Opportunistic infections are a problem of great relevance in public health and the precise detection and localization of infection in the early stages of the dis ease is of great importance for patient management as well as cost containment. Our proposal seeks to contribute to developing a new agent that meets the needs of diagnosis and follow-up of fungal and bacterial infections, focused on the de sign of a radiotracer with the potential for recognition of hidden infection foci. Defensins are plant antimicrobial peptides that not only show activity against plant pathogens but also against human ones. A short analogue of EcgDf1 de fensin, EcgDf21d (NH2-ERFTGGHCRGFRRRCFCTKHC-COOH), was labelled through the formation of a 99mTc-HYNIC complex which was assessed for phys icochemical and biological behaviour both in vitro and in vivo. The [99mTc]Tc HYNIC-EcgDf21 labelling procedure rendered a single product with remarkably high RCP and stability in the labelling milieu. The Log p value indicated that [ 99mTc]Tc-HYNIC-EcgDf21 has a hydrophilic behaviour, confirmed by the biodis tribution profiles. The optimal uptake value was obtained for Candida albicans infection model reaching a lesion/muscle ratio of 3, this correlates with in vitro binding studies, and the lesion can be definitely observed in the scintigraphic images.

Research on behavioral and social science has demonstrated that transitions throughout a person’s life course, as the life event of becoming a parent, represent an opportunity for dietary changes. However, research in this area has been mostly restricted to developed European and North American countries and has shown ambiguous results. The present work aimed to gain an in-depth understanding on the changes in eating habits caused by the transition to parenthood and to explore factors influencing those changes in Uruguay, a Latin American country. Forty-two in-depth interviews with Uruguayan parents from diverse social-economic backgrounds were conducted. The narrations by the interviewees evidenced mostly positive changes during three stages in the transition: pregnancy, the first months with the baby, and the beginning of complementary feeding. Most informants perceived changes mainly in terms of increased consumption of fruits, vegetables, and pulses; increased consumption of homemade meals; and decreased consumption of ultra-processed foods, fast foods, and fried foods. Reasons for these changes included adaptation of meals to the child’s needs and schedule, feeling of responsibility for the child’s health, and willingness to be a good role model. Factors influencing changes in eating habits were identified in some levels of the socioecological model and included socioeconomic status; price of fruits, vegetables, and fish; low accessibility of good quality fish; perceived time pressure; father’s low interest on healthy eating; social support; and access to nutrition information. Findings suggest that parenthood represents a window of opportunity for favorable changes in eating habits. However, barriers constraining those changes should be addressed by stakeholders.

The synthesis of cyclotetrapeptides analogues of the natural products tentoxin and versi cotide D was achieved in good yield by solid phase peptide synthesis (SPPS) of their linear precursors and solution phase cyclization. All the cyclopeptides and several open precursors were evaluated as herbicides. Five cyclopeptides and five lineal peptides showed a significant inhibition (>70%) of Ryegrass seed’s radicle growth at 67 µg/mL. The evaluation at lower concentrations (4–11 µM) indi cates two cyclopeptides analogs of tentoxin, which present one (N-Methyl-D-Phe), and two N-MeAA (N-Methyl-Ala and N-Methyl-Phe), respectively, as the most active of them, showing remarkable phytotoxic activity. In two cases, the open precursors are as active as their corresponding cyclopeptide. However, many linear peptides are inactive and their cyclization derivatives showed herbicidal activ ity. In addition, two cyclopeptide analogues of versicotide D showed more improved activity than the natural product. The results indicate that the peptide sequence, the amino acid stereochemistry and the presence of N-methyl group have important influence on the phytotoxic activity. Moreover, several compounds could be considered as lead candidates in the development of bioherbicides

