<p>The addition of free radicals and the 1,3 dipolar cycloaddition onto pristine and <strong>lithium-doped</strong> C60 were studied by means of the Perdew–Burke–Ernzerhof (PBE) and M06-2X density functionals. In all cases, <strong>lithium</strong> increased the reactivity even though for the 1,3 dipolar cycloaddition onto C60 the change observed with respect to bare C60 was minimal. Both functionals employed gave similar encapsulation energies for Li@C60 namely, 33.1 and 38.2kcal/ mol at the PBE/6-31G* and M06-2X/6-31G*, respectively. However, the increased reactivity because of lithium doping determined at the PBE level is smaller as compared with that computed with the M06-2X functional, whereas that determined at the second-order Møller–Plesset (MP2) level is the largest one. For example, using the M06-2X functional the binding energy of fluorine to Li@C60 is 28.5kcal/mol larger than that determined for C60, whereas at the PBE/6-31G* level it is predicted to be increased by 24.7kcal/mol. The results clearly suggest that Li@C60 is a much better free radical scavenger than C60. Finally, the complex hindered rotations of <strong>lithium</strong> inside C60 are expected to be strongly inhibited because lithium doping increases the well depth between the cage center and the equilibrium position near the addition site of the lithium atom.</p>

<p>(Acetonitrile-1κN)[μ-1H-benzimidazole-2(3H)-thione-1:2κ2 S:S][1H-benzimidazole-2(3H)-thione-2κS]bis(μ-1,1-dioxo-1λ6,2-benzothiazole-3-thiolato-1:2κ2S3:N;1:2κ2 S3:S3)-dicopper(I)(Cu-Cu), [Cu2(C7H4NO2S2)2(C7H6N2S)2(CH3CN)] or [Cu2(tsac)2(Sbim)2(CH3CN)] [tsac is thiosaccharinate and Sbim is 1H-benzimidazole-2(3H)-thione], (1), is a new copper(I) compd. that consists of a triply bridged dinuclear Cu-Cu unit. In the complex mol., two tsac anions and one neutral Sbim ligand bind the metals. One anion bridges via the endocyclic N and exocyclic S atoms (μ-S:N). The other anion and one of the mercaptobenzimidazole mols. bridge the metals through their exocyclic S atoms (μ-S:S). The 2nd Sbim ligand coordinates in a monodentate fashion (κS) to one Cu atom, while an acetonitrile mol. coordinates to the other Cu atom. The CuI-CuI distance [2.6286(6) Å] can be considered a strong cuprophilic interaction. In the case of [μ-1H-benzimidazole-2(3H)-thione-1:2κ2 S:S]bis[1H-benzimidazole-2(3H)-thione]-1κS;2κS-bis(μ-1,1-dioxo-1λ6,2-benzothiazole-3-thiolato)-1:2κ2 S3:N;1:2κ2S3:S3-dicopper(I) (Cu-Cu), [Cu2(C7H4NO2S2)2(C7H6N2S)3] or [Cu2(tsac)2(Sbim)3] (2), the acetonitrile mol. is substituted by an addnl. Sbim ligand, which binds one Cu atom via the exocylic S atom. In this case, the CuI-CuI distance is 2.606</p>

La mitocondria es un el sitio principal de generacion de superoxido y otras especies reactivas y es uno de los principales sitios de formacion de 3-nitrotirosina proteica tanto en condiciones basales como patologicas. En este trabajo se estudiaron dos proteinas mitocondriales, el citocromo c y la superoxido dismutasa de manganese (MnSOD) y de cobre y cinc (CuZnSOD), estudiando tanto la nitracion de estas proteinas como cambios estructurales en condiciones similares alas encontradas in vivo. En el caso del citocromo c se estudio la nitracion de tirosinas proteicas evaluando la presencia de un lipide mitocondrial, la cardiolipina. En los experimentos realizados se observo por diversas tecnicas que en presencia de este Iipido la nitracion de citocromo c expuesto a peroxinitrito se encuentra aumentada, siendo este aumento mediado por un cambio conformacional mediado por la cardiolipina. A nivel estructural, se observaron los cambios sufrido por citocromo 0 en presencia de cardiolipina por tecnicas de NMR para y diamagnetico. Estos experimentos, si bien no fueron concluyentes mostraron que la proteina sufre un cambio conformacional global. Estudios de la reactividad de cada tirosina de citocromo c utilizando mutantes mostro que, en presencia de cardiolipina, tirosinas que no son preferencialmente nitradas 0 modificadas en ausencia de este Iipido estan involucradas en procesos oxidativos tales como la actividad peroxidasa de citocromo c mediada por cardiolipina. Esta actividad peroxidasa se estudio tam bien utilizando un derivado carboximetilado en me\ionina de ci\ocmmo c que, a di~efencia de\ ci\ocmmo c sin modi~icaf pefmi\e \a deteccion de un Compuesto I de peroxidasa. EI agregado de cardiolipina a citocromo c carboximetilado expuesto a peroxido de hidrogeno aumenta la formacion de compuesto I de citocromo c, y por 10 tanto su actividad peroxidasa. Para profundizar el estudio de los cambios estructurales sufridos por citocromo c frente a diferentes oxidantes se sintetizo y purifico citocromo c mononitrado a nivel de Tyr 97 y 74, para generar anticuerpos monoclonales contra formas conformacionalmente modificadas de esta proteina que permitan la deteccion de las mismas tanto en estudios in vitro como in vivo En el caso de las superoxido dismutasas (SOD) se estudiaron las modificaciones sufridas en presencia de flujos de oxido nitrico y superoxido evaluando la capacidad de esta enzima de competir con el oxido nitrico por su sustrato, observandose que en presencia de estos flujos se forma peroxinitrito en concentraciones capaces de inactivar a la enzima y en el caso de MnSOD capaz de nitrarla. Por otra parte se reeestudio la reaccion de MnSOD con peroxinitrito determinando un nuevo valor para la constante de reaccion de 1,9 x 104 M·1s·1 por tetramero. A nivel estructural se estudio el efecto de la nitracion en la accesibilidad del sustrato al sitio activo utilizando metodos in silico donde se determino que la nitracion de Tyr 34, responsable de la inactivacion de MnSOD por peroxinitrito genera un aumento en la barrera de energia libre para el acceso del superoxido al sitio activo, 10 cual impide el acceso al sustrato, tanto por repulsion esterica como electrostatica. Tambien a nivel estructural, se determino el efecto de mutaciones puntuales de MnSOD en la estructura cuaternaria de la enzima, evaluando, a su vez, el efecto de estas mutaciones en la sensibilidad de sus tioles. ----- No se encontraron virus en este mensaje. A summary of the resource.
<p>The goal of the present work was to establish the essential oil yields and chemical compositions of nine Basil (Ocimum basilicum L.) cultivars. The plants were grown in the fields, in an averaged experimental design, using ten plants of each of the nine cultivars in each parcel, making three repeats of each experiment. The parameters measured were fresh and dry weight of the aerial parts, yield and chemical compositions of the essential oils. The yields ranged from 6.21 to 42.51 L/ha, the Genovese cultivar being the most productive. The chemical composition of all cultivars had high contents of linalool, with the highest being that of the Toscano "lettuce leave" cultivar.</p>

