A summary of the resource.
Dendritic cell-specific ICAM-3 grabbing non-integrin (DC-SIGN) expressed on a variety of DCs, is a C-type lectin receptor that recognizes glycans on a diverse range of pathogens, including parasites. The interaction of DC-SIGN with pathogens triggers specific signaling events that modulate DC-maturation and activity and regulate T-cell activation by DCs. In this work we evaluate whether F. hepatica glycans can immune modulate DCs via DC-SIGN. We demonstrate that DC-SIGN interacts with F. hepatica glycoconjugates through mannose and fucose residues. We also show that mannose is present in high-mannose structures, hybrid and trimannosyl N-glycans with terminal GlcNAc. Furthermore, we demonstrate that F. hepatica glycans induce DC-SIGN triggering leading to a strong production of TLR-induced IL-10 and IL-27p28. In addition, parasite glycans induced regulatory DCs via DC-SIGN that decrease allogeneic T cell proliferation, via the induction of anergic/regulatory T cells, highlighting the role of DC-SIGN in the regulation of innate and adaptive immune responses by F. hepatica. Our data confirm the immunomodulatory properties of DC-SIGN triggered by pathogen-derived glycans and contribute to the identification of immunomodulatory glyans of helminths that might eventually be useful for the design of vaccines against fasciolosis.

Glycans present in biological glycoconjugates have several structural and functional roles. Elucidation of glycan structure and biological function is critical to understand their role in physiological and pathogenic process, enabling the development of diagnostic methods and disease treatment. Immobilized glycosidases are powerful tools for glycan analysis, as they are able to remove specific carbohydrates without altering the protein structure. Here we describe the individual immobilization of Aspergillus oryzae β-galactosidase and Canavalia ensiformis α-mannosidase onto agarose and silica magnetic nanoparticles activated with cyanate ester groups. High immobilization yields (70–90%) were achieved, keeping above 60% of its original activity. Immobilized glycosidases were effective in the selective deglycosylation of model glycoproteins and a Fasciola hepatica lysate, evidenced by a decrease in specific lectin recognition of 40–50% after enzymatic deglycosylation. Immobilized glycosidases were reused for several deglycosylation cycles without loss of effectiveness. Their use was extended to the elucidation of the glycan role of native glycoconjugates. A decrease in the recognition of lactoferrin treated with α-mannosidase by a C-type lectin receptor, DC-SIGN was found. Also the specific deglycosylation of a F. hepatica lysate demonstrated the relevance of mannosylated glycans in the induction of Th2/Treg immune responses during the infection. Our results show successful immobilization of specific glycosidases in nano-supports and validate their utility to identify glycans biological functions.

We recently reported the structures of alfa-dictalediol monoacetate |b beta-dictalediol monoacetate |b and chromophycadiol monoacetate |b tricarbocyclic diterpenoids from a Canary Islands brown alga Dictiota sp. |b whose structures differ considerably from already-known diterpenes of terrestrial or marine origen. As a result of intensive investigation in our laboratory |b the biosynthetic pathway for these diterpenes is now reasonably well understood. This work includes the structural determination of five new diterpenes |b the precise structure of one of them determined by single-crystal X-ray diffraction analyses.

Los aceites de pescado son la principal fuente de EPA y DHA en la dieta. Estos acidos grasos de la famnia w3 tienen efectos importantes sobre el metabolismo y se recomienda aumentar su ingesta. Se estudiaron las fuentes de EPA y DHA disponibles para el consumidor: filetes de pescado y aceite de pescado encapsulado. Los pescados habitual mente consumidos en Montevideo son de especies consideradas magras (clasificaci6n correspondiente al mes de julio), y se requieren entre 100 y 400 gramos de filetes crudos para consumir 1 .gramo de EPA + DHA. Tambien se estudi6 la composici6n de aceites de pescado comerciales y de tipo comestible. Una alternativa a la inclusi6n de pescado en la dieta son los concentrados de w3. Para lograr un alto % de estos acidos grasos en el preparado, es necesario romper el enlace ester de los trigliceridos y fraccionar entonces 10s acidos grasos libres 0 sus derivados. El metodo elegido para esto fue la formaci6n de aductos de urea. La caracteristica de los compuestos de inclusi6n con urea es que s610 aquellos compuestos que encajan en ellumen de los cristales de urea pueden formarlos. Los polienos de aceites de pescado, al tener 5 6 6 insaturaciones de configuraci6n cis e interrumpidas por un metileno, tienen una mayor dificultad para formar los aductos que los acidos grasos saturados. Se estudi6 la reacci6n de complejaci6n con urea como forma de preparar concentrados de w3. La temperatura de reacci6n y la cantidad de solvente utilizada no son factores tan determinantes como la relaci6n urea:acidos graso (peso:peso de acidos grasos complejeables) para lograr un alto % de EPA + DHA en la FNCU. Variando las condiciones de reacci6n se logra recuperar selectivamente el DHA. El interes en los acidos grasos w3 ha dado lugar a la revisi6n de las tecnicas de anaIisis de composici6n de aceites de pescado. Se optimiz6 el uso del equipo cromatografico disponible. Un aspecto importante de la quimica de los aceites de pescado para uso alimentarlo es la prevenci6n de su deterioro oxidativo. Se estudi6 la estabilidad oxidativa del aceite de Higado de Merluza por medida del periodo de inducci6n en un equipo Rancimat. Se estabiliz6 el aceite con varios antioxidantes, siendo el mas eficiente el TBHQ a 200 ppm y a 100 ppm.

