There is increasing interest in the development and use of polyphenolic-rich antioxidant extracts as functional ingredients. However, their bitterness, astringency and characteristicflavour could limit their incorporation in foods and beverages. Four alternatives to reduce the bitterness, astringency, and characteristicflavour of antioxidant extracts of two Uruguayan native plants (Achyrocline satureioides and Baccharis trimera) were studied: sucrose, sucralose, polydextrose, and milk. All the evaluated alternatives highly significantly (p &lt; 0.001) reduced the bitterness, astringency and characteristicflavour of the extracts. However, their effectiveness depended on the type and concentration of the antioxidant extract being considered. Milk was the most effective inhibitor of the bitterness, astringency and characteristic flavour of A. satureioidesextracts; whereas when extracts from B. trimera were considered, sucrose was the most effective alternative. Results from the present work suggest that sweetened dairy products could be interesting carriers for the development of functional foods containing polyphenolic-rich antioxidant extracts. A second-order transition state, instead of a minimum, is found at the experimental geometry with a linear hydrogen bond. It collapses to a first-order transition state with a bifurcated hydrogen bond (similar to the ab initio one) from which the true AM1 minimum is obtainable. This structure has a stabilization energy of &minus;5.47 kcal mol^&minus;1 and shows three hydrogen bond interactions. This failure of the method is attributed to an exceedingly weak bending potential of the hydrogen bond within AM1. The conclusions reached in the study of the dimers are further tested by the calculation of a water trimer and a water tetramer and the linear and bifurcated hydrogen bonded complexes between H₂O and NH₄⁺. It is concluded that although it fails in the case of a dimer, AM1 may be useful in the treatment of larger clusters of hydrogen bonded molecules. En Uruguay ocurren entre 50 y 70 accidentes por ofidios al a&ntilde;o, todos causados por las especies Bothrops alternatus y Bothrops pubescens. El veneno de &eacute;stos ofidios es una mezcla compleja de prote&iacute;nas y p&eacute;ptidos biol&oacute;gicos activos cuyo efecto t&oacute;xico se caracteriza principalmente por importantes alteraciones en la coagulaci&oacute;n y da&ntilde;o local pronunciado en el miembro afectado por la mordedura. A los fines m&eacute;dicos, la identificaci&oacute;n de la especie del ofidio es secundaria puesto que la sintomatolog&iacute;a es similar, el protocolo de aplicaci&oacute;n es el mismo para ambos casos e incluye el mismo suero antiof&iacute;dico polivalente o bivalente. Esto tiene considerables ventajas a la hora del tratamiento ya que aunque no se pueda observar la mordedura del ofidio (seg&uacute;n el tipo de terreno a veces solo se siente el dolor), la presencia de sintomatolog&iacute;a y la realizaci&oacute;n de los an&aacute;lisis cl&iacute;nicos correspondientes pueden indicar el inicio del tratamiento, que en todos los casos es adecuado. Por otra parte esta importante ventaja puede inducir al error de considerar ambas especies como la misma y simplificar el accidente a solo una especie, cuando existen diferencias importantes entre las mismas. Seg&uacute;n su peso molecular y ensayos bioqu&iacute;micos realizados las toxinas m&aacute;s abundantes en Bothrops alternatus corresponder&iacute;an a la familia de metaloproteasas, principalmente del tipo PIII. Estas toxinas corresponden a casi 1/3 del total del veneno. Por otra parte el componente m&aacute;s abundante detectado en Bothrops pubescens corresponder&iacute;a a una Fosfolipasa A2 con cerca de 1/3 del total de prote&iacute;nas existente en el veneno. En lo que refiere a la determinaci&oacute;n de veneno en los pacientes, la cantidad promedio encontrada en la sangre