To assess recalcitrant pesticide bioremediation it is necessary to gradually increase the complexity of the biological system used in order to design an effective biobed assembly. Each step towards this effective biobed design needs a suitable, validated analytical methodology that allows a correct evaluation of the dissipation and bioconvertion. Low recovery yielding methods could give a false idea of a successful biodegradation process. To address this situation, different methods were developed and validated for the simultaneous determination of endosulfan, its main three metabolites, and chlorpyrifos in increasingly complex matrices where the bioconvertor basidiomycete Abortiporus biennis could grow. The matrices were culture media, bran, and finally a laboratory biomix composed of bran, peat and soil. The methodology for the analysis of the first evaluated matrix has already been reported. The methodologies developed for the other two systems are presented in this work. The targeted analytes were extracted from fungi growing over bran in semisolid media YNB (Yeast Nitrogen Based) with acetonitrile using shaker assisted extraction, The salting-out step was performed with MgSO4 and NaCl, and the extracts analyzed by GC-ECD. The best methodology was fully validated for all the evaluated analytes at 1 and 25mgkg(-1) yielding recoveries between 72% and 109% and RSDs <11% in all cases. The application of this methodology proved that A. biennis is able to dissipate 94% of endosulfan and 87% of chlorpyrifos after 90 days. Having assessed that A. biennis growing over bran can metabolize the studied pesticides, the next step faced was the development and validation of an analytical procedure to evaluate the analytes in a laboratory scale biobed composed of 50% of bran, 25% of peat and 25% of soil together with fungal micelium. From the different procedures assayed, only ultrasound assisted extraction with ethyl acetate allowed recoveries between 80% and 110% with RSDs <18%. Linearity, recovery, precision, matrix effect and LODs/LOQs of each method were studied for all the analytes: endosulfan isomers (α & β) and its metabolites (endosulfan sulfate, ether and diol) as well as for chlorpyrifos. In the first laboratory evaluation of these biobeds endosulfan was bioconverted up to 87% and chlorpyrifos more than 79% after 27 days.

Chlorpyrifos is one of the most employed insecticides worldwide. It is moderately persistent in the environment. It degrades to 3, 5, 6- trichloro-2-pyridinol (TCP), the metabolite of toxicological concern to both human health and the environment. In the field, pesticides point contaminations occur during the operation of the application machinery. Biobeds made up of a biomixture containing straw, peat, and soil have been successfully developed to minimise these incidents. The retained pesticides are degraded by the microorganisms in the biomixture. Although the dissipation of chlorpyrifos in different biobeds has been studied, the fate of TCP received scarce attention. Biodegradation in specific bioreactors can only be assessed if it is evaluated through proper analytical methods. In this work chlorpyrifos and TCP dissipation in biobeds was studied with a previously fit for purpose analytical method. The lyophilised biomixture was extracted with ethyl acetate in an aqueous solution of NaCl and sodium tetraborate. The extract was cleaned up with alumina. Chlorpyrifos was determined by gas chromatography with electron capture and mass spectrometer detectors and TCP by liquid chromatography coupled to tandem mass spectrometry, using the negative Multiple Ion Mode. Recoveries of the two analytes, up to 2 mg kg−1, were between 70% and 113% with RSDs below 16% and RSDwR below 19%. Further TCP confirmation was evaluated through Enhanced Product Ion mode for each ion. The dissipation of both analytes in laboratory biobeds inoculated with Abortiporus biennis using two different types of Uruguayan soils was assayed. Both bioreactors were able to dissipate the compounds after 60 days following different dissipation kinetics. Similar results were obtained after scaling up in a biobed under field conditions.

Current lifestyle, which seeks to optimize time, has led the industry to offer elaborated meals with minimal consumer involvement. The industry has been forced to offer new alternatives to produce novel products. One technique already known in the realm of haute cuisine and restaurants is sous vide cooking. Although there are ready-to-heat products prepared with sous vide cooking in some markets, they have not been disseminated worldwide. In this paper the perception of Uruguayan consumers of a product with sous vide cooking was explored. Six Focus Groups (n = 40) were used to explore the generic knowledge of the participants on the different convenience foods and the objective product (sous vide cooking), complemented by the projective technique of completing text in a figure. A relative ignorance of the cooking technique by the consumer – and therefore of its advantages – was determined, with a negative opinion towards keeping/containing food in a bag and a distrust in terms of/distrust regarding the possible use of additives. The sous vide product was associated with a superior or premium quality. In this study, useful information was obtained regarding the introduction of a sous vide cooking product into the market.