<h3><span style="text-decoration: underline;"><strong>Resumen</strong></span></h3>
<p>La temática de las <strong>Plantas Medicinales</strong>, es motivo de creciente interés a nivel nacional y mundial. Los Químicos y botánicos de Nuestra Casa de Estudios, se han interesado en el tema, realizando investigaciones científicas, con resultados valiosos y útiles. El Departamento de Documentación y Biblioteca, ha querido brindar su colaboración en este sentido, en la creación de un <strong>“Tesauro sobre Plantas Medicinales”</strong> de la región. Es un vocabulario controlado y estructurado, para el tratamiento normalizado de la información sobre el tema. El Tesauro, constituye un elemento indispensable para todo Centro de Información con la finalidad de indizar documentos sobre el tema. Su importancia deriva, en que los autores, los documentalístas y los usuarios, utilizan terminos diferentes para disignar un mismo concepto. Más concretamente sirve para crear un lenguaje normalizado de uso universal. El Tesauro sobre Plantas Medicinales de la <strong>Facultad de Química</strong>, no es un diccionario ni una enciclopedia, sino un intrumento de trabajo documental, que permite asegurar la coincidencia entre los diferentes lenguajes empleados por los emisores y receptores de la información, pudiendo así acceder rápidamente a la información. Uno de los objetivos de este grupo de trabajo multidisciplinario, es una continua adaptación y puesta al día del Tesauro, contando muy especialmente con la sugerencia de los usuarios, para mejorar el producto, en una sociedad dinámica y en constante evolución.</p>

<p>The first syntheses of two deoxythiocyanocyclitols (4-deoxy-4-thiocyano-L-chiro-inositol and deoxythiocyanoconduritol F) and two deoxysulfonylcyclitol acetals are reported by a chemoenzymatic enantioselective route. The compounds were prepared by a sequence of enzymatic and ruthenium-catalyzed dihydroxylations, and the results were studied regarding reaction conditions and co-catalyst for different derivatives. The new compounds were included in a minilibrary of deoxygenated cyclitols and evaluated for their capacity to influence the feeding behavior of Epilachna paenulata (Coleoptera: Coccinellidae), a common pest of the Curcubitaceae crops.</p>

<p>The first synthesis of two fagopyritol β-analogues (β-d-galactopyranosyl-(1′→1)-conduramine F-4 and β-d-galactopyranosyl-(1′→3)-4-aminodeoxy-l-chiro-inositol) has been accomplished by a chemoenzymatic route in satisfactory yields. The key step of the synthesis is the TMSOTf-promoted glycosylation reaction of a deoxyconduritol derivative. The methodology is amenable to scale-up and expandable to the preparation of other pseudofagopyritols.</p>

<p>The first synthesis of two selenyldeoxycyclitols (4-bromo-2-phenylselenyl conduritol F and 6-phenylselenylconduritol F) is reported via a chemoenzymatic enantioselective route. The key step of the synthesis is the selenolysis of a vinyl epoxide. The new compounds were evaluated for their capacity to inhibit the growth of different microorganisms using a modification of the agar diffusion technique with thin layer chromatography plates as support.</p>

