colour, chlorophyll, carotenoid and antioxidant compounds remained almost constant. The yield of essential oil did not change or it showed an apparent increase after 21 days at 0 �C, depending on plant growth stage. The characteristic flavour components of peppermint (menthone and menthol) increased, while the contents of the main constituents of spearmint essential oil showed minor variations after storage. The conditions assayed for packaging and storing fresh-cut mints were adequate to achieve a relatively long shelf life and they retained their antioxidant properties. Dada la relevancia del desarrollo de técnicas sensibles y confiables para la detección de especies oxidantes en sistemas biológicos, en este período se trabajó intensamente en la obtención y validación de sondas fluorescentes para la<br />detección de especies nucleofílicas, en particular peroxinitrito.<br />Analizando el estado del arte sobre métodos de detección y cuantificación de peroxinitrito, donde la inmensa mayoría de sondas reportadas carecen de la apropiada validación fisicoquímica y biológica, principalmente respecto al peroxinitrito producido endógenamente, y además la alta demanda del área redox en la necesidad de contar con métodos sensibles, conocidos y confiables es que se enfocó la presente Tesis en el estudio de este tema.<br />En el desarrollo del trabajo se exploraron diferentes fluoróforos derivados de fluoresceína y de cumarina soportando ácidos o ésteres borónicos como centro electrofílico capaz de reaccionar con oxidantes nucleofílicos. Además, modificaciones<br />estructurales de los derivados boronados de cumarinas, permitieron modular sus propiedades espectroscópicas e incluso direccionarlas a un sitio subcelular específico, desarrollando nuevas moléculas para la detección de este oxidante.<br />Se desarrolló Fl-B (fluoresceína-boronato) y mediante su caracterización in vitro, análisis inéticos, ensayos de citotoxicidad y evaluación en sistemas celulares, se validó a Fl-B como una sonda sensible para la detección directa de peroxinitrito producido endógenamente. Además, los datos obtenidos permitieron estimar el flujo intracelular de peroxinitrito en células endoteliales normales, y en un modelo de parásitos internalizados por macrófagos.<br />Estos estudios fueron punto de partida para colaborar tanto con los integrantes de nuestro grupo de investigación, como con otros grupos de investigación nacionales y extranjeros. Estas investigaciones utilizaron a Fl-B aquí desarrollada, como herramienta para el entendimiento de los mecanismos moleculares y celulares en el desarrollo de patologías relacionadas con el estrés nitroxidativo generando un gran avance para el área redox.<br />Se realizó un estudio mecanístico con oxidantes marcados isotópicamente para completar la elucidación del mecanismo de reacción de boronatos con oxidantes de relevancia biológica. Como resultado se demostró inequívocamente el origen de los<br />átomos de oxígeno en los productos formados. En este contexto, fue posible producir oxidantes marcados isotópicamente como radical superóxido (18O2•—), peroxinitrito marcado (16ON18O18O—), y peroximonocarbonato marcado (HC16O218O2—). Se demostró por primera vez de forma directa que la formación de 2-hidroxietidio (2-OH-E+) a partir de la sonda hidroetidio (HE) ampliamente utilizada para la detección de superóxido, implica la incorporación de un átomo de oxígeno proveniente del 18O2 •—. También se demostró que la oxidación de boronatos por H2 18O2 o 16ON18O18O— se acompaña de la incorporación de átomos de oxígeno provenientes de estos oxidantes, y no del medio de reacción. La incorporación de átomos de oxígeno provenientes del oxidante elimina cualquier tipo de ambigüedad sobre la identidad de la especie que se va a detectar. Conocer el mecanismo de reacción completo de las sondas reporteras con analitos específicos, es crucial para un correcto y riguroso entendimiento del sistema en estudio tanto in vitro como in vivo.</p> <p>Por último, se trabajó en el diseño de una nueva clase de compuestos con una mejor capacidad antioxidante que actúen previniendo o modulando el daño nitroxidativo, permitiendo el estudio y tratamiento de disfunciones redox. In this study, several approaches were tested to optimise the production and recovery of the widely used anticancer drug Taxol® (paclitaxel) from culturable vascular stem cells (VSCs) of Taxus baccata, which is currently used as a suc‑ cessful cell line for paclitaxel production. An in situ product recovery (ISPR) technique was employed, which involved combining three commercial macro-porous resin beads (HP-20, XAD7HP and HP-2MG) with batch and semi-continu‑ ous cultivations of the T. baccata VSCs after adding methyl jasmonate (Me-JA) as an elicitor. The optimal resin combi‑ nation resulted in 234±23 mg of paclitaxel per kg of fresh-weight cells, indicating a 13-fold improved yield compared to the control (with no resins) in batch cultivation. This resin treatment was further studied to evaluate the resins’ removal capacity of reactive oxygen species (ROS), which can cause poor cell growth or reduce product synthesis. It was observed that the ISPR cultivations had fourfold less intracellular ROS concentration than that of the control; thus, a reduced ROS concentration established by the resin contributed to increased paclitaxel yield, contrary to previous studies. These paclitaxel yields are the highest reported to date using VSCs, and this scalable