Polyreactive antibodies (pAb) bind to a broad range of unrelated structures, providing hosts with functional components able to rapidly recognize and protect against different pathogens. However, their roles against helminth parasites are still unexplored. Here, pAb profiles were analysed in cystic echinococcosis (CE), a zoonosis caused by the cestode Echinococcus granulosus sensu lato. Levels of anti-DNP (2,4-dinitrophenyl-hapten) antibodies were measured as a surrogate parameter of pAb in different biological settings. Firstly, levels of serum and peritoneal pAb were measured during early experimental secondary CE, using both high (Balb/c) and low (C57Bl/6) susceptible mouse strains. Serum pAb mostly differed in normal mice, being pAb levels of IgG subclasses with poor anti-parasite activities predominant in Balb/c animals. Conversely, peritoneal pAb isotypes/subclasses with efficient anti-parasite activities predominated in normal and infected C57Bl/6 mice. Secondly, sera from potentially resistant patients, susceptible individuals and healthy donors were analysed, showing higher pAb levels of the IgA and IgG—particularly IgG1—isotypes in potentially resistant individuals compared to control groups. Finally, since remarkable differences were observed in pAb profiles according to the intrinsic host susceptibility to the infection, we proposed here that pAb might be considered as potential humoral biomarkers for host resistance to CE. The aim of the present work was to study the hydrogel-forming polysaccharides carrageenan, guar and xanthan gum as potential absorbents in moisture activated dry granulation (MADG). Preferences of formulators regarding absorbents tend to incline towards microcrystalline cellulose and colloidal silicon dioxide. Besides, only few works exploring modified drug release achieved by MADG are reported in literature. A factorial design of experiments was introduced to study the effect of formulation variables on granulates and their tablets. Contact angle and drop penetration time provided useful information for guidance on excipient selection. Metronidazole was used as a highly soluble model drug to verify the release performance achieved with the polysaccharides. All of the granulates yielded tablets with appropriate physical properties, and significantly different disintegration behaviors and drug release profiles. Absorbent, binder and their mutual interaction had significant effects on formulation performance. The type of absorbent affected tablet disintegration time and drug release. The presence of copovidone, alone or in combination with povidone, proved to be a good choice for granulating a high load of not directly compressible materials by MADG. Carrageenan, guar and xanthan gum should be regarded as promising moisture absorbents for either immediate or extended release tablet formulations produced by MADG, an energy and time efficient granulation process. A broader range of absorbents could provide more versatility in MADG formulation and this advantageous process become more eligible for producing modified release products. Dietary PUFAs (polyunsaturated fatty acids) co-ordinately suppress transcription of a group of hepatic genes encoding glycolytic and lipogenic enzymes. Suppression of Fasn (fatty acid synthase) transcription involves two PUFA-responsive regions, but the majority of PUFA sensitivity maps to a region within the proximal promoter containing binding sites for NF-Y (nuclear factor-Y), Sp1 (stimulatory protein 1), SREBP (sterol-regulatory-elementbinding protein), and USF (upstream stimulatory factor). Promoter activation assays indicate that altered NF-Y is the key component in regulation of Fasn promoter activity by PUFA. Using electrophoretic mobility-shift assay and chromatin immunoprecipitation analysis, we demonstrate for the first time that PUFAs decrease in vivo binding of NF-Y and SREBP-1c to the proximal promoter of the hepatic Fasn gene and the promoters of three additional genes, spot 14, stearoyl-CoA desaturase and farnesyl diphosphate synthase that are also down-regulated by PUFA. The comparable 50% decrease in NF-Y and SREBP-1c binding to the promoters of the respective PUFA-sensitive genes occurred despite no change in nuclear NF-Y content and a 4-fold decrease in SREBP-1c. Together, these findings support a mechanism whereby PUFA reciprocally regulates the binding of NF-Y and SREBP-1c to a subset of genes which share similar contiguous arrangements of sterol regulatory elements and NF-Y response elements within their promoters. PUFA-dependent regulation of SREBP-1c and NF-Y binding to this unique configuration of response elements may represent a nutrient-sensitive