Los parásitos helmintos expresan diferentes glicoconjugados en su superficie y liberan productos de excreción-secreción ricos en diferentes glicanos que pueden ser muy importantes en su ciclo de vida, infección y patología. Evidencias recientes sugieren que los glicoconjugados de parásitos helmintos podrían desempeñar un papel en la evasión de la respuesta inmune, lo que lleva a una polzarización de las respuestas inmunes hacia un perfil Th2 modificado, que favorece la supervivencia del parásito en el hospedero. Sin embargo, existe poca información sobre la naturaleza o función de glicanos producidos por el trematodo Fasciola hepatica, el agente causante de la fasciolosis en nuestro país. En esta tesis, se investigó si las moléculas glicosiladas de F. hepatica participan en la modulación de la inmunidad del hospedero, enfocándonos en las células dendríticas. Nuestros resultados indican que glycoconjugados de F. hepatica son capaces de promover la producción de IL-4 e IL-10, al igual que suprimir de la producción de IFNγ. Durante la infección, este parásito es capaz de inducir un fenotipo semi-maduro en células dendríticas que expresan bajos niveles de MHCII y secretan IL-10. Además, se observó que los glicoconjugados parasitarios median la modulación de la maduración de células dendríticas inducida por LPS, ya que su oxidación con m-periodato de sodio restaura la capacidad de las células dendríticas tratadas con LPS para secretar altos niveles de las citoquinas pro-inflamatorias IL-6 e IL-12/23p40 y bajos niveles de la citoquina antiinflamatoria IL-10. Los ensayos de inhibición utilizando carbohidratos sugieren que la modulación inmune está mediada, al menos en parte, por el reconocimiento de CLRs específicos de manosa y mediante la senalización a través de la fosfatasa SHP2. Con el fin de identificar las estructuras glucídicas que median este efecto, se utilizaron glicosidasas específicas específicas (α-manosidasa y β-galactosidasa), que fueron inmovilizadas sobre nanopartículas magnéticas. La eficacia de las glicosidasas inmovilizadas se demostró mediante el estudio de la degalactosilación y demanosilación selectiva de glicoproteínas modelo. Por último, la desglicosilación específica de un extracto total del parásito nos permitó mostrar la importancia de glicanos manosilados en la inducción de Th2 durante la infección. Además, se evaluaron las propiedades inmunomoduladoras de los glicanos de F. hepatica sobre células dendríticas humanas, mostrando que éstos son capaces de aumentar la producción de las citoquinas IL-10 e IL-27p28 inducidas por ligandos de TLR. Asimismo, se observó que las propiedades inmunomoduladoras de glicanos están mediadas DC-SIGN, que reconoce glicanos pararasitarios que contienen fucosa y manosa. Por otra parte, los glicoconjugados de F. hepatica inducen a través de DC-SIGN un programa regulador sobre las células dendríticas, capaces de Resumen Página xii disminuir la proliferación de células T alogénicas, lo cual pone de manifiesto el papel de DC-SIGN en la regulación de la respuesta inmune innata y adaptativa por F. hepatica. Por último, se observó que un homólogo murino de DC-SIGN, CD209a, limita la producción de IL-12 por BMDCs pero no influye en su capacidad de estimular linfocitos T CD4+. Por otro lado, se observó un efecto diferente en las células dendríticas cuando se utilizan ligandos parasitarios inmovilizados. Nuestros resultados sugieren que el antígeno Tn expresado en la superficie de F. hepatica puede modular la maduración de las células dendríticas inducida por ligandos de TLR, en un proceso mediado por hMGL, conduciendo a un aumento en la producción de IL-10 y TNFα, y a la capacidad de dichas células de polarizar respuestas Th2. Asimismo, se observó que células CD11c+ mMGL2+ F4/80low son reclutadas al peritoneo de ratones infectados, las cuales expresan las citoquinas reguladoras IL-10, TNFα y TGFβ, al igual que una variedad de marcadores asociados a la regulación del sistema inmune y presentan una menor capacidad estimuladora de linfocitos T alogénicos. Los resultados presentados aquí contribuyen a comprender mejor la función de las moléculas glicosiladas de F. hepatica en la modulación de la inmunidad del hospedero y podrían ser útiles en el diseño de vacunas contra la fasciolos