Human African trypanosomiasis (sleeping sickness) and leishmaniasis are prevalent zoonotic diseases caused by genomically related trypanosomatid protozoan parasites (Trypanosoma brucei and Leishmania spp). Additionally, both are co-endemic in certain regions of the world. Only a small number of old drugs exist for their treatment, with most of them sharing poor safety, efficacy, and pharmacokinetic profiles. In this work, new multifunctional Ru(II) ferrocenyl compounds were rationally designed as potential agents against these trypanosomatid parasites by including in a single molecule 1,1′ -bis(diphenylphosphino)ferrocene (dppf) and two bioactive bidentate li gands: 8-hydroxyquinoline derivatives (8HQs) and polypyridyl ligands (NN). Three [Ru(8HQs)(dppf)(NN)](PF6) compounds were synthesized and fully characterized. They showed in vitro activity on bloodstream Trypanosoma brucei (IC50 140–310 nM) and on Leishmania infantum promastigotes (IC50 3.0–4.8 μM). The compounds showed good selectivity towards T. brucei in respect to J774 murine macrophages as mammalian cell model (SI 15–38). Changing hexafluorophosphate counterion by chloride led to a three-fold increase in activity on both parasites and to a two to three-fold increase in selectivity towards the pathogens. The compounds affect in vitro at least the targets of the individual bioactive moieties included in the new chemical entities: DNA and generation of ROS. The compounds are stable in solution and are more lipophilic than the free bioactive ligands. No clear correlation between lipophilicity, interaction with DNA or generation of ROS and activity was detected, which agrees with their overall similar anti-trypanosoma potency and selectivity. These compounds are promising candidates for further drug development.

Microbial agents exert their biocontrol effects through different mechanisms of action. Clavispora lusitaniae AgL21, a yeast capable of controlling postharvest diseases in lemon, produces volatile organic compounds (VOCs) impairing phytopathogenic fungi. Here we elucidate the nature of such compounds and check individual effects on mycelial growth of citrus fungal pathogens: Penicillium digitatum, Penicillium italicum, and Geotrichum citri-aurantii. VOCs characterization was carried out deploying solid-phase microextraction/gas chromatography mass spectrum techniques. Among the compounds produced by C. lusitaniae AgL21, ethyl acetate, isoamyl ac etate, 3-methyl butanol, and phenethyl alcohol displayed the highest relative abundance. Synthetic ethyl acetate, tested in vitro on P. digitatum, had no discernable effect on mycelial growth, whereas isoamyl acetate and both of the above alcohols exhibited fungistatic and fungicidal effects, respectively. The data provided in this study prove for the first time, that VOCs released by C. lusitaniae AgL21 are able to take part in antagonistic activities against citrus postharvest fungal pathogens.

Background: Vaccination against severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has pro ven to be a successful strategy for prevent severe infections. CoronaVac and BNT162b2 are the most used vaccines worldwide, but their use in heterologous vaccination schedules is still subjected to evaluation. Methods: Fifty healthy individuals who received heterologous prime-boost vaccination with CoronaVac and BNT162b2 were enrolled in a post-vaccination serological follow-up longitudinal prospective study. We evaluated specific serum anti-receptor binding domain (RBD) IgG antibody levels, and their capacity to block RBD-ACE2 interaction with a surrogate neutralization assay. In 20 participants, we assessed anti body binding kinetics by surface plasmon resonance, and Fc-mediated functions by ADCC and ADCP reporter assays. Results: Our baseline seronegative cohort, displayed seroconversion after two doses of CoronaVac and an important decrease in serum anti-RBD IgG antibodies levels 80 days post-second dose. These levels increased significantly early after the third dose with BNT162b2, but 73 days after the booster we found a new fall. Immunoglobulin functionalities showed a similar behavior. Conclusions: The heterologous prime-boost vaccination with CoronaVac and BNT162b2 generated an impressive increase in serum anti-RBD specific antibody levels followed by a drop. Nevertheless, these titers remained well above those found in individuals only vaccinated with CoronaVac in the same elapsed time. Serum IgG levels showed high correlation with antibody binding analysis, their capacity to block RBD-ACE2 interaction, and Fc-effectors mechanisms. Our work sheds light on the humoral immune response to heterologous vaccination with CoronaVac and BNT162b2, to define a post vaccination correlate of protection against SARS-CoV-2 infection and to discuss the scheduling of future vaccine boosters in general population.