motif through which PUFA selectively and co-ordinately targets subsets of hepatic genes involved in lipid metabolism. Nevirapine (NVP) extensive data obtained after oral single dose administration of 200 mg in a crossover study involving 16 healthy subjects was used to develop a descriptive population pharmacokinetic model including drug recirculation, since secondary peaks were observed in plasma concentration-time profiles for all subjects. Through implementation of model event time feature in NONMEM 7.3.0, a simple mechanistic model physiologically consistent with the process of drug cycling was able to describe multiple peaks phenomena and quantify its pharmacokinetic parameters, achieving a better performance than its analogue conventional one. Absorption process, between subject and-between occasion variability of pharmacokinetic parameters was also assessed. Estimated mean fraction of NVP bioavailable dose undergoing recirculation process was 51.6 %, a magnitude which could hardly be explained by enterohepatic cycling. In this work we propose an alternative disposition process to explain NVP drug recirculation: gastric secretion with posterior intestinal reabsorption. Due to physicochemical and pharmacokinetic properties of the drug, this neglected phenomenon is more likely to explain the results obtained, and might be present in disposition of several basic drugs. Hemos caracterizado la cinética de reacción de un conjunto de porfirinas de manganeso (MnPorfirinas) solubles en agua frente a peroxinitrito (ONOO–) y radical carbonato (CO3·–). La reacción, entre ONOO– y el complejo de MnIII, tiene constantes de velocidad entre 105 y 107 M-1 s-1 a 37 ºC con un comportamiento sigmoide en función del pH, pKa aparentes cercanos al del par ONOOH/ONOO– y máximos a pH alcalino. La reacción produce el complejo O=MnIV y ·NO2. Los valores de estas constantes de velocidad tienen una relación lineal de energía libre con parámetros fisicoquímicos de la porfirina tales como el potencial redox del par MnIII/MnII o la acidez de Brønsted de los nitrógenos pirrólicos o de las moléculas de agua axiales. La reacción, entre ONOO– y el complejo de MnII, tiene constantes de velocidad <106 M-1 s-1 a pH 7.4 y 37 ºC y produce estequiométricamente O=MnIV y NO2–. La reacción, entre CO3·– y el complejo de MnIII tiene constantes de velocidad extrapoladas a pH neutro entre 2 y 12 x 108 M-1 s-1 con un comportamiento ácido base congruente con la ionización de las moléculas axiales de agua presentes en el complejo. La reacción entre CO3·– y el complejo de MnII tiene constantes de velocidad a pH = 10.4 entre 1 y 5 x 109 M-1 s-1. En ambos casos el CO3·– produce oxidaciones por un electrón. Los complejos de O=MnIV se reducen en forma rápida con ascorbato o urato hasta MnIII en presencia de oxígeno y, con bajas concentraciones de O2, la reducción puede llegar hasta MnII si los reductores son flavoenzimas reducidas, entre ellas succinato deshidrogenasa y NADH deshidrogenasa de la cadena mitocondrial de transporte de electrones. Estas reacciones de reducción completan dos posibles ciclos para catalizar la reducción de ONOO– o CO3·– con una eficiencia suficiente para proteger blancos biológicos en concentraciones micromolares de MnPorfirinas. Se presentan resultados de uso del ciclo catalítico MnIII/MnIV con urato como reductor en la protección de LDL in vitro y del ciclo catalítico MnII/MnIV con succinato/succinato deshidrogenasa como reductor para proteger actividades enzimáticas de partículas submitocondriales in vitro. Pseudocapacitive materials can enhance the energy storage performance of supercapacitors by making use of surface redox reactions. In recent years, different iron compounds have been investigated as pseudocapacitive materials, showing promising features for supercapacitor electrode applications. Carbon nanocomposites containing iron/phosphorus compounds have been prepared from porous carbon, followed by thermal treatment at different temperatures (700°C to 1000°C). The obtained supercapacitor electrodes were evaluated by electrochemical analyses using sulfuric acid electrolyte. The as-prepared nanocomposite contained nanostructured iron oxides or oxyhydroxides, whereas the nanocomposites prepared at 700°C to 900°C were composed of nanostructured iron phosphates. On the other hand, heat treatment at 1000°C caused the formation of nanocrystalline iron phosphides (mostly Fe2P nanoparticles). The Fe-containing samples showed enhanced specific capacitance (246 F g−1 to 447 F g−1 at 10 A g−1), which can be related to the pseudocapacitive contribution of the iron compounds. The sample heat treated at 1000°C exhibited favorable electrochemical