Holistic methodologies such as Projective Mapping and sorting have gained popularity for sensory characterization of products with both trained assessors and consumers. One of their main disadvantage is that all samples should be simultaneously evaluated in the same session. An alternative to overcome this limitation is to evaluate samples by comparing them with a fixed set of products, as proposed in Polarized Sensory Positioning (PSP). In the present work a combination of Projective Mapping and Polarized Sensory Positioning, called polarized Projective Mapping (PPM), is presented and compared with the two original ways of performing PSP (scale based and triadic-PSP), in terms of conclusions regarding differences between samples and difficulty for consumers. Nine orange-flavored powdered drink samples (including one blind repeated sample) were evaluated by three groups of 45 consumers using PSP, triadic-PSP and PPM using a between-subjects design. Although the three methodologies provided similar sensory spaces, some differences were identified in terms of discriminative ability, conclusions regarding similarities between samples and perceived difficulty. Polarized Projective Mapping seems to be an interesting approach that combines the advantages of Polarized Sensory Positioning and the holistic character of Projective Mapping, providing the possibility of comparing samples evaluated in different sessions.

Polarized sensory positioning (PSP) is a novel methodology for sensory characterization which is based on the comparison among samples and a set of reference products, called poles. This is particularly useful when working with samples with persistent flavours, when evaluating a large number of samples over a long period of time, or during quality control. Although the key step of this methodology is the selection of the products to be considered as poles, no studies have been published addressing this issue. In this context, the aim of the present work was to contribute to bridging this gap by studying the influence of the poles on results from sensory characterization of two widely different products (make-up foundations and orange-flavoured powdered drinks) using Polarized Sensory Positioning with consumers. Results from both studies provided preliminary evidence about the stability of sensory spaces obtained with different set of poles (RV coefficients higher than 0.908). However, differences in conclusions regarding similarities and differences among samples were identified between the sets of poles in both studies, suggesting the need to carefully select stable poles for PSP tasks.

<p>A practical method is described for the extraction of uranium from monazite sands and its subsequent polarographic determination.</p>
<p>The sample is decomposed by fusion with potassium hydrogen difluoride followed by fusion with potassium pyrosulphate. Uranium is extracted from nitric acid solution with tributyl phosphate in 2,2,4-trimethylpentane, with aluminium nitrate as a salting-out agent, and back-extracted from the organic phase with water.</p>
<p>The final polarographic determination is carried out in 2 M acetic acid-2 M ammonium acetate-0·1 M ascorbic acid solution as supporting electrolyte. Neither a maximum suppressor nor removal of oxygen is needed.</p>
<p>The interference by lead and some factors influencing the extraction of uranium are studied.</p>
<p>The results are reproducible and agree with those obtained by other, more laborious, techniques. The proposed procedure is suitable for the determination of uranium in monazites and monazite sand concentrates containing not less than 0·005 per cent. of uranium oxide, and is superior in speed, reliability and convenience to other methods previously reported.</p>

Polarized Sensory Positioning (PSP) is a relatively new methodology for sensory characterization, based on the comparison of samples to reference products, usually called poles. The key step in the implementation of this methodology is the selection of the poles. In this context, the aim of the present work was to gain insights on the strategies used by consumers to complete this task and to study the influence of the number and characteristics of the poles on results from PSP. Three consumer studies were carried out, on three product categories: chocolate-flavored milks, vanilla milk desserts and orange-flavored powdered drinks. In each study, three groups of consumers (n = 40) evaluated samples using one of three sets of two or three poles. After the PSP task consumers indicated how they evaluated the degree of difference between the samples and the poles, and listed the sensory characteristics of the poles they took into account for the evaluation. Consumers tended to evaluate the degree of difference between the samples and each of the poles by evaluating the intensity of one or two main sensory characteristics. Sample configurations obtained using two and three poles did not largely differ, especially when the poles clearly represented the sensory characteristics responsible for the main differences among samples. This work demonstrates that it is possible to unfold a multidimensional sensory space with the use of just two well-selected poles, and that the number of poles should be selected considering the main sensory characteristics that discriminate among samples.