Herein, we investigated the adsorption of helicenes over graphene, boron nitride, graphane, and fluorographane utilizing density functional calculations. Our results indicated that helicenes experience significant structural distortions when adsorbed on the title monolayer. The strong interaction with the surfaces can partially over come the steric repulsions of helicenes. Although the characteristics of the surfaces considered are very different, the adsorption energies computed for graphene, BN, and graphane are comparable. However, over fluo rographane, they were slightly weaker. In the case of bilayer graphene, helicene[4], and helicene[5], when intercalated, their height can be reduced up to 0.3 and 0.4 Å, respectively.

Biomass-derived chemical platforms such as 5-hydroxymethylfurfural (5-HMF) can offer straight forward access to new value-added products. The presence of an aldehyde and hydroxymethyl functional groups in this molecule are interesting structural motifs to explore the synthesis of new bioactive compounds. In this contribution, a strategy to quickly and efficiently synthesize nitro vinylfurans starting from 5-HMF under greener conditions is described and the antiproliferative activity of the synthesized compounds evaluated. 5-HMF obtained by microwave-assisted satu rated biphasic dehydration from fructose was used to produce furaldehyde derivatives. Next, through a nitroaldolic condensation, the desired nitrovinylfurans were generated using efficient heating sources, ultrasound, or a non-microwave instant heating reactor, under solvent-free con ditions. Most of the newly synthesized compounds showed growth inhibition when assayed for their in vitro antiproliferative activity against six cancer cell lines (A549, HBL-100, HeLa, SW1573, T-47D, and WiDr), with lower toxicity on the human macrophage-like cells (THP-1). Our strategy allowed the rapid identification of nitrovinylfurans 16, 17, 19, and 20 as future can didates for the development of new selective anticancer drugs.

This work studied ultrasound assisted extraction (UAE) as a green chemistry technique to improve aqueous extraction of bioactive compounds from marcela (Achyrocline satureioides Lam.), a native plant from South America with reported beneficial health effects. Dried intact marcela inflorescences were extracted using solely water as solvent in an ultrasonic bath, applying 100 and 150 W/L ultrasound power at 40 and 60 °C, respectively. Extraction was followed throughout 30 min by determining antioxidant capacity (AOC) and total polyphenol content (TPC) every 5 min. Extracts were analysed by LC-MS and LC-DAD to identify extracted bioactive compounds and analyse the impact of temperature and ultrasound application on compound profile. Solidliquid extraction at 40 and 60 °C and maceration at room temperature were studied as reference extraction methods. UAE extracts were higher in AOC by 140% and 25% at 40 and 60 °C respectively, compared to solid-liquid extraction. For TPC, UAE showed an increase of 110% and 20% at 40 and 60 °C, respectively. LC results indicated that ultrasound application allowed faster and enhanced hydroxycinnamic acids extraction, whereas an increase in temperature resulted in a more diverse compound profile. UAE could be used to extract caffeic acid when applied at 40 °C.

Forestry and agricultural industries constitute highly relevant economic activities globally. They generate large amounts of residues rich in lignocellulose that have the potential to be valorized and used in different industrial processes. Producing renewable fuels and high-value-added compounds from lignocellulosic biomass is a key aspect of sustainable strategies and is central to the biorefinery concept. In this study, the use of biomass-derived monosaccharides for the enzymatic synthesis of sugar fatty acid esters (SFAEs) with antimicrobial activity was investigated to valorize these agro-industrial residues. With the aim to evaluate if lignocellulosic monosaccharides could be substrates for the synthesis of SFAEs, D-xylose, L-arabinose, and D-glucose, lauroyl and stearoyl monoesters were synthetized by transesterification reactions catalyzed by Lipozyme RM IM as biocatalyst. The reactions were performed using commercial D-xylose, L-arabinose, and D-glucose separately as substrates, and a 74:13:13 mixture of these sugars. The proportion of monosaccharides in the latter mixture corresponds to the composition found in hemicellulose from sugarcane bagasse<br />and switchgrass, as previously described in the literature. Products were characterized using nuclear magnetic resonance (NMR) spectroscopy and showed that only the primary hydroxyl group of these monosaccharides is involved in the esterification reaction. Antimicrobial activity assay using<br />several microorganisms showed that 5-O-lauroyl-D-xylofuranose and 5-O-lauroyl-L-arabinofuranose have the ability to inhibit the growth of Gram-positive bacteria separately and in the products mix. Furthermore, 5-O-lauroyl-L-arabinofuranose was the only product that exhibited activity against Candida albicans yeast, and the four tested filamentous fungi. These results suggest that sugar fatty acid esters obtained from sustainable and renewable resources and produced by green methods are promising antimicrobial agents.