de &eacute;stos al ingresar al sanatorio fue de 57 ng/mL, con un rango de 20 a 140 ng/mL. Tanto en Colombia como en Martinica los resultados son similares a los obtenidos en Uruguay. Con relaci&oacute;n a la determinaci&oacute;n de la evoluci&oacute;n de los niveles de suero antiof&iacute;dico en los accidentados se detectan 2 tiempos de eliminaci&oacute;n que sumado a informaci&oacute;n perteneciente a referencia bibliogr&aacute;ficas se puede presumir la existencia de al menos 4 tiempos distintos de eliminaci&oacute;n del suero antiof&iacute;dico: el de distribuci&oacute;n corporal, el de neutralizaci&oacute;n del veneno existente en sangre, el de neutralizaci&oacute;n del veneno que difunde de los tejidos, y el de eliminaci&oacute;n del suero antiof&iacute;dico excedente. Por otra parte se pudo determinar la respuesta contra el suero antiof&iacute;dico. &Eacute;ste genera una respuesta humoral en IgM e IgG contra las inmunoglobulinas de caballo en la mayor&iacute;a de los pacientes a los cuales se les administra. La respuesta var&iacute;a en un incremento del t&iacute;tulo de anticuerpos de 4 a m&aacute;s de 100 veces. Adem&aacute;s existe una relaci&oacute;n entre el t&iacute;tulo de anticuerpos anti suero antiof&iacute;dico alcanzado en pacientes y la cantidad de viales de suero antiof&iacute;dico administrado. El veneno of&iacute;dico tambi&eacute;n genera una respuesta humoral pero de menor proporci&oacute;n que el suero antiof&iacute;dico y principalmente de tipo IgM, a los 15 d&iacute;as. La respuesta humoral frente al veneno es contra la mayor&iacute;a de, o todos, los componentes del veneno, detect&aacute;ndose reactividad cruzada entre ambos venenos en los anticuerpos desarrollados contra el veneno de una especie. Adem&aacute;s del sistema de obtenci&oacute;n de agua, el almacenamiento debe hacerse de forma controlada, y el estancamiento debe ser el menor posible. A los efectos de la realizaci&oacute;n del siguiente trabajo se seleccion&oacute; como modelo un sistema de obtenci&oacute;n de agua de un centro de di&aacute;lisis, que es similar al de obtenci&oacute;n de agua purificada para uso farmac&eacute;utico y que est&aacute; regulada por Farmacopeas. La primera parte del trabajo consisti&oacute; en el estudio de la situaci&oacute;n del sistema mediante la utilizaci&oacute;n de m&eacute;todos cl&aacute;sicos de cultivo, se estudiaron las bacteri presentes en el sistema de agua as&iacute; como tambi&eacute;n la situaci&oacute;n de todos los puntos involucrados en el proceso de hemodi&aacute;lisis ( izador, l&iacute;quido de di&aacute;lisis y concentrados para hemodi&aacute;lisis) cuando ocurrieron c hos en pacientes (episodios de presunta endotoxemia) durante la di&aacute;lisis. La novedad de la propuesta es que se emplearon simult&aacute;neamente m&eacute;todos cl&aacute;sicos de cultivo, siguiendo las normativas establecidas por organismos nacionales e internacionales, junto con otros m&eacute;todos de an&aacute;lisis como son t&eacute;cnicas moleculares y citometr&iacute;a de flujo. Se espera mejorar la sensibilidad de los m&eacute;todos de detecci&oacute;n por m&eacute;todos de concentraci&oacute;n, y PCR en tempo real de microorganismos de importancia cl&iacute;nica como . Con los resultados obtenidos fue posible responder a preguntas tales como: 1) &iquest;En qu&eacute; medida se subestiman los microorganismos cultivables cuando se emplea PCA o TSA en lugar de R2A en un sistema de tratamiento de agua particular? 2) &iquest;Tienen los microorganismos aislados del agua de entra y salida del monitor despu&eacute;s de producido un chucho en un paciente durante la di&aacute;lisis, una capacidad de liberaci&oacute;n endotoxinas mayor que otros microorganismos aislados del sistema en otras oportunidades? 