performance, showing high electrical capacitance and good rate capability at 40 A g−1. These results reveal that porous carbon/iron phosphide nanocomposites are promising materials for use in supercapacitor electrode applications. Herein, we study new fullerene receptors which are constructed employing porphyrins, corannulene pincers and metallic centers. Theoretical calculations indicated that if porphyrin and corannulene pincers are merged, the strongest hosts for fullerenes can be built. We found that there is a linear increase of the interaction energy with respect to the number of pincers. The linear trend is broken when the pincers interact amongst themselves. The strongest host–C60 interaction was observed for the porphyrin which has 4 corannulene pincers and Ir or Rh centers, which exhibited interaction energies significantly larger than the one computed for the dimeric porphyrins. In addition, we report that some of the porphyrin receptors can host two fullerene molecules very efficiently, given that the host can be deformed without increasing its energy too much. Finally, we observed a good sensitivity of our hosts towards C70 with respect to C60. Synthetic routes of the hosts designed are discussed. The occurrence of ripples in an eight‐atom vacancy graphene system was analyzed through first principles calculations based on density functional theory. The study focuses on the conditions that should be considered in order that ripples could occur in the single layer defected graphene. The vacancies concentration, especially the pentagonal figures formed after relaxation of the vacant system, and the distances between neighbor pentagons, are found to be key aspects for rippling occurrence, with no charge distribution consequence. Two different configurations of rippling have been found where the position of the pentagonal figures is discussed as the driving force for any of them to occur. In this report, we used ab initio calculations in order to study the effect of Pt doping in monolayer MoS2. The Pt doping introduces important distortions at the geometrical levels, that induces the appearance of impurity levels, expressed as flat bands, in the proximity of the Fermi level, thus reducing the band gap. The vibrational analysis confirms the reduction of symmetry, and the simulated off-resonance Raman spectra presents important differences when comparted with pristine MoS2 monolayer, with relevant bands located at &sim;50, 150, 220, 300 and 330 cm−1. All this confirm that Pt doping could be possible, allowing electronic and vibrational tuning of their properties and this work provides the tool for a potential identification of these modifications at the experimental level. Persulfides (RSSH/RSS− ) are species closely related to thiols (RSH/RS− ) and hydrogen sulfide (H2S/HS− ), and can be formed in biological systems in both low and high molecular weight cysteine-containing compounds. They are key intermediates in catabolic and biosynthetic processes, and have been proposed to participate in the transduction of hydrogen sulfide effects. Persulfides are acidic, more acidic than thiols, and the persulfide anions are expected to be the predominant species at neutral pH. The persulfide anion has high nucleophilicity, due in part to the alpha effect, i.e., the increased reactivity of a nucleophile when the neighboring atom has high electron density. In addition, persulfides have electrophilic character, a property that is absent in both thiols and hydrogen sulfide. In this article, the biochemistry of persulfides is described, and the possible ways in which the formation of a persulfide could impact on the properties of the biomolecule involved are discussed. The saliva/plasma ratio showed a higher value during the first two hours and after food intake than that observed between 2 and 5 hours.Bile was collected in five patients with bladder lithiasis |b during the operation.Forty miligrams of total Paracetamol (free and conjugated) were detected in bile after hydrolysis with stomach homogenate of Helix pomatia L. All measurements were performed by reversed phase HPLC technique. A reabsorption of Paracetamol can be concluded after food intake |b probably due to an enterohepatic circulation of the drug. A simulation was done with population parameters of Paracetamol using a pharmacokinetic model. When a concentration of 1 × 108 cells ml−1 of the antagonist bacterium was applied to the fruit, disease incidence was reduced by 95%, and the lesion diameter of anthracnose decreased by 85%, which offered greater protection than the synthetic fungicide. This is the first report of antagonistic mechanisms of the marine bacterium