Lung cancer remains the leading cause of cancer mortality worldwide. Thus, the development of strategies against this type of cancer is of high value. Parasite infections can correlate with lower cancer incidence in humans and their use as vaccines has been recently explored in preclinical models. In this study, we investigated whether immunisations with a Trypanosoma cruzi lysate from epimastigotes protect from lung tumour growth in mice. We also explore the role of parasite glycans in the induction of the protective immune response. A pre-clinical murine cancer model using the lung tumour cell line LL/2 was used to evaluate the anti-tumour potential, both in preventive and therapeutic settings, of a T. cruzi epimastigote-derived protein lysate. Immunisation with the parasite lysate prevents tumour growth and induces both humoral and cellular anti-tumour immune responses to LL-2 cancer cells. The induced immunity and tumour protection were associated with the activation of natural killer (NK) cells, the production of interferon- (IFN- ) and tumour cell cytotoxicity. We also show that mannose residues in the T. cruzi lysate induce Toll-like receptor (TLR) signalling. The evaluated T. cruzi lysate possesses anti-tumour properties likely by activating innate and adaptive immunity in a process where carbohydrates seem to be essential.

The antiproliferative effects of four series of VIVO- and VVO-based compounds containing 8-hydroxyquinoline ligands on the bacterium Mycolicibacterium smegmatis (M. smeg) were investigated. The effects on M. smeg were compared to the antiproliferative effects on the protozoan parasite Trypanosoma cruzi (T. cruzi), the causative agent for Chagas disease. In this study, we investigate the speciation of these compounds under physiological conditions as well as the antiproliferative effects on the bacterium M. smeg. We find that the complexes are more stable the less H2O is present, and that the stability increases in lipid-like environments.<br />Only one heteroleptic complex and two homoleptic complexes were found to show similar antiproliferative effects on M. smeg as reported for T. cruzi so the responses generally observed by M.smeg. is less than observed by the pathogen. In summary, we find that M. smeg is more sensitive to the detailed structure of the V-complex but overall these complexes are less effective against M. smeg compared to T. cruzi.

Persulfides (RSSH/RSS− ) are species closely related to thiols (RSH/RS− ) and hydrogen sulfide (H2S/HS− ), and can be formed in biological systems in both low and high molecular weight cysteine-containing compounds. They are key intermediates in catabolic and biosynthetic processes, and have been proposed to participate in the transduction of hydrogen sulfide effects. Persulfides are acidic, more acidic than thiols, and the persulfide anions are expected to be the predominant species at neutral pH. The persulfide anion has high nucleophilicity, due in part to the alpha effect, i.e., the increased reactivity of a nucleophile when the neighboring atom has high electron density. In addition, persulfides have electrophilic character, a property that is absent in both thiols and hydrogen sulfide. In this article, the biochemistry of persulfides is described, and the possible ways in which the formation of a persulfide could impact on the properties of the biomolecule involved are discussed.

turgeons are chondrostean fish of high economic value and critically endangered due to anthropogenic activ ities, which has led to sturgeon aquaculture development. Russian sturgeon (Acipenser gueldenstaedtii), the second most important species reared for caviar, is successfully farmed in subtropical countries, including Uruguay. However, during the Uruguayan summer, sturgeons face intolerable warmer temperatures that weaken their defences and favour infections by opportunistic pathogens, increasing fish mortality and farm economic losses. Since innate immunity is paramount in fish, for which the liver plays a key role, we used deep RNA sequencing to analyse differentially expressed genes in the liver of Russian sturgeons exposed to chronic heat stress and challenged with Aeromonas hydrophila. We assembled 149.615 unigenes in the Russian sturgeon liver tran scriptome and found that metabolism and immune defence pathways are among the top five biological processes taking place in the liver. Chronic heat stress provoked profound effects on liver biological functions, up regulating genes related to protein folding, heat shock response and lipid and protein metabolism to meet en ergy demands for coping with heat stress. Besides, long-term exposure to heat stress led to cell damage triggering liver inflammation and diminishing liver ability to mount an innate response to A. hydrophila challenge. Accordingly, the reprogramming of liver metabolism over an extended period had detrimental effects on fish health, resulting in weight loss and mortality, with the latter increasing after A. hydrophila challenge. To our knowledge, this is the first transcriptomic study describing how chronic heat-stressed sturgeons respond to a bacterial challenge, suggesting that liver metabolism alterations have a negative impact on the innate anti bacterial response.