Nowadays, fertilization and pest control are carried out using chemical compounds that contaminate soil and deteriorate human health. Plant growth promoting bacteria endophytes (PGPBEs), are a well-studied group of bacteria that offers benefits to the host plant, such as phytostimulation, biofertilization, and protection against other microorganisms. The study of Gluconacetobacter diazotrophicus–which belongs to PGPBEs-aids the development of alternative strategies of an integrated approach for crop management practices. Ralstonia solanacearum is responsible for bacterial wilt disease. This phytopathogen is of great interest worldwide due to the enormous economic losses it causes. In this study the action of G. diazotrophicus as a growth promoting bacterium in Arabidopsis thaliana seedlings is analyzed, evaluating the antagonistic mechanisms of this beneficial endophytic bacterium during biotic stress produced by R. solanacearum. Effective colonization of G. diazotrophicus was determined through bacterial counting assays, evaluation of anatomical and growth parameters, and pigments quantification. Biocontrol assays were carried out with Ralstonia pseudosolanacearum GMI1000 model strain and R. solanacearum A21 a recently isolated strain. Inoculation of A. thaliana (Col 0) with G. diazotrophicus Pal 5 triggers a set of biochemical and structural changes in roots, stems, and leaves of seedlings. Discrete callose deposits as papillae were observed at specific sites of root hairs, trichomes, and leaf tissue. Upon R. pseudosolanacearum GMI1000 infection, endophyte-treated plants demonstrated being induced for defense through an augmented callose deposition at root hairs and leaves compared with the non-endophyte-treated controls. The endophytic bacterium appears to be able to prime callose response. Roots and stems cross sections showed that integrity of all tissues was preserved in endophyte-treated plants infected with R. solanacearum A21. The mechanisms of resistance elicited by the plant after inoculation with the endophyte would be greater lignification and sclerosis in tissues and reinforcement of the cell wall through the deposition of callose. As a consequence of this priming in plant defense response, viable phytopathogenic bacteria counting were considerably fewer in endophyte-inoculated plants than in not-inoculated controls. Our results indicate that G. diazotrophicus colonizes A. thaliana plants performing a protective role against the phytopathogenic bacterium R. solanacearum promoting

<p>This study presents a novel application of the heterogeneous solar photo-Fenton process using an iron-pillared clay (Fe-PILC) as a photocatalyst under natural sunlight. The Fe-PILC is characterized by thermogravimetric analysis, EDXRF, SEM and nitrogen adsorption–desorption isotherms. The solar photo-reactor consists of a tubular borosilicate tube with a compound parabolic collector (CPC) and two different materials as reflective material for performance comparison. Phenol is used as model pollutant. The catalytic tests are monitored by the<br />concentration of the pollutant, intermediates and the iron leaching, as well as the accumulated energy during the reaction time. Complementary catalytic tests allowed to establish the influence of parallel processes that could contribute to phenol removal. Experimental data proved that the Fe-PILC is active as heterogenous catalyst in photo-Fenton like processes promoted by natural solar radiation. The catalyst showed good stability in the process and allowed a 99%+ removal of the pollutant and its intermediates. Fenton, photolysis and adsorption do not contribute significantly to phenol removal. Finally, when comparing different reflective materials in the CPC, the aluminium sheet proved to be more efficient than the polyester film metallized with aluminium.</p>