Introducción: la apolipoproteína E (ApoE) es un constituyente de las lipoproteínas plasmáticas con un importante rol en el metabolismo como ligando de receptores de lipoproteínas. Las variaciones en el locus de ApoE se vinculan a variaciones en las concentraciones de lipoproteínas de baja densidad (LDL) plasmáticas. El alelo E4 se ha vinculado a aumento del riesgo cardiovascular, a progresión de la nefropatía diabética así como a severidad y progresión de neuropatía y retinopatía diabéticas. El alelo E2 se asocia a un efecto final protector para el desarrollo de enfermedad cardiovascular. Objetivos: estudiar la asociación entre este polimorfismo y las repercusiones macroangiopáticas, en especial cardiopatía isquémica, y el lipidograma en los pacientes diabéticos. Material y método: se estudiaron 78 pacientes diabéticos tipo 2, menores de 85 años de edad, valorando la asociación entre el polimorfismo de ApoE y las repercusiones macroangiopáticas y microangiopáticas de la diabetes, el lipidograma y la edad al diagnóstico de la diabetes. La determinación genotípica se realizó por métodos estándar de amplificación- restricción previo consentimiento informado. Las variables se analizaron utilizando chi cuadrado y prueba de Fisher. Error alfa menor a 0,05. Resultados: la frecuencia de portadores del alelo E4 de la Apo E entre los individuos con complicaciones macroangiopáticas (43,5%) fue mayor que entre los que estaban libres de estas complicaciones (26,2%), si bien las diferencias no fueron estadísticamente significativas. La presencia de cardiopatía isquémica se asoció en forma significativa con el alelo E4, 56% versus 25%. La presencia del alelo E4 se asoció significativamente con un diagnóstico de diabetes antes de los 40 años. Los pacientes con alelos de riesgo presentaron valores de LDL y triglicéridos (TG) aumentados. Conclusiones: el alelo E4 se asoció a un mayor riesgo de cardiopatía isquémica y una edad al diagnóstico de DM2 menor a 40 años. Se evidenció un efecto marcado del genotipo de ApoE sobre el perfil lipídico de los pacientes diabéticos tipo 2.

Significance: Major pathogenic enterobacteria and protozoan parasites from the phylum Euglenozoa, such as trypanosomatids, are endowed with glutathione (GSH)-spermidine (Sp) derivatives that play important roles in signaling and metal and thiol-redox homeostasis. For some Euglenozoa lineages, the GSH-Sp conjugates represent the main redox cosubstrates around which entire new redox systems have evolved. Several proteins underwent molecular adaptations to synthesize and utilize the new polyamine-based thiols. Recent Advances: The genomes of closely related organisms have recently been sequenced, which allows mining and analysis of gene sequences that belong to these peculiar redox systems. Similarly, the three-dimensional structures of several of these proteins have been solved, which allows for comparison with their counterparts in classical redox systems that rely on GSH/glutaredoxin and thioredoxin. Critical Issues: The evolutionary and structural aspects related to the emergence and use of GSH-Sp conjugates in Euglenozoa are reviewed focusing on unique structural specializations that proteins developed to use N1,N8-bisglutathionylspermidine (trypanothione) as redox cosubstrate. An updated overview on the biochemical and biological significance of the major enzymatic activities is also provided. Future Directions: A thiol-redox system strictly dependent on trypanothione is a feature unique to trypanosomatids. The physicochemical properties of the polyamine-GSH conjugates were a major driving force for structural adaptation of proteins that use these thiols as ligand and redox cofactor. In fact, the structural differences of indispensable components of this system can be exploited toward selective drug development. In this report, we present the synthesis of few layer MoS2 and polyaniline/MoS2 nanocomposites with a complete structural characterization by atomic force microscopy, small-angle X-ray scattering, X-ray diffraction, high-resolution transmission electron microscopy, Raman spectroscopy followed by AC impedance spectroscopy and thermoelectric characterizations. Our structural studies revealed that the intercalation of MoS2 nanosheets into polyaniline fibers produces a decrease in the MoS2 interlayer distance and a slight increase in the degree of order of the nanocomposites. In addition, an increment in the polaron Raman signature was observed with the addition of MoS2 nanosheets into polyaniline nanocomposites in agreement with the enhancement of the electrical conductivity, i.e. up to ~ 2.5 S cm−1. The addition of low amounts of MoS2 also lead to an increment of the seebeck coefficient (up to 5.3 µV K−1) and power factor (up to 0.0070 µV m−1 K−2). This report showed excellent synergistic effect between PANI and MoS2 nanosheets which could be of great importance for future applications of these nanocomposites, mainly as thermoelectric materials.