Quillaja saponins have an intrinsic capacity to interact with membrane lipids that self-assembles in nanoparticles (immu nostimulating complexes or ISCOM-matrices) with outstanding immunoadjuvant activity and low toxicity profle. However, the expensive and laborious purifcation processes applied to purify Quillaja saponins used to assemble ISCOM-matrices show an important drawback in the large-scale use of this vaccine adjuvant. Thus, in this study, we describe a protocol to appropriately formulate ISCOM-matrices using the raw aqueous extract (AE) of Quillaja lancifolia leaves. In the presence of lipids, AE was able to self-assemble in nanostructures that resembles immunostimulating complexes (ISCOM). These nega tively charged nanoparticles of approximately 40 nm were characterized by transmission electron microscopy and dynamic light scattering. In addition, well-known saponins with remarkable immunoadjuvant activity, as QS-21, were detected into nanoparticles. Thus, the easier, robust, cheaper, and environmentally friendly method developed here may be an alternative to the classical methods for ISCOM-matrices production that use high-purifed saponins.

Nitrogen cycle has been poorly investigated in Antarctic ecosystems. In particular, how extreme conditions of low temperature, dryness, and high radiation select the microorganisms involved in the cycle is not yet understood. Denitrification is an important step in the nitrogen cycle in which nitrate is reduced stepwise to the gases NO, N2O, and N2. Denitrification is carried out by a wide group of microorganisms spread in the phylogenetic tree. The aim of this work was to isolate and characterize denitrifying bacteria present in different cold environments from Antarctica. Bacterial isolates were obtained from lake, meltwater, sea, glacier ice, ornithogenic soil, and penguin feces samples from King George Island, Fildes peninsula in the Antarctic. Samples were taken during the deicing season in five sampling campaigns. From all the samples we were able to isolate denitrifying strains. A total of 199 bacterial isolates with the capacity to grow in anaerobic mineral media reducing nitrate at 4°C were obtained. The characterization of the isolates by 16S rRNA gene sequence analysis showed a high predominance of the genus Pseudomonas, followed by Janthinobacterium, Flavobacterium, Psychrobacter, and Yersinia. Other minor genera detected were Cryobacterium, Iodobacter, Kaistella, and Carnobacterium. The capacity to denitrify was not previously described for most of the bacteria related to our isolates and in many of them denitrifying genes were not present suggesting the presence of new genes in this extreme environment. Our work demonstrates the ubiquity of denitrification in the Maritime Antarctica and gives important information linking denitrification at cold temperature with taxa in an unequivocal way.

We successfully synthesized two types of bismuth tri-iodide nanoparticles: uncapped (BiI3) and capped with aniline (BiI3_ani), in order to study their potential as inorganic semiconductor for an organic–inorganic hybrid material with poly(3-hexylthiophene-2,5-diyl) (P3HT). We developed a novel direct synthesis method that avoids ligand exchange (for BiI3_ani), as well as the necessity of capping agent removal steps (for BiI3) and the formation of sec ondary compounds obtained by other techniques. Additionally, we evaluated and compared the properties of solution-processed P3HT:nanoparticles layers with the film sequence ITO/PEDOT:PSS/P3HT:nanoparticles, where ITO is indium tin oxide and PEDOT:PSS is poly(3,4-ethylenedioxythiophene) poly styrene sulfonate. We analyzed the layers by UV–Vis spectroscopy, X-ray diffraction, focused ion beam scanning electron microscopy and photolumi nescence analysis. We proved that aniline restricts the growth of nanoparticles and improves the processability of the material compared to nanoparticles without capping agent. We demonstrated that both types of nanoparticles can be successfully suspended in a solvent in which P3HT is soluble, improve the UV–Vis absorption of P3HT and interact with this polymer leading to an improved crystalline ordering on the blends compared to pristine P3HT layers. We also demonstrated that BiI3 nanoparticles produce a nanoscopic mixture with P3HT and that it is possible to exploit the photoluminescence emission of P3HT:BiI3_ani layers. Our results show that both types of nanoparticles can be good candidates for use in solution-processed organic–inorganic hybrid material with P3HT, which could be employed for the fabrication of hybrid polymeric–inorganic solar cells with BiI3 nanoparticles as electron acceptor and P3HT as electron donor.