En segundo lugar se presentan los antecedentes, el uso de soportes audiovisuales en la enseñanza en general, con énfasis en el uso de los mismos en la enseñanza de la química. La parte empírica se compone de dos secciones. La primera sección corresponde a la investigación llevada a cabo con el objetivo de analizar las relaciones entre el rendimiento en Química de los estudiantes liceales y el uso de soportes audiovisuales como medio de instrucción. Para ello se realizaron dos estudios en 2007 y 2008, de tipo cuasiexperimental dado que no fue posible asignar los sujetos aleatoriamente a los grupos de trabajo, sino que se trabajó con los grupos existentes. En ambos se trabajó con dos grupos de estudiantes que presentaban características sociodemográficas similares. En un grupo se estudió una unidad programática aplicando una metodología tradicional, utilizando principalmente información extraída de textos en español, mientras que con otro grupo se estudió el mismo tema, pero facilitando el acceso al mismo a partir de material audiovisual, elaboración de software, etc. Como medida del rendimiento en Química se adoptó la calificación obtenida por los alumnos en diferentes pruebas de conocimiento. Para validar las conclusiones se realizó el control de las variables que podían afectar el rendimiento (motivación, rendimiento previo, estrategias de estudio, antecedentes educativos de los padres, etc.). En la segunda sección se detalla cómo se llevó a cabo la producción de un material audiovisual (Video ). El material audiovisual fue producido en el marco de la pasantía que formó parte del plan de trabajo de la tesista. Dicha pasantía fue realizada en el Centro de Educación Flexible (C.E.F.) de la Facultad de Química entre los meses de diciembre de 2006 y noviembre de 2007. Por último, con la finalidad de recolectar datos que permitieran interpretar el resultado del uso del video producido en el C.E.F., se realizó una evaluación a un grupo de tercer año de Ciclo Básico que trabajó con dicho material audiovisual. Para la selección de los indicadores de rendimiento se adoptó un enfoque conductual eligiendo medidas de rendimiento centradas en los resultados de las actividades de evaluación escritas realizadas por los estudiantes (Algarabel y Dasi, 2001). Lo anterior no significa que no se le haya otorgado importancia a los procesos cognitivos de integración y estructuración de la información mediante los cuales se lograron dichos resultados, sino solamente que en este trabajo se pone el énfasis en éstos. Si bien la metodología de investigación no permite establecer relaciones de causalidad, los resultados obtenidos son consistentes con la hipótesis de que el uso de soportes audiovisuales mejora el rendimiento de los alumnos en Química. Como limitaciones se mencionan el haber trabajado con muestras de conveniencia de tamaño reducido, lo que no permite la generalización de estos hallazgos a otras poblaciones de estudiantes, así como el tiempo estipulado para la toma de datos, que impidió trabajar con un mayor número y tipo de apoyos audiovisuales. Estos resultados pueden ser tomados como punto de partida para nuevas investigaciones que puedan llevarse a cabo utilizando otros instrumentos así como también realizando otro tipo de tratamiento de los datos obtenidos.

<p>In the search for new therapeutic tools against Chagas' disease (American Trypanosomiasis) four novel mixed-ligand vanadyl complexes, [V(IV)O(L(2)-2H)(L(1))], including a bidentate polypyridyl DNA intercalator (L(1)) and a tridentate salycylaldehyde semicarbazone derivative (L(2)) as ligands were synthesized, characterized by a combination of techniques, and in vitro evaluated. EPR suggest a distorted octahedral geometry with the tridentate semicarbazone occupying three equatorial positions and the polypyridyl ligand coordinated in an equatorial/axial mode. Both complexes including dipyrido[3,2-a: 2',3'-c]phenazine (dppz) as polypyridyl coligand showed IC(50) values in the muM range against Dm28c strain (epimastigotes) of Trypanosoma cruzi, causative agent of the disease, being as active as the anti-trypanosomal reference drug Nifurtimox. To get an insight into the trypanocidal mechanism of action of these compounds, DNA was evaluated as a potential parasite target and EPR, and (51)V NMR experiments were also carried out upon aging aerated solutions of the complexes. Data obtained by electrophoretic analysis suggest that the mechanism of action of these complexes could include DNA interactions.</p>

<p>In the search for new metal-based drugs for the treatment of tumoral and parasitic diseases a vanadyl complex, [V(IV)O(SO(4))(H2O)(2)(dppz)].2H(2)O, that includes the bidentate polypyridyl DNA intercalator dipyrido[3,2-a:2',3'-c]phenazine (dppz), was synthesized, characterized by a combination of techniques, and in vitro evaluated on the human acute promyelocytic leukemia cell line HL-60 and against Dm28c strain epimastigotes of the parasite <strong>Trypanosoma cruzi</strong>, causative agent of Chagas' disease. EPR spectroscopy suggests a distorted octahedral geometry for the complex with the dppz ligand acting as bidentate, binding through both nitrogen donor atoms in an axial-equatorial mode. An oxo group, two water molecules and a sulphate donor occupy the remainder coordination positions. The complex, as well as the anti-trypanosomal reference drug Nifurtimox, showed IC(50) values in the muM range against T. cruzi Dm28c strain. In addition the complex exhibited excellent in vitro anti-tumor activity against leukemia (HL-60 cell line) comparable to that of cisplatin, inducing cell death by apoptosis with IC(50) values in the micromolar range. Data from gel electrophoresis and atomic force microscopy indicate that the complex interacts with DNA, suggesting that its mechanism of action may include DNA as a target. EPR and (51)V NMR experiments were also carried out with aged aerated solutions of the complex to get insight into the stability of the complex in solution and the species responsible for the in vitro activities observed.</p>

<p>Insect pheromone research has a long and rich history built up primarily by studies conducted in the Northern hemisphere. Not surprisingly, these studies have largely targeted species relevant to these regions of the world, for the most part agricultural and forest pests. Pheromone research in South American countries came a few decades behind, albeit their strong dependence in agriculture and therefore in pest management. In the last 20 years, a combination of economic, environmental and technical factors have come together to generate a small but rising number of chemical ecologists working in pheromone chemistry and biology in South America. In this article we summarize the results of this trend. We review pheromone studies in which South American scientists have participated in collaboration with foreign scientists, mostly chemistry groups, as well as the ever-growing number of studies carried out completely within the region. We have focused mainly in research involving the characterization of pheromones from native species, which involve the most important insect orders, namely Lepidoptera, Coleoptera, Heteroptera and Hymenoptera. We also present a simple meta-analysis including geographical distribution, trends in collaborative or independent work, and a 20-year evolution of published articles in the field. Finally, we emphasize the importance of a coordinated effort to further promote the growth of this field in Latin America, through the endorsement of international collaborations within the region. Such goal would be facilitated by a regional academic organization, which would in turn initiate the occurrence of regular scientific meetings.</p>