A summary of the resource.
Glass ceramics of PbB4O7 doped with 0.15 and 0.50% molar concentration of Ce and Eu were prepared by means of controlled crystallization of the glass powder. Crystallization was carried out at 590 °C for the undoped and at 640 °C for the doped glass ceramic samples. Heat treatment time was 60 and 180 min for both kinds of samples. Crystalline phase analysis reveals that the only present phase was lead diborate. Thermal analysis characterization shows that Europium may act as nucleation promoter, lowering the energy barrier for crystallization. On the other hand Cerium acts as inhibitor of the crystallization process. Optical characterization was done by means of absorption spectroscopy and luminescence measurements at room temperature. A shift in absorption edges was observed for both samples; this shift could be attributed to a large amount of BO4 groups as dopant concentration increases. From the luminescence spectra it was showed that the 5D0 to 7F2 red emission (613 nm) occurs through an electric dipole mechanism which is dominant in all the Europium doped samples, while in Cerium doped samples the emission peaks are due to the 5d–2F5/2 (528 nm) and 5d–2F7/2 (472 nm) transitions. Thermoluminescence analysis was performed with different irradiation doses (1 and 10 Gy) on samples at different concentrations. Results show the incorporation of localized states near the conduction band.

A summary of the resource.
Borate glasses with the composition MO·2B2O3 (M: Pb, Sr and Ba) and N2O·2B2O3 (N: Li and Na) were prepared. Glass-ceramics of the same composition were obtained by heat treating glass powder. The structure of the glasses, was studied by FTIR and 11B NMR resulted in a similar structure (having BO3 and BO4 species). The PbO·2B2O3 sample shows the lowest fraction of four coordinated boron, N4, of the set. Glass crystallization was analyzed by thermal analysis, using the Kissinger method. The evaluated activation energy shows that the BaO·2B2O3 sample presents the highest value; meanwhile Li2O·2B2O3 shows the lowest one. Thermally stimulated luminescence was measured in glasses and glass-ceramics using a 90Sr radiation source. In most cases, glass-ceramics present a higher sensitivity than glasses. Defects present in glasses are increased by the crystallization to improve the TL response of these materials. When we compare the response, as the area under the curve, (which is related to the absorbed dose) barium borate glass-ceramics present the best response compared to the other studied borates.

Photodynamic therapy has advantages over conventional cancer superfcial tumor therapies, such as insignifcant side efects, minimal cumulative toxicity, excellent functional and cosmetic results, precise target treatment minimizing damage to neigh boring normal tissues, and optimum long-term tumor regression. Upconversion nanoparticles have been proposed to extend photodynamic therapy for treating non-superfcial tumors. In particular, compounds with low phonon energy, like KMgF3, with an optimal synthesis design and a suitable doping system, could be used for this therapy application. This particular matrix allows the incorporation of dopants as transition metals ions (as Mn2+) besides rare-earth elements, minimizing the use of the former. To obtain nanoparticles of suitable size for nanomedicine, we used a factorial experimental design to determine statistical synthesis conditions that signifcantly afect particles’ size. With the proposed synthesis method, it was possible to obtain KMgF3 nanoparticles with sizes ranging from 13.46±0.30 nm to 32.18±0.60 nm, values estimated with the XRD technique and with size distribution suitable for photodynamic therapy. We proved the good correlation between the particle size estimated from transmission electron microscopy images and powder X-ray difraction measurements. According to the statistical analysis, the temperature and the interaction of temperature with MgCl2 concentration signifcantly afect the particle size (signifcance level of 0.05). The present work describes the infuence of solvothermal synthesis parameters on the KMgF3 nanoparticle size for the frst time. The results are particularly interesting for further doping the system and its functionalization, foreseeing the fnal application in cancer treatment.