S. rhizophila against anthracnose disease in mango, which in this study was found to be more effective than the synthetic fungicide. Cyclic voltammetry and electron spin resonance techniques were used in the investigation of several potential antiprotozoal containing thiosemicarbazone and carbamate nitrofurans. In the electrochemical behaviour, a self-protonation process involving the nitro group was observed. The reactivity of the nitro anion radical for these derivatives with glutathione, a biological relevant thiol, was also studied in means of cyclic voltammetry. These studies demonstrated that glutathione could react with radical species from 5-nitrofuryl system. Furthermore, from the voltammetric results, some parameters of biological significance as E1 7 (indicative of the biological nitro anion radical formation), and KO2 (thermodynamic indicator the of oxygen redox cycling) have been calculated. We also evaluated the stability of the nitro anion radical in terms of the dimerization constant (kd). The nitrofuran-free radicals from cyclic voltammetry were characterized by electron spin resonance. A clear dependence between both the thiosemicarbazone or carbamate substructure and the length of the linker, furyl- or furylpropenyl-spacer, and the delocalization of the unpaired electron was observed. In the search for new therapeutic tools against Chagas disease (American trypanosomiasis) palladium and platinum complexes of the bioactive ligand pyridine-2-thiol N-oxide were exhaustively characterized and evaluated in vitro. Both complexes showed high in vitro growth inhibition activity (IC50 values in the nanomolar range) against Trypanosoma cruzi, the causative agent of the disease. They were 39–115 times more active than the antitrypanosomal drug Nifurtimox. The palladium complex showed an approximately threefold enhancement of the activity compared with the parent compound. In addition, owing to their low unspecific cytotoxicity on mammalian cells, the complexes showed a highly selective antiparasite activity. To get an insight into the mechanism of action of these compounds, DNA, redox metabolism (intraparasite free-radical production) and two parasite-specific enzymes absent in the host, namely, trypanothione reductase and NADH-fumarate reductase, were evaluated as potential parasite targets. Additionally, the effect of metal coordination on the free radical scavenger capacity previously reported for the free ligand was studied. All the data strongly suggest that trypanocidal action of the complexes could mainly rely on the inhibition of the parasite-specific enzyme NADH-fumarate reductase. To characterize novel tools for insect control, essential oils from two Asteraceae (Artemisia absinthium and Eupatorium buniifolium) were studied for their potential toxicity to beneficial insects [honey bees (Apis mellifera L.)] and phytotoxic effects against tomato seeds and vegetative parts. Our results show that seed germination was affected at application rates needed to control the leafminer T. absoluta but not at rates needed to control the whitefly T. vaporariorum. The same trend was found for honeybee toxicity: the use of these essential oils at the amounts needed to control T. vaporariorum would be not acutely toxic for bees. Finally, an experimental greenhouse trial showed that the application of the essential oil from E. buniifolium at 3% on whitefly-infested plants can cause whitefly adult mortality without affecting the crop yield. In the search for new metal-based drugs against diseases produced by trypanosomatid parasites, four organoruthenium(II) compounds [Ru2(p-cymene)2(L)2] X2, where L are bioactive 5-nitrofuryl-containing thiosemicarbazones and X=Cl or PF6, had been previously obtained. These compounds had shown activity on Trypanosoma brucei, the etiological agent of African trypanosomiasis. Because of genomic similarities between trypanosomatides, these ruthenium compounds were evaluated, in the current work, on Trypanosoma cruzi, the parasite responsible of American trypanosomiasis (Chagas disease). Two of them showed significant in vitro growth inhibition activity against the infective trypomastigote form of T. cruzi (Dm28c clone, IC50=11.69 and 59.42 μM for [Ru2(p-cymene)2(L4)2]Cl2 and [Ru2(p-cymene)2(L1)2]Cl2, respectively, where HL4=5- nitrofuryl-N-phenylthiosemicarbazone and HL1=5- nitrofurylthiosemicarbazone), showing fairly good selectivities toward trypanosomes with respect to mammalian cells (J774 murine macrophages). Moreover, [Ru2 (p-cymene)2(L2)2]Cl2, where HL2=5-nitrofuryl-Nmethylthiosemicarbazone, was synthesized in order to evaluate the effect of improved solubility on biological behavior.