<p>Background: All-trans retinoic acid (atRA) plays an essential role in the regulation of gene expression, cell growth and differentiation and is also important for normal cardiovascular development but may in turn be involved in cardiovascular diseases, i.e. atherosclerosis and restenosis. The cellular atRA levels are under strict control involving several cytochromes P450 isoforms (CYPs). CYP26 may be the most important regulator of atRA catabolism in vascular cells. The present study describes the molecular cloning, characterization and function of atRA-induced expression of a spliced variant of the CYP26B1 gene.<br />Methodology/Principal Findings: The coding region of the spliced CYP26B1 lacking exon 2 was amplified from cDNA synthesized from atRA-treated human aortic smooth muscle cells and sequenced. Both the spliced variant and full length CYP26B1 was found to be expressed in cultured human endothelial and smooth muscle cells, and in normal and atherosclerotic vessel. atRA induced both variants of CYP26B1 in cultured vascular cells. Furthermore, the levels of spliced mRNA transcript were 4.5 times higher in the atherosclerotic lesion compared to normal arteries and the expression in the lesions was increased 20-fold upon atRA treatment. The spliced CYP26B1 still has the capability to degrade atRA, but at an initial rate one-third that of the corresponding full length enzyme. Transfection of COS-1 and THP-1 cells with the CYP26B1 spliced variant indicated either an increase or a decrease in the catabolism of atRA, probably depending on the expression<br />of other atRA catabolizing enzymes in the cells.<br />Conclusions/Significance: Vascular cells express the spliced variant of CYP26B1 lacking exon 2 and it is also increased in atherosclerotic lesions. <p>Synthesis and cytotoxicity of imidazo[5,4-f]benzimidazolequinones and iminoquinone derivatives is described, enabling structure–activity relationships to be obtained. The most promising compound (an iminoquinone derivative) has undergone National Cancer Institute (NCI) 60 cell line (single and five dose) screening, and using the NCI COMPARE program, has shown correlation to NQO1 activity and to other NQO1 substrates. Common structural features suggest that the iminoquinone moiety is significant with regard to NQO1 specificity. Computational docking into the active site of NQO1 was performed, and the first comprehensive mitomycin C (MMC)-NQO1 docking study is presented. Small distances for hydride reduction and high binding affinities are characteristic of MMC and of iminoquinones showing correlations with NQO1 via COMPARE analysis. Docking also indicated that the presence of a substituent capable of hydrogen bonding to the His194 residue is important in influencing the orientation of the substrate in the NQO1 active site, leading to more efficient reduction.</p>

Estudio genómico comparativo de las cepas de Ralstonia solanacearum causantes de la marchitez bacteriana de la papa en Uruguay Ralstonia solanacearum es el agente responsable de la marchitez bacteriana, una de las enfermedades de plantas más destructivas a nivel mundial. En Uruguay este patógeno constituye uno de los principales problemas sanitarios asociados al cultivo de papa. El objetivo general de esta tesis es generar conocimiento sobre las cepas de R. solanacearum que afectan los cultivos de papa en Uruguay, de forma de contribuir al desarrollo de estrategias efectivas para su control. Para ello, se generó una colección de cepas uruguayas de R. solanacearum encontrando que todas pertenecen al filotipo IIB, secuevar 1 (IIB-1), un grupo especialmente adaptado a la papa como hospedero y que se asocia a climas templados. Se encontraron diferencias de agresividad entre las cepas sobre varios hospederos incluyendo tomate, papa y la especie silvestre Solanum commersonii, una especie silvestre nativa de Uruguay considerada una valiosa fuente de resistencia en programas de mejoramiento de papa. La diversidad genética se evaluó mediante marcadores BOX-PCR y por hibridación genómica comparativa (CGH) usando la tecnología de microarrays. Globalmente estos análisis revelaron una baja diversidad genética entre las cepas presentes en Uruguay y confirmaron su clasificación como filotipo IIB, secuevar 1. El análisis genómico comparativo por CGH permitió establecer el contenido genético de cada una de las cepas uruguayas analizadas. Esta información se analizó con dos enfoques diferentes. Por un lado, se comparó el contenido genético entre cepas con diferente nivel de agresividad u origen de aislamiento, identificando genes involucrados en la virulencia de R. solanacearum. Por otro lado, los datos generados se utilizaron para establecer una lista de genes específicos de cepas IIB-1, los cuales se aplicaron al desarrollo de métodos de detección específicos que permitan evitar la diseminación del patógeno. Los resultados generados en esta tesis contribuyen a aumentar el valor biológico de los datos de secuencia actualmente disponibles y apuntan a su aprovechamiento para el desarrollo de estrategias de control de este importante patógeno.