The preparation of oxyfluoroborate glasses and glass–ceramics doped with Yb3+ and Er3+ with improved luminescent properties is reported. Glasses with compositions (100 − x − y) (MO·2B2O3) + xPbF2 + y(YbF3:ErF3) (% in mol), where M is Sr, Ba, Ca, x = 10–40% and y = 0, 1 or 5% were prepared by the melt/quenching technique. Glass crystallization was studied using thermal analysis and X-ray diffraction techniques. Optical absorption and infrared up-conversion were studied on both glasses and glass–ceramics. The incorporation of a fluoride compound into the borate glass resulted to depend on the ionic radius of the glass modifier: as it increases, glasses become more stable against crystallization. On the other hand, alkali–fluoride compounds such as BaF2 and SrF2 can be crystallized from these systems by selecting an appropriate proportion between their components. Furthermore, the up-conversion response can be tuned by changing the glass modifiers (Sr, Ba and Ca) of the borate matrix, which also influence the type of fluoride crystallized compound. The strontium glass–ceramics have the highest luminescence response due to the crystallization of SrF2 compound in the system SrO–B2O3–PbF2. Meanwhile, the lowest luminescence signal was obtained for samples in the system BaO–B2O3–PbF2 where the Pb1.33Ba2.66B11FO20 phase crystallizes. With this strategy, new materials with improved luminescence properties that can be used as up converters, were obtained.

In this work we present the preparation and characterization of new fluoroborate glasses doped with Yb3 + and Er3 + and modified by Ca, Sr and Ba oxides. Glasses with compositions (100-x-y) (MO ⋅ 2B2O3) + x NaF + y (YbF3:ErF3) (% in mol), where M is Sr, Ba, Ca, x = 10–30% and y = 0, 1 or 5% were prepared by the melt/quenching technique. Glass forming ability and stability were studied by thermal analysis and it is shown that calcium samples presented the highest stability. The nature of the modifier cation has also shown a direct influence on the absolute and relative intensities of the rare earth ions transitions in the visible and near-infrared regions. Though the calcium samples present lower absorption intensities, they display the best up- and down-conversion efficiencies. There are evidences that the incorporation of the fluoride compound leads to a fluorine rich environment on the first coordination sphere of the RE, decreasing the probability of OH– groups – well known quenchers of RE emissions. Though preliminary, the results in this study can become of relevance for the field of solar cell technology improvement via betterment of solar light harvesting in the infrared and ultraviolet regions.

Response surface methodology (RSM) was applied to optimise laccase production by a new strain of Pycnoporus sanguineus, isolated from the Uruguayan forests, grown on bark shavings under semi-solid-state fermentation conditions. Enhancement of laccase production was studied, with a full 24 factorial design with two replicates and four central points. Operating under the optimal conditions obtained (pH 3.0, yeast extract 10 g L−1, glucose 5 g L−1 and bark shavings 3 g), a laccase activity of nearly 3000 U L−1 was achieved. Subsequently, the effect of different potential inducers on laccase production (Cu+2, Mn+2 and lignin) was tested using the one-factor-at-a-time approach. It was found that the addition of 0.5 mM Cu+2 into the cultures considerably increased laccase production and, thus, maximum activity values of about 16 000 U L−1 were achieved. Further, the optimised culture broth (containing mainly laccase) showed high capacity to decolourise different recalcitrant synthetic dyes such as Acid Red 88 (AR88), Reactive Black 5 (RB5) and Lanaset Grey G (LG). After selecting enzyme concentration and time of reaction, almost 70% of AR88 could be decolourised in the absence of redox mediators. However, mediators were necessary to bleach RB5 and LG. Methyl syringate was the most effective mediator for AR88 (reaching a decolouration degree over 90%) and RB5 (nearly 70%). Around 80% of LG could be decolourised in the presence of violuric acid. These promising results make this enzyme a very useful tool for developing wastewater treatments.

Snakins are antimicrobial peptides (AMPs) found, so far, exclusively in plants, and known to be important in the defense against a wide range of pathogens. Like other plant AMPs, they contain several positively charged amino acids, and an even number of cysteine residues forming disulfide bridges which are considered important for their usual function. Despite its importance, studies on snakin tertiary structure and mode of action are still scarce. In this study, a new snakin-like gene was isolated from the native plant Peltophorum dubium, and its expression was verified in seedlings and adult leaves. The deduced peptide (PdSN1) shows 84% sequence identity with potato snakin-1 mature peptide, with the 12 cysteines characteristic from this peptide family at the GASA domain. The mature PdSN1 coding sequence was successfully expressed in Escherichia coli. The purified recombinant peptide inhibits the growth of important plant and human pathogens, like the economically relevant potato pathogen Streptomyces scabies and the opportunistic fungi Candida albicans and Aspergillus niger. Finally, homology and ab initio modeling techniques coupled to extensive molecular dynamics simulations were used to gain insight on the 3D structure of PdSN1, which exhibited a helix–turn–helix motif conserved in both native and recombinant peptides. We found this motif to be strongly coded in the sequence of PdSN1, as it is stable under different patterns of disulfide bonds connectivity, and even when the 12 cysteines are considered in their reduced form, explaining the previous experimental evidences.