To date, there are a few technologies for the development of noncompetitive immunoassays for small molecules, the most common of which relies on the use of anti-immunocomplex antibodies. This approach is laborious, case specific, and relies upon monoclonal antibody technology for its implementation. We recently demonstrated that, in the case of monoclonal antibody-based immunoassays, short peptide loops isolated from phage display libraries can be used as substitutes of the anti-immunocomplex antibodies for noncompetitive immunodetection of small molecules. The aim of this work was to demonstrate that such phage ligands can be isolated even when the selector antibodies are polyclonal in nature. Using phenoxybenzoic acid (PBA), a major pyrethroid metabolite, as a model system, we isolated the CFNGKDWLYC peptide after panning a cyclic peptide library on the PBA/anti-PBA immunocomplex. The sensitivity of the noncompetitive enzyme-linked immunosorbent assay (ELISA) setup with this peptide was 5-fold (heterologous) or 400-fold (homologous) higher than that of the competitive assay setup with the same antibody. Phage anti-inmunocomplex assay (PHAIA) was also easily adapted into a rapid and highly sensitive dipstick assay. The method not only provides a positive readout but also constitutes a major shortcut in the development of sensitive polyclonal-based assays, avoiding the need of synthesizing heterologous competing haptens.

<p>The aim of this study was to assess the risk of exposure to polycyclic aromatic hydrocarbons (PAH) from yerba mate infusions in Uruguay using the margin of exposure approach (MOE) and a probabilistic method (Monte Carlo simulation). Servings/day, portion size, weekly frequency of mate consumption and body weight were the factors considered. The amount in infusions of benz[a]pyrene (B[a]P), PAH2 (sum of chrysene and B[a]P), and PAH4 (sum of benz[a]anthracene, chrysene, benz[b]fluoranthene and B[a]P) were used as markers of PAH exposure. Total content of PAH in infusions had large inter-brand variability (48–54 %) with significant differences among brands. PAH content in infusions prepared as habitually consumed was about 40 % of total content. The probability of occurrence of MOE < 10,000 varied according to the infusion preparation and the marker of exposure used, being higher for infusions prepared for total content and when B[a]P was used as marker of exposure. When the average B[a]P amount in infusion as habitually consumed was used in the simulation model, the probability of MOE < 10,000 was 9 %. The main factors contributing to B[a]P MOE variance were B[a]P amount (28.4 %), servings/day (17.3 %), and portion size (9.6 %). Heavy drinkers of yerba mate with high B[a]P content are those at risk to PAH exposure from mate infusions.</p>

With the aim to improve the performance of enzyme bound to hydrophilic solid phases, their immobilization with polyethylene glycol (PEG) tether have been studied. Sweet potato �-amylase, which hydrolyses the high molecular weight substrate starch and �-galactosidase, which acts on low molecular weight substrates, were used as model enzymes and beaded thiol–agarose as solid phase. Several two step methods for the introduction of the tether using a bis-oxirane homobifunctional PEG as well as a heterobifunctional derivative with a hydroxysuccinimide ester and a maleimide group have been evaluated. Amino groups, native and de novo thiol groups in the enzymes were utilized for immobilization. The best approach was found to be to first introduce the PEG derivative via one of its reactive groups to the enzyme. Subsequently the formed conjugate was bound to the solid phase by the remaining reactive group. Attempts to first introduce the PEG tether into the solid phase were not successful. Ahigh degree of substitution with PEG chains on the enzyme leads to high immobilization yields for both �-amylase and �-galactosidase, but relatively lower gel-bound activity for the former enzyme which is acting on a high molecular weight substrate and thus more sensitive for steric shielding effects. With optimal degree of PEG substitution (which occurred at five times molar excess of the heterobifunctional reagent) the gel-bound activity of �-amylase was increased from 12% (for the derivative without tether) to 31%.

mmobilised enzymes for industrial applications should be very active and operationally stable. Reversible binding, if possible, is desirable to allow matrix reuse. Usually these aims come into conflict: strong binding may imply low expressed activity and poor reversibility. Immobilisation of Aspergillus oryzae β-galactosidase to conventional ionic exchangers is efficient but the derivatives exhibit noticeable enzyme leakage when used for the hydrolysis of lactose from whey. This problem can be avoided by covalent cross-links to the matrix, at the expense of losing reversibility. We have optimised the conditions for polyethylenimine (PEI)-coating of agarose supports to achieve a β-galactosidase derivative that allows a high lactose conversion from whey in a steady bed-reactor with no enzyme leakage, together with good elution properties. Therefore, PEI-coating appears to be a very suitable strategy to convert the agarose surface to an adequate support for a reversible, but strong and non-distorting, ionic protein immobilisation.