El presente trabajo de tesis se enmarca dentro del estudio de la estructura de diferentes complejos entre una proteína de interés farmacológico y ligandos, y el análisis de las consecuencias que dichos modelos estructurales tienen sobre los mecanismos tanto de inhibición como catalítico. En particular, este trabajo apunta hacia el refinamiento a resolución mediana de la enzima humana PTP1B y, más específicamente, al estudio estructural por cristalografía y modelización del movimiento del lazo flexible WPD de esta enzima frente a la presencia de diferentes iones halógenos. La proteína de interés fue co-cristalizada en diferentes concentraciones de buffer acetato y de iones halógeno (Br – y I -). Estos cristales fueron medidos en fuentes de radiación sincrotrónica, y las estructuras tridimensionales de los complejos fueron determinadas. Estos datos experimentales permitieron comprobar que en los complejos formados por la proteína y el ion bromuro el lazo WPD fue encontrado tanto en la conformación cerrada como la abierta, mientras que en los complejos con el ion ioduro sólo se obtuvieron estructuras en las que el mencionado lazo se encontraba en conformación cerrada. Esto estaría indicando una relación entre la identidad del ion presente en el complejo y la fuerza de las interacciones establecidas entre éste y los residuos proteicos circundantes. Los estudios cristalográficos también permitieron determinar una correlación entre la concentración de haluro presente en la mezcla en la que el cristal fue crecido y laconformación del lazo en estudio en dicho cristal. A mayor concentración del haluro, mayor es la ocupación tanto del ion como de la conformación cerrada del lazo. Con respecto a esto, se observó que cuando la solución presentaba concentraciones elevadas tanto de acetato como del haluro, el efecto del primero prevalecía, y la ocupación del haluro disminuía. En busca de una correlación entre el tipo de ion presente en el complejo y la ocupación relativa de cada uno de los iones presentes, se llevó a cabo una serie de estudios teóricos, empleando diferentes técnicas y niveles de teoría. Los movimientos colectivos de la enzima PTP1B en complejo con los dos haluros considerados fueron estudiados a través de una simulación de Dinámica Molecular, mpleando el módulo SANDER del programa AMBER10. La información obtenida respecto al movimiento del mencionado lazo en ambos complejos fue consistente con las observaciones experimentales obtenidas en este mismo trabajo: mientras que el complejo proteína-bromuro presenta un cambio conformacional a lo largo de la simulación, esto no se observó para el complejo proteína-ioduro. Esto apoya la teoría previamente planteada de la existencia de interacciones diferenciadas entre los dos haluros y los residuos circundantes. Para lograr una mejor comprensión de las interacciones actuantes entre ambos iones y los residuos proteicos de la región catalítica, se están llevando a cabo optimizaciones de geometría para cuatro de los complejos (PTP1B-bromuro con el lazo en conformación cerrada, el mismo complejo con el lazo en la conformación abierta, PTP1B-ioduro en conformación abierta y el mismo complejo en conformación cerrada). Estos cálculos se están llevando a cabo empleando el modelo multicapa ONIOM del programa Gaussian09, el cual nos permite estudiar las partes más relevantes de la estructura a un nivel de funcionales de la densidad, empleando el funcional M06, y empleando la base DGDZV para representar los orbitales de Kohn-Sham de ambos iones, y la base 6-31G* para representar los residuos proteicos del sitio catalítico. El resto de la proteína se está estudiando a nivel de Mecánica Molecular, empleando el campo de fuerza AMBER para representar las interacciones existentes. Una etapa crítica para poder emplear métodos híbridos es la correcta determinación de la región a estudiar al nivel más elevado de teoría (DFT). En este trabajo se presenta la metodología empleada para lograrlo, y se deja constancia de los residuos que se consideran dentro de la región más relevante de la estructura. El presente trabajo muestra que ciertas interacciones entre los residuos del sitio catalítico y el ligando presente en el complejo serían los responsables de la conformación adoptada por el lazo flexible WPD de la proteína PTP1B. Una vez se terminen los estudios híbridos y se logre determinar cuáles son los principales residuos catalíticos, se tendrá un punto de partida confiable para el diseño costo-efectivo de posibles inhibidores específicos para esta enzima.

Abstract

La madera, material natural degradable es susceptible a diversos agentes<br />bióticos y abióticos de deterioro. Microorganismos e insectos constituyen los<br />agentes bióticos; dentro de los primeros, los hongos de la podredumbre blanca<br />de la madera poseen la habilidad de mineralizar completamente el polímero de<br />lignina, segundo componente estructural más abundante de la madera. Esta<br />potencialidad está basada en su sistema enzimático extracelular compuesto<br />por diferentes tipos de enzimas (peroxidasas y oxidasas) y compuestos de bajo<br />peso molecular. En consecuencia la madera debe ser protegida a través de<br />tratamientos con productos efectivos, habiendo una tendencia mundial al<br />desarrollo de productos alternativos de menor toxicidad que los existentes.<br />El presente trabajo plantea al uso de las sales de Zn y Mn como conservantes<br />para maderas. Se estudia su eficacia antifúngica, empleando tanto sales puras<br />como aquellas provenientes de un proceso de reciclado de pilas. Dichas sales<br />han sido utilizadas a nivel industrial, interrumpiéndo u uso en 1930 debido a<br />razones básicamente comerciales. Se plantea su aplicac ido al<br />desarrollo en la Unidad de Gestión Tecnológica de la Facultad de Química de<br />un proceso por vía hidrometalúrgica de reciclado de pi comunes y alcalinas.<br />El mismo es simple, de bajo costo y permite el aprovechamiento de los<br />materiales que contienen a la vez que evita la posible contaminación que<br />provoca su eliminación. El producto principal del proceso es una solución rica<br />en Zn y Mn.<br />Se evalúa el efecto de las mencionadas soluciones sobre la fisiología de<br />dos especies<br />representativas de los hongos causantes de podredumbre blanca en la madera,<br />que poseen diferente sistema ligninolítico. Se estudia la influencia de dichos<br />metales en la fisiología de ambos hongos, con dos estrategias. Una global que<br />analiza el efecto sobre el crecimiento (desarrollomicelial y germinación de<br />conidias) y una más específica que analiza la función inolítica (actividad<br />enzimática de las peroxidasas y expresión de los genes que codifican<br />manganeso peroxidasas). Para este último punto se iden caron por primera<br />vez las MnPs de y se caracterizaron sus genes. xiv Phane rochae te chrys os porium y Punctularia atropurpuras cens , P.atropurpurascens , Fisiología<br />de hong os filamentos os : e fe c to de l Zn y Mn<br />Los resultados obtenidos permiten constatar la actividad antifúngica de la<br />solución proveniente del proceso de reciclado, así como de la mezcla de sales<br />puras de Zn-Mn. Inhibe el crecimiento de ambos basidiomycetes, si bien<br />es más sensible a los metales. Durante el crecimiento sobre<br />madera en ensayos de laboratorio se confirma el poder antifúngico.. En campo,<br />es lixiviada de la madera, pero un segundo tratamiento con bórax impide la<br />pérdida de los metales. A 24 meses del comienzo del en as así<br />tratads se comparan con aquellas con CCA. A nivel del sistema ligninolítico, la<br />solución es inhibidora de la actividad enzimática, del mismo modo que la<br />mezcla de sales puras de Zn y Mn. El Mn presenta en términos generales un<br />efecto inductor de los parámetro del metabolismo estud con la excepción<br />de la germinación de conidios y le expresión de los genes de<br />Se constató que tiene por lo menos 7 MnPcon alta<br />similaridad con MnP de otros basidiomycetes. <p>An important issue in the development of metal-based radiopharmaceuticals is the selection of the labelling strategy in order to couple the metal to the pharmacophore without losing the biological activity. With the aim to evaluate the correlation between ligand denticity and biological behaviour of the corresponding 99mTc complexes, we designed a tridentate and a bidentate 5-nitroimidazole derivatives suitable for 99mTc(I) tricarbonyl complexation and with potential use as radiopharmaceuticals towards hypoxic tissue diagnosis. Ligands were synthesized using metronidazol, a pharmaceutical containing the bioreductive pharmacophore as starting material. The chelating units were connected to the pharmacophore using the click reaction of Huisgen. Both 99mTc complexes were obtained in high yield and were hydrophilic and stable in labelling milieu. The complex obtained from the tridentate ligand exhibited high stability in human plasma, low protein binding and a favourable biodistribution characterized by low blood and liver uptake, fast elimination and negligible uptake in other organs or tissues. Selective uptake and retention in tumour together with favourable tumour/muscle ratio makes this 99mTc-complex a promising candidate for further evaluation as potential hypoxia imaging agent in tumours. The bidentate ligand, on the other hand, yielded a less stable 99mTc-complex that experimented hydrolysis in vitro and decomposition in human plasma and showed high protein binding, high blood and liver uptake and moderate excretion. Although selective uptake and retention in tumour was also observed physicochemical and biological behaviour are inadequate for in vivo use, demonstrating that denticity of the ligand is particularly important and that tridentate ligands are preferable in order to prepare 99mTc-tricarbonyl complexes for Nuclear Medicine imaging</p>