Imidazolium salts are best known for their applications in organic synthesis as room-temperature ionicliquids, or as precursors of stable carbenes, but they also show important biological properties suchas anti-oxidative effects, induction of mitochondrial membrane permeabilisation and inhibition of theinfection cycle of Plasmodium falciparum. For these reasons, and since chemotherapy for Chagas diseaseis inefficient, the aim of this study was to test the use of imidazolium compounds against the kineto-plastid haemoflagellate aetiological agent for this disease, namely Trypanosoma cruzi. The results showthat five of the tested compounds are more effective than the reference drug benznidazole against theepimastigote and trypomastigote forms of T. cruzi. Moreover, intracellular amastigotes were also affectedby the compounds, which showed lower toxicity in host cells. Transmission electron microscopy anal-ysis demonstrated that the tested agents induced alterations of the kinetoplast and particularly of themitochondria, leading to extraordinary swelling of the organelle. These results further demonstrate thatthe test agents with the best profile are those bearing symmetrical bulky substituents at N1and N3, dis-playing promising activity against all forms of T. cruzi, interesting selectivity indexes and exceptionalactivity at low doses. Accordingly, these agents represent promising candidates for the treatment ofChagas disease.

The kinetic isotope effects, KIEs, for hydrogen abstraction from the isotopologs CH4, 13CH4, CH3D, and CD4 by chlorine atoms have been studied by the dual-level direct dynamics approach with the MORATE computer program. Low-level calculations of the potential energy surface were carried out at the NDDO-SRP level (in particular the AM1-SRP level), using two different sets of specific reaction parameters labeled SRP4 and SRP13. High-level structural and energetic properties of the reactants, saddle point, and products were obtained at the MP2-SAC and MP2 levels using the 6-311G(2d,d,p) basis set and were used to interpolate corrections to the low-level calculations. The dual-level calculations were carried out using the ICL-Eckart improved interpolated corrections algorithm. Tunneling was included by the microcanonical optimized multidimensional tunneling (μOMT) method, and we find that large-curvature tunneling paths usually provide the dominant contribution, with significant participation of excited vibrational states. Both rectilinear and curvilinear coordinates were applied to the unsubstituted reaction. The 12C/13C KIEs calculated at the MP2-SAC///SRP4 level using MP2 frequencies are in very close agreement with the experimental ones, with values of 1.07 and 1.06 at 243 and 297 K, respectively, as compared to experimental values of 1.07 at both temperatures. For the reaction CH3D + Cl, the calculated H/D KIEs are equal to 1.55 and 1.45 at 223 and 296 K, while the measured values are equal to 1.59 and 1.50, respectively. The H/D KIE for the reaction CD4 + Cl is calculated to decrease from 11 to 4.7 as the temperature increases from 300 K to 450 K, whereas the experimental value decreases from 12 to 3.9. We also make comparison with previous results for the unsubstituted reaction CH4 + Cl.

Two Neisseria gonorrhoeae isolates from Seattle and two isolates from Uruguay were resistant to erythromycin (MIC, 4 to 16 μg/ml) and had reduced susceptibility to azithromycin (MIC, 1 to 4 μg/ml) due to the presence of the self-mobile rRNA methylase gene(s)ermF or ermB and ermF. The two Seattle isolates and one isolate from Uruguay were multiresistant, carrying either the 25.2-MDa tetM-containing plasmid (Seattle) or a β-lactamase plasmid (Uruguay). Sixteen commensal Neisseria isolates (10 Neisseria perflava-N. sicca, 2 N. flava, and 4N. mucosa) for which erythromycin MICs were 4 to 16 μg/ml were shown to carry one or more known rRNA methylase genes, includingermB, ermC, and/or ermF. Many of these isolates also were multiresistant and carried thetetM gene. This is the first time that a complete transposon or a complete conjugative transposon carrying an antibiotic resistance gene has been described for the genus Neisseria.

All rat ovaries tested showed spontaneous contractions both in vivo and in vitro and addn. of oxytocin [50-56-6] stimulated the ovaries by increasing the tone and amplitude and(or) frequency of contractions. In vitro the response to oxytocin was similar for all stages of estrus, while in vivo the response was highest at proestrus. The higher ovarian smooth muscle sensitivity to oxytocin in proestrus suggests the action of the hormone is facilitated near ovulation.

A summary of the resource.
<p>This investigation was designed to determine the influence of the estrous cycle on the response of ovarian smooth muscle to oxytocin. Muscular activity in vivo was studied by recording intraovarian pressure. Intra-aortic injections of oxytocin were given to the rats at all stages of the estrous cycle. The effect of oxytocin was evaluated by the number of responding ovaries and the magnitude of the responses. According to these parameters, a greater response was observed during proestrus, compared with the other cycle stages. The significance of these results is discussed.</p>

Abstract
A summary of the resource.
Oxytocin stimulated ovarian contractility in 29 of 44 animals. Doses of 0.5, 5 and 25 mU/ml were used in all experiments. Statistical analysis showed no significant differences in the sensitivity of the ovaries to oxytocin in the different phases of the estrous cycle. These results suggest that the action of the peptide is not influenced by the hormonal status of the animal.