Monodisperse cationic core-shell latex particles have been prepared using a shot polymerization process, with N-(3-aminopropyl)-methacrylamide-hydrochloride (APMH) as the functional monomer. The final latexes were characterized with respect to final polymerization conversion, water soluble polymer formation, particle size and size distribution, surface charge density and electrokinetic properties. Then the covalent grafting of maleic anhydride-alt-methyl vinyl ether (MAMVE) copolymer onto aminated latex particles was investigated. The most efficient conditions to obtain derivatised particles with no alteration of the colloidal stability were to control both polymer amount/latex particles concentration ratio and the mixing method of the two species. The charge inversion of the hydrolysed MAMVE functionalized particles was demonstrated by measuring the electrophoretic mobility as a function of pH. Finally, the covalent binding approach was implemented with peptide-MAMVE conjugates, confirming the great potential of this promising methodology for the preparation of reactive latex particles bearing peptides.

Using the flexible ligand oxydiacetic acid (H2oda), ten new heteropolynuclear compounds with general formula [Ln2Zn3(oda)6]·xH2O (Ln = lanthanide ion, oda = oxydiacetate) were synthesized and structurally characterized. They were prepared by the direct reaction of oda in aqueous solution with stoichiometric amounts of Zn and Ln salts, followed by slow organic solvent diffusion. Two different structures were obtained: the larger lanthanides Pr, Nd, Eu and Tb gave compounds belonging to the hexagonal system, space group P6/mcc; whereas, the smaller ones, Ho, Er, Yb and Y formed cubic systems, space group Fd[3 with combining macron]c. In the case of Dy, the two polymorphs were obtained by slight variations in the synthetic conditions. The phase change observed is discussed in comparison with previously reported [Ln2M3(oda)6]·xH2O compounds (M = bivalent metal ion). Luminescence studies were also carried out. Compounds containing Eu and Tb showed the most intense luminescence, also bearing relatively long lifetimes for the excited state deactivation. The luminescence profiles of both Dy systems are independent of the hexagonal or cubic arrangements, due to the similar environment of the Ln having the same point symmetry.

A summary of the resource.
The nitrilotripropionic acid (H3ntp) is a flexible polycarboxylate ligand that has been used in the preparation of polynuclear compounds with several metal cations. In this work, we present the in solution study of the interaction of ntp with a wide range of metal ions (M): Mn(II), Fe(II), Co(II), Ni(II), Cu(II), Zn(II), Cd(II), Pb(II) and Fe(III). The species [Mntp]−, [M(Hntp)], and [M3(Hntp)2]2+ were detected for most metal ions. In the case of Cu(II), the potentiometric model was also supported through additional thermodynamic characterization by spectroscopy. We also performed computational calculations at the UB3LYP/LANL2DZ SMD level of theory that allowed us to predict the electronic spectra of the proposed species. The computational model showed a satisfactory performance, as demonstrated by the calculated electronic spectra fit to the experimental data. A possible assembly mechanism for the reported capsule-shaped complex [Cu3(Hntp)2]2+ is proposed.

Abstract
A summary of the resource.
Polyreactive antibodies (pAb) bind to a broad range of unrelated structures, providing<br />hosts with functional components able to rapidly recognize and protect against different<br />pathogens. However, their roles against helminth parasites are still unexplored.<br />Here, pAb profiles were analysed in cystic echinococcosis (CE), a zoonosis caused by<br />the cestode Echinococcus granulosus sensu lato. Levels of anti-DNP (2,4-dinitrophenyl-<br />hapten) antibodies were measured as a surrogate parameter of pAb in different<br />biological settings. Firstly, levels of serum and peritoneal pAb were measured during<br />early experimental secondary CE, using both high (Balb/c) and low (C57Bl/6) susceptible<br />mouse strains. Serum pAb mostly differed in normal mice, being pAb levels<br />of IgG subclasses with poor anti-parasite activities predominant in Balb/c animals.<br />Conversely, peritoneal pAb isotypes/subclasses with efficient anti-parasite activities<br />predominated in normal and infected C57Bl/6 mice. Secondly, sera from potentially<br />resistant patients, susceptible individuals and healthy donors were analysed, showing<br />higher pAb levels of the IgA and IgG—particularly IgG1—isotypes in potentially resistant<br />individuals compared to control groups. Finally, since remarkable differences<br />were observed in pAb profiles according to the intrinsic host susceptibility to the<br />infection, we proposed here that pA