3) &iquest;Son eficientes los procesos de desinfecci&oacute;n establecidos para el sistema de agua? &iquest;Y los sistemas empleados para la conservaci&oacute;n de los dializadores? La presente investigaci&oacute;n propone analizar las concepciones de ciencia y de cient&iacute;fico que tienen estudiantes y docentes universitarios del &Aacute;rea Cient&iacute;fico- Tecnol&oacute;gica de la Universidad de la Rep&uacute;blica. El inter&eacute;s de la did&aacute;ctica de las ciencias naturales por las llamadas metaciencias, es decir por la epistemolog&iacute;a, la historia de la ciencia y la sociolog&iacute;a de la ciencia, proviene de un reconocimiento consensuado de que ellas pueden contribuir de maneras muy diversas y potentes a la ense&ntilde;anza de las ciencias naturales en las aulas de los distintos niveles educativos. Profundizar en el estudio de las ideas de los docentes y estudiantes permitir&aacute; posteriormente reflexionar acerca de ellas, buscando contribuir al desarrollo y a la mejora del aprendizaje de las ciencias en todos los niveles. El trabajo de investigaci&oacute;n se inscribe en una modalidad descriptiva-explicativa con un abordaje cuali - cuantitativo. Se aplican cuestionarios de respuesta cerrada y abierta, se realizan entrevistas y observaciones de clase no participantes. Dentro de la poblaci&oacute;n objeto de este estudio se encuentran docentes de diferentes grados universitarios y estudiantes tanto ingresantes como avanzados pertenecientes a las Facultades de Ciencias, Ingenier&iacute;a y Qu&iacute;mica de la Universidad de la Rep&uacute;blica. Se analizan los datos a partir de tres dimensiones principales: la imagen de ciencia, la imagen del cient&iacute;fico y el conocimiento sobre personalidades vinculadas a la actividad cient&iacute;fica de nuestro pa&iacute;s. A trav&eacute;s de esta investigaci&oacute;n se constata que los estudiantes ingresantes a las Facultades del &Aacute;rea Cient&iacute;fico &ndash; Tecnol&oacute;gica tienen visiones sobre la ciencia que se alejan de las concepciones actuales sobre la naturaleza de la ciencia afirmando que la ciencia es una actividad neutral y que la experimentaci&oacute;n es la base fundamental del conocimiento cient&iacute;fico. Muestra tambi&eacute;n que tanto los estudiantes avanzados como los docentes manifiestan que la ciencia no es una actividad neutra y que no se puede hablar de verdades absolutas ni de m&eacute;todos &uacute;nicos. Sin embargo es a partir de las entrevistas y las observaciones de clases donde se constata que, a pesar de explicitar una visi&oacute;n de ciencia m&aacute;s alineada con las teor&iacute;as actuales, los docentes transmiten en el aula una concepci&oacute;n de la actividad cient&iacute;fica que es contrapuesta a lo manifestado expl&iacute;citamente. Los resultados muestran que a trav&eacute;s de la ense&ntilde;anza se est&aacute; transmitiendo una imagen de ciencia y del trabajo cient&iacute;fico que no concuerda con las visiones actuales acerca de la naturaleza de la ciencia. Se presentan las teor&iacute;as cient&iacute;ficas sin ninguna conexi&oacute;n con los problemas que trataban de resolver en el momento de su surgimiento, se promueve una visi&oacute;n inductivista del trabajo cient&iacute;fico en la que las observaciones anteceden a las hip&oacute;tesis te&oacute;ricas, se transmite una concepci&oacute;n simplista de la experimentaci&oacute;n en la que existe una &uacute;nica interpretaci&oacute;n posible para los resultados. Este diagn&oacute;stico evidencia la necesidad de la formaci&oacute;n epistemol&oacute;gica del profesorado de ciencias y puede brindar nuevas orientaciones curriculares que pongan el conocimiento acerca de la naturaleza de la ciencia en un lugar central de la educaci&oacute;n cient&iacute;fica general. Due to their biological properties, these molecules have been considered useful for drug development, as novel agents in disease therapeutics, applicable to both agriculture and medicine. New technologies of massive sequencing open opportunities to discover novel AMP encoding genes in wild plant species. This work aimed to identify cysteine-rich AMPs from Peltophorum dubium, a legume tree from South America. We performed whole-transcriptome sequencing of P. dubium seedlings followed by de novo transcriptome assembly, uncovering 78 AMP transcripts classified into five families: hevein-like, lipid-transfer proteins (LTPs), alpha hairpinins, defensins, and snakin/GASA (Giberellic Acid Stimulated in Arabidopsis) peptides.