Anecdotal reports and open-label case studies in humans indicated that the psychedelic alkaloid ibogaine exerts profound antiaddictive effects. Ample preclinical evidence demonstrated the efficacy of ibogaine, and its main metabolite, noribogaine, in substance-use-disorder rodent models. In contrast to addiction research, depression-relevant effects of ibogaine or noribogaine in rodents have not been previously examined. We have recently reported that the acute ibogaine administration induced a long-term increase of brain-derived neurotrophic factor mRNA levels in the rat prefrontal cortex, which led us to hypothesize that ibogaine may elicit antidepressant-like effects in rats. Accordingly, we characterized behavioral effects (dose- and time-dependence) induced by the acute ibogaine and noribogaine administration in rats using the forced swim test (FST, 20 and 40 mg/kg i.p., single injection for each dose). We also examined the correlation between plasma and brain concentrations of ibogaine and noribogaine and the elicited behavioral response. We found that ibogaine and noribogaine induced a dose- and time-dependent antidepressant-like effect without significant changes of animal locomotor activity. Noribogaine's FST effect was short-lived (30 min) and correlated with high brain concentrations (estimated >8 μM of free drug), while the ibogaine's antidepressant-like effect was significant at 3 h. At this time point, both ibogaine and noribogaine were present in rat brain at concentrations that cannot produce the same behavioral outcome on their own (ibogaine ∼0.5 μM, noribogaine ∼2.5 μM). Our data suggests a polypharmacological mechanism underpinning the antidepressant-like effects of ibogaine and noribogaine.

Transforming lignocellulosic biomass into C5 and C6 sugars suitable to produce biofuels, building blocks, and high-value-added<br />compounds is a key aspect of sustainable strategies and is central to the biorefinery concept. Xylan is found acetylated and bound<br />to cellulose and lignin forming an insoluble complex in nature, and its degradation involves a collection of enzymes acting<br />together. To gain a better understanding of this process, the present study focuses on the elucidation of the main products<br />resulting from the hydrolysis of delignified Eucalyptus dunnii bark by an enzymatic extract from Pseudozyma sp. with xylanase<br />and acetylxylan esterase activities but no cellulase activity. Scanning electronmicroscopy (SEM) studies of the insoluble fraction<br />after hydrolysis revealed cracking on the surface of the material. The enzymatic activity of the crude yeast extract was evidenced<br />by TLC and HPLC analysis of the hydrolysate, which allowed us to detect xylose, acetylxylobiose, and acetic acid. Finally, the<br />principal low molecular weight products obtained from this process were characterized by nuclear magnetic resonance (NMR)<br />spectroscopy as xylose and 3-O-acetylxylobiose. Based on these spectroscopic and chromatographic results, it was possible to<br />estimate a 4:1 ratio of xylose to 3-O-acetylxylobiose. These results highlight the importance of using an enzymatic system for<br />effective xylan degradation.

Forestry and agricultural industries constitute highly relevant economic activities globally. They generate large amounts of residues rich in lignocellulose that have the potential to be valorized and used in different industrial processes. Producing renewable fuels and high-value-added compounds from lignocellulosic biomass is a key aspect of sustainable strategies and is central to the biorefinery concept. In this study, the use of biomass-derived monosaccharides for the enzymatic synthesis of sugar fatty acid esters (SFAEs) with antimicrobial activity was investigated to valorize these agro-industrial residues. With the aim to evaluate if lignocellulosic monosaccharides could be substrates for the synthesis of SFAEs, D-xylose, L-arabinose, and D-glucose, lauroyl and stearoyl monoesters were synthetized by transesterification reactions catalyzed by Lipozyme RM IM as biocatalyst. The reactions were performed using commercial D-xylose, L-arabinose, and D-glucose separately as substrates, and a 74:13:13 mixture of these sugars. The proportion of monosaccharides in the latter mixture corresponds to the composition found in hemicellulose from sugarcane bagasse<br />and switchgrass, as previously described in the literature. Products were characterized using nuclear magnetic resonance (NMR) spectroscopy and showed that only the primary hydroxyl group of these monosaccharides is involved in the esterification reaction. Antimicrobial activity assay using<br />several microorganisms showed that 5-O-lauroyl-D-xylofuranose and 5-O-lauroyl-L-arabinofuranose have the ability to inhibit the growth of Gram-positive bacteria separately and in the products mix. Furthermore, 5-O-lauroyl-L-arabinofuranose was the only product that exhibited activity against Candida albicans yeast, and the four tested filamentous fungi. These results suggest that sugar fatty acid esters obtained from sustainable and renewable resources and produced by green methods are promising antimicrobial agents.