Efflux transporter and enzyme overexpression can be induced by certain antiepileptic drugs. Phenytoin (PHT) is at the same time substrate and inducer of CYP2C isoenzymes and efflux carriers. Its inductive effect has been postulated to be concentration and timedependent. Since verapamil (VPM) is a well known substrate and inhibitor of P-glycoprotein, its administration could modify PHT systemic exposure. The objective of this work was to determine if single doses (40 mg/kg) of VPM might change PHT body fate in the same way when given at the beginning or several days after 100 mg/kg of PHT daily doses were started. Both drugs were administered intraperitoneally to female Sprague Dawley rats. VPM increased plasma PHT concentrations after one day of treatment, while a decrease in PHT plasma exposure was observed when VPM was added at the fifth day of the antiepileptic treatment. These results suggested that VPM would have different impact on PHT pharmacokinetics, depending on the level of expression of both efflux transporters and enzymes. Before the hepatic cells could acquire a high content of enzymes due to the inductive effect of PHT dosing, VPM decreased the predominant intestinal clearance of PHT. But, once the enzymatic machinery at the hepatocyte became more important than that at the intestine, although ineffective because of the high hepatobiliary efflux transporter overexpression, VPM blockade from the liver resulted in an increased total PHT clearance.

An increased production of cherry and gourmet tomato cultivars that are harvested at advanced colour stages and sold at a higher price has been observed in the last 10 years. In this context, producers need information on the sensory characteristics of new cultivars and their perception by potential consumers. The aim of the present work was to obtain a sensory characterisation of nine cherry tomato cultivars produced under Brazilian organic cultivation conditions from a chef and consumer perspective. Nine organic cherry tomato genotypes were evaluated by ten chefs using an open-ended question and by 110 consumers using a check-all-that-apply question.

Background The horn fly Haematobia irritans is a blood-sucking ectoparasite responsible for substantial economic loss of livestock. Like other hematophagous arthropods species, the successful blood-feeding of H. irritans is highly dependent on the modulation of the host’s hemostasis and immune system. Here, we evaluated the biological activity of hematobin (HTB), a protein recently identified in the H. irritans saliva, on macrophage biology. The goal was to understand the putative interactions between the components of H. irritans saliva and the early host immune responses. Results Thioglycolate-elicited peritoneal macrophages from BALB/c mice were stimulated by lipopolysaccharide (LPS) plus interferon-γ (IFN-γ) in the presence or absence of recombinant HTB. The presence of the salivary protein in the cultures inhibited nitric oxide production and decreased the inducible nitric oxide synthase (iNOS) expression induced by LPS plus IFN-γ. The tumor necrosis factor-α (TNF-α) and interleukin-12p40 (IL-12p40) levels were also reduced in the macrophages pre-incubated with HTB; these findings correlated to the decreased NF-κB expression. The biological activities described here were not associated with changes in annexin V binding to macrophages suggesting that HTB does not induce cell death. In addition, the activity of HTB seems to be specific to macrophages because no changes were observed in lymphocyte proliferation or cytokine production. Conclusions We describe here the first bioactive salivary protein of H. irritans. We characterized its ability to modulate macrophage inflammatory response, and the results can help explain how horn flies modulate the host immune system to feed on blood.

American Trypanosomiasis is a chronic infection discovered and described in 1909 by the Brazilian scientist Carlos Chagas. It is caused by the protozoan parasite Trypanosoma cruzi. Although it affects about 10 million people in Latin America, the current chemotherapy is still inadequate. The discovery of new drugs is urgently needed. Our group is focused on the development of prospective metal-based drugs mainly based on bioactive ligands and pharmacologically interesting metal ions. In this work three new rhenium(I) tricarbonyl compounds fac-[ReI(CO)3Br(HL)] where HL = 5-nitrofuryl containing thiosemicarbazones were synthesized and fully characterized in solution and in the solid state. The in vitro evaluation of the compounds on T. cruzi trypomastigotes (Dm28c strain) showed that the Re(I) compounds are 8 to 15 times more active than the reference drug Nifurtimox and show a 4 to 17 fold increase in activity in respect to the free (HL) ligands. Obtained compounds also show good selectivity indexes (IC50 endothelial cells Ea.hy926/IC50 T. cruzi (Dm28c tripomastigotes)). 1H NMR and MS studies, performed with time, showed that the fac-[Re(CO)3Br(HL)] species convert into the dimers [Re2(CO)6(L)2] in solution. Crystal structure of [ReI2(CO)6(L2)2], the product of complexes' dimerization, was solved. Related to the mechanism of action, the studied compounds do not generate radical oxygen species in the parasite (as 5-nitrofuryl derived thiosemicarbazones do) probably due to the unfavorable nitro reduction potential of the generated dimeric species. On the contrary, the compounds produce a decrease of the oxygen consumption rate of the parasites, maybe inhibiting their mitochondrial respiration.