<br />No transcripts with similarity to cyclotide or thionin genes were identified. Genomic DNA analysis by PCR confirmed the presence of 18 genes encoding six putative defensins and 12 snakin/GASA peptides and allowed the characterization of their exon-intron structure. The present work demonstrates that AMP prediction from a wild species is possible using RNA sequencing and de novo transcriptome assembly, regarding a starting point for studies focused on AMP gene evolution and expression. El trabajo que se describe a continuaci&oacute;n surge como parte de un proyecto de investigaci&oacute;n de s&iacute;ntesis de bengamidas y an&aacute;logos, el cual se desarrolla en la C&aacute;tedra de Qu&iacute;mica Org&aacute;nica de la Facultad de Qu&iacute;mica, bajo la direcci&oacute;n el Dr. Gustavo Seoane. Dicho grupo de investigaci&oacute;n hace uso de una cepa mutante de F39/D, para obtener sintones quirales a partir de compuestos arom&aacute;ticos diversos, los cuales se utilizan como material de partida en s&iacute;ntesis asim&eacute;tricas de compuestos naturales con actividad biol&oacute;gica. Las bengamidas son una clase de productos naturales ma inos que se aislaron originariamente en 1986 de esponjas del g&eacute;nero recolectadas cerca de las Islas Fiji. Est&aacute;n formadas por un n&uacute;cleo central de 7 miembros, correspondiente a una - caprolactama, al que se unen una cadena polioxigenada tambi&eacute;n, en la mayor&iacute;a de los casos, una cadena lipof&iacute;lica correspondiente a un &aacute;cid graso saturado que esterifica a la mol&eacute;cula. Estos compuestos, que fueron aislados como parte de un programa de b&uacute;squeda de metabolitos marinos con actividad antiparasitaria, muestran una muy interesante actividad antihelm&iacute;ntica y citot&oacute;xica. En consecuencia, existe u gran inter&eacute;s en su preparaci&oacute;n, ya que la opci&oacute;n de extracci&oacute;n a partir de esponjas produ cantidades limitadas y discontinuas. La alta oxigenaci&oacute;n de la cadena lateral de las bengamidas y la funcionalizaci&oacute;n del anillo central los hace buenos candidatos para intentar su s&iacute;ntesis usando la metodolog&iacute;a de biotransformaci&oacute;n de arenos. En el presente trabajo se describen cuatro rutas de aproximaci&oacute;n a la s&iacute;ntesis del anillo central a partir de halo-dihidrodioles quirales como &uacute;nico sint&oacute;n de partida; se discute la apertura nucleof&iacute;lica de a-halooxiranos quirales derivados de ; y se presentan los resultados de evaluaci&oacute;n biol&oacute;gica (contra helmintos) una serie de intermedios sint&eacute;ticos. The aim of this work was to perform a groundwater quality monitoring in Uruguay, from the Medical Geology perspective, focusing attention on arsenic levels and its correlations with other relevant inorganic parameters. For this purpose, a total of 46 groundwater samples from private wells, out of the scope of the state-run water utility company, were analyzed. The accuracy of the analytical methods was ensured by using certified reference materials. Arsenic concentration range was 1.72&ndash;120.5 &mu;g L&minus;1, half of the samples being above the limit of 10 &mu;g L&minus;1 recommended by WHO for drinking water, with the corresponding risks for human health. Pearson correlations were performed, resulting in strong positive correlations for the pairs As/Cl&minus;, As/F&minus;, As/Na and As/V.