Las biotransformaciones son modificaciones químicas pr ducidas sobre un sustrato, mediadas por organismos vivos o sus partes. Estas reacciones ocurren generalmente en condiciones suaves, a menudo son ester oselectivas, y muchas veces rinden productos de difícil obtención por síntesis clá ca. Concomitante con esto generan menos residuos tóxicos y subproductos, comparadas con los procesos químicos tradicionales. Por ello son un área de desarrollo dentro de la química verde, y una interesante herramienta a aplicar en un contexto de desarrollo sustentable. En este trabajo se buscó profundizar en aspectos quími os y metodológicos de las biotransformaciones de d-limoneno y 1,8-cineol. Para ello se utilizaron hongos aislados de fuentes naturales: una cepa de Aspergillus niger para biotransformar el limoneno y una de Aspergillus terreus para el cineol. Ambos monoterpenos son subproductos de industrias nacionales. El d-limoneno es el principal componente del aceite esencial de naranja y otros cítricos, sin contribuir en mayor medida a las propiedades organolépticas de la esencia. El 1,8-cineol es el componente mayoritario del aceite esencial de eucalipto, presente en las hojas de estos árboles, las que son un residuo de la creciente industria forestal. Mediante las bioconversiones se buscó obtener productos de mayor valor agregado per se o con potencial actividad biológica. En particular se estudió la potencialidad herbicida de los productos de biotransformacióndel cineol y derivados. La bioconversión de d-limoneno dio lugar a diferentes productos de oxidación, mayoritariamente alcoholes, con porcentajes de bioconv rsión relativamente bajos. Se estudió la recuperación de los productos, lográndose sistematizar el método de extracción del limoneno y sus productos de biotransformación desde el medio de cultivo. Del estudio de la influencia del medio de cultivo y la geometría del sistema de fermentación surgió que la biotransformación está notablemente condicionada por dichos parámetros. Mediante el uso de un inhibidor enzimático se logró una biotransformación regioespecífica del limoneno obtenié e alcohol perílico, un potencial anticancerígeno, como único producto. La biotransformación de 1,8-cineol arrojó cuatro alcoholes secundarios como principales productos, en todas las condiciones ensayadas. La geometría del sistema de fermentación no afectó significativamente el resultado de la biotransformación, pero el medio de cultivo resultó condicionante de la regio y estereoselectividad de la reacción. El principal producto fue el 2-exo-hidroxicineol, obtenido prácticamente como racemato en el medio YMPG y con un exceso enantiomérico del 96 % de su forma dextrógira en el medio CM. Sin embargo el rendimiento global de la bioconversión fue muy bajo (3,3 % en el día 7 de reacción). Los alcoholes obtenidos fueron sometidos a esterificaciones enzimáticas buscando derivados con potencial fitotoxicidad. Se ens ron dos lipasas y cuatro donadores de acilo. La esterificación con acetato de vinilo catalizada por una lipasa de Candida rugosa dio como único producto el (-)- acetato de 2-exohidroxicineol. La hidrólisis de este acetato dio lugar al (-)-2-exo-hidroxicineol con un exceso enantiomérico del 99 %. El trabajo que se describe a continuación surge como parte de un proyecto de investigación de síntesis de bengamidas y análogos, el cual se desarrolla en la Cátedra de Química Orgánica de la Facultad de Química, bajo la dirección el Dr. Gustavo Seoane. Dicho grupo de investigación hace uso de una cepa mutante de F39/D, para obtener sintones quirales a partir de compuestos aromáticos diversos, los cuales se utilizan como material de partida en síntesis asimétricas de compuestos naturales con actividad biológica. Las bengamidas son una clase de productos naturales ma inos que se aislaron originariamente en 1986 de esponjas del género recolectadas cerca de las Islas Fiji. Están formadas por un núcleo central de 7 miembros, correspondiente a una - caprolactama, al que se unen una cadena polioxigenada también, en la mayoría de los casos, una cadena lipofílica correspondiente a un ácid graso saturado que esterifica a la molécula. Estos compuestos, que fueron aislados como parte de un programa de búsqueda de metabolitos marinos con actividad antiparasitaria, muestran una muy interesante actividad antihelmíntica y citotóxica. En consecuencia, existe u gran interés en su preparación, ya que la opción de extracción a partir de esponjas produ cantidades limitadas y discontinuas. La alta oxigenación de la cadena lateral de las bengamidas y la funcionalización del anillo central los hace buenos candidatos para intentar su síntesis usando la metodología de biotransformación de arenos. En el presente trabajo se describen cuatro rutas de aproximación a la síntesis del anillo central a partir de halo-dihidrodioles quirales como único sintón de partida; se discute la apertura nucleofílica de a-halooxiranos quirales derivados de ; y se presentan los resultados de evaluación biológica (contra helmintos) una serie de intermedios sintéticos.