The protein DBC1 is the main SIRT1 regulator known so far, and by doing so, it is involved in the regulation of energy metabolism, especially in liver and fat adipose tissue. DBC1 also has an important function in cell cycle progression and regulation in cancer cells, affecting tumorigenesis. We recently showed that during quiescence, non-transformed cells need DBC1 in order to re-enter and progress through the cell cycle. Moreover, we showed that deletion of DBC1 affects cell cycle progression during liver regeneration. This novel concept prompted us to evaluate the role of DBC1 during adult neurogenesis, where transition from quiescence to proliferation in neuronal progenitors is key and tightly regulated. Herein, we analyzed several markers of cell cycle expressed in the dentate gyrus of the hippocampus of controls and DBC1 KO adult mice. Our results suggest a reduced number of neuroblasts therein present, probably due to a decline of neuroblast generation or an impairment in neural differentiation. In agreement with this, we also found that adult DBC1 KO mice had a reduction in the volume of the granule cell layer of the dentate gyrus. Interestingly, behavioral analysis of KO and control mice revealed that deletion of DBC1 parallels to specific cognitive impairments, concerning learning and possibly memory forma tion. Our results show, for the first time, that DBC1 plays an active role in the nervous system. In particular, specific anatomical and behavioral changes are observed when is absent.

The information included on food packages has a crucial role in influencing consumer product associations and purchase decisions. In particular, visual and textual cues on processed and ultra-processed products can convey health-related associations that influence consumer healthiness perception and purchase decisions. In this context, the present work aimed to explore the use of health-related cues on the packages of processed and ultra processed products sold in Uruguay to provide insights for policy making. A total of 3813 products from 34 different food categories found in four of the most important supermarket chains in Uruguay were surveyed. The textual and visual information included on the packages as well as the nutritional composition of the products were analyzed. Results showed that 67% of the products included at least one health-related cue. Pictures of culinary ingredients, natural and minimally processed foods were the most frequent health-related cue, followed by references to naturalness and claims related to critical nutrients. The prevalence of health-related cues largely differed across product categories, ranging from 100% to 17%. The relationship between the presence of health-related cues on the packages and the excessive content of nutrients associated with non-communicable diseases was assessed using a gradient boosting model, which showed limited predictive ability. This suggests that the inclusion of health-related cues on food packages was not strongly related to the nutritional composition of products and therefore cannot be regarded as a healthiness indicator. These results stress the need of develop stricter labeling regulations to protect consumers from misleading information.

<p>Perceived (or subjective) wellbeing is regarded as key to understanding consumer food choices and the devel opment of strategies to promote desirable eating habits. Yet, in-depth understanding of the specific factors that contribute to people’s perceived wellbeing across cultures is lacking. These factors motivated the present research that used word associations to conduct an exploratory analysis of consumer conceptualisations of food related wellbeing (WB). Adults (n = 4945) living in the United Kingdom, Australia, Singapore or Germany, and speaking respectively, English or German, took part in the study. Health, pleasure, food quality, positive emo tions and social aspects of food consumption were the main associations with food-related wellbeing. Absence hereof was associated with unhealthiness, disgust, negative emotions and poor mental health. The differences in these main associations emphasised the importance of exploring wellbeing, as well as lack hereof. Not doing so leads to an incomplete understanding of this multidimensional construct. The research was conducted with four terms related to wellbeing (each in their positive and negative versions): ‘sense of wellbeing’ and ‘lack of wellbeing’, ‘feeling good’ and ‘feeling bad/unhappy’, ‘satisfied with life’ and ‘dissatisfied with life’ and ‘fulfilled in life’ and ‘unfulfilled in life’. Because these different terms gave rise to different wellbeing associations, re searchers in this area must choose their empirical approach with care. The terms ‘sense of wellbeing’ and ‘feeling good’ tended to more frequently give rise to health-related associations. Conversely, ‘satisfied with life’ and ‘fulfilled in life’ tended to more frequently give rise to positive spiritual and emotional associations of food related wellbeing. The main conceptualisations of food-related wellbeing were cross-culturally similar, but extension of the present research to other Asian countries was recommended based on several differences be tween Singaporean participants and those from other countries. In ethnically diverse countries like Singapore, further within-country investigations of different cultures will also have merit.</p>