production method could be applied for a diverse range of similar compounds utilising plant cell culture. Nematode parasites have a profound impact on humankind, infecting nearly one-quarter of the world’s population, as well as livestock. There is a pressing need for discovering nematicides due to the spread of resistance to currently used drugs. The free-living nematode Caenorhabditis elegans is a formidable experimentally tractable model organism that offers key advantages in accelerating nematicide discovery. We report the screening of drug-like libraries using an overnight high-throughput C. elegans assay, based on an automated infrared motility reader. As a proof of concept, we screened the “Pathogen Box” library, and identical results to a previous screen using Haemonchus contortus were obtained. We then screened an in-house library containing a diversity of compound families. Most active compounds had a conjugation of an unsaturation with an electronegative atom (N, O, or S) and an aromatic ring. Importantly, we identified symmetric arylidene ketones and aryl hydrazine derivatives as novel nematicides. Furthermore, one of these compounds, (1E,2E)-1,2-bis(thiophen-3-ylmethylene)hydrazine, was active as a nematicide at 25 µm, but innocuous to the vertebrate model zebrafish at 50 µm. Our results identified novel nematicidal scaffolds and illustrate the value of C. elegans in accelerating nematicide discovery using a nonlabor-intensive automated assay that provides a simple overnight readout. Searching for prospective vanadium-based agents against Trypanosoma cruzi, the parasite causing Chagas disease, four new [VVO(8HQ–H)(L–2H)] compounds, where 8HQ is 8-hydroxyquinoline and L are tridentate salicylaldehyde semicarbazone derivatives L1–L4, were synthesized and characterized in the solid state and in solution. The compounds were evaluated on T. cruzi epimastigotes (CL Brener) as well as on VERO cells, as mammalian cell model. Compounds showed activity against T. cruzi (IC50 6.2–10.5 μM) of the same order than Nifurtimox and 8HQ, and a four- to sevenfold activity increase with respect to the free semicarbazones. For comparison, [VVO2(L–H)] series was prepared and the new [VVO2(L3–H)] was fully characterized. They showed negligible activity and low selectivity towards the parasite. The inclusion of 8HQ as ligand in [VVO(8HQ–H)(L–2H)] compounds led to good activities and increased selectivity towards the parasite with respect to 8HQ. 51V NMR experiments, performed to get insight into the nature of the active species, suggested partial decomposition of the compounds in solution to [VVO2(L–H)] and 8HQ. Depending on the dose, the compounds act as trypanocide or trypanostatic. A high uptake of vanadium in the parasites (58.51–88.9% depending on dose) and a preferential accumulation in the soluble protein fraction of the parasite was determined. Treated parasites do not seem to show a late apoptotic/necrotic phenotype suggesting a different cell death mechanism. In vivo toxicity study on zebrafish model showed no toxicity up to a 25 µM concentration of [VVO(8HQ–H)(L1–2H)]. These compounds could be considered prospective anti-T. cruzi agents that deserve further research. In this work, an exploratory study was first carried out using a focus group (FG) technique to determine attributes and aspects that influence the decision to purchase mandarins. Secondly, the consumer panel recruited 100 regular mandarin consumers that were randomly divided into two groups. Group “A” received segments of 15 mandarins. Group “B”, instead of receiving the same mandarins in segments, they received them as a whole fruit having to peel them by themselves. Both groups evaluated the total acceptability with a 9-point hedonic scale and answered a CATA questionnaire composed of 38 terms for Group “A” and 52 terms for Group “B”. Although the consumers in the FG described an "ideal" mandarin as easy-to-peel, no significant difference (p > 0.05) was found in the acceptability<br />between groups “A” and “B”. The aim of this study was to determine the prevalence of Ser315Thr substitution in isoniazid (INH)-resistant strains of Mycobacterium tuberculosis in Uruguay. The katG gene of 62 INH-resistant strains was analysed by an RFLP-PCR assay. PCR products were digested with MspI to detect Ser315Thr and Arg463Leu substitutions. A total of 16 of the 62 (26 %) INH-resistant strains analysed had a Ser315Thr substitution. Only one INH-resistant strain had an Arg463Leu substitution and two strains had a deletion in katG. Of the 16 strains with Ser315Thr, 15 showed different profiles using a double-repetitive-element PCR assay, demonstrating that there was no local dissemination of any particular strain. These findings are in agreement with published data from regions where the prevalence of tuberculosis (TB) is intermediate and may be due in part to the success of the local TB control programme. Searching for prospective agents against infectious diseases, four new ferrocenyl derivatives, [M(L)(dppf)4](PF6), with M = Pd(II) or Pt(II), dppf = 1,1'-bis(dipheny1phosphino) ferrocene and HL = tropolone (HTrop) or hinokitiol (HHino), were synthesized and characterized. Complexes and ligands were evaluated against the bloodstream form of T. brucei, L. infantum amastigotes, M. tuberculosis (MTB) sensitive strain and MTB clinical isolates. Complexes showed a significant increase of the anti-T. brucei activity with respect to the free ligands (>28- and >46-fold for Trop and 6- and 22-fold for Hino coordinated to Pt-dppf and Pd-dppf, respectively), yielding IC50 values < 5 μM. The complexes proved to be more potent than the antitrypanosomal drug Nifurtimox. The new ferrocenyl derivatives were more selective towards the parasite than the free ligands. The Pt compounds were less toxic on J774 murine macrophages (mammalian cell model), than the Pd ones, showing selectivity index values (SI = IC50 murine macrophage/IC50T. brucei) up to 23. Generation of the {M-dppf} compounds lead to a slightly positive impact on the anti-leishmanial potency. Although the ferrocenyl derivatives were more active on sensitive MTB than the free ligands (MIC90 = 9.88-14.73 μM), they showed low selectivity towards the pathogen. Related to the mechanism of action, the antiparasitic effect cannot be ascribed to an interference of the compounds with the thiol-redox homeostasis of the pathogen. Fluorescence measurements pointed at DNA as a probable target of the new compounds. [Pt(Trop)(dppf)](PF6) and [Pt(Hino)(dppf)](PF6) could be considered prospective anti-T. brucei agents that deserve further research. In this work, five new Pd–Fe heterobimetallic [PdII(L)(dppf)](PF6) compounds, including 8-hydroxyquinolyl derivatives HL1–HL5 as bioactive ligands and dppf = 1,1’-bis(diphenylphosphino)<br />ferrocene as the organometallic co-ligand, were synthesized and fully characterized in the solid state and in solution. Molecular structures of three compounds were solved by single crystal X-ray diffraction methods. The compounds displayed submicromolar or micromolar IC50 values against bloodstream<br />T. brucei (IC50: 0.33–1.2 μM), and good selectivity towards the pathogen (SI: 4–102) with respect to mammalian macrophages (cell line J774). The new Pd complexes proved to be 2-fold to 45-fold more potent than the drug nifurtimox but most of them are less active than their Pt analogues. Potential molecular<br />targets were studied. The complexes interact with DNA but they do not alter the intracellular thiolredox homeostasis of the parasite. In order to understand and predict the main structural determinants on the anti-T. brucei activity, a search of quantitative structure–activity relationships (QSAR) was performed including all the [M(L)(dppf)](PF6) complexes, where M = Pd(II) or Pt(II), currently and previously<br />developed by us. The correlation obtained shows the relevance of the electronic effects, the lipophilicity and the type of metal. According to the QSAR study, compounds with electron-withdrawing ligands, higher lipophilicity and harboring Pt would result in higher T. brucei cytotoxicity. From the whole series of<br />[M(L)(dppf)](PF6) compounds developed, where M = Pt(II) or Pd(II) and HL = 8-hydroxyquinolyl derivatives, Pt-dppf-L4 (IC50 = 0.14 μM, SI = 48) was selected to perform an exploratory pre-clinical study in infected mice. Con el fin de elucidar las vías de señalización med iadas por PknG, recientemente estudiamos su interactoma e identificamos nuevos su stratos de esta quinasa. En el presente trabajo nos centramos en una de las proteí nas fosforiladas por PknG: la proteína FhaA. Esta proteína tiene una estructura q ue se asemeja a las proteínas de andamiaje que participan en vías de señalización de eucariotas, con un dominio N- terminal de función desconocida y un dominio C-term inal FHA que reconoce específicamente de treoninas fosforiladas, ambos unidos por una secue ncia de aproximadamente 300 residuos sin estructura definid a. Como primer paso hacia la elucidación del rol de esta proteína nos propusimos la caracterización a nivel molecular de los complejos de señalización formados in vivo por FhaA, así como sus cambios dinámicos frente a diferentes estímulos que simulen el ambiente del hospedero. La estrategia experimental utilizada se basa en la sobre-expresión de FhaA conteniendo una etiqueta (Strep-tag) en Mycobacterium smegmatis en combinación con el entrecruzamiento de proteínas in vivo , purificación por afinidad y espectrometría de masa para obtener una instantánea de los complejos de señalización. Nuestros resultados nos permitieron i dentificar 56 interactores de FhaA, entre los que se incluye el único interactor previa mente reportado [1]. Los basidiomicetos son un grupo de importancia entre los hongos, pues tienen esencial participación en la naturaleza por la versatilidad de las especies que lo constituyen; unas forman ectomicorrizas, otras son causantes de enfermedades como las royas y los carbones, y se encuentran también las especies comestibles que se cultivan con fines nutritivos. Pertenecen también a este grupo especies con importancia en la medicina por presentar metabolitos con actividad biológica contra una amplia gama de patologías clínicas, incluso influyen positivamente en la profilaxis y el tratamiento del sida. Dentro de este grupo se encuentran, además, los hongos de la podredumbre blanca, únicos microorganismos que son capaces de degradar eficientemente compuestos aromáticos y heterogéneos porque presentan un complejo enzimático inespecífico con actividad oxidativa contra una amplia variedad de sustancias tóxicas y recalcitrantes que les permite aplicaciones como biorremediadores de suelos y aguas contaminadas en las industrias textil y papelera, y tienen también marcada importancia en la agricultura, pues además pueden presentar actividad antifúngica, fitotóxica y nematicida, capacidades útiles para el manejo de plagas.