They are cysteine-rich molecules, highly sequence diverse but with a conserved and stable structure. In this work, a defensin gene (EcgDf1) was isolated from Erythrina crista-galli, a legume tree native from South America. The predicted peptide presents eight cysteines, with a γ-core motif GXCX3-9C and six cysteines distributed like the typical defensin αβ motif. The mature EcgDf1 coding sequence was heterologously expressed in Escherichia coli strains and purified by affinity chromatography. Possible dimer and oligomers of EcgDf1 were visible in SDS electrophoresis. Moreover, its 3D structure, determined by homology modeling, docking, and molecular dynamics simulations, was found to be compatible with the formation of homodimers between the β3 and β1-loop-α1, leaving the β2-loop-β3 free to interact with lipid membranes. The purified recombinant peptide inhibited the growth of several critical plant and human pathogens, like the opportunistic fungi Candida albicans and Aspergillus niger and the plant pathogens Clavibacter michiganensis ssp. michiganensis, Penicillium expansum, Botrytis cinerea, and Alternaria alternata. EcgDf1 is a promising candidate for the development of antimicrobial products for use in agriculture and medicine.

Inflammation plays a major role in the onset and development of chronic non-communicable diseases like obesity, cardiovascular diseases and cancer. Combined, these diseases represent the most common causes of death worldwide, thus development of novel pharmacological approaches is crucial. Electrophilic nitroalkenes derived from fatty acids are formed endogenously and exert anti-inflammatory actions by the modification of proteins involved in inflammation signaling cascades. We have developed novel nitroalkenes derived from α-tocopherol aiming to increase its salutary actions by adding anti-inflammatory properties to a well-known nutraceutical. We synthesized and characterized an α-tocopherol-nitroalkene (NATOH) and two hydrosoluble analogues derived from Trolox (NATxME and NATx0). We analyzed the kinetics of the Michael addition reaction of these compounds with thiols in micellar systems aiming to understand the effect of hydrophobic partition on the reactivity of nitroalkenes. We studied NATxME in vitro showing it exerts non-conventional anti-inflammatory responses by inducing Nrf2-Keap1-dependent gene expression and inhibiting the secretion of NF-κB dependent pro-inflammatory cytokines. NATxME was also effective in vivo, inhibiting neutrophil recruitment in a zebrafish model of inflammation. This work lays the foundation for the rational design of a new therapeutic strategy for the prevention and treatment of metabolic and inflammation-related diseases.

<p>Chronic metabolic diseases, like obesity, type II diabetes and atherosclerosis often involve a low-grade and sterile systemic inflammatory state, in which activation of the pro-inflammatory transcription factor NF-kB and the NLRP3 inflammasome play a major role. It is well established that genetic inhibition of the NLRP3 inflammasome ameliorates acute and chronic inflammation. Indeed, accumulating experimental evidences in murine models and also in humans suggest that inhibition of the NLRP3 inflammasome might be a suitable approach to tackle the deleterious effects of chronic metabolic diseases. In this work, we explored our previously synthesized nitroalkene-Trolox™ derivative named NATx0, as a non-conventional anti-inflammatory strategy to treat chronic inflammatory diseases, such as obesity-induced glucose intolerance. We found that NATx0 inhibited NF-kB nuclear translocation and pro-inflammatory gene expression in macrophages in vitro. In addition, treatment with NATx0 prevented NLRP3 inflammasome activation after LPS/ATP stimulation in macrophages in vitro. When tested acutely in vivo, NATx0 inhibited neutrophil recruitment in zebrafish larvae, and also diminished IL- 1β production after LPS challenge in mice. Finally, when NATx0 was administered chronically to diet-induced obese mice, it decreased muscle tissue inflammation and glucose intolerance, leading to improved glucose homeostasis. In conclusion, we propose that this novel nitroalkene-Trolox derivative is a suitable tool to tackle acute and chronic inflammation in vitro and in vivo mainly due to inhibition of NF-kB/NLRP3 activation.</p>