Proteins in whey from casein production were hydrolyzed with proteolytic extracts from the latex of Araujia sericifera var. hortorum. Hydrolysate fractions that were partially purified by RP-HPLC and evaluated using the ORAC method resulted in an antioxidant capacity of 112.4 ± 4.2 Trolox equivalents (μM). LC–MS/MS analysis of peptide fractions showed that peptides derived from β-lactoglobulin could be responsible for antioxidant activity. The sum of these results along with the fact that a controlled and reproducible production process was used, involving the use of natural resources and reaction conditions without excessive costs at an industrial scale, makes a promising contribution to improving antioxidant dairy products.

The reversibility of the thiol-thioester linkage has been broadly employed in many fields of biochemistry (lipid synthesis) and chemistry (dynamic combinatorial chemistry and material science). When the transthioesterification is followed by a S-to-N acyl transfer to give an amide bond, it is called Native Chemical Ligation (NCL), a high-yield chemoselective process used for peptide synthesis. Recently, we described thioglycolic acid (TGA) as a useful reagent for thioester deprotection both in solution and anchored to a solid-support under mild conditions. Inspired by NCL, in this work, we extended this approach and explored the use of 2-aminothiols for the deprotection of thiols bearing an acyl group. The best results were obtained using cysteamine or L-cysteine in an aqueous buffer pH 8 at room temperature for 30 min. The described approach was useful for S-acetyl, S-butyryl, and S-benzoyl heterocycles deprotection with yields up to 84%. Employing this methodology, we prepared six new analogs 2 of mercaptomethyl bisthiazolidine 1, a useful inhibitor of a wide-range of Metallo-β-Lactamases (MBLs). Compared with the previous methodologies (TGA polymer supported and TGA in solution), the biomimetic deprotection herein described presents better performance with higher yields, shorter reaction times, less timeconsuming operations, easier setup, and lower costs.

Caenorhabditis elegans is a free-living nematode that has been validated for anthelmintic drug screening. However, this model has not been used to address anthelmintic dose-response-time and drug-drug interactions through matrix array methodology. Eprinomectin (EPM) and Ivermectin (IVM) are macrocyclic lactones widely used as anthelmintics. Despite being very similar, EPM and IVM are combined in commercial formulations or mixed by farmers, under the assumption that the combination would increase their efficacy. However, there is no data reported on the pharmacological evaluation of the combination of both drugs. In this study, we assessed the pharmacodynamics and drug-drug interactions of these two anthelmintic drugs. Since the action of these drugs causes worm paralysis, we used an infrared motility assay to measure EPM and IVM effects on worm movement over time. The results showed that EPM was slightly more potent than IVM, that drug potency increased with drug time exposure, and that once paralyzed, worms did not recover. Different EPM/IVM concentration ratios were used and synergy and combination sensitivity scores were determined at different exposure times, applying Highest Single Agent (HSA), Loewe additivity, Bliss and Zero Interaction Potency (ZIP) models. The results clearly indicate that there is neither synergy nor antagonism between both macrocyclic lactones. This study shows that it is more relevant to prioritize the exposure time of each individual drug than to combine them to improve their effects. The results highlight the utility of C. elegans to address pharmacodynamics studies, particularly for drug-drug interactions. Models in vitro can be integrated to facilitate preclinical and clinical translational studies and help researchers to understand drug-drug interactions and achieve rational therapeutic regimes.