Although several efforts to promote breastfeeding have been implemented worldwide, the prevalence of exclusive breastfeeding in the first 6 months of life remains low. One of the factors responsible for this trend is the widespread sales and marketing of breast-milk substitutes. In this context, the present work aimed at exploring mothers and health professionals' social representations of breastfeeding and infant formula. The study was conducted in Montevideo (Uruguay), as part of the periodic assessment performed by the Uruguayan government to monitor the marketing of breast-milk substitutes according to the OMS/UNICEF NetCode toolkit. A total of 330 mothers of children under 24 months old and 154 health professionals were recruited at 33 health care facilities. Participants were asked to complete two word association tasks, one with breastfeeding and the other with infant formula. Inductive coding was used to classify the responses into categories and dimensions. The structure of the social representations was explored considering the frequency of mention and the average rank of appearance of the categories. Results showed that mothers’ social representations mainly positioned breastfeeding as a feeding practice, whereas infant formulas were mainly perceived as a complement to breast-milk that provides support. In the case of health professionals, the social representations of breastfeeding and infant formulas were in line with scientific evidence and health recommendations. These results suggest that efforts to promote breastfeeding should focus on raising public awareness of the risks associated with breast-milk substitutes and on overcoming the systemic barriers to breastfeeding, particularly among working mothers.

<p>En el presente estudio preliminar se analiza el contenido en minerales de uno de los residuos más voluminosos de la industria alimentaria nacional; los sueros provenientes de la elaboración de quesos y manteca.</p>
<p>Se señala la importancia que, desde el punto de vista trófico tiene tanto la composición como el tenor de minerales en los sueros lácteos.</p>
<p>De los resultados obtenidos hasta el presente se deduce el interés que tiene un futuro estudio en profundidad del tema, a fin de considerar un aprovechamiento integral de estos residuos para alimentación humana.</p>

Plant-herbivore interactions have been extensively studied in tomato plants and their most common pests. Tomato plant chemical defenses, both constitutive and inducible, play a role in mediating these interactions. Damaged tomato plants alter their volatile profiles, affecting herbivore preferences between undamaged and damaged plants. However, previous studies on tomato volatiles and herbivore preferences have yielded conflicting results, both in the volatile chemistry itself as well as in the attraction/repellent herbivore response. This study revisits the volatile-mediated interactions between tomato plants and two of their main herbivores: the leafminer Tuta absoluta and the whitefly Trialeurodes vaporariorum. Tomato plant volatiles were analyzed before and after damage by each of these herbivores, and the preference for oviposition (T. absoluta) and settling (T. vaporariorum) on undamaged and damaged plants was assessed both after conspecific and heterospecific damage. We found that both insects consistently preferred damaged plants over undamaged plants. The emission of herbivore-induced plant volatiles (HIPVs) increased after T. absoluta damage but decreased after T. vaporariorum damage. While some of our findings are in line with previous reports, T. absoluta preferred to oviposit on plants damaged by conspecifics, which differs from earlier studies. A comparison of HIPVs emitted after damage by T. absoluta and T. vaporariorum revealed differences in up- or down-regulation, as well as significant variations in specific compounds (12 for T. absoluta and 26 for T. vaporariorum damaged-plants). Only two compounds, β-caryophyllene and tetradecane, significantly varied because of damage by either herbivore, in line with the overall variation of the HIPV blend. Differences in HIPVs and herbivore preferences may be attributed to the distinct feeding habits of both herbivores, which activate different defensive pathways in plants. The plant’s challenge in simultaneously activating both defensive pathways may explain the preference for heterospecific damaged plants found in this study, which are also in line with our own observations in greenhouses.

Abstract
A summary of the resource.
Research background. New sources of docosahexaenoic acid have recently been investigated aiming at infant formula fortification and dietary supplementation, among which the single cell oil with 40–50 % of this acid. Experimental approach. For this purpose, such an oil was blended with caprylic acid in amount substance ratio ranging from 1:1 to 5:1 and the blends were interesterified using either Novozym 435 or Lipozyme TL IM as the catalyst. The influence of the amount of excess free caprylic acid in the substrate, as well as the type of enzyme on the triacylglycerol rearrangement resulting from the synthesis of the structured lipids were evaluated. Results and conclusions. The regiospecific lipase Lipozyme TL IM seemed to induce transesterification among single cell oil triacylglycerols preferably by acidolysis with caprylic acid, which was directly proportional to the ratio of this acid in the substrate. In reactions catalyzed by the non-regiospecific lipase Novozym 435, a higher incorporation of caprylic acid into single cell oil triacylglycerols was observed than when using Lipozyme TL IM, independently of the oil/caprylic acid molar ratio. Novelty and scientific contribution. These results revealed the importance of combining the choice of the type of lipase, either regiospecific or not, with the amount ratios of free fatty acids and the substrate in acidolysis when aiming to produce structured lipids as a source of docosahexaenoic acid.