In the search for a more effective chemotherapy for the treatment of Chagas’ disease and human African trypanosomiasis, caused by Trypanosoma cruzi and Trypanosoma brucei parasites, respectively, the use of organometallic compounds may be a promising strategy. In this work, eight new heterobimetallic compounds are described including four 5-nitrofuryl containing thiosemicarbazones as bioactive ligands (HL1–HL4) and dppf = 1,1′-bis(diphenylphosphino) ferrocene as an organometallic co-ligand. Complexes of the formula [MII(L)(dppf)](PF6) with M = Pd or Pt were synthesized and fully characterized in the solid state and in solution, including the determination of the molecular structure of four of them by single crystal X-ray diffraction methods. Most compounds showed activity in the low micromolar or submicromolar range against both parasites, with the platinum compounds being more active than the palladium analogues. Activity was significantly increased by generation of the M-dppf compounds (3–24 fold increase with respect to free ligands HL for T. cruzi and up to 99 fold increase with respect to HL for T. brucei). The inclusion of the organometallic co-ligand also led to lower toxicity in mammalian cells and higher selectivity towards both parasites when compared to the free HL compounds. The complexes interact with DNA and affect the redox metabolism of the parasites. Furthermore, the most active and selective compound of the new series showed no in vivo toxicity in zebrafish embryos.

Background The timing of milk donations to human milk banks ranges from a few days to more than 1 year after delivery, and the Holder method is used for pasteurization. We evaluated the effect of temporal variation and thermal treatment on the immunological properties of milk. Methods We analyzed 73 milk samples, raw and after pasteurization, donated at different lactation stages. We studied antibodies, lysozyme, cytokines, soluble receptors, and factors with impact on barrier function. We also evaluated in vitro the capacity of milk to modulate nuclear factor-κB (NF-κB) signaling in an HT-29 epithelial cell line stimulated with tumor necrosis factor-α (TNF-α). Results With few exceptions, immune components exhibited their highest levels in colostrum, and were stable in the various stages of mature milk. Pasteurization altered the immunological composition of milk, and very drastically for some components. Raw milk of the first year reduced NF-κB activation in HT-29 cells treated with TNF-α to approximately the same extent, and Holder pasteurization significantly affected this capacity. Conclusions Overall, the present work reports that mature donated milk is equally valuable over the first year of lactation, but warns about drastic losses of anti-inflammatory properties during Holder pasteurization that could be critical for the health of preterm infants.

Introduction: The gastrointestinal and immune systems of premature infants are not fully developed, rendering them more vulnerable to severe complications like necrotizing enterocolitis. Human milk offers a rich array of bioactive factors that collectively contribute to reducing the incidence of gut infections and inflammatory conditions. When a mother's milk is unavailable, preterm infants are often provided with donor human milk processed in Human Milk Banks. However, it remains uncertain whether pasteurized milk confers the same level of risk reduction as unprocessed milk. This uncertainty may stem from the welldocumented adverse effects of heat treatment on milk composition. Yet, our understanding of the comprehensive impact on protective mechanisms is limited. Methods: In this study, we conducted a comparative analysis of the effects of raw versus pasteurized milk and colostrum versus mature milk on cellular functions associated with the gut epithelial barrier and responses to inflammatory stimuli. We utilized THP-1 and HT-29 cell lines, representing monocyte/macrophages and gut epithelial cells, respectively. Results: Our observations revealed that all milk types stimulated epithelial cell proliferation. However, only raw colostrum increased cell migration and interfered with the interaction between E. coli and epithelial cells. Furthermore, the response of epithelial and macrophage cells to lipopolysaccharide (LPS) was enhanced solely by raw colostrum,with a milder effect observedwith mature milk. In contrast, both raw and pasteurized milk diminished the LPS induced response in monocytes. Lastly, we examined how milk affected the differentiation of monocytes into macrophages, finding that milk reduced the subsequent inflammatory response of macrophages to LPS.Discussion: Our study sheds light on the impact of human milk on certain mechanisms that potentially account for its protective effects against necrotizing enterocolitis, highlighting the detrimental influence of pasteurization on some of these mechanisms. Our findings emphasize the urgency of developing alternative pasteurization methods to better preserve milk properties. Moreover, identifying the key components critically affected by these protective mechanisms could enable their inclusion in donor milk or formula, thereby enhancing immunological benefits for vulnerable newborns.