Abstract
A summary of the resource.
Con este trabajo se pretendió obtener sensores para adquisición de imágenes digitales de radiación X y γ a temperatura ambiente, con área de al menos 2” x 2”, basados en films cristalinos de semiconductores compuestos. En principio se estudia BiI3 como material para dichos sensores porque tiene propiedades físicas que lo hacen apropiado como sensor de radiaciones a temperatura ambiente, y también adecuadas para crecer un film. El trabajo comienza con la síntesis del BiI3 a partir de subcarbonato de bismuto y yoduro de potasio. Luego de esto se busca identificar las impurezas por absorción atómica con “inductively coupled plasma” (ICP), y optimizar un método para su reducción al nivel de ppb. También se determina la estequiometría del compuesto en sus distintas etapas de purificación mediante volumetría. El paso siguiente es el crecimiento de los films cristalinos sobre sustratos adecuados para, en el paso final, obtener imágenes digitales. Se utilizan como sustrato vidrio, vidrio paladizado y vidrio recubierto con óxido de estaño e indio (ITO). Los films se crecen por el método de deposición física de vapor (physical vapor deposition, PVD). Si bien para obtener imágenes digitales es suficiente tener films policristalinos, la experiencia indica que serán mejores films orientados o epitaxiales. Por tanto, se tiende a lograr el crecimiento de estos dos tipos de estructuras, y para ello se estudian los procesos de nucleación y coalescencia de los films. Sobre las mejores nucleaciones coalescidas se crecen films. Luego que un film ha crecido es necesario caracterizarlo. Se comienza por determinar sus propiedades físicas (perfilometría, microscopía óptica, de barrido electrónico (SEM) y de fuerzas atómicas (AFM) para estudiar su superficie, y difracción de rayos X, de polvo para estudiar su orientación). Después se determinan sus propiedades eléctricas mediante la construcción de detectores. También se mide la respuesta de los detectores a la radiación X. Por último se comparan los resultados obtenidos con los reportados para la obtención de imágenes con dispositivos alternativos.

Con el objetivo de desarrollar potenciales radiofármacos de 99mTc para imágenes de zonas de hipoxia, se han diseño, sintetizado y evaluado una serie de complejos de 99mTc conteniendo ligandos nitroimidazólicos. Dicho farmacóforo en condiciones de hipoxia es capaz de sufrir una reducción irreversible, transformándose en grupos citotóxicos, dentro de la célula. Para la síntesis de los ligandos se seleccionó el grupo 5-nitroimidazol como farmacóforo y a partir del fármaco comercial Metronidazol® (1-(2-metil-5- nitroimidazol-1-il)etanol), el que se puede obtener en cantidad y calidad necesaria, se realizaron las modificaciones químicas necesarias para acoplarle las distintas unidades quelantes necesarias para coordinar el metal, así como modificar la distancia através de una cadena espaciadora (linker). Para unir al Tc se seleccionaron los complejos tricarbonílicos de Tc(I) (COMPLEJOS 1 y 2), complejos Tc(V) nitrido (COMPLEJOS 3 y 4) y los complejos Tc(III) 4+1 (COMPLEJOS 5 y 6) a fin de poder evaluar la influencia del core empleado en las propiedades finales de los co jos obtenidos. Para ello se prepararon 6 ligandos, 2 por cada sistema de unión al metal utilizado, variando en cada caso la cadena espaciadora y la unidad quelante. Todos los complejos son preparados por sustitución utilizando el precursor adecuado en cada caso, el cual contiene al metal en el estado de oxidación requerido que puede ser fácilmente sustituidos por el ligando final. Los estudios fisicoquímicos revelan que los complejos preparados presentan variada lipofilicidad, siendo los mas hidrofílicos los complejos tricarbonílicos de Tc(I), presentando un logP de -0.44±0.04 para el COMPLEJO 1 y -0.75±0.08 para el COMPLEJO 2. Los compuestos mas lipofílicos fueron los complejos 4+1 de Tc(III) con logP de 0.7±0.1 y 1.2±0.3 para el COMPLEJO 5 y 6 respectivamente. Los complejos de Tc(V) nitrido presentan una lipofilicidad similar a los complejos 4+1 de Tc(III). destacar que, como era esperado, los COMPLEJOS 4 y 6 que contienen en cadena espaciadora un anillo piperacina presentan la mayor lipofilia dentro de la serie. Todos los complejos presentan una muy buena estabilidad en plasma, siendo la pureza radioquímica en todos los casos superior al 85% a las 4 horas de incubación a 37 ºC. La unión a proteínas determinada por exclusión molecular resultó adecuada para los complejos 1, 2, 3 y 5, estando en valores del 12 al 30 %. Los complejos que presentan el anillo piperacina (complejos 4 y 6), en cambio presentaron una elevada unión (69±3% y 75±4% respectivamente). Se desarrolló una metodología que permitió realizar la evaluación de los complejos en células tumorales humanas (HCT-15) en condiciones de hipoxia, obteniéndose para todos los complejos relaciones de captación entre hipoxia e oxia superiores a 1, siendo la mayor para el COMPLEJO 1 (2.2±0.3). Los estudios de biodistribución en ratones CD1 normales, realizados entre 30 min. y 4 horas post-inyección,mostraron una buena correlación con las propiedades fisicoquímicas. El complejo de mejor perfil es el COMPLEJO 1, ya que presenta una baja actividad sanguinea, rapida d ón de tejidos blandos y eliminación combinada por vía urinaria y hepatobiliar (64.86±4.95% y 46.86±2.39% respectivamente a las 4 horas). Los complejos 4 y 6 (como era de esperar) mostraron una muy alta captación en hígado (54-55% a las 4h) y una muy baja eliminación (10-12 % a las 4h). La captación en tumor evaluada en animales portadores de sarcoma de Lewis murino (células 3LL), demostró la captación y retención de los complejos en el tumor. Todos los complejos presentan una relación de % Dosis/g en Tumor/Músculo mayor a 1 a las 4 h post inyección debid ción de la actividad en el tumor y a la depuración de los demás tejidos. La mayor relación la presentó el COMPLEJO 1 con un valor de 4.22±0.07 a las 4 h. Los estudios estructurales empleando Re estable indicarían que es correcta la estructura propuesta para los complejos 5 y 6, siendo necesarios más estudios a fin de poder confirmar o descartar la e sta en los otros casos. Se pudieron realizar en la tesis todas las etapas involucradas en el desarrollo de un nuevo radiofármaco, desde la selección del farmacóforo, síntesis orgánica, marcación, estudios fisicoquímicos hasta la evaluación biológica.