The structures and molecular interactions of established synthetic chalcones were correlated with their release profiles from asolectin liposomes. The effects of chalcones on the properties of liposomes were evaluated by dynamic light scattering (DLS), ultraviolet-visible spectroscopy (UV-VIS), horizontal attenuated total reflection Fourier transform infrared (HATR-FTIR), 31P nuclear magnetic resonance (31P NMR), zeta (ζ) potential and differential scanning calorimetry (DSC). The profiles and mechanisms of release were accessed according to the Korsmeyer-Peppas model. Results obtained allowed the establishment of a relationship between the chalcone release profile and 1) the ordering effects of chalcones in different membrane regions, 2) their polar or interfacial location in the lipid layer, 3) the influence of hydroxy and methoxy substituents, 4) their effect on reorientation of lipid choline-phosphate regions. The obtained data may improve the development of chalcone-based systems to be used in the therapy of chronic and acute diseases.

A summary of the resource.
<p>Syringic and cinnamic acids were previously identified as the major components on Rapanea ferruginea0s leaf extract. In this work, these phenolic acids were investigated and correlated in two aspects. The first one was referred to their role against DPPH and hydroxyl radicals, considering the concentration detected in R. ferruginea extract. The second one considered their effects on the physico-chemical properties of soy asolectin liposomes, such as hydration degree, trans-gauche isomerization, vibrational, rotational and translational motions, state-of-phase, size and surface charge. The half-maximum inhibitory concentration (IC50) related to syringic acid in DPPH assays was detected as 7.36 � 10�3 mM.<br />At the cinnamic acid highest tested concentration (8.23 � 10�3 mM), the activity against DPPH was<br />around 4.6%. Regarding hydroxyl radical-induced lipid peroxidation, the IC50 detected for syringic acid free in solution (free-form) was equivalent to 0.082 � 10�3 mM, a value 3.6 times lower than the detected for its liposome-loaded form. At the highest tested concentration (0.24 � 10�3 mM), the free-form cinnamic acid inhibited lipid peroxidation by 4 times the observed to its liposome-loaded form. Results showed that hydrogen donation is not the preferential mechanism in which the tested phenolic acids inhibit hydroxyl radical-induced lipid peroxidation. Syringic acid showed higher antioxidant potential than cinnamic acid, probably having a primordial role in the properties related to R. ferruginea. Liposomes’ physico-chemical data provided correlations between the phenolic acids’ antioxidant potential and their influence in the mobility of specific lipid groups. The findings of this work will contribute to the design and development of phenolic acids-based and plant extract-based pharmacological systems.</p>

Complexes [RuCl(H4NO2Fo4M)(bipy)(dppb)]PF6 (1), [RuCl(H4NO2Fo4M)(Mebipy)(dppb)]PF6 (2), [RuCl (H4NO2Fo4M)(phen)(dppb)]PF6 (3), [RuCl(H4NO2Ac4M)(bipy)(dppb)]PF6 (4), [RuCl(H4NO2Ac4M) (Mebipy)(dppb)]PF6 (5) and [RuCl(H4NO2Ac4M)(phen)(dppb)]PF6 (6) with N4-methyl-4-nitrobenzalde hyde thiosemicarbazone (H4NO2Fo4M) and N4-methyl-4-nitroacetophenone thiosemicarbazone (H4NO2 Ac4M) were obtained from [RuCl2(bipy)(dppb)], [RuCl2(Mebipy)(dppb)], and [RuCl2(phen)(dppb)], (dppb ¼ 1,4-bis(diphenylphospine)butane; bipy ¼ 2,20-bipyridine; Mebipy ¼ 4,40-dimethyl-2,20-bipyridine; phen ¼ 1,10-phenanthroline). In all cases the thiosemicarbazone is attached to the metal center through the sulfur atom. Complexes (1e6), together with the corresponding ligands and the Ru precursors were evaluated for their ability to in vitro suppress the growth of Trypanosoma cruzi. All complexes were more active than their corresponding ligands and precursors. Complexes (1e3) and (5) revealed to be the most active among all studied compounds with ID50 ¼ 0.6e0.8 mM. In all cases the association of the thiosemicarbazone with ruthenium, dppb and bipyridine or phenanthroline in one same complex proved to be an excellent strategy for activity improvement.