Enamel defects in permanent and deciduous teeth may be oral manifestations of celiac disease. Sometimes they are the only sign that points to this underdiagnosed autoimmune pathology. However, the etiology of these specific enamel defects remains unknown. Based on previously reported cross-reactivity of antibodies to gliadin with the enamel proteins, amelogenin and ameloblastin, we analyzed (using immunohistochemistry) the ability of anti-gliadin IgG, produced during untreated disease, to recognize enamel organ structures. We used swine germ teeth as a tissue model because they are highly homologous to human teeth in terms of proteins and development biology. Strong staining of the enamel matrix and of the layer of ameloblasts was observed with serum samples from women with celiac disease; high IgG reactivity was found against both gliadin peptides and enamel matrix protein extract, but there was no IgG reactivity against tissue antigens. In line with these findings, the gamma globulin fraction from gliadin-immunized BALB/c mice showed a similar staining pattern to that of amelogenin-specific staining. These results strongly suggest a pathological role for antibodies to gliadin in enamel defect dentition for both deciduous and permanent teeth, considering that IgG can be transported through the placenta during fetal tooth development.

El color es un atributo importante en la elección del producto dulce de leche por parte del consumidor. El mismo se desarrolla en el proceso de elaboración debido principalmente a las reacciones de pardeamiento no enzimático, las cuales se ven influenciadas por varios factores. En este trabajo se evaluó el desarrollo de color, con el tiempo de tratamiento térmico y el pH inicial como variables tecnológicas, en un sistema modelo compuesto de caseinato, lactosa, sacarosa y agua destilada cuya composición fue similar a la del dulce de leche comercial. El estudio del color se realizó utilizando los parámetros de medición de color del sistema CIELAB y el índice de Kubelka Munk (K/S). Con ambos métodos se constató un aumento significativo del dE*ab y del K/S con el tiempo de tratamiento térmico y el pH inicial. Con el sistema CIELAB además se observó una tendencia a disminuir la luminosidad con el tiempo y el aumento del pH inicial, y el aumento de a* con el tiempo. Browning and gelation kinetics of a sheep milk and sucrose model system with 70% total solids (simulating dulce de leche) and the influence of temperature and sucrose content on this process were studied. The Kubelka–Munk index and subjective heat stability tests were used to monitor nonenzymatic browning and to determine gelation time. Browning and gelation processes showed different kinetics where gelation was shown to occur much faster than browning. Both independent variables (temperature and sucrose content) had a significant influence in both processes, where temperature had the higher impact.

Melanoidins, the brown-colored compounds formed through the Maillard reaction, are responsible for color development in dulce de leche (DL), a popular confectionary dairy product in the Río de la Plata region, particularly in Uruguay and Argentina. Color is a critical quality parameter that strongly influences consumer preference. This work aimed to develop a method to perform preliminary structural characterization of the chromophores produced by the Maillard reaction. Melanoidins are present in a water-insoluble fraction, linked to a protein backbone, conforming melanoproteins of high molecular weight. The insoluble melanoprotein fraction (10% total solids) was isolated, and the chromophores released by proteolysis and isolated by gel-permeation chromatography. The analysis of the products revealed that they present a high degree of molecular weight (MW) polydispersity, in a range of 300 to 2000 Da, where the compounds of higher molecular weight contributed the most to the color of the product. The isolated fractions were further analyzed by RP-HPLC using a diode array detector (DAD) detector. These results, together with H-NMR data, suggested that the chromophores isolated belonged to a relatively simple mixture of aromatic products with higher hydrophobic character relative to other products of the melanoprotein digestion.

Holder pasteurization, a process that consists of heating the sample at 62.5 ◦C for 30 min, is the most widely used protocol to ensure microbial inactivation in human milk. Aiming to evaluate vitamin losses after pasteurization, three extraction methods and the subsequent chromatographic conditions were optimized. For the simultaneous determination of retinol, α-tocopherol, and β-carotene a saponification procedure was employed, while folic acid and ascorbic acid were extracted using solvent extractions, followed by high-performance liquid chromatography – diode array detection.