En esta tesis se aplicaron técnicas de Análisis en Flujo para la automatización de las determinaciones de cobre, zinc, selenio total, nitrato y nitrito en leche en polvo, leche fluida y fórmulas infantiles. Se mostraron las ventajas de utilizar sistemas en flujo para estas determinaciones. Se diseñaron, optimizaron y validaron sistemas de Análisis por Inyección en Flujo (FIA) para la determinación de cobre y zinc en leche en polvo y fórmulas infantiles, con detección mediante espectrometría de absorción molecular UV-visible. Para la determinación de nitrato, nitrito y zinc se diseñaron, optimizaron y validaron también sistemas en flujo con inyección secuencial (SIA) con detección mediante espectrometría de absorción molecular UV-visible. Se obtuvieron datos sobre el contenido de selenio total en leche en polvo y leche fluida de nuestro país utilizando un sistema en flujo multiconmutado (MCFA) con detección mediante absorción atómica con generador de hidruros, el mismo fue optimizado y validado, resultando adecuado para estas determinaciones. Para cada determinación fue necesario optimizar un tratamiento de preparación de muestra adecuado. En el caso particular de la determinación de selenio se optimizó un método para mineralización de leche en polvo vía húmeda con HNO3 y H2O2 utilizando microondas. Para leche fluida se optimizó un método utilizand descomposición de la matriz con ultrasonido y ácidos. Los resultados de las determinaciones realizadas con las técnicas en flujo propuestas resultaron comparables con los métodos oficiales actualmente utilizados por la AOAC (Association of Oficial Analytical Chemist), e IDF (Internacional Dairy Federation). Se utilizó material de referencia certificado, siempre que fue posible, para la validación. Las frecuencias de muestreo de los métodos desarrollados estuvieron entre 30-160 hora-1. Los resultados obtenidos son importantes debido a que pueden contribuir a completar las tablas nutricionales nacionales que permanecen incompletas agregando el contenido de selenio en leche. Los valores de selenio total obtenidos estuvieron en el rango de 11.5 – 34.1μg.L-1 para leche fluida, 31 – 130 μg.kg-1 para leche en polvo y 42 -138 μg.kg-1 para fórmulas infantiles comerciales. El desarrollo de esta tesis permitió emplear diversas técnicas de Análisis enFlujo rápidas y eficientes para las determinaciones para las que fueron propuestas con las ventajas de la automatización.

Abstract
A summary of the resource.
<p>Herpes simplex virus type 1 (HSV-1) infection has a prevalence of 70% in the human population. Treatment is based on acyclovir, valacyclovir, and foscarnet, three drugs that share the same mechanism of action and of which resistant strains have been isolated from patients. In this aspect, innovative drug therapies are required. Natural products offer unlimited opportunities for the<br />discovery of antiviral compounds. In this study, 28 extracts corresponding to 24 plant species and 4 alga species were assayed in vitro to detect antiviral activity against HSV-1. Six of the methanolic extracts inactivated viral particles by direct interaction and 14 presented antiviral activity when incubated with cells already infected. Most interesting antiviral activity values obtained are<br />those of Limonium brasiliense, Psidium guajava, and Phyllanthus niruri, which inhibit HSV-1 replication in vitro with 50% effective concentration (EC50) values of 185, 118, and 60 μg/mL, respectively. For these extracts toxicity values were calculated and therefore selectivity indexes (SI) obtained. Further characterization of the bioactive components of antiviral plants will pave the way for the discovery of new compounds against HSV-1.</p>
